    ژن گیرنده پروتئین مورفوژنتیک استخوان (BMPR1B) یکی از ژن های بزرگ اثر است که نقش مهمی در افزایش میزان تخمک‏گذاری در گوسفندان دارد. بر این اساس، هدف از این مطالعه بررسی و مقایسه چندشکلی های موجود در اگزون 8 ژنBMPR1B و ارتباط آن با صفت چندقلوزایی در جمعیت گوسفندان نژاد لری بختیاری، شال، قزل و افشاری بود. لذا، نمونه خون از قوچ ها و میش های تک قلو و چندقلوزا اخذ و پس از تکثیر این جایگاه،جهت تعیین الگوی ژنوتیپی نمونه ها از روش PCR-SSCP استفاده شد. نتایج SSCP حضور سه الگوی ژنوتیپی را در این 4 جمعیت گوسفندان نژاد ایرانی نشان داد که بیشترین فراوانی مربوط به الگوی ژنوتیپی 1 (73%) و کمترین آن مربوط به الگوی ژنوتیپی 2 (8%) بود. همچنین نتایج نشان داد که در رابطه با اثر الگوهای ژنوتیپی اگزون 8 ژن BMPR1B بر میانگین تعداد نتاج در هر زایش، بین سه الگوی ژنوتیپی اختلاف معنی داری وجود داشت. گوسفندان دارای الگوی 2 با 7/1 بره در هر زایش و گوسفندان دارای الگوی 3 با 4/1 بره در هر زایش به ترتیب بیشترین و کمترین تعداد بره در هر زایش را داشتند. سپس، توالی یابی این قطعه تکثیر شده از اگزون 8 انجام شد. نتایج توالی یابی نشان دهنده وجود چهار جهش در توالی mRNA بود. جایگزینی باز C در موقعیت 855 که موجب ایجاد یک کدون توقف در موقعیت 266 (I266*) می شود. همچنین جهش تبدیلی تک نوکلئوتیدی در موقعیت-های G812T، T854A و C855A نیز مشاهده گردید که به موجب آن ها به ترتیب اسیدآمینه تریپتوفان به لیزین در موقعیت 219 (W219L)، فنیل آلانین به تیروزین در موقعیت 233 (F233Y) و فنیل آلانین به لیزین در موقعیت 233 (F233L) تبدیل گردید. به طور کلی، نتایج این تحقیق نشان داد که در اگزون 8 ژن BMPR1B در گوسفندان ایرانی نژادهای مختلف جهش های مهمی وجود دارد که میتوانند بر صفات مرتبط با تعداد نتاج در هر زایش موثر باشند.

    یکی از مهم ترین تنش های محیطی موثر در درختان پسته که به طور قابل توجهی باعث کاهش محصول می گردد، تنش شوری می باشد. در این آزمایش، الگوی بیان پروتئین و بررسی برخی خصوصیات مورفولوژیکی و بیوشیمیایی در پایه های پسته بادامی سفید و بادامی زرند تحت تنش شوری انجام گردید. این آزمایش به صورت فاکتوریل در قالب طرح کاملا تصادفی با سه تیمار و 20 تکرار در گلخانه ای با شرایط کنترل شده اجرا شد. در این پژوهش سه سطح تنش شوری شامل تیمار شاهد (آب مقطر)، تنش متوسط (60 میلی مولار کلریدسدیم) و تنش شدید (120 میلی مولار کلریدسدیم) اعمال گردید. زمانیکه اندازه دانهال به طول 15-20 سانتی متر رسید، تنش شوری اعمال و به مدت شش ماه ادامه یافت و در این مدت علائم تنش شوری (کاهش رشد رویشی و کاهش سطح برگ) مشخص و الگوی بیان پروتئین و برخی خصوصیات مورفولوژیکی و بیوشیمیایی تعیین گردید. نتایج نشان داد که الگوی بیان پروتئین و خصوصیات رشدی و بیوشیمیایی تحت تاثیر تنش شوری واقع شدند و دو پایه عکس -العمل های کاملا متفاوتی نشان دادند، به طوریکه کاهش شدت بیان و یا حذف باندهای پروتئینی در برگ پسته بادامی سفید بیشتر از برگ پسته بادامی زرند بود. نتایج نشان داد که کاهش رشد رویشی و کاهش میزان کلروفیل کل در پایه بادامی سفید بیشتر از پایه بادامی زرند تحت تنش شوری واقع شده است. همچنین نتایج نشان داد که با افزایش تنش شوری، میزان پرولین برگ و فعالیت آنزیم های آنتی اکسیدان در پایه بادامی زرند نسبت به پایه بادامی سفید افزایش یافت. با توجه به نتایج این پژوهش، به نظر می-رسد پسته بادامی زرند توانایی بیش تری در حفظ پروتئین های برگ و مقاومت بالاتری نسبت به تنش شوری دارد.

    تغییرات کارکرد ژنوم تحت شرایط تنش می تواند راهکاری برای درک بهتر سازوکارهای تحمل به تنش در گیاهان باشد. در این پژوهش سعی شده است تا با بررسی توالی های EST دو کتابخانه کنترل و تنش شوری در گندم، تغییرات کارکردی ژنوم در تنش شوری و ژن های موثر در تحمل به شوری شناسایی شوند. بدین منظور توالی های EST از وب گاه هاروارد و Graingenes دریافت شدند. به منظور دسته‏بندی توالی های EST، تعیین پروتئین های مرتبط با یونی ژن ها (کانتیگ ها و سینگل تون ها)، تعیین گروه های کارکردی و آزمون های آماری، به ترتیب از بانک اطلاعاتی Egassembler، بلاست وب گاه NCBI، وب گاه موسسه ماکس پلانک و نرم افزار IDEG6 استفاده شد. نتایج نشان دهنده وجود اختلاف معنی دار بین 20 گروه کارکردی در شرایط کنترل و تنش شوری بود. در هر دو شرایط کنترل و تنش شوری، گروه های کارکردی پروتئین و RNA بیشترین و گروه های کارکردی متابولیسم کربوهیدرات اصلی، تبدیل مواد آلی و چرخه کربس کمترین فعالیت کارکردی ژنوم را نسبت به سایر گروه های کارکردی ژنوم به خود اختصاص دادند. تحت تنش شوری، فعالیت گروه های کارکردی سنتز ATP/ انتقال الکترون میتوکندریایی، متابولیسم چربی، اکسایش-کاهش، پروتئین، RNA، DNA و سلول افزایش یافت. بنابراین می‏توان پیشنهاد کرد که این گروه های کارکردی دارای نقش مهمی در مکانیسم پاسخ به تنش شوری هستند. تجزیه و تحلیل بیان ژن‏ها در شرایط کنترل و تنش شوری نشان داد 271 ژن دارای بیان افتراقی معنی دار بودند که در 23 گروه کارکردی مختلف قرار گرفتند. ژن های شناسایی شده در این پژوهش می توانند جهت دست-ورزی ژنتیکی با هدف بهبود تحمل به تنش شوری در گیاهان زراعی مورد استفاده قرار گیرند.

    خارمریم (Silybum marianum L.) یکی از گیاهان دارویی مهم است که از زمان‏های باستان برای درمان بیماری کبد و بیماری های مرتبط با صفرا و همچنین مسمومیت حاد ناشی از مصرف قارچ‏های سمی مورد استفاده قرار می گرفت. شناخت تنوع ژنتیکی در گیاه خارمریم همانند سایر گیاهان کمک بزرگی در جهت تحقیقات به‏نژادی و بررسی و شناسایی تنوع موجود در ذخایر ژرم‏پلاسم این گیاه می‏نماید. هدف از این مطالعه بررسی تنوع ژنتیکی 23 اکوتیپ خارمریم مربوط به مناطق زیستی مختلف ایران با استفاده از نشانگرهای مولکولی ISSR بود. به این منظور از 9 نشانگر ISSR استفاده شد که برمبنای آنها بین اکوتیپ‏ها چندشکلی مشاهده گردید. این آغازگرها مجموعا 41 باند چندشکل و تکرارپذیر ایجاد نمودند. در این بررسی میانگین میزان محتوای اطلاعات چندشکلی (PIC)، میانگین شاخص اطلاعاتی شانون و میانگین شاخص ضریب تنوع نی به ترتیب برابر با 38/0، 30/0 و 20/0 بدست آمد. تجزیه خوشه‏ای با استفاده از الگوریتم UPGMA و ماتریس تشابه جاکارد، اکوتیپ‏های خارمریم را در 4 گروه اصلی تقسیم‏بندی نمود. در مجموع بین این گروه بندی و مناطق پراکنش جغرافیایی اکوتیپ ها تطابق کاملی وجود نداشت. نتایج این پژوهش مبین وجود تنوع ژنتیکی مناسب در بین اکوتیپ‏های خارمریم ایران جهت آغاز مطالعات پروژه های بهنژادی است.

    نقشه های ژنتیکی با تراکم و پوشش بالای ژنومی نقش بسزایی در تحقیقات پایه ای و کاربردی ژنتیک ایفا می کنند. در دهه های اخیر با پیدایش نشانگرهای DNA تحول عظیمی در تهیه و اشباع نقشه های ژنتیکی در گیاهان مختلف فراهم شده است. در این تحقیق از نشانگرهای رتروترنسپوزونی IRAP و REMAP و نشانگرهای ISSR جهت اشباع نقشه ژنتیکی جو در 149 فرد هاپلوئید مضاعف حاصل از تلاقی دو والد Clipper و Sahara استفاده شد. از چهارچوب نقشه ژنتیکی این جمعیت به عنوان نقشه پایه در تجزیه و تحلیل های بعدی استفاده شد. از 120 آغازگر به کار رفته، 6 آغازگر IRAP، 7 آغازگر REMAP و 3 آغازگر ISSR بین والدین چندشکلی نشان دادند که از این میان 11 باند چندشکل برای نشانگر IRAP، 8 باند چندشکل برای نشانگر REMAP و 4 باند چندشکل برای نشانگر ISSR بدست آمد. درکل 18 نشانگر چندشکل در جمعیت تفرق نشان دادند که از این تعداد 12 نشانگر به هفت گروه لینکاژی جو منتسب شدند. دو نشانگر از نسبت مندلی 1:1 انحراف نشان دادند. بیشترین تعداد نشانگرها به گروه لینکاژی دوم منتسب شدند. نشانگرهای رتروترنسپوزونی در این مطالعه به خوبی توانستند بخشی از نواحی خالی گروه های لینکاژی 1، 3، 5 و 6 را در نقشه ژنتیکی جو پر کنند. نتایج تحقیق نشان داد که نشانگرهای رتروترنسپوزونی می تواند بطور موثری جهت اشباع نقشه ژنتیکی جو مورد استفاده قرارگیرند.

    الگوهای حاصل از داده های مولکولی می توانند جهت شناسایی ارتباط میان جمعیت های یک گونه به کار گرفته شوند. در ایران، شتر به عنوان یکی از منابع مهم جهت تامین شیر، گوشت و الیاف کاربرد گسترده ای دارد. با استفاده از 8 جفت نشانگر ریزماهواره (YWLL08، VOLP03، VOLP08، YWLL38، CVR01، YWLL44، VOLP32 وVOLP67 ) ساختار و فاصله ژنتیکی جمعیت 81 نفره شتر، ارزیابی گردید. با استفاده از روش نمکی بهینه یافته، DNA ژنومی شترها استخراج و تکثیر جایگاه-های ریزماهواره از طریق واکنش زنجیره ای پلیمراز با موفقیت انجام شد و فرآورده های حاصل روی ژل پلی آکریل آمید 8% واسرشته ساز الکتروفورز شدند. بیشترین فاصله ژنتیکی بین جمعیت شهربابک و صحرای جهاد (56/0) و کمترین آن بین دو جمعیت رفسنجان (11/0) مشاهده گردید. نتایج PCA به بهترین شکل تعداد گروه های ژنتیکی موجود در مجموعه داده ها را توجیه کرد و نشان داد که کل نمونه ها به 3 مولفه اصلی تفکیک شدند. هم چنین نتایج خوشه بندی سلسله مراتبی نشان داد که در اولین سطح خوشه بندی، جمعیت های شهربابک، رفسنجان 1 و رفسنجان 2 در خوشه مشترکی قرار دارند و جمعیت شمشیرآباد خوشه متمایز دیگری را به خود اختصاص داده است، در حالی که جمعیت صحرای جهاد در یک خوشه کاملا مستقل قرار گرفته است. در کل نتایج حاصل از این مطالعه نشان دهنده مطابقت خوشه-بندی ژنتیکی جمعیت ها با فواصل جغرافیایی آنهاست.

    بیماری اسکلروتینیا از بیماری های قارچی مهم آفتابگردان در ایران می باشد که باعث کاهش رشد و عملکرد محصول می شود. در پژوهش حاضر واکنش 100 لاین آفتابگردان روغنی به 6 جدایه قارچ اسکلروتینیا مورد بررسی قرار گرفت و سپس به منظور شناسایی مکان های ژنی دخیل در مقاومت به بیماری از 128 جایگاه ژنومی تکثیر شده توسط آغازگرهای رتروترنسپوزونی IRAP استفاده شد. نتایج نشان داد که تنوع بالایی در ژر م پلاسم مورد مطالعه برای مقاومت به بیماری پوسیدگی اسکلروتینیایی ساقه وجود دارد. بررسی ساختار جمعیت، لاین های مورد مطالعه را به 3 زیرجمعیت احتمالی (3=K) تقسیم نمود. در تجزیه ارتباط با دو مدل GLM و MLM به ترتیب 14 و 11 مکان ژنومی پیوسته با مقاومت به بیماری (P≤0.01) شناسایی شد. نشانگرهایLTR 1063-65 ،LTR 1064-65 وLTR 1062 با ژن های کنترل کننده مقاومت به چندین جدایه قارچ عامل بیماری پیوسته بودند. نشانگرهای مشترک به دلیل تسهیل گزینش همزمان برای ایجاد مقاومت به چندین جدایه قارچی، می توانند در برنامه های به نژادی آفتابگردان نظیر انتخاب به کمک نشانگر (MAS) بخوبی استفاده شوند.

    میوستاتین عضوی از خانواده بزرگ فاکتور رشد تغییر شکل یافته B است که یک تنظیم کننده منفی مهم در رشد ماهیچه در مهره داران می باشد. در این مطالعه بخشی از توالی ژن میوستاتین (اینترون 1) در جمعیت شترهای ایران مورد بررسی قرار گرفت. با استفاده از روش توالی یابی، دو چندشکلی و یک حذف و اضافه در اینترون 1 این ژن شناسایی شد. جهش ها باعث دو جابجایی شامل تغییر C به T در موقعیت 16 و تغییر T به C در موقعیت 136 و همینطور یک حذف یا اضافه شدن باز T در موقعیت 218 شدند. جهش های مشاهده شده در سه موقعیت اینترون 1 باعث ایجاد سه هاپلوتیپ متفاوت شدند که هر سه هاپلوتیپ در دو گونه شترهای دوکوهانه و یک کوهانه مشترک بود و هیچ هاپلوتیپی محدود به یک منطقه خاص یا نژاد خاص نبود. آزمون تاجیما D به جز جمعیت ایستگاه طرود مثبت به دست آمد ولی از نظر آماری تفاوت معنی داری با صفر نداشت.

    Bone morphogenetic protein receptor-1B is one of the major genes significantly affect ewe ovulation rate. The aim of this study was to screen exon 8 of BMPR1B in single- and twin-birth ewes of Iranian Lori-Bakhtyari, Shal, Ghezel, and Afshari breeds. DNA was extracted from blood of ewes and rams from numerous sheep flocks and the genotype patterns were screened by PCR-SSCP approach. Results of SSCP analysis showed that there are three genotypic patterns in the four above-mentioned breeds. The highest abundant pattern and the lowest pattern were pattern 1 (73 %) and pattern 2 (8 %), respectively, in the whole flocks. There was a significant difference between these three genotypic patterns in terms of litter size attribute, so that ewes harboring Pattern 2 had the highest litter size average (1.7 lamb per parturition) while ewes carrying Pattern 3 had the lowest litter size average (1.4 lamb per parturition). Furthermore, the PCR amplicon from exon 8, containing these genotypic patterns, were sequenced. The sequencing results of candidate samples from different classes of SSCP showed that there were four new mutations in BMPR1B of Iranian sheep. Three of these mutations were nucleotide conversions, including G812T, T854A, and C855A mutations which caused to tryptophan to lysine (W219L), phenyl alanine to tyrosine (F233Y), and phenyl alanine to lysine (F233L) conversions, respectively. More importantly a C nucleotide insertion mutation corresponding to 855 nt of BMPR1B CDs was also observed which can cause to producing a stop codon at 266 amino acid position and consequently a dysfunctional polypeptide. In conclusion, these results clearly indicate the presence of important mutation in BMPR1B in Iranian sheep breeds which can significantly affect ewe fecundity attributes.

    Salinity stress is one of the most important environmental stresses affecting pistachio trees and causes decrease its production. In this experiment the effect of salinity stress on protein expression pattern, the content of proline and antioxidant enzymes (catalase, gayacol peroxidase, ascorbate peroxidase and superoxide dismutase) were carried out on two pistachio rootstocks including of Badami-Sefid and Badiami-Zarand. The experiment was performed as factorial based on Completely Randomized Design in a controlled greenhouse condition with three treatments and 20 replications. In this research, three salinity treatments were used include control (distilled water), moderate stress (60 mM NaCl) and severe stress (120 mM NaCl). After appearing the salt stress symptoms (decrease of vegetative growth and leaf area), protein pattern and some morphological and biochemical characteristics were determined. Results showed that protein expression pattern, vegetative growth and biochemical characteristics were affected by salinity treatments and responses of the two rootstocks were completely different. Under salinity stress, Badami-Sefid rootstock exhibited more protein bands reduction and elimination compared to Badami-Zarand. Results also showed that vegetative growth and total chlorophyll content in Badami-Sefid rootstock were decreased more than Badami-Zarand under salinity stress. Also, it has been found that by increasing salinity levels, proline content and antioxidant enzyme activities were higher in BadamiZarand compared to Badami-Sefid. It seems that Badami-Zarand rootstock has more ability to keep leaf proteins and therefore, higher resistance to salinity stress.

The goal of this study was to identify new genes related to salt stress and functional analysis based on expressed sequence tags (EST). Therefore, two EST libraries under control and salt stressed conditions were downloaded from Harvard and Graingenes databanks. EGassembler, NCBI BLAST, MaxPlanck and IDEG6 software tools were applied for clustering and assembling EST sequences, determination of proteins related to unigenes (contigs and singletons), functional categories and statistics test, respectively. Results showed that twenty functional categories are significantly different between control and stress conditions. Most of contigs and singletons fell into protein and RNA categories whereas CHO metabolism and TCA and organic transformation had the minimum of numbers of contigs and singletons in both control and salt stressed conditions. Percentage of unigenes in mitochondrial electron transport/ATP synthesis, Lipid metabolism, redox, protein, RNA, DNA, and cell categories were significantly higher in stress condition compared with control condition. Therefore, these functional categories 7Tcould be involved in7T 7Tthe mechanism of7T 7Tresponse7T 7Tto salt stress. 7TTwo hundred and seventy-one genes were differentially expressed that grouped into 23 functional categories. The wheat genes identified in this study can be considered as candidates genes to improve salt tolerance through genetic manipulation.

    Milk Thistle (Silybum marianum L.) is one of the important ancient medicinal plants that have been used to treat liver disease, bile-related diseases and poisoned individuals by poisonous mushrooms. Assessment of genetic variation in Milk Thistle like the other important plants is a key component in breeding programs. The aim of this study was studying genetic variation of some Iranian Milk Thistle ecotypes using ISSR marker. A total of 41 repeatable polymorphic marker loci were produced by 9 ISSR primers. The average of polymorphism information content (PIC), Shannon’s information index and Nei’s gene diversity were 0.38, 0.49 and 0.33, respectively. Using cluster analysis by UPGMA algorithm and Jaccard similarity matrix, different ecotypes of Milk thistle grouped in 4 main clusters. We did not find an exact match between dendrogram grouping and geographical grouping of ecotypes. This could be due to the high diversity of this plant or relocation of ecotypes. The results indicate existence of a significant variation among Iranian Milk thistle ecotypes to start the breeding programs.

    Genetic maps with high density and wide genome coverage play significant role in basic and applied genetic researches. In recent decades, with the advent of DNA markers, the huge development has been created in genetic map preparation and saturation in various plant species. In this study, IRAP, REMAP and ISSR markers were applied to saturate the genetic map of barley in a population with 149 double haploid individuals derived from a cross of Clipper and Sahara. The basic genetic map of this population was used as a frame for further analysis. In general, among the 120 primers used, 6 IRAP, 7 REMAP and 3 ISSR primers showed polymorphism between parents which generated 11 IRAP, 7 REMAP and 4 ISSR markers. Eighteen markers segregated in population, out of which 12 markers assigned to 7 barley linkage groups. Two markers deviated from the ratio of 1:1. The maximum number of markers was assigned to linkage group 2. In the current investigation, retrotransposon-based markers were able to saturate the gaps of barley genetic map on linkage groups 1, 3, 5 and 6. The results showed that retrotransposon markers can be effectively used for saturation of barley genetic maps.

    Patterns in heritable molecular data can be used to summarize the relationships between populations of a species. In Iran, camels are providers of milk, meat and fibers. Using of 81 reproductive individuals belonging to five sampling locations of Camelus dromedarius in Kerman province, eight microsatellite markers 3T(3TYWLL08, VOLP03, VOLP08, YWLL38, CVR01, YWLL44, VOLP32 and VOLP673T)3T were analyzed to genetic structure and genetic distance in these populations. DNA extraction was conducted with optimized and modified salting-out method. The polymerase chain reactions for 81 individuals were successfully done with all primers and then amplification products were resolved on 8% polyacrylamid gel and stained with silver nitrate. The highest genetic distance obtained between Shahr-e Babak and Sahra-e Jahad (0.56) populations and the lowest genetic distance was observed between the two populations of Rafsanjan (0.11). Principal component analysis indicated that all samples could be mainly regrouped into three main clusters and most suitable number of genetic groups in dataset was explained. Results of this study indicated that in the first level of clustering, Shahr-e Babak, Rafsanjan1 and Rafsanjan 2 populations shared the same cluster and Shamshir Abad had another separate cluster while Sahra-e Jahad population appeared as an independent cluster. A similarity of geographical distribution was in good accordance with founded genetic relationships in this study.

    Sclerotinia disease is important fungal disease of sunflower in Iran that reduces its growth and yield. In this study, reactions of 100 oily sunflower lines to 6 fungal isolates of Sclerotinia were studied and then 128 retrotransposon-based molecular markers (IRAP) were used to identify genes controlling disease resistance. The results showed that there is high diversity in the studied germplasm for resistance to the Sclerotinia stem rot. Population structure analysis subdivided the studied lines into 3 subpopulations (K=3). Association analysis with general and mixed linear models (GLM and MLM) identified 14 and 11 loci, respectively that are significantly associated (P≤0.01) with Sclerotinia disease resistant genes. Markers LTR 106365, LTR 1064-65 and LTR 1062 were commonly associated with resistant to some fungal isolates. The common markers due to facilitating simultaneous selection for resistance to several fungal isolates can be well used in sunflower breeding programs like marker-assisted selection (MAS).

    Myostatin (MSTN) is a member of the transforming growth factor-β superfamily (TGFβ) and is an important negative regulator of muscle growth in vertebrates. In this study, a partial sequence of MSTN gene (intron 1), in the camel population of Iran, was evaluated. Two single nucleotide polymorphisms and one insertion or deletion (InDell) were identified in intron 1 of the myostatin gene, using sequencing method. The mutations caused, two substitutions including C to T and T to C in positions 16 and 136, respectively and one InDell of T in position 218. The mutations observed in three positions of intron 1, created three different haplotypes. These haplotypes were shared among Bacterianus and Dromedaries camel populations with no haplotype restricted to a specific region or a single camel breed. Tajima D were positive for all populations except Trod station, but it wasn’t significant.
