    مقدمه و هدف
    تولیدآنزیم بتالاکتامازهای وسیع الطیف (Extended spectrum beta lactamase، ESBL) امروزه به عنوان یک تهدید عمده در سراسر جهان با توجه به درمان های محدود می باشد. این مطالعه جهت بررسی الگوهای حساسیت آنتی بیوتیکی و بررسی حضور ژن TEM در ایزوله های اشریشیاکلی و کلبسیلا پنومونیه جداسازی شده از نمونه های مختلف بالینی انجام شد.
    روش بررسی
    نمونه های مختلف بالینی در طول سال 90 از مراکز درمانی دانشگاه علوم پزشکی شهید بهشتی جمع آوری و جهت تشخیص آن به آزمایشگاه دانشگاه فرستاده شد؛ برای شناسایی مقاومت آنتی بیوتیکی از روش دیسک دیفیوژن و برای تعیین فراوانی محصولات ژن TEM از روش PCR با استفاده از پرایمرهای اختصاصی آن استفاده شد.
    یافته ها
    از میان 50 ایزوله کلبسیلا پنومونیه تعداد 17 ایزوله (34%) تولیدکننده بتالاکتامازهای وسیع الطیف ESBL و دارای ژن بتالاکتاماز TEMبودند، مقاومت به سفوتاکسیم90 درصد، بیشترین مقدار و کمترین درجه مقاومت مربوط به ایمی پنم 4 درصد گزارش شدند. در میان 50 سوش اشرشیاکلی از نظر مقاومت آنتی بیوتیکی، مقاومت به سفکسیم 90 درصد بیشترین مقدار و مقاومت به مروپنم 6 درصد کمترین مقدار گزارش شد. در بین سوش های اشرشیاکلی7 مورد (14%) تولیدکننده بتالاکتامازهای وسیع الطیف ESBLو دارای ژن بتالاکتاماز TEMبودند.
    نتیجه گیری
    با توجه به شیوع بالای مقاومت به داروهای بتالاکتام در بین باکتری های مورد مطالعه، انجام دقیق تست آنتی بیوگرام جهت درمان مناسب توصیه می گردد.

    سابقه و هدف
    مطالعات در سرطان سینه بیان کننده حضور جمعیتی از سلول ها با ویژگی های سلول های بنیادی میباشد که نقش مهمی در اغاز، پیشرفت و مقاومت به درمان های جاری دارد.از انجاییکه کشت سه بعدی در شرایط ازمایشگاهی محیط مناسبی برای مطالعه سلول های بنیادی سرطانی در مقابل محیط پیچیده بافتی ایجاد می کند.هدف ما در این مطالعه بررسی اثر کشت سه بعدی بر ویژگی های سلول های بنیادی سرطانی می باشد.
    مواد و روش ها
    سلول های MDA-MB468 که به صورت سه بعدی و دو بعدی کشت داده شده اند از نظر قدرت ایجاد کلنی،اسفروئید،بیان مارکر های سطحی بنیادینگی(CD44،CD133)توسط فلوسایتومتری، و همینطور بیان ژن های بنیادینگی(ALDH،Sox2،Nanog،Oct4،c-Myc) توسط Real time PCR مورد بررسی قرار گرفتند.
    یافته ها
    سلول های MDA-MB468 که به صورت دو بعدی و سه بعدی کشت داده شده اند تفاوت معناداری از نظر قدرت کلنی زایی و ایجاد اسفروئید نشان ندادند. کشت سه بعدی سبب ایجاد کاهش معناداری در بیان مارکر CD133+ نسبت به کشت دو بعدی شد.(5.84±52.69% در کشت سه بعدی در مقابل 0.34±98.55% در کشت دو بعدی) در حالیکه تفاوت معناداری در بیان CD44+ در این دو نوع کشت مشاهده نشد.همینطور کشت سه بعدی سبب سرکوب بیان ژن های بنیادینگی شامل oct4،nanog،ALDH،sox2 نسبت به کشت دو بعدی شد.
    نتیجه گیری
    در مجموع نتایج حاصل از پژوهش انجام شده موید این میباشد که کشت سه بعدی که منعکس کننده معماری بافتی و شرایط In vivo میباشد سبب کاهش ویژگی های سلول های بنیادی سرطانی در رده سلولی MDA-MB468 میشود.

    سابقه و هدف
    اطلاع از روابط فیلوژنتیک و تنوع ژنتیک در مرکبات جهت تشخیص روابط ژنتیکی، شناسایی ژرم پلاسم و ثبت ارقام جدید، امری مهم و ضروری می باشد. در کلکسیون های مرکبات کشور، تعدادی بیوتیپ وجود دارد که شناسایی آنها عمدتا بر اساس صفات مرفولوژی بوده و تحقیقات ژنتیکی چندانی بر روی آنها انجام نشده است. نشانگرهای ملکولی می توانند در این زمینه مفید باشند.
    مواد و روش ها
    به منظور بررسی تنوع ژنتیکی و روابط خویشاوندی بین 45 ژنوتیپ از ارقام ناشناخته و تجاری مرکبات موجود در ایستگاه تحقیقات کترا، از نشانگر SSR استفاده گردید. استخراج DNA از برگ های جوان مرکبات با روش موری و تامسون انجام شد و سپس تکثیر با ده جفت آغازگر ریزماهواره صورت گرفت. محصولات تکثیر شده با استفاده از ژل پلی آکریلامید 6% دارای اوره 7 مولار و تحت شرایط واسرشتگی تفکیک شده و با رنگ آمیزی نیترات نقره قابل رویت گردید. الگوی باندی براساس حضور باند (یک) یا عدم حضور باند (صفر) نمره دهی شده و محتوای اطلاعات چندشکلی برای هر جفت نشانگر محاسبه گردید و با استفاده از نرم افزار NTSYS، فاصله ژنتیکی ژنوتیپ ها براساس ضریب تشابه جاکارد محاسبه وروش تجزیه خوشه ایدورترین همسایه برای گروه بندی استفاده شد.
    یافته ها
    میانگین چندشکلی 0/685 برآورد شد. تعداد آلل ها از 3 تا 13متغیر بوده و در مجموع 72 آلل شناسایی شد. تجزیه کلاستر 45 ژنوتیپ مورد بررسی را براساس نشانگرهای SSR به شش گروه تقسیم نمود. ارقام پرتقال، نارنگی و 24 ژنوتیپ ناشناخته در گروه A، گریپ فروت و پوملو و نارنج و شش ژنوتیپ ناشناخته در گروه B، سه ژنوتیپ ناشناخته در گروهC، دو ژنوتیپ ناشناخته و یوزو به ترتیب در گروه های D و E و در نهایت ارقام لیمو و بالنگ و دو ژنوتیپ ناشناخته در گروه F قرار گرفتند.
    نتیجه گیری
    هر یک از این نشانگرها سطوح مختلفی از تنوع را در سطح ژنوم ارایه می دهند که می تواند در مدیریت ژرم پلاسم و ذخایر توارثی مفید باشد. تجزیه تنوع ژنتیک و روابط خویشاوندی در مرکبات، اطلاعات مفیدی را برای برنامه های به نژادی، انتخاب و ثبت ارقام جدید فراهم می کند.

    سابقه و هدف
    زیتون گیاهی با ارزش اقتصادی بالائی است که از روغن و کنسرو میوه آن استفاده می شود. روش های سنتی ازدیاد زمان بر می باشد. با استفاده از تکنیک کشت بافت میتوان گیاهانی با کیفیت بالا وسرعت رشد بیشتر به دست آورد. هدف از پژوهش حاضر، بررسی و مقایسه تاثیر نانو کلات آهن و کلات آهن بر سرعت رشد ریز قلمه های زیتون رقم دزفول بود.
    مواد و روش ها
    جداکشت های تک گره های از سرشاخه های جوان گیاه 5 ساله زیتون رقم دزفول سترون در محیط کشت Dkwدارای هورمون (2ip(4mgl -1کشت شد.ریز قلمه های سترون حاصل برای واکشت در محیط های کشت دارای غلظت های مختلف نانو آهن و آهن (Mμ 180، 120،60) استفاده شد. 45 روز پس از واکشت شاخص های رشد بررسی شد.
    یافته ها
    بررسی ها نشان داد که شاخص های رشد نانو آهن باعث کاهش تعداد گره ها، شاخساره ها، تعداد برگ ها و طول ساقه شد. درغلظت بالای سولفات آهن رشد ساقه و تعداد برگ ها افزایش معنی داری را نشان داد. شاخه زائی در غلظت متوسط به اکثر خود رسید. میزان کلروفیل b و کاروتنوئیدها نیز با افزایش غلظت آهن و نانو آهن در محیط کشت افزایش یافت. سولفات آهن بیش از نانو آهن موجب افزایش این رنگیزه ها شد.
    نتیجه گیری
    در بین 8 تیمار بکار برده شده بالاترین غلظت آهن و نانو آهن در هفته اول باعث از بین رفتن نمونه ها شد. برای ریز ازدیادی زیتون رقم دزفول، کلات آهن (F1.5 (Mμ180، در محیط کشت DKW مناسب تر ازسایر غلظت ها می باشد.

    سابقه و هدف
    هدف از انجام این تحقیق بررسی پلی مورفیسم ناحیه کد کننده اگزون 17 ژن DGAT1 در گوسفندان مهربان بود.
    مواد و روش ها
    استخراج DNA به روش کیت از تعداد 120 نمونه گوسفند نژاد مهربان به دست آمد و واکنش زنجیره ای پلی مراز برای تکثیر 309 جفت باز از اگزون 17 ژن DGAT1 با استفاده از یک جفت آغازگر اختصاصی انجام گرفته و محصولات PCR با روش RFLP و با استفاده از آنزیم برشی AluI هضم شدند و هر سه ژنوتیپ TT، TC، CC به دست آمد.
    یافته ها
    یافته ها بیانگر وجود فرکانس های 0/25، 0/24 و 0/51 به ترتیب برای ژنوتیپ های CC، TC و TT می باشد. عدم وجود تعادل هاردی- واینبرگ با توجه به شرایط نگهداری دام در منطقه می تواند مربوط به مهاجرت و انتخاب باشد. یافته ها هم چنین حاکی از مناسب بودن روش PCR-RFLP در بررسی چندشکلی های ژن DGAT1 می باشد.
    نتیجه گیری
    در جمعیت گوسفندان مهربان دو آلل T و C به ترتیب با فراوانی 0/37 و 0/63مشاهده شد. همچنین جمعیت مورد مطالعه در تعادل هاردی- واینبرگ قرار نداشت.

    سابقه وهدف
    یکی از مشکلات اساسی و شایع مربوط به مجرای ادراری بروز عفونت و تشکیل سنگ های ادراری در انسان است. هدف از این بررسی تعیین جنس سنگ و نوع باکتری های موجود در مجرای ادراری (لگنچه) بیماران مراجعه کننده به بیمارستان لبافی نژاد تهران بوده است.
    مواد و روش ها
    از لگنچه و ادرار 100 بیمار (نوزاد 2روزه تا فرد 75 ساله) مبتلا به سنگ مجرای ادراری که تحت عمل جراحی PCN قرار گرفتند نمونه برداری شد و نوع باکتری ها، با روش استاندارد میکروبیولوژی شناسایی شد. قطعه کوچکی از سنگ مجرای ادراری بیماران در شرایط استریل در دو لوله حاوی محیط کشت BHI جهت کشت و شناسایی باکتری ها قرار داده شد و بخش دیگری از سنگ برای تجزیه شیمیایی و تعیین جنس با استفاده از کیت های تشخیصی سنگ نگاه داشته شد.
    یافته ها
    در طی این بررسی، از 100 نمونه کشت سنگ که انجام شد، باکتری ها در 31 مورد (31%) رشد کردند و در بقیه موارد 69 مورد (69%) هیچ گونه میکروارگانیسمی مشاهده نگردید و در برخی از نمونه ها دو باکتری رشد کردند. میکروارگانیسم های ایزوله شده از کشت لوله حاوی سنگ شامل استافیلوکوکوس اورئوس (9/6%)، اشرشیا کلی (29/3%)، پروتئوس (6/4%)، سیتروباکتر (9/6%)، سودوموناس(9.6%)، باسیلوس سوبتیلیس (06/4%)، بودند. در بررسی باکتریایی نمونه ادرار لگنچه نیز دقیقا همین نتایج حاصل گردید. سنگ های اگزالات کلیسمی (62%)، فسفات کلسیمی (18%)، سنگ فسفات آمونیوم منیزیوم (سنگ عفونی) (11%)، و سنگ اسیداوریکی (7%) و سنگ سیستینی (2%) از کل سنگ ها را تشکیل دادند.
    نتیجه گیری
    در 69% از نمونه لگنچه هیچ گونه باکتری رشد نکرد. احتمالا باکتری های دیگری نظیر مایکوپلاسما، اوره آپلاسما اوره آلیتیکوم، نانوباکتری ها و غیره وجود داشتند که در محیط های کشت معمول آزمایشگاهی قادر به رشد نبودند. بنابراین پیشنهاد می شود به جای استفاده از محیط کشت معمولی از محیط های کشت اختصاصی برای باکتری های نامبرده استفاده شود و یا می توان از تکنیک PCR برای شناسایی عفونت های مجرای ادراری بهره جست. شناسایی ترکیب سنگ های مجرای ادراری جهت درمان و پیشگیری از عود مجدد سنگ ها، امری ضروری و اجتناب ناپذیر است و برای پیشگیری از عود تشکیل سنگ باید رژیم غذایی و درمان دارویی ویژه ای را به دارندگان سنگ توصیه کرد.

    سابقه و هدف
    امروزه تقاضا برای انرژی پاک رو به افزایش است. یکی از روش های آسان و پاک تولید انرژی استفاده از پیل های سوختی به ویژه پیل زیست سوختی می باشد. پیل زیست سوختی وسیله ای است که قادر به تبدیل انرژی شیمیایی به انرژی الکتریکی می باشد. انواع مختلفی از پیل های زیست سوختی طراحی شده اند که می توانند از بیوکاتالیست ها، آنزیم ها و حتی میکروارگانیزم ها، با هدف تولید انرژی الکتریکی استفاده نمایند.
    مواد و روش ها
    در این پژوهش، از سول – ژل در کپسوله کردن و تثبیت آنزیم لاکاز روی الکترود، استفاده گردیده است. در این روش تثبیت، رله آنزیمی داخل حفره های کربن- سیلیکا که از ساختار هتروژن الکترود کربن- سرامیک (CCE) ناشی می شود، تثبیت و ساختار ABTS-CCE تشکیل می گردد. از سول - ژل به دلیل دارا بودن ABTS به عنوان یک ماتریکس برای دستیابی به انتقال الکترون واسطه ای لاکاز استفاده گردید.
    یافته ها
    ولتاموگرام چرخه ای مقدار انتقال الکترون پایین را به دلیل ارتباط ضعیف آن بین آنزیم و سطح الکترود را نشان داد. بیشینه پیک ردوکس در 10 میکروآمپر در یک محلول حاوی 15 میکرومولار او- دیانیزیدین به عنوان سوبسترا به دست آمده است.
    نتیجه گیری
    با استفاده از نانولوله های کربنی اتصال بهبود یافته و بیشینه پیک ردوکس در 14 میکروآمپر در غلظت مشابه به دست آمد.

    سابقه و هدف
    مهندسی بافت حوزه رو به رشدی برای ترمیم و جایگزینی عملکرد معیوب بافت یا ارگان آسیب دیده می باشد و امروزه به عنوان یک درمان نوین، جایگزین روش های مرسوم پیوند مطرح گردیده و به این منظور مواد زیستی پلیمری(داربست ها) و سلول های زنده را به کار می گیرد. داربست هایی در مقیاس زیر میکرون و نانو ساخته می شود که ساختاری مشابه ماتریکس طبیعی خارج سلولی (ECM) بوده و از چسبندگی، تکثیر و تمایزسلولی حمایت می کند و در عین حال پایداری فیزیکی و شیمیایی و ساختار زیستی مناسب را برای بافت جدید فراهم می کند. در پژوهش حاضر از نانوکامپوزیت کیتوسان/لامینین به عنوان داربست برای رشد و تکثیر سلول استفاده شده است.
    مواد و روش ها
    داربست های نانوکامپوزیت کیتوسان/لمینین برای کاشت و تکثیر سلولهای فیبروبلاست رده ی L929 با استفاده از روش خشک کن انجمادی استفاده گردید. ویژگی های فیزیکوشیمیایی داربست توسط میکروسکوپ الکترونی به طور کامل بررسی شد. سپس میزان زیست سازگاری داربست مورد بررسی قرار گرفت.
    یافته ها
    مشاهدات حاصل از میکروسکوپ الکترونی حاکی از تشکیل ریزساختارهای متخلخل سیلندری شکل و حفرات به هم پیوسته بودند.
    نتیجه گیری
    نتایج نشان داد که استفاده از این داربست به دلیل عدم سمیت و زیست سازگاری مناسب، امکان رشد و اتصال مناسب سلول های فیبروبلاست رده ی L929 را فراهم می کند.

    سابقه و هدف
    Symbiodinium جلبک تک سلولی دو تاژکی است که با تعدادی از بی مهرگان هم زیستی دارد و تاکنون 9 کلاد از آن با روش های مولکولی شناسایی شده است. کلادهای مختلف Symbiodinium خصوصیات فیزیولوژیکی متنوعی دارند که می توانند تاثیرات متفاوتی بر دوام و بقای میزبان آن ها در برابر شرایط مختلف محیطی و موقعیت های جغرافیایی داشته باشند. در این تحقیق، کلادهای زوگزانتله هم زیست با مرجان های آبسنگ ساز در شرق جزیره کیش با نمونه های مورد بررسی در سال 2007 مقایسه گردیدند.
    مواد و روش ها
    از 5 گونه مرجان Acropora arabiensis، Acropora downingi، Porites compressa، Platygyra daedalea، Psammacora contigua، از عمق 2 تا 5 متری نمونه برداری انجام گرفت. بعد از جدا کردن بافت نرم حاوی Symbiodinium مرجان-ها توسط پمپ هوا، استخراج DNA به روش CTAB-Chloroform به منظور بررسی کلادها و زیرکلاد های Symbiodinium، با استفاده از ژن(LSU (Large Subunit Ribosomal DNA انجام شد. پس از انجام واکنش زنجیره ای پلیمراز، نمونه ها توالی یابی شدند.
    یافته ها
    نتایج تعیین ترادف ژنی نشان داد که هیچ تفاوتی بین کلادهای به دست آمده و کلادهای گزارش شده در سال 2007 وجود ندارد. هیچ یک از گونه های مرجانی بررسی شده تغییر و جابجایی در جوامع هم زیست خود ایجاد نکرده اند.
    نتیجه گیری
    با توجه به عمق کم و شرایط سخت در ناحیه شرقی جزیره کیش، حضور کلاد D ممکن است یک پاسخ سازشی یا اقلیمی شدن به شرایط محیطی غالب در منطقه باشد.

    سابقه و هدف

    ال-آسپاراژیناز ΙΙ کاربرد موثری در درمان لوسمی لنفوبلاستیک (ALL) دارد. این آنزیم از منابع باکتریایی جدا شده و به صورت تجاری به عنوان داروی ضدسرطان عرضه می شود. سلول های سرطانی برخلاف سلول های نرمال، نیاز شدیدی به ال-آسپاراژین دارند، در صورت به کارگیری ال-آسپاراژیناز (E.C. 3.5.1.1) ال-آسپاراژین به ال-آسپارتات و آمونیوم تبدیل می شود که در نهایت به تخریب سلول های سرطانی منجر می شود. هدف از این پژوهش، کلون کردن ژن ال-آسپاراژیناز E. coli II در B. subtilis جهت تولید انبوه این آنزیم انجام گرفت.

    مواد و روش ها

    ژن ال-آسپاراژیناز ansB) II) با روش PCR از E. coli BL21 جدا گردید. قطعه تکثیر یافته و شاتل وکتور بیانی pMR12 توسط آنزیم های HindΙΙΙ، BamHΙ هضم شد. واکنش اتصال بین قطعه PCR و شاتل وکتور بیانی برش خورده با روش استاندارد انجام گرفت. در مرحله بعد، وکتور نوترکیب با روش شوک با CaCl2 سرد بهE. coli JM101 ترانسفورم شد. در نهایت به میزبان B. subtilis با روش شیمیایی انتقال یافت.

    یافته ها

    در این مطالعه، ژن ansB به وسیله PCR از E. coli BL21 جدا و توسط تجزیه و تحلیل آنزیمی محصول PCR تایید گردید و در شاتل وکتور بیانی pMR12 کلون شد. سپس وکتور نوترکیب ابتدا در E. coli کلون گردید و وجود ژن 1047bp با تجزیه و تحلیل آنزیمی و واکنش PCR تایید شد به دنبال آن داخل B. subtilis کلون شد و استخراج پلاسمید انجام گرفت و با باند شاتل وکتور بیانی pMR12 دارای ژن ansB به صورت صحیح، مقایسه و تایید گردید.

    نتیجه گیری

    در این تحقیق با استفاده از شاتل وکتور بیانی pMR12 توانستیم ژن ansB را در B. subtilis کلون کنیم. این مطالعه، اولین گزارش کلونینگ ژن ansB در B. subtilis است.

    سابقه و هدف
    Charybdis hellerii گونه ی بسیار شناخته شده ای در اقیانوس هند است. بررسی های مولکولی فراوانی بر روی این گونه در سایر نقاط دنیا انجام شده است. در ایران مطالعات مولکولی کمی بر روی خرچنگ ها صورت گرفته شده است، با این حال مطالعات ریخت شناسی بسیار جامع و مفیدی بر روی آنها انجام گرفته شده است. در تحقیق حاضر برای اولین بار در ایران به بررسی مولکولی جنس Charybdis گونه ی Charybdis hellerii پرداخته شده است.
    مواد و روش ها
    در بهار سال 1392 و از سواحل صخره ای خلیج چابهار نمونه ها جمع آوری گردیدند. پس از نگهداری در دمای 20-، برش بافتی از آبشش نمونه ها جدا گردید. عمل استخراج DNA با استفاده از روش CTAB انجام گرفت. واکنش زنجیره ای پلیمراز (PCR) انجام شد. پس از توالی یابی، درخت فیلوژنی گونه ی مورد نظر رسم گردید.
    یافته ها
    با بررسی های مورفولوژی گونه مورد مطالعه Charybdis hellerii شناسایی گردید و بررسی های مولکولی داده های مورفولوژی را تایید کردند. توالی نوکلئوتیدی به دست آمده در قطعه ی 1 ژن سیتوکروم اکسیداز(CO1) مورد آنالیزهای مولکولی قرار گرفت. با آنالیزهای انجام شده و ترسیم درخت فیلوژنی مشخص شد که گونه ی ایرانی با 94 درصد بوت استرپ حمایت با گونه C. hellerii در یک کلاد قرار گرفته است.
    نتایج
    نتایج حاصل از آنالیز ML نشان داد که گونه مورد نظر در کلاد Eubrachyura قرار می گیرد که 94 درصد شبیه به Charybdis hellerii است و در یک گروه خواهری قرار گرفتند. آنالیزهای مولکولی نتایج حاصل از بررسی های موفولوژی را تایید می کند.

    سابقه و هدف
    ژن سرکوبگر تومور P53 که به عنوان «محافظ ژنوم» شناخته می شود، در بیشتر از نیمی از تمام انواع سرطان-های انسانی جهش یافته است. پروتئین P53 یک عامل رونویسی است که وظیفه آن کنترل رونویسی از ژن های مسئول آپپتوز و توقف چرخه سلولی است. هم چنین پروتئین P53 می تواند رونویسی از ژن هایی که در پیشرفت چرخه سلولی نقش دارند جلوگیری نماید. بنابراین بدیهی است که از دست دادن فعالیت این ژن با فراوانی بالا در انواع مختلفی از سرطان ها مشاهده شده است. مشاهدات تجربی هم زمانی بین افزایش اسیدیته و نیز جهش در کدون های خاص ژن P53 (به طور خاص کدون 248) را نشان داده اند. این امر می تواند تا حدود زیادی ناشی از پروتونه شدن این توالی سه نوکلئوتیدی باشد. جهش در این کدون خاص باعث جایگزینی اسید آمینه می گردد که نتیجه آن ایجاد پروتئینی با خاصیت انکوژنی میباشد. در این مطالعه تاثیر pH اسیدی را بر پایداری کدون 248 در ژن P53 بررسی گردید.
    مواد و روش ها
    در این مطالعه از روش مکانیک مولکولی برای پاسخ پرسش این مطالعه استفاده شد. در این مطالعه از نرم-افزار HYPERCHEM استفاده شد. هم چنین به منظور دست یافتن به بهترین پیش بینی از میدان های نیروی AMBER، BIO+، MM+ و OPLS استفاده گردید.
    یافته ها
    نتایج به دست آمده در میدان های نیروی مختلف همگی نشان دادند که در pHاسیدی سطح انرژی توالی مورد بررسی به میزان زیادی بالاتر از شرایط فیزیولوژیک می باشد.
    نتیجه گیری
    نتایج این مطالعه پاسخ قابل ملاحظه ای را درباره هدف مطرح شده ارائه نمود و نشانداد کهpH اسیدی می تواند به لحاظ تئوریک ناپایداری مولکول را نسبت به pH فیزیولوژیک افزایش دهد.

    زمینه و هدف
    جنس میکرومونوسپورآ منبع پرکاربرد متابولیت های مختلف فعال زیستی مانند آنتی بیوتیک ها و مهار کننده های آنزیم است. اعضای میکرومونوسپورآ به طور گسترده در انواع زیستگاه ها، به ویژه خاک غنی توزیع شده اند. مطالعه حاضر با هدف جداسازی، شناسایی ایزوله ها توسط تکثیر ژن 16S rRNA تا سطح جنس و تعیین فعالیت ضد میکروبی آنها صورت پدیرفت.
    مواد و روش ها
    60نمونه خاک از نقاط مختلف ایران جمع آوری شد. هر نمونه خاک تحت تیمار با فنل 1/5 درصد در محیط کشت های مختلف مناسب برای جداسازی میکرومونوسپورآ کشت داده شد. شناسایی جنس ایزوله های جدا شده با استفاده از تکثیر ژن 16S rRNA با پرایمرهای اختصاصی جنس صورت گرفت. سپس عصاره متعلق به میکرومونوسپورآ جدا شده، در برابر تعدادی از باکتری های بیماری زا برای تعیین فعالیت ضد میکروبی آنها مورد آزمایش قرار گرفت.
    یافته ها
    از 60 نمونه خاک مورد بررسی 200 اکتینومیست جدا شد، که 15 ایزوله به عنوان میکرومونوسپورآ با استفاده از تکثیر ژن 16S rRNA با پرایمرهای اختصاصی جنس تایید شدند. از عصاره جدا شده هر ایزوله 5 ایزوله فعالیت ضد میکروبی روی استافیلوکوکوس اورئوس مقاوم به متی سیلین (MRSA) و باسیلوس سرئوس داشتند.
    نتیجه گیری
    میکرومونوسپوراهای جدا شده در این مطالعه توان تولید آنتی باکتریال های موثر بر گونه های MRSA به عنوان یکی از معضلات سیستم های بهداشتی را دارا بودند.

    غربال گری اکتینومیست ها توسط محققین، برای تولید آنتی بیوتیک های نو در سال های اخیر شتاب چشمگیری پیدا کرده است.
    مواد و روش ها
    از بیابان های کویر لوت، کویر استان سیستان و بلوچستان و بیابان های اهواز 30 نمونه خاک جمع آوری گردید. اکتینومیست های هر نمونه خاک جداسازی و خواص ضد میکروبی آنها بر علیه تعدادی باکتری پاتوژن بررسی شد.
    یافته ها
    تعداد 300 ایزوله اکتینومیست از نمونه های خاک جداسازی شد. از این تعداد ایزوله، 24 ایزوله جدا شده دارای خواص ضدمیکروبی مناسبی بر علیه حداقل یکی از باکتری های پاتوژن مانند S. aureus، E. faecium، K.pneumonia و A. baumannii بودند.
    نتیجه گیری
    این مطالعه نشان داد که خاک های بیابانی دارای گونه های متنوعی از اکتینومیست ها می باشند که تعدادی از آنها دارای خواص وسیع الطیف ضد میکروبی هستند و منابع بالقوه ای برای آنتی بیوتیک ها می باشند.

    Background
    Extended spectrum β-lactamases (ESBLs) have emerged as a major threat worldwide with limited treatment options. This study aimed to determine the antibiotic susceptibility pattern and the evaluation of TEM genes producing in Escherichia coli and Klebsiella pneumonia isolates collected from clinical samples.
    Materials And Methods
    Bacteria were isolated and identified from the different samples in patients sent to laboratory of shahid Beheshti University in Tehran in 2011. Isolates were then tested for antimicrobial susceptibility by disc diffusion and examined for TEM genes production by polymerase chain reaction using specific primer.
    Results
    Out of 100 studied nosocomial infection specimens, 50 isolates were klebsiella pneumonia of which 34 percent were ESBL producer and all were positive for TEM gene, resistance of Cefotaxime was 90 percent which is the highest degrees of resistance and lowest degrees of resistance to Imipenem was 4 percent. Among the 50 isolates of Escherichia coli of which 14 percent were ESBL producer and all were positive for TEM gene, resistance of Cefexime was 90 percent which is the highest degrees of resistance and lowest degrees of resistance to Meropenem was 6 percent.
    Conclusion
    Due to relatively high prevalence of ESBL-producing bacteria in the studied population, antibiogram test are advised for appropriate treatment.

    Background
    The cancer stem cell model postulates that a small subset of cancer cells with stem like properties drive tumor initiation, progression and drug resistance. Since Three-dimensional (3D) in vitro models provide a well-defined environment for cancer research in contrast to the complex host environment of an in vivo model, our aim in this study is to compare cancer stem cell properties in 3D and 2D culture.
    Materials And Methods
    MDA-MB468 Cells cultured in 2D and 3D culture were compared for their colony formation and spheroid formation efficiency, stemness markers expression (CD133,CD44) by flowcytometry and stemness gene expression (Oct4,Sox2,Nanog,Aldh, Cmyc) by Real time PCR.
    Results
    Cells cultured in 3D showed no significant difference in colonogenic and spheroid forming capacity in comparison with those cultured in 2D. While Cells in 2D showed higher proportion of CD133+ phenotype than those in 3D (98.55±0.34 vs. 52.69±5.84), no significant difference was observed in CD44+ expression. Additionally, stemness genes including Aldh, Cmyc, Oct4, Nanog, and Sox2 were strikingly down regulated in 3D culture.
    Conclusion
    We demonstrated that 3D culture which could mimic the tumor architecture and in vivo conditions decrease cancer stem cell properties of MDA-MB468 cell line.

    Background
    Understanding phylogenic relationships and genetic diversity in Citrus is considered to be important in clarifying their genetic relationships, germplasm characterization and the registration of new varieties. There are some Citrus biotypes in the country Citrus collections which have been classified merely based on their morphological traits and many genetic researches were not done on them. Molecular markers would help to infer their relations with known cultivars.
    Materials And Methods
    In order to investigate about phylogenic relationships among 45 Citrus genotypes including 33 unknown local genotypes and 12 commercially important Citrus varieties at the collection in Agricultural Research Institute of Kotra, Ten SSR primer pairs were used. DNA was extracted from young leaf tissues according to the procedure of Murry and Thompson (1980). One SSR primer was chosen from each corn chromosome and totally ten primers were assayed using the sample of 45 genotypes. The amplified products were separated using 6% polyacrylamide gel with 7 M urea under denaturing conditions and visualized by staining with silver nitrate. Gels were then scored based on either the presence or absence of bands. Polymorphism information content (PIC) for each SSR marker was determined with 1- Σfi 2- Σ2fi2 fj2 where fi is the frequency of the ith allele and fj is frequency of the ith allele + 1. Estimation of genetic similarities was done among these lines using the NTSYS program. The 45 genotypes were clustered based on the matrix of Jaccard genetic similarity using the complete clustering algorithm.
    Results
    Ten primers revealed 0.685 polymorphism rate. The number of alleles per locus ranged from 3 to 13. Among all, 72 alleles were identified. Cluster analysis was resulted in designating the 45 samples into six major groups. Orange, mandarin and 24 unknown genotypes into group A, grapefruit, Pummelo, sour orange and six unknown genotypes into group B, three unknown genotypes into group C, two unknown genotypes and Yuzo into groups D and E, citron, Eureka lemon, sweet lime, Mexican lime and two unknown genotypes into group F.
    Conclusion
    SSR markers revealed different levels of diversity which can be useful in germplasm and genetic resources management. Genetic diversity and phylogenetic analysis in Citrus provide useful information for further breeding programs, selection, and registration of new varieties.

    Background
    Olive tree (Olea europaea L.) has a high economic value and many countries such as Iran and Mediterranean countries use its oil and canned fruits. Conventional propagation programs are time consuming. Tissue culture technique allows producing high quality and rapid growing plants. The objective of the present study was to evaluate the effect of chelated iron and nano-chelated iron on the growth rate of in vitro microshoots.
    Material And Methods
    Uninodal explants of young shoots from a mature olive Oleaeuropaea L((cv. Dezful), were cultured in DKW medium supplemented with 2ip (4m〖g.L〗^(-1)). Sterile microshoots were subcultured in DKW medium supplemented with different concentrations of nano-Fe and Fe chelate (60, 120,180, 240 µM). Forty-five days after subculture, growth parameters were investigated.
    Results
    Results indicated that the growth parameters (number of nodes, microshoots, leaves and stem length) significantly decreased in the presence of nano Fe. The maximum level of leaves number and stem length was achieved in the high concentration of Fe (180 µM). Upon increasing the Fe or nano Fe content in the medium, the chlorophyll b and carotenoid levels were significantly increased. Fe chelate was more effective than nano Fe.
    Conclusion
    Microshoots couldnt survive in the highest concentration of nano Fe and Fe (240 µM). The best results were obtained in the presence of Fe (120, 180 µM)

    Background
    The aim of this study to compare the coding area of the exon 17 DGAT1 gene polymorphism in Mehraban sheep.
    Materials And Methods
    DNA was extracted using Kit method, 120 samples of Mehraban sheep were obtained, and also polymerase chain reaction was performed to amplify 309 bp of exon 17 DGAT1 gene using a specific primer. In RFLP method, PCR products using AluI enzyme digestion were digested and the three genotypes TT, TC and CC were found.
    Conclusion
    Mehraban sheep in two alleles T and C respectively, with frequency of 0.37 and 0.63 were observed. Also the population was not found to follow Hardy-Weinberg equilibrium.

    Background
    The infection outbreak and formation of urinary tract stones is one of the basic and general problems in humans. The purpose of this research was to study on stones and identify bacteria in patient’s urinary tract (pelvic) attending Tehran Labafi-Nejad hospital.
    Material And Methods
    Specimens of the pelvic and urine of 100 patients from 2-day infants to 75-years old persons) with urinary tract stones were collected during PCNL operation and bacteria were identified by microbiological standard methods. The small piece of patient’s urinary tract stones was put (in sterile condition) in two tube containing BHI medium for culture and identification of bacteria, then, another part of stone was retained for chemical analysis using stones diagnosis kits.
    Result
    In this study, out of 100 stone culture samples, the bacteria grew in 31 cases (31%) and there weren’t any microorganisms in the remaining 69 cases (69%). Two bacteria were grown together in some cases. The isolated microorganisms from tube’s culture, containing stone included: E. coli (29%, 3), Staphylococcus aureus (9%,6), Proteus (6%,4). Isolated bacteria from pelvic urine cultures were the same as stone’s culture. The percentage of obtained stones include: calcium oxalate stones (62%), calcium phosphate (18%), magnesium ammonium phosphate (struvite stones) (11%), uric acid (7%) and cystein stones (2%). In 69% of pelvic sample, no bacteria were grown. It might be due to other bacteria such as (Mycoplasma urealyticum, Nanobacteria, etc.), which couldn’t grow in general laboratory media. Therefore, it’s recommended to use specific media instead of general cultures, for the identification of bacteria. Also PCR technique can be used for the detection of urinary tract infection. Determination of compound of urinary tract stones is essential and inevitable for the treatment and prevention of recurrent stone formation. It’s necessary for stone formers to follow food diet and use special drug treatment, in order to prevent the recurrent stone formation.

    Nowadays demands for clean power source is highly enhanced. Bio-fuel cell (BFC) can convert chemical energy to electrical energy. The enzyme-based bio fuelcells (BFC) is a special fuel cell using enzyme as catalyst and can directly convert chemical energy to electrical energy. Bio-fuel cells are energy conversion devices based on bio- electrocatalysis leveraging on enzymes or microorganisms.
    Materials And Methods
    In the present paper, Sol-gel is used to laccase Encapsulation and immobilization on the electrode. Mediator is immobilized into porous silicate–carbon heterogeneous structure of carbon ceramic electrode (CCE). In the next step, laccase is immobilized in hydrophilic silicate thin film deposited on the ABTS modified CCE (ABTS-CCE) surface. ABTS polymer is located in sol-gel function as the mediator for the electron transfer.
    Results
    Cyclic voltammetric results indicate low electron transfer rate because of weak contact between enzyme and electrode surface while maximum redox pick achived in 10 micro amper in a solution containing 15 micro molar o-dianisidin as substrate.
    Conclusion
    By using of carbon nanotubes connection were improved and maximum redox pick achived in 14 micro amper in the same concentration.

    Background
    Tissue engineering is a promising area in biomedical engineering to repair or replace the function of defective or damaged tissues or organs. Recently, tissue engineering has provided a new medical therapy as an alternative to conventional transplantation methods by using polymeric biomaterials with living cells. The scaffold provides the microenvironment (synthetic temporary extracellular matrix) for regenerative cells, supporting cell attachment, proliferation, differentiation, and neo tissue genesis due to their suitable chemical, physical and biological. In this research, chitosan/laminin nanocomposite was exploited as scaffold for suitable cell proliferation.
    Materials And Methods
    Freeze-drying technique was used to fabricate chitosan/laminin-nanocomposites for L929 fibroblast cellsseeding, proliferation and attachment. The physico-chemical properties of the scaffold were fully characterized by using scanning electron microscopy (SEM). Consequently, the biocompatibility of scaffold was evaluated by biocompatibility test.
    Results
    The microstructure observation with SEM suggests the formation of cylinder-shaped porous structure and interconnected porosity.
    Conclusion
    Our results demonstrated that the nano-sized chitosan/laminin scaffolds are nontoxic and biocompatible which can promote proliferation and attachment of L929 fibroblast cells. And their appropriate adhesion to nanocomposite for improved tissue engineering applications.

    Background
    Symbiodinium is a unicellular biflagellate alga that performs symbiosis with certain invertebrates. Different clades of Symbiodinium show variety of physiological features that could have uneven affect on the hermatypic corals stability and survival in various environmental conditions and geographical locations. In this study, Clades of symbiotic zooxanthellae of hermatypic corals of eastern Kish Island were compared to samples taken in 2007.
    Materials And Methods
    Samples were taken at depth of 2 to 5 meters from 5 coral species including Acropora arabiensis, Platygyra daedalea, Porites compressa, Acropora downingi and Psammacora contigua. Coral colonies were air brushed in order to separate Symbiodinium cells. DNA was extracted using CTAB-Chloroform method. LSU (Large Subunit Ribosomal DNA) region was used as target for amplification. Finally PCR products were sequenced.
    Results
    Results did not show any difference between Corals’ symbionts and the study in the area during 2007. None of the studied coral species have switched or shuffled their symbionts populations.
    Conclusion
    Taking into account inhospitable condition and shallowness of eastern parts of the Kish Island, presence of clade D might be an adaptive or acclimatized response due to dominant regional condition.

    Aim and Background

    L-asparaginaseΙΙ (E.C. 3.5.1.1) has effective usage for treatment of acute lymphoblastic leukemia. This enzyme is isolated from bacterial sources and it is commercially available as an anti-cancer drug. This enzyme catalyzes the hydrolysis of L-asparagine to ammonium and aspartate. Unlike Normal cells, the cancer cells strongly require L-Asparagine leading to destruction of these cells. The propose of this project is cloning of L-asparaginaseΙΙ gene from E. coli in B. subtilis to produce the mass of this enzyme.

    Materials And Methods

    L-asparaginaseΙΙ gene is isolated from E. coli by PCR approach. The amplified fragment and expression shuttle vector pMR12 are digested by HindΙΙΙ and BamHΙ enzymes. The ligation between DNA fragment and the cloning shuttle vector is done by standard method. In the next step recombinant vector is transferred to E. coli JM101 by cold calcium chloride treatment and finally introduced into B. subtilis by a chemical method.

    Results

    in this experiment, ansB gene is isolated from E. coli by PCR and cloned into the expression shuttle vector pMR12. Existence of gene with 1047 bp lenght is confirmed by enzymatic analysis and PCR reaction. Then recombinant vector cloned in E. coli at first and then in B. subtilis. Finally the plasmid extracted and compared with the band of expersion shuttle vector containing ansB gene to confirm.

    Conclusions

    In this study, we tried to clone the ansB gene in B. subtilis using expression shuttle vector pMR12. This is the first report of ansB gene cloning in B. subtilis.

    Background
    Charybdis hellerii is a well-known species in the Indian Ocean. There are many molecular researches on this species all over the word. Molecular researches on Iranian water crabs are very scarce; however there are several comprehensive morphological studies. In this paper, for the first time, phylogeny of Charybdis hellerii from Iranian waters are studied.
    Materials And Methods
    Specimens were collected in 2013 from sandy sediments of shallow-waters in Chabahar Bay at full tide. Collections were transported to the laboratory and preserved in -20 degrees. The gill tissue was used for DNA extraction. The Hexadecyltrimethylammonium bromide (CTAB) method was used to extract DNA. Polymerase Chain Reaction (PCR) was then carried out. The sequencing alignments were conducted and phylogenic tree was obtained.
    Results
    The crab sample was identified as Charybdis hellerii by morphological analysis confirmed by molecular analysis. Nucleotides sequence of the cytochrome oxidase subunit 1 (CO1) gene was analyzed. The phylogenetic analysis was conducted and found that the Iranian species with other Charybdis hellerii are in a clade and support with 94% bootstrap.
    Conclusion
    The maximum Likelihood phylogenetic analysis showed that the Iranian species is in the Eubrachyura clad and with 94% bootstrap support is a sister group Charybdis hellerii. The molecular results were also confirmed by morphological features.

    P53 tumor suppressor gene, also known as “genome guardian” is mutated in more than half of all kinds of cancers. P53 protein acts as a tetramer and is a transcription factor, controlling the transcription of genes that are necessary for apoptosis or cell proliferation arrest. Also it inhibits the transcription of genes that are responsible for cell growth. Experimental researches revealed that acidic pH raised the rate of cancer and mutation in 248CGG codon of P53 gene. This may be due to protonation of this three-nucleotide codon. Mutation in this codon changes the encoding amino acid and subsequently produces a protein, which has oncogenic features instead of tumor suppressor characteristics of original p53 protein. In current study, we perform an investigation on the impact of protonation on stability of codon 248CGG in this gene.
    Materials And Methods
    Molecular mechanic methods used in this study to calculate energy levels. HYPERCHEM software used in AMBER, BIO+, MM+ and OPLS.
    Results
    Obtained results from different force fields all suggest that acidic condition can increase molecular instability in given sequence in comparison with physiological pH.
    Conclusion
    Our results suggested a reliable answer about the effect of protonation on mentioned codon and its stability. From theoretical point of view, acidity can increase the instability of this specific codon. Along with the experimental investigations, our results can, to some extent, elaborate the mutagenesis of acidic pH.

    Aim and
    Background
    The genus Micromonospora is a prolific source of various bioactive metabolites, such as antibiotics and enzyme inhibitors. Members of Micromonospora are widely distributed in a variety of habitats, notably soil rich. The aim of current study was to isolation, identification of the isolates to genus level by 16S rRNA gene amplification and determines the isolates antimicrobial activity.
    Material And Methods
    Sixty soil samples collected from different parts of Iran. Each soil sample treated by 1.5% phenol and was cultured on different suitable media to isolation of Micromonospora. The isolates were subjected to genus identification by means of 16S rRNA amplification with genus specific primers. Extract of the isolates belongs to Micromonospora, were tested agianst pathogenic bacteria to determine their antibacterial activity.
    Results
    From 60 soil samples, 200 actinomycets were isolated which 15 isolates were confirmed as Micromonospora by means of 16S rRNA amplification with genus specific primers. Extract of the 5 isolates had antimicrobial activity against Methicillin-resistant Staphylococcus aureus (MRSA) ATCC 33591 and Bacillus cereus ATCC 1399.
    Conclusion
    Isolated Micromonospora spp during this study, were able to produce the antimicrobial components against MRSA, one of the most challenge in the clinical setting.

    Aim and
    Background
    Screening of actinomycest for the production of novel antibiotics has been intensively evaluated for many years by scientists.
    Material And Methods
    From Lout desert, Sistan and Balouchestan and Khuzestan deserts, 30 soil samples were collected. The actinomycets from each sample were isolated and their antibiotics acivities agains pathogenic bacteria were investigated.
    Results
    A total of 300 actinomycetes isolates from soil samples were isolated. Out of 300 isolates, 24 isolates shown antimicrobial activity agains S. aureus, E. faecium, K. pneumonia and A. baumannii.
    Conclusion
    The study indicated that desert soil had diverse group of actinomycetes which some of the isolates had broader spectrum antibacterial activity which showed potential as a source of antibiotics for pharmaceutical interest.
