صفحات 1-18هدفگون Astragalus verus یکی از گونه های خوب برای تولید کتیرا است. این مطالعه با هدف بهینه سازی کشت سوسپانسیون سلولی در این گیاه انجام شد.مواد و روش هاابتدا بهینه سازی کالوس زایی و سپس بهینه سازی سوسپانسیون سلولی انجام شد. آزمایش کالوس زایی در قالب فاکتوریل با ریزنمونه های ریشه، هیپوکوتیل و کوتیلدون در محیط کشت MS حاوی BAP در ترکیب با 2,4-D و NAAدر پنج تکرار انجام شد. در آزمایش بهینه سازی سوسپانسیون سلولی، کالوس های حاصل از محیط کشت NAA و 2,4-D در قالب طرح پایه کاملا تصادفی با 16 تیمار هورمونی در سه تکرار بررسی شدند. همچنین کاربرد تیمار های تاریکی، اسیدآسکوربیک، پلی وینیل پیرولیدون و کلات کننده آهن EDTA جهت کنترل ترکیبات فنولی بررسی شد.نتایجبیشترین درصد کالوس زایی در محیط کشت 2,4-D از ریزنمونه ریشه با میانگین 67 درصد بدست آمد. بهترین تیمار هورمونی سوسپانسیون سلولی شامل سه میلی گرم در لیتر2,4-D در ترکیب با 5/0 میلی گرم در لیتر Kin با میانگین 277/0 زنده مانی، و 040/0 گرم وزن خشک (در پنج میلی لیتر) بود. در مطالعه تاثیر مهار کننده های قهوه ای شدن بیشترین وزن خشک از تیمار 100 میلی گرم در لیتر اسید اسکوربیک با میانگین 76/0گرم (در پنج میلی لیتر) بدست آمد که در جلوگیری از قهوه ای شدن بسیار موثر بود.نتیجه گیرینتایج این پژوهش نشان داد که 2,4-D می تواند به تنهایی جهت القای کالوس مناسب و در ترکیب با Kin برای تولید سوسپانسون سلولی گون قابل استفاده باشد. همچنین مشخص شد که اسید اسکوربیک می تواند به خوبی فرایند قهوه ای شدن سلول های سوسپانسیون سلولی را کنترل نماید. نتایج این مطالعه می تواند جهت تولید درون شیشه ای کتیرا مورد استفاده قرار گیرد.کلیدواژگان: Astragalu sverus، اسیداسکوربیک، سوسپانسیون سلولی، قهوه ای شدن

صفحات 19-34هدفقارچ های میکوریز با دامنه وسیعی از گیاهان همزیست هستند و نقشی کلیدی در پایداری اکوسیستم دارند. برخی از گونه های قارچ های میکوریز قابلیت سازگاری با شرایط سخت را دارند. تحقیقات کمی بر روی تنوع قارچ های میکوریز در ایران انجام گرفته است. این تحقیق به منظور شناسایی قارچ های میکوریز درونزی همزیست با درختان پسته در شرایط شوری و خشکی شدید و مقایسه نتایج با جدایه های برخی گیاهان غیر زراعی انجام شده است.مواد و روش هادر این بررسی ابتدا تعداد 56 نمونه ریزوسفری از درختان پسته و 40 نمونه ریزوسفری از گیاهان غیرزراعی جمع آوری و درصد کلنیزاسیون ریشه آنها تعیین شد. سپس 37 جدایه که درصد کلنیزاسیون بالاتری داشتند انتخاب و با استفاده از گیاه سورگوم در گلدان تکثیر شدند. در مرحله بعد تعداد هشت جدایه برتر (چهار جدایه از پسته و چهار جدایه از گیاهان غیر زراعی) بر روی گیاه اندروپگون (Andropogon gerardii) تلقیح و شناسایی مولکولی به روش qPCR انجام شد.نتایجدرصد کلنیزاسیون نمونه های پسته در گلخانه بر روی سورگوم نسبت به پسته در شرایط مزرعه بیشتر بود در حالی که در گیاهان بومی درصد کلنیزاسیون نسبت به سورگوم بیشتر بود. قارچ Rhizophagus بر اساس ویژگی های ریخت شناختی اسپورها در خاک، در تمام نمونه های پسته بجز نمونه 42 شناسایی گردید. سه جنس Claroideoglomus،Funneliformis و Rhizophagus جنس های غالب باغ های پسته بودند.نتیجه گیریتنوع گونه های قارچ میکوریز در مناطق غیر کشاورزی بیشتر از باغ های پسته بود. بیشترین سرعت کلنیزاسیون ریشه مربوط به Diversispora می باشد و جدایه های Funneliformis، Rhizophagus، Claroideoglomus و Racocetra در رتبه های بعدی قرار گرفتند.کلیدواژگان: اندروپگون، تنوع گونه ای، جدایه غالب، شناسایی مولکولی

صفحات 35-53هدفسیب با نام علمی Malus domestica Borkh از تیره گلسرخیان (Rosaceae) یکی از مهمترین درختان میوه در جهان است. تخمین میزان تنوع ژنتیکی به عنوان یکی از گام های پایه ای و اساسی در نگهداری و حفاظت مواد ژنتیکی در بانک های ژنی و اجرای برنامه های بهنژادی است. هدف این تحقیق تعیین تنوع ژنتیکی 22 رقم سیب جمع آوری شده از کلکسیون مرکز تحقیقات جهاد کشاورزی خراسان رضوی و برخی ارقام موجود در شهرستان جیرفت با استفاده از نشانگر مبتنی بر واکنش زنجیره ای پلیمراز ISSR بود.مواد و روش هاپس از استخراج DNA از گیاهان مورد مطالعه، PCR با پنج آغازگر انجام شد.نتایجکولتیوارهای سیب 48/81 درصد چندشکلی داشتند. با آنالیز خوشه ایارقام در شش گروه طبقه بندی شدند. در گروه اول ارقام محلی گناباد، طبسی شماره 3، قاسم آبادی، محمدی، گلشاهی اوغاز، گلاب اصفهان، سیب قرمز دماوند و گرانی اسمیت قرار گرفتند. گروه دوم شامل ارقام زرد لبنان، سیب قرمز خونی لبنان، محلی ساردو، گلاب ساردو، مشعلی، مربایی مشهد، علی موری دوم رس، اربابی و سیب شماره 3 بود. گروه سوم و چهارم به ترتیب شامل ارقام شیخ احمد تبریز و شفیع آبادی بودند. گروه پنجم گل مشهد و گروه ششم عباسی ازغده و اطلسی طبس را شامل شدند. در این مطالعه آغازگر 842 بیشترین چند شکلی را در بین ارقام نشان داد و بین گروهبندی ژنتیکی و مناطق جغرافیایی ارقام رابطه ای وجود نداشت. قرار گرفتن ژنوتیپ های مناطق مختلف در گروه های یکسان می تواند احتمالا به دلیل انتقال ژنوتپ ها بین مناطق در زمان گذشته باشد.نتیجه گیرینتایج این مطالعه کارآمد بودن نشانگرهای ISSR برای شناسایی نواحی چند شکلی، تخمین فاصله ژنتیکی و مدیریت ژرم پلاسم ارقام سیب را نشان می دهد.کلیدواژگان: تنوع ژنتیکی، سیب، نشانگر مولکولی، ISSR

صفحات 55-74هدفشناسایی مسیرهای موثر در ایجاد تحمل نسبت به تنش شوری یکی از مباحث جذاب در علوم گیاهی است که در این راستا روش های جدید داده کاوی نگرش جدیدی برای محققان ایجاد کرده است. در این پژوهش، الگوریتم های انتخاب ویژگی (feature selection) و درخت تصمیم گیری (decision tree, DT) به منظور شناسایی نشانگرهای بیوشیمیایی در تحمل به شوری استفاده شد.مواد و روش هادر این راستا، به منظور بررسی برخی نشانگرهای بیوشیمیایی از جمله میزان پروتئین، آنزیم های آنتی اکسیدانی سوپراکسید دیسموتاز، پراکسیداز، کاتالاز، آسکوربات پراکسیداز، پرولین، میزان سدیم و پتاسیم در ارقام گندم و خویشاوند وحشی آنAegilops crassa ، آزمایشی به صورت فاکتوریل در قالب طرح کاملا تصادفی با سه تکرار انجام شد. فاکتورهای آزمایشی شامل ژنوتیپ (ارقام گندم زراعی ارگ (مقاوم به شوری) و الموت (حساس به شوری)، و یک خویشاوند وحشی (Ae. crassa)) و تنش شوری (کلرید سدیم mM150 و mM 0) بودند.نتایجنتایج این دو راهکار نشان داد که ترکیب سلسله مراتبی پرولین، سوپراکسید دیسموتاز و آسکوربات پراکسیداز می تواند به عنوان نشانگرهای بیوشیمیایی برای انتخاب ژنوتیپ متحمل به تنش شوری استفاده گردد. دو درخت تصمیم گیری دارای بیشترین کارایی در پیش بینی ژنوتیپ حساس و متحمل بودند. این درخت های تصمیم گیری عبارت بودند از مدل درخت تصمیم گیری Ratio DT Parallel Gain با کارایی 5/97 درصد که روی پایگاه داده ای Info Gain Ratio اجرا شد و دیگری DT Gain Ratio با کارایی 67/91 درصد که روی پایگاه داده ای Ruleاجرا شد.نتیجه گیریدر مجموع، نتایج حاصله بیانگر آن بود که تلفیق مطالعات بیوانفورماتیک و آزمایشگاهی می تواند منجر به شناسایی مسیرهای مرتبط با تنش شوری و فهم بهتر مکانیسم های کنترل کننده تحمل به تنش شود.کلیدواژگان: انتخاب ویژگی، درخت تصمیم گیری، گندم، نشانگرهای بیوشیمیایی

صفحات 76-92هدفگیاه گل راعی (Hypericum perforatom) متعلق به خانواده هایپریکاسه بوده و از زمان یونان باستان به عنوان یک گیاه دارویی برای درمان اضطراب، افسردگی، زخم و سوختگی استفاده شده است. در مطالعه حاضر، اثر غلظتهای مختلف CPPU و BA (هر کدام در سه غلظت) در ترکیب با دو غلظت IAA بر کالوسزایی و باززایی شاخساره گل راعی از ریزنمونههای برگ و میانگره به منظور تعیین مناسبترین غلظت تنظیم کنندههای رشد اکسینی و سیتوکینینی و مشخص نمودن مناسب ترین ترکیب هورمونی برای به دست آوردن بیشترین باززایی شاخساره در محیط کشت پایه MSانجام گرفت.
مواد و روشها: این آزمایش در قالب آزمایش فاکتوریل بر پایه طرح کاملا تصادفی در سه تکرار و پنج ریزنمونه در هر تکرار مورد بررسی قرار گرفت و صفات وزن تر کالوس، تعداد شاخساره، طول شاخساره و تعداد گره بلندترین شاخساره اندازهگیری و بررسی شد.نتایجبراساس نتایج حاصل در بین تیمارهای هورمونی، ترکیب هورمونی 5/1 میلیگرم در لیتر CPPU و 1 میلیگرم در لیتر IAA بیشترین اثر را روی باززایی گل راعی داشت که اثبات کننده نقش تنظیم کنندگی سیتوکینینیCPPU در باززایی شاخساره در گل راعی به خوبی مشاهده گردید.نتیجه گیریغلظت 1 میلیگرم درلیتر IAA به همراه 5/1 میلیگرم در لیتر CPPU یا BA باعث افزایش معنیداری در باززایی شاخساره گردید. همچنین، غلظت 1 میلیگرم در لیتر IAA در ترکیب با 5/0 میلیگرم در لیتر BA باعث افزایش وزن تر کالوس در گل راعی شد.کلیدواژگان: باززایی، IAA، علف چای، CPPU، BA -
صفحات 93-109هدفزیتون به عنوان یکی از مهم ترین گیاهان باغی دارای تعداد ارقام فراوانی در جهان است. لذا، هدف این پژوهش، مطالعه تنوع ژنتیکی مجموعه ای از ارقام زیتون ایستگاه طارم ایران بود.مواد و روش هادر این تحقیق از 25 نشانگر CBDP برای بررسی روابط و تنوع ژنتیکی 20 رقم زیتون تجاری و امیدبخش ایرانی و خارجی استفاده شد.نتایج25 آغازگر مورد استفاده در مجموع 755 قطعه (آلل) چند شکل تکثیر دادند و میانگین محتوای اطلاعات چند شکلی (PIC) و شاخص نشانگری (MI) آغازگرها به ترتیب 941/0 و 64/5 بود که بیانگر کارایی و قدرت تمایز بالای نشانگرهای CBDP می باشد. پارامترهای ژنتیکی تنوع ژنی نی (H) و شاخص شانون (I) در ارقام خارجی نسبت به ارقام ایرانی بالاتر بودند. جریان ژنی (NM) بالایی (46/4) بین ارقام زیتون مشاهده شد که این نتیجه مقادیر پایین شاخص های تمایز بین جمعیتی (Gst) و FsT را تایید کرد. تجزیه واریانس ملکولی (AMOVA) به ترتیب 17 و 83 درصد از تغییرات کل ژنتیکی را به تنوع بین گروهی (ارقام ایرانی و خارجی) و درون گروهی تفکیک کرد. تجزیه خوشه ایبا استفاده از ضریب تشابه دایس و روش UPGMA ارقام ایرانی و خارجی را در پنج گروه قرار داد که رقم امیدبخش T24 و رقم خارجی Koroneiki با حداکثر تفاوت با بقیه هر کدام در یک گروه جداگانه قرار گرفتند. تجزیه به مختصات اصلی (PCoA) توانست به خوبی ارقام امیدبخش ایرانی را از ارقام تجاری ایرانی و خارجی تفکیک کند. تجزیه ساختار (Structure) ژنتیکی جمعیت نیز نتایج حاصل از تجزیه کلاستر و PCoA را تایید کرد و دو گروه ارقام ایرانی و خارجی به وضوح از هم تفکیک شدند.نتیجه گیریدر جمع بندی کلی مشاهده شد که رقم امیدبخش T24 با دارا بودن شباهت ژنتیکی مشترک به هر دو گروه ارقام ایرانی و خارجی می تواند رقم کاندید مناسبی جهت تجاری سازی با دارا بودن خصوصیات مکمل هر دو گروه باشد. همچنین استفاده از نشانگرهای CBDP برای بررسی تنوع ژنتیکی سایر ارقام زیتون نیز توصیه می شود.کلیدواژگان: تجزیه ساختار، جعبه CAAT، رقم امیدبخش، نشانگر آگاهی بخش

صفحات 111-131مقدمهسرما یکی از تنش های مهم مناطق سردسیر و معتدل است و در برنامه های اصلاحی عدس، بهبود تحمل به سرما در اولویت قرار دارد. فاکتور های رونویسی NAC در واکنش به تنش های مختلف غیرزیستی نقش مهمی دارند.مواد و روش هادر این پژوهش به منظور ارزیابی بیان ژن های MfNAC، MtNAC57 و GmNAC2، گیاهان رقم گچساران عدس تحت پنج تیمار مدت زمان تنش سرما در قالب طرح تصادفی کامل با سه تکرار قرار گرفتند. تیمارهای مدت زمان تنش سرمایی °C4-2 شامل h6، h12، h24 و h48 بود که روی گیاهان 29 روزه اعمال شد و گیاهان رشد یافته در شرایط گلخانه (دمای °C23) به عنوان شاهد در نظر گرفته شدند. استخراج RNA از بافت ها، با استفاده از روش لیتیم کلراید و ساخت cDNA با استفاده از کیت TaKara انجام شد. واکنش Real-time PCR برای ژن های مورد نظر در هر دو نوع اندام هوایی و زمینی در سه تکرار بیولوژیک و دو تکرار تکنیکی انجام شد. از ژن Actin به عنوان کنترل داخلی استفاده شد.نتایجنتایج تجزیه واریانس بیان ژن ها نشان داد که بین سطوح مختلف تیمارهای مدت زمان تنش دمایی در هر دو اندام اختلاف معنی داری وجود داشت. بیان ژن GmNAC2 در اندام هوایی، 6 ساعت پس از تنش کاهش معنی داری یافت در حالی که در اندام زمینی در تیمار 24 ساعت پس از تنش بیان این ژن کاهش نشان داد. بیان ژن MtNAC57 در تیمارهای h6، h12 و h24 نسبت به شاهد کاهش نشان داد ولی در اندام زمینی تیمار h12 افزایش معنی دار مشاهده شد. ژن mfNAC در اندام هوایی در همه تیمارها نسبت به شاهد کاهش بیان معنی دار نشان داد در حالی که در اندام زمینی فقط در تیمار بلند مدت h48 افزایش بیان معنی دار مشاهده شد.نتیجه گیریدر این تحقیق، بیان برخی از ژن های خانواده NAC تحت تنش سرما در عدس برای اولین بررسی شد و نتایج حاصل می تواند برای مطالعات به نژادی این گیاه و سایر حبوبات مفید واقع شود.کلیدواژگان: اندام هوایی، ریشه، سرما، عدس، PCR زمان واقعی

صفحات 134-146هدفاولین آنزیم مسیر اختصاصی بیوسنتز آلکالوئیدهای پیرولیزیدینی (PA)، هموسپرمیدین سینتاز (HSS) است. HSS انتقال NAD1 وابسته به یک گروه آمینوبوتیل اسپرمیدین را به پوترسین کاتالیز می کند. پوترسین، اسپرمیدین و اسپرمین سه پلی آمینی هستند که بطور طبیعی دارای بار مثبت بوده و تقریبا در تمام سلول های زنده یافت می شوند. این ترکیبات به DNA متصل شده و در تعدادی از فرآیندهای حیاتی مانند تقسیم سلولی، تمایز و عملکرد غشاء دخالت دارند. گزارش های بسیار اندکی در مورد بیان ژن هموسپرمیدین سینتاز 1 (HSS1) در گیاه دارویی زلف پیر (Senecio vulgaris) به خصوص در شرایط طبیعی گزارش شده است. هدف از پژوهش حاضر بررسی بیان ژن هموسپرمیدین سینتاز 1 در بافت های مختلف گیاه دارویی زلف پیر (S. vulgaris)با استفاده از PCR در زمان واقعی بود.مواد و روش هاRNA کل از ریشه ها، شاخساره ها (ساقه ها و برگ ها) و گل ها استخراج شد و به cDNA تبدیل شد. آغازگرهای اختصاصی برای ژن HSS1 و نیز برای Pol II(به عنوان ژن خانه دار مرجع در آزمایش)، طراحی شد و بیان نسبی ژن با استفاده از روش پی سی آر در زمان واقعی بررسی شد.نتایجمنحنی ذوب دو ژن نشان دهنده تکثیر اختصاصی در PCR بود. ارزیابی سطح بیان نرمال شده ژن HSS1 نشان داد که این ژن به صورت متفاوتی در اندام های مختلف S. vulgaris بیان شده است. حداکثر بیان نسبی (تغییرات بیش از 59 برابر نسبت به ژن مرجع Pol II) در ریشه هایی که هموسپرمیدین سینتاز فعال است و هموسپرمیدین تولید می شود مشاهده شد. علاوه بر آن این ژن در گل ها نیز به میزان کمتری (افزایش 4 برابر نسبت به ژن خانه دار Pol II). بیان شد. طبق انتظار، این ژن بیان نسبی قابل توجهی در شاخساره ها یعنی جایی که هموسپرمیدین حمل می شود، نشان نداد.نتیجه گیریبیان ژن HSS1 در گیاه زلف پیر به صورت ویژه بافتی است و در ریشه ها بیشتر از دیگر بافت ها می باشد.کلیدواژگان: بیان ژن، پی سی آر در زمان واقعی، زلف پیر، شرایط طبیعی، هموسپرمیدین سینتاز

صفحات 147-170هدفاپتوژنتیک به عنوان یک شاخه علمی جدید، از فتورسپتورها به منظور اجرای تحقیقات پایه ای و کاربردی استفاده می کند. یکی از راه های توسعه اپتوژنتیک، شناسایی فتورسپتورهای جدید و تعیین خصوصیات آن هاست. کریپتوکروم های DASH متعلق به خانواده پروتئینی کریپتوکروم/فتولیاز، فتورسپتورهای دارای قابلیت ترمیم DNA بوده و در مسیرهای پیام رسانی علامت نقش دارند. هدف این تحقیق، تولید کافی پروتئین پیش گویی شده کریپتوکروم DASH1 ولوکس (VcCryDASH1)، برای استفاده در مطالعات طیف جذبی و نیز بررسی قابلیت ترمیم DNA می باشد.مواد و روش هاابتدا توالی کدکننده پروتئین VcCryDASH1 با استفاده از آغازگرهای اختصاصی، از کتابخانه cDNA جلبک Volvox carteri جداسازی و در پلاسمید بیانی pGEX-2TK همسانه سازی شد. تولید پروتئین نوترکیب در دو سویه متفاوت باکتری E. coli و همچنین تحت تاثیر زمان های مختلف القاء با IPTG و همچنین میزان انحلال پذیری آن توسط روش SDS-PAGE مورد ارزیابی قرار گرفت. بررسی قابلیت اتصال به DNA در این پروتئین، با روش همردیف سازی انجام شد.نتایجتولید کافی پروتئین، در سویه Rosetta، در °C32، و زمان القاء بیش از 3 ساعت بدست آمد. در زمان القاء 1 و 3 ساعت، نیمی از پروتئین به صورت نامحلول بود. همردیف سازی پروتئین VcCryDASH1 مشخص نمود که درصد بالایی از آمینواسیدهای متصل شونده به DNA، در این پروتئین حضور دارند.نتیجه گیریاین نتایج نشان می دهند که پروتئین VcCryDASH1 تحت شرایط بهینه ، دارای قابلیت انحلال پذیری دوگانه بوده اما روش ما می تواند مقدار کافی آن را در باکتری E. coli سویه Rosetta تولید کند. همچنین بدلیل داشتن آمینواسیدهای حفاظت شده ، این پروتئین دارای قابلت اتصال به DNA می باشد که آن را به عنوان یک منبع احتمالی جهت طراحی مولکول های میانجی نور-DNA در اپتوژنتیک معرفی می نماید.کلیدواژگان: سویه Rosetta، کریپتوکروم DASH1، وکتوربیانی، Volvox carteri

Pages 1-18ObjectiveAstragalus verus is one of best species for tragacanth production. The aim of this study was optimization of cell suspension culture in this plant.Materials and methodsFirst, the callus production was optimized then optimization of cell suspension was performed. The callus production experiment was conducted in a factorial arrangement with explants of root, hypocotyl and cotyledon in MS medium containing BAP in combination with 2,4-D and NAA. In experiment of cell suspension optimization, callus that derived from medium containing NAA and 2, 4-D were investigated using 16 hormonal treatments and three replications in a completely randomized design. Also, it was investigated the application of dark treatments, ascorbic acid, poly-vinyl pyrrolidone and EDTA iron chelate to avoid phenolic compounds.ResultsThe results showed that the highest callus production percentage (average of 67%) was obtained in the medium of 2,4-D with root explant. The best hormone treatment of cell suspension was 3 mg /L 2,4-D in combination with 0.5 mg/L Kin with an average of 0.277 viability, and 0.040 g dry weight (at 5ml). In the study of the effect of browning inhibitors, the highest dry weight was obtained with 100 mg/l ascorbic acid in cell culture medium with an average of 0.76 g (at 5ml). This treatment was effective in prevention phenolic compounds.ConclusionsThe results of this study showed that 2,4-D could be used alone for inducing proper callus and in combination with Kin for the production of cell suspension. It was also found that ascorbic acid can well control the process of browning cells of cell suspension. Pages 19-34ObjectiveArbuscular mycorrhizal fungi (AMF) establish symbiosis with a great number of plant species. This group of fungi plays an important role in ecosystem stability. Some AMF species can quickly adapt to stress conditions. The diversity of native AMF is less studied in Iran. In the present study, we identified AMF symbionts of pistachio trees under salinity and drought stress. Furthermore, the results were compared with some native plants in Kerman province.Materials and methodsIn this study, 56 and 40 samples from the rhizosphere of pistachio trees and native plants were collected respectively. After root colonization assessment, 37 samples with more colonization percentages were selected as initial inoculums. Inoculums prepared with sorghum plant in a pot experiment. In the next step, molecular identification of eight isolates (four isolates from pistachio and four isolates from the native plant) was performed using qPCR method in root samples of Andropogon gerardii.ResultsThe result showed that root colonization was lower in pistachio than sorghum plants which were in contrast to native plants. Morphological identification of AMF spores showed all samples were contained Rhizophagus except for sample ID 42 of pistachio orchards. Claroideoglomus, Funneliformis, and Rhizophagus were found as dominant species in pistachio orchards.ConclusionsThe present study shows that AMF diversity was higher in native plants than pistachio. Evaluation of genetic diversity is one of the basic steps for plant breeding and preserving of germ plasm through gene banks.Materials and methodsThe aim of this study was evaluation of the genetic diversity of 22 varieties of apples collected from Khorasan Aagricultural Research Center and some region of Jiroft using ISSR markers based on the polymerase chain reaction (PCR).Materials and methodsDNA extraction was done and then PCR conducted by five ISSR-primers.ResultsApple cultivars had 81.48% polymorphism. Cluster analysis classified them into six groups. The first group had five apple cultivars from Khorasan Razavi including Mohali Gonabad, Tabasi number3, Ghasemabai, Mohamadi and Golshahi Oghaz as well as Golab Esfehan, Sibe Ghermez and Gerani Esmith from Jiroft. Genotypes of Zard Lobnan, Sib Ghermez Khoni, Mohali Sardo, Golab Sardo from Jiroft and Mashali, Morabei Mashhad, Ali Mori Dovom Ras, Arbabi, SIB number3 from Khorasan Razavi categorized into the second group. Genotypes of Shaikh Ahmad Tabriz and Shafiabadi were placed in the third and forth group respectively. The fifth and sixth groups include Gol Mashhad and Abasi Azghade, Atlasi Tabas from Khorasan Razavi. Also, ISSR primer 842 showed the highest polymorphism among apple cultivars. There is no correlation between the geographical origin and genetic grouping of apple cultivars. Probably, setting of different regions genotypes in the same genetic group is due to transport of genotypes among regions in past.ConclusionsResults indicated that ISSR is an effective technique for identification of polymorphism, genetic distance and germplasm management among apple cultivars.Keywords: Apple, Genetic Diversity, ISSR, Molecular Marker

Pages 55-74ObjectiveThe recognition of pathways involved in salt tolerance mechanisms is one of the interesting topics in plant sciences and the new data-mining techniques provide a new insight for researchers. In this research, various attribute weighting and decision tree (DT) algorithms were executed to discover biochemical markers linked to salinity tolerance.Materials and methodsIn this regard, to assess some biochemical markers such as protein, superoxide dismutase, peroxidase, catalase, ascorbate peroxidase, proline, sodium and potassium in wheat and its wild relative Aegilops crassa, a factorial experiment was conducted in a completely randomized design with three replications. The factors in the study were genotypes (Arg (salt tolerance) and Alamout (salt sensitive), and a wild relative (Ae. crassa)) and salinity (sodium chloride 0 mM and 150 mM).ResultsAccording to these approaches, proline, superoxide dismutase and ascorbate peroxidase can be used as biochemical markers to screen wheat genotypes for salt tolerance. Feature selection and decision tree results showed that hierarchy combination of proline, superoxide dismutase and ascorbate peroxidase can be used as biochemical markers for selection of salinity tolerant wheat. The DT Parallel Gain Ratio model when run on the Info Gain Ratiodataset and the DT Gain Ratiomodel when run on the Ruledataset were the best model in distinguishing sensitive and tolerant genotypes with 97.5% and 91.67% performances, respectively.ConclusionsOverall, the results showed that combining bioinformatics, and laboratory studies can lead to identifying pathways associated with salinity and provide a better understanding of the mechanisms underpinning stress tolerance.

Pages 76-92ObjectiveSt. John’s wort belongs to Hypericaceae family and has been used since ancient Greeks as a herb for the treatment of anxiety, depression, wounds and burns. In present study, the effects of different concentrations of CPPU and BA (each in three concentrations) in combination with two concentrations of IAA on callus production and shoot regeneration of Hypericum perforatum from leaf and stem explants were evaluated to identify the best growth regulator combination for obtaining the highest yield of in vitro regenerated shoots.Materials and methodsThis was implemented in a CRD-based factorial experiment with three replicates and five samples in each replicate and the traits including callus fresh weight, number of shoots, shoot length, length of the tallest shoots, and the number of nodes of the tallest shoots were measured and examined.ResultsResults showed that the most effective combination for regeneration was 1.5 mg/l CPPU and 1 mg/l IAA, which may be a proof for more effectiveness of CPPU in comparison with BA as cytokinin source on shoot regeneration in Hypericum perforatum.ConclusionsThe use of 1 mg/l IAA in combination with 1.5 mg/l CPPU or BA resulted in a significant increase in shoot regeneration. Pages 93-109ObjectiveThe olive is considered as one of the most important horticultural crops, with more abundant number of cultivars. Thus, the aim of this research was to study the genetic diversity in a set of olive cultivars in Taroum station of Tran.Materials and methodsIn this research, 25 CBDP markers were used to study the genetic diversity and relationships of 20 promising and commercial Iranian and foreign varieties of olive.ResultsThe 25 used primers amplified a total of 755 polymorph alleles, and the mean of PIC and MI indices were 0.941 and 5.64, respectively, indicating the high efficiency and differentiation power of CBDP markers. The genetic parameters of Nei and Shannon indices were higher in foreign cultivars than those of Iranian cultivars. The high gene flow (4.46) was observed among olive cultivars, which confirmed the low values of the inter-population differentiation (Gst) and FsT indices. Analysis of molecular variance (AMOVA) differentiated the total genetic variation into inter-group (Iranian and foreign cultivars) (17%) and within-group (83%) diversity. The cluster analysis using Dice coefficient and UPGMA method divided the Iranian and foreign cultivars into five groups, which the promising cultivar T24 and Koroneiki with the maximum difference with others were placed in separate groups. The Principal Coordinate Analysis (PCoA) was able to distinguish promising Iranian cultivars from the other commercial cultivars. The population structure analysis confirmed the results of the cluster and PCoA analysis and clearly separated the two groups of Iranian and foreign cultivars.ConclusionsIn conclusion, it was observed that the promising cultivar T24 with having a genetic similarity to both groups of Iranian and foreign cultivars can be a suitable candidate for commercialization with complementary characteristics of both groups. Pages 111-131ObjectiveCold is one of the important stresses in cold and temperate regions and improving cold tolerance is in the top priority of the lentil breeding programs. NAC transcription factors play an important role in response to various biological stresses, including cold stress.Materials and MethodsIn this study, to evaluate the expression of MfNAC, MtNAC57 and GmNAC2 genes, the plants of Gachsaran lentil cultivar were subjected to five temprature treatments in a completely randomized design with three replications. Twenty-nine old plants were subjected to duration of cold stress treatments including 6h, 12h, 24h and 48h at 2°-4°c and plants grown under greenhouse conditions (23°C) were considered as controls. Total RNA extraction from tissues was performed using lithium chloride method and cDNA was synthesized using TaKara kit. For each treatment, real-time PCR was performed in both shoot and root in three biological and two technical replications. Actin gene was used as internal control.ResultsThe results of analysis of variance of all studied genes showed that there was a significant difference between the different duration of cold treatments in both shoot and root. GmNAC2 gene expression decreased significantly in the shoot, 6 hours after stress, while in the root, the expression decreased 24 hours after stress. The expression of MtNAC57 gene in 6h, 12h, and 24h treatments decreased compared to control, but in the root in 12h treatment, a significant expression was observed. MfNAC gene in shoot showed a significant reduction in expression in all treatments, while in root a significant increase was only observed during long-term treatment of 48h.ConclusionsIn the present study, for first time, the expression pattern of some members of NAC gene family was examined in lentils under cold stress and the results could be useful for studies on this plant and other legumes.Keywords: Cold, lentil, real time PCR, root, shoot

Pages 134-146ObjectiveHomospermidine synthase (HSS) is the first enzyme in the specific pathway of pyrrolizidiene alkaloid (PA). HSS catalyzes the NAD1-dependent transfer of an aminobutyl group of spermidine to putrescine. Putrescine, spermidine and spermine are three polyamines which are naturally positively charged compounds found in virtually all living cells. These compounds bind to DNA and have been implicated in a number of crucial processes such as cell division, differentiation and membrane function. Very few studies on expression of homospermidine synthase gene in Senecio vulgaris, particularly in natural conditions have been reported. The objective of current research was to study homospermidine synthase gene expression in different tissues of S. vulgaris by real-time PCR.
Material andmethodsThe total RNA was extracted from roots, shoots (stems and leaves) and flowers, and was converted to cDNA. Specific primers were designed for HSS1 gene and also for the Pol II which was applied as a reference housekeeping gene in the experiment and expression level of HSS1 was assessed using real-time PCR.ResultsMelting curves of two genes showed the specificity of PCR amplification. Evaluation of normalized expression level of HSS1 gene showed that the gene was differentially expressed in different tissues of S. vulgaris. Maximum relative expression (>59-fold change relative to Pol II reference gene) was observed in roots where the homospermidine synthase is activated and homospermidine is produced. Interestingly, the gene is also expressed in flowers but with a lower extent (4-fold change relative to housekeeping Pol II gene). As expected the gene showed non-significant relative expression in shoots where the homospermidine is transported.ConclusionsExpression of HSS1 gene in S. vulgaris is tissue-specific and is higher in the roots relative to other tissues.Keywords: Homospermidine synthase, Gene expression, Natural condition, Real-time PCR, Senecio vulgaris -
Pages 147-170ObjectiveOptogenetics, as a new scientific branch, uses the photoreceptors to conduct basic and applied research. Identification of the novel photoreceptors and their characteristics is a way to develop the optogenetics. Cryptochrome/Photolyase protein family DASH crypthochromes have DNA repair ability and play role in signaling pathways. Current research goal was the adequate production of the Volvox predicted DASH1 cryptochrome protein (VcCryDASH1) to use in spectroscopy studies as well as DNA repair assay.Materials and methodsFirst, the coding sequence of VcCryDASH1 was isolated from Volvox carteri cDNA library using the specific primers and cloned into pGEX-2TK expression vector. The recombinant protein production was analyzed using two different strains of E. coli under different incubation times, using IPTG to induce the protein expression and then its solubility was investigated by SDS-PAGE. DNA binding ability of the protein was studied by alignment method.ResultsThe appropriate production of the protein obtained in Rosetta strain at 32°C under more than 3 hours incubation. Approximately, 50 percent of the protein was insoluble under 1 and 3 hours incubations. VcCryDASH1 alignment analysis showed that there is high percentage of amino acids in this protein with DNA binding ability.ConclusionThese results revealed that under optimum conditions, VcCryDASH1 has dual solubility but our approach can produce the adequate quantity of the protein in the Rosetta. Also, because of having conserved amino acids, the protein can bind to DNA, nominating it as a possible resource to design the DNA- light mediator molecules in optogenetics.Keywords: DASH1 cryptochrome, Expression vector, Rosetta strain, Volvox carteri
