    پوسیدگی صورتی ناشی از Phytophthora erythroseptica بیماری خاک برد مهمی در سیب زمینی است که سبب بروز خسارت قابل توجهی در مزرعه و انبار می شود. مجموعه ای از جدایه های P. erythroseptica به دست آمده از مناطق تولید سیب زمینی در چهار قاره ی مختلف شامل آمریکا، آسیا، اروپا و استرالیا مورد ارزیابی های فیلوژنتیکی، ریخت شناختی و فیزیولوژیک قرار گرفت. در تمام درخت های فیلوژنتیکی رسم شده بر اساس توالی های هسته ای (فاصله ی ترانویسی شده ی داخلی، بتاتوبولین و پروتئین شوک حرارتی 90) و میتوکندریایی (زیرواحد یک سیتوکروم اکسیداز سی) و درخت برآیند توالی های هسته ای، جدایه های P. erythroseptica همراه با جدایه ی تیپ، یک دودمان فیلوژنتیکی مجزا تشکیل دادند. همچنین هیچ گونه تنوع ریخت شناختی و فیزیولوژیک در جدایه ها مشاهده نشد. کلیه ی جدایه ها هم تال بوده، آنتریدیوم های انتهایی پیراماده، ااگونیوم های گرد و ناپرساز و اسپورانژیوم های بدون پستانک، بیضی تا تخم مرغی شکل تولید کردند. نتایج نشان که داد جدایه های P. erythroseptica متعلق به مناطق جغرافیایی مختلف از نظر خصوصیات فیلوژنتیکی و ریخت شناختی یک نواخت اند. این یکنواختی ممکن است ناشی از طبیعت هم تال P. erythroseptica یا انتشار یک همسانه ی منفرد توسط غده های آلوده ی سیب زمینی باشد.

    این تحقیق با هدف مقایسه بیماری‏زایی و برخی ویژگی‏های زیست‏شناختی و ریخت‏سنجی ماده‏های چهار جمعیت‏ نماتد سیستی Heterodera schachtii، جمع‏آوری شده از مزارع چغندرقند استان‏های اصفهان، شیراز، آذربایجان غربی و خراسان و واکنش ژنوتیپ‏های چغندرقند در برابر آن‏ها انجام شد. واکنش 12 ژنوتیپ چغندرقند در گلخانه و بر اساس تعداد ماده بالغ تشکیل شده بر روی ریشه‏ها ‏بررسی شد. همچنین در هر جمعیت اندازه ماده‏های تشکیل شده روی هر ژنوتیپ اندازه‏گیری و میزان تفریخ تخم و خروج لارو از سیست در جمعیت‏ها ‏نیز در شرایط آزمایشگاهی مقایسه گردید. تجزیه خوشه‏ای ‏ژنوتیپ‏ها ‏بر اساس تعداد و اندازه ماده‏های تشکیل شده روی گیاهان نشان داد که ارقام Toucan، Cactus، Fernando، Sanetta و رقم مقاوم Pauletta در یک خوشه و ژنوتیپ‏های 035، 7112*SB36*SB30، 031، 034، 7112*SB36*SB28 و رقم حساس 7112*SB36*SB29 نیز در یک خوشه قرار گرفتند. نتایج نشان داد که در ژنوتیپ‏های مقاوم علاوه بر کاهش تعداد ماده‏های تشکیل شده روی ریشه، اندازه آن‏ها ‏نیز کوچک‏تر بود. نتایج مقایسه جمعیت‏ها ‏نشان داد که جمعیت آذربایجان غربی از نظر بیماری‏زایی تفاوت معنی‏دار‏ی با جمعیت‏ اصفهان داشته و ماده‏های کمتر و کوچک‏تر روی گیاهان آلوده به این جمعیت تشکیل داد. بیشترین تفریخ تخم و خروج لارو از سیست در جمعیت مشهد صورت گرفت.

    یکی از عوامل زردی، کوتولگی و پیچیدگی در حبوبات که باعث کاهش محصول می شود ویروس برگ قاشقی باقلا (Bean leaf roll virus, BLRV) می باشد. به منظور بررسی تنوع جدایه های ایرانی این ویروس از مزارع حبوبات در استان های مرکزی، فارس، خوزستان، کهگیلویه و بویر احمد، گلستان، زنجان و قزوین بازدید و از گیاهان یونجه، عدس، نخود، لوبیا، شبدر، باقلا، نخودفرنگی و شنبلیله نمونه برداری انجام شد. درمورد نمونه های انتخابی مراحل استخراج آر ان ای ویروس، تکثیر بخشی از ژن کد کننده ی پروتئین پوششی (از نوکلئوتید 3250 تا نوکلئوتید 3638) با آغازگر های اختصاصی، همسانه سازی و تعیین ترادف به عمل آمد. به این ترتیب، BLRV در گیاهان یونجه، عدس، نخود، لوبیا و شبدر عمدتا با علائم زردی و در گیاه باقلا با علائم قاشقی شدن برگ ها و زرد شدن برگ های جوان تشخیص داده شد. همردیف سازی چندگانه ترادف بخشی از ژن پروتئین پوششی 15 جدایه ی ایرانی با ترادف ناحیه مشابه سایر جدایه های BLRV موجود در بانک ژن نشان داد که میزان شباهت نوکلئوتیدی و ترجمه آمینواسیدی این ناحیه از ژن پروتئین پوششی به ترتیب 98–94% و 100–96% می باشد. مطالعات تبارزایی نشان داد که تمام جدایه های BLRV از ایران در یک گروه مجزا از جدایه های غیرایرانی قرار می گیرند. میزان شباهت ژن در این بخش از ژنوم می تواند نشان دهنده ی پایستگی و ثبات نسبی ژنتیکی ژن پروتئین پوششی ویروس مورد بررسی باشد.

    ویروس موزاییک ایرانی ذرت یک گونه منحصر به فرد در جنس نوکلئورابدوویروس می باشد. تنوع ژنتیکی جدایه های مختلف ویروس موزاییک ایرانی ذرت جمع آوری شده از مناطق جغرافیایی مختلف ایران و از میزبان های مختلف ویروس با علایم متفاوت در سال های 1394 و 1395 مورد بررسی و مطالعه قرار گرفت. توالی مربوط به ژن های نوکلئوکپسید، گلیکوپروتیین و یک قطعه با اندازه 500 جفت باز از ژن پلی مراز و نواحی غیر کد شونده بین ژن ها تعیین شد. تنوع ژنتیکی در نواحی ژن های نوکلئوکپسید و گلیکوپروتیین به میزان 3/1 در صد و درژن پلی مراز 8/0 درصد بود. تنوع ژنتیکی در نواحی غیرکد کننده بیشتر از نواحی ژنی بودکه در ناحیه بین ژن فسفوپروتیین و ژن 3 به بیش از 14% می رسید. در تجزیه و تحلیل های فیلوژنتیکی جدایه ها، ارتباطی بین گروه بندی فیلوژنتیکی و منطقه جغرافیایی، دامنه میزبانی و نوع علایم ویروسی دیده نشد. نسبت جانشینی های نامترادف به مترادف کمتر از یک بود که بیانگر وجود فشار انتخاب منفی بر روی ژن های مورد مطالعه بود. با توجه به این که ذرت یک گیاه وارداتی به ایران است و کشت آن در سال های اخیر توسعه یافته است به نظر می رسد ویروس موزاییک ایرانی ذرت از علف های هرز و یا میزبان های زراعی به ذرت منتقل شده است. احتمالا وابستگی شدید ویروس موزاییک ایرانی ذرت به زنجرک ناقل به عنوان تنها راه انتقال شناخته شده برای ویروس، می تواند دلیل پایین بودن تنوع ژنتیکی ویروس باشد. تنوع ژنتیکی پایین در ویروس موزاییک ایرانی ذرت، می تواند به عنوان ابزاری مفید، جهت تولید واریته هایی با مقاومت پایدار استفاده شود.

    Phytophthora parsiana به عنوان یک اامیست مقاوم به دمای بالا با دامنه ی میزبانی محدود به گیاهان چوبی، مورد توجه است. در مطالعه ی حاضر در یک آزمایش مقدماتی، دامنه میزبانی بیمارگر روی گونه های مختلف گیاهان علفی با استفاده از شش جدایه ی P. parsiana، شامل جدایه های تیپ این گونه از منابع مختلف، در شرایط گل خانه بررسی شد. دو جدایه ی قارچ، از میان گیاهان علفی شامل کدوییان، فلفل، حبوبات و گیاهان روغنی، تنها روی فلفل بیماریزا بودند. سپس به مننظور بررسی واکنش ارقام مختلف فلفل به بیمارگر، بیماریزایی 16 جدایه ی P. parsiana روی سه رقم فلفل بررسی شد. نتایج نشان داد که P. parsiana تنها روی فلفل قرمز و نه فلفل دلمه ای بیماریزا بود. این اولین گزارش از میزبان علفی برای P. parsianaاست. بین جدایه های P. parsianaبیماریزا روی فلفل قرمز و دو جدایه ی دیگر از این گونه که از پسته رفسنجان و یزد جداسازی شده بودند و روی فلفل قرمز بیمارگر نبودند، ارتباطات فیلوژنتیکی مورد بررسی قرار گرفت. بر اساس توالی سنجی ناحیه ی فاصله ی ترانویسی شده ی داخلی (ITS) ، جدایه ی بیمارگر روی فلفل و جدایه ی رفسنجان همراه با دیگر جدایه های P. parsiana در تبار 10 از درخت فیلوژنتیکی قرار گرفتند ولی جدایه ی یزد هیچ شباهتی با P. parsianaنداشت ودر تبار شش درخت فیلوژنتیکی با شباهت بسیار نزدیک (شباهت نوکلئوتیدی 100-99 درصد) همراه باPhytophthora taxon Walnutگروه بندی شد. این اولین گزارش ازPhytophthora taxon Walnut از پسته در ایران است.

    بررسی جنگل های دست کاشت کاج الدار Pinus eldarica Medw. (Pinaceae) در منطقه هزار پیچ گرگان (استان گلستان) در فروردین سال 1391 حاکی از آلودگی برگ های کاج الدار به قارچ مولد زنگ بود. بررسی نمونه ها مشخص نمود که عامل بیماری زنگ به گونه ای از جنس Coleosporium از تیره Coleosporiaceae تعلق داشت. گفتنی است فقط اعضا جنس Coleosporium قادر به ایجاد آلودگی و تولید مرحله اسیومی روی برگ های جنس Pinus در منطقه اروپا-آسیا می باشند. سایر جنس های زنگ گزارش شده روی کاج یعنی گونه های Melampsora و Cronartium سرشاخه ها، شاخه ها و حتی تنه کاج را آلوده می نمایند. طی بررسی های میکروسکوپی ویژگی های زیر روی نمونه های آلوده مشاهده گردید: اسپرموگونیوم ها روی هر دو سطح برگ و زیراپیدرمی بودند. اسیوم ها، شکوفا به شکل جوش های برآمده سفید رنگ از نوع Peridermium غالبا در سطح زیرین و بعضا روی سطح فوقانی برگ، دارای پریدیوم مشخص توسعه یافته زبانه ای شکل و آماس کرده بودند. اسیوم ها به طول 3-5/1 میلیمتر، پهنای غالبا 5/0 میلیمتر و ارتفاع تا 2/1 میلیمتر اندازه گیری شدند. اسیوسپورها به صورت زنجیری در اسیوم ها تشکیل شده بودند. اسیوسپورها بیضوی، بیضوی کشیده یا کم و بیش کروی، دارای دیواره پوشیده از زگیل های میله مانند (rod-like verrucae) ، ضخامت دیواره با احتساب زگیل ها 4-5/2 میکرومتر و ارتفاع زگیل ها تا 5/2 میکرومتر اندازه گیری شدند. بعضا تعدادی از زگیل ها در سطح اسپور به هم پیوسته و حالت شبه-مشبک روی دیواره اسپور مشاهده گردید. همچنین در برخی اسپورها کاهش ارتفاع زگیل ها به یک سمت اسپور قابل مشاهد بود

    جنسCampydoraCobb, 1920 تنها جنس متعلق به خانواده CampydoridaeThorne, 1935 و زیرراسته Campydorina و راسته Enoplia از نظر شکل سر و ساختار مری دارای حالتی منحصر به فرد در میان اعضا شاخهNematoda است و از این جنس منفرد فقط یک گونه Cobb, 1920Campydora demonstrans تاکنون شناسایی شده است. به استناد مطالعات انجام شده بر پایه توالی ناحیه 18SDNA ریبوزومی، این جنس دارای جد مشترک با جنس هایAlaimus ، Tripylaو Ironus می باشد (Mulin et al 2003). بر اساس گزارش اشتورهان (Sturhan 2010) این نماتد، در نمونه خاک های جمع آوری شده از قزوین، مشهد و خرم آباد به ترتیب با پوشش علفی، توت فرنگی و یونجه و مزارع گندم وجود دارد، ولی تاکنون به غیر از این گزارش ، از خصوصیات ریخت شناختی و ریخت سنجی این جمعیت ها اطلاعاتی ارائه نشده است.

    Pink rot, caused by Phytophthora erythroseptica, is an important soil-borne disease of potato causes significant losses in the field and storage. A selection of P. erythroseptica isolates from potato-growing regions in four continents including America, Asia, Europe and Australia was assessed using phylogenetic, morphological and physiological analyses. In all nuclear (internal transcribed spacers, ß-tubulin and heat shock protein 90) and mitochondrial (cytochrome c oxidase subunit I) trees as well as combined nuclear and mitochondrial tree, P. erythroseptica isolates formed a separate phylogenetic lineage containing the type isolate. Additionally, no morphological and physiological diversities were observed among the isolates. All isolates were homothallic and produced amphigynous terminal antheridia and globose aplerotic oogonia as well as non-papillate and ellipsoid to ovoid sporangia. The results showed that P. erythroseptica isolates belonged to different geographic regions are phylogenetically and morphologically similar. This uniformity could be due to the homothallic nature of P. erythroseptica or distribution of a single clone of pathogen by infected potato.

    The aim of this research was to compare the pathogenicity andsome biological and morphological characters of four populations of the sugar beet cyst nematode Heterodera schachtii, collected from infested farms of Isfahan, Shiraz, West Azerbaijan and Khorasan provinces. The responses of 12 sugar beet genotypes were evaluated in greenhouse based on the female number on the roots. Moreover, the sizes of females of the nematode populations on each genotype were determined, and egg hatching and emergence of the second stage juveniles (J2s) from the cysts were compared. The cluster analysis of genotypes based on the number and size of the females on the plants showed that the genotypes Toucan, Cactus, Fernando, Sanetta cultivars were clustered with the resistant cultivar Pauletta in one group, and the genotypes035,7112*SB36*SB30, 031, 034 and 7112*SB36*SB28 were clustered with the susceptible control (7112*SB36*SB29) in another group. Fewer and smaller females were produced on the resistant genotypes. Nematode population of West Azerbaijan showed a noticeable difference in pathogenicity with Isfahan population and produced fewer and smaller females. Mashhad population had the highest rate of egg hatching and juvenile emergence from cysts compared to the other populations.

    Legume diseases characterized by yellowing, stunting and leaf rolling cause heavy losses to legume crops. One of the causal agents of these diseases is Bean leaf roll virus (BLRV) which is widespread in legume species throughout Iran. In the present research, genetic variation of BLRV isolates from different plants and different geographical regions is investigated. Samples of Vicia faba, Pisum sativum, Cicer arietinum, Medicago sativa, Lens culinaris, Trifolium spp., Trigonella foenum-graecum and Phaseolus vulgaris showing symptoms of yellowing, dwarfing and leaf rollingwere collected from the fields of Markazi, Fars, Khuzestan, Kohgiluye-va-Boyerahmad, Golestan, Zanjanand Qazvin provinces in 2016-2017. Total RNA was extracted from symptomatic samples and subjected to RT-PCR using specific primers amplifying a fragment of 389 bp in size from the CP region of the virus which was cloned and sequenced. BLRV was detected in faba bean, alfalfa, lentil, chickpea, bean and clover. Comparison of obtained nucleotide (nt) and amino acid (aa) sequences of Iranian isolates of BLRV with corresponding nt and aa sequences of other BLRV isolates available in GenBank showed a 94-98% and a 96-100% nt and aa similarity, respectively. Phylogenetic analyses indicated that all Iranian BLRV isolates constituted a group district from the group formed by non-Iranian BLRV isolates. These data indicated that the CP region of this virus is relatively conserved among different isolates of BLRV.

    Maize Iranian mosaic virus (MIMV) is a distinct member of the genus Nucleorhabdovirus. The genetic diversity of maize Iranian mosaic virus (MIMV) isolates collected from different geographical regions of Iran, different hosts and different symptoms was determined during 2015-2016. Selection pressure analyses were also performed. Sequences of the nucleocapsid protein, glycoprotein, and a 500 bp fragment of polymerase genes and non-coding regions were determined. Genetic diversity in coding regions was less than 1.3 % in nucleocapsid protein and glycoprotein genes and 1 % in polymerase gene. The diversity in non-coding sequences was higher compared to ORFs especially between P and ORF 3 (up to 14.7 %). In phylogenetic analysis no correlation was found between MIMV isolates and geographical regions, type of symptoms and host differences. The ratio of non-synonymous to synonymous polymorphic sites indicated purifying selection has acted upon the analyzed genes. Likely, the dependence of MIMV on planthoppers for spreading can explain its low diversity. The information on the low genetic diversity of MIMV will be helpful in selection of maize cultivars with durable resistance

    The high temperature-tolerant oomycete Phytophthora parsiana was considered to be limited to woody plants. In the preliminary study, the host range of the pathogen on annual herbaceous plant species using six isolates of P. parsiana including type strains from different sources was examined under greenhouse conditions. Among herbaceous species including cucurbits, pepper, pulse and oil crops, two isolates were pathogenic to pepper. Then three cultivars of pepper were used to discriminate pepper cultivars to sixteen isolates of P. parsiana. Results showed P. parsiana were pathogenic to red pepper (‘Anheim’ and ‘Casabel’cultivars) but not bell pepper. This is the first report of an herbaceous host of P. parsiana (sensu lato). Phylogenetic relationships of the pathogenic isolates of P. parsiana from pistachio to pepper and two isolates of the species from pistachio from Rafsanjan and Yazd were not pathogenic to pepper were examined. Based on sequences of the internal transcribed spacer (ITS) region of the ribosomal RNA, isolates pathogenic to pepper and one from Rafsanjan were in a group of P. parsiana belonging to clade 10 but the Yazd isolate had no genetic similarity to P. parsiana, being placed in clade 6 of the ITS tree, closely matching that of Phytophthora taxon Walnut (99-100 nucleotide identity). This is the first report of Phytophthora taxon Walnut from pistachio in Iran.

    Eldar pine (Pinus eldarica Medw., Pinaceae) is among the most popular and commonly planted ornamental trees in Northern and Central Iran. Rust infected specimens of Eldar pine were collected from pine cultivated forest at Hezaar-Pich, Gorgan, Golestan province, in April 2012 and submitted to the Botany Department of Iranian Research Institute of Plant Protection, Tehran for diagnosis. The specimens showed heavy rust diseases symptoms including aecia on needles of Eldar pine. A description of the causal agent follows: Spermogonia subepidermal, on both sides of infected leaves. Aecia mostly hypophyllous, develop as conspicuous white columnar pustules on pine needles, peridermioid, with strongly developed bullate or tongue-shaped peridium, 1.5-3 x 0.5 mm and up to 1.2 mm high. Aeciospores catenulate, 25-40 x 17.5-26.5 µm, ellipsoid, oblong ellipsoid or more or less globoid, spore wall covered by rod-like verrucae (verrucose), wall including verrucae 2.5-4 µm thick, discrete rods up to 2.5 µm high, occasionally reduced on one side of spores or united in irregular or pseudoreticulate patterns (Fig. 1). Based on infection pattern and above mentioned morphological features of aecia and aeciospores, the rust fungus was identified as Coleosporium tussilaginis (Pers.) Lev. (Majewski 1977). The voucher specimen, 17001F, was deposited in IRAN, Iranian Research Institute of Plant Protection, Tehran. This is the first report of C. tussilaginis on a member of Pinaceae in Iran. Moreover, Pinus eldarica, is reported as a new host for the rust. Coleosporium tussilaginis is a heteromacrocyclic rust with telial stage on members of Asteraceae family including species of the genera Tussilago, Senecio, Petasites and Sonchus (Braun 1981).

    The genus Campydora Cobb, 1920, has a unique status among the phylum Nematoda based on head shape and the structure of esophagus. The genus has shown a common ancestor with the genera Alaimus, Tripyla, and Ironus under the order Enoplida as inferred from 18S ribosomal DNA sequence studies (Mullin et al., 2003). The family Campydoridae Thorne, 1935 contains only one genus and a single species without any additional species or genus. Sturhan (2010), found and reported Campydora demonstrans from Qazvin, Mashhad and Khorramabad regions in Iran from rhizosphere soils of herbaceous, tomatoes, alfalfa and wheat fields. He did not provide information on the morphological and morphometrical characteristics of the populations, so far. A population of this species was collected from the soil around Miyaneh city, East Azarbaijan province and the nematodes extracted using Jenkins (1964) and transferred to anhydrous glycerin by De Grisse (1969) methods. They were mounted on microscopic slides and the morphological and morphometrical characters studied using an Olympus BX 41 optical microscope and digital images prepared by a DP50 camera.
