    بیماری کپک خاکستری با عامل Botrytis cinerea یکی از بیماری های مهم پس از برداشت به ویژه در میوه سیب و توت فرنگی است. به منظور مهار زیستی این بیمارگر از 14 جدایه مخمری استفاده شد. جدایه ها از منابع طبیعی و گیاهان جداسازی شدند. پس از شناسایی مولکولی، جدایه ها برای ارزیابی توانایی مهار زیستی علیه B. cinerea در قالب طرح کاملا تصادفی با سه تکرار به روش کشت متقابل روی محیط کشت سیب زمینی دکستروز آگار مورد استفاده قرار گرفتند.
    مخمرهای شناسایی شده عبارت بودند از: Pichia kudriavzevii ، Pichia galeiformis، Galactomyces candidum، Meyerozma guilliermondii، Saccharomyces cerevisiae، Zygoascus meyerae، Pichia sp. ، Candida parapsilosis، Metschnikowia sp. ، Candida boidinii، Lecythophora sp. . و Candida catenulatea که بیشترین تاثیر در بازدارندگی را گونه G. candidum (85/47 درصد) و کمترین تاثیر را گونه P. kudriavzevii (53/6 درصد) داشت. نتایج نشان داد که اکثر مخمرهای آسکومیستی شناسایی شده قدرت مهار کنندگی این بیمارگر را با درصدهای مختلف از 48 - 7 داشتند.

    در تحقیق حاضر، بهینه سازی و مدل سازی فعالیت بازدارندگی رشد میسلیومی و سمیت تدخینی اسانس آویشن باغی با استفاده از روش سطح پاسخ، به ترتیب روی دو قارچ مهم بیماری زای Fusarium graminearum و Botrytis cinerea و سوسک زیان آور غلات (Rhyzopertha dominica F.) مورد بررسی قرار گرفت. همچنین اجزای شیمیایی اسانس مذکور با استفاده از دستگاه گاز کروماتوگرافی-طیف سنج جرمی بررسی شد و کارواکرول (99/27 درصد) ، تیمول (76/12 درصد) ، ژرانیول (91/8 درصد) و ژرانیل استات (60/7 درصد) به عنوان ترکیبات عمده آن شناسایی شدند. غلظت های مورد استفاده اسانس و افزایش زمان در معرض قرارگیری به صورت معنی داری باعث کاهش رشد میسلیومی قارچ ها و افزایش مرگ و میر حشره آفت شد. شرایط بهینه جهت حصول بیشینه بازدارندگی از رشد میسلیومی در قارچ F. graminearum (67/88 درصد) ، زمان 59/88 ساعت و غلظت 67/486 پی پی ام به دست آمد. همچنین زمان 01/74 ساعت و غلظت 96/552 پی پی ام، سبب ایجاد بیشترین بازدارندگی رشدی در قارچ B. cinerea (43/94 درصد) می شود. غلظت 10/43 میکرولیتر بر لیتر و مدت زمان 60 ساعت به عنوان شرایط بهینه برای دستیابی به مرگ و میر 23/72 درصدی سوسک زیان آور غلات برآورد شد. نتایج نشان دادند که اسانس آویشن باغی پتانسیل لازم را برای مدیریت قارچ های بیماری زای F. graminearum و B. cinerea و سوسک زیان آور غلات دارا می باشد.

    در حال حاضر، مدیریت مبارزه با نماتود مولد زخم ریشه ی چای (Pratylenchus loosi) ، با تکیه بر راه کارهای مهار زیستی، از اولویت های پژوهشی در محصول چای به حساب می آید. در این پژوهش، پس از نمونه برداری از خاک و ریشه ی درختچه های چای در بهار و تابستان 1392 در مناطق چای کاری استان های گیلان و مازندران، قارچ ها از خاک ناحیه فراریشه ی چای جداسازی شدند. قارچ های مورد آزمایش شامل Paecilomyces lilacinus (T1) و P. lilacinus (N1، N3) ، Fusarium culmurom، Clonostachys rosea و Acremonium strictum بودند. پس از شناسایی و خالص سازی جدایه ها و انجام آزمایش های مربوط به هم ستیزی در شرایط آزمایشگاهی (محیط کشت مایع زاپک داکس 34/3 درصد و مالت 5/1 درصد) نتایج نشان داد در محیط کشت مایع زاپک داکس، قارچ P. lilacinus (T1) پس از 72 ساعت، موجب غیر فعال شدن 01/52 درصد از لاروهای P. loosi گردید و همین جدایه تاثیر معنی داری روی مرگ ومیر بالغین از خود نشان نداد. در محیط کشت مایع مالت، قارچ P. lilacinus (T1) پس از 72 ساعت، 41/32 درصد از نوزادان و 54/30 درصد از بالغین P. loosi را غیر فعال نمود. در شرایط گلدان، آزمایشات همستیزی در سه حالت مایه زنی قارچ و نماتود همزمان، مایه زنی قارچ و دو هفته بعد مایه زنی نماتود و مایه زنی نماتود و دو هفته بعد مایه زنی قارچ، انجام شد. نتایج نشان داد که قارچ C. rosea در صورتی که دو هفته قبل از نماتود مایه زنی شود، می تواند در شاخص های وزن کل نهال، وزن تر ریشه، حجم ریشه و جمعیت نماتود در ریشه و خاک تاثیر قابل توجهی از خود نشان دهد

    ااکتینوباکتری ها به واسطه داشتن ویژگی‏های آنتاگونیستی بسیار در برابر طیف گسترده‏ای از بیمارگرهای گیاهی به ویژه قارچ‏ها، به عنوان عوامل بالقوه کنترل بیولوژیک توجهات زیادی را به خود معطوف کرده‏اند. Sclerotinia Sclerotiorum (Lib.) De Bary عامل پوسیدگی سفید ساقه خیار که دارای سه خصوصیت عمده چون دامنه میزبانی وسیع، توان بیماریزایی بالا و دوام طولانی اسکلروت‏ها در شرایط نامناسب محیطی می‏باشد یکی از مخرب‏ترین بیمارگرهای گیاهی در جهان است. با توجه به خسارت شدید و دشواری‏های کنترل توسط روش‏های شیمیایی، روش‏های جایگزینی مانند کنترل بیولوژک توسط گونه‏های اکتینوباکتربرای این بیمارگر مورد ارزیابی قرار گرفته است. در این پژوهش اثر آنتاگونیستی 109 جدایه اکتینوباکتر در برابر قارچ پوسیدگی سفید خیار بررسی گردید و سه جدایه UTS13، UTS19 و UTS49 با بیشترین میزان ممانعت از رشد پرگنه قارچ بیمارگر درون تشتک‏های پتری برای بررسی‏های بیشتر انتخاب شدند. خصوصیات فیزیولوژیکی جدایه‏های برتر حاکی تولید آنزیم‏های پروتئازی و کیتینازی بود. جدایه UTS13 به عنوان جدایه برتر در آزمون‏های گلخانه‏ای مورد شناسایی قرار گرفت و با توجه به توالی rDNA 16S مورد بررسی قرار گرفت و به عنوان Streptomyces albidoflavus شناسایی گردید. نتایج نشان داد که جدایه S. albidoflavus UTS 13 باعث کاهش علائم بیماری به میزان 50 درصد در شرایط گلخانه‏ای و افزایش صفات رشدی گیاه در مقایسه با تیمار شاهد سالم شد. پژوهش حاضر سعی بر معرفی باکتری S. albidoflavus UTS 13 به عنوان یک عامل بیوکنترل موثر به همراه طیف وسیعی از فعالیت‏های آنتاگونیستی در برابر قارچ بیمارگر S. sclerotiorum را داشته است.

    یکی از معیارهای مهم برای ارزیابی کارایی دشمنان طبیعی مطالعه ی ویژگی های رفتاری آن ها می باشد. در این بررسی ترجیح مرحله ی سنی طعمه، تغییر رفتار و تداخل لاروهای پشه ی Aphidoletes aphidimyza Rondani روی سنین مختلف پورگی شته ی جالیز، Aphis gossypii Glover بررسی شد. آزمایش ها در ظروف پتری پلاستیکی حاوی دیسک برگی خیار در اتاقک رشد انجام شد. بر اساس نتایج به دست آمده، اختلاف معنی داری از نظر ترجیح مراحل مختلف پورگی شته ی جالیز توسط لاروهای پشه ی A. aphidimyza وجود نداشت، اما بیش ترین بیوماس مصرف شده روی پوره های سن چهارم محاسبه شد. در آزمایش بررسی تغییر رفتار، در ترکیب های 20:40 و 30:30 (پوره ی سن سوم: سن اول) ، لارو شکارگر ترجیح بیش تری را به پوره ی سن سوم شته ی جالیز نشان داد. در بررسی تداخل لاروهای پشه ی A. aphidimyza، قدرت جستجوگری لارو شکارگر بین 0623/0 تا 1805/0 بر ساعت محاسبه شد که مقدار منفی شیب خط رگرسیون نشان دهنده ی بروز تداخل بین لاروهای پشه ی A. aphidimyza بود. نتایج این بررسی نشان داد که ویژگی های رفتاری لارو پشه ی A. aphidimyza می تواند تحت تاثیر تراکم آفت و نیز خود شکارگر قرار گیرد.

    قارچ Fusarium graminearum عامل بیماری بلایت فوزاریومی سنبله است، این بیماری یکی از بیمار ی های مهم غلات است و این قارچ باعث خسارت کمی و کیفی زیادی به گندم می شود. کاهش عملکرد محصول گندم به علت کاهش تعداد دانه ها و چروکیده شدن دانه ها است و کاهش کیفیت دانه به دلیل تجمع مایکوتوکسین ها (زهرابه های قارچی) در آنهاست. در این تحقیق اثر عصاره های شیرین بیان، دارچین و ریحان بر مهار رشد F. graminearum و بیان ژن های اصلی دخیل در زیست ساخت زیرالنون بررسی شد. پس از کشت F. graminearum در محیطPotato Dextrose Broth، حداقل غلظت بازدارندگی (MIC) رشد قارچ تحت اثر عصاره ها با استفاده از روش میکروتیتر پلیت اندازه گیری شد. همچنین بیان ژن های PKS4، PKS13، FG12056 و FG02398 با تکنیکReal-time PCR ارزیابی شد. عصاره شیرین بیان کمترین مقدار MIC (25 میلی گرم بر میلی لیتر) و عصاره های دارچین و ریحان بیشترین مقدار MIC (50 میلی گرم بر میلی لیتر) را داشتند. عصاره شیرین بیان با کمترین مقدار حداقل غلظت قارچکشی (MFC) (50 میلی گرم بر میلی لیتر) ، بالاترین خاصیت قارچکشی روی F. graminearum را داشت و عصاره های دارچین و ریحان بیشترین مقدار MFC (100 میلی گرم بر میلی لیتر) را داشتند. میزان بیان ژن های PKS4، PKS13، FG12056 و FG02398 به طور معنی داری توسط عصاره شیرین بیان کاهش یافت. نتایج این تحقیق نشان داد که عصاره شیرین بیان دارای خاصیت قارچکشی و مهارکنندگی بر رشد قارچ F. graminearum است و میزان بیان ژن های اصلی در مسیر زیست ساخت زیرالنون را کاهش می دهد.

    در پژوهش حاضر، جداسازی جدایه های مختلف قارچ Beauveria bassiana از خاک مناطق مختلف و بررسی زهرآگینی آن ها علیه سن گندم (Hem. : Scutelleridae) Eurygaster integriceps، سوسک Zophobas morio (Col. : Tenebrionidae) و شب پره آرد (Lep. : Pyralidae) Ephestia kuehniella انجام شد. از نمونه های خاک مناطق کرج، ارومیه، ماکو و دزفول، 23 جدایه قارچ B. bassiana توسط تله گالریا، جداسازی و شناسایی شد. حشرات به روش غوطه وری با این جدایه ها آلوده شدند، بر اساس نتایج پوره های سن 5 سن گندم و لاروهای سن 4 شب پره آرد به تمام جدایه ها حساس بودند درحالی که لارو سن آخر سوسک مقاوم بود و هیچ مرگی ثبت نشد. بیش ترین تلفات به وسیله ی جدایه های TV و TZ با حدود 90 درصد مرگ روی هر دو میزبان E. kuehniella و E. integriceps مشاهده شد که در بالاترین گروه زهرآگینی قرار گرفتند. بر اساس زهرآگینی، جدایه ها به سه دسته تقسیم بندی شدند: جدایه های با زهر آگینی کم (ایجاد 10 تا 33 درصد مرگ) ، جدایه های با زهر آگینی متوسط (ایجاد 34 تا 67 درصد مرگ) و جدایه های با زهر آگینی زیاد (ایجاد 68 تا 100 درصد مرگ). روی محیط کشت PDA دو نوع تندش یک طرفه و دوطرفه برای کنیدی های جدایه ها تشخیص داده شد. درصد این دو نوع تندش بین جدایه ها تفاوت معنی داری داشت، بین نوع تندش و زهرآگینی همبستگی معنی داری دیده نشد.

    در این مطالعه به منظور به دست آوردن ریزوباکترهایی با قدرت تشکیل بیوفیلم بالا و بررسی نقش آن ها در بیوکنترل Pectobacterium carotovorum subsp. carotovorum، ابتدا از مناطق مختلف استان های آذربایجان شرقی و غربی 100 نمونه از ناحیه ریزوسفر گیاهان مختلف زراعی جمع آوری و 214 جدایه باکتری با روش سری رقت جداسازی و خالص-سازی شد. جدایه های به دست آمده براساس میزان بازدارندگی علیه پکتوباکتریوم و توانایی تشکیل بیوفیلم در شرایط آزمایشگاهی غربالگری شدند. از 214 جدایه مورد بررسی، 13 جدایه علیه پکتوباکتریوم فعالیت آنتاگونیستی بالایی در شرایط آزمایشگاهی نشان دادند و 12 جدایه توانایی تشکیل بیوفیلم بالایی از خود نشان دادند. در آزمون برش های سیب زمینی جدایه هایی با قدرت تشکیل بیوفیلم بالا مانند G177 و G19-1 به صورت کامل از استقرار بیمارگر جلوگیری کرده و مانع ایجاد پوسیدگی نرم شدند. با این وجود در شرایط گلخانه ای تنها جدایه G177 بیماری را کنترل نمود و علاوه بر آن موجب افزایش رشد طولی ساقه نیز شد. به طور کلی نتایج به دست آمده نشان دهنده آن بود که تشکیل بیوفیلم به تنهایی به عنوان مکانیسم، مسئول بیوکنترل پکتوباکتریوم نبوده است و این فاکتور تحت تاثیر عوامل مختلفی قرار می گیرد.

    حشره ‏کش‏های زیستی مبتنی بر باکتری Bacillus thuringiensis (Bt) مهم ترین و پرکاربردترین عوامل کنترل زیستی حشرات آفت در جهان هستند. یکی از مهترین عواملی که باعث غیرفعال شدن باکتری Bt در طبیعت می شود، اشعه فرابنفش است. حفظ مقاومت باکتری در برابر شرایط محیطی باعث افزایش گسترده از این باکتری به عنوان عامل کنترل آفات می شود. هدف از اجرای تحقیق حاضر، بررسی اثر حفاظتی نانوصفحات گرافن اکسید بر زنده مانی اسپورهای باکتری جدایه KD-2 در مقابل اشعه فرابنفش بود. در این راستا، زیست سنجی فرمولاسیون‏های نانو و غیرنانو روی لارو سن دو Ephestia kuehniella انجام شد. زنده مانی اسپورها در فرمولاسیون نانو و فرمولاسیون اسپور آزاد بعد از 120 ساعت قرار گرفتن در معرض اشعه فرابنفش A به ترتیب 36/0±43/28 % و 69/0± 29/19% به دست آمد. درصد مرگ لارو سن دو E. kuehniella تیمار شده با فرمولاسیون نانو و فرمولاسیون غیر نانو اشعه دیده پس از 10 روز 88/2±35% و 05/3±22% حاصل شد. لذا نتایج تحقیق حاضر نشان داد که نانوصفحات گرافن اکسید می‏تواند به عنوان محافظ اشعه فرابنفش A در فرمولاسیون های باکتری Bt استفاده شوند.

    بیماری بلایت باکتریایی برنج با عامل Xanthomonas oryzae pv. oryzae، یکی از مهمترین بیماری های برنج در سراسر دنیا می باشد. وقوع این بیماری در ایران نیز گزارش گردیده است. در تحقیق حاضر اثرات ضد باکتریایی تعدادی از گیاهان دارویی موجود در طبیعت استان کهگیلویه و بویراحمد بر این باکتری و همچنین باکتری غیربیماری‏زای Pseudomonas fluorescens CHA0 مورد بررسی قرار گرفت. بررسی اثر ضدباکتریایی اسانس‏های آویشن دنایی، رزماری، رازیانه، پونه، نعناع و شمعدانی ‏معطر در شرایط آزمایشگاه به روش نشر در آگار انجام گرفت. همچنین حداقل غلظت مهار کنندگی و حداقل غلظت کشندگی اسانس‏ها تعیین گردید. یافته‏های این پژوهش نشان داد که بیشترین اثر ضدباکتریایی در غلظت mg/ml4 و mg/ml100، مربوط به گیاه آویشن دنایی می باشد، همچنین در غلظت mg/ml4، کمترین اثر متعلق به گیاه پونه می باشد. در غلظتmg/ml 1، بیشترین اثر ضد باکتریایی مربوط به گیاهان نعناع، پونه و رازیانه بود. باکتری Xanthomonas oryzae pv. oryzae بیشترین حساسیت را به اسانس آویشن دنایی با حداقل غلظت مهار کنندگی mg/ml122/0 و حداقل غلظت کشندگی mg/ml245/0 و باکتری Pseudomonas fluorescens CHA0 بیشترین حساسیت را به اسانس رازیانه با حداقل غلظت مهار کنندگی mg/ml95/1 و حداقل غلظت کشندگی mg/ml90/3 نشان دادند.

     کنترل بیولوژیک بیماری های گیاهی شاخص مهم تولید محصولات کشاورزی ارگانیک است. جهت یافتن جدایه های بومی مناسب تریکودرما برای کنترل بیولوژیک پوسیدگی فوزاریومی خیار، از خاک گلخانه در مناطق عمده کاشت استان یزد نمونه برداری انجام شد. پس از جداسازی و خالص سازی و براساس ویژگی های مورفولوژی و مولکولی جدایه های بومی تریکودرما از هم تفکیک شدند. 20 ایزوله تریکودرما, متعلق به گونه های T. harzianum و T. atroviride شناسایی شدند. نتایج تجزیه واریانس داده های آزمون های کشت متقابل و تولید متابولیت های فرار نشان داد که ایزوله های بومی در سطح احتمال یک درصد از رشد میسلیو م قارچ F. oxysporum ممانعت کردند. نتایج نشان داد که سرعت رشد این جدایه ها بسیار بیشتر از عامل بیماری پوسیدگی فوزاریوم بوده، به طوریکه پس از چند روز پرگنه های آن را پوشاندند و ضمن اشغال محل رشد و مصرف غذای مورد نیاز، روی میسلیوم پاتوژن نیز رشد کردند و با تولید ترکیبات فرار، مانع تشکیل میسلیوم قارچ پاتوژن شدند. نتایج نشان داد که بیشترین درصد بازدارندگی از رشد میسلیو م مربوط به گونه T. atroviride (Ta8) بود.

    بره موم یکی از فرآورده های جانبی با ارزش زنبورعسل می باشد که زنبورهای کارگر با جمع آوری صمغ گیاهان و مخلوط کردن آن با آنزیم های خود، ماده ای چسبناک، موم مانند و معطر را تولید می کنند. برخی خواص بیولوژیکی از قبیل خواص ضدباکتریایی، ضدقارچی و آنتی اکسیدانی برای این ماده به اثبات رسیده است. در این پژوهش، فعالیت مهار کنندگی عصاره اتانولی بره موم روی چهار قارچ بیماریزای پس از برداشت گیاهی شامل Aspergillus flavus (عامل کپک زرد پسته) ، Botrytis cinerea، A. tubingensis وCladosporium cladosporioides (قارچ های عامل پوسیدگی میوه انگور) بررسی شد. برای این منظور، ابتدا عصاره اتانولی بره موم در غلظت های 50، 100، 250، 500 و 1000 میکروگرم بر میلی لیتر به محیط کشت PDA افزوده شد. سپس دیسک میسلیومی جوان قارچی در مرکز محیط کشت حاوی عصاره، مایه زنی و میزان رشد آنها اندازه گیری و میزان درصد بازدارندگی عصاره تعیین شد. نتایج نشان داد که بین گونه های قارچی، غلظت های عصاره و همچنین برهمکنش بین گونه های قارچی و غلظت های مختلف آن تفاوت معنی داری (P≤0. 01) وجود دارد. گونه های قارچی، میزان حساسیت متفاوتی در مواجهه با غلظت مشخص از عصاره مزبور نشان دادند، ولی به طور کلی بیشترین درصد بازدارندگی در بیشترین غلظت مورد استفاده مشاهده شد. قارچ های A. tubengensis و C. cladosporioides به ترتیب با %88/80 و %11/79 بیشترین درصد بازدارندگی از رشد را در غلظت μg/L 1000 عصاره مزبور نشان دادند. با توجه به نتایج این تحقیق می توان گفت که بره موم به عنوان فرآورده ای طبیعی، پتانسیل ضدقارچی دارد و می توان پس از تحقیقات تکمیلی از آن برای مهار زیستی بیماری های پس از برداشت قارچی بهره برد.

    The gray mold disease caused by Botrytis cinerea is one of the important post-harvest diseases especially in apple and Strawberry. In order to biological control of B. cinerea, we used 14 isolates of ascomycetous yeasts obtained from natural resources and plants. For molecular identification of yeast isolates, genomic DNA extracted and D1/D2 domain of the large subunit (LSU) rDNA gene sequenced. Nine species including Pichia kudriavzevii, Pichia galeiformis, Galactomyces candidum, Meyerozma guilliermondii, Saccharomyces cerevisiae, Zygoascus meyerae, Pichia sp., Candida parapsilosis, Metschnikowia sp., Candida boidinii, Lecythophora sp. and Candida catenulatea were identified based on sequence similarity of D1/D2 domain of the LSU rDNA gene of the isolates with those in NCBI database. The dual culture method used for assay of their biocontrol activity against of B.cinerea on potato dextrose agar medium in a completely randomize design with tree replications. Our results show that highest inhibitor effect related to G. candidum with 47.85 percent and lowest effect related to P. kudriavzevii with 6.53 percent. In this study, yeast isolates obtained from natural resource and plants showed high biocontrol activity against of B. cinerea.

    In the present study, optimization and modeling of the mycelial growth inhibitory and fumigant toxicity of Thymus vulgaris essential oil were investigated using Response Surface Methodology (RSM) against the pathogenic fungi Fusarium graminearum and Botrytis cinerea and the lesser grain beetle (Rhyzopertha dominica), respectively. The chemical composition of this essential oil was also assessed through Gas Chromatography-Mass Spectrometry (GC-MS) and carvacrol (27.99%), thymol (12.76%), and geraniol (8.91%) were identified as its main components. Mycelial growth inhibition of pathogenic fungi and insect pest mortality was significantly enhanced with increasing of the essential oil concentrations and the exposure times. Optimum conditions to achieve maximum mycelial growth inhibitory (88.67%) were evaluated as time of 88.59 h and the concentration of 486.66 ppm. Also, a time of 74.01-h and the concentration of 552.96 ppm were caused the maximum growth inhibitory of B. cinerea (94.43%). The concentration of 43.10 µl/l and 60 h exposure time were estimated as optimum conditions for achieving 72.23% mortality of R. dominica. Results demonstrated that the essential oil of T. vulgaris has a high potential for management of F. graminearum and B. cinerea and the lesser grain beetle

    Currently, the management of the struggle against tea root lesion nematode (Pratylenchus loosi) based on biological control is one of the research priorities in tea product. In this research, after sampling the soil and root of tea bushes in tea cultivated areas included Guilan and Mazandaran provinces during the spring and summer of 2013, fungi were isolated from tea rhizosphere soil area. The fungi tested included Paecilomyces lilacinus (T1), P. lilacinus (N1, N3), Fusarium culmurom, Clonostachys rosea and Acremonium strictum. After identifying, purifying of strains and performing experiments related to the fungi antagonistic under laboratory conditions (Czapek-Dox 3.34% and malt 1.5% as culture filtrates). Results showed that in Czapek-Dox culture, P. lilacinus (T1) fungi deactivated 52.01% of P. loosi juveniles after 72 hours and this strain didn’t show significant effect on adult mortality. In malt culture, P. lilacinus (T1) fungi deactivated juveniles and adult 32.41% and 30.54%, respectively. In greenhouse condition, antagonistic experiments were carried out in three modes of fungi and nematode inoculation simultaneously, fungi inoculation and two weeks later nematode inoculation and nematode inoculation and two weeks later fungi inoculation. The results showed that if C. rosea inoculated two weeks before nematode inoculation, it could be considerable effect on some indices such as total seedling weight, root fresh weight, root volume and nematode populations in root and soil in comparison with other strains

    Actinobacteria have attracted high interests as potential biocontrol agents due to their antagonistic properties against wide range of plant pathogens particularly fungi. Sclerotinia sclerotiorum (Lib) De Bary, the causal agent of cucumber white stem rot, is one of the most destructive phytopathogens throughout the world which has three important characters as wide host range, severe pathogenicity and prolonged survival of the Sclerotia in unfavorable environmental conditions. Considering the serious damage of this pathogen that cannot be easily controlled by chemical methods, alternative approaches such as biocontrol method using several species of actinobacteria were evaluated against this pathogen. Antifungal bioactivity of 109 isolates of actinobacteria collected from soils of Alborz province of Iran was investigated against S. sclerotiorum through agar dual culture bioassays. Among them, Streptomyces UTS13, UTS19 and UTS49 revealed reasonable inhibitory capabilities in dual culture procedure. In physiological studies the isolates showed enzymatic activities of chitinase, and protease. The results showed that UTS13 reduced the disease upto 50% in greenhouse conditions and significantly increased plant growth compared with the control. UTS13 isolate was identified with molecular features as Streptomyces albidoflavus UTS13. Future perspective includes production of commercial biocontrol products and resistant transgenic plants having antifungal properties originated from biologically active genes of S. albidoflavus UTS13. This investigation introduced S. albidoflavus UTS13 as an effective biological agent with wide spectrum of antagonistic activities against Sclerotinia sclerotiorum.

    One of the important criteria for assessing the efficiency of natural enemies is to study their behavioral characteristics. In the current study host stage preference, switching, and mutual interference of the predatory gall midge larvae Aphidoletes aphidimyza Rondani were studied on different nymphal instars of melon aphid, Aphis gossypii Glover. The experiments were carried out in Petri dishes with cucumber leaf discs in a growth chamber. Based on the results, no significant differences were found for the preference of A. aphidimyza larvae for each nymphal instars of the melon aphid. But, its highest eaten biomass was recorded on forth instars. In switching trial, at 20: 40 and 30: 30 (third instar: first instar) ratios, the predator had more preference for third instars of the melon aphid. In mutual interference, the per capita searching efficiency of the predator varied from 0.0623 to 0.1805 h-1, which the negative value of the regression slope indicated the mutual interference of A. aphidimyza larvae. The results showed that the behavioral characteristics of A. aphidimyza may be affected by the density of the pest as well as the predator larvae

    Fusarium graminearum causes Fusarium head blight, an important disease of cereals that leads to extensive yield and quality loss of wheat. Yield losses results from reduction in the number of kernels and shriveled kernels and grain quality is reduced due to accumulation of mycotoxins. In this study, the effects of ethanolic extracts of Glycyrrhiza glabra, Cinnamomum zeylanicum and Ocimum basilicum on F. graminearum growth and the expression of some essential genes in zearalenone biosynthetic pathway were investigated. Minimal inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) were measured and the expression of PKS4, PKS13, FG12056, and FG02398 genes were evaluated by using quantitative real-time PCR (qRT-PCR) technique. Glycyrrhiza glabra extract had the the lowest MIC value (25 mg/ml) and the extracts of C. zeylanicum and O. basilicum had the highest MIC value (50 mg/ml). Glycyrrhiza glabra extract had the lowest MFC value (50 mg/ml) and the highest fungal property on F. graminearum, and the extracts of C. zeylanicum and O. basilicum had the highest MFC value (100 mg/ml). The expression of PKS4, PKS13, FG12056, and FG02398 genes were significantly decreased by G. glabra extract. The results of this study showed that the G. glabra extract has fungicidal and inhibitory effects on F. graminearum and reduces the expression of essential genes in zearalenone biosynthetic pathway.

    In the present study, some isolates of Beauveria bassiana were isolated from different soil samples and their virulence was investigated against Sunn pest Eurygaster integriceps (Hem., Scutelleridae), flour moth, Ephestia kuehniella (Hym., Pyralidae), and king meal worm, Zophobas morio (Col., Tenebrionidae). Twenty three isolates were isolated using Galleria-baiting method from collected soil samples in different regions; Karaj, Urmia, Mako and Dezful and were identified. The insect hosts were infected using dipping method. The results showed that the fifth nymphal instars of Sunn pest and fourth larval instars of flour moth were susceptible to all isolates of B. bassiana while last larval instars of the beetle were resistant and no mortality was observed. The most virulent isolates were TV and TZ with approximately 90% mortality on E .kuehniella and E. integriceps. The isolates were categorized broadly into three classes: low virulent (0-33% mortality), medium virulent (34-67% mortality) and high virulent (68-100% mortality). On PDA medium, two distinct germination types of conidia were revealed including conidia with single and dual germ tubes. There were significant differences among isolates in their percentages of germination types but there was no significant correlation between virulence and conidial germination types.

    In this research for obtaining rhizobacteria with high ability of biofilm formation and investigation of their role in biocontrol of Pectobacterium carotovorum subsp. carotovorum, first 100 samples from East and West Azarbaijan provinces were collected and 214 bacterial isolates were isolated and purified by serial dilution method. Obtained isolates were screened based on their inhibition against Pectobacterium and the ability of biofilm formation under in vitro. Out of 214 isolates, 13 isolates showed high antagonistic activity against Pectobacterium under in vitro condition and 12 isolates showed high ability of biofilm formation. In potato slices bioassay, isolates with high ability of biofilm formation such as G19-1 and G177 inhibited the establishment of pathogen and they prevent soft rotting. However, in greenhouse studies only isolate G177 controlled the disease and in addition, it increased the stem growth as well. In general, obtained results from this study showed that biofilm formation as a mechanism solely is not responsible for biocontrol of Pectobacterium and this factor is effected by different agents.

    Biological insecticide Bacillus thuringiensis (Bt) is known as the most important and most commonly biological control agents in the world. One of the most important factors that disable Bt in nature is ultraviolet radiation. Maintaining bacterial resistance against environmental conditions causes widespread increases of this bacterium as a pest control agent. The objective of the present study was to evaluate the protective effectiveness of graphene oxide (GO) on spore viability of Bt subsp. KD-2 against Ultra Violet-A (UV-A) radiation. In this regards, bioassay of nano and non-nano formulations were carried out against second-instar larvae of Ephestia kuehniella. After 120 h exposure to UV-A, spore viability of the nano-formulation and free spore formulation were 28.43± 0.36 % and 19.29 ± 0.69 %, respectively. The mortality of irradiated free spore formulations and non-nano formulation on the second-instar larvae of E.kuehniella were 22 ±3.05% and 35±2.88% on the 10th day of the experiment. Therefore, the results of this study showed that nanosheet of GO can be used as protective UV-A for Bt formulation.

    Bacterial blight disease of rice, caused by Xanthomonas oryzae pv. oryzae, is one of the most important diseases of rice worldwide. Incidence of this disease has been reported in Iran. In present study, antibacterial effects of some medicinal plants of Kohgiluyeh and Boyer-Ahmad province on this bacterium as well as on Pseudomonas fluorescens CHA0 was investigated. Antimicrobial effect of essential oils of Thymus daenensis, Rosmarinus officinalis, Foeniculum vulgar, Mentha spicata, Mentha piperita and Pelargonum graveolens was determined by agar diffusion method, under laboratory condition. Minimal inhibitory concentration (MIC) and Minimal bactericide concentration (MBC) of essential oils were also determined. The results showed that, at the concentrations of 4 and 100 mg/ml, the essential oil of T. daenensis had the highest antibacterial effect and at the concentration of 4 mg/ml, Mentha spicata had the lowest antibacterial effect. At the concentration of 1 mg/ml, essential oils of Mentha piperita, Mentha spicata and Foeniculum vulgar showed the most antibacterial activity. It was found that Xanthomonas oryzae pv. oryzae was the most sensitive to Thymus daenensis with MIC of 0.122 mg/ml and MBC of 0.245 mg/ml, while Pseudomonas fluorescens CHA0 was the most sensitive to Foeniculum vulgar with MIC of 1.95 mg/ml and MBC of 3.90 mg/ml.

    Biocontrol of plant diseases is a significant indicator for the production of organic agricultural products. In order to find suitable native Trichoderma spp., for the purpose of biological control of Fusarium wilt of cucumber, samples were taken from the greenhouse soil of major areas of greenhouse cucumber cultivation in Yazd province. After the processes of separation and purification, and based on morphological and molecular characteristics, native Trichoderma spp. isolates were identified. Twenty Trichoderma isolates were identified which belonged to T. harzianim and T. atroviride species. The results of analysis of variance of dual-culture experiments and the test data relating to the production of volatile metabolite substances showed that native isolates inhibited the F. oxysporum mycelium growth (α=1%). The results indicated that the rapidity of growth in native isolates was notably higher than that of Fusarium wilt agent, so that after some days they covered its colonies. Besides, while having occupied the place of growth and using the needed nourishment, they grew on pathogen mycelium, and through producing volatile substances they inhibited the formation of pathogen mycelium. The results showed that T. atroviride (Ta8) isolate had the maximum growth inhibition effect.

    Propolis is a valuable bee-product collected by worker bees from plants and mixing with their enzymes, a sticky, waxy and aromatic material is produced. Various biological properties e.g. antibacterial, antifungal and antioxidant activity had been proven for this substance. In this study, the inhibitory activity of propolis ethanolic extract in vitro against four pathogenic fungi including Aspergillus flavus (causal agent of pistachio yellow rot), Botrytis cinerea, A. tubingensis and Cladosporium cladosporioides (causal agents of grape rot) was studied. First the ethanolic extract of propolis was provided and then was added to PDA medium in 50, 100, 250, 500 and 1000 µg/ml. Mycelial disks of fresh fungi cultures were incubated in the center of treated medium and the growth rate of fungi was measured and the inhibition percent was determined. Results showed that there is significantly different (P≤0.01) among fungal species, concentrations and interaction between fungal species and concentration. The sensitivity of fungal species was different in contract to specified concentration of the extract, generally the highest inhibition was observed at the highest used concentration. A. tubengensis and C. cladosporioides showed the highest growth inhibition with 80.88% and 79.11% respectively. The results of this study showed propolis possesses high antifungal potential and it can use for biological control of plants fungal diseases after further researches.
