صفحات 1-15سلول های بنیادی اسپرماتوگونی اساس و پایه سیستم تولید مثل مذکر را تشکیل می دهند و تنها سلول های بنیادی اند که قادر به انتقال صفات ژنتیکی از نسلی به نسل بعد هستند. یکی از راه های حفظ باروری در افراد مذکر که نیاز به درمان های شیمی درمانی دارند، جداسازی سلول های بنیادی اسپرماتوگونی و حفظ طولانی مدت آن است تا برای از سرگیری اسپرم زایی استفاده شوند. پیشبرد اسپرم زایی در شرایط آزمایشگاهی یکی از راه های دست یابی به این هدف است. اسپرم زایی آزمایشگاهی برای کوتاه کردن یک روند پیچیده به قسمت های کوچک تر به منظور آزمایشها، دستکاری ها و فهم آن در سطح سلولی و مولکولی ضروری است و اجازه دستکاری محیط پاراکرین و همچنین بررسی تاثیر نقش هر یک عوامل رشد به صورت جداگانه در روند اسپرم زایی را می دهد. سیستم های مختلف کشت در شرایط آزمایشگاهی به منظور معرفی جایگزین مناسب برای پیشبرد اسپرم زایی و در نهایت رسیدن به اسپرماتوزوای بالغ و عملکردی برای استفاده در روشهای درمان ناباروری است. در این مقاله سعی شده است روش های مختلف حفظ طولانی مدت سلول های بنیادی اسپرماتوگونی و سیستم های کشت در آزمایشگاه به منظور پیشبرد فرآیند اسپرم زایی را بررسی شود و یک جمع بندی مفید و کاربردی برای استفاده در بالین ارایه شود.کلیدواژگان: اسپرم زایی، سلول های بنیادی اسپرماتوگونی، حفظ طولانی مدت

صفحات 17-31هدفمیکرووزیکول های مشتق از سلول به عنوان مکانیسمی نوین در زمینه ارتباطات سلول-سلول شناخته می شوند. اگزوزوم های مشتق از سلول های بنیادی مزانشیمی به عنوان مکانیسمی نوین در زمینه عملکرد پاراکراین سلول های بنیادی مزانشیمی قلمداد می شوند. در این راستا، اگزوزوم ها نقشی مهم در ارتباطات بین سلولی مابین سلول های بنیادی مزانشیمی و سلول های توموری بر عهده دارند.مواد و روش هااگزوزوم ها از محیط کشت سلول های بنیادی مزانشیمی توسط سانتریفوژ افتراقی تخلیص شد. اندازه و شکل ظاهری اگزوزوم های مشتق از سلول های بنیادی مزانشیمی توسط میکروسکوپ الکترونی نگاره بررسی شد. همچنین، اندازه اگزوزوم ها توسط تفرق دینامیکی نور سنجیده شد. آنالیز وسترن بلات برای نشانگر اگزوزومی CD9 انجام شد. اگزوزوم های تخلیص شده با رنگ فلورسنت PKH نشاندار شد. سلول های توموری تخمدان SKOV3 با اگزوزوم های نشاندار شده تیمار و جذب سلولی توسط میکروسکوپ فلورسنت بررسی شد.نتایجمیکروسکوپ الکترونی نگاره نشان داد که اگزوزوم های مشتق از سلول های بنیادی مزانشیمی دارای شکلی کروی با قطر مابین 30 الی 100 نانومتر است. همچنین اندازه اگزوزوم ها توسط سنجش فرق دینامیکی نور توزیع اندازه زنگوله ای شکل با بیشینه فراوانی 80 نانومتر را نشان داد. آنالیز وسترن بلات نیز بیان CD9 (نشانگر ویژه اگزوزومی) در اگزوزوم های تخلیص شده را نشان داد. همچنین بررسی میکروسکوپ فلورسنت نشان داد که اگزووزم های نشاندار شده با رنگ PKH26 می تواند توسط سلول های توموری SKOV3 با کارآمدی بالا جذب شود.نتیجه گیریروش ارایه شده در زمینه جداسازی اگزوزوم ها از سلول های بنیادی مزانشیمی و تایید ماهیت و جذب آن ها توسط سلول های توموری، گامی مهم و پیش نیاز در زمینه درک عملکرد اگزوزوم ها در سلول های توموری برای مطالعات آینده است. بدیهی است توانایی در مطالعه زیستی جذب اگزوزوم ها توسط سلول های سرطانی فرصت های جدیدی در زمینه بررسی های عملکردی این نانووزیکول های طبیعی و محتویات آن ها در زمینه درمان سرطان فراهم می آورد.کلیدواژگان: سلول های بنیادی مزانشیمی، اگزوزوم، سانتریفوژ افتراقی، جذب سلولی، سلول های توموری

صفحات 33-44هدفنگرانی از وجود آفلاتوکسین در مواد غذایی و خطراتی که این سم برای سلامت انسان و حیوانات دارد، باعث پیدایش راه های مختلف حذف یا کاهش این سم شده است؛ از جمله این روش ها، کنترل زیستی قارچ توسط ریززنده های دیگر است. در این تحقیق از باکتری باسیلوس آمیلولیکوفاسینس جدا شده از خاک باغ پسته از باغات شهرستان دامغان به عنوان عامل کنترل زیستی برای مهار رشد و تولید آفلاتوکسین قارچ آسپرژیلوس پارازیتیکوس استاندارد NRRL2999 استفاده شد.مواد و روش هاپس از 72 ساعت کشت باکتری در دمای 30 درجه سانتی گراد، مایع رویی آن به عنوان منبع ترکیبات ضد قارچی جداسازی شد. غلظت های مختلف از مایع رویی در مجاورت سوسپانسیون قارچی در محیط کشت GYB به مدت چهار روز در دمای 28 درجه سانتی گراد قرار گرفت. پس از این مدت میزان مهار رشد قارچ به روش محاسبه وزن خشک توده قارچی محاسبه شد. میزان آفلاتوکسین B1 نمونه ها نیز به روش کروماتوگرافی لایه نازک و HPLC سنجش کیفی و کمی شد.نتایجبر اساس نتایج، با افزایش میزان مایع رویی کشت باکتری به عنوان ماده آنتاگونیست در محیط رشد قارچ، کاهش بیشتری برای رشد قارچ مشاهده شد. آفلاتوکسین B1تولیدی در نمونه های کنترل به حدودppm 35/2 رسید که این میزان در نمونه های تیمار شده با مایع رویی باکتری به شدت کاهش پیدا کرده بود. میزان این کاهش نیز وابسته به غلظت مایع رویی بود.نتیجه گیریبا توجه به دست آوردهای این تحقیق و با توجه به بومی بودن سویه باکتریایی مورد نظر، می توان از این سویه به عنوان عامل کنترل زیستی قارچ های مولد آفلاتوکسین استفاده کرد.کلیدواژگان: کنترل زیستی، آفلاتوکسین B1، آسپرژیلوس پارازیتیکوس، باسیلوس آمیلولیکوفاسینس

صفحات 45-58هدفبیماریآلزایمر شایع ترین شکل زوال عقل در پیری است. مهمترین عامل آسیبشناختی این بیماری رسوبات خارج سلولی پپتید بتا آمیلویید است که از طریق مکانیسمهای مختلف باعث مرگ سلولهای عصبی میشود. درمانهای جدید روی ترکیباتی تمرکز میکنند که از طریق چند مکانیسم بتوانند در بهبود این بیماری موثر باشند. گیاهان دارویی، به خاطر دارا بودن ترکیبات متعدد، با آثار فارماکولوژیک گوناگون، از طریق مکانیسمهای مختلف، میتوانند در درمان این بیماری، موثر باشند. هدف از این مطالعه، بررسی اثر حفاظتی عصاره متانولی هفت گیاه از گیاهان دارویی ایرانبر سمیت پپتید بتا آمیلویید در مدل سلولی PC12 است.
مواد و روشها: عصارهگیری از گیاهان با حلال متانول به روش ماسیراسیون در دمای اتاق انجام گرفت. سلولهای PC12 طبق برنامه استاندارد کشت داده شدند. سپس سلولها با پپتید بتا آمیلویید به تنهایی و به همراه غلظتهای مختلف عصاره ها بهمدت 24 ساعت انکوبه شدند و میزان بقای سلولی با استفاده از روش MTT اندازهگیری شد.نتایجاز بین عصاره های مورد مطالعه، عصاره گیاهان Sanguisorba minor، Cerasus microcarpa،Ferulago angulata،Rosa canina توانستند مرگ سلولی ناشی از پپتید بتا آمیلویید را در کشت سلولهای PC12 کاهش دهند. اثر حفاظتی مشاهده شده برای این عصاره ها وابسته به دوز بود. عصاره گیاهان Stachys pilifera، Amygdalus scoparia و Alhagi pseudalhagi آثار حفاظتی نشان ندادند.
نتیجه گیری: با توجه به این نتایج عصاره گیاهان Sanguisorba minor، Cerasus microcarpa، Ferulago angulata، Rosa canina میتوانند در درمان بیماری آلزایمر مورد توجه قرار گیرند.کلیدواژگان: بتاآمیلویید، آلزایمر، گیاهان دارویی، سمیت عصبی، کشت سلول -
صفحات 59-72هدفکاندیدا آلبیکنس مخمر فرصت طلبی است که در افراد دارای نقص سیستم ایمنی و در شرایط مناسب، به حالت بیماریزا مبدل میشود. مواد تشکیل دهنده گیاهان دارویی مانند کامفور میتوانند بیان ژنهای دخیل در ویرولانس قارچها را به واسطه خواص ضد قارچی خود، کاهش دهند. محصول ژنهای INT1 و EFG1 نقش مهمی در چسبندگی کاندیداآلبیکنس به بافت میزبان و هیفی شدن آن ایفا میکنند که این خصوصیات میتواند از عوامل بسیار مهم بیماریزاییآن باشند. مطالعه حاضر به بررسی اثر کافور بر تغییرات بیان ژنهای INT1 و EFG1 در سه زمان تیمار 24، 48 و 72 ساعت با استفاده از RealtimePCRپرداخته است.
مواد و روشها: رقتهای سریالی از کامفور در غلظتهای 512، 256، 128، 64، 32، 16، 8، 4، 2 و 1 میلیگرم در هر میلیلیتر تهیه شد و سپس سوسپانسیون سلولی کاندیدا آلبیکنسسویه ATCC 10231با غلظت 103×5/1 سلول در میلیلیتر بهمدت 48 ساعت در 35 درجه سانتیگراد با آن تیمار شد. سپس حداقل غلظت مهارکننده 50 درصد و 90 درصد (MIC50/90) رشد قارچ و حداقل غلظت کشنده قارچ (MFC) این ترکیب به روش رقت میکروبراث تعیین شد. کاندیدا آلبیکنس بهمدت 24، 48 و 72 ساعت با کامفور در حداقل غلظت مهارکننده 50 درصد رشد قارچ (MIC50) تیمار شد. استخراج RNA مخمرها قبل و بعد از مجاورت با این ترکیب صورت گرفت، سپس ترجمه معکوس به cDNA و در نهایت ارزیابی به کمک Real time PCR انجام شد.نتایجحداقل غلظت مهارکننده 50 درصد رشد قارچ (MIC50)، حداقل غلظت مهارکننده 90 درصد رشد قارچ (MIC90) و حداقل غلظت کشنده قارچ (MFC) برای کامفور به ترتیب 16، 32 و 64 میلیگرم در هر میلیلیتر تعیین شد. بررسی تغییرات بیان این ژنها در مخمر نیز نشان داد که کامفور بیان ژن INT1 را در زمان 24 ساعت پس از تیمار به میزان 87 درصد، 48 ساعت پس از تیمار 97 درصد و 72 ساعت پس از تیمار 86 درصد نسبت به نمونه تیمار نشده کاهش داده است. این موضوع در خصوص ژن EFG1 نیز نشان داد که بیان این ژن در تیمارهای 24 ساعتی با کامفور به میزان 58 درصد، در تیمارهای 48 ساعتی 93 درصد و در تیمارهای 72 ساعتی 49 درصد کاهش داشته است.
نتیجه گیری: در سالهای اخیر به دنبال بروز مقاومتهای دارویی و بروز عوارض جانبی داروهای شیمیایی، استفاده از گیاهان دارویی افزایش یافته است. این گیاهان ممکن است بتوانند بهعنوان عوامل ضد قارچی کارآمد عمل نمایند. نتایج این پژوهش نشان داد که استفاده از ترکیبات خالص شیمیایی موجود در ساختار این گیاهان مانند کامفور، میتواند بهطور چشمگیری در کاهش بیان ژنهای ویرولانس قارچی از جمله INT1 و EFG1 تاثیر داشته باشد.کلیدواژگان: کاندیدا آلبیکنس، کامفور، عوامل بیماری زا -
صفحات 73-88هدفژنوم انسان حاوی ژنهای محافظت کننده است که دارای توالی عوامل پاسخگو به آنتی اکسیدانها در پروموترشان است که بیان آنها بهطور خاص تحت تاثیر عامل رونویسی Nrf2قرار میگیرد. این مسیر پیام رسانی (ARE\KEAP1) عمدهترین مکانیسم دفاعی در برابر استرس اکسیداتیو است. رده سلولی حاوی عوامل پاسخگو به آنتی اکسیدانها (ARE)، سلولهای گزارشگر حساسی هستند که برای تشخیص عوامل فعال کننده پاسخگو به آنتی اکسیدانها بهکار میروند و میتوانند در شناسایی مواد اکسیداتیو همچون سرب کمک کننده باشند.
مواد و روشها: در این مطالعه از رده سلولی پایدار Huh7-1x-ARE-luc تولید شده، استفاده شد. مهارفعالیتمتابولیکی سلولهای Huh7 در غلظتهای مختلف سرب (0 تا 80 میکرومولار) توسط آزمون MTT سنجیده شد. در ادامه رده سلولی Huh7-1x-ARE-luc برای بررسی آثار استرس اکسیداتیو با سرب بهعنوان یک ترکیب القا کننده استرس اکسیداتیو و نانوکورکومین بهعنوان یک آنتی اکسیدان در غلظتهای متفاوتی از این ترکیبات تیمار شد و سپس میزان فعالیت لوسیفرازی زیستحسگر بررسی شد. همچنین میزان بیان ژنهای مسیر ARE\KEAP1 توسط Real time PCR تعیین شد.نتایجنتایج نشان میدهد که غلظت 30 میکرومولار سرب باعثمهار متابولیکی50درصدیسلولهای Huh7 میشود. فعالیت لوسیفرازی سلولهای تیمار شده با غلظتهای 4 و 8 میکرومولار از نانوکورکومین و غلظت 30 میکرومولار از سرب نسبت به کنترل ( 30 میکرومولار سرب) کاهش یافته است و با افزایش غلظت نانوکورکومین به 16 میکرومولار این فعالیت افزایش مییابد. نتایج تحلیل داده های حاصل از Real time-PCR نشان میدهد که بیان تمامی ژنهای مورد بررسی شامل Nrf2 و NQO1 در رده سلولی Huh7-1x-ARE-luc تیمار شده با سرب 30 میکرومولار و نانوکورکومین 4 و 8 میکرومولار نسبت به تیمار همان ژنها با سرب 30 میکرومولار کاهش یافته است. از سوی دیگر؛ بیان این ژنها در سلولهای تیمار شده با سرب 30 میکرومولار و نانوکورکومین 16 میکرومولار نسبت به تیمار همان ژنها با سرب 30 میکرومولار افزایش داشته است.
نتیجه گیری: نانوکورکومین بهعنوان یک آنتیاکسیدان توانسته اثر سمیت سرب را احتمالا از طریق مسیر ARE\KEAP1 بهطور معنیداری کاهش دهد؛ در نتیجه میتواند بهعنوان یک آنتیاکسیدان در کاهش استرس اکسیداتیو استفاده شود.کلیدواژگان: عوامل پاسخ گو به آنتی اکسیدان ها، استرس اکسیداتیو، رده سلولی گزارشگر، نانوکورکومین

Pages 1-15Spermatogonial stem cells are foundation of the male reproductive system. These cells are the only conduit capable of transferring genetic traits from one generation to the next. Isolation and long-term preservation of spermatogonial stem cells for use in inducing spermatogenesis is one technique to preserve fertility in male patients who need chemotherapy. In vitro spermatogenesis is an alternative to achieve this goal. The use of an optimal model of human spermatogenesis is a major step in understanding the physiology and genetic pathways in the male reproductive system. In vitro spermatogenesis is crucial to reducing a complex process into smaller parts for experimentation, manipulation, and deriving cellular and molecular level knowledge. Is it possible to manipulate the paracrine environment and separately evaluate the effects of growth factors. Different in vitro culture systems are used to explore alternatives to spermatogenesis and obtain mature, functional spermatozoa for ultimate use in infertility treatment. Pages 17-31ObjectiveCell-derived microvesicles are described as a new mechanism for cell-to-cell communication. Stem cell-derived exosomes have been described as a new mechanism for the paracrine effects of mesenchymal stem cells (MSCs). In this regard, exosomes may play a relevant role in the intercellular communication between MSCs and tumor cells.MethodsExosomes were purified from the conditioned medium of MSCs by differential centrifugation. Exosome size and morphology were examined by scanning electron microscope and sized with dynamic light scattering (DLS). Western blot analysis confirmed the exosomes by using CD9 as a marker. Purified exosomes were labeled with a PKH26 red fluorescent labeling kit. The labeled exosomes were incubated with SKOV3 ovarian tumor cells for 12 h at 37°C, and we used an inverted fluorescence microscope to monitor cellular uptake.ResultsScanning electron microscopy revealed that the purified MSCs-derived exosomes had a spherical shape with a diameter of approximately 30-100 nm. Exosome size measurement by dynamic light scattering analysis also showed a single bell-shaped size distribution with a peak of ~80 nm. Western blot analysis also demonstrated the presence of CD9 (a representative marker of exosomes) in the purified exosomes. These data confirmed that the vesicles isolated from MSCs-conditioned media were the exosomes based on their size and presence of the protein marker CD9. Florescent microscopy showed that PKH26-labeled exosomes could be taken up by SKOV3 tumor cells with high efficiency.ConclusionOur approach for isolation, characterization and cellular uptake of exosomes derived from MSCs is valuable and a prerequisite for future studies that intend to discover exosome function in tumor cells. Pages 33-44ObjectiveThe concern about the presence of aflatoxin and its risk for human and animal health has resulted in the introduction of different methods to eliminate or reduce this toxin. One of these methods is biological control of the fungus by other microorganisms.MethodsWe isolated a strain of Bacillus amyloliquefaciens from the soil of a pistachio orchard, Damghan, Iran. This strain was applied as a biological control to examine the inhibition of growth and toxin production by Aspergillus parasiticus NRRL2999. After 72 hours of incubation of the bacterium at 30°C, we separated the supernatant as a potential source for antifungal compounds. Different doses of the supernatant were co-cultured with the fungus suspension in glucose yeast extract broth (GYB) at 28°C for 4 days. After the incubation period, we measured the inhibition of fungal growth by dry weight assessment of the fungal mass. We performed qualitative and quantitative assessment to determine the amount of aflatoxin B1 by TLC and HPLC, respectively.ResultsIncreased bacterial culture supernatant as the antagonist in fungal growth medium resulted in decreased fungal growth. AFB1 production was 2.35 ppm in control samples, whereas the amount considerably decreased in samples treated by the bacterial supernatants. The toxin reduction was dose-dependent.ConclusionThe results of this study have shown that this bacterial strain, by taking into consideration its native origin, can be used as a biological control against aflatoxigenic fungi.Keywords: biological control, Aflatoxin B1, Aspergillus parasiticus, Bacillus amyloliquefaciens

Pages 45-58ObjectiveAlzheimer's disease (AD) is the most common form of dementia in the elderly. One of the major pathological hallmarks of AD is extracellular deposition of aggregated beta-amyloid (Aβ) peptide in the brain. Aβ causes neural cell death through several mechanisms. New treatments for AD focus on compounds that can modify disease progression and reduce symptoms through several mechanisms. Medicinal plants include several compounds with heterogeneous pharmacological effects that can be effective in the treatment of AD through different mechanisms. The aim of this study is to evaluate the protective effect of a methanolic extract of seven medicinal plants from Iran on Aβ induced toxicity in a cell culture model.MethodsWe cultured PC12 cells according to standard protocols. The cells were incubated with Aβ alone or with different concentrations of extracts for 24 hours. Cell viability was measured using the MTT assay.ResultsSanguisorba minor, Cerasus microcarpa, Ferulago angulate,andRosa caninasignificantly reduced Aβ-induced cell death in the PC12 cell culture. The observed protective effects of extracts were dose-dependent. We did not observe any protective effects with the Stachys pilifera,Amygdalus scoparia, and Alhagi pseudalhagi extracts.ConclusionSanguisorba minor, Cerasus microcarpa,Ferulago angulate, andRosa canina extracts could be considered for treatment of AD.Keywords: Beta amyloid, Alzheimer's disease, Medicinal plants, Neurotoxicity, cell culture

Pages 59-72ObjectiveCandida albicans (C. albicans) is an opportunistic yeast that can lead to pathogenesis in immunocompromised individuals and under suitable conditions. Medicinal plants ingredients such as camphor can reduce the expressions of genes involved in virulence of the fungi through their antifungal properties. The products of INT1 and EFG1 are implicated in inducing filamentous growth and adhesion of C. albicans to the host tissues. Both of these characteristics are very important in its virulence. The present study focuses on the evaluation of the effects of camphor on INT1 and EFG1 expressions at three time points of treatment (24, 48, and 72 hours) via real-time PCR.MethodsWe prepared serial dilutions of camphor (1, 2, 4, 8, 16, 32, 64, 128, 256, and 512 mg/ml) and co-cultured them with 1.5×103 cells/ml of a C. albicans ATCC 10231 suspension for 48 hours at 350C. Next, we determined the MIC50/90 and MFC. C. albicans cells were treated with the MIC50 concentration of camphor for 24, 48, and 72 hours. RNA from C. albicans was extractedbefore and after treatment, back translated into cDNA, and analyzed with real-time PCR.ResultsMIC50, MIC90 and MFC of camphor were determined at 16 mg/ml, 32 mg/ml and 64 mg/ml, respectively. Evaluation of gene expression changes in yeast showed that camphor reduced the INT1 gene expression about 87% at 24 hours, 97% at 48 hours, and 86% at 72 hours after treatment compared to the untreated sample. EFG1 expression reduced about 58% at 24 hours, 93% at 48 hours, and 49% at 72 hours after treatment with camphor.ConclusionIn recent years, advancements have been seen in herbalism due to the increased drug resistance and adverse effects of chemical drugs. These plants may efficiently act as antifungal agents. The results of this study have shown that the use of camphor can significantly reduce the expression of virulence genes INT1 and EFG1 in C. albicans.Keywords: Candida albicans, Camphor, Virulence factors

Pages 73-88ObjectiveThe human genome consists of protective genes, which contain a sequence known as the antioxidant responsive element (ARE) located in their promoter regions. ARE is specific to the transcription factor NF-E2 related factor2 (Nrf2). This signaling pathway is the major defense mechanism against oxidative stress that comprises the chemoprotective response. The cell line that expresses the ARE reporter is sensitive for the detection of ARE activating compounds. It can help to identify toxicity risk and antioxidant activity of chemicals and drugs.MethodsWe used a stable Huh7 ARE-reporter cell line in this study. Metabolic activity of these cells in different concentrations of lead (0 to 80 micromole) was evaluated by the MTT test. We assessed the effects of oxidative stress. We exposed the Huh7 ARE-reporter cell line to different concentrations of lead, an oxidative stress inducer, and nanocurcumin, an antioxidant compound, after which we investivated luciferase activity. Real-time PCR was used to detected AREKEAP1 pathway gene expression.ResultLead, at 30 μM, suppressed 50% of the cells metabolic activity.Cells treated with both lead (30 μM) and nanocurcumin at 4 μM and 8 μM had decreased luciferase activity compared to those treated with only lead. This activity increased when we increased the nanocurcumin concentration to 16 μM. Real-time PCR analysis showed decreased NQO1 and NRF2 expressions in cells treated with both lead (30 μM) and nanocurcumin (4 μM and 8 μM) compared to just lead treated cells. However, NQO1 and NRF2 had increased expressions in cells treated with both lead (30 μM) and nanocurcumin (16 μM) compared to just lead treated cells.ConclusionNano curcumin, as an antioxidant, significantly reduced the toxic effects of lead toxic. This effect probably occurred through the AREKEAP1 pathway.
