    بیان ژن های مقاومت توسط پیشبرهایی که صرفا در پاسخ به بیمارگرها فعال می شوند مزیت بزرگی در تولید گیاهان مقاوم به بیماری محسوب می شود. در این بررسی ژن کیتیناز کایمری تحت پیشبر القاءپذیر با بیمارگر SP-DDEE به گیاه کلزا منتقل شد و پتانسیل این گیاهان در مهار رشد بیمارگرهای قارچی در محیط in vivo مورد ارزیابی قرار گرفت. در این راستا، سه سازه ی pGDEC، pGMPC و pBISM2 به ترتیب حاوی پیشبر SP-DDEE (شامل دوعنصر D و دو عنصر E17 بالادست پیشبر حداقل)، SP-MP (شامل فقط پیشبر حداقل) و CaMV35S (شامل پیشبر دائمی) جهت تراریختی گیاهان مورد استفاده قرار گرفت. بررسی ها با استفاده از آزمون فعالیت آنزیمی مشخص نمود که پیشبر القائی مورد نظر نسبت به محرک متیل جاسمونات القاءپذیر بوده ولی پاسخ مشخصی به محرک سالیسیلیک اسید نشان نداد. بعلاوه، نتایج مشخص نمود عصاره دیسک های برگی گیاهان تراریخت حاوی پیشبر SP-DDEE تیمار شده با متیل جاسمونات به نسبت گیاهان شاهد قادر به مهار رشد بیمارگرهای قارچی Sclerotinia sclerotiorum و Rhizoctonia solani می باشند. در مجموع مشخص شد پیشبر SP-DDEE، نه تنها نسبت به محرک متیل جاسمونات به عنوان مولکول پیام رسان در مسیر مقاومت به بیمارگرهای نکروترف القاءپذیر می باشد، بلکه میزان بیان ژن کیتیناز کایمری، تحت کنترل این پیشبر نیز جهت افزایش مقاوت نسبت به بیمارگرهای قارچی مناسب به نظر می رسد.

    گل جالیز (Orobanche spp.) انگل مطلق ریشه گیاهان دولپه و دارای خسارت فراوان می باشد. بنزوتیادیازول به عنوان ماده محرک، پتانسیل القای مقاومت در برخی گیاهان را دارد. در این پژوهش، تاثیر غلظت های مختلف BTH (10، 100 و 500 میلی گرم در لیتر) در القای مقاومت در گیاه گوجه فرنگی در برابر گل جالیز Loefl Orobanche cernua موردمطالعه قرار گرفت. ابتدا حساسیت هفت رقم نسبت به بیمارگر ارزیابی شد. دو رقم ارلی اربانا و سوپرچف به ترتیب به عنوان متحمل ترین و حساس ترین ارقام جهت آزمایش گلخانه ای بعدی انتخاب شدند. نشاءها قبل از انتقال به گلدان های حاوی 20 میلی گرم بذر گل جالیز، به مدت یک ساعت در غلظت های مختلف BTH خیسانده و همچنین 15 روز بعد، مجددا آبیاری شدند. پس از سه ماه، شاخص های رشدی میزبان و بیمارگر بررسی و اندازه گیری شد. نتایج نشان داد که رقم متحمل و تمامی غلظت های BTH در مهار بیمارگر موثر بودند. غلظت mg/l100 و 500 در رقم ارلی اربانا مهار صددرصدی بیمارگر را دربر داشت. همچنین تغییرات فعالیت آنزیم فنیل آلانین آمونیالیاز و پراکسیداز در زمان های 24، 48، 72 و 96 ساعت بعد از کاربرد BTH موردبررسی قرار گرفت. بر اساس نتایج، بیشترین فعالیت آنزیمی در رقم ارلی اربانا در غلظت mg/l 500 در حضور گل جالیز حاصل شد. درمجموع، استفاده از ترکیبات تجاری BTH به عنوان یک فعال کننده سیستم دفاعی گیاه و یک ترکیب جدید، ساده و سازگاربامحیط زیست می تواند جهت مهار گل جالیز در کشاورزی پایدار بکار رود.

    در سال های اخیر با توجه به گسترش بیماری شانکر درختان زردآلو و بادام مشکوک به آلودگی با عامل Pseudomonas syringae pv. syringae (Pss) در استان آذربایجان شرقی، جهت ردیابی و شناسایی برخی از خصوصیات فنوتیپی و ژنوتیپی جدایه های بیمارگر Pss، نمونه برداری از 13 منطقه مختلف جغرافیایی استان انجام شد. از نمونه های مشکوک با علایم ظاهری شانکر، تعداد 14 جدایه باکتری گرم منفی و تولیدکننده رنگدانه فلورسنت، بر اساس آزمون های بیوشیمیایی و افتراقی LOPAT و GATTa به عنوان Pss شناسایی شدند. جدایه ها اختلاف بسیار جزیی در آزمون های مورفولوژیکی، بیوشیمیایی و فیزیولوژیکی نشان دادند. تعیین ترادف بخشی از ناحیه ژنی rpoD، نتایج آزمون های مورفولوژیکی و فنوتیپی را تایید نمود. جدایه ها تفاوت معنی داری در مهار رشد رویشی پرگنه قارچ Geotrichum candidum با تولید زهرابه داشتند. در تمامی جدایه های Pss قطعه 725 جفت بازی ژن syrB دخیل در سنتز زهرابه سیرینگومایسین ردیابی گردید. جدایه ها بر اساس آزمون بیماری زایی روی سرشاخه های بریده زردآلو به دو گروه پرآزار و کم آزار تقسیم شدند. تعیین ترادف بخشی از ژن syrB و رسم تبارنما با روش بایزین نشان داد که در میان ده جدایه مورد بررسی تنوع ژنتیکی وجود داشته و جدایه ها به دو گروه کاملا متمایز تقسیم شدند. گروه بندی حاصله با گروه بندی به دست آمده بر اساس تعیین ترادف بخشی از ژن خانه دار rpoD مشابهت داشت.

    نماتد سیستی کلم (Heterodera cruciferae Franklin، 1945) از جمله نماتدهایی است که احتمالا به علت خسارت نسبتا کم و پراکندگی محدود جغرافیایی، تحقیقات کم تری در مقایسه با سایر نماتدهای سیستی بر روی آن انجام شده است. تمامی اعضای خانواده کلم سانان (Brassicaceae) و برخی از گیاهان خانواده نعناعیان (Lamiaceae) توسط این نماتد آلوده می شوند. آلودگی به نماتد سیستی کلم در سبزی کاری های اطراف تبریز روی انواع کلم و هم چنین علف های هرز سلمه تره (Chenopodoum album)، شیرتیغی (Sonchus asper) وSisymbrium loeseliiمشاهده گردید. جمعیتی از این نماتد از مزارع آلوده جمع آوری و بر اساس صفات ریخت شناختی، ریخت سنجی و مولکولی شناسایی شد. تکثیر و نگهداری نماتدها روی میزبان اصلی آن در شرایط گلخانه صورت پذیرفت. استخراج RNA از نماتدها و واکاوی ترانسکریپتوم جمعیتی شامل مراحل لاروی سن سه، چهار و بالغ انجام گردید. نتایج نشان داد که بسیاری از ژن های بیماری زایی گزارش شده از سایر نماتدهای انگل گیاهی مانند نماتد طلایی سیب زمینی، نماتد سیستی چغندرقند، نماتد قلوه ای و نماتد چوب کاج، در نماتد سیستی کلم نیز بیان می‏گردند. ژن های بتا-1،4-اندوگلوکوناز (سلولاز)، 3-3-14، گلوتاتیون پراکسیداز، پکتات لیاز، کیتیناز،major sperm protein ، کالریتیکولین، کالمودولین، venum allergene like protein، ubiquitin extension protein، پراکسی ردوکسین، کوریسمات میوتاز، آلدولاز، آنکسین، گالکتین،VRFamid receptor و آرژینین کیناز در این نماتد شناسایی و در پایگاه اطلاعات داده ها (NCBI) ثبت گردید.

    به منظور شناسایی نماتودهای جنس‏های Ditylenchus Filipjev، 1936 و Nothotylenchus Thorne، 1941 طی سال‏های 1392 تا 1395 تعداد 341 نمونه‏ بافت گیاهی و خاک از مزارع، باغات و مراتع استان‏های فارس، کهگیلویه و بویراحمد، بوشهر و هرمزگان، همچنین 32 نمونه از استان‏های مازندران، گلستان، همدان، لرستان و ایلام جمع آوری گردید. پس از استخراج، کشتن، تثبیت و انتقال نماتودها به گلیسیرین خالص، شناسایی گونه ها با استفاده از میکروسکوپ نوری مجهز به دوربین دیجیتال و بر اساس ویژگی های ریخت‏شناختی و ریخت‏سنجی انجام گرفت. نتایج نشان داد گونه های این جنس‏ها در 7/54 درصد نمونه ها وجود داشتند. در این بررسی 12 گونه از جنس ‏Ditylenchus و پنج گونه از جنس Nothotylenchus شناسایی گردید. گونه های D. myceliophagus و D. medicaginis به ترتیب با 8/38 و 1/35 درصد و گونه N. geraerti با 60% دارای بیشترین فراوانی بودند. سه گونه N. basiri، D. geraerti و D. solani برای اولین بار از ایران گزارش و توصیف می‏شوند. علاوه بر آن شرح کاملی از گونه های N. adasi و N. medians و کلید شناسایی گونه های شناخته شده جنس‏های Ditylenchus و Nothotylenchus در ایران ارائه شده است.

    پیچیدگی برگ از بیماری های خسارت زای گوجه فرنگی است که توسط چندین ویروس از جنس Begomovirus (خانواده Geminiviridae) از جمله ویروس پیچیدگی برگ زرد گوجه فرنگی (TYLCV) ایجاد می شود. این ویروس توسط سفید بالک Bemisia tabaci منتقل می شود. بمنطور بررسی تغییرات وقوع و شدت بیماری و تخمین جمعیت ناقل ویروس در سه رقم گوجه فرنگی شامل YRMT، PANDA-F1 و BRST، 15 گیاه از هر رقم که در گلخانه پرورشی-تولیدی دانشکده کشاورزی شیراز کشت شده بودند بصورت تصادفی انتخاب و میزان آلودگی طبیعی آنها به جدایه ایرانی TYLCV (TYLCV[Ab]) در فواصل زمانی 20 روز (از 20 روز پس از تاریخ کاشت تا انتهای 100 روز) بوسیله آزمون PCR با استفاده از آغازگرهای اختصاصی TYLCV[Ab] تعیین شد. پس از بروز علائم، شدت بیماری با روش چشمی و با مقیاس ارزیابی ترتیبی اندازه گیری و به مقیاس نسبی تبدیل شد. به منظور برآورد جمعیت B. tabaciتعداد 50 عدد شفیره از برگ های هر گیاه جمع آوری و با استفاده از خصوصیات ظاهری شفیره، شفیره های B. tabaci از شفیره های سفیدبالک گلخانه (Trialeurodes vaporariorum) تفکیک و شمارش گردیدند. تجزیه آماری نشان داد که تفاوت معنی دار در میزان وقوع و شدت بیماری و جمعیت B. tabaci بین ارقام وجود دارد. وقوع بیماری از 6/6% تا 6/86% بترتیب در 40 و 100 روز پس از تاریخ کاشت برآورد شد. وقوع و شدت بیماری و جمعیت حشره ناقل در رقم های YRMT و PANDA-F1 نسبت به رقم BRST روندی صعودی تری نشان دادند و با شیب بیشتری افزایش یافتند. رقم BRST نسبتا« متحمل تر از دو رقم دیگر (YRMT و PANDA-F1) که نسبت به TYLCVحساس بودند تشخیص داده شد.

    زنگ زرد گندم با عامل Puccinia striiformis f. sp. tritici یکی از مخرب ترین بیماری های گندم در جهان است. این بیماری یک تهدید دایمی برای تولید گندم در بیشتر کشورهای تولید کننده این محصول می باشد. تعداد زیادی از پاتوتیپ های (نژادهای) زنگ زرد با دارا بودن فاکتورهای بیماریزایی (ویرولانس= پرآزاری) جدید به طور پیوسته در نواحی مختلف جهان تکامل یافته و موجب شکسته شدن مقاومت منابع مورد استفاده می شوند (Chen 2005).

    بیماری زوال یک تهدید بالقوه بقاء حیات درختان بلوط در بسیاری از کشور های اروپایی، آمریکایی و آسیای مرکزی به شمار می رود (Luchi et al. 2005). این بیماری منجر به مرگ تدریجی و یا سریع درختان بلوط در عرصه های جنگلی می شود. قارچ Biscogniauxia mediterranea، عامل بیماری پوسیدگی ذغالی بلوط، به عنوان یکی از عواملدخیل در زوال درختان بلوط در ایران شناخته شده است (Mirabolfathi et al. 2013، Ghasemi-Esfahlan et al. 2016). با توجه به اینکه اطلاعاتی در خصوص شیوع بیماری پوسیدگی ذغالی بلوط در جنگل های بلوط استان آذربایجان غربی در دسترس نبود، در این راستا، در تابستان و پاییز سال 1395 بازدید هایی از جنگل های سردشت به عمل آمد و از قسمت های تنه و شاخه درختان بلوط (Quercus brantii) دارای علایم زوال نمونه برداری شد. در این تحقیق شش جدایه از گونه B. mediterranea از درختان بلوط دارای علایم زوال جداسازی گردید. هویت جدایه های قارچی با استفاده از داده های توالی ناحیه ITS-rDNA و استفاده از آغازگر های اختصاصی گونه تایید گردید. آزمون بیماری زایی با استفاده از دو جدایه B. mediterranea مطابق روش قاسمی اسفهلان و همکاران (Ghasemi-Esfahlan et al. 2016) انجام گردید.

    The expression of defensive genes from a promoter that is specifically activated in response to pathogen invasion is highly desirable for engineering disease-resistant plants. In this study, chimeric chitinase gene under the control of SP-DDEE pathogen inducible promoter transformed to the canola plants and these plants were evaluated for their in vivo biocontrol potential against fungal pathogens. In this regard, three constructs, pGDEC, pGMPC, pBISM2 containing synthetic promoters, SP-DDEE (two D and two E elements minimal promoter), SP-MP (minimal promoter), and the CaMV35S constitutive promoter, respectively were used. Enzyme activity assay demonstrated that the synthetic pathogen-inducible promoter was responsive to the Methyl jasmonate (MJ) elicitor, but not responsive to the salicylic acid. Moreover, results indicated that leaf total protein from transgenic lines harboring the SP-DDEE promoter treated with MJ, inhibited the growth of the fungal pathogens of Sclerotinia sclerotiorum and Rhizoctonia solani. Overall, data show that not only the SP-DDEE synthetic promoter is highly responsive to MJ, as an important chemical signal in necrotrophic pathogen defense, but the inducible expression of the chimeric chitinase gene, when controlled by the SP-DDEE promoter, is also seems to be appropriate to increased resistance to fungal pathogens.

    Benzothiadiazole (BTH) is as an elicitorwith potential to induction of resistance in some plants. In this research, the effect of different concentrations of BTH (0, 10, 100, 500 mg/ l) for induction of resistance in tomato against Orobanche cernua Loeflwas investigated. For following experiment in greenhouse condition, Early Urbana(EU) and Super Chef respectively the most tolerant and sensitive cultivars were selected. Before transplanting of tomato into contaminated pots with 20 mg broomrape seeds, the roots of plant were soaked in BTH for an hour. Also fifteen days later, the seedlings were irrigated with BTH. Growth parameters of tomato plants and pathogen were assessed 3 months later. The results demonstrated that all concentrations of BTH and tolerance variety were effective in inhibition of pathogen. The best broomrape controls were recorded by 100 percent at 100 and 500 mg/l on EU cultivar. The activity of the enzyme phenylalanine ammonia lyase (PAL) and peroxidase (POX) at 24, 48, 72 and 96 hours were estimated after irrigated plants with BTH. The results showed that there was a significant effect of the concentrations of BTH and pathogen on the activity of PAL and POX enzymes that are involved in plant defense mechanisms. The highest enzymatic activity was obtained in EU at concentration of 500 mg/l in the presence of O. cernua. In conclusions, the use of BTH commercial compounds as plant defense activator is a new, simple strategy to control broomrape and compatible with the environment for the sustainable agriculture.

    In recent years, due to the increase in canker symptoms on almond and apricot trees suspected to Pseudomonas syringae pv. syringae (Pss) strains in East Azerbaijan province, detection and determination of some phenotypic characters and genetic characterizations of Pss strains were performed after sampling from 13 different geographical regions. A total of 14 Gram negative and fluorescent producing bacterial strains isolated from apparently diseased samples were identified as Pss based on biochemical and differential LOPAT and GATTa tests. Strains showed slight differentiation in morphological, biochemical and physiological tests. Partial sequences of the rpoD gene confirmed the results of the morphological and phenotypical tests. Pss strains had significant differences in regard to inhibition of Geotrichum candidum mycelial growth by toxin production. A 725-bp fragment of the syrB gene required for synthesisof syringomycin was amplified in all Pss strains. Based on pathogenicity tests on apricot twigs, strains were divided into two groups, one of higher and one of lower virulence. Partial sequences of the syrB gene and constructing a dendrogram using Bayesian inference showed genetic diversity among ten studied strains and divided them into two main groups.This grouping was similar to strains’ grouping based on partial sequences of the rpoD gene.

    Cabbage cyst nematode (Heterodera cruciferae, Franklin, 1945) is a nematode that probably due to less geographical distribution and relatively lower crop losses in comparison to related species has gained less attention. All members of Brassicaceae and some species of Lamiaceae are infected by the nematode. Cabbage cyst nematodes was observed in vegetable growing areas of suburb of Tabriz on cabbage and some weeds such as Chenopodoum album, Sonchus asper and Sisymbrium loeselii as well. A population of this nematode collected from the infected vegetable fields of Tabriz and its identity was confirmed based on morphological, morphometrical and molecular data. Propagation and maintainance of the nematode was carried out on its main host (white cabbage and kohlrabi) under greenhouse conditions. RNA was extracted from the nematode and transcriptome of the population including third, fourth stage juveniles and adult females analysed. The results revealed that most already reported parasitism genes from other plant parasitic nematodes such as potato cyst nematode, sugar beet cyst nematode, reniform nematode and pine wood nematode are expressed in cabbage cyst nematode, too. Furthermore, it was found that, β-1,4-endogluconase, 14-3-3, glutathione peroxidase, pectatelyase, chitinase, major sperm protein, calreticulin, calmodulin, venum allergen like protein, ubiquitin extension protein, peroxiredoxin, chorismate mutase, aldolase, annexin, galectin, VRFamid receptor and arginine kinase could be considered as a part of the nematode parasitism effectors.     For identification of nematode species of the genera Ditylenchus and Nothotylenchus, 341 plant and soil samples from farms, orchards and pastures of Fars, Kohgiluye and Boyer-Ahmad, Bushehr and Hormozgan provinces, and 32 samples from Mazandaran, Golestan, Hamadan, Lorestan and Ilam provinces were collected, during 2013-2016. After extraction, killing and fixing, the extracted nematodes were transfered to anhydrous glycerol. Species identification was done using light microscope equipped with digital camera, based on morphometric and morphological characters. The results showed that the genera were existed in 54.7% of the samples. In this study, 12 species of Ditylenchus and five species of Nothotylenchus were identified. D. myceliophagus and D. medicaginis with 38.8% and 38.1% frequency, respectively, and N. geraerti with 60% frequency were the most common species. The species N. basiri, D. geraert and D. solani are new records and were described for the first time from Iran. Moreover, descriptions of N. adasi and N. medians, and the identification key for the Iranian species of the genera Ditylenchus and Nothotylenchus were provided.

    Tomato leaf curl disease (TLCD) is one of the most destructive diseases of tomato caused by a number of begomoviruses including Tomato yellow leaf curl virus (TYLCV) belonging to the genus Begomovirus (family Geminiviridae). TYLCV is transmitted by the whitefly Bemisia tabaci. In order to investigate changes in occurrence and severity of TLCD and evaluate the viral vector population on three tomato cultivars namely Yellow Round Multiple Truss (YRMT), PANDA F1 and Black Russian ST 175 (BRST) in a greenhouse in College of Agriculture, Shiraz University, 45 plants (15 plants from each cultivar) were randomly selected. Specific primers of an Iranian isolate of TYLCV (TYLCV[Ab])were used to detect plants naturally infected with TYLCV by PCR. After symptom emergence, both disease incidence (DI) and severity (DS) were evaluated visually using an ordinal rating scale. To estimate vector’s population, 50 whitefly puparia were collected from two leaves of each cultivar and based on the vasiforme and outward specifications of puparia, B. tabaci distinguished from Trialeurodes vaporariorum. The data analysis by SAS software indicated significant differences in DI, DS and vector population dynamics between three tomato cultivars. The DI was estimated to be 6.6% to 86.6%, at 40 and 100 days after planting date, respectively. The incidence and severity of disease and insect vector populations in YRMT and PANDA-F1 cultivars showed a higher trend and rose more sharply compare to those in the BRST cultivar. BRST was comparatively more resistant than the two other cultivars (YRMT and PANDA) which were susceptible to TYLCV.

    Stripe (yellow) rust caused by Puccinia striiformis f. sp. tritici is the most devastating disease of bread wheat (Triticum aestivum) in the world. The disease represents a constant threat to wheat production in many wheat-producing countries. A wide range of virulent yellow rust pathotypes (races) is evolving in different regions of the world causing the breakdown of widely utilized sources of resistance in wheat (Chen 2005). On the other hand, comprehensive information on pathogen virulences and its variation and also epidemiological information on pathogen movements provides a basis for the development of an early effective warning system (Yahyaoui et al. 2001).

    Decline disease is a potential threat for survival and longevity of oaks species in many European, American and Central Asian countries (Luchi et al. 2005). Biscogniauxia mediterranea, the causal agent of oak charcoal rot disease, is known as one of the fungal species involved in oak decline in Iran (Mirabolfathi et al. 2013, Ghasemi-Esfahlan et al. 2016).
