    ویروس موزائیک معمولی لوبیا (BCMV Bean common mosaic virus،) از مهم ترین عوامل بیماریزای لوبیا در سراسر جهان می باشد. این تحقیق به منظور ردیابی این ویروس از مزارع لوبیای استان زنجان انجام شد. برای این منظور، تعداد 130 برگ از نمونه های لوبیا با علایم موزائیک، رگبرگ نواری، بد شکلی و قاشقی شدن از مزارع استان زنجان در سال 1393 جمع آوری و توسط آزمون سرولوژیکی الایزا (DAS- ELISA) با استفاده از آنتی بادی چند همسانه ای، آلودگی آنها به ویروس تایید شد. جهت خالص سازی بیولوژیک، ویروس روی گیاه محک و سپس روی لوبیای رقم اختر مایه زنی و تکثیر شد. سپس با استفاده از آغازگرهای اختصاصی طراحی شده براساس توالی پروتئین پوششی، قسمتی از ژنوم ویروس تکثیر و توسط شرکت ماکروژن توالی یابی گردید. براساس نتایج آزمون الایزا بیش ترین میزان آلودگی نمونه های جمع آوری شده مربوط به نمونه های مزارع دانشگاه زنجان و کمترین میزان آلودگی در مزارع سوکهریزی و نعلبندان مشاهده شد. نتایج مقایسه توالی بخشی از پروتئین پوششی با توالی سایر ویروس ها نشان داد که جدایه غالب این ویروس به جدایه ای از آفریقای جنوبی با رس شمار AF361337 شباهت بالایی (99%) دارد. همچنین ترسیم درخت فیلوژنتیک مربوط به توالی تعیین شده نشان داد، این جدایه ویروسی با سایر جدایه های گزارش شده از ایران در یک خوشه قرار می گیرند که بیانگر تنوع پایین این ویروس در یک محدوده خاص جغرافیایی است.

    هدف از این مطالعه بررسی نشانگرهای چند شکلی تک نوکلئوتیدی (SNP) مشترک موثر بر صفات وزن بدن در دو و چهار هفتگی، وزن سینه و لاشه (کشتار شده در چهار هفتگی) بود. بدین منظور از داده های فنوتیپی 105 قطعه بلدرچین و 12798 نشانگر SNP استفاده شد. همبستگی فنوتیپی بین صفات مورد نظر با استفاده از مدل چهار صفتی برآورد شد به گونه ای که اثرات جنس و هچ به عنوان اثرات ثابت در نظر گرفته شدند. هم چنین روش تابعیت تک نشانگری برای برآورد اثر هر نشانگر برای صفات مورد نظر به کار گرفته شد. نتایج نشان داد که اثر جنس و هچ بر چهار صفت مورد مطالعه معنی دار بود (p < 0.0001). بیش ترین همبستگی بین وزن چهار هفتگی و وزن لاشه برآورد گردید (96/0). تعداد SNPهای مشترک موثر بر چهار صفت مذکور 3 و 36 عدد به ترتیب در سطوح معنی داری 1 و 5 درصد به دست آمدند. با توجه به همبستگی بالای بین صفات مورد بررسی، احتمالا SNPهای موثر مشترک در بین چهار صفت بر روی ژن های کنترل کننده این صفات و یا در نزدیکی آن ها قرار گرفته҅اند.

    کنترل زمان گل دهی یکی از مهم ترین اجزای اثر متقابل بین گیاهان و محیط رشد آن ها می باشد که نه تنها برای میزان محصول تولیدی بلکه برای کیفیت دانه برنج نیز عامل مهمی به حساب می آید. در این تحقیق مطالعات فنوتیپی و مولکولی بر روی 45 رقم برنج محلی و اصلاح شده انجام شد. ابتدا چندشکلی ژن های Ehd1 و Ehd3 در بین ارقام و سپس ارتباط این دو ژن با زمان خوشه دهی مورد بررسی قرار گرفت. نتایج مطالعات فنوتیپی حاکی از وجود تنوع بیشتر در ارقام محلی نسبت به ارقام اصلاح شده بود. ارقام محلی به طور متوسط 8 روز زودرس تر از ارقام اصلاح شده بودند و تفاوت زمان خوشه دهی آن ها معنی دار بود. آغازگرهای اختصاصی طراحی شده براساس هم ردیف سازی چندگانه، توانست چندشکلی مورد انتظار را بین ارقام مختلف آشکار سازد. تجزیه ارتباط در جمعیت مورد مطالعه نشان داد که ژن Ehd1 نسبت به ژن Ehd3 تاثیر بیشتری بر زمان خوشه دهی داشت (1/12 در مقابل 4/5 درصد) و اثر افزایشی آن ها به ترتیب 4/3 و 2/2 روز برآورد شد. با توجه به کمی بودن صفت مورد مطالعه و اثر کم دو ژن فوق بر زمان خوشه دهی، می توان نتیجه گیری نمود که به نژادگر هنگام اصلاح ارقام زودرس باید به نقش سایر ژن ها و اثرات متقابل آن ها نیز توجه نماید.

    شوری یکی از مهم ترین تنش های غیر زیستی است که تولید محصولات زراعی را محدود می نماید. به منظور بررسی تاثیر سطوح مختلف شوری (صفر، 2، 4، 6 و 8 دسی زیمنس بر متر) بر صفات رشدی گیاهچه های آفتابگردان و تجزیه ژنتیکی این صفات در رابطه با تحمل به شوری، آزمایشی به صورت فاکتوریل بر پایه طرح بلوک های کامل تصادفی انجام گرفت. در این مطالعه، طول ریشه چه و ساقه چه، وزن تر ریشه چه و ساقه چه و وزن خشک ریشه چه و ساقه چه در گیاهچه ها ارزیابی شدند. نتایج نشان داد که بین لاین ها در سطوح مختلف شوری، از نظر تمامی صفات مورد مطالعه تفاوت معنی دار (P≤0.05) وجود دارد. به منظور تجزیه ژنتیکی صفات گیاه چه در رابطه با تحمل به شوری در گیاهچه های آفتابگردان، از نقشه پیوستگیSSR/SNP با 221 نشانگر و متوسط فاصله 44/7 سانتی مورگان بین نشانگرها استفاده گردید. نتایج مکان یابی ژنی نشان داد که در سطوح شوری صفر، 2، 4، 6 دسی زیمنس بر متر به ترتیب 18، 13، 4 و 10 QTL در کنترل صفات رشدی آفتابگردان نقش دارند. در این مطالعه، در سطح شوری 8 دسی زیمنس بر متر به دلیل عدم جوانه زنی گیاهچه ها، هیچ QTLای یافت نشد. مهم ترین QTL های شناسایی شده با بالاترین مقدار درصد تغییرات فنوتیپی توجیه شده در سطوح شوری صفر، 2، 4، 6 دسی زیمنس بر متر به ترتیب مربوط به صفات وزن تر ریشه چه (72/61%)، طول ریشه چه (34/59% و 37%) و وزن خشک ساقه چه (39/21%) بود. نتایج نشان داد که QTL های هم مکان برای صفات مختلف در چندین گروه پیوستگی پراکنده هستند که وجود چنین حالتی می تواند موجب افزایش کارآیی انتخاب به کمک نشانگر و پیش برد برنامه های به نژادی گیاهی شود.

    خانواده bZIP یکی از گسترده ترین عوامل رونویسی در گیاهان است که در پاسخ به تنش های زیستی و غیرزیستی نقش دارد. تنظیم بیان ژن در مرحله ترجمه بر اساس خصوصیات مولکول mRNA تعیین می شود که یکی از مهم ترین آن ها نواحی غیرترجمه شونده (UTR) هستند. در این مطالعه ویژگی های نواحی غیرترجمه شونده خانواده bZIP در گیاه جو (شامل 78 عضو) و اثر آن ها بر رونویسی و ترجمه، بر اساس روش های بیوانفورماتیکی مورد بررسی قرار گرفت. بر اساس نتایج، میانگین طول ناحیه 5''-UTR کمتر و محتوای GC آن بیشتر از ناحیه 3''-UTR بود. زمینه کدون آغاز برای موقعیت 4+ در 23/49% از اعضا توسط گوانین و موقعیت 3- در 70/56% توسط نوکلئوتیدهای پورینی اشغال شده بود. در مجموع 18 نوع عناصر فعال سیس موثر در تنش های غیرزیستی به دست آمد که عوامل رونویسی تنظیم شونده توسط اسید جیبرلیک بیش ترین فراوانی را داشتند. ریبوزوم برای رسیدن به کدون آغاز 50 عضو باید انرژی بیش از Kcal/mol 50 صرف کند، اگرچه 23 عضو دارای نواحی داخلی برای ورود ریبوزوم (IRES) بودند. یک توالی هدف sRNA در توالی های 5''-UTR (HvbZIP4) و هم چنین 3''-UTR (HvbZIP44) مشاهده شد. 225 عدد توالی چهارچوب باز خواندنی (با حداقل طول 12 نوکلئوتید) در بالادست ناحیه رمزکننده اصلی مشاهده شد. شباهت قابل توجهی بین توالی چهارچوب باز خواندنی بالادست (چهارچوب 3+) در HvbZIP24 و یک پروتئین بالادست در Theobroma cacao (XM_007017561.1) مشاهده شد. به طور کلی، مولکول های mRNA اعضا این خانواده دارای ساختارهای متفاوت تنظیمی بودند، این امر راه حلی طبیعی برای کنترل دقیق میزان تولید این نوع پروتئین های تنظیمی است.

    باکتری Ralstonia solanacearum قادر است بالغ DNA بر 30 خانواده مختلف گیاهی اعم از گیاهان زراعی و وحشی شامل سیب زمینی، توتون، گوجه فرنگی، بادنجان و بادام زمینی را آلوده نماید. یکی از بیماری های مهم سیب زمینی پژمردگی باکتریایی می باشد که در اثر باکتری Ralstonia solanacearum به وجود می آید. با استفاده از محیط کشت TTC از ساقه های پژمرده ارقام مختلف سیب زمینی جمع آوری شده از مناطق عمده سیب زمینی کاری استان کرمان شامل جیرفت، ساردوئیه، لاله زار و صوغان تعدادی استرین از باکتری Ralstonia solanacearum جداسازی، سپس این استرین ها با استفاده از آزمون های بیوشیمیای تعیین بیووار و بیماری زایی آنها بر روی گوجه فرنگی رقم Red Cloud ثابت شد. پس از آن با استفاده از آغازگرهای BOX، ERIC و REP تنوع ژنتیکی استرین ها با روش واکنش های زنجیره ای پلی مراز بررسی گردید. براساس درخت فیلوژنتیکی ترسیم شده با نرم افزار NTSYSpc2.0، استرین ها در دو گروه ژنتیکی قرار گرفتند که در بین استرین های هر گروه نیز تنوع مشاهده شد. به احتمال زیاد وجود تنوع ژنتیکی باکتری R. solanacearum در مناطق سیب زمینی کاری استان کرمان ناشی از ورود غده های بذری آلوده از سایر منابع آلوده به این استان است.

    هدف از این مطالعه ارزیابی تاثیر رقم، ساکارز و هورمون بر ریزغده زایی سیب زمینی در شرایط کشت درون شیشه ای بود. بدین منظور آزمایشی به صورت فاکتوریل در قالب طرح کاملا تصادفی با سه تکرار بر روی سه رقم سیب زمینی به نام های آگریا، مارفونا و ساتینا انجام شد. در این تحقیق، چهار سطح ساکارز (40، 60، 80 و 100 گرم در لیتر) در ترکیب با دو محیط فاقد هورمون و استفاده از هورمونBAP و NAA (غلظت10 و 5/0 میلی گرم در لیتر) بر روی ریز نمونه های ارقام مذکور مورد مطالعه قرار گرفت. دوازده هفته پس از شروع کشت تعداد، وزن، قطر و تعداد چشم ریزغده ها اندازه گیری گردید. نتایج تجزیه واریانس اختلاف معنی داری بین ارقام، سطوح ساکارز و هورمون برای همه صفات مورد مطالعه نشان داد. در این آزمایش، رقم ساتینا بیش ترین مقادیر را برای تمامی صفات نشان داد درحالی که رقم مارفونا برای کلیه صفات به استثنای تعداد ریزغده که نسبت به رقم آگریا برتری داشت، کم ترین مقادیر را دارا بود. در بین سطوح مختلف ساکارز بهترین پاسخ از کاربرد 80 گرم در لیتر به دست آمد و مقادیر 60، 40 و 100 گرم به ترتیب در مراتب بعدی قرار داشتند. اگر چه غده زایی در غیاب هورمون انجام شد اما افزودن هورمون به محیط کشت موجب ارتقاء عملکرد غده زایی گردید. به طورکلی نتایج نشان داد که بیش ترین تعداد، وزن و قطر ریزغده و تعداد چشم در رقم ساتینا، غلظت 80 گرم بر لیتر ساکارز در شرایط استفاده از 10 میلی گرم در لیتر BAP و 5/0 میلی گرم در لیتر NAA حاصل شد.

    تنش شوری، منجر به تنش اکسیداتیو و افزایش رادیکال های آزاد اکسیژن می شود که برای سلول سمی می باشد. گیاهان از چند سیستم آنتی اکسیدانتی برای محافظت از سلول های خود علیه این سموم استفاده می کنند. در این مطالعه اثر تنش بلندمدت شوری بر فعالیت آنزیم های آنتی اکسیدانی پراکسیداز و آسکوربات پراکسیداز، کلروفیل، نشت غشائی و روابط آب برگ در گیاه هالوفیت چمن شور (Aeluropus littoralis) مورد مطالعه قرار گرفت. بدین منظور ریشه و برگ گیاهان تحت 60 روز تنش شوری با غلظت های صفر، 250، 450 و 650 میلی مولار NaCl قرار گرفتند. نتایج نشان دادند که فعالیت آنزیم های پراکسیداز و آسکوربات پراکسیدازها در برگ تمام تیمارها افزایش یافته است. فعالیت آنزیم پراکسیداز در تنش 450 و 650 میلی مولار و آنزیم آسکوربات پراکسیداز در 250 و 450 میلی مولار در ریشه افزایش داشت. در 650 میلی مولار میزان نشت غشایی افزایش و میزان کلروفیل و محتوای آب نسبی (RWC) کاهش پیدا کرد.

    وجود تنوع ژنتیکی پایه و اساس اصلاح گیاهان است که گزینش گیاهان با خصوصیات مطلوب و یا انتقال صفات به گیاهان را ممکن می سازد. مطالعه ی تنوع ژنتیکی بر پایه نشانگرها و از جمله نشانگرهای پروتئینی در برنامه های به نژادی و به ویژه انتخاب والدین نقش مهمی دارد. در تحقیق حاضر، تنوع ژنتیکی 16 رقم گندم نان (سرخ تخم، شهریار، توس، الموت، زرین، آرام، گاسپارد، بزوستایا، پیشگام، سایسون، گاسکوژن، میهن، امید، نوید، بک کراس روشن زمستانه و زارع) به کمک روش الکتروفورز SDS-PAGE بررسی شد. غلظت پروتئین توسط روش برادفورد مشخص گردید. پس از رنگ آمیزی ژل پلی اکریل آمید با کوماسی بلو R-250 و امتیازدهی باندها طبق روش صفر (عدم وجود باند) و یک (وجود باند)، تجزیه خوشه ایبر مبنای ضریب تشابه جاکارد به روش UPGMA انجام گردید. بر این اساس ارقام در چهار گروه مجزا قرار گرفتند. بر این اساس ژنوتیپ سرخ تخم در گروه دوم، ژنوتیپ گاسپارد در گروه سوم و سایر ژنوتیپ ها در گروه اول خوشه بندی شدند. نتایج نشان داد که ژنوتیپ بک کراس روشن زمستانه با ژنوتیپ گاسپارد بیش ترین تفاوت فاصله ای را براساس آلل های پروتئینی مورد مطالعه، دارا هستند. بنابراین، با توجه به فواصل ژنوتیپ ها نسبت به هم براساس ضریب تشابه، در برنامه های بیومتری اصلاحی، تلاقی ژنوتیپ های امید، سایسون، بک کراس روشن زمستانه با ژنوتیپ گاسپاردقابل توجیه می باشد.

    در این پژوهش وقوع ویروس موزائیک چغندرقند (BtMV) در مزارع چغندرقند استآن های خراسان رضوی، خراسان شمالی و البرز بررسی شد و به منظور تعیین جایگاه تاکسونومیکی جدایه های ایرانی BtMV و بررسی رابطه فیلوژنتیکی آن ها با سایر جدایه های گزارش شده از دنیا، بخشی از ژنوم ویروس شامل منطقه ژنی رمزکننده پروتئین پوششی و انتهای NIb تعیین ترادف شد. به این منظور، استخراج RNA و واکنش RT-PCR با آغازگرهای اختصاصی روی نمونه های الایزا مثبت انجام و محصول پی سی آر در پلاسمید PTZ57R/T همسانه سازی و تعیین ترادف شد. منطقه ژنی رمزکننده پروتئین پوششی و بخشی از انتهای NIb در این توالی ها به همراه سایر توالی های موجود در GenBank هم ردیف سازی شدند. سپس ماتریس درصد برابری و مولفه های تکاملی به دست آمد. ترسیم درخت فیلوژنتیکی و ردیابی وقوع نوترکیبی نیز انجام شد. براساس درخت فیلوژنتیکی رسم شده، جدایه های BtMV به دو گروه تقسیم شدند؛ جدایه های چین، اسلواکی و استرالیا (Euroasia) در یک گروه و جدایه امریکا در گروهی دیگر قرار گرفتند. نتایج نشان داد که جدایه های ایران در دسته مجزایی درگروه Euroasia قرار گرفتند. این اولین گزارش از ترادف بخشی از ژنوم ویروس موزائیک چغندرقند از ایران می باشد.

    کلزا که به منظور استحصال روغن آن تولید می گردد، همانند سایر گیاهان زراعی متاثر از شوری می شود. شوری خاک به عنوان یکی از مهمترین عوامل نامساعد و تنش زای غیرزنده اثر نامطلوب بر عملکرد و کیفیت محصولات کشاورزی دارد. باکتریPseudomonasfluorescens جزو باکتری های محرک رشد گیاه می باشد و در همیاری با گیاهان باعث افزایش رشد مقاومت گیاه در برابر عوامل بیماری زا و محدود کننده محیطی می گردد. در این تحقیق شوری در دو سطح صفر و 300 میلی مولار نمک NaCl اعمال شد. حضور یا عدم حضور باکتری به عنوان تیمارهای دیگر با سه تکرار اعمال گردید و برای تجزیه های آماری از طرح کرت های خرد شده با طرح پایه ی کاملا تصادفی استفاده شد. به منظور بررسی پروتئین های دخیل در بهبود مقاومت گیاه کلزا به شوری در همیاری با باکتری Pseudomonas fluorescens الگوی الکتروفورز دو بعدی پروتئوم ریشه آن مورد بررسی قرار گرفت. نتایج حاصل نشان داد که از 216 لکه ی ظاهر شده 16 لکه ی پروتئینی در اثر اعمال تنش شوری، 13 لکه ی پروتئینی در حضور باکتری محرک رشد گیاه و 23 لکه در صورت اعمال هر دو تیمار تغییر بیان نشان می دهند. پس از شناسایی احتمالی پروتئین های مربوطه با استفاده از معیارهای وزن مولکولی و نقطه ی ایزوالکتریک مشخص شد این پروتئین ها که با افزایش یا کاهش بیان همراه بوده اند جزو پروتئین های دخیل در مسیرهای متابولیکی/ انرژی زایی، پیام رسانی، حفاظت و دفاع سلولی، کانالی، ساختار و مکان یابی پروتئین های دیگر، ساختمان سلولی و رونویسی و ترجمه هستند. با اعمال تیمار شوری یا باکتری یا شوری به همراه باکتری، بیشترین تعداد لکه های تغییر بیان یافته مربوط به گروه پروتئین های متابولیکی/ انرژی زایی بود.

    پروتئین های ذخیره ای آندوسپرم از مهم ترین عوامل موثر بر کیفیت دانه گندم می باشند. زیرواحدهای گلوتنین با وزن مولکولی بالا (HMW-GS) نقش کلیدی را در شکل و ساختار گلوتن بازی می کنند و ارتباط تنگاتنگی با کیفیت گندم دارند. جهت بررسی تنوع آللی در مکان های ژنی کدکننده زیرواحدهای گلوتنین با وزن مولکولی بالا در 25 رقم از گندم های نان ایرانی و استرالیایی، نه جفت آغازگر آلل اختصاصی برای مکان های ژنی (Glu-A1،Glu-B1وGlu-D1) به کار گرفته شدند. نتایج نشان داد که برای مکان ژنی Glu-A1، سه آلل (AxNull، Ax1 و Ax2*) شناسایی گردیدند که در بین آن ها بیش ترین فراوانی مربوط به آلل Ax2* با فراوانی 56 درصد بود. برای مکان ژنی Glu-B1 در مجموع شش آلل شناسایی شدند، که ترکیب آللی Bx7+By8با فراوانی 32 درصد بیش ترین فراوانی را داشت. برای مکان ژنی Glu-D1، در مجموع دو آلل شناسایی شد که ترکیب آللی Dx2+Dy12 با فراوانی 82 درصد بیش ترین فراوانی را در بین آلل های شناسایی شده به خود اختصاص داد. در نهایت رتبه بندی ژنوتیپ ها براساس سیستم امتیازدهی پی فلاگر به منظور پیش بینی کیفیت پخت نان نشان داد که اکثر ژنوتیپ های مورد بررسی رتبه 8، 9 و 10 را داشتند که نشان دهنده ی کیفیت پخت نان خوب تا عالی آن ها است. نتایج تجزیه خوشه ایبراساس زیرواحدهای مختلف نیز این ارقام با کیفیت بالا را در یک گروه مجزا قرار داد. از تنوع شناسایی شده در این پژوهش می توان به عنوان منبع با ارزش تنوع آللی در برنامه های اصلاحی گزینش به کمک نشانگر به منظور بهبود کیفیت محصولات نهایی حاصل از گندم بهره برد.

    Bean common mosaic virus (BCMV) is one of the most important diseases that reduce the yield of common bean worldwide. To detect and identify of BCMV from common bean fields in Zanjan province, 130 samples were collected that showing mosaic, vein stripe, chlorotic spot and leaf deformation symptoms in 2014. BCMV infection in these samples was assayed by DAS-ELISA using polyclonal antibody. Indicator plant, Chenopodium quinoa, was used to purify virus biologically and then pathogenicity test was carried out on susceptible bean variety, Akhtar. A fragment of viral genome was amplified by polymerase chain reaction using specific primers designed for a part of the coat protein gene and then sequenced by Macrogen Company. The results of ELISA test showed the highest disease incidence in collected samples from Zanjan University fields and lowest number was observed in Sookakrizi and Nalbandan. Moreover, the comparison of the obtained sequence with other viruses in GenBank database showed the highest similarity (99%) with a BCMV isolate with Genbank accession number AF361337 from South Africa. A phylogenetic tree was also constructed by MEGA6 software. This isolate was grouped in the same clade as other isolates from Iran which indicates a low genetic variation for this virus in an especial geographical region.

    The objective of this study was to investigate common effective single nucleotide polymorphism (SNP) on two and four week body weight, breast and carcass weights traits (slaughtered at 28 days of age) in Japanese quail. For this purpose, phenotypic data of 105 birds and 12798 SNPs were used. Phenotypic correlation among the interested traits were estimated using a multiple (four) trait model, such that the effects of sex and hatch were considered as a fixed effects.     Flowering time control is one of the most important components of interactions between plants and their growth environment that is a crucial factor not only for yield rate but also for rice grain quality. In this research phenotypic and molecular studies were conducted in 45 local and improved rice cultivars. Firstly polymorphism of Ehd1 and Ehd3 genes between the cultivars was studied and then the association of the two genes with heading date was investigated. Results of phenotypic studies indicated the existence of higher variation of heading date in local cultivars than that of improved cultivars. Local cultivars had on average 8 days earliness relative to improved ones, with being significant difference between their heading dates. Specific primers designed based on multiple alignments could detect polymorphism between different cultivars. Association studies showed that pooled population Ehd1 had a higher association with heading date than Ehd3 (12.1 vs. 5.4%) and their additive effects were estimated, respectively, 3.4 and 2.2 days. Regarding to quantitative nature of the studied trait and minor effects of the two genes on heading date, it can be concluded that when improving early-mature cultivars utilizing the two genes breeders might consider the roll of other genes and their interactions.

    Salinity is one of the important abiotic stresses, which limit the crop production. In order to study the effect of several salinity levels (0, 2, 4, 6 and 8 (dS/m)), exerted by NaCl, on some growth traits of sunflower line and genetic analysis of several traits related to salinity tolerance, A factorial experiment carried out based on RCBD. In this study, some growth traits incuding shoot length, root length, shoot fresh weight, root fresh weight, shoot dry weight and root dry weight were evaluated in seedings. Results revealed statistical significant difference among RILs in several salinity levels. To genetic analysis of tolerance to salinity in sunflower seedlings, linkage map of SSR/SNP with 221 markers and average distance of 7.44 CM between markers used. Results of gene mapping manifested 18, 13, 4 and 10 QTLs in salinity levels of 0, 2, 4 and 6 (dS/m) for studied characteristics. In the salinity level of 8 (dS/m) due to lack of germination of seedlings, no QTL was found. The most significant QTLs with high explained phenotypic varition in salinity levels of zero, 2, 4, 6 dS/m were identified for root fresh weight (61.72%), root length (59.34 and 37%) and shoot dry weight (21.39%), respectively. Results showed some co-localized QTLs for several characteristics, which dispersed in some linkage groups. The co-localized QTLs could lead to increase the efficiency of marker-assisted selection in plant breeding programs.

    bZIP family is one of the most diverse transcription factors in plants playing critical roles in response to biotic and abiotic stresses. Regulation of gene expression at translation level is determined based on the mRNA features which untranslated regions (UTRs) are among the most important ones. In the present study, we attempted to survey the features of UTRs and their probable effect on transcription and translation of bZIP family in barley (78 members) through in silico approaches. Based on the results, mean length of 5´-UTRs were more than 3´-UTR but 5´-UTRs had more GC content. Context of initial codons had 49.23% guanine at and 56.70% purine at -3 positions. Totally, 18 abiotic stress responsive Cis-acting elements were found which GA-regulated myb element was prevalent. Ribosome should consume more than 50 Kcal/mol energy to reach the start codon of 50 members. However, 23 members had Internal Ribosome Entry Site (IRES). sRNA targets were only detected in 5´-UTR (HvbZIP4) and in 3´-UTR(HvbZIP44). 225 Upstream Open Reading Frame (uORFs) were extracted where the minimum length was 12 nt. uORF of HvbZIP24 (at frame ) was similar to an upstream protein of Theobroma cacao (XM_007017561.1). In general, mRNAs of the bZIP family in barley had different regulatory structures, which might be a natural solution for accurate regulation of these regulatory proteins.

    Ralstonia solanacearum affects more than 30 plant families, including crops and wild species. Highly susceptiblecrops are potato, tobacco, tomato, eggplant and peanut. Bacterial wilt, caused by Ralstonia solanacearum, is one of the most important diseases of potato (Solanum tuberosum) in world. Strains of Ralstonia solanacearum were isolated from the wilted stems of potato cultivars from the major poato-growing areas, Jiroft, Sardueyeh and Soghan in Kerman province, using TTC medium. The strains were differentiated into biovars using biochemical tests and their pathogenicity was confirmed on tomato Cv. Red Cloud. Possible genetic diversity of R. solanacearum strains recovered from different areas was evaluated by rep-PCR, using REP, ERIC and BOX primers. A phylogenetic tree generated by NTSYS-pc2.0 software placed the strains into two major clusters, with some variations within each cluster. The genetic diversity of R. solanacearum strains infecting potato in Kerman province has probably arisen from the importation of latently infected seed tubers.

    The aim of this study was to evaluate the effects of cultivar, sucrose and hormone on in vitro microtuberization of potato (Solanum tuberosum L.). For this purpose, the experiment was done as a factorial based on completely randomized design with three replications on three potato cultivars so called Agria, Marfona and Satina. In this research, four levels of sucrose (40, 60, 80 and 100 g.l-1) were studied in combination with with two hormonal treatments (hormone free and medium containing 10 mg.l-1BAP and 0.5 mg.l-1NAA) on explants of these cultivars. The number, weight, diameter and eye of microtubers induced per plantlet were measured at 12 weeks after culture initiation. Analysis of variance showed significant differences between cultivars, sucrose and hormone levels for all traits. In this experiment, the highest values for all traits studied among cultivars belong to Satina, while Marfona had the lowest values for all of traits except number of microtubers that was better than Agria. Among the four concentrations of sucrose, the highest values for all traits studies was observed in the medium containing 80 g.l-1 sucrose, followed by mediums containing 60, 40 and 100 g.l-1 sucrose respectively. Although tuberization was initiated in the absence of hormone because of sucrose, nevertheless, the presence of hormone accelerated microtuberization. In general, results showed that the highest number, weight and diameter of microtuber and eye number were obtained in the Stina cultivar, 80 g.l-1 sucrose and hormonal combination10 mg.l-1BAP and 0.5 mg.l-1 NAA.

    Salinity stress generates oxidative stress and the reactive oxygen radicals which is toxic for cell. Plants adapted to develop antioxidant system to protect the cells from these toxins. In this study, the effect of long term salinity stress was studied on antioxidant enzymes activity like peroxidase and ascorbate peroxidase, chlorophyll, membrane leakage and water relation of leaves in halophyte plant Aeluropus littoralis. The leaves and root of growth plant treated with NaCl at different salt concentration (0, 250, 450 and 650 mM) for a period of 60 days. The results indicated enzyme activity of peroxidase and ascorbate peroxidase increased in all concentration of NaCl in leaves. In root, the activity of peroxidase in 450 and 650 mM and in ascorbate peroxidase in 250 and 450 mM of NaCl increased. In 650 mM of treatment the amount of membrane leakage was high while chlorophyll content and relative water content (RWC) were decrease.

    Knowledge of genetic diversity in breeding programs based on various marker, including parental choice is important. The most important part of plant breeding programs understanding of genetic diversity to classify population based on different markers for selecting the suitable parents. In this research, genetic diversity of 16 genotypes of wheat was evaluated using SDS-PAGE protein markers. Extraction of endosperm proteins and then concentration of proteins was measured by Bradford method. For separating and patterning of the extracted proteins, SDS-PAGE technique via poly acrylamide electrophoresis was used. After staining of proteins by coomassie blue R-250 and scoring of the bands, cluster analysis was computed based on Jaccard coefficient which that the genotypes classified into 3 separate groups. Thus according to genetic distances between the genotypes, we can use cross between Omid and Gaspard for obtain the highest amount of heterosis in future breeding program.

    In this study incidence of Beet mosaic virus (BtMV) was assessed in sugar beet fields in Razavi Khorasan, North Khorasan and Alborz provinces and to perform phylogenetic analysis of Iranian BtMV isolates and to study the phylogenetic relationship among them with other reported BtMV isolates, a part of the viral genome consists of NIb and coat protein genes was sequenced. To do this, RNA was extracted from ELISA positive samples and Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) was performed by specific designed primers and amplified DNA fragments were cloned in PTZ57R / T plasmid and sequenced. The NIb and coat protein (CP) were aligned with other sequences available in GenBank. Then, percent equity matrix and evolutionary parameters were obtained. Phylogenetic tree was drawn and recombination analyses were performed. Based on the phylogenetic tree, BtMV isolates were divided into two groups, China, Slovakia and Australia (Euroasia) isolates were placed in a same group and American isolate was located in another group. Results showed that the Iranian isolates were placed in a separate cluster in the Euroasia group. This is the first report of partial sequence of Iranian BtMV genome.

    Canola which is being used for its oil is affected by salinity stress. Salinity is one of the most important abiotic stressful conditions which affects productivity and quality of agricultural products. Pseudomonas fluorescens is a Plant growth promoting rhizobacteria having symbiotic relation with plants that reduces adverse effects and increases plant growth. In this study salinity stress has been applied in two levels (0 and 300 mM NaCl). Absence and presence of P. fluorescens was the other treatment applied to plant root with three replications. Statistical analysis was carried out as split plot based on completely randomized design. In order to evaluate proteome analysis of canola root under salinity stress inoculated with P. fluorescens 2D-PAGE approach was carried out. To reach this goal proteome extract of root was extracted via TCA-acetone method. Isoelectric focusing and SDS-PAGE for 1st and 2nd dimensions, were used, respectively. Acquired gels were analyzed using PD-quest software after staining with silver nitrate. Results demonstrated 216 spots on the gels. Sixteen spots showed significant change as result of salinity stress. Thirteen spots showed change in quantity as result of presence of P. fluorescens FY32. After applying salinity and inoculation PGPR, 23 spots presented changes significantly. After identification of proteins using molecular weight and PI, it has been recognized that most of the responding proteins to salinity and PGPR presence conditions are categorized in metabolic/ energy related pathways. Then, signaling, maintenance/ defense, channel proteins and protein structure/ location related proteins were the ones that had high frequency respectively.

    Endosperm storage proteins are important factors affecting wheat grain quality, so that high molecular weight glutenin subunits play a key role in the gluten formation and structure and are closely related to wheat quality. To investigate the allelic variation at the loci coding high molecular weight glutenin subunits (HMW-GS) among 25 bread wheat genotypes of, nine allele-specific primer pairs (Glu-A1, Glu-B1, Glu-D1) were used. Results showed that at the Glu-A1 locus, three allele (AxNull, Ax1 and Ax2*) were identified. The highest frequency belonged to allele Ax2* (56%). At the Glu-B1 locus, six alleles were detected, totally. The allelic combination of Bx7CDܯ was 32% in frequency which showed the highest frequency. At the Glu-D1 locus, two alleles were detected. At this locus, allelic combination of Dx2­༭ had the highest frequency of 82% among the detected alleles. The Pfluger scoring system was utilized to score Glutenin composition. The studied genotypes showed different allelic combinations with a different level of quality score. Most of genotypes were ranked as good to excellent bread-making quality. Cluster analysis grouped all high quality cultivars in a separate cluster. The variations identified in this study can be a valuable resource of allelic diversity for breeding programs to improve the quality of wheat.
