    بیماری سل گاوی یکی از مهم ترین بیماری های مشترک بین انسان و دام است. این بیماری به لحاظ شیوع و زیان اقتصادی همواره مورد توجه محققین مختلف بوده است. علیرغم تلاش های بسیار جهت تهیه واکسن موثر علیه این بیماری، هنوز مطالعات گسترده در این زمینه ادامه دارد. از آنجا که واکسیناسیون BCG در گاو مصونیت متغیری ایجاد کرده و مطلوبیت کافی نداشته، لذا ایمن سازی با پلاسمید نوترکیب حاوی ژن های کد کننده پروتئین های محافظت کننده، راهکاری امید بخش برای ساخت واکسن به شمار می آید. مایکوباکتریوم بویس (M. bovis) پاتوژن مهم بیماری سل گاوی است و آنتی ژن ESAT-6 می تواند به عنوان ایمونوژن حفاظتی و نیز کاندید مناسب برای DNA واکسن محسوب شود. از این رو، انجام این مطالعه با هدف کلونینگ و تایید بیان پروتئین نوترکیب به عنوان کاندیدای ایمنی زایی در سلول های میوبلاست موش BALB/c مورد بررسی قرار گرفت. آنتی ژن ESAT-6 در پلاسمید بیانی یوکاریوتی pcDNA3.1(+)کلون شد. صحت پلاسمید نوترکیب pcDNA3.1(+)/ESAT-6، با روش کلونی PCR، نقشه یابی آنزیمی و توالی یابی تایید شد. پلاسمید نوترکیب pcDNA3.1(+)/ESAT-6 به سلول های میوبلاست موش تزریق و پس از یک هفته از بافت ماهیچه نمونه گیری شد. نتایج تجزیه وسترن بلات، بیان پروتئین نوترکیب ESAT-6 را در سلول های میوبلاست تایید کرد. پیش بینی ساختاری نشان داد که پروتئین نوترکیب از شاخص آنتی ژنیسیتی و هیدروفوبیسیتی برخوردار است. نتایج آزمایش نشان داد که بیان پروتئین rESAT-6 در سلول های میوبلاست موش تاییدی بر عملکرد پلاسمید نوترکیب در سلول های میوبلاست موش است و پلاسمید نوترکیب pcDNA3.1(+)/ESAT-6 پتانسیل استفاده در بررسی ایمنی زایی و تشخیص را در تحقیقات دارد.

    ریحان (Ocimum basilicum)، گیاه دارویی از خانواده نعناعیان (Lamiaceae) است که دارای ترکیبات حلقوی و اسانس بوده و خواص ضد باکتری و آنتی اکسیدانی دارد. کیتوزان از ترکیبات اصلی دیواره سلولی بسیاری از گونه های قارچی است که به عنوان الیسیتور زیستی در بهبود بیوسنتز متابولیت های ثانوی استفاده می شود. در این پژوهش، اثر کیتوزان بر ترکیبات فنیل پروپانوئیدی و بیان ژن و فعالیت آنزیم فنیل آلانین آمونیالیاز (PAL) مورد بررسی قرار گرفت. بدین منظور گیاه ریحان در مرحله پیش گلدهی با غلظت دو گرم در لیتر کیتوزان تیمار شد و سپس در زمان های یک، 2، 3 و 5 روز بعد از اعمال تیمار کیتوزان برداشت شد. تجزیه اسانس نشان داد که میزان متیل چاویکول و متیل اوژنول تحت تاثیر کیتوزان در مقایسه با شاهد افزایش یافت، به طوری که بیشترین میزان آنها به ترتیب در زمان های اول و دوم بعد تیمار بود. بیان ژن و فعالیت آنزیم PAL نیز تحت تاثیر کیتوزان نسبت به شاهد نشان داد که بیشترین میزان بیان و فعالیت آنزیم یک روز بعد از اعمال تیمار کیتوزان بوده و در روز پنجم بعد از اعمال تیمار کاهش یافت. تغییرات میزان بیان ژن و فعالیت آنزیم PAL با روند تغییرات ترکیبات فنیل پروپانوئیدی در زمان های مختلف برداشت مطابقت داشت. بنابراین کیتوزان به عنوان الیسیتور زیستی از طریق افزایش بیان ژن و فعالیت آنزیم PAL سبب افزایش ترکیبات فنیل پروپانوئیدی شد.

    امروزه مطالعه تجمع غیر طبیعی پروتئین ها مورد توجه محققین حوزه های مختلف علوم به ویژه پزشکی و زیست فناوری قرار گرفته است. تجمع فیبریل های آمیلوئیدی پروتئین ها می تواند در بروز بیماری های مهمی ازجمله آلزایمر، دیابت قندی نوع II و پارکینسون نقش داشته باشد. در زیست فناوری از مشکلات اصلی تولید طیف وسیعی از پروتئین ها و پلی پپتیدهای نوترکیب تجمع برگشت ناپذیر در حین خالص سازی می باشد. در پروتئین های نوترکیب معمولا دنباله هایی در انتهای کربوکسیل یا انتهای آمینی قرار می دهند که می توانند در ساختار، خصوصیات فیزیکی و شیمیایی پروتئین ها تاثیر بگذارند. تاکنون مطالعه جامعی بر روی تاثیر این دنباله ها درتمایل تجمع پذیری پروتئین های نوترکیب انجام نگرفته است. در این مطالعه اثر دنباله های مختلف درتمایل تجمع پذیری پروتئین های نوترکیب با الگوریتم بیوانفورماتیکی AGGRESCAN مورد بررسی قرار گرفت. بررسی ها نشان داد که دنباله ها می توانند بر روی تجمع پذیری پروتئین های نوترکیب اثر بگذارند. برای تعیین صحت نتایج پیش گویی، فرآیند تجمع و فیبریلاسیون پروتئین آلفاسینوکلئین به همراه دنباله هیستیدینی و بدون دنباله با روش های CD، AFM و فلوریمتری و استانداردهای ویژه فیبریل های آمیلوئیدی بررسی شد. آلفا سینوکلئین از پروتئین های مهمی است که تشکیل پلاک های آمیلوئیدی گسترده ای در بیماری های نورودجنریتیو می دهد. برنامه بیو انفورماتیکی نشان داد که دنباله اضافه شده توانست باعث افزایش تمایل پروتئین برای فیبریلاسیون شود. در مطالعه برون تنی داده های حاصل از تجزیه فرآیند فیبریلاسیون نیز موید همین مسئله بود و نشان داد که حضور دنباله موجب افزایش میزان فیبریلاسیون آلفاسینوکلئین شد. در نتیجه پیشنهاد می شود، در بسیاری مطالعات، به ویژه مطالعات مربوط به چگونگی بیماری زایی یا جستجوی داروهای مهارکننده فیبریلاسیون بهتر است که دنباله ها از پروتئین حذف شوند.

    وجود تنوع ژنتیکی برای تداوم و پیشرفت برنامه های به نژادی گیاهان زراعی و افزایش کارایی انتخاب ضروری است. در این راستا نشانگرهای مولکولی بطور موفقیت آمیز در ارزیابی تنوع ژنتیکی انواع گیاهان زراعی مورد استفاده قرار گرفته اند. بدین منظور 20 رقم گندم نان در قالب طرح کاملا تصادفی کشت شدند و 5 صفت فیزیولوژیک کلروفیل کل، کلروفیل a، کلروفیل b، کارتنوئید و پایداری غشا سیتوپلاسمی در آنها اندازه گیری شد و همچنین در سطح مولکولی نیز از 12 جفت آغازگر ریزماهواره استفاده شد. جهت تعیین روابط ژنتیکی بین ارقام، تجزیه خوشه ایبه روش Ward انجام و ارقام مورد بررسی به چهار گروه تقسیم شدند. سطح اطلاعات چند شکلی از 0/32 (مکان ژنی Xgwm10) تا 0/725 (مکان ژنی Xgwm33) برای نشانگرهای SSR متفاوت بود. با استفاده از روش رگرسیون گام به گام ارتباط بین هر کدام از 5 صفت فیزیولوژیک و 40 نشانگر چند شکل مورد بررسی قرار گرفت. نتایج حاصل از رگرسیون گام به گام ارتباط معنی دار دو نشانگر را با 5 صفت فیزیولوژیک در دمای شاهد (C°8) و ارتباط معنی دار 6 نشانگر را با 5 صفت تحت تنش شدید (C°2-) نشان داد که می توان از آنها در انتخاب مقدماتی در برنامه های اصلاحی استفاده کرد. همچنین نتایج نشان داد که برخی ازنشانگرها با بیش از یک صفت ارتباط دارند که بیانگر پیوستگی صفات با یکدیگر و یا تاثیر ژن های چند اثره می تواند باشد.

    نشانگرهای ریزماهواره در بررسی تکامل نژادی و تهیه نقشه های ژنی و لینکاژی نقش موثری دارند. در سال های اخیر مطالعات بسیاری برای شناسایی نشانگرهای ریزماهواره (با مبدا ژنوم یا ترنسکریپتوم) در گونه های مختلف مرکبات با استفاده از روش های متداول انجام شده است. در این مطالعه برای اولین بار از داده های بدست آمده از توالی یابی عمیق ترنسکریپتوم نارنگی کلمانتین (Citrus clementina) برای شناسایی مکان های ریزماهواره در مرکبات استفاده شد. به منظور ارزیابی کارآیی ریزماهواره های بدست آمده از این روش در مطالعات روابط فیلوژنی، برای تعدادی از این مکان ها آغازگر طراحی و الگوی باندی آن ها در 34 ژنوتیپ مرکبات بررسی شد. در مجموع 9082 مکان ریزماهواره از 75659 یونی ژن بدست آمده از توالی یابی عمیق ترنسکریپتوم نارنگی کلمانتین، شناسایی شد که بیشترین فراوانی به ترتیب مربوط به ریزماهواره های دو و سه نوکلئوتیدی بود. تعداد بالای EST-SSRهای بدست آمده نشان دهنده بازده بالای روش نسل جدید توالی یابی در شناسایی نشانگرهای مولکولی بود. 25 مکان ریزماهواره دو و سه نوکلئوتیدی برای طراحی آغازگر انتخاب شدند. در مجموع 65 آلل با میانگین 4/25 آلل در هر مکان و میانگین آلل موثر 2/53 از 16 جفت آغازگر چندشکل و با کیفیت تفکیک بالا بدست آمد. میانگین محتوای اطلاعات چندشکلی 0/48 بود که بیشترین مقدار آن در مکان های ABRII7 و ABRII9 مشاهده شد. مطالعه روابط ژنتیکی 34 ژنوتیپ مرکبات به روش تجزیه خوشه ایبر اساس الگوریتم NJ و فاصله Uncorrected P انجام شد. بر این اساس همه ژنوتیپ ها در چهار گروه پوملو، سیترون، ماندرین و نارنج سه برگ قرار گرفتند. شباهت بالای طبقه بندی بدست آمده با سایر مطالعات تنوع ژنتیکی مرکبات نشان دهنده کیفیت بالای نشانگرهای بدست آمده از روش توالی یابی عمیق ترنسکریپتوم است.

    شناسایی و انتخاب ارقام با خصوصیات کمی و کیفی بهبود یافته، از اهمیت خاصی در اصلاح رقم برخوردار است. موتاژن های شیمیایی با اثرات موتاژنی، خزانه ژنی متنوعی را در منابع گیاهی ایجاد کرده و به عنوان یک ابزار تکمیلی در فرایندهای به نژادی به کار گرفته می-شوند. در این مطالعه به منظور ایجاد تنوع ژنتیکی در برنج (رقم ندا) از موتاژن اتیل متان سولفونات (EMS) استفاده شد و نقش جهش های حاصل از آن در بهبود تحمل به تنش شوری مورد بررسی قرار گرفت. لاین های موتانت نسل M2 در مرحله گیاهچه ای در شرایط شوری (NaCl (-1/2 مگاپاسکال ارزیابی شدند که منجر به شناسایی 9 لاین متحمل به شوری شد. این لاین ها به همراه رقم مادری در مرحله گیاه کامل در شرایط مزرعه با اعمال شوری 7 دسی زیمنس بر متر مربع نیز مورد ارزیابی قرار گرفتند که بر اساس شاخص تحمل به تنش (STI) سه لاین متحمل به شوری به نام های MT41، MT189 و MT196 شناسایی شدند. اما از نظر افت کمتر عملکرد در شرایط شوری، سه لاین موتانت MT184، MT189 و MT196 به عنوان لاین های متحمل تر به شوری شناسایی شدند. برای بررسی سطح تنوع مولکولی حاصل از تیمار جهش در لاین های موتانت، از نشانگرهای ISSR استفاده شد که 50 درصد نشانگرها چندشکلی بین رقم مادری و لاین های موتانت را نشان دادند. تجزیه خوشه ایبا روش UPGMA ژنوتیپ ها را به دو گروه عمده تفکیک کرد که گروه لاین های موتانت نسبت به گروه رقم مادری بیش از دو برابر عملکرد و شاخص تحمل به تنش را نشان داد. بر اساس این تحقیق، می توان دو لاین MT189 و MT196 را به عنوان لاین های متحمل به شوری در هر دو مرحله گیاهچه ای و گیاه بالغ معرفی کرد. همچنین، نتیجه گیری می شود که جهش القایی EMS تاثیر مطلوبی در ایجاد لاین های موتانت متحمل به شوری داشته و از نشانگرهای ISSR می توان برای جداسازی لاین های موتانت متحمل به شوری در جمعیت های موتانت برنج استفاده کرد.

    برخی از میکروارگانیسم ها که به عنوان افراطی دوست از آن ها یاد می شود قادر به زندگی در نواحی هستند که برای میکروارگانیسم های دیگر مرگ آور است. دریاچه های نمک با شوری در حد اشباع یکی از این محیط ها هستند. توانمندی های زیست فن آوری میکروارگانیسم ها در محیط های پرشور باعث اهمیت بررسی آن ها شده است. دریاچه ارومیه بزرگترین سطح آبی کشور و یکی از دریاچه-های فوق شور در جهان، در شمال غربی کشور قرار دارد که شبیه دریای پر شورGreat در آمریکاست. هدف از این تحقیق بررسی تنوع پروکاریوت های نمک دوست افراطی مناطق مختلف دریاچه ارومیه با روش مبتنی بر کشت می باشد. از مناطق مختلف شرق و غرب دریاچه نمونه برداری شد و برای جداسازی میکروارگانیسم های نمک دوست از محیط های کشتMGM، SWN وHM با درصد نمک مختلف استفاده شد. جدایه ها براساس ویژگی های ریخت شناسی و بیوشیمیایی اولیه تفکیک شدند. از میان سویه های بدست آمده از آب، خاک، لجن و نمک، تعدادی به صورت تصادفی انتخاب و ژن 16S rDNA آنها تکثیر و تعیین ترادف شدند. از بین 228 سویه حاصل، قطعه-ای از 16S rDNA برای 36 سویه منتخب، تکثیر و ترادف یابی شد که از نظر فیلوژنتیک سویه های آرکی در 8 گونه و سه جنس (Halorubrum (44 درصد، (Haloarcula (38/8 درصد و (aloterrigena (11/1 درصد قرار گرفتند. باکتری های جدا شده هم به دو جنس Salicola و Pseudomonas تعلق داشتند که از این میان Pseudomonas باکتری نمک دوست افراطی نیست. میزان شباهت سویه ها با تاکسون های شناخته شده میکربی بین 100-96/5 درصد بود. از بین سویه ها، 5 سویه شباهت کمتر از 98/7درصد با سویه های استاندارد نشان داد که محدوده مرزی برای ارایه گونه جدید میکروبی بومی است. نتایج این پژوهش نشان داد که دریاچه ارومیه محیط غنی از میکروارگانیسم ها بوده که مشابه نتایج سایر مناطق پرشور بررسی شده است.

    تنش غیرزیستی عامل محدود کننده در تولیدات گیاهی محسوب می شود. تجمع پرولین رایج ترین واکنش تحت تنش خشکی و شوری بوده و به عنوان محافظت کننده اسمزی فعالیت می کند. تنش خشکی از طریق کاهش آب گیاه سبب تولید انواع گونه های فعال اکسیژن (ROS) می شود. ROS ها با تخریب پروتئین ها، اسیدهای آمینه، کلروفیل و DNA به سلول آسیب می رسانند. برای کم کردن این آسیب ها، گیاهان دارای سیستم دفاعی آنتی اکسیدانی مانند کاتالاز (CAT) هستند. یکی از روش های مقابله با تنش خشکی استفاده از تنظیم کننده های رشد گیاهی است که سبب افزایش رشد و تقسیم سلولی می شوند، همچنین بر روی بیان ژن، تولید اسیدهای نوکلئیک و پروتئین ها اثر می گذارند. در این پژوهش تاثیر هورمون های سیتوکنین و براسینواستروئید بر میزان فعالیت و بیان ژن آنزیم کاتالاز و پرولین تحت شرایط تنش خشکی در دو رقم کلزا به نام های GKH و OKAPI بررسی شد. این تحقیق به صورت آزمایش فاکتوریل در قالب طرح پایه کاملا تصادفی با سه تکرار انجام شد. در هر دو رقم کلزای پاییزه GKH2005و OKAPI بیشترین فعالیت کاتالاز در شرایط تنش خشکی در سطح هورمونی براسینواستروئید بوده و اختلاف معنی داری را با شاهد (عدم هورمون پاشی) نشان داد. در هر دو رقم تحت تنش خشکی بیشترین بیان ژن cat16در سطوح هورمونی براسینواستروئید و ترکیب سیتوکنین 100 میکرومولار + براسینواستروئید بود. نتایج مقایسه میانگین میزان بیان ژن cat54 نیز نشان داد که بیشترین بیان متعلق به سطح هورمونی براسینواستروئید بوده و نسبت به سطح عدم هورمون پاشی دارای بیان بیش از سه برابر داشت. محتوای پرولین و بیان ژن p5cs در سطوح هورمون پاشی رقم GKH به صورت افزاینده بود و در رقم OKAPI روندی کاهشی را نشان داد.

    نژادهای تجاری قارچ دکمه ای به خاطر اینکه در طی فرایندهای اصلاحی تولید شده و منشا والدینی غالبا مشابه ای دارند شباهت ژنتیکی بالایی با هم دارند. شناسایی و تفکیک ژنوتیپ های این قارچ می تواند راه گشای مسائل اصلاحی و ثبت مالکیت حقوقی در این قارچ باشد. امروزه روش های مولکولی ابزار مناسبی را جهت ارزیابی تنوع ژنتیکی و بررسی روابط خویشاوندی فراهم کرده اند. نشانگرهای ISSR توانایی جداسازی ژنوتیپ های با رابطه خویشاوندی نزدیک را دارا می باشد. از سوی دیگر، توالی های منحصر به فرد نواحی ITS و IGS ریبوزومی قارچ ها به منظور بررسی روابط تکاملی استفاده می شوند. در این مطالعه توانمندی این ابزارهای مولکولی در تفکیک ژنوتیپ های قارچ دکمه ای مورد بررسی قرار گرفت. 23 ژنوتیپ از سه گونه قارچ دکمه ای با استفاده از 20 آغازگر ISSR مورد بررسی قرار گرفتند و نواحی ITS و IGS ریبوزومی آنها نیز به کمک آغازگرهای اختصاصی تکثیر و توالی یابی شد. از 20 آغازگر ISSR، تعداد 13 آغازگر با ایجاد 245 باند قابل امتیازدهی و 242 باند چندشکل جهت بررسی روابط ژنتیکی میان ژنوتیپ ها مناسب تشخیص داده شدند. کمترین ضرایب چندشکلی (0/92 و0/95) به ترتیب به آغازگرهای 841 و 809 بیشترین ضریب چندشکلی (1/0) به 11 آغازگر دیگر تعلق گرفت. کمترین و بیشترین ضریب شباهت ژنتیکی به ترتیب بین ژنوتیپ های P4 و Dezful با 0/100 و 512 و Yousefi با 0/961 بدست آمد. آغازگرهای 808، 809، 841 و 842 توانایی جداسازی کلیه ژنوتیپ ها را با وجود روابط خویشاوندی نزدیک آنها دارند. از سوی دیگر، نتایج توالی یابی نواحی ITS و IGS نشان داد که این نواحی علیرغم عدم قابلیت تفکیک درون گونه ای، در تفکیک بین گونه ای کارآمد می باشند. بنابراین نشانگرهای ISSR در مقایسه با نواحی ITS و IGS از قابلیت بالاتری در تفکیک ژنوتیپ های با رابطه خویشاوندی نزدیک در قارچ خوراکی دکمه ای برخوردار بوده و می توانند به عنوان ابزاری کارآمد در انگشت نگاری و ثبت حقوقی نژادهای تجاری درون یک گونه مورد استفاده قرار گیرند.

    در این تحقیق پاسخ های القا شده گیاهان سازگارشده و سازگار نشده گندم نان (Triticum aestivum L.) (رقم نورستار) و گندم دوروم (Triticum turgidum L.) (ژنوتیپ های گردیش و SRN) به تنش سرما از طریق اندازه گیری شاخص های خسارت (ELI)، پراکسیداسیون چربی های غشا (MDA) و فعالیت آنزیم های آنتی اکسیدان (سوپر اکسید دیسموتاز، کاتالاز، آسکوربات پراکسیداز و گایاکول پراکسیداز) ارزیابی شد. ژنوتیپ ها پاسخ های متفاوتی به تیمارهای دمایی (23 و 5- درجه سانتی گراد) نشان دادند. تحت تنش سرما، به موازات کاهش شاخص های خسارت، فعالیت آنزیم های آنتی اکسیدان به خصوص در گیاهان سازگارشده افزایش یافت، طوری که افزایش فعالیت آنزیم ها در نورستار بیشترین و در SRN کمترین بود. سازگاری به سرما، آمادگی بیشتری را در مقابله با تنش سرما در نورستار در مقایسه با ژنوتیپ های دوروم ایجاد کرد و کمترین میزان خسارت و بیشترین میزان فعالیت سیستم دفاعی سلول در این شرایط بدست آمد. نتایج نشان داد که تفاوت فراوانی در شدت و سرعت پاسخ های سلولی در این ژنوتیپ ها وجود دارد. تغییر همزمان الگوی فعالیت آنزیم های آنتی اکسیدان که با تغییر شاخص های خسارت (ELI و MDA) مرتبط است، گویای آن است که فعالیت این آنزیم ها احتمالا در همراهی با سایر مکانیسم های دفاع سلولی، تحمل به تنش سرما یا توان زیستی گندم را افزایش داده و یا سبب بهبودی گیاه پس از اعمال تنش شدید می شود. دوره های سازگاری کوتاه مدت سبب افزایش ظرفیت ژنتیکی تحمل به سرما در ژنوتیپ های گندم شد به طوری که درجه تحمل در نورستار بیشتر از ژنوتیپ های دوروم بود. چنین شاخص هایی ممکن است در ارزیابی ژنوتیپ های گندم به تنش سرما به همراه تیمارهای تنش کوتاه مدت که موجب صرفه جویی در زمان و هزینه پژوهش شده، موثر باشند.

    اسید سالیسیلیک به عنوان یک عامل کلیدی در القای تحمل گیاهان به تنش های محیطی از جمله بیماری ها نقش دارد. پس از هجوم پاتوژن ها به خصوص عوامل قارچی، گیاهان با القای مقاومت اکتسابی سیستمیک که با افزایش میزان اسید سالیسیلیک درونی گیاه همراه است مسیرهای پیام رسانی گسترده ای از قبیل رونویسی ژن های رمزکننده پروتئین های وابسته به بیماری زایی (Pathogenesis related proteins)، آنزیم های دخیل در سنتز فیتوالکسین ها و پمپ های ناقل ABC-transporter را فعال می سازند. در این پژوهش اثر اسید سالیسیلیک بر بیان تعدادی از ژن های مرتبط با سازوکار دفاعی شامل PR1، PDF1.2، Thionin، NPR1، PDR3، PDR4، PDR5وPDR8 با استفاده از تکنیک Real time PCR در دو رقم خزر (مقاوم به بلاست) و هاشمی (حساس به بلاست) در زمان های صفر، 6، 12، 24 و 48 ساعت پس از اعمال اسید سالیسیلیک بررسی شد. نتایج نشان داد مصرف اسید سالیسیلیک سبب افزایش بیان ژن های مورد بررسی شده که این افزایش در زمان های مختلف بین دو رقم متفاوت بود به طوری که PR1، PDF1.2 وPDR3 در رقم خزر افزایش بیان نشان دادند ولی در رقم هاشمی در هیچ زمانی تفاوت معنی داری مشاهده نشد. همچنین بیان دو ژن PDR5 وPDR8 در رقم هاشمی نسبت به خزر در 48 ساعت پس از تیمار افزایش معنی داری نشان داد. بنابراین مصرف اسید سالیسیلیک می تواند سبب افزایش بیان ژن های دفاعی و احتمالا القای تحمل گیاه در برابر حمله پاتوژن ها شود. همچنین الگوی بیان ژن ها در اثر تیمار اسید سالیسیلیک تا حد زیادی وابسته به ماهیت ژنتیکی ژنوتیپ می باشد.

    این پژوهش با هدف شناسایی ژن های موثر در تحمل به شوری در گندم از طریق بررسی توالی های EST و بررسی نقش ژن رمز کننده BAX Inhibitor 1-like protein (BI-85) در پاسخ به تنش شوری اجرا شد. تجزیه و تحلیل اطلاعات EST نشان داد که ژن BI-85 در پاسخ به تنش شوری افزایش بیان داشت. به همین دلیل این ژن جهت بررسی بیشتر و تایید نقش آن در پاسخ به تنش شوری در ژنوتیپ-های زراعی متحمل و حساس گندم و خویشاوند وحشی Aegilops crassaانتخاب شد. بررسی بیان این ژن در بافت ریشه با استفاده از Real-time PCR در زمان های صفر، 12 ساعت و 3 هفته پس از اعمال تنش شوری نشان داد که میزان بیان این ژن تحت تنش شوری به طور معنی داری در Aegilops crassa (دارنده ژنوم DD) بیشتر از ژنوتیپ های زراعی متحمل و حساس افزایش می یابد اگرچه در شرایط بدون تنش میزان بیان آن در سه ژنوتیپ مورد آزمایش تفاوت معنی داری نداشت. میزان بیان ژن BI-85، 12ساعت بعد از اعمال تنش به طور معنی داری نسبت به زمان قبل از تنش در Aegilops crassa افزایش یافت و سه هفته بعد از اعمال تنش به شدت کاهش یافت در حالی که میزان بیان این ژن در ارقام گندم در زمان های قبل و بعد از تنش تفاوت معنی داری نداشت. به نظر می رسد که محصول ژنBI-85 در مسیرهای انتقال علائم خاصی که تنها در ژنوتیپ های متحمل وحشی وجود دارند و باعث افزایش تحمل نسبت به تنش شوری می شوند نقش دارد و همچنین محصول ژنBI-85 به صورت بازخورد منفی سبب کنترل بیان این ژن می شود.

    این تحقیق به منظور ارزیابی اثر جایگزینی بتائین با بخشی از متیونین بر بیان ژن آنزیم مالیک در مرغان تخمگذارتحت تنش اجرا شد. 96 قطعه مرغ (سویه تخمگذار تجاری های لاین W36 در سن 60 هفتگی در چرخه دوم تولید) به صورت طرح کاملا تصادفی در قالب آزمایش فاکتوریل با دو سطح جایگزینی بتائین با متیونین و دو سالن تیمار دمایی (تنش و بدون تنش) و سه تکرار و هشت مرغ در هر تکرار انجام شد. جهت اعمال تنش، مرغ های یک سالن را روزانه شش ساعت تحت دمای 35 درجه سانتی گراد قرار داده شد. دو سطح آزمایشی بتائین شامل جیره حاوی صددرصد متیونین به عنوان گروه شاهد و جیره حاوی87 درصد متیونین و 13 درصد بتائین به عنوان آزمون مورد بررسی قرار گرفت. پس از دو ماه تغذیه، در انتهای آزمایش چهار مرغ از هر تکرار کشتار شده و پس از استخراج کل RNA از بافت کبد، و ساخت cDNA بیان ژن مورد نظر با کمک Real time PCR بررسی شد. نتایج نشان داد که افزودن بتائین به جیره سبب کاهش معنی دار بیان ژن آنزیم به میزان 2/47 واحد در بافت کبد شد (P < 0.01). همچنین تحت تنش گرما بیان ژن آنزیم مالیک به طور معنی داری به میزان 1/96 واحد کاهش پیدا کرد (P < 0.01). بنابراین بتائین و تنش گرما بیان ژن آنزیم مالیک را در سطح mRNA مهار کرده و قادر است نقش مهمی در کاهش میزان چربی کبدی ایفا کند.

    شش نشانگر ریزماهواره به منظور بررسی تنوع و ساختار ژنتیکی دو جمعیت ماهی Oxynoemacheilus Kiabii رودخانه گاماسیاب در حوضه دو استان همدان و کرمانشاه مورد استفاده قرار گرفت. تعداد 60 نمونه از دو منطقه جمع آوری شد. هر شش نشانگر ریزماهواره مورد استفاده در این تحقیق پلی مورفیسم نشان دادند. در مجموع 70 الل برای شش جایگاه مورد استفاده بدست آمد طوری که تعداد الل در هر جایگاه در محدوده 16 – 6 بود. میانگین هتروزیگوسیتی مشاهده شده (Ho) و مورد انتظار (He) به ترتیب 0/515 و 0/849 بود. بیشتر مناطق انحراف از تعادل هاردی – واینبرگ را نشان داد. با توجه به میزان جریان ژنی بالا (14/554=Nm) بین دو منطقه و Fst پایین (0/021= Fst) به نظر می رسد تمایز پایینی بین جمعیت های این گونه در مناطق مورد بررسی وجود دارد.

    one of the most common diseases in zoonotic is Bovine tuberculosis. It has been more attended because of prevalence and economic loss. Although, many efforts have been done to produce the vaccine, but the research in this area is going on. Mycobacterium bovis (M. bovis) is the most important pathogen in bovine tuberculosis. The ESAT-6 antigen is considered as a protective immunogenic and also its important candidate for DNA vaccine. BCG vaccine makes a variable immunity period and it is not desirable. Immunization with recombinant plasmid encoding protective proteins is a promising vaccination technique Therefore, the aim of this study was cloning and expression of recombinant protein in the mice myoblast cells of BALB /c as the immunogenicity of candidate. The ESAT-6 antigen from M. bovis was cloned in a eukaryotic expression plasmid pcDNA3.1 (+). The integrity of the constructed plasmid was confirmed by using clone PCR, restriction enzyme mapping and sequencing. Recombinant plasmid pcDNA3.1 (+) /ESAT-6 injected into Myoblast cells. After one week the samples was taken from muscle tissue. The electrophoresis and western blotting confirmed the recombinant protein of ESAT-6 (rESAT-6) in Myoblast cells. Also the structure prediction showed that the recombinant protein antigenicity and hydrophobicity index is high. The current research indicated that rESAT-6 could be used as a research experimental tool in protection assays against bovine tuberculosis.

    Basil (Ocimumbasilicum), a medicinal plant of the Lamiaceae family, has ring-shaped compounds, oils, antibacterial and antioxidant characteristics. Chitosan is the main compound of fungal species and could be usedas biotic elicitor to improve secondary metabolites. In the present study, the effect of chitosan on phenylpropanoid compounds, gene expression, and activity of phenyl alanine ammonia lyase (PAL) was evaluated. The plants were treated at pre flowering stage with 2 g/L chitosan and harvested after 1, 2, 3, and 5 days after chitosan. Essential oils analysis showed that methylchavicol and methyleugenol increased under chitosan, so that the most increase was after 1 and 2 days after treatment,respectively. The results of PAL activity and gene expression showed that gene expression and PAL activity increased one day after chitosan and decreased five days after chitosan.Totally, changes in gene expression and PAL activity in different harvest stages are consistent with phenypropanoid compounds changes. Thus, chitosan, as a biotic elicitor, increased phenypropanoidcompounds by increasing PAL gene expression and activity.

    Nowadays, study on the abnormal protein aggregation has been interested by researchers in different areas of science especially medicine and biotechnology. The aggregated amyloid fibrils of proteins play the major role in several important diseases including Alzheimer’s’, Parkinson’s’ and type II diabetes. In biotechnology, a major problem during production and purification of the recombinant proteins and peptides is the formation of irreversible aggregates. In the recombinant proteins some tags are usually added in the C-terminal or N-terminal of the genes which can change the physicochemical properties of proteins. So far, no comprehensive study has been carried out on the effect of these sequences on the aggregation of recombinant proteins. In this study, by using AGGRESCAN bioinformatics algorithm, the effect of different tags on propensity of the aggregation of recombinant proteins was investigated. Results demonstrated that the tags can affect the protein aggregation. To determine accuracy of in silico prediction, the fibrillation of alpha-synuclein with or without His-tag was assessed by circular dichroism, atomic force microscopy, fluorimetry and the amyloid standard tests. Alpha-synuclein is the key protein in the development of neurodegenerative diseases by formation the extensive plaques. The bioinformatics program predicted that His-tag can induce the protein tendency to aggregation. Data obtained from experimental study also confirmed the in silico results and showed that the presence of the tag inspire the fibrillation process in alpha-synuclein. Thus it is suggested in many studies, especially studies related to the pathogenesis of fibrillation or exploring the inhibitors of protein aggregation such tags removed from proteins.

    Genetic diversity is a necessary factor for breeding programs and increasing genetic gain of selection in crop plants. Molecular markers have been successfully used for analysis of genetic diversity in various crops. For this purpose the 20 bread wheat cultivars were in a completely randomized design and five physiological traits, total chlorophyll, chlorophyll a, chlorophyll b, carotenoids and their stability was assessed in the cytoplasm membrane. Study of genetic diversity accomplished by 12 SSR marker primers. In order to determine genetic relation between cultivars, cluster analysis in Ward method was performed and cultivars were into four groups. The association between 40 microsatellite bands of 12 primer pairs and 5 physiological traits recorded on 20 wheat bread cultivars. Polymorphic information contents (PIC) of loci were in the range of 0.32 (Xgwm10) to 0.725 (Xgwm33). By multiple regression stepwise relationship of each 5 physiological traits and the 40 polymorphic markers were studied. The result of stepwise regression analysis showed the significant relationship of 2 markers with 5 physiological trait at the contort temperature and also 6 marker with those physiological marker at the severe level of spring cold stress, may facilitate preliminary selection of these traits for breeding purposes. Moreover, our results showed that some of the markers were related to more than one trait, implying close linkage and/or peliotropy of these genes on chromosomes.

    Microsatellite markers are important for phylogenetic and gene mapping studies. Several researches developed microsatellite markers (either genomic or transcriptomic) in different Citrus species during recent years. For the first time in this study, Clementine (Citrus clementina) deep transcriptome sequencing data was used for developing Citrus microsatellite markers. Primers were designed for some of these loci and their band patterns were assessed for phylogeny studies in 34 citrus genotypes. Totally 9082 SSRs were identified from 75659 Unigenes obtained from deep transcriptome sequencing of Clementine. Di- and tri- nucleotide repeats had the maximum frequencies respectively. A high number of developed EST-SSRs confirmed Next Generation Sequencing method to be highly efficiencent for molecular marker development. Totally 25 microsatellite loci were selected for primer designing. From these totally 65 alleles obtained with average of 4.25 alleles and 2.53 effective alleles per locus. The average of Polymorphic Information Content (PIC) was 0.48 and the highest PIC observed for ABRII7 and ABRII9. Phylogenetic study for 34 Citrus genotypes was done based on NJ algorithem and Uncorrected P distance. All of genotypes located in 4 groups: Pummelo, Citon, Mandarin and Trifoliate orange. This classification was similar to those obtained by previous studies illustrateed a high efficiency of microsatellite markers developed using deep transcriptome sequencing.

    Identification and selection of varieties with improved qualitative and quantitative traits has a particular importance in breeding process of current varieties and developing new cultivars. Chemical mutagens provide a diverse gene pool in plant resources and as a complementary tool are applicable in breeding programs. In this study, ethyl methane sulfonate (EMS) was used to create genetic variability in rice (cv. Neda), and the role of gained mutations was examined in improving salt tolerance. M2 mutant lines were evaluated at germinating and seedling stages under NaCl salinity (-1.2 MPa), that was resulted in identifying 9 salt tolerant lines. The lines along with original wild cultivar were also evaluated at whole plant stage under field condition with 7 dS m-1 soil EC, which three lines viz. MT41, MT189 and MT196 based on stress tolerance index (STI) were identified as salt tolerant mutants. However, regarding yield loss under salinity, three mutants viz. MT184, MT189 and MT196 were identified as salt tolerant lines. ISSR markers were used to assess genetic variability induced by mutation in mutant lines, which 50 percent of them detected polymorphism between wild cultivar and mutant lines. Cluster analysis using UPGMA method could classify studied genotypes into two distinct groups which mutant lines group showed over 2 times yield and stress tolerance index more than wild cultivar group. Based on this research, two mutant lines (MT189 and MT196) are introduced as salt tolerant lines at both seedling and whole plant stages. Also, it can be concluded that EMS-induced mutagenesis had a desire effect on the development of salt tolerant lines and that ISSR markers are applicable for differentiating salt tolerant mutants in rice mutant populations.

    Microbial life is present not only in our familiar world but also in extreme environments. Salt lakes with near or at saturating salinity are extreme environments that common all over the world. The study in detail of such environments would permit to determine not only the microbial diversity but also the gene pools and potential use of this information for biotechnological applications. Urmia Salt Lake in the northwestern of Iran is the second saltiest lake in the world and resembles the Great Salt Lake in the western USA. Water, soil, sediment and salt samples were taken from east and western sites in Urmia Salt Lake in July 2012. Direct plating, dilution plating and long incubation period were used to isolate organisms on MGM, MH, SWN medium. Isolates were taken from the samples by using the conventional culture-dependent methods. Of these, 36 isolates were selected for sequencing and phylogenetic analysis, based on their growth characteristics and colony morphology. Two hundred and twenty-eight of microorganisms were obtained from soil, salt, water and sediment samples collected from the east and western of the lake. Of these, 36 isolates were selected for sequencing and phylogenetic analysis. Results showed that 36 strains represented 8 species, belonging to 3 genera Halorubrum, Haloarcula, Haloterrigena. As total, bacterial isolates were belonged to Salicola, Pseudomonas. All strains showed 96.5 to 100 % similarity in 16S rDNA sequencing. Of these, 5 strains showed less than 98.7% sequence similarity to the closest known strains and were representatives as new taxa of Urmia Lake.The phylogenetic analysis of sequences of Urmia Lake indicated in overlaps with 16S rDNA sequences from other lakes with similar habitats.

    Abiotic stress is a limiting factor for plant products. The most common reaction under drought and saline stresses is proline accumulation as a protective osmotic action. Drought stress causes to produce reactive oxygen species by reducing plant water. This reactive oxygen species injured the cell by demolition proteins, amino acids, chlorophyll and DNA. To reducing these injurious, plants possess antioxidant defense systems such as catalase (CAT) which its main obligation is to decompose hydrogen peroxide. The best way to confrontation with drought stress is using the plant growth regulators that not only cause to plant growth and cell division, but also is a contributing factor for gene expression, producing nucleic acids and proteins. In this assay we conducted an experiment on the effect of Cytokinin and Brassinosteroied hormone's on proline content, catalase activity and their gene expression under drought stress in two Cultivars called OKAPI and GKH. This experiment was conducted as factorial based on CRD design with three replicates in greenhouse at laboratory of Agricultural Biotechnology Research Institute of Iran (ABRII) in 2012. The results showed that both varieties of rapeseed, GKH and OKAPI have the highest CAT activity in term of drought stress on Brassinosteroied hormone levels, moreover it cleared up that there is a significant difference with the control (non hormone spray). The results of Cat16 gene expression assay showed that the both cultivars in drought stress had the maximum gene expression in hormone levels Brassinosteroied and Cytokinin 100 mM + Brassinosteroied. Also the results showed that the highest mean values for cat54 gene expression was for Brassinosteroied hormone with expression level more than three times relation to non hormone spray. The content of proline and p5cs gene expression in levels of hormone spray was rising in GKH and reduced in OKAPI, respectively.

    The button mushroom commercial strains are supposed to be genetically very similar because they have originated from a limited heritage lines. Identification and strain-typing of this mushroom could solve issues of breeding and recording legal ownership. Molecular markers and sequencing of specific regions in the genome have provided appropriate tools for evaluating genetic diversity and relationship. ISSR marker has proved to be a promising marker for identification of close genotypes. On the other hand, sequences of ITS and IGS regions of ribosomal DNA in fungi are unique areas that offer useful evolution information. The objective of this work was to evaluate the potential of molecular tools for genotype identification in the button mushroom. For this purpose, 23 genotypes of three species of button mushroom were assessed for their similarity using 20 ISSR primers. ITS and IGS regions of ribosomal DNA also were amplified by specific primers and sequencing were done. Out of 20 ISSR primers, 13 proved to be discriminative in button mushroom, producing 245 scorable and 242 polymorphic bands. According to the similarity matrix, the lowest polymorphic coefficient (0.92 and 0.95) belonged to the primers 841 and 809, respectively and the highest (1.0) belonged to the other primers. Genotypes P4 and Dezful with similarity coefficient of 0.100 and genotypes yousefi and 512 with similarity coefficient of 0.961, were accordingly the least and the most similar genotypes. Data aggregation from primers No. 808, 809, 841 and 842 showed that these four primers are able to isolate all the genotypes with close relationship. On the other hand, sequencing results of ITS and IGS regions indicated that these areas are identical between genotypes of one species but are different between species. Therefore they are able to discriminate the button mushroom species. Our result demonstrated that ISSR markers compared with ITS and IGS regions are powerful enough for detection of polymorphism among closely related genotypes of button mushroom and can be used for fingerprinting and legal ownership.

    In this study, the induced responses of acclimated and non-acclimated genotypes of bread wheat (Triticum aestivum L.) (Norstar) and durum wheat (Triticum turgidum L.) (Gerdish and SRN) to cold stress have been assayed by detecting electrolyte leakage index (ELI), malondialdehyde content (MDA) and the activity of antioxidant enzymes (superoxide dismutase, catalase, ascorbate peroxidase and guiacol peroxidase). Wheat genotypes showed different responses to thermal treatments (23 and -5°C). After exposing plants into cold stress, along with decreasing of damage indices the antioxidative activities increased particularly in acclimated plants so that the maximum and minimum activity of enzymes belonged to Norstar and SRN plants. Acclimated plants showed more readiness in facing up cold stress in Norstar plants compared to durum wheat so that the minimum damage indices and the maximum enzyme activities were observed. The results have shown a potent variety in the level and speed of cell responses in these genotypes. Simultaneous changes of damage indices (ELI and MDA results) and antioxidative activities showed that these enzymes accompanied with other defense mechanisms increased cold tolerance and/ or enhance plant survive in wheat and can cause plant recovery even after sever cold stress. The short-term cold acclimation increased genetic capacity in wheat genotypes so that the degree of tolerance in Norstar was higher than that of durum genotypes. Such defense and damage indices may be used as cold tolerance marker in evaluation of genotypes in a short-term cold stress profitably in a short time and low cost.

    Different investigations have shown that salicylic acid (SA) plays a key role in plant resistance induction against environmental stresses specially diseases. Pathogen attack induced systemic acquired resistance (SAR) in plants accompanied with increasing of endogenous SA, which activates wide signal transduction pathways such as transcription of the pathogenesis related proteins (PRP), enzymes related to phytoalexins biosynthesis and ABC transporter pump. Therefore, the effect of salicylic acid on the expression of defense-related genes including PDR8, PDR5, PDR4, PDR3, NPR1, Thionin, PDF1.2 and PR1 was investigated using Real Time PCR technique in two cultivars Khazar (as resistant to blast) and Hashemi (as susceptible to blast) in 0, 6, 12, 24 and 48 hours after SA treatment. Results showed that the expression of the all studied genes was increased by SA treatment. The expression of the PR1, PDF1.2 and PDR3 was increased in cultivar Khazar while no significant differences was observed among different times of SA treatment in Hashemi. Also the expression of PDR5 and PDR8 was increased significantly in cultivar Hashemi 48 h after SA treatment. Totally, it could be concluded that SA exogenous treatment of rice might increase the expression of the defense genes and probably induce plant resistance against pathogen attack. In addition, the expression pattern of the defense related genes largely depends on plant genetic background.

    This study was carried out to identify new salt induced genes from wheat involved in salt stress tolerance using EST analysis and study the role of gene encoding BAX Inhibitor 1-like protein (BI-85) in response to salt stress. Using EST analysis, it is also cleared that BI-85 is one of the salt induced genes whose expression increased under salt stress condition. Hence, this gene was selected to determine its expression pattern in response to salinity stress in tolerant (Arg) and sensitive (Alamout) genotypes of wheat and its wild relative, Aegilops crassa (DD). Gene expression analysis at 0, 12 hours and 3 weeks after salt treatment using Real-time PCR revealed that the expression of BI_85 gene under salt stress in the root was markedly greater in Aegilops crassa compared with salt-tolerant and salt-sensitive cultivated genotypes although its expression was not significantly different between the three genotypes under unstressed conditions. Twelve hours after salt treatment, in Aegilops crassa the expression of BI-85 significantly increased in comparison with before treatment and its expression level was dramatically decreased after 3 weeks in these genotypes while the expression level of this gene was not different before and after salt treatment in cultivated genotypes. It looks there are some signal transduction pathways only in tolerant genotypes that increased salt tolerance in these genotypes and the product of BI-85 gene plays a critical role in these pathways and the protein encoded by this gene may contribute to a negative feedback regulation of BI-85 expression.

    This study was conducted to determine the effects of Betaine (Bet) substitution with a part of Methionine (Met) on malic enzyme gene expression in laying hens under heat stress. Ninety six hens (strains of commercial laying 65-wk-old Hy–Line Leghorn in second cycle of production) were performed as a completely randomized design (CRD) with a 2 ×2 factorial arrangement of treatments with three replicate cages and eight hens per cage. There were 4 dietary treatments: two levels of Bet (0 and 13%) substitute to Met combined with two environmental conditions (heat stress (HS) and thermoneutral (TN) conditions). To apply stress, the hens of a salon were exposed under 35 °C, for six hours, daily. Two experimental levels including 100% Met diet as control group and 87% Met + 13% Bet diet as trial group were surveyed. After two month feeding, four hens euthanasia by cervical dislocation and the livers were removed, frozen in liquid N2 and stored at −80 °C. After extracting the total RNA, quality was measured and was used to cDNA synthesis. Finally, mentioned genes expression evaluated by specific promoter and Real- time PCR set. The results indicated that supplementation of the basal diets with Betaine lead to decrease significantly (2.47 unit) in enzyme gene expression in liver tissue (P < 0.01). Also, heat stress decreases significantly (1.96 unit) in the malic enzyme gene expression (P < 0.01). Therefore, Betaine and heat stress could repress malic enzyme gene expression in mRNA level and play important role in decreasing liver fat.

    Six highly variable microsatellite loci were used to investigate the genetic diversity and population structure of the Oxynoemacheilus kiabii in the Gamasiab River in between the Hamedan and Kermanshah provinces. A total of 60 samples were collected from these areas. All of the 6 microsatellite loci screened in this study showed polymorphism. A total of 70 alleles were observed for six loci. The number of alleles per locus ranged from 6 to 16. The averages of observed (Ho) and expected heterozygosity (He) recorded as much as 0.515 and 0.849, respectively. Most cases deviated significantly from Hardy-Weinberg equilibrium (p≤0.01). It seems that there is a low differentiation between two populations of this species through the studied areas due to high level of gene flow (Nm=14.554) between two regions and low Fst (0.021).
