    در حالی که بروسلوزیس در کشورهای توسعه یافته و پیشرفته تقریبا ریشه کن شده است، در کشورهای در حال توسعه از نواحی مدیترانه تا شبه قاره هندوستان و کشورهای عربی سبب مشکلات جدی بهداشتی می شود که این موضوع در بررسی گزارش ها بسیار مشاهده می شود. بیش از صد سال است که روش های متعدد سرولوژیک در تشخیص این بیماری معرفی و به کارگیری شده اند، اما هیچ یک قادر نبوده اند تا به حال نتایج رضایت بخشی را فراهم کنند لذا پزشکان پیوسته در موارد مذکور براساس علائم بارز کلینیکی اقدام به درمان بیماران با رژیم های درمانی گوناگون و بعضا غیراختصاصی می کنند که حداقل نتیجه آن شکست درمان و عود بروسلوزیس است. در این مطالعه کوشش شده است با بررسی اجمالی عوامل موثر در تشخیص سرولوژیک بیماری، مشکلات کنونی بررسی شده و راهکارهایی برای بهبود وضعیت تشخیصی بروسلوزیس به ویژه برای آزمایشگاه های بومی پیشنهاد شود.

    زمینه و هدف
    ویتامین K2 به عنوان یکی از ویتامین های محلول در چربی، می تواند در برخی موارد آثار مهاری مناسبی بر فعالیت استافیلوکوکوس اورئوس مقاوم به متی سیلین داشته باشد. هدف از این مطالعه تعیین غلظت ویتامین K2 بر میزان بیان ژن های mecA و blaZ در ایزوله های استافیلوکوکوس اورئوس مقاوم به متی سیلین است.
    مواد و روش کار
    76 ایزوله بالینی استافیلوکوکوس اورئوس مقاوم به متی سیلین (MRSA) با تست های فنوتیپی از نمونه های بالینی مختلف (خون، ادرار، زخم و غیره) جداسازی شد. برای تیمار ایزوله های MRSA، از غلظت های 1، 10، 50، 100، 200، 300 و 500 میکروگرم بر میلی لیتر از ویتامین K2 و بازه های مختلف زمانی، استفاده شد. عملکرد ویتامین در زمان های 24، 48 و 72 ساعت گرمخانه گذاری، بررسی شد. برای سنجش کمی ژن ها از روش Real-time PCR استفاده شد. همچنین به منظور آنالیز نتایج به دست آمده از نرم افزار REST 2008 V3 و SPSS V16 استفاده شد.
    یافته ها
    میزان بیان ژن های blaZ و mecA تنها در غلظت 500 میکروگرم بر میلی لیتر از ویتامین 2K کاهش داشت. علاوه بر این، زمان های گرمخانه گذاری 48 و 72 ساعت بهترین اثر عملکردی را از نظر تاثیر ویتامین بر ژن های مورد مطالعه داشت. در ایزوله های جدا شده از نمونه های بالینی زخم و ادرار کاهش بیان ژنهای mecA و blaZ بیشترین مقدار را به خود اختصاص داد. همچنین، ارتباط معنی داری بین زمان گرمخانه گذاری و نوع نمونه بالینی بر کاهش بیان ژن های مورد نظر، مشاهده شد (0/01> p) و (0/05> p).
    نتیجه گیری
    ویتامین K2 می تواند نقش مهمی در کنترل سویه های استافیلوکوک اورئوس مقاوم به متی سیلین داشته باشد.

    زمینه و هدف
    استافیلوکوکوس اورئوس پاتوژن فرصت طلبی است که به ویژه سویه های مقاوم به متی سیلین آن، عامل طیف وسیعی از عفونت های بیمارستانی و اکتسابی از جامعه است. قدرت چسبندگی از عوامل مهم افزایش بیماری زایی این باکتری است. در پژوهش حاضر، میزان مقاومت به متی سیلین و فراوانی ژن های چسبندگی fnbA و fnbB در جدایه های بالینی استافیلوکوکوس ارئوس بررسی شد.
    مواد و روش کار
    جدایه های استافیلوکوکوس ارئوس از نمونه های بالینی بیماران مراجعه کننده به آزمایشگاه های تشخیص پزشکی رشت در بازه زمانی یک ساله شهریور 95-94 جداسازی و مقاومت جدایه ها به متی سیلین به روش انتشار دیسک و ارزیابی حضور ژن mecA بررسی شد. فراوانی ژن های چسبندگی fnbA و fnbB در سویه های حساس و مقاوم به متی سیلین در واکنش PCR با استفاده از پرایمر اختصاصی این ژن ها مشخص و با آزمون 2% مقایسه شد.
    یافته ها
    از 90 جدایه بررسی شده، 37 جدایه (41%) مقاوم به متی سیلین و mecA مثبت بودند. همچنین در 59 (65/5%) و 37 (43/3%) جدایه به ترتیب ژن های fnbA و fnbB شناسایی شدند. فراوانی ژن های fnb در سویه های مقاوم به متی سیلین استافیلوکوکوس اورئوس به گونه ای معنی دار بیشتر از سویه های حساس به متی سیلین بود (0/05> P).
    نتیجه گیری
    نتایج این مطالعه بیانگر شیوع بالای سویه های مقاوم به متی سیلین در نمونه های بالینی استافیلوکوکوس اورئوس در رشت و نیز فراوانی ژن های fnb در این جدایه ها است.

    زمینه و هدف
    با وجود اینکه روش های مختلفی برای تولید نانوذرات وجود دارد، روش تولید بیولوژیکی نانوذرات به دلیل خواص دوستدار طبیعت آن و صرفه جویی در انرژی، توجه محققان را به خود جلب کرده است. در مطالعه حاضر تولید بیولوژیکی نانوذرات طلا به وسیله سویه قارچ Fusarium oxysporum انجام و سمیت نانوذرات به دست آمده در کشت سلولی بررسی شد.
    مواد و روش کار
    Fusarium oxysporum خریداری و در محیط کشت سابوراد دکستروز براث (SDB) کشت داده شد. سوپرناتانت کشت در معرض محلول کلرواوریک اسید در غلظت نهایی 1 میلی مولار قرار گرفت. پس از تولید نانوذرات، محلول تغییر رنگ داده نانوذرات برای بررسی با اسپکتروفوتومتر و TEM استفاده شد و نانوذرات به دست آمده شست وشو و استریل شدند. در نهایت دو رده کشت سلولی CIRC-HLF به عنوان رده طبیعی و SW 872 به عنوان رده سرطانی برای آزمونMTT استفاده شدند.
    یافته ها
    رنگ سوپرناتانت قارچ پس از 24 ساعت از زرد به قرمز تغییر پیدا کرد. عکس های TEM نشان داد که نانوذرات کروی و چند وجهی با ابعاد در حدود50-70 نانومتر هستند. آزمون MTT و آزمون کشت سلولی نشان داد که نانوذرات طلای تولیدی هنگامی که در غلظت های بالا استفاده شود اثر سمی دارد و کشت سلولی رده CIRC-HLF به غلظت های بالا از نانوذرات طلا نسبت به رده سلولی SW 872 مقاوم تر بودند.
    نتیجه گیری
    به نظر می رسد که تولید نانوذرات طلا بیولوژیک با Fusarium oxysporum انجام شدنی است و می توان آن را به راحتی تولید، شست وشو و استریل کرد. این نانوذرات می توانند در شرایط درون بدن هم استفاده شوند.

    زمینه و هدف
    خوراک آبزیان از اجزای مختلفی تشکیل شده است که کیفیت نامطلوب هریک از آنها بر کیفیت گوشت تولید شده تاثیرگذار است. قارچ ها از عوامل فساد و تولید سم درموادغذایی هستند. در بین سموم قارچی آفلاتوکسین ها اهمیت زیادی دارند. این سم قابلیت آن را دارد که از خوراک به ماهی و سپس انسان منتقل شود. هدف از این پژوهش تعیین میزان آفلاتوکسین در خوراک و همچنین شناسایی فلور قارچی آن است.
    مواد و روش کار
    در این مطالعه 8 مزرعه پرورش ماهی قزل آلای رنگین کمان واقع در شهرستان دامغان برای بررسی فلور قارچی و اندازه گیری سطح آفلاتوکسین کل سنجش شدند. از انبار 8 مزرعه پرورش ماهی، 500 گرم نمونه خوراک کارخانه ای (پلت) به صورت تصادفی تهیه شد. فلور قارچی با روش کشت و کشت روی لام، جداسازی و شناسایی شدند. آفلاتوکسین کل با استفاده از روش الیزا اندازه گیری شد.
    یافته ها
    در مجموع 11جنس (آسپرژیلوس، فوزاریوم، پنسیلیوم، موکور، اسکوپولاریوپسیس، کرایزوسپوریوم، ریزوپوس، آبسیدیا، الوکلادیوم، آلترناریا، سودالشریا) شناسایی گردید. جنس ریزوپوس و آسپرژیلوس به ترتیب با 20/38% و 18/47% بالاترین میزان فراوانی را داشتند. در بین گونه های آسپرژیلوس، آسپرژیلوس فلاووس با 34/48% بالاترین و اکراسئوس با 10/34% پایین ترین فراوانی را دارا بودند. آ. نایجر نیز با 7 مورد گزارش،24/14% از فراوانی را به خود اختصاص داد. کمترین فراوانی با 1/28% متعلق به جنس کرایزوسپوریوم بود. میانگین میزان آفلاتوکسین کل ppb 5/36 در دامنه ppb 11/64-2/03 بود.
    نتیجه گیری
    میزان آفلاتوکسین در تمام نمونه های بدست آمده از پلت مصرفی قزل آلا، کمتر از میزان مجاز (ppb20) بود.

    زمینه و هدف
    افلاتوکسین ها گروه بزرگی از مایکوتوکسین ها و متابولیت های ثانویه از گونه های قارچ آسپرژیلوس هستند. aflR گروهی از ژن های مداخله کننده در نسخه برداری و تولید آفلاتوکسین در قارچ های آسپرژیلوس هستند. در این مطالعه هویت مولکولی قارچ آسپرژیلوس مولد سم افلاتوکسین در پسته های تازه شهرستان سیرجان بررسی شد.
    مواد و روش کار
    تعداد 100 نمونه پسته آلوده به قارچ از انبارهای پسته در شهرستان سیرجان به روش هدفمند اخذ شد. نمونه ها به محیط کشت سابورو دکستروز براث منتقل شدند. کنترل مثبت استفاده شده در این پژوهش، سویه قارچ آسپرژیلوس مولد سم افلاتوکسین و کنترل منفی سوش قارچی غیر آسپرژیلوس و آسپرژیلوس های غیر آفلاتوکسیک بودند. استخراج DNA با استفاده از کیت و از PCR برای شناسایی ژن aflR در نمونه های آلوده به قارچ انبارهای پسته استفاده شد.
    یافته ها
    از بین 100 نمونه مغز پسته تازه که به صورت هدفمند انتخاب شده بودند، 10 نمونهمورد سالم 64 نمونه آلوده به قارچ های آسپرژیلوس، 18 نمونه آلوده به قارچ پنیسلیوم، 1 مورد موکور، 5 نمونه ساپروفیت و 2 مورد آلودگی قارچی ناشناخته گزارش شد. از بین آسپرژیلوس ها فقط در 7 مورد ژن تنظیمی مولد افلاتوکسین حضور داشت.
    نتیجه گیری
    در بسیاری از کشورها آلودگی پسته به آفلاتوکسین به مقادیر مختلف وجود دارد، که می تواند تهدید کننده ی سلامت جامعه باشد. . بهترین اقدام برای جلوگیری از پیامدها و کاهش آفلاتوکسین در پسته، نظارت و کنترل بر تولید، نگهداری و توزیع آن ها و تشخیص آلودگی توسط تکنیک های دقیق مولکولی است. هم چنین ایجاد شرایط ایده آل برای به حداقل رساندن آلودگی امری لازم برای سلامت مصرف کنندگان است.

    زمینه و هدف
    ویروس پاپیلوم انسانی (HPV) یکی از مهمترین مشکلات بهداشت عمومی و عامل اصلی سرطان دهانه رحم است. عفونت HPV در کشورهای در حال توسعه با منابع مالی کم، شیوع زیادی دارد. این مطالعه با هدف تعیین فراوانی نسبی ژنوتیپ های HPV و خصوصیات مرتبط با آن در زنان مراجعه کننده به یکی از بیمارستان های عمومی شهر تبریز انجام شده است.
    مواد و روش کار
    این مطالعه مقطعی روی 400 زن با نمونه پاپ اسمیر، مراجعه کننده به یک بیمارستان عمومی در تبریز از شهریور 1394 تا شهریور 1395 انجام شد. شناسایی ژنوتیپ 28 ویروس پاپیلومای انسانی(HPV) با استفاده از روش PCR انجام شد.
    یافته ها
    عفونت HPV مثبت در 155 زن (38/75%) در محدوده سنی17 تا 85 سالگی تشخیص داده شد. شایع ترین نوع، 16HPV- (19/1%) بود که به دنبال آن 39- HPV (12/5%) و 18-HPV (8/9%) بود. بالاترین میزان عفونت HPV در سن 36 سالگی (7/7%) مشاهده شد. سطح تحصیلات و وضعیت اقتصادی هر زن نشان داد که بیشتر زنان مبتلا به HPV دیپلم (34/6%) داشته و وضعیت اقتصادی متوسطی (67/9%) دارند. 60/9% از این زنان خانه دار بودند.
    نتیجه گیری
    تعیین ژنوتیپHPV و عوامل خطر مرتبط با عفونتHPV در هر منطقه جغرافیایی می تواند منجر به تولید واکسن های موثر بر ویروسHPV شود. همچنین می تواند برای مدیریت بیماری و تشخیص حساسیت بالای نئوپلازی داخل اپیتلیال سرویکس مفید باشد.

    زمینه و هدف
    بیماری عروق کرونری قلب (Coronary Artery Disease) یا آترواسکلروزیس عامل اصلی مرگ و میر در جهان است. عوامل عفونی مثل ویروس هرپس سیمپلکس (HSV) ممکن است در بیماری‏زایی آترواسکلروزیس دخالت داشته باشند. هدف از این مطالعه بررسی سطح آنتی بادی های IgG اختصاصی HSV در افراد مبتلا به بیماری عروق کرونری قلب و بررسی میانگین پروتئین فاز حاد با حساسیت بالا (High Sensitivity- C Reactive Protein) در این بیماران بود.
    مواد و روش کار
    در این تحقیق 40 بیمار مبتلا به آترواسکلروزیس که بیماری آنها با آنژیوگرافی ارزیابی شده بود به همراه 40 فرد سالم به عنوان گروه شاهد آزمایش شدند. سنجش hs-CRP با روش نفلومتری و آنتی بادی های IgG اختصاصی ویروس با روش الایزا بررسی شد.
    یافته ها و
    نتیجه گیری
    نتایج نشان داد که میانگین سطح hs-CRP در بیماران در مقایسه با گروه شاهد به طوری معنی دار بالاتر بود (0/05>P). درصد بالاتری از بیماران دارای IgG اختصاصی HSV(78/4 %) در مقایسه با گروه شاهد (21/6%) بودند. ارتباط معنی داری بین سرولوژی مثبت HSV با CAD مشاهده شد. میانگین hs-CRP در افراد با سرولوژی مثبت HSV، نسبت به افراد با سرولوژی منفی به طور معنی داری بالاتر بود. در مجموع سرولوژی مثبت HSV همراه سطوح بالای hs-CRP ممکن است یک فاکتور خطر برای آترواسکلروزیس باشد.

    Brucellelosis has been almost eradicated in developed countries while in under developed areas from Mediterranean regions to Arabic countries and even in Indian subcontinental can cause serious health problems. This point can be found out easily in released reports. It is over a century that various serological methods have been introduced and applied for diagnosis of brucella infection, but all were not able to provide satisfactory results. Therefore, clinicians treat the patients with various unspecific treatment protocols based on clinical signs and symptoms, consequently causing treatment failure or possible relapse later on. This article aimed to have short survey on serological diagnosis of brucellosis and proposed an approach at present situation of brucellosis diagnostic specifically for local laboratories.

    Background And Aims
    As one of the fat-soluble vitamins, vitamin K, can in some cases have intrinsic inhibitory effects on the activity of methicillin-resistant Staphylococcus aureus. The aim of this study, was to determine the effect of vitamin K2 on the expression of mecA and blaZ genes in Multi Drug Resistant Methicilin Resistance S. aureus isolates.
    Materials And Methods
    76 clinical isolates of methicillin-resistant S.aureus were isolated by phenotypic tests. For treatment of isolates, concentrations of 1, 10, 50, 100, 200, 300 and 500 μg / ml of vitamin K2 and different time intervals were used. The q-PCR method was used to quantitatively evaluate the genes. It was also used to analyze the results of REST 2008 V3 and SPSS 16 software.
    Results
    The expression of blaZ and mecA genes was reduced only at a concentration of 500 μg / ml of vitamin K2. Additionally, the 48 and 72 hours incubation times had the best effect on the effect of vitamins on the genes studied. In the isolates from clinical specimens, ulcers and urine reduced the expression of mecA and blaZ genes. Also, there was a significant relationship between incubation time and clinical specimen type on decreasing the expression of the desired genes (P≤0.01, P≤0.05).
    Conclusions
    In accordance to the results of this study, the use of vitamin K2 can play an important role in the control of methicillin-resistant Staphylococcus aureus strains.

    Background And Aims
    Staphylococcus aureus is an opportunistic pathogen, which particularly its methicillin resistant strains, is responsible for a wide range of hospital and community acquired infections. Adhesion ability is one of the important virulence factors of this bacterium. In this study, the resistance to methicillin and the frequency of fnbA and fnbB adhesion genes in clinical isolates of S. aureus were investigated.
    Materials And Methods
    Isolates of S. aureus were collected from clinical samples of patients referred to Rasht medical diagnostic laboratories. Resistance of isolates to methicillin was investigated by disk diffusion method and determination of presence of mecA gene. Frequency of fnbA and fnbB adhesion genes in methicillin sensitive and methicillin resistant strains was determined using specific primers of these genes in PCR reaction and was compared using chi-square test.
    Results
    Out of 90 isolates, 37 isolates (41%) were resistant to methicillin and mecA positive. Also, in the PCR reaction, fnbA and fnbB genes were identified in 59 (65.5%) and 37 (43.3%) isolates, respectively.     Conclusions
    The results of this study indicate high prevalence of methicillin-resistant strains in clinical isolates of S. aureus in Rasht and the frequency of fnb gene in these isolates.

    Background And Aims
    Although there are several ways of nanoparticles production, however the biological method of nanoparticles production is under attention of researchers due to its eco friendly and energy saving properties. In the present study biosynthesis of gold nanoparticle was done by Fusarium oxysporum fungus strain and the cytotoxicity of the obtained nanoparticles was evaluated in the cell culture.
    Materials And Methods
    Fusarium oxysporum was purchased and cultured in Sabouraud dextrose broth medium. The culture supernatant was subjected to chloroauric acid solution at the final concentration of 1 mM. After nanoparticles production, the color changed reaction mixture was used for characterization with spectrophotometry, XRD and TEM microscopy. The obtained nanoparticles were washed and sterilized. Finally two cell lines (CIRC-HLF as normal and SW 872 as cancerous cell lines) were used for 3-(4,5 dimethyl thiazol-2yl)-2,5 diphenyl tetrazolium bromide (MTT) assay.
    Results
    The fungal supernatant color after 24 hours was changed from yellow to red. TEM images have shown the nanoparticles were spherical or hexagonal and their sizes were around 50-70 nanometer. MTT assay and cell culture assay showed that the produced GNPs had toxic effect when they were used at high concentrations and the CIRC-HLF cell line was more resistant than SW 872 one against the higher concentration of GNPs.
    Conclusions
    It seems that the biological production of gold nanoparticles by Fusarium oxysporum can be done and can be easily produced, washed, sterilized and can be used in vivo.

    Background And Aims
    Aquatic feed consists of different components; the undesirable quality of each of them affects the quality of the meat produced. Fungi are agents of contamination and poison production in food. Aflatoxins are of particular importance among the toxins of fungi. This poison has the ability to transfer through feeds to fish and then humans. The aim of this study was to determine the amount of aflatoxin in the feed and to identify the fungal flora.
    Materials And Methods
    In this study, 8 rainbow trout farms in Damghan city were evaluated for the assessment of fungal flora and total aflatoxin levels. 500 g sample of feed (pellet) was randomly prepared from the storeroom of the 8 fish farms. Fungi of Aspergillus Flavous and also fungal flora were isolated and identified by culture and slide culture method. Total aflatoxin measurement was performed using ELISA method.
    Results
    A total of 11 genera (Aspergillus, Fusarium, Penicillium, Mucor, Scopulariopsis, Chrysosporium, Rhizopus, Absidia, Ulocladium, Alternaria, Pseudallescheria) were identified. The genera of Rhizopus and Aspergillus were the highest with 20.38% and 18.47% respectively. Among Aspergillus species, Aspergillus flavus was the highest with 48.44% and A. ochraceus had the lowest incidence of 10.34%. A. niger was also reported with 24.74% of the reported cases. The lowest frequency was found in the genus Chrysosporium with 1.28%. The average of total aflatoxin concentration was 5.36 ppb in the range of 2.03-11.64.
    Conclusions
    The aflatoxin content in all samples from Rainbow trout consumption was less than the permitted level (20 ppb).

    Background And Aims
    Aflatoxins are a large group of mycotoxins and secondary metabolites of Aspergillus fungal species. aflR The interfering genes group in the transcription and production of aflatoxin plays an important role in Aspergillus fungi. In this study, the molecular identity of aflatoxigenic Aspergillus spp. in fresh pistachios in Sirjan city was investigated.
    Materials And Methods
    In This study 100 samples of fungal infected pistachio were purposefully collected from pistachio warehouses in Sirjan city. Samples were transferred to the sabouraud dextrose broth medium. The positive control used in this study were generator genes of aflatoxin in Aspergillus strains and negative control were non Aspergillus fungal strains and non aflatoxic Aspergillus. DNA extraction were performed by kit and PCR method was used for identification of AflR gene in samples of fungal infected pistachio warehouses for confirmation of the presence of the generator genes of aflatoxin.
    Results
    Molecular results showed that among 100 samples of fresh pistachio nuts that were purposefully selected, there were 10 healthy samples, and in 64 Aspergillus fungi ones, 18 samples contained pencillium, 1 sample mucor, 5 sample Saprophytic and 2 samples contained unknown fungal infection. Among Aspergillus, only in 7 cases were regulatory gene of aflatoxin.
    Conclusions
    By studying this subject, factors affecting on ways to reduce aflatoxin can be found. Since most countries are sensitive on the issue of aflatoxin in terms of health, this research can play an important role in the ways of increasing exports to countries.

    Background And Aims
    Human papillomavirus (HPV) is one of the major public health problems and the main causes of cervical cancer. The prevalence HPV infection in developing countries with low financial resources is high. This study aimed to determine the relative frequency of HPV genotypes and its sociodemographic characteristics in women referred to a general hospital in Tabriz, Iran from 2015-2016.
    Materials And Methods
    This cross-sectional study was performed in 400 women with Pap smear samples, referring to to a general hospital in Tabriz, Iran from 2015- 2016. The detection of 28 HPV genotypes was performed by using the PCR technique. The sociodemographic survey was conducted for each HPV positive woman.
    Results
    HPV-positive infection was detected in 155 (38.75%) women aged 17-85 years. HPV 16 (19.1%) was the most prevalent type, followed by HPV 39 (12.5%) and HPV 18 (8.9%). The highest rate of HPV infection was observed at the age of 36 years (7.7%). The level of education and economic situation of each woman were showed most of HPV-positive women had a high school diploma (34.6%) and average economic situation (67,9%). 60.9% of these women were a housewife, and 67.3% lived in the capital .
    Conclusions
    Determination of HPV genotype and risk factor related to HPV infection in each geographical region can lead to the production of effective vaccines against the HPV virus. It can also be useful for disease management and high sensitivity diagnosis of cervical intraepithelial neoplasia.

    Background And Aims
    Coronary artery disease (CAD) is a major cause of mortality worldwide. Infectious agents such as herpes simplex virus (HSV) may be implicated in the pathogenesis of atherosclerosis. The aim of this study was to survey the prevalence of IgG antibodies against HSV in patients with coronary artery disease and to determine the mean of high-sensitivity C-reactive protein(hs-CRP) in these patients.
    Materials And Methods
    40 patients being evaluated for atherosclerosis by coronary angiography along with 40 healthy individuals as a control group were examined in this research. Assessment of hs-CRP was carried out by nephelometry and virus-specific IgG antibodies by Enzyme-linked Immunosorbent Assay (ELISA).
