    سابقه و هدف
    یکی از مشکلات تزریق خون مزمن؛ آلوایمیونیزاسیون یا به وجود آمدن آنتی بادی علیه زیرگروه های خونی است. یکی از این زیرگروه ها، آنتی ژن های سیستم Kell می باشد. در این مطالعه بر آن شدیم که درصد فراوانی گروه خونی Kell را در کیسه خون های اهدایی که در درمانگاه تالاسمی مصرف می شود تعیین نماییم.
    مواد و روش ها
    در یک مطالعه مقطعی در سال 1392، بر روی کلیه کیسه خون هایی که برای تزریق خون به درمانگاه تالاسمی آورده می شد، ابتدا مشخصات هر کیسه خون شامل گروه خون، شماره کیسه خون، سن و جنس اهدا کننده ثبت شده و سپس از هر کیسه یک کورد جدا و با کیت آنتی Kell بررسی شد. نتایج با استفاده از نرم افزار 18SPSS و آزمون کای دو مورد تجزیه و تحلیل قرارگرفت.
    یافته ها
    تعداد 11557 کیسه خون در طول یک سال بررسی شد، 7/98% مربوط به مردان و 3/1% زنان بودند. کمترین سن اهدا 17 سال و مسن ترین اهداکننده 65 سال بود. در میان اهداکنندگان؛ 6/19% اهدا کننده بار اول، 4/27% با سابقه و 9/52% اهداکننده مستمر بودند. 11118 (2/96%) کیسه، آنتی ژن Kellمنفی و 439 (8/3%) کیسه، آنتی ژن Kellمثبت بودند.
    نتیجه گیری
    در این مطالعه نشان داده شد که کمتر از 4% افراد اهداکننده از لحاظ آنتی ژن Kell مثبت هستند و با توجه به عواقب یک واکنش همولیتیک در یک بیمار با تزریق خون مزمن، بهتر است که کیسه های خون برای این مصرف کنندگان غربال شود و چون بیش از 96% کیسه ها آنتی ژن منفی هستند، لذا می توان کیسه های آنتی ژن Kell مثبت را از چرخه تزریق خون حذف کرد.

    سابقه و هدف
    در طب انتقال خون، تضمین سلامت و کیفیت خون و فرآورده های آن، امری ضروری می باشد. استانداردهای ملی انتقال خون، کنترل کیفی حداقل 1% فرآورده های تولید شده را ضروری دانسته و برای هر فرآورده، شاخص های لازم جهت کنترل کیفی را تعریف کرده است. این مطالعه با هدف ارزیابی و تحلیل نتایج کنترل کیفی فرآورده گلبول قرمز در مراکز انتقال خون ایران طی سال های 1392-1391 انجام شد.
    مواد و روش ها
    مطالعه حاضر از نوع توصیفی- مقطعی بود.در بازه زمانی سال های 1392-1391، اطلاعات مربوط به فرآورده گلبول قرمز کنترل کیفی شده در سازمان انتقال خون ایران، از کلیه واحدهای کنترل کیفی پایگاه های سراسر کشور جمع آوری و سپس همه اطلاعات در نرم افزار 2010Excel وارد گردید. ابتدا نتایج با مقادیر استاندارد مقایسه شد، درصد موارد عدم انطباق هریک از شاخص ها، mean ± SD شاخص های متغیر، و نیز CV به عنوان یک شاخص پراکندگی محاسبه گردید.
    یافته ها
    درصد مطلوبیت شاخص های مختلف به این شرح ارایه شد؛ استریلیتی 100%، شاخص حجم 8/93%، هماتوکریت 2/91%، هموگلوبین 6/99%، شاخص همولیز 6/98%، عدم مشاهده لخته 8/97% و mean±SD شاخص حجم 9/20 ± 270 میلی لیتر، هماتوکریت4% ± 9/74%، هموگلوبین 9/7 ± 63 گرم در کیسه و شاخص همولیز 16/0% ± 23/0% بود.
    نتیجه گیری
    با توجه به ارزیابی و تحلیل یافته های کنترل کیفی فرآورده گلبول قرمز، شاخص های کیفی با الزامات کیفی ملی و بین المللی مطابقت دارد.

    سابقه و هدف
    سلول های بنیادی مزانشیمی، یک منبع سلولی ایده آل برای پیوند می باشند. اما استرس های مختلف در طول تکثیر آزمایشگاهی منجر به کاهش بقای سلولی آن ها پس از پیوند می شود. بنابراین به کارگیری راه کارهای عملی جهت افزایش میزان بقای آن ها در ریز محیط سرشار از استرس بسیار ضروری است. هدف این مطالعه، بررسی اثر سکرتوم ترشحی سلول های بنیادی مزانشیمی دست ورزی شده با دو ژن Nrf2 و HIF-1&alpha بر بقای این سلول ها، تحت استرس های سلولی مختلف بود.
    مواد و روش ها
    در یک مطالعه تجربی، پلاسمیدهای نوترکیب pcDNA3.1-HIF-1&alpha و pcDNA3.1-Nrf2 به طور جداگانه و هم زمان به درون سلول های بنیادی مزانشیمی بند ناف ترانسفکت شدند. 72 ساعت بعد، بیان HIF-1&alpha و Nrf2 در سلول های دست ورزی شده به روش RT-PCR بررسی شد. گروه های سلولی مختلف با استرس اکسیداتیو، فقر سرمی و کمبود اکسیژن مواجه شدند. سکرتوم سلول های ترانسفکت شده جمع آوری و تغلیظ شده و با سلول های بنیادی مزانشیمی بند ناف مجاور شد. بقای سلول ها پس از اعمال شرایط استرس مشابه با روش WST-1 بررسی شد.
    یافته ها
    سلول های هم زمان ترانسفکت شده، ژن های HIF-1&alpha و Nrf2 را بیان کردند. این سلول ها میزان بقای (5/5% ± 5/84%) بیشتری را در مقایسه با گروه کنترل(4% ± 3/55%) نشان دادند. هم چنین بقای سلول های کشت داده شده با سکرتوم پس از مواجهه با استرس های مختلف، 6% ± 3/81% و بقای گروه کنترل 3/4% ± 9/57% بود.
    نتیجه گیری
    سکرتوم سلول های دست ورزی شده، سلول های بنیادی مزانشیمی را در برابر استرس اکسیداتیو، فقر سرمی و کمبود اکسیژن محافظت می کند.

    سابقه و هدف
    پیشگیری، درمان و کنترل سرطان با استفاده از مواد طبیعی در حال حاضر به عنوان یک استراتژی امیدوارکننده در نظر گرفته شده است. در این خصوص، کافئین که یک آلکالوئید پورینی موجود در گیاهان مختلف از جمله قهوه، کاکائو، کولا و چای می باشد، به دلیل مهار چند منظوره سرطان از جمله لوسمی ها به تازگی توجه زیادی به خود جلب کرده است.
    مواد و روش ها
    در یک مطالعه تجربی به منظور بررسی اثر کافئین در لوسمی پرومیلوسیتیک حاد، سلول های NB4 در غلظت های 1، 2 و 4 میلی مولار کافئین کشت داده شد و متعاقبا از آزمون های MTT، Caspase 3، BrdU cell proliferation assay و Quantitative Real-Time PCR جهت بررسی اثر کافئین در القای مرگ سلولی و مطالعه مکانیسم احتمالی آن استفاده شد. آزمون paired student t test و نرم افزار 16 SPSS جهت تحلیل داده ها به کار رفت.
    یافته ها
    بررسی میزان ساخت DNA و بقای سلولی با استفاده از روش های BrdU و MTT، نشان داد که کافئین میزان تکثیر و بقای سلوهای NB4 را به صورت وابسته به دوز و زمان کاهش می دهد. هم چنین نتایج به دست آمده از این تحقیق نشان داد که افزایش دوز کافئین موجب القای فعال شدن آنزیم کاسپاز 3 و افزایش بیان mRNA ژن Bax و p21 می گردد.
    نتیجه گیری
    در مجموع، با توجه به این که مشخص شد کافئین می تواند موجب مهار تکثیر و القای آپوپتوز در سلول های لوسمی پرومیلوسیتیک حاد NB4 شود، لذا می توان چنین نتیجه گیری کرد که مصرف کافئین به عنوان ماده ای در چای ممکن است یک عامل مفید جهت القای مرگ سلولی این سلول های بدخیم در مبتلایان به لوسمی پرومیلوسیتیک حاد عمل کند.

    سابقه و هدف
    امروزه وارفارین به طور وسیعی توسط پزشکان تجویز می شود. عوامل مختلفی هم چون سن، جنس، رژیم غذایی، داروهای مصرفی و از همه مهمتر عوامل ژنتیکی در تعیین دوز وارفارین موثر می باشند. از عوامل موثر ژنتیکی می توان به سه آلل اصلی ژن CYP2C9 اشاره نمود. با توجه به روند رو به پیشرفت بیماری قلبی، این مطالعه جهت ارزیابی پلی مورفیسم های رایج در بیماران تحت درمان با وارفارین انجام شد.
    مواد و روش ها
    مطالعه تجربی حاضر در سال 1390-1389در آزمایشگاه پیوند انجام گرفت. بیماران به دلایل مختلف از جمله مشکلات قلبی وارفارین دریافت می کردند. پلی مورفیسم های ژن CYP2C9 به روش PCR-RFLP مورد بررسی قرار گرفتند و در نهایت یافته ها توسط نرم افزار 13 SPSS و آزمون آماری t-test تجزیه و تحلیل شدند.
    یافته ها
    مطالعه بر روی 100 بیمار انجام شده بود که 3/47% آن ها مونث و 7/52% آن ها مذکر بودند. فراوانی افراد با آلل *1*1، *1*2، *1*3 و *2*3 برای ژن CYP2C9 به ترتیب 42/68%، 52/10%، 94/18% و 1/2% بود. دوز روزانه وارفارین مصرفی در افراد با آلل *1*1 بیشتر از افراد با آلل *1*2، *1*3 و *2*3 بود(02/0 p=؛ 57/1 ± 58/4 میلی گرم در روز).
    نتیجه گیری
    نتایج تحقیق نشان داد حضور پلی مورفیسم ژن CYP2C9 اثر مهمی بر دوز وارفارین مورد نیاز برای حفظ INR در محدوده 3-2 دارد. در این مطالعه آلل *1*2 CYP2C9 شایع ترین پلی مورفیسم های موجود بود ولی افراد با ژنوتیپ های *2*2 و *2*3 نیز وجود دارند که اهمیت نیاز به بررسی ژنوتیپ وارفارین را در جامعه ایرانی نشان می دهد.

    سابقه و هدف
    تکنولوژی ریزآرایه، یک تصویر کلی از میزان بیان هزاران ژن به طور هم زمان ارایه می دهد. تفسیر داده های ریز آرایه بدون آنالیز آماری و روش های هوش مصنوعی ممکن نیست. هدف این مقاله، تشخیص انواع لوسمی حاد با استفاده از مجموعه داده های ریز آرایه و الگوریتم های داده کاوی بود.
    مواد و روش ها
    در این مطالعه توصیفی از داده های بیان 7129 ژن مربوط به 72 بیمار مبتلا به لوسمی استفاده شد. سپس با انتخاب ژن های مهم بر اساس روش های ضریب همبستگی، بهره اطلاعاتی، نسبت بهره و امتیاز Fisher و با استفاده از روش های جداکننده خطی، ماشین بردار پشتیبان، k نزدیک ترین همسایه، بیزین ساده، شبکه بیزین، نزدیک ترین میانگین، رگرسیون لجستیک، شبکه عصبی پرسپترون چند لایه و درخت تصمیم J48 برروی ژن های انتخاب شده به تشخیص لوسمی میلوژنیک و لنفوسیتیک حاد پرداخته شد.
    یافته ها
    روش های نزدیک ترین میانگین، ماشین بردار پشتیبان، k نزدیک ترین همسایه، بیزین ساده و شبکه عصبی پرسپترون چند لایه با استفاده از 39 ژن انتخاب شده توسط نسبت بهره با دقت 100٪، قادر به تشخیص لوسمی میلوژنیک و لنفوسیتیک حاد هستند. هم چنین روش ماشین بردار پشتیبان با استفاده از 87 ژن انتخاب شده توسط بهره اطلاعاتی و روش شبکه عصبی پرسپترون چند لایه با استفاده از 133 ژن انتخاب شده توسط بهره اطلاعاتی با دقت 100٪، قادر به تشخیص آن می باشند.
    نتیجه گیری
    نتایج این مطالعه نشان داد که انتخاب ژن ها و الگوریتم های داده کاوی قادر به تشخیص انواع لوسمی با دقت بسیار بالایی هستند، بنابراین با استفاده از این روش ها، می توان تصمیمات مناسبی در مورد نحوه تشخیص و درمان بیماران گرفت.

    سابقه و هدف
    تقارن، یک ویژگی مورفولوژی آنتروپومتری در جهت استقرار محورهای بدن در طول دوران جنینی به وجود آمده است. درماتوگلیفیک علمی است که به مطالعه صحیح و اصولی خطوط پوستی موجود در کف دست، پا و نوک انگشتان می پردازد. هدف این تحقیق، بررسی تقارن سنجی و ارتباط خطوط اثر انگشت و گروه های مختلف خونی در جمعیتی از زنان فارس ساکن خراسان رضوی بود.
    مواد و روش ها
    در یک مطالعه توصیفی، نوع گروه خونی تعیین و پرینت بند اول انگشتان دست چپ و راست 53 زن باردار سالم در محدوده سنی 18 تا 42 سال در خراسان رضوی با استفاده از جوهر چاپ تهیه شد. الگوهای درماتوگلیفیک آن ها از نظر کمی و کیفی مقایسه و فراوانی الگوها در هرنوع گروه خونی تعیین شد. در تحلیل داده ها از آزمون کای دو و نرم افزار 16 SPSS و 15MINITAB استفاده شد.
    یافته ها
    اختلاف معنادار نتایج تقارن سنجی خط شماری الگوهای درماتوگلیفیک بین انگشت اشاره راست با چپ زنان گروه خونی A، گویای عدم تقارن این الگو در دو دست بود(اختلاف میانگین: 72/2، انحراف معیار: 34/1)(05/0p<). بین الگوی اثر انگشت و نوع انگشتان در دست راست افراد با گروه خونی B و بین الگوی اثر انگشت و نوع انگشتان در دست چپ افراد با گروه خونی B و O تفاوت معناداری در سطح 5% دیده شد. بین الگوی اثر انگشت و نوع انگشتان در هر دو دست افراد با Rh مثبت تفاوت معنادار در سطح یک درصد وجود داشت.
    نتیجه گیری
    مطالعه حاضر، وجود ارتباط بین الگوی اثر انگشت و گروه خونی ABOو Rh را آشکار کرد.

    سابقه و هدف
    تالاسمی، شایع ترین بیماری ژنتیکی در انسان است که در پیشگیری از آن، آگاهی عمومی نقش مهمی دارد. لذا این مطالعه با هدف بررسی آگاهی و عملکرد زوجین ناقل تالاسمی بر اساس مدل اعتقاد بهداشتی طراحی و اجرا گردید.
    مواد و روش ها
    در این مطالعه توصیفی- تحلیلی از نوع مقطعی، جامعه آماری شامل کلیه زوجین ناقل تالاسمی منطقه سیستان در سال 1391 بودند. تعداد 137 نفر از زوجین ناقل تالاسمی منطقه سیستان با استفاده از روش نمونه گیری تصادفی سیستماتیک انتخاب و مورد مطالعه قرار گرفتند. برای گردآوری داده ها از پرسشنامه محقق ساخته که روایی آن توسط متخصصین و پایایی آن نیز به وسیله آزمون آماری آلفای کرونباخ تایید شده بود استفاده گردید. داده های جمع آوری شده توسط نرم افزار 15SPSS و با استفاده از آزمون های آماری t-test، آنالیز واریانس یک طرفه، کای اسکوئر و ضریب همبستگی اسپیرمن در سطح معناداری 05/0 p< مورد تجزیه و تحلیل قرار گرفت.
    یافته ها
    میانگین آگاهی و عملکرد افراد مورد پژوهش به ترتیب 8/2 ± 06/13 و 2 ± 8/10 نمره از 20 و 14 نمره بود. فقط 7/11% افراد مورد پژوهش از سطح آگاهی خوبی برخوردار بودند، 8/62% افراد، شدت درک بالایی داشتند. 9/48% (67 نفر) آزمایش مرحله اول PND و 7/38% (53 نفر) آزمایش مرحله دوم PND را انجام داده بودند. بین سازه های مدل اعتقاد بهداشتی و دانش و عملکرد افراد رابطه معنادار آماری وجود داشت (05/0 p<).
    نتیجه گیری
    از آن جایی که اکثر افراد مورد مطالعه از آگاهی مناسبی درباره روش های پیشگیری از تالاسمی ماژور برخوردار نبودند، لازم است برنامه های آموزشی برای ارتقای دانش آن ها تدارک دیده شود.

    سابقه و هدف
    تالاسمی، نوعی کم خونی است که فرد در اثر بیماری دچار تغییر شکل صورت و اندام ها می شود و تصویر ذهنی بدی از خود پیدا می کند که می تواند خطر بروز ناامیدی را افزایش دهد. اخیرا وضعیت سلامت روانی این بیماران، مورد توجه مسئولین بهداشتی قرار گرفته است. این پژوهش با هدف بررسی همبستگی تصویر ذهنی با ناامیدی در بیماران10 الی 21 سال مبتلا به تالاسمی ماژور در شهرکرد انجام شد.
    مواد و روش ها
    در این مطالعه توصیفی - همبستگی، جامعه مورد مطالعه بیماران 10 الی 21 سال مبتلا به تالاسمی ماژور مراجعه کننده به مراکز درمانی بیمارستان های آموزشی وابسته به دانشگاه علوم پزشکی شهرکرد در سال 1393 بودند. 110 بیمار مبتلا به تالاسمی ماژور در مطالعه شرکت کردند و تصویر ذهنی و ناامیدی آنان توسط پرسشنامه تصویر ذهنی آفر و پرسشنامه ناامیدی بک سنجیده شد. نتایج با استفاده از ضریب همبستگی اسپیرمن، t مستقل و من ویتنی یو تجزیه و تحلیل شدند.
    یافته ها
    نتایج حاکی از این بود که 7/66% این بیماران، تصویر ذهنی متوسط و 3/37% آن ها تا حدودی ناامیدی، داشتند. با ضریب همبستگی 554/0 و 001/0p<، بین میانگین تصویر ذهنی و ناامیدی ارتباط آماری معناداری وجود داشت.
    نتیجه گیری
    برنامه ریزی به منظور ارتقاء سطح آگاهی، شرکت در کلاس ها و ارائه خدمات مشاوره روان پرستاری، برنامه های آموزشی و مشاوره ای صدا و سیما در کنار ارائه خدمات کلینیکی و درمانی، جهت بیماران مبتلا به تالاسمی ماژور دارای اهمیت می باشد.

    سابقه و هدف
    امروزه بوتاکس، به منظور کاهش چروک های صورت و چانه استفاده فراوان دارد. عوارض جانبی تزریق آن، بسیار نادر است. هدف از تحقیق زیر، معرفی یک مورد از عوارض نادر تزریق بوتاکس بود.
    مورد: بیمار خانم 30 ساله ای بود که به دلیل پتوز، دیپلوپی، اختلال در بلع و صحبت کردن و ضعف عضلانی مراجعه نموده سابقه تزریق 3 نوبت بوتاکس در ماه گذشته را می داد. در معاینه، ضعف عضلات اکستانسور گردن، چشمی، حلقی و تنفسی دیده شد. بیمار با تشخیص میاستنی گراویس، تحت حمایت تنفسی و پلاسما فرزیس درمانی قرار گرفت و ضعف عضلانی وی بهبود یافت.
    نتیجه گیری
    میاستنی گراویس توسط اتوآنتی بادی های رسپتور استیل کولین یا صفحات انتهایی حرکتی در محل اتصال نوروماسکولر ایجاد می شود. پلاسمافرزیس درمانی، سطوح پلاسمایی این اتوآنتی بادی را پایین می آورد.

    Background And Objectives
    Alloimmunization is one of the side effects of regular blood transfusion in thalassemic patients. We studied the frequency of Kell antigen in donor blood bags used in Adult Thalassemia Clinic in Tehran.
    Materials And Methods
    Our aim was to screen Kell antigen on the blood bags which came for use in Adult Thalassemia Clinic. First, we recorded the bag number with which we could access the characteristics of the blood donors such as sex, age, number of donation, and so on. Then, for Kell study, we used anti Kell kit on each cord detached from the blood bags. The results were collected and analysed by SPSS18.
    Results
    In our one year cross sectional study, we checked Kell antigen for 11557 blood bags 98.7% were for male and 1.3% for female. The minimum age of our donors was 17 years old and maximium 65. Out of the total number of blood donors, 19.6 % were first time, 27.4% had the history of prior donation, and 52.9% were repeated donors. In our kell study for K or KEL1 antigen, we had 96.2% Kell negative bags and 3.8% were Kell positive.
    Conclusions
    We had the rate of less than 4% positivity of Kell antigen on our donors showing more than 96% Kell negativity. However, for the reduction of the risk of alloimmunization in chronically transfused patients, it is better to check all bags for Kell antigen and prevent the use of Kell positive bags.

    Background And Objectives
    In transfusion medicine, safety and quality assurance of blood and blood components are very critical. National blood transfusion standards ratify that the quality control should be performed on at least 1% of the total preparations for each type of blood component they also define the essential indicators for each product to be qualified. This study aims to evaluate and analyze the results of the quality control of red blood cells (RBC) in blood centers in Iran from 2013-2014.
    Materials And Methods
    This study was descriptive-sectional. The data of red blood cell components that had been qualified in Iranian Bood Transfusion Organization were gathered from all blood centers across Iran from 2013-2014 time period. Then, all data were enterd in software microsoft excel 2010. The results were compared with the standards the percentage of parameter non-conformity, the measures for the mean ± SD of parameters, and the CV as an index of dispersion were all calculated.
    Results
    Utility functions of different parameters were as follows. Sterility: 100%, volume: 93.8%, hematocrit: 91.2%, hemoglobin: 99.6%, hemolysis index: 98.6%, no clots: 97.8%, the mean ± SD of volume: 270 ± 9.20 mL, Hematocrit: 74.9 ± 4%, Hb: 63 ± 7.9 g/bag, and hemolysis index was 0.23 ± 0.16%.
    Conclusions
    According to the evaluation and analysis results, the quality parameters of red blood cell components were consistent with national and international quality requirements.

    Background And Objectives
    Mesenchymal Stem Cells (MSCs) are the ideal cell source for transplantation. But different stresses during MSCs in vitro expansion lead to decreased survival rate after transplantation. Therefore, applying practical strategies to enhance their viability in stressful microenvironment is quite necessary. This study aimed to survey effects of HIF-1&alpha-Nrf2-engineered-MSCs secretome on MSCs survival under different stress conditions.
    Materials And Methods
    Recombinant pcDNA3.1-Nrf2 and pcDNA3.1-HIF-1&alpha were transfected and co-transfected into umbilical cord MSCs (UC-MSCs) using FUGENE HD transfection reagent. After 72 hrs, expression of Nrf2 and HIF-1&alpha were verified by RT-PCR. Different cell groups were exposed to hypoxic, serum deprived and oxidative stress conditions. The HIF-1&alpha-Nrf2-engineered-MSCs secretome was harvested and concentrated. Then, UC-MSCs were cultured in presence of this secretome and their viability was assayed using trypan blue exclusion dye and WST-1 flowing by induction of the same stress conditions.
    Results
    HIF-1&alpha-Nrf2-engineered-MSCs expressed Nrf2 and HIF-1&alpha. HIF-1&alpha-Nrf2-engineered-MSCs indicated a higher survival rate (84.5 ± 5.5%) compared with the control group (55.3 ± 4%). Moreover, the survival rate of UC-MSCs cultured with HIF-1&alpha-Nrf2-engineered-MSCs secretome was 81.6 ± 6% and it was 57.9 ± 4.3% for the control group under the same stress conditions.
    Conclusions
    HIF-1&alpha-Nrf2-engineered-MSCs secretome protects MSCs against oxidative, serum deprived and hypoxic stress conditions.

    Background And Objectives
    Prevention, control and treatment of cancer using natural materials currently have been considered as the promising strategy. In this regard, caffeine as a purine alkaloid found in various plants, including coffee, cocoa, cola and tea has recently attracted great interest because of its remarkable multifunctional inhibitory effects on cancers such as leukemia. Regarding the anti-cancer effects of caffeine, we sought to investigate the effectiveness of this methylxanthine alkaloid against NB4 acute promyelocytic leukemia cells.
    Materials And Methods
    In order to examine the effects of caffeine in APL, NB4 cells were cultured in the presence of different concentrations of caffeine subsequently, MTT, Caspase 3 activity assay, BrdU cell proliferation assay and quantitative real-time PCR were performed to assess the effect of caffeine on cell viability and the possible mechanisms of inducing cell death.
    Results
    Evaluation of DNA synthesis and cell survival using BrdU and MTT methods indicated that caffeine reduces proliferation and survival of NB4 cells in a dose and time dependent manner. The results of this study showed that elevation in the dose of caffeine induces caspase-3 activation and increases Bax and p21 gene mRNA expression levels.
    Conclusions
    Overall, our data illustrate that caffeine inhibits cell proliferation and induces apoptosis in acute promyelocytic leukemia NB4 cells. Thus, it can be concluded that caffeine as a substance in tea may be a useful agent for induction of cell death in malignant cells of patients with acute promyelocytic leukemia.

    Background And Objectives
    Warfarin and other anticoagulant medications like Coumadin are widely prescribed by physicians to prevent ischemic events. A variety of factors such as sex, age and weight can affect warfarin dosage requirements. A clinical effect of warfarin depends on highly polymorphic drug-metabolizing (CYP2C9) enzymes. The objective of this study was to investigate the impact of CYP2C9*2, CYP2C9*3 polymorphisms on the variability of warfarin dosage requirements in Iranian population that were under warfarin therapy for different reasons.
    Materials And Methods
    The study included 100 patients with the mean age of 61.3 years. The restriction fragment length polymorphism method was used to identify polymorphisms of CYP2C9 (*1, *2, *3) in Iranian patients with ischemic heart disease in Peyvand Laboratory.
    Results
    Two-thirds (68.42%) of the patients had the wild-type (WT) CYP2C9*1*1 genotype 10.53%, 18.95%, and 2.1% of the patients had CYP2C9 *1*2, *1*3, and *2*3 genotype, respectively. WT CYP2C9*1*1 genotype was associated with a higher daily warfarin dosage (4.58 ± 1.57 mg/day p= 0.02) as compared to other CYP2C9 genotypes.
    Conclusions
    The Iranian study sample is characterized by high frequency *1*1 genotypes that determine a higher warfarin-loading dose. Analysis of CYP2C9 gene variants allows the prediction of warfarin dosage. These results can be used to individualize treatment with warfarin in Iranian patients.

    Background And Objectives
    Microarray technology represents the expression of thousands of genes simultaneously. Microarray analysis may not be possible without statistical analysis and artificial intelligence methods. The aim of this paper is to diagnose acute leukemia using microarray data and data mining algorithms.
    Materials And Methods
    The expression of 7129 genes of 72 patients with leukemia was used in this study. Then, by the selection of important genes based on correlation coefficient, information gain, gain ratio and fisher score criteria and by the use of linear discriminat, support vector machine, k nearest neighbor, naïve Bayes, Bayes net, nearest mean, logistic regression, multilayer perceptron neural network and J48 decision tree methods on the selected genes, acute myeloid and lymphoblastic leukemia were attemted to be diagnosed.
    Results
    The methods of nearest mean, support vector machine, k nearest neighbor, naïve Bayes, and multilayer perceptron neural network are able to detect acute myeloid and lymphoblastic leukemia using 39 selected genes by the gain ratio with 100 percent accuracy. Moreover, support vector machine method using 87 selected genes by information gain and support vector machine method using 133 selected genes by information gain are able to detect acute myeloid and lymphoblastic leukemia with 100 percent accuracy.
    Conclusions
    The results of this study showed that gene selection and data mining algorithm are able to diagnose leukemia with high accuracy. Therefore, appropriate decisions can be made using these methods about the how of the diagnosis and treatment of patients.

    Background And Objectives
    Asymmetry as a morphological-anthropometric characteristic is developed for body axis formation. Dermatoglyphics studies the skin lines in palms, toes and fingertips accurately and systematically. The objective of this research is to investigate the relevance of the asymmetry lines of fingerprints and blood groups in a population of women living in Khorasan Razavi.
    Materials And Methods
    For this purpose, after determining the blood groups, the fingerprints of the first phalange of the left and right hands of 53 healthy pregnant women (18 to 42 years old), were printed by ink and their dermatoglyphic patterns were compared in terms of quality and quantity and the patterns’ frequencies were determined in any blood group. In analyzing the data, we used SPSS 16 & MiniTaba 15 softwares.
    Results
    The findings suggest asymmetry in women with A blood group due to a significant difference in the ridge count of dermatoglyphic patterns between their right and left indexes (Mean difference: 2.72, Standard Deviation: 1.34) (p< 0.05). There was a significant difference between fingerprint patterns and the type of fingers in the right hand of women with B blood group and between fingerprint patterns and the type of fingers in the left hand of those with B and O blood groups (p< 0.05). There was a significant difference between fingerprint patterns and the type of fingers in the both hands of women with Rh+ blood group (p< 0.01).
    Conclusions
    The present study reveals an association between the pattern of fingerprints and ABO blood groups.

    Background And Objectives
    Thalassemia is among the most common genetic diseases in humans that can largely be prevented if adequate levels of public knowledge are obtained. This study was designed based on the educational health belief model to assess knowledge and practice of thalassemia couples in prevention of incidence of major thalassemia.
    Materials And Methods
    In this descriptive-analytical cross-sectional survey, 137 couples in Sistan area by systematic random sampling were selected. Data collection tool was a questionnaire whose validity and reliability were proven by health educational specialists and Cronbach Alpha test. The data were analyzed in spss.15 software using descriptive and analytical statistics including Chi square test, T-test, and spearman correlation coefficient. The significant level was set as 0.05.
    Results
    The mean scores of knowledge and practice of participants out of 20 and 14 were 13.6 ± 2.8 and 10.8 ± 2, respectively. Only 11.7 percent of participants had good level of knowledge, and 62.8% had good level of severity perceived 48.9% (n = 67) of them had done the first stage of prenatal diagnosis of beta-thalassemia (PND) and 38.7% the second stage of PND. There was a statistically significant relationship between the constructs of health belief model and the participants’ knowledge and practice (p< 0.05).
    Conclusions
    Since majority of participants did not have adequate information about genetically preventive procedures about major thalassemia, it is necessary to hold training programs to improve their knowledge about thalassemia.

    Background And Objectives
    Thalassemia is a kind of anemia that causes the patient's malformation of face and lips. As a result, the patient's mental image will be distorted, which can increase the risk for hopelessness among the patients. Recently‚ research efforts have started to be conducted on the patients’ mental health conditions to which the health officials are urged to attend. The present article studied the correlation between mental image and hopelessness in 10-21 year old patients who suffer from major thalassemia in Shahrekord.
    Materials And Methods
    In this correlational-descriptive study, the subject community included those 10-21 year old patients with major thalassemia who had referred to teaching universities of Shahrekord University of Medical University 110 patients participated in the study and their mental image and hopelessness were both measured via two distinctive questionnaires (the Offer self-image and the Beck hopelessness scale). The data were gathered, analyzed, and investigated using parametric and non-parametric methods.
    Results
    The findings indicated that 66.7% of the patients had average mental image and 37.3% were somehow hopeless. Thus, there was a significant correlation between the mental image average and that of hopelessness (r = 0.554, p < 0.001).
    Conclusions
    To help these kind of patients overcome their problems, it is necessary to improve their awareness level by (1) providing nursing and psychosis consultative services and the related training courses (2) broadcasting training and consultative programs on radio and TV channels, and (3) offering remedial-clinical services.

    Background And Objectives
    Botox decreases wrinkles in face and chin. This drug minimizes muscle activity with its side effects being rare. This article presents one of this rare cases.Case: A 30-year-old woman with petosis, diplopia, dysartheria, dysphagia and muscle weakness was admitted to the hospital. She had no history of disease. She had three Botox injections during the preceding month. In the examination, muscle weakness 4/5 (cervical extensor, ocular and pharynx) was detected. Her diagnosis was myasthenia gravis. With TPE, force enhancement after muscle stretch was seen and she was separated from artificial ventilation.
    Conclusions
    Myasthenia gravis is caused by antibody-acetylcholine receptor at the neuromuscular junction or motor end plate. Therapeutic plasmapheresis decreases the level of antibody-acetylcholine receptor.
