    بیولوژی سینتتیک یک رشته دانشی نوین است که با خلق یک میکروارگانیسم مصنوعی توسط کریگ ونتر که دارای حداقل ژنوم کمینه سازگار با زیست بود، موجب برافروختن بحث های پیشین فلسفی پیرامون چیستی حیات گردیده است. همچنین پیشرفت های اخیر بیولوژی سینتتیک موجب بحث و گفتگوهای نوین فلسفی پیرامون موجودات هیبرید ”ارگانیسم های سینتتیک“ و ”ماشین های زنده“ و تفاوت بنیادین میان ماشین های بیولوژیک و جهان زنده شده است. این نوشتار، ضمن تجزیه و تحلیل نقادانه به چشم اندازهای فلسفی درباره طراحی ارگانیسم های سینتتیک، زیست کمینه و حیات مصنوعی، به چگونگی خلق دانش در بیولوژی سینتتیک بر پایه فراز ریچارد فاینمن ”آنچه که من نتوانم خلق کنم، درک نمی کنم« نیز می پردازد.

    زمینه
    ژن سرکوبگر تومور TP53، در ترمیم آسیب های وارد شده به DNA و همچنین در آپوپتوزیس سلولی نقش دارد. این پژوهش به بررسی ارتباط بین پلی مورفیسم کدون 72 ژن TP53 در انواع نمونه های بافتی غیرطبیعی سرویکس در مقایسه با نمونه های زنان سالم به عنوان گروه کنترل و همچنین فراوانی ویروس های هرپس سیمپلکس انسانی در این نمونه ها پرداخته است.
    مواد و روش ها
    در این مطالعه موردی- شاهدی، تعداد 110 نمونه بیوپسی سرویکس با پاتولوژی غیرنرمال از آرشیو بخش پاتولوژی بیمارستان شهدای خلیج فارس بوشهر مورد بررسی قرار گرفت و تعداد 164 نفر از زنان سالم به عنوان گروه کنترل انتخاب شدند. جهت شناسایی ویروس هرپس سیمپلکس از آزمون PCR یک مرحله ای و به منظور آنالیز پلی مورفیسم کدون 72 ژن TP53 از روشAllele-specific PCR استفاده گردید. داده ها با نرم افزار SPSS ویرایش 17 مورد تجزیه و تحلیل قرار گرفتند.
    یافته ها
    توزیع ژنوتیپ های TP53 در دو گروه بیمار و کنترل، از نظر آماری معنی دار نبود. در بررسی فراوانی دو آلل آرژنین و پرولین مشخص گردید که افراد فاقد آلل پرولین در گروه بیمار (19 درصد) نسبت به گروه کنترل (1/30 درصد) کمتر می باشند که این تفاوت از نظر آماری معنی دار است (04/0=p) (36/3-03/1: CI 95 درصد، 86/1=OR). همچنین ارتباط آماری معنی داری میان نتایج مختلف پاتولوژی، سن، قومیت و محل سکونت با ژنوتیب TP53 مشاهده نگردید. در این بررسی، ویروسHSV در نمونه های مورد مطالعه شناسایی نشد.
    نتیجه گیری
    به دلیل مشاهده اختلاف معنی دار آلل پرولین بین دو گروه بیمار و کنترل به نظر می رسد که این آلل با نتایج پاتولوژی غیرنرمال دهانه رحم در استان بوشهر ارتباط داشته باشد.

    زمینه
    سرطان معده چهارمین سرطان شایع و دومین عامل مرگ ومیر ناشی از سرطان در سراسر جهان محسوب می شود. نواحی شمالی ایران از شمال غرب تا شمال شرق جزو مناطق دارای ریسک بالای این سرطان هستند. جهش ژنتیکی و تغییر در سطح بیان یکی از مهم ترین فاکتورهای تاثیرگذار در سرطان زایی و پیشرفت سرطان است. از جمله ژن هایی که دچار تغییر در سطح بیان می شوند می توان به ژن CFL1 اشاره نمود. CFL1 ایزوفرم غیرماهیچه ای خانواده پروتئینی ADF/cofolin هست. پروتئین کدشونده توسط این ژن، پلیمریزه و دپلیمریزه کردن F اکتین و G اکتین به روش وابسته به PH را انجام می دهد. در این مطالعه بیان این ژن درآدنوکارسینومای معده از نوع روده ای مورد بررسی قرار گرفت.
    مواد و روش ها
    در این مطالعه RNA تام از 26 نمونه بیوپسی آدنوکارسینومای معده از نوع روده ای استخراج شده و پس از سنتز cDNA آزمایش Real time PCR برای اندازه گیری بیان ژن انجام شد. داده های حاصل توسط نرم افزار SPSS ویرایش 16، REST و Sigmaplot مورد آنالیز قرار گرفت.
    یافته ها
    میزان بیان ژن کوفیلین در نمونه های توموری نسبت به نمونه های سالم حاشیه تومور به میزان معنی دار افزایش پیدا کرده است (05/0>P). همچنین شاخص زیستی برای ژن CFL1 88/0 به دست آمد.
    نتیجه گیری
    این ژن با شاخص 88/0 می تواند به عنوان بیومارکر در سرطان معده استفاده شود. در مجموع یافته های این تحقیق نشان داد که میزان بیان CFL1 با ماهیت سرطانی تومورها ارتباط معناداری داشته است که این بدین معناست CFL1 احتمالا می تواند پتانسیل بالایی به عنوان هدف جدید برای ژن درمانی در سرطان معده داشته باشد.

    زمینه
    انسان در جوامع مدرن در معرض سطوح قابل ملاحظه ای از انتشار میدان های الکترومغناطیس (EMF) با فرکانس های مختلف قرار دارد. اثرات نوروبیولوژیکی میدان های الکترومغناطیس موضوع بحث و تحقیقات بسیار گسترده ای در طول چند دهه گذشته بوده است. بنابراین ما اثرات EMF را برروی یادگیری بینایی و تغییرات آناتومیکی نواحی هیپوکمپ و پره فرونتال (PFA) را در میمون های رزوس نر مورد مطالعه قرار دادیم.
    مواد و روش ها
    در این پژوهش، 4 میمون رزوس نر گونه ماکاکا مولاتا در معرض میدان های الکترومغناطیس با فرکانس های 5 و 30 هرتز با شدت 7/0 میکروتسلا به مدت 30 روز، روزانه 4 ساعت قرار داده شدند. تغییرات حافظه و یادگیری بینایی یک هفته قبل و یک هفته بعد از دوره پرتودهی با استفاده از باکس طراحی شده که حیوانات را برای به دست آوردن پاداش به چالش می کشد، مورد آزمایش قرار گرفت. همچنین تغییرات آناتومیکی مغز میمون ها با استفاده از تکنیک تصویربرداری تشدید مغناطیسی (MRI) یک هفته قبل و یک هفته بعد از پرتودهی اسکن شد. میمون ها با تزریق داخل عضلانی کتامین هیدروکلراید (20-10 میلی گرم بر کیلوگرم) و زایلوزین (4/0-2/0 میلی گرم بر کیلوگرم) بیهوش شده و با استفاده از دستگاه تصویربرداری در سه سطح کرونال، آگزیال و ساجیتال در فاز T2 و اسلایس های با ضخامت 3 میلی متر اسکن شدند. تغییرات آناتومیکی نواحی هیپوکمپ و پره فرونتال با تکنیک حجم سنجی مورد بررسی و مطالعه قرار گرفت.
    یافته ها
    قرارگیری در معرض میدان الکترومغناطیس با فرکانس 30 هرتز موجب کاهش تعداد پاسخ های صحیح در فرایند یادگیری و تاخیر در شکل گیری حافظه در دو میمون مورد آزمایش شد. در حالی که فرکانس 5 هرتز تاثیری بر تغییرات یادگیری و حافظه بینایی نداشت. هیچ تغییر آناتومیکی در ناحیه پره فرونتال و هیپوکمپ در هر دو فرکانس مشاهده نشد.
    نتیجه گیری
    میدان الکترومغناطیس با فرکانس 30 هرتز موجب اختلال در فرایند یادگیری و حافظه بینایی می شود، که احتمالا این تغییرات را از طریق تاثیر بر فاکتورهای دیگری به غیر از تغییر در ساختار و آناتومی مغزی اعمال می کند.

    زمینه
    افتراق بین تومورهای خوش خیم و بدخیم پاروتید قبل از انجام عمل جراحی احتمالی حائز اهمیت می باشد زیرا می تواند تصمیم گیری های طرح درمانی بیماران را با تغییر مواجه کند. در این مطالعه نقش احتمالی magnetic resonance spectroscopy بر روی افتراق تومورهای خوش خیم از بدخیم پاروتید مورد بررسی قرار گرفت.
    مواد و روش ها
    40 بیمار با توده غدد بزاقی به ویژه پاروتید و ساب مندیبولر که کاندید عمل جراحی بودند وارد مطالعه شدند. بیماران از درمانگاه بیمارستان امیراعلم انتخاب و در بیمارستان امام خمینی تحت MRI قرار گرفتند. در این مطالعه بر روی این بیماران magnetic resonance spectroscopy (MRS) در زمان اکوی 135 و 270 میلی پایه انجام شد و نسبت کولین به کراتین تعیین گردید. بر حسب نتیجه سیتولوژی بیماران به سه گروه تومور خوش خیم غیر وارتین، وارتین و بدخیم تقسیم شدند و از منحنیROC curve برای به دست آوردن بهترین نقطه برش (cutoff points) استفاده شد.
    یافته ها
    40 بیمار وارد این مطالعه شدند که 25 بیمار تومور خوش خیم غیر وارتین، 9 بیمار تومور بدخیم و 6 بیمار تومور وارتین داشتند. برای افتراق تومورهای خوش خیم از بدخیم (TE)Time of Echo در زمان 135 قدرت بیشتری از خود نشان داد، درTE:135 بهترین نقطه برش برای افتراق خوش خیمی و بدخیمی برابر با نسبت کولین به کراتین 92/1 بود که ویژگی 89 درصد و حساسیت 81 درصد داشت، در حالی که بهترین نقطه برش در 270:TE برابر با 76/1 بود که حساسیت 89 درصد و ویژگی 71 درصد نشان داد. بر طبق این امر نسبت کولین به کراتین کمتر از 25/1 موارد بدخیم را رد می کند (حساسیت 92 درصد) و نسبت بالاتر از 76/1 قویا به نفع بدخیمی می باشد (با ویژگی 100 درصد). MRS توانایی تفکیک بین تومور خوش خیم غیروارتین با گروه وارتین را نداشت.
    نتیجه گیری
    بر طبق این مطالعه نسبت کولین به کراتین در ام ار اسپکتروسکوپی می تواند به عنوان یک روش غیر تهاجمی در افتراق تومورهای خوش خیم و بدخیم کمک کننده باشد.

    زمینه
    مشاهده، دسته بندی و شمارش انواع مختلف گلبول های سفید در نمونه خون، یکی از گام های اساسی در درمان بیماری های مختلف است. هدف از انجام این پژوهش طراحی و پیاده سازی سیستمی سریع، قابل اعتماد و مبتنی بر پردازش تصاویر میکروسکوپی نمونه خون برای طبقه بندی چهار نوع از گلبول های سفید است.
    مواد و روش ها
    در این مقاله، از روش خوشه بندی k-means اصلاح شده برای انجام عمل بخش بندی تصویر استفاده شده است. علاوه بر این، عمل طبقه بندی گلبول های سفید با استفاده از یک شبکه عصبی کانولوشنی عمیق و با کمک داده های موجود در پایگاه داده MISP – پایگاه داده رایگان و متشکل از تصاویر میکروسکوپی نمونه خون – انجام شده است. همچنین، روش های مختلف رگولاریزاسیون مثل حذف تصادفی و افزایش تعداد تصاویر پایگاه داده، برای جلوگیری از بیش برازش (Overfitting) مدل پیشنهادی مورد استفاده قرار گرفته اند.
    یافته ها
    در بخش طبقه بندی، دقت شبکه عصبی برابر 99 درصد اندازه گیری شده است که نسبت به بسیاری از پژوهش های پیشین موفق تر بوده است. همچنین در بخش بخش بندی، شاخص اطلاعات متقابل برابر 73/0 حاصل شد.
    نتیجه گیری
    نتایج حاصل از این پژوهش نشان می دهد طراحی و پیاده سازی سیستمی سریع و قابل اعتماد با کمک پردازش تصاویر میکروسکوپی نمونه خون با استفاده از روش های مختلف پردازش تصویر و یادگیری ماشین امکان پذیر است.

    با توجه به افزایش شدید وزن افراد و عوارض آن در جوامع مختلف و نیز با پیشرفت تکنیک های جراحی چاقی، لازم است که مداخلات روانشناختی جهت کاهش عوارض بعد از عمل و نیز بالا بردن نتایج مثبت آن برای این افراد صورت پذیرد. بنابراین هدف از این پژوهش مطالعه اثربخشی مدل اطلاعات- انگیزش- مهارت های رفتاری بر بهبود اندازه وزن و محیط اندام ها در زنان تحت عمل جراحی چاقی بود.
    مواد و روش ها
    طرح پژوهش حاضر نیمه تجربی و از نوع پیش آزمون - پس آزمون با گروه آزمایش و کنترل بود. جامعه آماری شامل کلیه زنان چاقی بود که طی نیمه دوم سال 1395 در بیمارستان فوق تخصصی مادر و کودک غدیر شیراز عمل جراحی اسلیوگاسترکتومی را انجام دادند، از بین آنها 30 نفر به صورت در دسترس انتخاب شده و به صورت تصادفی در گروه آزمایش و گروه کنترل، گمارده شدند. از ابزارهای ترازوی خط کش دار، متر نواری و پرسشنامه اطلاعات جمعیت شناختی، در دو مرحله پیش آزمون و پس آزمون برای هر 2 گروه استفاده شد. برای گروه آزمایش، مدل اطلاعات- انگیزش- مهارت های رفتاری طی چهار هفته (هر هفته یک جلسه 5/1 ساعته)، در کلینیک چاقی بیمارستان اعمال گردید. داده ها با روش تحلیل کوواریانس چندمتغیره توسط نرم افزار آماری SPSS ویرایش 19 تجزیه و تحلیل شدند و سطح معنی داری 05/0P< برای نتایج معنی دار تلقی گردید.
    یافته ها
    نتایج تحلیل کوواریانس چندمتغیره نشان داد که مدل اطلاعات- انگیزش- مهارت های رفتاری بر اندازه وزن و محیط اندام ها در زنان تحت عمل جراحی چاقی تاثیر معنی داری گذاشته است (05/0P<).
    نتیجه گیری
    نتایج این پژوهش نشان داد که مدل اطلاعات- انگیزش- مهارت های رفتاری می تواند بر بهبود اندازه وزن و محیط اندام ها در زنان تحت عمل جراحی چاقی اثربخش باشد.

    Synthetic biology is a novel branch of biological sciences. The creation of a synthetic microorganisms with minimal genome compatible with life by Craig Venter had provoked the prior hot philosophical discussions about the nature of life. The recent advancements in synthetic biology brings new philosophical analysis about hybrid entities “synthetic organisms” and “living machines” and fundamental difference between biological machines and the living world. This paper is a critical analysis of the philosophical perspective on the design of synthetic organisms, minimal genome and artificial life. It also presents a critical view on knowledge-making practices in synthetic biology on Richard Feynman’s statement: “What I cannot create, I do not understand.”

    Background
    TP53 tumor suppressor gene play a role in repairing DNA damages and also in cell apoptosis. This research have paid to the association between codon 72 polymorphism of TP53 gene in a variety of abnormal cervical tissue samples compared to samples of healthy women as a control group and also frequency of human herpes simplex viruses (HSV) in these samples.
    Material and
    Methods
    In this case-control study, 110 biopsies of the cervix with abnormal pathology were examined from archives of the Department of Pathology at Shohadaye Khalij Fars Hospital, and 164 healthy women were selected as control group. PCR test was used for detection of HSV, and Allele-specific PCR was used for analysis of codon 72 polymorphism of TP53 gene. Data was analyzed with SPSS software.
    Results
    Distribution of codon 72 of TP53 genotypes in patient and control groups was not statistically significant. Evaluation of the frequency of arginine and proline alleles in patient and control groups revealed that those without proline allele were lower in patients (19%) compared to controls (30.1%), and this difference was statistically significant (p=0.04), (OR: 1.03-3.36¡ CI 95%=1.86). Also there were not significant correlation between the various results of pathology, age, ethnicity and place of residence with the TP53 genotype. In this study, HSV was not detected in samples.
    Conclusion
    Because of significant difference in proline allele between the case and control groups, it seems that this allele is associated with the abnormal cervical pathology results in Bushehr province.

    Background
    Gastric cancer is considered as the fourth most common cancer and the second most common cause of cancer-related death around the world. Northern areas of Iran from NorthWest to NorthEast have high risk of this cancer. Genetic mutations and changing in expression level are the most effective factors in carcinogenesis and cancer progression. CFL1 is one of the genes that is affected by changes in the level of expression. CFL1 is isoform of the ADF/cofilin protein familyThe protein encoded by this gene can polymerize and depolymerize F-actin and G-actin in a pH-dependent manner. In this research, we studied expression of this gene in intestinal samples of gastric adenocarcinoma.

    Methods
    In this study, mRNA in intestinal type of gastric adenocarcinoma of 26 biopsy samples was extracted, then after cDNA synthesis, real-time polymerase chain reaction was performed to measure gene expression.

    This gene can be used as a biomarker in gastric cancer. Overall, the results of this study showed that the expression of CFL1 has significant association with the nature of cancer tumors. This implies that CFL1 can have high potential as novel targets for gene therapies in gastric cancer.

    Background
    Humans in modern societies expose to substantially elevated levels of electromagnetic field (EMF) emissions with different frequencies.The neurobiological effects of EMF have been the subject of debate and intensive research over the past few decades. Therefore, we evaluated the effects of EMF on visual learning and anatomical dimensions of the hippocampus and the prefrontal area (PFA) in male Rhesus monkeys.
    Materials And Methods
    In this study, four rhesus monkeys were irradiated by 0.7 microtesla ELF-EMF either at 5 or 30 Hz, 4 h a day, for 30 days. Alterations in visual learning and memory were assessed before and after irradiation phase by using a box designed that cchallenging animals for gaining rewards Also, the monkeys’ brains were scanned by using MRI technique one week before and one week after irradiation. The monkeys were anesthetized by intramuscular injection of ketamine hydrochloride (10–20 mg/kg) and xylazine (0.2–0.4 mg/kg), and scanned with a 3-Tesla Magnetom, in axial, sagittal, and coronal planes using T2 weight­ed protocol with a slice thickness of 3 mm. The anatomical changes of hippocampus and the prefrontal area (PFA) was measured by volumetric study.
    Results
    Electromagnetic field exposure at a frequency of 30 Hz reduced the number of correct responses in the learning process and delayed memory formation in the two tested monkeys. While, ELF-EMF at 5 Hz had no effect on the visual learning and memory changes. No anatomical changes were found in the prefrontal area and the hippocampus at both frequencies.
    Conclusion
    ELF-EMF irradiation at 30 Hz adversely affected visual learning and memory, pprobably through these changes apply through effects on other factors except changes in brain structure and anatomy.

    Background
    Differentiation between benign and malignant parotid tumors before surgery is important because it can change the therapeutic planning for patients. In this study we evaluated the role of Magnetic resonance spectroscopy (MRS) in differentiation of benign from malignant parotid tumors.
    Materials And Methods
    40 patients entered with salivary gland tumor especially parotid) that candidated for surgery in this study. Patients selected in Amiralam hospital and did MRI in Imam Khomeini hospital. In this study, on these patients Magnetic resonance spectroscopy (MRS) was performed at 135 and 270 Time of Echo, and the choline/creatine ratio was determined. Regarding to the cytological results, patient divided to warthin, benign and malignant tumors and ROC curves used to determine the best cut off points.
    Results
    40 patients entered in this study, 25 had benign non warthin tumors, 9 malignant tumors and 6 warthin tumors. In differentiation benign from malignant tumors TE136 was stronger. The best cut off point of Choline to Creatine ratio in TE: 136 was 1.92 with sensitivity 81 % and specificity 89% and in TE 270 was 1.76 with Sensitivity 89% and specificity 71%. Choline/creatine ratio 1.76 strongly suggest malignancy (with10% specificity). MRS was not able to differentiate benign non warthin from warthin tumors.
    Conclusion
    According to this study, Choline/ creatine ratio in MRSpectroscopy can be helpful as noninvasive method in differentiation of benign from malignant tumors.

    Background
    Observation, categorize and count various types of white blood cells in a blood sample is a One of the most important steps in the treatment of various diseases. The aim of this study was to design and implement a fast and reliable and based on the processing of microscopic images of blood samples for the classification of four types of white blood cells.
    Materials And Methods
    In this article, the modified k-means clustering method is used to perform image segmentation. Furthermore, The classification of white blood cells was done using a deep convolutional neural network and with the help of data in the MISP database, a free database composed of microscopic blood sample images. Moreover, Several regularization techniques such as dropout and image augmentation were applied to prevent the network from overfitting.
    Results
    In the classification category, the accuracy of the neural network is measured to be 99%, which has been more successful than many earlier studies. In the segmentation section, the cross-reference index was 0.73.
    Conclusion
    The results of this research show that rapid and reliable system design and implementation is possible by processing the microscopic images of the blood sample using different methods of image processing and machine learning.

    Background
    Considering the high increase in individual's weight and its complications in different societies, as well as the progression of bariatric surgery techniques, psychological interventions are needed to reduce postoperative complications and to increase their positive outcomes for these patients. Therefore, the purpose of this research was to study the effectiveness of Information-Motivation-Behavioral (IMB) model on improving weight and body size in women undergoing bariatric surgery.
    Materials And Methods
    The design of present semi- experimental study was pre-test, post-test with experimental and control groups. The statistical population included all obese women that undergone laparoscopic sleeve gastrectomy surgery in Shiraz Ghadir Mother & Child Multi Super Specialty Hospital, during second half of 2016. The 30 of them were selected by using available sampling method and they randomly were assigned to 2 groups, experimental (n=15) and control (n=15). The Balance with ruler, Tape measure& demographic information questionnaire was used to collect data for each 2 groups in two steps (pre-test and post-test).
