    طراحی و تولید صحیح و مناسب یک ساختار فیوژن؛ از مهمترین مشکلات در زمینه تولید مقادیر زیاد پروتئین های فانکشنال برخوردار از پیچ خوردگی های مناسب است. چنین ساختاری باید دارای کلیه اجزاء مورد نیاز یک ژن واقعی باشد. در صورتیکه ساختار فیوژن به خوبی طراحی شده باشد، می توان آن را در داخل یک میزبان مناسب کلون کرد. کلستریدیوم پرفرینجنز از عوامل مهم بیماری زای انسان و دام بوده و تعداد زیادی توکسین از جمله اپسیلون و بتا که مسئول بیماری های شدیدی هستند را تولید میکند. در مطالعه حاضر یک ساختار جدید شامل partial cds ژن توکسین اپسیلون کلستریدیوم پرفرینجنز تیپ D و partial cds ژن توکسین بتا کلستریدیوم پرفرینجنز تیپ B طراحی شد. در اولین مرحله این ژن ها با استفاده از دو جفت پرایمر آمپلیفای شده و در مرحله دوم با استفاده از پرایمرهای اپسیلون فوروارد و بتا ریورس و با بکار گرفتن قطعه AEAAAKEAAAKAبه عنوان لینکر با یکدیگر فیوز شدند. این کار بر مبنای روش fusion PCR وبا استفاده از آنزیم DNA Pfu پلیمراز که قابلیت تصحیح خطا را دارد انجام شد. قطعه فیوژن پس از اینزرت شدن در پلاسمید pJET1.2blunt در باکتری E.coli سویه TOP10 کلون شد. براساس آخرین اطلاعات موجود این مقاله اولین گزارش در خصوص طراحی و کلونینگ فیوژن ژن اپسیلون- بتا و اولین گزارش در مورد استفاده از استراتژی فیوژن واکنش زنجیره ای پلیمراز در کلستریدیا است که می تواند برای توسعه و تکوین یک واکسن نوترکیب از فیوژن پروتئین اپسیلون-بتا استفاده شود. همچنین این ساختار می تواند به عنوان مدلی برای توسعه و تکوین ساختار های جدید فیوژن پروتئین برای سایر پروتئین ها و توکسین های بالقوه نیز بکار گرفته شود.

    بیماری آگالاکسی مسری یکی از معمولترین بیماری ها در نشخوارکنندگان کوچک است که عامل آن مایکوپلاسما آگالاکتیه است. این بیماری بطور گسترده در جهان وجود دارد و ایران یکی از کشورهای درگیر می باشد. هدف از این مطالعه جداسازی و تشخیص مایکوپلاسما آگالاکتیه با روش کشت و واکنش زنجیره ای پلیمراز در نمونه های شیر گرفته شده در استان کردستان، ایران بوده است. 367 نمونه شیر از گوسفند و بز جمع آوری گردید. جهت تست واکنش زنجیره ای پلیمراز از پرایمر اختصاصی مایکوپلاسما آگالاکتیه که قطعه 375 جفت بازی را تکثیر می دهد استفاده گردید.20 نمونه (5/5%) از 367 نمونه مورد آزمایش در محیط کشت PPLO آگار مثبت بودند و5 نمونه (25%) از 20 نمونه جدا شده با پرایمر مایکوپلاسما آگالاکتیه مثبت گردیدند. از 5 نمونه جدا شده 4 نمونه(75%) مربوط به گوسفند و 1 نمونه(25%) مربوط به بز بود. نتایج تست واکنش زنجیره ای پلیمراز بر روی 367 نمونه شیر نشان داد که 11 مورد(3%) با پرایمر مایکوپلاسما آگالاکتیه مثبت می باشند. جداسازی مایکوپلاسما آگالاکتیه نشان می دهد که بیماری آگالاکسی مسری در استان کردستان وجود داشته و نتایج این مطالعه پیشنهاد می کند که روش واکنش زنجیره ای پلیمراز با کاهش زمان انجام آزمایش می تواند بعنوان یک روش آلترناتیو در کنار کشت استفاده شود.

    اندازه گیری پاسخهای آنتی بادی در حیواناتی که بطور طبیعی با ویروس BVDV مواجه می شوند ویا درحیوانات واکسینه هنوز یک روش استاندارد برای تشخیص است. برای اسهال ویرسی گاوان الایزا یک آزمایش تشخیصی ارزشمند برای اندازه گیری سطح آنتی بادی های اختصاصی ویروس اسهال ویروسی ونیز سطح آنتی ژنهای ویروسی در نمونه های خون میباشد. در مطالعه حاضر 120 نمونه خون از گاوهای باسابقه سقطی از گله های مختلف گاوهای شیری صنعتی در اطراف مشهد در مراحل مختلف آبستنی جمع آوری شد. همچنین تعداد 30 نمونه بعنوان کنترل از دامهای سالم (فاقد سابقه سقط جنین) از همان دامداری ها جمع آوری شد. حضور آنتی بادی بر علیه ویروس اسهال ویروسی گاوان در 120 نمونه خون اخذ شده به روش الایزای غیر مستقیم بررسی گردید. از 120 نمونه خون اخد شده، 89 نمونه (%74.16) مثبت بودند. از نمونه های مثبت، 12 نمونه (%13.48) متعلق به سه ماهه اول، 54 نمونه (%60.68) متعلق به سه ماهه دوم و 23 نمونه (%25.84) مربوطه به سه ماهه سوم آبستنی بودند. از 89 نمونه مثبت در آزمایش الایزا، 12 نمونه (%13.48) مربوط به گاوهای با سابقه مرده زایی و 8 نمونه (%8.99) با سابقه جنین مومیایی شده بودند. از 89 نمونه مثبت، 71 نمونه (%79.78) مرتبط با گاوهای بین 2-5 سالگی و 18 (%20.22) مرتبط با گاوهای بالای5 سالگی بودند. در گروه کنترل، 20 نمونه (%66.66) دارای آنتی بادی مثبت بودند. در این آزمایش نیز حضور آنتی ژن اسهال ویروسی گاوان در سرمها نیز بوسیله الایزای ساندویچی مورد ارزیابی قرار گرفت واز 120 نمونه خون اخذ شده، 2 نمونه (67/1%) مثبت بودند که این نمونه ها به سه ماهه دوم آبستنی تعلق داشتند. در گروه کنترل، هیچکدام از نمونه ها دارای آنتی ژن مثبت نبودند. نتایج حاصله نشاندهنده آن است که شیوع عفونت اسهال ویروسی گاوان در بین گاوهای سقطی در مشهد بالاست. هرچند میزان شیوع عفونت در این دامها بیشتر از گروه کنترل می باشد، اما این اختلاف معنی دار نیست.

    به منظور برآورد میانگین آنتی بادی مادری در گوساله های 4-1 ماه علیه ویروس هرپس نوع یک این مطالعه در جمعیت گاوی غیر واکسینه در دو دامداری انجام گرفت. تعداد 113 سرم خون از512 راس گوساله های 1-4 ماهه در استان قزوین گرد آوری گردید. با استفاده از روش الیزای مهاری تعداد 89 سرم مثبت و 24 سرم منفی خون مورد تجسس قرار گرفت. نتایج آنالیز واریانس یک طرفه بیانگر وجود اختلاف معنی دار در تیتر آنتی بادی مهار شده در سنین یک تا چها ماهگی بود. (P<0.001). همچنیین نتایج این مطالعه نشانگر نزولی بودن آنتی بادی نسبت به سن بود بطوریکه از 4/ 84(در صد فاصله اطمینان 95%: 6/90-1/78) در سن یکماهگی به 6/57 (درصد فاصله اطمینان 95%: 2/68-1/47) در سن چهار ماهگی رسید که نزدیک حد آستانه که برابر 55 تعیین شده بود می باشد. همچنیین با استفاده از روش توکی اختلاف معنی داری بین درصد آنتی بادی مهاری در ماه های 1و4 (P<0.001) 2و4 (P=0.034) مشاهده گردید. آزمون مجذور کای برای استقلال حاکی از وجود ارتباط معنی دار بین سن و موارد مثبت سرمی بود (P=0.005).

    باکتری های تولید کننده توکسین شیگا مرتبط با بیماری های سندرم اورمیک و کولیت خونی در انسان است. نمونه های مخاطی رکتوم در گوسفند و گاو بهترین جایگاه ماندگاری این باکتری ها هستند. از 1246 باکتری اشریشیا کلی جدا شده از 782 حیوان بیمار و سالم وجود ژنهای stx1، stx2، eae با روش Multiplex PCR مورد ارزیابی قرار گرفت. در کل 75/9 % از این باکتری ها از 52 راس گاو و 9/7% از 28 راس گوسفند جداشد. ژن stx2 به مراتب بیشتر از stx1 از نمونه های جداشده از گاو وجود داشت، در مقابل در گوسفند برعکس بود. ارتباط آماری معنی داری بین گوسفندان آلوده و تماس با مزارع گاوهای آلوده بدست آمد. وجود گوسفند و گاو بطور همزمان در یک مزرعه توزیع مشابهی از ژنها را نشان دادند. بنابراین وجود گاو و گوسفند با هم به عنوان یک فاکتور خطر محسوب می شود. حیواناتی که در آغل نگهداری می شوند آلودگی بیشتری نسبت به حیوانات مزرعه داشتند. دفع مدفوعی باکتری در حیوان سالم و بیمار یکسان بود. بین گوسفند و گاوهای شیری و فصل ارتباط آماری معنی داری برای میزان آلودگی باکتری بدست نیامد. دفع زیاد مدفوعی مفهوم مهمی در مطالعات اپیدمیولوژی سلامت جامعه، کنترل O157 در کشتارگاه و مزارع و قطع حیوانات آلوده و محافظت زنجیره غذایی دارد.

    پاستورلوزیس یکی از بیماری های مهم اقتصادی در نشخوارکنندگان خصوصا گاو و گاومیش می باشد که بیشتر در اثر پاستورلا مولتوسیدا و گاهی منهمیا همولیتیکا ایجاد می شود. هدف از این تحقیق جداسازی پاستورلا مولتوسیدا از ریه های پنومونیک در گاو و گاومیش های کشتاری کشتارگاه صنعتی ارومیه است. از مجموع 240 ریه پنومونیک در طی 12 ماه تحقیق از ریه6 (% 5/2) مورد پاستورلا مولتوسیدا جداسازی گردید که عواملی همچون نژاد، جنس، سن و فصل را می توان در بروز پاستورلوزیس دخیل دانست. موارد مثبت در گاومیشهای نر دورگه (4 مورد) در فصل زمستان و در نژاد هلیشتاین (2 مورد) در فصل بهار است.

    همافیلوس آنفولانزای تیپ b باکتری است گرم منفی، پلئو مورفیک و مهمترین عامل مننژیت در کودکان زیر 5 سال به خصوص تا2 سال می باشد. در این مطالعه جدایه های مختلف Hib که از بچه های مشکوک به مننژیت جدا شده بودند با استفاده از روش های مختلف میکروبی، بیوشیمیایی و سرولوژیکی تشخیص و مورد شناسایی قرار گرفتند. در این تحقیق محیط های مختلف کشت از جمله محیط مایع بهبود پیدا کرده و تهیه شده(GC medium Base GCB) در آزمایشگاه موسسه رازی ومحیط های تجاری(Brain Heart Infusion BHI) و (TSB) Trypticase Soy Broth برای مقایسه و انتخاب بهترین میزان رشد (bacterial biomass) جدایه هامورد استفاده قرار گرفتند. به تمام محیط های مذکور 2 فاکتور مهم mg/lit 10 X (hemin) و ساپلیمنت IsovitalX حاوی فاکتور V به میزان /ml 01/0 که لازم برای رشد این میکرو ارگانیزم می باشد اضافه گردید. میزان رشد با شروع cfu 104 باکتری در هر لیتراز محیط کشت آغاز گردید. نتایج بدست آمده مقدار حجم رشدی بسیار نزدیکی را در محیط ها نشان داد اگرچه محیط GCB (modified گردیده) میزان رشد بیشتری را نسبت به سایر محیط های تجاری داشت و این محیط برای رشد، استخراج و تخلیص PRP(CPS) استفاده گردید. در این مطالعه تولید PRP بر اساس تکنیک ها و امکانات موجود در بخش از جمله فنل کلروفرم بجای Ultrafiltration برای به حداقل رساندن موادی مانند اندوتاکسین ومواد پروتئینی استفاده گردید. همچنین کاهش بعضی از پروسه های شیمیایی و اضافه نمودن روش هایی از جمله استفاده از آنزیم هایی مانند Pronease و Phospholipase برای از بین بردن پروتئین باقیمانده و مواد فسفاتی در تخلیص PRP صورت گرفته و مقدار PRP حاصله با استفاده از روش بیال تعیین گردید. نتایج این مطالعات نشان داد اگر چه تفاوت چندانی در مقدار PRP بدست آمده در بین سه تا از سویه ها با مقدار متوسط mg/lit 108 مشاهده نگردید، اما سویه ATF2 مقدار PRP بسیاربالایی را (mg/lit 192) نشان داد و همچنین پایین ترین مقدار PRP مربوط به سویه (H.inf1 (16mg/lit بود. به نظر می رسد که داده های بدست آمده از آزمایش می توانند متعلق به یک جامعه نرمال با میانگین 4/106 و انحراف استاندارد 5/62 باشد. بنا به نتیجه ی آزمون یک نمونه ای کولموگروف اسمیرنوف فرضیه فوق تایید شد (P=0.5). حال با مشاهده mg/lit 192≤ PRP مقدار تولید PRP تولید شده قابل توجه می باشد. مقدار پروتئین و نوکلئیک اسید موجود در PRP در طول موج های 280 و 260 نانومتر به ترتیب 10% و 5% تخمین زده شد.

    لاکتوکوکوس گارویه آعامل لاکتوکوکوزیز بیماری نوظهوری است که چندین گونه از ماهی ها را بیمار نموده و باعث خسارات اقتصادی قابل توجهی در آبزی پروری آب شور و آب شیرین می گردد. لاکتوکوکوزیز معمولا در مواقعی از سال که درجه حرارت آب از 15 درجه سانتیگراد بیشتر می شود اتفاق می افتد. این بیماری باعث سپتی سمی فوق حاد و هموراژیک در ماهی ها می گردد. این مقاله روش تولید واکسن تجربی علیه لاکتوکوکوزیز ماهی با توجه به عامل بیماریزا، قدرت بیماریزایی، تولید انبوه، ضریب اطمینان، تاثیر و آزمایش مزرعه ای برای استفاده بصورت غوطه ورسازی آن را مورد بررسی قرار داده است. در آزمایش تاثیر واکسن و به دنبال چالش ماهی های واکسینه با باکتری بیماریزا (107 باکتری در هر میلی لیتر محلول تهیه شده) و زیر نظر گرفتن ماهی ها به مدت 72 ساعت، تنها 10 درصد ماهی های مورد آزمایش بیمار شده و تلف شدند در حالی که در گروه کنترل میزان تلفات 60 درصد بود. در آزمایش مزرعه ای میزان تلفات از زمان واکسیناسیون تا اندازه بازاری در ماهی های واکسینه و غیر واکسینه ثبت شد. تلفات ثبت شده در ماهی ها واکسینه 11 درصد و در ماهی های غیر واکسینه 23 درصد ثبت گردید. این مطالعه نشان داد تلفات در ماهی های واکسینه 50 درصد کاهش یافته است. واکسن تجربی علیه لاکتوکوکوزیز ماهی برای اولین بار در ایران تولید و مورد آزمایش قرار می گیرد.

    انگل های جنس لیشمانیا از جمله تک یاخته های انگلی بوده و مسبب طیف وسیعی از بیماری های عفونی با تظاهرات جلدی، احشائی وجلدی-مخاطی در سراسر جهان و به خصوص کشور های نواحی گرمسیر و از جمله کشور ایران بوده و موارد جلدی و احشائی آن هر ساله از برخی نقاط کشور گزارش می گردد. متاسفانه هنوز دارو یا واکسن موثری جهت این بیماری ایجاد نگردیده است. از ملزومات اولیه جهت تحقیقات واکسن، کشت انبوه انگل می باشد که در این مسیر از انواع محیط ها همراه با سرم جنین گاوی استفاده می شود و از آنجائیکه سرم جنین گاوی فرآورده ای گران قیمت و وارداتی می باشد در این تحقیق تلاش در استفاده از جایگزینی مناسب برای آن بود. با مراجعه به کشتارگاه طیور مقادیر فراوان سرم مرغ های کشتار شده استحصال و در رقت های مختاف در محیط کشت جهت انبوه سازی پروماستیگوت های انگل لیشمانیا ماژور استفاده گردید. نتایج تحقیق حاضرنشان دهنده آن بود که محیط کشت RPMI-1640 غنی شده با 10% سرم مرغ می تواند به عنوان گزینه ای مناسب جهت کشت انبوه انگل ها باشد.

    Designing and producing a proper fusion construction is the most important problem of producing large quantities of a properly folded functional protein. This construction should have all necessary components of a real gene. A good designed fusion gene construction could be cloned into a good and suitable host. Clostridium perfringens is an important pathogen of humans and livestock and produces numerous toxins including epsilon and beta which are responsible for severe diseases. In the present study a new construction containing Clostridium perfringens type D epsilon toxin gene and type B beta toxin gene was designed. At the first step two pairs of primers were used for these genes amplification. At the next step epsilon forward and beta reveres primers were used to produce a chimeric gene containing amplified partial cds of etxD and partial cds of cpbB which are linked together by the AEAAAKEAAAKA fragment as a small linker. The method was based on fusion PCR and using of Pfu DNA polymerase, which has a proofreading activity. The fusion gene inserted into pJET1.2blunt and cloned into E.coli strain TOP10. Based on the latest information, this is the first design and cloning of epsilon-beta fusion gene and also this is the first time that PCR fusion strategy is used for Clostriadial gene fusion, which could be used for development of a recombinant epsilon-beta fusion protein vaccine. This construction also could serve as a model for development and production of novel fusion protein for other potential proteins and toxins.

    Contagious agalactiae (C.A.) is one of the most common disease affecting small ruminants which is caused by Mycoplasma agalactiae. This disease is particularly widespread around the world and Iran is one of the countries that C.A. is present. The aim of this study was isolation and identification of M. agalactiae (MG) with culture and PCR technique in milk samples in Kordestan province, Iran. A total of 367 milk samples were collected from sheep and goat. Specific published primers amplify a 375 bp gene of MG were used for PCR. Twenty (5.5%) out of 367 were positive in PPLO agar and 5 (25%) out of these isolates were positive with Mycoplasma agalactiae primers. Four (75%) out of 5 isolates was from sheep and 1(25%) from goat. Result of PCR with 367 milk samples showed that 11(3%) of them were positive with these primers. The isolation of M. agalactiae showed that C.A is present in Kordestan province and our results suggested that PCR method because of reduces the time consuming could be an alternative method beside culture.

    The measurement of antibody responses of animals exposed to BVDV either through a natural exposure or an immunization protocol is still a standard procedure. For BVDV, the test formats have been largely limited to ELISA which is a valuable diagnostic test to measure the level of BVDV specific antibodies as well as antigen in blood samples. In the present study, 120 blood samples were collected from the cows with the history of abortion in different period of pregnancy from different industrial dairy cattle herds of Mashhad area of Iran. Also 30 samples were collected from the cows with no history of abortion as control. The presence of antibody against BVDV from the 120 serum samples was investigated by indirect ELISA. From 120 serum samples which were collected from aborted cows, 89 samples were positive (%74.16). From these positive samples, 12(13.48%), 54 (60.68%) and 23 (25.84%) samples belong to the first, second and third trimester of pregnancy, respectively. From 89 positive samples, 12 (13.48%) samples were related to stillbirth and 8 (8.99%) samples were belongs to the mummified fetus. From 89 positive samples, 71 (79.78%) were related to cattle between 2-5 years old and 18 (20.22%) were associated to cattle more than 5 years old. In control group, 20 samples (66.66%) were antibody positive. Also the presence of BVDV antigen in serum samples was investigated by Ag-capture ELISA. From 120 serum samples, 2 samples were positive (1.67%), which belongs to the second period of pregnancy. In control group, none of the samples were antigen positive. The results of this study showed that the prevalence of BVDV infection is high among the aborted cows of Mashhad area. Although this prevalence is higher than the control group, the observed difference is not significant.

    In order to estimate the mean of maternal antibody in calves against BHV-1 (Bovine Herpes virus type 1), this study was carried out in a population of calves from non-vaccinated dairy cattle at 2 livestock in Qazvin province. One hundred thirteen sera out of 512 were collected from 1-4 months unvaccinated calves. We used Blocking –Percentage of maternal antibodies against BHV-1, which obtained by Blocking ELISA assay in 1-4 months calves sera. The result of one way analysis of variance determined that there was a significant difference among blocking percentage of maternal antibodies against BHV-1 in 1-4 months (P<0.001). Comparing percentage of mean titer indicated a decreasing trend with respect to age i.e. from 84.4(a 95% CI: 78.1- 90.6) in 1 month to 57.6(a 95% CI: 47.1- 68.2) in 4 months, which was near to 55 as cut off point. Tukey's method showed a significant difference between the Blocking percent of mean titer between 1 and 4(P<0.001), 2 and 4 (P=0.034) months. Chi-square test for independency showed a significant association between age and seroactivity (P=0.005).

    Shiga toxin producing Escherichia coli have been associated with HUS, HC and TTP in human. We found recto-anal mucosal sample in sheep as well in cattle is the main site for E. coli O157 localization. 1246 E. coli isolates from 872 both healthy and diarrheic animals were analyzed, by screening for the presence of Shiga toxin-producing (VT 1 and VT 2) and intimin (eae) genes used Multiplex PCR. 87(9.75%) VTEC from 52 cattle and 28(7.90%) from 28 sheep were isolated. VT2 gene was found to be more frequent than VT1 in cattle (54.02% vs 26.43%), in contrast the same genes in sheep (21.42%vs25%). There was observed significant difference in the origin of VT positive sheep in close contact with farms of cattle origin. Having cattle and sheep with each other was a possible risk factor. The animal was kept in pen was more localized than tethered. Young cattle were documented strongly significant high prevalence rate in E. coli O157:H7 than older, but no effect of age was observed on the occurrence of E. coli O157 in sheep. Both diarrheic and healthy animals were shed E. coli O157:H7 in their feces. Sheep and dairy cow were not illustrated any significance differences geographical region and seasonal variation with E. coli O157:H7 prevalence rate.

    Pasteurellosis is one of the important economic diseases in ruminants, especially in cows and buffaloes. It is caused by Pasteurella multocida and occasionally by Mannheimia haemolytica. The aim of this study was to isolate Pasteurella multocida from lungs with probable mentioned bacterial agents in cows and buffaloes in Urmia's slaughter house. 240 lung samples over a period of 12 months were cultured. The results have revealed 6 (2.5 %) Pasteurella multocida results suggest that the animal, its breed, sex, age and season can be effective in the occurrence of these positive cases. The positive samples were all from male beef cattle of hybrid breeds (4 samples) in winter and Holstein breeds (2 samples) in spring.

    Heamophilus influenzae type-b (Hib) is a gram - negative pleomorphic bacterium that causes meningitis infections in children with the age of less than 5 years particularly in two years old infants. In the present study various isolates of Heamophilus influenzae from infants suspected to meningitis were collected, identified, characterized and were used throughout our experiments. Different culture media namely Brain Heart Infusion Broth (BHIB), Tryptic Soy Broth (TSB) and GC medium Base (GCB) which this medium was modified and prepared in our own laboratory, were compared to determine the highest bacterial yield. All media were added supplements 10mg/ml hemin & 0.01/ml IsovitaleX containing V factor. The bacterial yield for all Hib strains present in our laboratory were measured with an initial inoculums of 104 cfu per ml. The result showed very closed amount of biomass for all isolates however, GCB had slightly higher yield and ultimately we chose this medium for cultivation and extraction of capsular polysaccharide (CPS-b). In our laboratory we have adapted the PRP production according to our technical and instrumental availabilities which exists in our laboratories replacing ultra centrifugation to phenol chloroform to remove contaminants like endotoxin and proteins to the minimum level and also decreased the number of some chemical treatments while some steps were added in purification process. Our study showed although there were not significant differences between the PRP extract of the three isolates with average amount of 108 mg/lit, however, isolate ATCC10210 (ATF2) showed the highest amount with 192mg/lit and the least PRP was produced by isolate H.inf.1, with 16 mg/lit. It seems that the data can be categorized to a normal distribution with the mean of 106.4 and standard deviation of 6.25. This result was confirmed by one sample kolmogorov-Smirnov test, hence the PRP ≥192 mg/lit is statistically significant at a significant level of α =0.10 (P=0.085). The amount of contaminants e.g protein along with nucleic acid present in PRP was estimated at optical density 280 and 260nm, were under 10% and 5% respectively.

    Lactococcus garvieae is the etiological agent of lactococcosis, an emerging disease which affects several fish species and causes important economic losses both in marine and freshwater aquaculture. Lactococcosis usually happens when water temperature increases over 15°C during the year. Normally, it causes a hyperacute and haemorrhagic septicemia in fish. This paper presents a procedure for producing experimental vaccine for rainbow trout (Oncorhynchus mykiss) lactococcosis including aspects such as pathogen characterization, pathogenicity, mass cultivation, safety, potency and field trial tests for immersion use. In the potency test, after challenging the vaccinated fish with live pathogenic bacteria (1×107 bacteria per milliliter of immersing solution) and observing for 72 hours thereafter, 10% of fish died while the control group showed 60% mortality within the observation time. In the field trial from vaccination time onward till marketing of the fish, those mortalities that occurred in groups of vaccinated and non-vaccinated fish were recorded. Total death occurred in the vaccinated group was 11%, while in non vaccinated group this number was approaching 23%. This observation indicates a 50% reduction in mortality in the vaccinated group. This is the first report on experimental vaccine against lactococcosis in fish that is produced and tested in Iran.

    Genus Leishmania is a protozoan parasite that causes different severe diseases. Fetal Calf Serum (FCS) is the major part of the Leishmania culture media, for mass cultivation and the most expensive ingredient in these media. In the present work, the efficacy of chicken serum was evaluated in Leishmania culture media. The results indicated that, the (10%) chicken serum enriched culture medium supported the growth of large scale cultures of the parasites and can be used for primary and mass cultivation of Leishmania parasites. In conclusion, the chicken serum was effective, easy available and cheap replacement for FCS.
