    به منظور شناسایی و مطالعه گونه های Cylindrocarpon همراه با ریشه و طوقه کلزا و ذرت در استان خوزستان طی سال زراعی 1386- 1385، از مزارع کلزا و ذرت در مناطق ایذه، دزفول، شوشتر، شوش، اندیمشک و ملاثانی نمونه برداری به عمل آمد. در این بررسی پنج گونه از جنس سیلندروکارپن شناسایی شد که به ترتیب فراوانی شامل C. destructans(96 جدایه)، C. didymium (69 جدایه)، hederae C. (25 جدایه)، C. obtusisporum (14 جدایه) و C. macrodidymum (3 جدایه) بودند. جداسازی گونه های مذکور از ذرت و کلزا برای اولین بار در دنیا انجام می شود. در این تحقیق از جفت آغازگرهای ITS4 & ITS5 برای شناسایی گونه های سیلندروکارپن استفاده شد. محصول تکثیر شده از DNA جدایه های سیلندروکارپن با استفاده از آغازگرهای اختصاصی، قطعه ای به وزن 560-500 جفت باز تکثیر نمود. مقایسه توالی های به دست آمده از گونه های فوق با توالی های موجود در بانک ژن، با استفاده از ابزار جستجوی B LAST(Basic Local Alignment Search Tool)، نشان داد که تمامی توالی های به دست آمده از نمونه های مورد بررسی به گونه های سیلندروکارپن تعلق دارند. گروه بندی گونه های سیلندروکارپن با استفاده از توالی ITS مناطق ریبوزومی مطابق با طبقه بندی بر اساس خصوصیات مرفولوژیکی ارائه شده است. این مطلب نشان می دهد که تعیین توالی ITS مناطق ریبوزومی یک ابزار مفید و کار آمد در ارزیابی رابطه فیلوژنتیکی گونه های جنس سیلندروکارپن است.

    سوختگی برگی که توسط قارچRhynchosporium secalis ایجاد می شود یکی از مهم ترین بیماری های جو در کشور می باشد. تنوع پاتوتیپی 47 جدایه از این بیمارگر از پنج استان کشور با استفاده از هشت رقم افتراقی جو ارزیابی گردید. از میان جدایه های مورد بررسی Rs55 و Rs86از استان لرستان دارای بیشترین قدرت (توان) بیماری زایی و جدایه Rs45 از استان خوزستان دارای کمترین قدرت بیماری زایی بودند. براساس واکنش ارقام افتراقی در برابر این جدایه ها تعداد 20 پاتوتیپ شناسایی شد. این پاتوتیپ ها دارای طیف وسیع بیماری زایی بوده و پاتوتیپ یک در همه مناطق پراکنش داشت و پاتوتیپ های 15، 12 و 13 دارای بالاترین شدت بیماری زایی بودند. در بین ارقام افتراقی مورد بررسیIgri و Armele با ژن های مقاومتBRR4 و BRR1 بالاترین مقاومت و ارقام WI و Digger بیشترین حساسیت را در برابر پاتوتیپ ها نشان دادند. براساس دندروگرام ترسیم شده و با در نظر گرفتن خط برش 76/0 جدایه های مورد بررسی در هفت گروه متمایز قرار گرفتند. تنوع پاتوتیپی R. secalis در این مناطق با موقعیت جغرافیایی آنها مطابقت نداشت و این امر لزوم توسعه منابع مقاومت مختلف در مناطق عمده کشت جو در کشور را تایید می کند.

    نمونه های جنس Septoria روی 45 جنس از 24 تیره گیاهان میزبان بررسی گردیدند. نمونه های مورد بررسی شامل نمونه های تعیین نام نشده موجود در مجموعه مرجع قارچ های وزارت جهاد کشاورزی واقع در بخش تحقیقات رستنی های موسسه تحقیقات گیاه پزشکی کشور و نیز نمونه های جدید جمع آوری شده بودند. بر اساس نتایج این بررسی تعداد نه آرایه شامل Septoria sp. aff. alhagiae، S. botuliformis، S. bromi var. bromi، S. caricina، S. caricicola، S. eremuricola، S. festucae، S. sigesbeckiae و S. meliae به ترتیب از میزبان های Alhagi sp.، Populus sp.، Bromus sp.، Carex depaupertata، C. pendula، Eremurus sp.، Festuca sp، Siegesbeckia orientalis و Melia azedarach برای میکوبیوتای ایران جدید هستند که به طور کامل و مصور در این مقاله توصیف شده اند.

    بوته های برنج دارای علائم پوسیدگی ناحیه طوقه و لکه های بزرگ قهوه ای تیره روی غلاف و ساقه در مزرعه ای در مازندران مشاهده گردید. بر اساس مشخصات مورفولوژیک، قارچ رشد یافته در اطراف قطعات کشت داده شده، Bipolaris sorokiniana شناسایی شد. پس از توالی یابی ناحیه ITS از ژن rDNA نسبت به ارزیابی همولوژی آن با توالی های موجود در بانک ژن اقدام گردید که با هشت استرین sorokiniana B. و Cochliobolus sativus (تلئومورف) با درجه بسیار بالا همولوژی نشان داد. فیلوگرام ترسیم شده نشان داد که در مقایسه با گونه های دیگر Bipolaris، Cochliobolus و جنس های قارچی خویشاوند آنها، استرین B. sorokiniana جدا شده از مازندران به همراه هفت استرین دیگر B. sorokiniana و یک استرین C. sativus در یک گروه قرار گرفتند. آزمون بیماری زایی روی دو رقم برنج و با روش های مختلف انجام شد. این اولین گزارش از بیماری زایی B. sorokiniana روی گیاه برنج در ایران می باشد.

    در این پژوهش زوال چمن از نظر آلودگی به بیمارگرهای قارچی بررسی گردید. نمونه برداری طی سال های 86 تا 88، از مناطق مختلف چمن کاری در سطح فضای سبز شهر شیراز که علائم زردی و زوال داشت، انجام گرفت و عوامل قارچی از برگ و ریشه چمن های آلوده جداسازی و شناسایی شدند، که در بین این عوامل قارچی، عوامل بیماری زای چمن که به میزان بیشتری جداسازی شد، شامل گونه های Fusarium culmorum، F. equiseti، F. solani، F. crookwellense، F. semitectum، F. polyphialidicum وF. sambucinum، Pythium aphanidermatum، P. vexans، P. torulosum،P. oligandrum،P. ultimum var sporangiiferum، P.deliense، P.vanterpooli، Bipolaris australiensis، B. sorokiniana، B. cynodontis، B. spicifera، Curvularia lanuta، Exserohilum rostratum، Rhizoctonia solani، R. zeae و R. spp. دوهسته ای بودند و درصد بیماری زایی گونه های هر جنس با استفاده از آزمون چند دامنه ای دانکن مورد بررسی قرار گرفت.گونه F. semitectum در بین گونه های فوزاریوم، Pythium aphanidermatum در بین گونه های پیتیوم، B. sorokiniana در بین شبه جنس های هلمینتوسپوریومی و در بین گونه های Rhizoctonia، R. solani AG2.2IIIB بیشترین درصد بیماری زایی را دارا بودند.

    در این بررسی نمونه های متعددی از گیاهان دارای لکه برگی از مناطق مختلف استان گیلان بررسی شدند. پس از شناسایی جنس Cercospora و قارچ های شبه سرکوسپورا، گونه های آن پس از بررسی های ریخت شناسی، با بهره گیری از نوشته ها و منابع معتبر موجود شناسایی و تعیین نام شدند. بر اساس نتایج به دست آمده تاکنون سه گونهCercospora texensis، C. flagellaris، Pseudocercospora daturina شناسایی شده اند که به عنوان گونه های جدید برای میکوفلور ایران گزارش می شوند. هم چنین گونه های C. althaeina (از Althaea rosea)،C. apii s.l. (شامل گونه های C. avicennae،C. brachiata، C. brunkii، C. daturicola، C. hydrangeae،C. rumicis، kikuchii C.، C. xanthiicola و C. zinniae) و Passalora circumscissa (از Prunus avium) شناسایی شدند که پیش از این از ایران گزارش شده اند. با وجود این، سرکوسپورا روی جنس های Amaranthus (C. brachiata)، Datura (C. daturicola)، (rumicis C.) Rumex، Xanthium (C. xanthiicola) و Zinnia (C. zinniae) برای اولین بار از ایران گزارش می شود.

    باکتری (Pss) Pseudomonas syringae pv. syringae عامل بیماری های بسیاری روی محصولات زراعی و باغی می باشد یکی از مهم ترین بیماری های ناشی از این باکتری شانکر، لکه برگی و نکروز پوست در درختان میوه هسته دار (هلو، شلیل، زردآلو، آلو و گیلاس) است. به منظور بررسی همسانی یا تنوع جدایه های عامل بیماری، طی سال های 1386-1388 از باغات درختان میوه هسته دار واقع در مناطق مختلف استان های اردبیل، گیلان، مازندران و خراسان رضوی نمونه برداری صورت گرفت. باکتری های جداشده از بافت های آلوده جوانه، برگ، شاخه و تنه بر اساس آزمون های گروه LOPAT و GATTa، Pss شناسایی شدند. تعداد 70 جدایه مورد بررسی های فنوتیپی (مورفولوژیکی، فیزیولوژیک و بیوشیمیایی) و الکتروفورز پروتئین های سلولی (SDS-PAGE) قرار گرفتند. جدایه ها از نظر خصوصیات فنوتیپی و الگوی پروتئین های سلولی اختلاف های جزئی نشان دادند. جهت ارزیابی تنوع ژنتیکی، DNA ژنومی جدایه ها استخراج و در واکنش زنجیره ای پلیمراز (rep-PCR) با استفاده از آغازگرهای ERIC و REP بررسی شد. آنالیز خوشه اینتایج به دست آمده نشان داد که در ERIC-PCR در سطح تشابه 75 درصد، جدایه ها به 8 گروه و با REP-PCR به 5 گروه و با ترکیب همه آغازگرها به 9 گروه تقسیم شدند. نتایج نشانگر وجود تنوع ژنتیکی درون جدایه های Pss عامل بیماری شانکر درختان میوه هسته دار بود.

    گونه Heterodera schachtii، یکی از مهم ترین عوامل بیماری زای چغندرقند در دنیاست. این نماتود در اکثر مناطق تحت کشت این گیاه در ایران گسترش دارد و موجب کاهش عملکرد و میزان قند موجود چغندر تولید شده در مزارع آلوده می شود. جمعیت های مختلف schachtii H. از لحاظ خصوصیات مرفولوژیکی بسیار متفاوت هستند و شناسایی آنها براساس خصوصیات مرفولوژیکی و مرفومتریکی سیست هاولاروهای سن دوم بسیار وقت گیر بوده و نیازمند مهارت و دقت زیاد می باشد. در سال های اخیر امکان استفاده از روش ITS-PCR-RFLP برای تفکیک این گونه از سایر گونه های موجود بررسی شده است. در این بررسی 120 جمعیت از نماتود سیستی چغندرقند از مزارع مختلف در استان خراسان رضوی جمع آوری شد و سیست ها از لحاظ خصوصیات مرفولوژیکی و مرفومتریکی مورد بررسی قرار گرفتند. در میان این جمعیت ها، 88 جمعیت که تفاوت های مرفولوژیکی نسبتا زیادی با هم داشتند، انتخاب شدند. پس ازاستخراج DNA، تکثیر ناحیه rDNA-ITS با استفاده از آغازگرهای عمومی این ناحیه انجام شد. محصول PCR هر نمونه با استفاده از آنزیم برشی MvaI هضم شد. تمامی 88 نمونه الگوی باندی مشابهی را تولید کردند که با الگوی ارائه شده برای این گونه در منابع معتبر همخوانی دارد. پس از تایید گونه، گروه بندی جمعیت های ذکر شده بر اساس خصوصیات مرفومتریکی نشان داد که جمعیت های مناطق شمالی استان ازجمعیت های مناطق جنوبی قابل تفکیک است، هر چند بر اساس مناطق کوچک تر جغرافیایی تفکیک قابل قبولی صورت نگرفت. هم چنین بر اساس طول باندهای کوتیکولی ناحیه واژن و برجستگی های گره مانند موجود در مخروط انتهای بدن سیست ها، چهار تیپ مرفولوژیکی نشخیص داده شد. نتایج این تحقیق نشان داد که روش ITS-PCR-RFLP می تواند در شناسائی جمعیت های مختلف H. schachtii با دامنه وسیع تفاوت های مرفولوژیکی بسیار کارآمدتر و سریع تر از روش های مرفولوژیکی باشد.

    به منظور شناسایی نماتودهای انگل گیاهی، تعداد 45 نمونه خاک از شهرستان کرج، جیرفت و جویبار (به ترتیب از استان های تهران، کرمان و مازندران) جمع آوری شد. نماتودهای موجود در آنها با استفاده از روش الک ها و سانتریفوژ استخراج گردید. نماتودهای استخراج شده پس از تثبیت، به گلیسیرین منتقل و پس از تهیه اسلایدهای دائمی با استفاده از میکروسکوپ نوری مورد شناسایی قرار گرفت. در این تحقیق چهار گونه Neodolichorhynchus sulcatus، Paraphelenchus micoletzkyi، Paratrichodorus minor و Tylenchorhynchus annulatus شناسایی شد. از بین گونه های شناسایی شده سه گونه N. sulcatus، P. micoletzkyi و minor P. برای اولین بار از ایران گزارش می شود. هم چنین گونه T. annulatus به دلیل نبودن شرح کاملی در ایران، در این مقاله شرح داده شده است.

    در این مطالعه تاثیر سه ماده افزوده سوکروز، آلجینات سدیم و صمغ عربی روی پایداری سلول های مخمر Pichia guilliermondii در فرمولاسیون های پودری مورد بررسی و مقایسه قرار گرفت. سلول های مخمر پس از تکثیر در محیط کشت ملاس نیشکر با حامل های پودری تالک، کائولین، سبوس گندم و سبوس برنج و مواد افزوده مختلف شامل سوکروز، صمغ عربی و سدیم آلجینات مخلوط شدند. جمعیت مخمر در 16 فرموله تهیه شده طی دوره شش ماهه مورد بررسی قرار گرفت. در پایان دوره شش ماهه نگهداری در دمای 4 درجه سانتی گراد، بیشترین جمعیت سلول زنده مخمر در هر گرم فرمولاسیون با تعداد1011×4/1 و 1010×5/6 به ترتیب در فرمولاسیون های شماره 11(سبوس گندم همراه با صمغ عربی) و 15 (سبوس گندم همراه با سوکروز) و کمترین جمعیت سلول زنده مخمر با تعداد107×6/7 سلول در فرمولاسیون شماره 2 (کائولین) دیده شد. در فرمولاسیون های نگهداری شده در 24 درجه سانتی گراد نیز بیشترین جمعیت سلول زنده مخمر با تعداد 1010×5/6 در فرمولاسیون شماره 15 مشاهده شد. در دمای 24 درجه نیز کمترین تعداد سلول زنده در فرمولاسیون شماره 2 مشاهده گردید. در بررسی اثر فرمولاسیون در کنترل بیماری کپک آبی سیب در شرایط انبار نتایج قابل قبولی حاصل شد. در بررسی کارایی فرمولاسیون های نگهداری شده در هر دو دمای 4 و24 درجه سانتی گراد، بیشترین کاهش مساحت لکه بیماری روی میوه سیب با کاربرد فرمولاسیون شماره 15 دارای سوکروز و سبوس گندم مشاهده شد. تحقیق حاضر نشان داد که نوع و مقدار مواد به کار رفته در فرمولاسیون باعث حفظ قدرت حیات و کارایی جدایه آنتاگونیست در طی مدت انبارداری شده و موجب حفظ شدت اثر آن در کنترل بیماری در شرایط انبار می شود

    طی سال های 1386 و 1387 از خاک مزارع و باغ های استان مازندران نمونه برداری به عمل آمد. نمونه های خاک از عمق 10 تا 15 سانتی متری تهیه گردید. سپس با روش طعمه گذاری با بذر چغندر قند و خلال دندان ضدعفونی شده، 121 جدایه رایزوکتونیا به دست آمد. شناسایی گونه و گروه آناستوموزی جدایه های جمع آوری شده از طریق بررسی خصوصیات مرفولوژیک و ویژگی های آنها در محیط کشت، آزمون آناستوموز و با استفاده از کلیدهای معتبر انجام گردید. 101 جدایه چند هسته ای و 20 جدایه دو هسته ای شناسایی شدند. از بین جدایه های چند هسته ای، 7 جدایه به گروه آناستوموزی AG1، 38 جدایه به AG2، 5 جدایه به AG4، 3 جدایه به AG5، 13 جدایه به AG6، 11 جدایه به AG9، 3 جدایه به AG11 از R. solaniو 21 جدایه به WAG-Z (R. zeae) واز بین جدایه های دو هسته ای، 15 جدایه به گروه آناستوموزی AG-K، 3 جدایه به R. ramicola و دو جدایه دیگر به دو گروه آناستوموزی نامشخص تحت عنوان BNR-1 و BNR-2 نسبت داده شدند.

    بیماری برق زدگی نخود ناشی از Didymella rabiei، مهم ترین بیماری نخود معمولی در استان کرمانشاه است. عامل بیماری روی بقایای گیاهی و بذرهای نخود بقا می یابد. مرحله جنسی قارچ روی بقایای آلوده نخود که در سطح خاک مزرعه باقی می مانند، تشکیل شده و در بقا و پراکنش بیماری به نقاط دور نقش اساسی دارد. طی سال های زراعی 83-1382 و 84-1383 بقایای آلوده نخود از اوایل مهرماه روی سطح خاک چند مزرعه در شهرستان های مختلف استان کرمانشاه قرار داده شد. در سال زراعی 83-1382 مرحله جنسی قارچ روی اغلب نمونه های قرار گرفته در سطح مزارع تشکیل نگردید و یا در صورت تشکیل به مرحله بلوغ نرسید. در سال زراعی 84-1383 سودوتسیوم های بالغ قارچ به فراوانی روی همه نمونه ها تشکیل شد. سودوتسیوم ها حدود یک ماه تا 45 روز پس از قرار دادن بقایای آلوده نخود در سطح مزرعه (اوایل تا اواسط آبان ماه) روی ساقه و غلاف های آلوده شروع به تشکیل نمودند. متعاقبا سودوپارافیزهای قارچ در بین بافت استرومایی سودوتسیوم رشد و توسعه یافت. سودوتسیوم ها و آسک های بالغ در نیمه اول اسفند ماه در نمونه ها دیده شد. رها سازی آسکوسپورها از اواخر اسفند شروع و تا اواخر اردیبهشت ادامه داشت. همه سودوتسیوم های مورد بررسی در اواسط خرداد، کاملا خالی بودند.

    در این تحقیق تشکیل بیوفیلم در تعدادی از استرین های Pseudomonas fluorescens به صورت غیر مستقیم توسط ارزیابی سلول های باکتریایی رنگ شده توسط کریستال ویوله بررسی و با هم مقایسه گردید. استرینPseudomonas fluorescens UTPF98 به دلیل توانایی فراوان در تشکیل بیوفیلم انتخاب شد. سپس مراحل تشکیل بیوفیلم(شامل اتصال برگشت پذیر و برگشت ناپذیر به سطح، تشکیل میکروکلنی و تشکیل ماکروکلنی همراه با تولید اگزوپلی ساکارید) در این استرین بر روی اسلاید های شیشه ای ردیابی شد. در نهایت تاثیر برخی عوامل تغذیه ای شامل کاتیون ها (آهن، منیزیم، کلسیم، روی، مس، مولیبدن، منگنز، بور و کبالت)، منابع کربن (گالاکتوز، آرابینوز، زایلوز، مانوز، گلوکز و رامنوز)، اسید های آمینه (آسپارتیک اسید، فنیل آلانین، پرولین، تیروزین، لوسین، آسپارژین، آلانین، ایزولوسین، گلایسین، گلوتامیک اسید، هیستیدین، ترئونین، آرژنین، لیزین و گلوتامین) و فسفر بر تشکیل بیوفیلم در استرین نام برده بررسی شد. تاثیر کاتیون ها در تشکیل بیوفیلم متفاوت می باشد. هم چنین همه منابع کربن و اسید های آمینه آزمایش شده تاثیر مثبت بر تشکیل بیوفیلم داشتند. از طرفی تشکیل بیوفیلم با میزان فسفر موجود در محیط کشت رابطه معکوس داشت. نتایج به دست آمده از این پژوهش نشان می دهد که شرایط تغذیه ای محیط کشت بر روی تشکیل بیوفیلم تاثیر می گذارد. برای بیوکنترل موفق توسط سودوموناد های فلورسنت، شناخت شرایط تغذیه ای تاثیر گذار بر تشکیل بیوفیلم و چگونگی این تاثیر گذاری الزامی می باشد.

    در اواخر فصل زراعی سال 1388، روی نمونه های بذور آلوده رقم عنبربو قرمز برنج جمع آوری شده از مزارع موسسه تحقیقات کشاورزی واقع در شاوور از توابع شهرستان اهواز، لکه هایی به رنگ قهوه ای مشاهده گردیدند. پس از انتقال نمونه های آلوده به آزمایشگاه، قطعاتی از حد فاصل بخش های آلوده و سالم بذور انتخاب شدند و به مدت 20 دقیقه زیر جریان آب شیر شستشو شدند. پس از ضدعفونی سطحی بذور به مدت یک دقیقه در محلول هیپوکلریت سدیم رقیق شده (محتوی 1% کلر در محلول تجاری) و سه بار شستشو با آب مقطر سترون و سپس خشک کردن با کاغذ صافی سترون، قطعات روی بستری متشکل از کاغذ صافی مرطوب (Blotter method) درون تشتک های پتری سترون قرار داده شدند. تشتک های پتری درون انکوباتور با دمای 25-22 درجه سلسیوس قرار گرفتند. پس از گذشت 17 روز، پرگنه های رشد کرده قارچ در اطراف هر قطعه به روش تک اسپور خالص سازی شدند. قطر پرگنه قارچ پس از چهار شبانه روز رشد در شرایط تاریکی و دمای 25 درجه سلسیوس روی محیط غذایی سیب زمینی دکستروز آگار (PDA)، به حدود چهار سانتی متر رسید. رنگ پرگنه قارچ در مرکز تشتک پتری به رنگ خاکستری و در حاشیه به رنگ قهوه ای تیره بود. به منظور تحریک تشکیل کنیدیوم در پرگنه قارچ، جدایه به دست آمده، روی محیط غذایی حاوی کاه گندم و آب شیر- آگار دو درصد (TWA plus wheat straw) در دمای 25-22 درجه سلسیوس و با شرایط نور متناوب (12 ساعت تاریکی و 12 ساعت روشنایی) کشت داده شد. Mycologia 1992)) پس از گذشت حدود 10 روز، کنیدیوم های قارچ به وفور روی ساقه های گندم تشکیل شدند. کنیدیوم ها به رنگ قهوه ای روشن تا متمایل به تیره بوده و سطح صاف داشتند. کنیدیوم ها اغلب استوانه ای شکل و دارای 6-3 و اغلب پنج دیواره عرضی بودند. به طور تصادفی طول و عرض حدود 100 کنیدیوم و 30 کنیدیوفور از طریق میکروسکوپ دوچشمی شکل 1. کنیدیوم و کنیدیوم های گونه Biopolaris hawaiiensis Fig. 1. Conidiophor and Conidia of Bipolaris hawaiiesis olympus مدل BH-2 ساخت کشور ژاپن و با عدسی شیئی 40 اندازه گیری شد. میانگین طول کنیدیوم ها 37-75/8 میکرومتر و عرض آنها 10 - 5 میکرومتر بود. کنیدیوفورها منفرد و به حالت افراشته قرار داشتند. کنیدیوفورها به رنگ قهوه ای روشن تا متمایل به تیره بوده و میانگین طول آنها حداکثر 120 و عرض آنها 7-2 میکرومتر بود. بر اساس خصوصیات ریخت شناختی مورد بررسی و مقایسه این خصوصیات با توصیفات انجام شده توسط الیس (Ellis، dematiaceous hyphomycetes، 1976) و سیوانسان (Sivanesan، Graminicolous species of hyphomycetes، 1987)، جدایه به دست آمده تحت گونه Bipolaris hawaiiensis تعیین نام گردید. طبق بررسی های انجام گرفته، این اولین گزارش از وجود این گونه قارچی روی بذرهای برنج در ایران می باشد. گونه hawaiiensis B. برای ارزیابی قابلیت ایجاد بیماری این گونه روی گیاه برنج، آزمون بیماری زایی در شرایط گلخانه ای انجام شد. سوسپانسیون اسپور با غلظت 104×5 تهیه شد و مایه زنی صورت گرفت. بیماری زایی این گونه در محیط گلخانه ثابت نشد و برگ ها بدون علائم بودند. در شکل 1 عکس میکروسکوپی از اندام های مختلف قارچی شامل کنیدیوم، کنیدیوفور قارچ نشان داده شده که توسط دوربین Sony مدل متصل به میکروسکوپ Olympus با بزرگنمایی 100 گرفته شده است.

    بیماری آتشک با عامل Erwinia amylovora یکی از مهم ترین و مخرب ترین بیماری های خانواده گل سرخیان است. روش هایی که با سرعت و حساسیت زیاد پاتوژن را ردیابی می کنند ابزار مهمی در کنترل بیماری هستند. در این تحقیق حساسیت و دقت چهار روش تشخیصی مختلف بدون غنی سازی و به همراه غنی سازی مقایسه و ارزیابی گردید. روش کشت روی محیط نیمه انتخابی CCT تا CFU/ml1 باکتری E. amylovora را از عصاره گیاه آلوده تشخیص داد اما قادر به ردیابی آلودگی پنهان نبود. با استفاده از غنی سازی تشخیص و ردیابی باکتری در آلودگی های پنهان امکانپذیر شد. روشLateral flow immuno Chromatography غلظت CFU/ml 105 باکتری در عصاره گیاه آلوده را شناسایی نمود، ولی پس از غنی سازی علاوه بر توان ردیابی باکتری در شرایط آلودگی پنهان، دقت تشخیص این آزمون صد هزار برابر یعنی تا CFU/ml 1 افزایش پیدا کرد. روش PCR معمول و PCR آشیانه ای با استفاده از آغازگرهای اختصاصی با موفقیت قادر به ردیابی آلودگی پنهان و جمعیت های پایین باکتری در حد CFU/ml1 باکتری در عصاره گیاهی با غنی سازی و بدون غنی سازی شد. غنی سازی تاثیری در افزایش دقت این آزمون مولکولی نداشت هرچند که در ردیابی سلول های زنده باکتری بسیار موثر است.

    بیماری لکه خرمایی گندم با عامل قارچی Pyrenophora tritici-repentis(Died.) Drechsler در حال حاضر در مناطق شمالی کشور گسترش زیادی پیدا کرده است. هر چند گزارشی از جداسازی فرم غیرجنسی قارچ با نام Drechslera tritici-repentis(Died.) Shoemaker از مازندران قبلا ارائه شده (فروتن و همکاران 1995) ولی تولید فراوان کنیدیوم در محیط کشت و نیز فرم جنسی قارچ تاکنون از کشور گزارش نشده است. در این تحقیق برگ های آلوده گندم از مزارع استان های گلستان و مازندران جمع آوری و در آزمایشگاه و شرایط گلخانه ای بررسی شدند. نتایج نشان داد که پرگنه های هفت روزه قارچ در محیط V8-Caco3 به رنگ خاکستری تیره، کنیدیوفورها در پایه متورم به رنگ زرد تا قهوه ای با ابعاد 300-100 × 8-7 میکرومتر،کنیدیوم ها زرد و دارای 7-5 بند و به ابعاد200-100 × 18-14 میکرومتر با یاخته پایه مخروطی می باشند(شکل 1 a). با کشت پلاگ هایی از محیط کشت فعال V8-Caco3 روی محیط کشت آب آگار(WA) 2% حاوی قطعات برگ گندم سترون شده و نگهداری آنها در شرایط نوری و دمایی خاص شامل 12 ساعت نور فلئورسنت و 12 ساعت نور near UV در دمای 22 درجه سلسیوس، پس از دو هفته پسودوتسیوم های تیره با ابعاد 250-200 میکرومتر روی بافت برگ تشکیل شوند (شکل 1 b و c). برای بلوغ بیشتر اندام های جنسی پتری های محتوی پسودوتسیوم های قارچ به مدت دو هفته دیگر در یخچال نگهداری شدند. آسکوسپورها تخم مرغی شکل و دارای سه بند عرضی مشخص و یک بند طولی و با ابعاد50-40 × 20-16 میکرومتر بودند (شکل1d). این مشخصات با مشخصات ارائه شده برای قارچ Pyrenophora tritici-repentis در (1987)Sivanesan کاملا مطابقت داشت. جهت رعایت اصول کخ، آزمون بیماری زایی بر اساس مایه زنی سوسپانسیون آسکوسپورهای قارچ با غلظت 6 10 اسپور در هر میلی لیتر آب روی ارقام حساس بولانی و تجن گندم انجام شد. گیاهچه های گندم در مرحله 2-3 برگی مایه زنی شدند و سپس به مدت 48 ساعت زیر اتاقکی با پوشش پلاستیکی سیاه در گلخانه، تحت دمای22 درجه سلسیوس و تناوب نوری 16 ساعت (16 ساعت روشنایی و 8 ساعت تاریکی) قرار گرفتند. سپس اتاقک از روی گیاهچه ها برداشته شده و گیاهچه های مایه زنی شده همچنان در شرایط نوری و دمایی ذکر شده قرارگرفتند.

    در سال های اخیر کشت توت فرنگی (Fragaria × ananassa Duchesne) گسترش نسبتا زیادی داشته و به 3500 هکتار رسیده است. در سال 1387 از طوقه و ریشه بوته های توت فرنگی با علائم پژمردگی و در حال خشک شدن جدایه هایی شبیه به Macrophomina از استان های کردستان، مازندران و گلستان جدا سازی شدند. جدایه های حاصل در محیط کشت سیب زمینی- دکستروز- آگار (PDA) (ساخت شرکت مرک) برای بررسی مشخصات ظاهری و نرخ رشد کشت داده و در دمای 25 درجه سلسیوس نگهداری شدند. پس از 6 روز میکرواسکلروت های گرد تا دوکی شکل و تیره رنگ قارچ مشاهده و قطر پرگنه به 90 میلی متر در ظروف پتری رسید. پس از 10 روز از هر یک از شش پرگنه ده میکرواسکلروت جدا و اندازه گیری شدند. طول میکرواسکروت ها 200-50 و عرض آنها 130-40 میکرومتر تعیین شد. این مشخصات با کلیدهای شناسایی معتبر مقایسه و جدایه ها Macrophomina phaseolina (Tassi) Goid شناسایی شدند ((Holliday and Punithalingam 1970. شش جدایه از جدایه های بدست آمده برای اثبات بیماری زایی انتخاب و پس از تکثیر به خاک گلدان های حاوی بوته های سالم توت فرنگی (رقم کاماروسو) که چهار هفته از کشت آنها گذشته بود اضافه گردیدند. برای تهیه مایه اولیه عامل بیماری از محیط کشت ماسه-پرلیت-زاپک مایع سترون شده استفاده شد. از هریک از جدایه ها یک قرص پرگنه به ارلن های 250 میلی لیتری حاوی محیط کشت مذکور اضافه و پس از دو هفته نگهداری در دمای 25 درجه سلسیوس کاملا مخلوط شدند. ده گرم از مایه اولیه با خاک استریل هر گلدان یک لیتری مخلوط و در هر گلدان یک بوته توت فرنگی کشت و برای هر جدایه چهار بوته در نظر گرفته شد. گیاهان در دمای oC2±26 در دوره نور 12 ساعت روشنایی در گلخانه نگهداری شدند (Avilés et al. 2008). پس از سه هفته، در 100-50% بوته ها یک یا دو برگ پژمرده مشاهده گردید و اولین مرگ گیاه بعد از شش هفته در بوته های آلوده شده اتفاق افتاد. پس از ده هفته 100% بوته های تیمار شده پژمرده شده یا مردند. عامل بیماری مجددا از بوته های آلوده جدا سازی شد. M. phaseolina از گیاهان بسیاری از جمله یونجه، لوبیا، ذرت، پنبه، آفتابگردان، کنجد، سویا و زیتون جدا و به عنوان یکی از عوامل بیماری زای مهم گیاهی در ایران شناخته می شود (Ershad 2009). در این بررسی مشخص شد، 60-40 درصد مزارع توت فرنگی آلوده به بیماری بوده و حدود 1% از بوته ها در این مزارع بیمار تشخیص داده شدند. بیماری پوسیدگی ماکروفومینایی ریشه و طوقه توت فرنگی از کشورهای آمریکا، فرانسه، هند، اسرائیل و اسپانیا اخیرا گزارش شده (Mertely et al. 2005; Maas 1998; Avilés et al. 2008) و این نخستین گزارش از وجود بیماری در ایران است.

    بیماری پوسیدگی کف آلود چوب درختان یا چوب خیس از جمله عوارضی است که باعث ایجاد پوسیدگی در نواحی مرکزی چوب برخی از درختان سایه دار و جنگلی نظیر چنار، زبان گنجشک، صنوبر، بلوط، افرا و نارون می گردد(Scortichini et al. 1991). علائم اصلی آلودگی، تغییر رنگ چوب بخصوص در مناطق مرکزی تنه است که توام با حالت خیسی و پوسیدگی می باشد. در این مناطق مقداری گاز تولید شده که در اثر فشار آن ترشحات کف آلود زیادی از بافت خارج می گردد. از درختان آلوده چندین نوع باکتری جداسازی شده است، اما دخالت قطعی هر یک از عوامل در ایجاد بیماری مورد تردید می باشد (Murdoch & Campana 1983). در برخی از منابع به دخالت باکتری Enterobacter nimipressuralis در ایجاد بیماری اشاره گردیده است(Brenner et al. 2005). در ایران گزارش مستدلی از وجود این بیماری در دسترس نیست. در بهار 1390 علائمی از وجود یک بیماری مشابه در درختان نارون شهرستان رفسنجان دیده شد. درختان بیمار دچار زوال تدریجی بوده، خطوط سفید تا خاکستری در پوست آنها مشاهده می شد و ترشحات کف آلودی از منافذ پدید آمده در تنه و سرشاخه ها، جاری بود. در چند نوبت، از پوست ناحیه کامبیوم درختان آلوده نمونه هایی جداسازی و پس از ضدعفونی سطحی بر روی محیط های کشت NA، Kings B، YDC و EMB کشت شد. پس از 48 ساعت پر گنه های رشد یافته در سطح محیط کشت، خالص سازی و برخی از ویژگی های مرفولوژیک و بیوشیمیایی پر گنه های غالب، با استفاده از روش های متداول باکتری شناسی تعیین شد (Schaad et al.، 2001). در تمامی نمونه های به دست آمده جمعیتی نسبتا بالا از یک باکتری گرم منفی، که روی محیط کشت EMB تولید هاله سبز متالیک می نمود، مشاهده شد. جدایه های مذکور بی هوازی اختیاری بوده و واکنش اکسیداز و آرژنین دی هیدرولاز در آنها، منفی بود. جدایه ها قادر به القای واکنش فوق حساسیت در برگ های شمعدانی، هیدرولیز ژلاتین و تویین 80 نبودند و واکنش MR/VP در آنها، مثبت ارزیابی گردید. این جدایه ها قادر به استفاده از آرابیتول، آدونیتول، دلسیتول، گلیسرول و مایواینوزیتول به عنوان تنها منبع کربن نبوده اما ازگلوکز، مانیتول، مانوز، سوربیتول و رامنوز اسید تولید کردند. به منظور شناسایی دقیق تر، DNA تعدادی از جدایه ها به روش لیز قلیایی استخراج و بخشی از ژن 16S rDNA به وسیله آغازگرهای عمومی 63f و 1387r تکثیر شد(Marchesi et al. 1998). محصول PCR به وسیله کیت Accu Prep PCR purification Kit ساخت شرکت بایونر کره جنوبی تخلیص و سپس تعیین توالی گردید. نتایج نشان داد که توالی نوکلئوتیدی جدایه نارون، 99 درصد مشابه با توالی ناحیه 16S rDNA در باکتری E. nimipressuralis است و بر اساس دندروگرام ترسیم شده، با این گونه در یک گروه قرار می گیرد. با عنایت به نتایج محدود آزمون های بیوشیمیایی و تعیین توالی، جدایه های مذکور E. nimipressuralis تشخیص داده شد. بر اساس اطلاعات ما این نخستین گزارش از همراهی این باکتری با بیماری چوب خیس درختان نارون در ایران است. برای جلوگیری از گسترش بیماری، درختان آلوده توسط شهرداری رفسنجان امحا گردید.

    بیماری فیتوپلاسمایی غنچه درشت یا تورم جوانه(Big bud) یکی از بیماری های مهم گوجه فرنگی(Lycopersicum esculentum) در دنیاست و در ایران تاکنون از استان های فارس، خراسان رضوی،اردبیل، اصفهان و بوشهر گزارش شده است. در بازدید های تابستان سال 1389 از مزارع گوجه فرنگی شرق استان لرستان در منطقه درود علائم بیماری غنچه درشت گوجه فرنگی دیده شد. علائم بارز بیماری عبارت بودند از: کوچک ماندن، باریک شدن، ضخیم شدن و رنگ پریدگی برگ ها، ارغوانی شدن رگبرگ ها در قسمت زیرین برگ، کاهش فاصله میانگره ها، ضخیم شدن ساقه، رشد جوانه های جانبی ساقه و تغییرات گل شامل بزرگ شدن کاسبرگ ها و تبدیل کاسه گل به جسمی کیسه مانند و گل سبزی، راست ایستادن شاخه های انتهایی، عدم تشکیل میوه و کوتولگی گیاه. عامل بیماری به وسیله پیوند از یک بوته گوجه فرنگی دارای علائم بارز بیماری در مزرعه به سه بوته گوجه فرنگی سالم منتقل گردید و در آنها علائم بیماری غنچه درشت ظاهر شد. با روش ژانگ و همکاران (1998، J. Virol. Methods 71: 45-50) از بافت رگبرگ میانی 5 بوته گوجه فرنگی علائم دار در طبیعت، بوته های گوجه فرنگی آلوده شده در شرایط گلخانه و یک بوته فاقد علائم دی ان ای کل استخراج گردید. در آزمون واکنش زنجیرهای پلیمراز (PCR) مستقیم با استفاده از جفت آغازگر P1/P7 آلودگی نمونه های دی ان ای استخراج شده از گوجه فرنگی علائم دار در طبیعت و بوته های گوجه فرنگی مایه زنی شده با پیوند به فیتوپلاسما به اثبات رسید و قطعات 1800 جفت باز ی تکثیر شد. تحت همین شرایط در نمونه های دی ان ای گوجه فرنگی سالم چنین قطعه ای تکثیر نشد. بر اساس علائم بیماری، انتقال با پیوند و واکنش مثبت در آزمون PCR مستقیم، بیماری غنچه درشت گوجه فرنگی در استان لرستان ماهیت فیتوپلاسمایی دارد.

    Root and crown rot of corn and rape seed is one of the most important disease attack to these crops at various stages of their growth and cause considerable damage to these crops. For study and identification of the Cylindrocarpon species associated with corn and rape seed plants in Khuzestan province, this study was conducted during growing season of 2006-2007. Samples were collected from different field in different region of Khuzestan In that study 207 isolates of Cylindrocarpon were recovered. isolates belonged to the 5 species which based on there frequency are as follow: C. destructans (96), C. didymum(69), C. hederae(25),), C. obtusisporum(14), C. macrodidymum (3). This is the first report of the association of these species of Cylindrocarpon with corn and rape plants in the World. Cylindrocarpon isolates specific primers ITS4 & ITS5 were used. specific primers amplified a 500-560 bp band which is a confirmation of Cylindrocarpon species. Comparsion of the generated sequences from the mentioned species with the avalaible sequences in the gene Bank, using Blast Browers indicated that all of the generated sequences beloneged to the Cylindrocarpon species. Cylindrocarpon grouping using ITS sequences was corresponding to the classification of this geneus based on morfhological characteristics. This showen that sequences determination of ribosomal its rigon is a valuable tool in the assessment of the phylogenetic relation of Cylindrocarpon species.

    Leaf blotch of barley, caused by Rhynchosporium secalis, is a major disease of barley in Iran. Pathogenic variation of barley scald pathogen was determined by testing 47 isolates collected from five provinces of Iran. Among the isolates tested Rs55 and Rs86 were the most virulent and Rs45 was the least virulent. According to their reactions against eight differential barley cultivars containing known resistance genes, 20 different patotypes were determined and these pathotypes represent a very broad virulence spectrum. Pathotype 1 (pt1) was found in all five surveyed provinces. Pathotypes pt15, pt12 and pt13 caused susceptible reaction in 62, 50 and 50% of cultivars, respectively. Of these cultivars Igri and Armele containing BRR4 and BRR1 resistance genes were the most resistant whereas cultivars WI and Digger were the most susceptible against these pathotypes, respectively. There was no association between pathogenic variability and geographical location of R. secalis.

    In an attempt to provide scientific monograph on the genus Septoria Sacc. in Iran, several specimens of Septoria, associated with 45 host plant genera belonging to 24 families were investigated. Most studied materials were dried unidentified herbarium specimens from Mycological Collection of the Ministry of Jihad-e Agriculture (IRAN) located at the Iranian Research Institute of Plant Protection, Tehran. Some newly collected specimens were also studied. Based on our taxonomic study 59 Septoria species were identified. Among identified taxa 9 species were new to Iran. All newly recorded species were described and illustrated by microphotographs and drawings. The nine newly recorded taxa mentioned in current paper are as follow: Septoria sp. aff. alhagiae on Alhagi sp., Septoria botuliformis on Populus sp., Septoria bromi var. bromi on Bromus sp., Septoria caricina on Carex depaupertata, Septoria caricicola on Carex pendula, Septoria eremuricola on Eremurus sp., Septoria festucae on Festuca sp., Septoria sigesbeckiae on Siegesbeckia orientalis and Septoria meliae on Melia azedarach.

    Diseased rice plants with foot rot as well as dark brown lesions on sheaths and stems were observed in a paddy field in Mazandaran province, Iran. Single lesions were excised and surface sterilized and then plated on water agar. The arising fungi were purified and refrigerated in agar slants. On the basis of morphological characteristics the fungus was identified as Bipolaris sorokiniana. The ITS sequence of the fungus was submitted to a BLAST search to find most similar sequences in GenBank. The search results showed highest similarity to eight strains of B. sorokiniana and Cochliobolus sativus (teleomorph). For phylogenetic comparison, the sequences of Bipolaris spp., Cochliobolus spp. together with other species belonging to closely related fungal genera were included. The resulted phylogram made with the neighbor-joining method using the program PAUP, showed that the Iranian strain (B54) formed a monophyletic group with seven B. sorokiniana and one C. sativus strain. To prove pathogenicity, several methods were done on two popular rice cultivars. This is the first record of B. sorokiniana on Oryza sativa in Iran.

    The objective of the present study was to investigate fungal pathogens associated with turfgrass decline in Shiraz parks and other city landscapes. During 2007-2008 numerous samples of turfgrass showing chlorosis and decline were collected from different turfgrass growing regions. Several fungi were isolated from root and leaves of declining turfgrasses. The results showed that the prevalent species were as follows: Fusarium culmorum, F. equiseti, F. solani, F. crookwellense, F. semitectum, F. polyphialidicum, F. sambucinum, Pythium aphanidermatum, P. vexans, P. torulosum, P. oligandrum. P. ultimum var sporangiiferum, P. vanterpoolii,P. deliense, Bipolari australiensis, B. sorokiniana, B. cynodontis, B. spicifera, Exserohilum rostratum, Curvularia lanuta and species of Rhizoctonia solani, R. zeae and binucleate Rhizoctonia species. Pathogenicity test for each species was carried out under greenhouse condition using standard methods. The virulence of the fungal isolates was evaluated and the means of disease incidence were compared using Duncan multiple rang test. Among fungal species recovered, F. semitectum, P. aphanidermatum, R. solani AG2-2IIIB and B. sorokiniana were more pathogenic than others.

    Numerous newly collected & herbarium specimens belonging to the genus Cercospora on a variety of host plants were examined using light microscopy. Six species including Cercospora althaeina, C. flagellaris, C. texensis, Passalora circumscissa, Pseudocercospora daturina & Cercospora apii s.l. (C. avicennae, C. brachiata, C. brunkii, C. daturicola, C. hydrangeae, C. rumicis, C. kikuchii, C. xanthiicola & C. zinniae) were identified. Among these species, C. flagellaris, C. texensis, Pseudocercospora daturina are recorded for the first time from Iran. Species of Amaranthus, Datura, Rumex, Xanthium & Zinnia, respectively, are reported as new host for C. brachiata, C. rumicis, C. xanthiicola, C. zinniae in Iran.

    Pseudomonas syringae pv. syringae (Pss) is the causative agent of several bacterial diseases on a range of agricultural and horticultural crops that causes canker, leafspot and necrosis of the bark of stone fruit trees (peach, nectarine, apricot, plum and cherry). In this research samples were collected from various areas in Ardebil, Guilan, Mazandaran and Khorasane-‌Razavi provinces during 2007-2009. Bacterial strains identified as Pss, on the basis of LOPAT and GATTa were selected. Number of 70 bacterial isolates were compared based on their physiological and biochemical characteristics and total cellular protein profiles (SDS-PAGE). Pss isolates showed slight differences in phenotypic characteristics and protein profiles. To assess genetic diversity among the strains, genomic DNA was extracted from strains and used in ERIC and REP-PCR analysis. Strains formed 8 and 5 clusters in the ERIC-PCR and REP-PCR, at 75% similarity level, respectively and by the combination data set of both ERIC and REP-PCR, strains formed 9 clusters. The results demonstrated the existence of a considerable genetic diversity among Pss strains causing canker of stone fruit trees in the northern provinces of Iran.

    Heterodera schachtii is one of the most economically important pests of sugar beet worldwide. It is also widespread in most sugar beet producing regions in Iran and causes serious yield reduction and decreasing sugar content of sugar beet in infested fields. Populations of H. schachtii show differences in morphological characters. Traditional identification based on morphological and morphmetrical characters of cysts and J2s is time consuming and demands careful study. In recent years, the DNA technique based on ITS-PCR-RFLP has been a useful tool for separating of H. schachtii from the similar species. 120 populations of H. schachtii were collected from different sugar beet fields in Khorasan Razavi province. The populations were studied based on morphological and morphometrical characters. 88 populations with high variation in morphological characters were selected for further studies. DNA was extracted from full cyst and the ITS-rDNA regions were amplified with using of universal primers. The PCR product of each sample was digested with MvaI. All populations showed similar ITS-PCR-RFLP profiles, which were in agreement with the published data. Clustering of these populations based on morphometrics features separated the south regions populations from the northern populations, however, there was not logical separation among the smaller geographic areas. Four morphological types with different underbridge and bullae in vulval cones were revealed. It was demonstrated in this study that ITS-PCR-RFLP could be used as a helpful tool for identifying different populations of H. schachtii with various morphological characters.

    In order to identify the plant parasitic nematodes, a number of 45 soil samples were collected from different fields in Joubar, Karaj and Jiroft (Mazandaran, Tehran and Kerman provinces, respectively). The nematodes were extracted from soil by centrifugal flotation technique. The extracted nematodes were transferred to glycerin. The permanent slides were mounted. The nematodes were identified by morphological and morphometrical characters. In this study four species were identified viz Neodolichorhynchus sulcatus, Paraphelenchus micoletzkyi, Paratrichodorus minor and Tylenchorhynchus annulatus. Among these species N. sulcatus, P. micoletzkyi and P. minor are new records for nematode fauna of Iran.

    In this study the efficacy of sucrose, sodium alginate and Gum arabic as adjuvants on yeast cells (Pichia guilliermondii) viability in powder formulations was determined. Yeasts, grown on a sugarcane molasses-based medium, were combined with talc, kaolin, wheat bran or rice bran carriers and three adjuvants (sucrose, sodium alginate, Gum arabic) and the viability of yeast in 16 formulations was determined over a six month periods. Formulation no. 11, containing wheat bran with sucrose, and formulation no. 15, containing wheat bran with sucrose had significantly higher viable yeast cells content over a six months storage period in 4. Formulation no. 2, containing kaolin had a significantly lowest viable yeast cell. In 24˚C, formulation no. 15 had significantly highest viable yeast cells and formulation no. 2 had significantly lowest viable yeast cells content over a six months storage period. These formulations were tested against Penicillium expansumapple, the blue mold pathogen and all formulations effectively controlled the disease. Formulation no. 15, containing wheat bran and sucrose were the best formulation in controlling blue mold of apple.

    During 2007-2008, soil samples were collected from the 10-15 cm depth of the soil profiles at the fields and orchards in Mazandaran province. Using baiting methods, 121 isolates of Rhizoctonia spp. were recovered, of them 101 and 20 isolates were belonged to multinucleate (MNR) and binucleate (BNR) Rhizoctonia spp., respectively. Among the MNR isolates, 7 ones were assigned to R. solani anastomosis group one (AG-1), 38 isolates to AG-2, 5 isolates to AG-4, 3 isolates to AG-5, 13 isolates to AG-6, 11 isolates to AG-9, 3 isolates to AG-11 and 21 isolates to WAG-Z (R. zeae). Among the BNR isolates, 15 and 3 isolates were identified as AG-K and R. ramicola, respectively. AG group determination was not sufficient to identify two BNR isolates. For this reason we named them as BNR-1 and BNR-2.

    Chickpea blight caused by Didymella rabiei is the most important disease of chickpea in Kermanshah province in west of Iran. The pathogen survives in infected chickpea debris and seeds. The telemorph develops on overwintering chickpea debris and can play a roll on distribution and survival of the pathogen. Pseudothecial development of Didymella rabiei on naturally infected chickpea debris was investigated in some major growing areas of chickpea production in west of Iran. In this regard chickpea debris, stems and pods of naturally infected by D. rabiei were collected at harvest time. They were placed on soil surface of some fields in different regions of the province in September during 2003 to 2005. In 2003 D. rabiei did not develop on chickpea debris but in 2004 mature pseudothecia and pycnidia of D. rabiei formed on chickpea debris in all locations. Differentiation of pseudothecia initials happened in November, 30 to 45 days after placement of debris on the soil surface. Ascospore maturation occurred in early March. Discharge of ascospores occurred in late March and continued by mid-May. Completely empty pseudothecia were observed in mid-June.

    Biofilm formation on abiotic surfaces initiates in response to environmental cues, and the same is true for attachment to the plant surfaces. In this study the detection of biofilm formation of some Pseudomonas fluorescens strains was done indirectly by determining the extent of CV-stained cells attached to a surface. Pseudomonas fluorescens UTPF98 was selected because of its great ability of biofilm formation. And then the stages of biofilm formation (reversible and irreversible attachment, microcolony formation and macrocolony formation with exopolysacharid (EPS) production) in mentioned strain was investigated on glass slides. At last the effect of some nutritional factors like cations (Mg2+, B3+, Cu2+, Co2+, Mo6+, Zn2+, Mn2+, Ca2 + and Fe2+), carbon sources (arabinose, rhamnose, glucose, mannose, galactose and xylose), aminoacids (aspartic acid, asparagine, phenylalanine, leucine, threonine, proline, glutamic acid, glutamine, arginine, tyrosine, histidine, alanine, lysine, isoleucine and glycine) and phosphorus on biofilm formation was investigated in this study. Cations had various effects on biofilm formation. All carbon sources and aminoacids tested promoted biofilm formation. In contrast, phosphorus reduced biofilm formation. These results clearly show that nutritional status of the medium can influence biofilm formation in vitro. For successful biocontrol by fluorescent pseudomonads, one needs to understand which and how nutritional status affects the biofilm formation in potential biocontrol products.

    Rice plant is one of the most important and strategic agricultural product with having highest cultivation area after wheat in the world. Several factors including plant pathogenic fungi are causing agents of quantitative and qualitative losses in this plant. Brown spot is an important seed borne disease of rice that caused by different species of Bipolaris. Brown spots observed on infected grains in Shavor of Ahvaz at the end of crop season in 2009. Infected samples were transferred to laboratory and were cut parts between infected and healthy tissue. These parts were washed under water flow for 20 minutes. Seeds before putting into Petri dishes containing wet filter paper, were disinfected with sodium hypochloride 20% (1% active chlorine) for about 1 minutes and more was washed with sterile distilled water thrice and dried with sterile filter paper. Petri dishess were incubated under temperature of 22-25˚C. After 17 days, fungal clony formed around cultivated seed part was purified through single spore method. Colony diameter under darkness and optimum temperature was reached about 4 cm on PDA medium. Center and margin of colony was gray and dark brown, respectively. For sporulation, subcultures of colony were transfered on TWA+Wheat straw medium under optimum temperature and alternative light (12 hours darkness and 12 hours lighting). After ten days, conidia were formed on medium and wheat straw abundantly. Conidia were light to dark brown, smooth, often cylindrical, with 3-6 and often 5 pseudoseptum. Length and width of conidia were 8.75-37µm and 5-10µm, respectively. Conidiophores were single, cylindrical, light to dark brown with maximum height 120 µm and width 2-7 µm. According to the morphological characteristics and based on Ellis (1976) and Sivanesan (1987), the isolate identified as B. hawaiiensis that did not isolated from rice in Iran, before. Thus this is first report of the fungus associated to rice. Pathogenicity test of this fungus was done in greenhouse. Spore a suspension of inoculums 5×104 conidia/ml was prepared. No symptom was showed on the leaves and thaus pathogenicity of this strain did not proved in greenhouse.

    Fire blight disease caused by Erwinia amylovora is deemed to be one of the most important and devastating disease affecting in Rosacea. Sensitive and rapid detection protocols are important tools in disease control. In this research sensitivity and specificity of four different detection methods with and with out enrichment were evaluated. 1CFU/ml E. amylovora could be detected by selective culture method in infected plant extract but not in latent infections. However, enrichment was increased the susceptibility of this method to detect E. amylovora in latent infections. Lateral flow immunochromatography detect 105 CFU/ml bacteria in infected plant extract and enrichment made this method to detect latent infections and increased the sensitivity to detect 1CFU/ml bacteria. Using specific primers, the standard and nested PCR were detected 1CFU/ml bacteria in infected plant material.

    Recently, tan spot disease of wheat caused by Pyrenophora tritici-repentis(Died.) Drechsler has become very important disease in northern parts of Iran and causes severe yield loss. Although the anamorph of the fungus has been reported previously on infected leaves from Mazandaran province (Forotan et al., 1995), but there is no report on mass production of conidia in laboratory and the teleomorph of the fungus in Iran. Infected leaves were collected from wheat fields in Golestan and Mazandaran provinces and were investigated in laboratory and greenhouse conditions. The results showed that after seven days the colonies of the fungus on V8-CaCo3 medium was dark grey color, conidiophores swollen at the base, yellow to brown color, 7-8 μm thick and 100-300 μm long, conidia were yellow, with 5-7 pseudosepta, 14-18 × 100-200 μm and the basal cells were conical. 5-mm plug taken from an active colony margin of a young culture grown on V8-CaCo3 were used to inoculate 2%WA medium containing wheat leaves and these media were kept in special treatment including 12 hours photoperiod of white fluorescent and near UV light at 22 ºC. After two weeks, dark colored pseudothecia with 200-250 μm size, were produced on leaf tissues, Ascospores were oval shape with three transverse and one longitudinal septa, and 16-20 × 40-50 μm size. Based on morphological characters the fungus was identified as Pyrenophora tritici-repentis (Sivanesan, 1987). Pathogenicity test was carried out on susceptible cvs, Bolani and Tajan seedlings at 2-3 leaf stage with suspension of ascospores with concentration of 106 /ml in greenhouse. Inoculated seedlings were kept under black plastic bags for 48 hours under 22 ºC and 16 hours photopriod. Plastic bags were removed, and inoculated seedlings were left under the same condition mentioned. Symptoms of the disease including distinct necrotic and chlorotic lesions were observed after six days on inoculated leaves. The fungus was isolated from the infected leaf tissues. This is the first report of sexual stage of P. tritici-repentis and ability of ascospores to infect wheat from Iran.

    Strawberry (Fragaria × ananassa Duchesne) is produced on 3500 ha in Iran. During surveys on strawberry root and crown rot disease (February-April 2007), a fungus resemble Macrophomina was isolated from crowns and roots of collapsed and dying strawberry plants which were collected from Golestan, Mazandaran and Kordestan provinces of Iran. To study rate of growth and cultural characteristic of isolates, a plug of each isolate was cultured on potato dextrose agar (PDA) (Merck) medium and incubated at 25ºC. After 6 days the fungal grew with dark oblong microsclerotia and filled 90 mm in diameter Petri plates. After 10 days ten microsclerotia were recovered from six colonies and ranged in size from 50 to 200 μm in length and 40 to 130 μm wide and were round to oblong or irregular in shape. This description matches that of Macrophomina phaseolina (Tassi) Goid (Holliday and Punithalingam 1970). Six representative isolates were used for a pathogenicity assay on nursery runner plants (cv. Camarosa) grown on perlite for 4 weeks in a greenhouse. Inoculum of M. phaseolina was produced by blending 2-week-old cultures on sand-perlite-Czapek-dox broth in Erlenmeyer flasks (250 ml) that were incubated at 25ºC. Each of six isolates was inoculated on four plants by adding 10 g of inoculum per pot (1 l). The plants were incubated at 26±2ºC with 12-h day/night conditions in a greenhouse. Sterile, sandperlite- Czapek-dox broth was applied in control pots. After 3 weeks 50 to 100% of inoculated plants developed wilting in one or more leaves. First mortality of plants was seen approximately 6 weeks after inoculation and 100% mortality was recorded after 10 weeks. Uninoculated plants remained symptomless. The pathogen was readily reisolated from inoculated plants. This fungus has been observed to cause disease in many plants (alfalfa, bean, corn, cotton, sunflower, sesame, soybean, olive) (Ershad, 2009) but to our knowledge, this is the first report of M. phaseolina crown and root rot of strawberry in Iran. Initial studies showed that these areas were infested with about 40-60% and approximately 1% of plants were infected in fields. Similarly, this pathogen has been reported from strawberry-growing areas of the United States of America, France, India, Spain and Israel (Mertely et al. 2005; Maas 1998; Avilés et al. 2008).

    Bacterial wet-wood or bacterial slime is a common disease that affects the central core of many shade and forest trees like ash, fir, maple, oak, sycamore and elm (Scortichini et al., 1991). Symptoms of this disorder include a yellow-brown discoloration of the wood, generally confined to the central core of the tree. This affected wood is wetter than surrounding wood and is under high internal pressure. The gas pressure and high moisture content cause an oozing or bleeding of slime, from pruning cuts, through bark cracks and branch crotches. Several bacteria often are associated with wet-wood. It has not been conclusively demonstrated that these bacteria cause the disease, (Murdoch & Campana, 1983). Some recent references indicated that the bacterium Enterobacter nimipressuralis may be involved in the development of wet-wood (Brenner et al., 2005). There is no incidence of the diseas in Iran. In spring of 2011 a similar disease was observed on elm trees in repont on Rafsanjan, Kerman province. The infected tress showed liquid exuding from wounds including origin, branch stubs or barks cracks, and vertical streaks of white to gray encrustations of dried effluent on bark surfaces. Bark was removed from infected trees and segments of cambium bearing lesions were cut and surface sterilized and teased apart in a few drops of sterile water. After few minutes’ incubation, loopfuls of the suspension were streaked onto plates of NA, KINGs B, YDC and EMB media and incubated at 25oC for 2 days. Some biochemical and physiological characteristics of the isolates that produced metalic green pigment on EMB medium, was determined (Schaad et al., 2001). Strains were gram negative, and positive in tests for MR/VP but negative for oxidase, arginine dhydrolase, hydrolysis of Tween-80, gelatin liquefaction and hypersensitive reaction on tobacco (Nicotiana tabacum). All strains utilized glucose, mannitol, manose, rhamnose, and sorbitol. Arabitol, adonitol, dulcitol, mayo-Inositol and glycerol were not used as carbon source for growth. For further investigation, total DNA was extracted from some isolates using alkaline lysis procedure. A segment of 16s rDNA gene were amplified by PCR using 63f and 1387r as a reverse and forward universal primers, respectively (Marchesi et al., 1998). The amplified fragments (PCR products) were purified using Accu Prep PCR purification Kit (Bionner, South Korea) and were sequenced. Sequencing of the entire 16S rDNA gene of representative elm isolates allowed us to compare their relatedness. Elm isolates clustered in the same clade with E. nimipressuralis. Nucleotide sequence analysis in GenBank showed that the nucleotide sequence of 16s rRNA gene of Elm strains had 99% similarity to reference strains of E. nimipressuralis. Based on our knowledge this is the first report of isolation of E. nimipressuralis associated with bacterial wet-wood from elm trees in Iran. The infected trees were eradicated by municipality organization.

    Phytoplasmal big bud is one of the important diseases of tomato (Lycopersicon esculentum) in the world. The disease has been reported from Iranian provinces of Bushehr, Isfahan, Fars, Khorasane Razavi and Ardabil. During a summer surrvey in 2010, symptoms of big bud disease was observed in tomato fields of Dorood area in Lorestan province. Characteristic symptoms of the disease were small thickened and chlorotic leaves, purple coloration along the veins of the underside of young leaves, proliferation of auxillary buds, abnormal flowers with enlarged and united calyx segments, and virescent petals, sterility and dwarfing The disease agent was successfully transmitted by side veneer grafting from a naturally symptomatic tomato to 3 healthy tomato plants causing big bud symptoms. Total DNA was extracted from midrib tissue of 5 naturally symptomatic, 3 experimentally inoculated and a healthy tomato plant using Zhang et al. (1998, J. Virol. Methods 71: 45-50) procedure. Direct polymerase chain reaction (PCR) with primer pair P1 ⁄ P7 yielded fragments of approximately 1.8 kbp from five of five field collected big bud affected plants in Dorood and all symptomatic experimentally inoculated plants. No products were amplified with DNA samples from asymptomatic tomato plant. On the basis of disease symptoms, graft transmission and positive reaction with the phytoplasma-specific primed PCR, tomato big bud disease in Dorood has phytoplasmal etiology. This is the first report of tomato big bud disease in Lorestan province.
