Swing.]

Molecular responses of susceptible (Marfona) and tolerant (Els) cultivars of Solanum tuberosum, and a resistant genotype of S. phureja were comparatively studied using a cDNA-AFLP technique to identify candidate genes involved in defense mechanisms against Ralstonia solanacearum. Transcriptional changes were studied at 0, 2, 4 and 7 days post inoculation of the plants by R. solanacearum through root inoculation under in vitro conditions. Of the 2820 transcript derived fragments (TDFs) detected, 206 TDFs showed variable patterns. In total 20 TDFs with stronger differential induction were sequenced. These TDFs showed significant homologies with the genes involved in physiological processes, stress responses, cell structure, defense, transcription and protein synthesis/folding. Approximately 5% of the sequenced TDFs were identified with unknown function. Induction of a higher number of genes was altered in response to R. solanacearum in S. phureja compared to S. tuberosum.

South citrus growing belt of Iran including Hormozgan, Sistan-Baluchestan, Bushehr, Fars and Kerman provinces, has a long history of cultivation of acid lime (Mexican lime) [Citrus aurantifolia (Chritm.) Swing.]. In two recent decades due to witches' broom disease of lime (WBDL), caused by 'Candidatus Phytoplasma aurantifolia', thousands of acid lime trees have been killed in the south of Iran. Using resistant or tolerant varieties is one of the important strategies for controlling WBDL . The aim of this study was producing new acid citrus hybrids resistant to the WBDL disease. Four types of crosses were carried out. The female parent was ‘Eurekaʼ lemon [C. limon (L.) Burm. F.] and the male parents were ‘Yuzuʼ [C. junos Sieb. ex Tan.], ‘Cleopatraʼ mandarin [C. reshni Hort. ex Tan.], ‘Kusaieʼ lime (C. limonia Osb.) and ʻHamlinʼ orange [C. sinensis (L.) Osb.]. From the above parents, 79 hybrid plants were successfully inoculated with the 'Ca. P. aurantifolia' by inverted T-budding. Appearance of disease symptoms and nested-PCR assay were used to assess inoculated hybrids for resistance. Two years post-inoculation 63 hybrids remained symptomless. Among symptomless hybrids, 35 and 28 hybrids were negative and positive for presence of WBDL phytoplasma, respectively. Symptomless hybrids negative for WBDL phytoplasma are under more phenotypic evaluation and may be promising as new acid citrus resistant to WBDL.

At present, replant disorder and peach (Prunus persica (L.) Batsch) short life has become an important problem in peach production, and it has seriously restricted the development of the peach industry in Chahar Mahal va Bakhtiary province. During 2014-2017, 108 samples of plant tissues and rhizosphere were collected from the peach orchards of this province. Samples were investigated to isolation and identification microorganisms and pathogens associated with symptoms of peach replant problem. Symptoms including: yellowing, stunting, abscission of leaf, leaf discoloration and chlorosis, crown and root rot, canker and death of young trees. From plant and soil samples, the frequency respectively, isolates of Bacillus sp, Fusarium sp, Pseudomonas flourescens, Mesocriconema xenoplax, Aspergillus sp, Alternaria sp, Pantoea agglomerans, Arthrobacter sp, Phytophthora sp, Pseudomonas syringae, Pseudomonas putidae, Verticillium dahliae, Dematophora necatrix, Trichoderma sp, Streptomyces sp, Meloidogyne sp, Pratylenchus sp, Paratylenchus sp, Stenotrophomonas maltophilia, Pythium spand Rhizoctonia spwere isolated and identified. About 11% of isolates were unknown. The amount of replant disorder and isolation of these microorganisms in orchards that were planted immediately after the destruction of previous trees or planted the same previous species was about 30% more than the orchards that had not been cultivated (fallow) for at least one year. On this basis canbe say these microorganisms are the most important agents associated with replant problem and life short in peach orchards in Chahar Mahal va Bakhtiary province. Koch's postulates investigations are necessary to determine the role of these microorganisms in this replant disease

Cucumber mosaic virus (CMV) belongs to the genus Cucumovirus of the family Bromoviridae. Compared to other plant viruses, it has the broadest host range, and is reported from different regions of Iran. In the present study, the genetic diversity of Iranian CMV isolates was estimated through the sequence analysis of all the CMV genes and compared to those in other countries. To this purpose we report the genome sequence of two CMV strains isolated from squash (SqSh) and melon (MeEs) in Shiraz and Esfahan. The two isolates were closely related based on the analysis of all gene sequences. Phylogenetic analysis revealed that most of the Iranian CMV isolates belonged to the subgroup IA, however, they did not share a most recent common ancestor based on RNAs 2 and 3. The reassortment type of IA-IA-IB was fount to have played a very important role in the evolution and genetic diversity of CMV population in Iran. 1a and CP ORFs had the lowest and highest genetic diversity among Iranian CMV population of the subgroup I, respectively. We suggested that migration in long and short distances have played a role in evolution and genetic structure of CMV strains in Iran.

Cultivation of medicinal and aromatic plants has increased in the tropical countries to meet the needs of the pharmaceutical, cosmetics and food industries. Many plants are susceptible to various diseases and pests. Phylum nematoda includes the group of multicellular and vermiform animals, which found in all different habitats. Plant parasitic nematodes are estimated to be about 10% of the phylum. Using of medicinal plants for the human disease treatment is almost universal among non-industrial societies and countries. In current study, in addition to main hosts of cabbage cyst nematode (kohlrabia, Brassica oleracea L. var. gongylodes, and white cabbage, Brassica oleracea L. var. captita alba) that are naturally infected by cyst nematode in vegetables growing area of Tariz, thirteen species of medicinal plants namely Dracocephalum moldavica L., Melissa officinali L., Lallemantia iberica F., Sisymbrium sophia L., Sinapis alba L., Brassica nigra L., Portulac oleracea L., Foeniculum vulgare Mill, Ocimum basilicum L., Alyssum homolocarpum L., Cichorium intybus L., Brassica oleracea var. gongylodes, Vicia ervilia L., Eruca sativa L. were selected and inoculated with the cabbage cyst nematode under greenhouse conditions. The plants were randomly sampled in different phenological stages between germination to flowering periods. The sampled roots roots were stained using lactoglycerin-fushin acid solution and microscopic slides prepared. The results showed that beside positive control plants (kohlrabi) different developmental stages of the nematode were detected in Black mustard (Brassica nigra. L), Yellow mustard (Sinapis alba L.), Turnip (Eruca sativa L.), Alyssum (Alyssum homolocarpum L.) and Soothe (Sysymbrium Sophia L.) roots. The infected plants as weed in vegetable field of the area could increase the spreading of the nematode and the level of yield losses. Meanwhile, the unaffected medicinal plants could be considered as potential rotation candidates for the cabbage.

Root and crown rot is one of the most important fungal plant diseases which is observed in fields annually. One of these agents are Fusarium species which cause lots of damages to crops such as maize. In a survey in cereal fields in Lorestan and Ilam provinces in 2016, plants with fusarium root and crown rot symptoms were collected and were transferred to laboratory. In vitro experiments were started quickly. After isolation in Nash & Snyder and DCPA, isolates were purified and then were transferred to PDA, CLA, KCl, SNA and strile distilled water to microscopic observations. Macrocinidia, microconidia, conidiogenous cells, chlamydospores and colony features were studied according to diagnistic keys (Marasas, 1986; Leslie and Summerell, 2006). Results showed that isolated species were F. polyphialidicum Marasas. Average growth rate, white to pale orange aerial mycelium, white to yellow color in reverse colony on PDA were observed. Robust and large macroconidia with moderately curvature were produced in white sporodochium. Size of these spores were 4.5-6 × 35-65 µm, they were 5-7 septate, with foot shaped basal cell and tapering apical cell. Microconidia were clavate and 0-septate, and 2.5-3.5 × 35-65 µm. Monophialids and polyphialides and single, in pairs, clumps and in chains chlamydospores were another features of this species. According to above properties and identification, pathogenicity test were examined in greenhouse conditions according to Burgess et al. (1994) method. This is the first report of F. polyphialidicum on maize in Iran.
