    شناخت تنوع ژنتیکی و طبقه بندی ذخایر توارثی (ژرم پلاسم) در پیش برد اهداف به نژادی و مدیریت حفظ ذخایر ژنتیکی گیاهان دارای نقش اساسی می باشد. از این رو، به منظور ارزیابی تنوع ژنتیکی 20 ژنوتیپ رز با استفاده از 6 نشانگر CDDP و 20 نشانگر RAPD صورت گرفت. برای تعیین تشابه بین ژنوتیپ های رز از ضریب تشابه جاکارد استفاده گردید و گروه بندی ژنوتیپ ها به روش اتصال همسایگی (NJ) انجام شد. دامنه ضریب تشابه ژنتیکی بین ژنوتیپ ها 04/0 تا 77/0 برای نشانگر CDDP و برای نشانگر RAPD بین 05/0 تا 79/0 به دست آمد. درصد چند شکلی نشانگرهای CDDP و نشانگر RAPD، 100 درصد برآورد گردید. تجزیه خوشه اینشانگرهای CDDPو RAPD ژنوتیپ ‏ها را به سه گروه تقسیم- بندی کرد. مطالعه حاضر نشان داد نشانگرCDDP به خوبی ژنوتیپ های رز را از هم تفکیک نمودند و این اولین گزارش آنالیز تنوع ژنتیکی رز با استفاده از نشانگر مولکولی که براساس ناحیه DNA هدف گذاری-شده (CDDP) در مقایسه با نشانگر RAPD، است. نشانگر CDDP به عنوان سیستم مولکولی جدید برای ارزیابی ژرم پلاسم رز معرفی شد.

    این بررسی با هدف نقشه یابی نشانگرهای مولکولی پیوسته با ژن های مرتبط با وزن هزار دانه و عملکرد دانه در جو زراعی اجرا گردید. به منظور شناسایی QTLها، یک جمعیت شامل 148 هاپلوئید مضاعف (DH) برای صفات وزن هزار دانه و عملکرد دانه در طرح کاملا تصادفی با 3 تکرار در شرایط گلخانه ای مطالعه گردید. جمعیت هاپلوئید مضاعف حاصل تلاقی بین رقم اصلاح شده Clipper (با عملکرد بالا) و رقم محلی از الجزایر به نامSahara3771 (با عملکرد پایین) بود. در این تحقیق 30 نشانگر جدید ISSR به 466 جایگاه قبلی روی نقشه پیوستگی جمعیت اضافه گردید. نشانگرها در مجموع 1460 سانتی مورگان از ژنوم جو را پوشش داده و متوسط فاصله دو نشانگر مجاور 3 سانتی مورگان بود. تجزیه QTL منجر به شناسایی پنج QTL مرتبط با عملکرد دانه (بر روی کروموزوم های2H ،4H ،5H و 6H) شد که این مکان ها در مجموع 57 درصد از تنوع فنوتیپی عملکرد را توجیه می کردند. QTL واقع بر کروموزوم 2H با 20 درصد بزرگ اثرترین QTL بود. برای وزن هزار دانه نیز 3 عدد QTL شناسایی گردید که QTL واقع بر کروموزوم 2H با 69 درصد بزرگ اثرترین QTL بود. شناسایی QTLهای بزرگ اثر برای وزن هزار دانه و عملکرد، سودمندی استفاده از مارکر های مولکولی در نقشه یابی ژنی برای صفات وزن دانه و عملکرد دانه را نشان می دهد که از این پتانسیل می توان برای انتخاب به کمک نشانگر (MAS) در آینده ای نزدیک در برنامه های اصلاح برای بهبود عملکرد جو استفاده کرد.

    پنبه یکی از گیاهان زراعی مهم و اقتصادی در دنیا و به عنوان چهارمین محصول مهم صنعتی در ایران به شمار می رود. در گذشته، تلاش های زیادی جهت اصلاح کیفیت پنبه توسط اصلاحگران سنتی صورت گرفته است که به علت محدودیت های موجود در روش های سنتی، استفاده از روش های مهندسی ژنتیک رواج یافت. به دنبال رواج روش های نوین، گیاهان تراریخته پنبه مقاوم به حشرات، بیماری ها، علفکش، تنش های غیر زیستی و همچنین الیاف با کیفیت بالاتر تولید شدند. امروزه، تعدادی از این ارقام تجاری سازی شده و از پذیرش عمومی بالایی در سراسر جهان برخوردار هستند. اکنون نیز با ظهور روش-های توالی یابی نسل جدید و تسهیل شناسایی ژن های کاندید و مناسب جهت دست ورزی و وجود روش-های ویرایش ژنوم () با قابلیت ایجاد تغییرات هدفمند در ژن ها، افق تازه ای در حوزه اصلاح ژنتیکی پنبه ایجاد شده است. این مقاله ضمن مروری بر تاریخچه اصلاح پنبه و بررسی موانع موجود، به تحقیقات صورت گرفته در زمینه تولید ارقام مقاوم به تنش های زیستی و غیر زیستی و افزایش کیفیت پنبه با روش های مهندسی ژنتیک و ویرایش ژنومی می پردازد.

    در این پژوهش با توجه به اهمیت تنش خشکی و همچنین گیاه کلزا، آزمایشی بصورت فاکتوریل در قالب طرح بلوک های کامل تصادفی با استفاده از ده رقم کلزای بهاره در مرحله گیاهچه ای به روش کشت هیدروپونیک و با اعمال تنش خشکی ناشی از PEG6000 انجام شد. دو هفته پس از اعمال تنش و در پایان مرحله روزت نمونه برداری انجام شد. نتایج نشان داد که در شرایط تنش ارزش صفات مورفولوژیک و فیزیولوژیک کاهش یافت. همچنین بین ارقام مورد مطالعه از لحاظ پاسخ به تنش خشکی تنوع وجود داشت و در مجموع متحمل ترین و حساس ترین ارقام از نظر صفات مطالعه شده بترتیب رقم SW5001 و Sarigol بودند که برای مطالعات تکمیلی روی این دو رقم تجزیه پروتئوم انجام شد. برای بررسی الگوی پروتئینی، استخراج پروتئین از بافت برگی انجام و الکتروفورز بعد اول به روش نوارهای IPG و الکتروفورز بعد دوم با تکنیک SDS-PAGE اجرا شد و پس از رنگ آمیزی با آبی کوماسی تصویربرداری از ژل ها و تجزیه لکه های پروتئینی با نرم افزار PDQuest انجام شد. در نهایت تعداد 25 لکه پروتئینی معنی دار بین گیاهان شاهد و تحت تنش خشکی برای هر دو رقم، تشخیص داده شدند که از این تعداد 15 لکه پروتئینی بین دو رقم مشترک بودند و تعداد شش لکه پروتئینی منحصر به رقم متحمل و چهار لکه پروتئینی منحصر به رقم حساس بودند. پس از شناسایی این پروتئین ها با طیف سنجی جرمی، در مجموع بیشترین گروه های پروتئینی مشترک بین دو رقم، پروتئین های دخیل در واکنش نوری فتوسنتز، چرخه کالوین و پروتئین های سم زدا بودند. در مجموع مهمترین دلیل حساسیت و تحمل ارقام در کلزا بیان متفاوت و القای منحصر به فرد پروتئین ها و در نهایت تاثیر آنها روی سایر صفات بدست آمد.

    خلر (Lathyrus sativus L.)، یکی از مهم ترین گیاهان زراعی و علوفه ای دنیاست که به علت پروتئین و لایسین بالا گیاهی شناخته شده است. به منظور ارزیابی تنوع ژنتیکی 33 ژنوتیپ خلر، از 20 نشانگر نیمه تصادفی اینترون-اگزونی یا ISJ استفاده شد. استخراج DNA با روش CTAB صورت گرفت. اطلاعات حاصل از آغازگرهای ISJ مورد تجزیه و تحلیل قرار گرفت و از مجموع باند ها، 77 % باندها مربوط به آغازگرهای ET و 81 % مربوط به آغازگرهای IT چند شکل بودند. نتایج نشان داد که متوسط تعداد باند چند شکل به ازای هر آغازگر 95/3 عدد بود و بیشترین میزان اطلاعات چند شکلی و شاخص نشانگر را مربوط به آغازگر ET18-6 بود. بر اساس محاسبه ضریب تشابه جاکارد، محدوده تشابه ژنوتیپ-ها از 42/0 تا 89/0 متغیر بود. تجزیه گروه بندی بر اساس روش UPGMA صورت گرفت و براساس دندروگرام بدست آمده و در ضریب تشابه 70/0، ژنوتیپ ها به پنج گروه اصلی تقسیم شدند. نتایج تجزیه به مختصات اصلی نشان داد که سه مولفه اول در مجموع 65/74 درصد از کل واریانس را توجیه نمودند که بیانگر پراکنش محدود این نشانگرها در سطح ژنوم این گیاه می باشد. این تحقیق اولین گزارش از کاربرد نشانگر ISJ در گیاه علوفه ای مهم خلر در دنیا می باشد که امید می رود در توسعه اصلاح ملکولی این گیاه موثر واقع گردد.

    لپتین توسط سلول های چربی تولید می شود و نقش مهمی در تنظیم وزن بدن از طریق کاهش مصرف مواد غذایی و تحریک مصرف انرژی ایفا می کند. ژن لپتین می تواند در بسیاری از اعمال فیزیولوژیکی بدن و تنظیم مصرف غذا شرکت کند. در این تحقیق به منظور شناسایی چند شکلی در ناحیه اینترون شماره 2 ژن لپتین از 150 بز کرکی راینی بصورت تصادفی خونگیری انجام شد. سپس DNA ژنومی آنها استخراج و قطعه ای به طول 442 جفت باز از ناحیه اینترون 2 ژن لپتین با استفاده از روش PCR-RFLP تکثیر گردید. تجزیه و تحلیل الگوهای باندی منجر به شناسایی سه الگوی باندی AA،AB و BB شد، که فراوانی آنها به ترتیب برابر با 57/0، 3/0 و 13/0 بدست آمد. دو آلل A و B با فراوانی 72/0 و 28/0 در جمعیت مورد مطالعه شناسایی شد. شاخص شانون و تعداد آلل موثر برای این جایگاه از ژن لپتین به ترتیب 5898/0 و 6673/1 بدست آمد. نتایج نشان داد وزن تولید ارتباط معنی داری با ژن لپتین ندارد، اما وزن از شیرگیری، طول تنه، ارتفاع بدن و دور سینه حیوان ارتباط معنی داری (P<0.05)با چندشکلی یافت شده در این ژن دارند.

    بیماری موزائیک انجیر گسترش جهانی دارد و به عنوان شایع ترین بیماری ویروسی در انجیر شناخته می شود. یکی از مهمترین ویروس هایی که در این بیماری نقش دارد، ویروس بادنای یک انجیر (FBV-1) می باشد. در تحقیق حاضر، روابط فیلوژنتیکی بین جدایه های ایرانی و سایر جدایه های گزارش شده از این ویروس مورد بررسی قرار گرفت. با استفاده از آغازگرهای اختصاصی ژن رمز کننده پروتئین حرکتی ویروس، قطعه ی مورد انتظار به اندازه تقریبی 1090 جفت باز از ژنوم ویروس تکثیر شد. قطعه ژنی 12 جدایه در پلاسمید pTZ57R/T همسانه و تعیین توالی شد. ترادف های به دست آمده با ترادف-های موجود در بانک ژن مقایسه و پس از همردیف سازی چندگانه، درخت فیلوژنتیکی بر اساس ژن پروتئین حرکتی FBV-1 ترسیم شد. بر اساس آنالیز فیلوژنتیکی، جدایه های FBV-1 در دو گروه متمایز دسته بندی شدند و جدایه های ایرانی در دو زیرگروه با 99% مشابهت نوکلئوتیدی در گروه اول قرار گرفتند. در بررسی تبارزایی بر مبنای توالی اسید آمینه ای، جدایه های مورد بررسی در سه گروه با 99-98 % مشابهت قرار گرفتند. جدایه های ایرانی از تنوع ژنتیکی پایین تری در مقایسه با جدایه های آمریکایی برخوردار بودند و شرایط جغرافیایی در تنوع ژنتیکی جدایه های موجود تاثیر داشت.

    چغندرقند یکی از مهمترین گیاهانی است که در تامین غذای مردم جهان نقش کلیدی دارد و از نظر ارزش غذایی در ردیف برنج، ذرت، گندم، سیب زمینی و حبوبات قرار دارد. تعیین میزان تنوع ژنتیکی در مواد گیاهی گام اولیه برای شناسایی، حفظ و نگهداری ذخایر توارثی و طراحی برنامه های اصلاحی می باشد. به منظور بررسی تنوع ژنتیکی در 20 ژنوتیپ چغندرقند، از 20 آغازگر ISSR استفاده گردید. بعد از استخراج DNA و تکثیر قطعات با استفاده از آغازگرهای ISSR، نوارهای حاصله امتیازبندی شدند و تجزیه های آماری صورت گرفت. شاخصهای تنوع ژنتیکی بهدست آمده در ژنوتیپهای چغندرقند مورد مطالعه نشان داد که بیشترین میزان محتوای اطلاعات چند شکلی (PIC) مربوط به مکان UBC853 (38/0) و کمترین میزان PIC مربوط به مکان UBC847 (13/0) بود. همچنین بیشترین میزان شاخص شانون که نشان دهنده تنوع بین جمعیتی است، مربوط به مکانUBC830 (32/4) در حالی که مکان ژنی UBC847 دارای کمترین میزان شاخص شانون (58/0) بود. تجزیه خوشه ایبراساس داده های مولکولی با استفاده از ضریب تشابه جاکارد و روش UPGMA، ارقام را در 4 گروه اصلی قرار داد. براساس نتایج حاصل از تجزیه به مولفه های اصلی، سه مولفه اول به ترتیب 67/72، 99/5 و 05/4 درصد از واریانس کل را توجیه کردند. گروهبندی حاصل از مولفه های اصلی، نتایج تجزیه خوشه ایرا تا حدی تایید کرد. از نتایج حاصل از این مطالعه می توان در برنامه های به نژادی چغندرقند استفاده نمود.

    Identification of genetic diversity and classification of genetic resources (germplasm) are of important and essential activities in breeding and management of plant genetic resources. In order to study the genetic diversity of rose, twenty genotypes were analyzed using six Conserved DNA-Derived Polymorphism (CDDP) and 20 RAPD markers. Matrix genetic distance ranged from 0.04 to 0.77 in CDDP and from 0.05 to 0.79 in RAPD marker analysis. The level of polymorphism generated by CDDP markers (100%) was similar to RAPD marker. Cluster analysis for CDDP and RAPD markers revealed that genotypes were grouped in three clusters. The current study showed CDDP marker very well differentiated rose genotypes from each other. This is the first report of using targeted DNA region molecular marker (CDDP) for genetic diversity analysis in rose in comparison with RAPD markers. We introduce CDDP as a new molecular markers system for evaluating Rose germplasm.

    To identify the QTLs affecting grain weight and yield, 148 doubled-haploid (DH) population derived from a cross between Clipper (high yield) and Sahara3771 (low yield) were screened under controlled conditions. The experiment was conducted in CRD with 3 replications. The new 30 ISSR marker loci were added to a backbone of 466 loci on the Clipper × Sahara DH linkage map. The map spanned 1460 centimorgans (cM) and had a mean density of 2 loci per 3 cM. The QTL analysis led to identification of 5 QTLs on 2H, 4H, 5H and 6H chromosomes for grain yield. These QTLs could explain 57% of the total phenotypic variation, and the QTL on chromosome 2H (ksuF2-mwg892) with 20% had the largest effect. For grain weight, 3 QTLs were identified, and the QTL on 2H could explain 69% of total phenotypic variation. The identification of QTLs with large effects on grain weight and yield illustrates the usefulness of molecular markers in gene mapping and suggest that marker-assisted selection will be feasible in the near future in barley breeding programs.

    Cotton is considered as one of the most important crops in the world and ranks the fourth economic crop in Iran. In past years, conventional plant breeders endeavored to increase cotton quality. To overcome the constraints that conventional breeding programs comprised, new genetic engineering technologies were introduced. Since then, many cotton events resistant to pests, disease, pesticides, abiotic stresses and also high quality of lint have been produced and commercialized around the world which has confronted a noticeable public acceptance. Today and with the advent of next generation sequencing methods in favor of easier identification of suitable candidate genes for their manipulation and novel targeted genome editing technology (CRISPR/Cas9), outstanding prospects regarding cotton breeding have been developed. In present article, history of cotton breeding including genetic engineering and genome editing researches which led to production of resistant cultivars to biotic and abiotic stresses and high quality fiber are reviewed.

    In this research, considering the importance of drought and canola, an experiment was done as factorial in a Randomized Complete Block Design using ten spring canola cultivars with hydroponic method in seedling stage and with induced of drought stress by PEG6000. Two weeks after of the stress induction and at the end of the rossete stage, samples were taken. The results showed that the value of morphological and physiological traits was declined under drought stress. Also the studied cultivars were varied in response to drought stress and in general, the most tolerant and sensitive cultivars for studied traits were SW5001 and Sarigol cultivars, respectively that to graduate studies on these two cultivars proteome analysis was performed. To study the pattern of protein, extraction of protein from leaf tissue was performed and the first dimension electrophoresis using IPG strips and second dimension electrophoresis was performed by SDS-PAGE technique and after the gels staining with commassie blue, gels imaging with scanner and protein analysis with PDQuest software was done. Finally a total of 25 protein spots between control plants and under drought stress for both cultivars were detected that of these, 15 protein spots were common between two cultivars and six unique protein spots for tolerant cultivar and four unique protein spots for susceptible cultivar. After detection these proteins with mass spectrometry, overall, the most common protein groups between two cultivars were involved proteins in photo-reaction of photosynthesis, Calvin cycle and detoxifying enzymes. In total, the most important cause of the sensitivity and tolerance of canola cultivars different expression and unique expression of proteins into cultivars and finally effects of them on other were obtained.

    Grass pea (Lathyrus sativus L.), as one of the most important forage crop plants, has high content of protein and Laysin. In order to assess the genetic diversity of 33 genotypes of grass pea, 20 semi-random primers were used in the present study. DNA was extracted by CTAB method. The ISJ data were analyzed and 77% of IT bands and 81% of ET bands were polymorphic. The results showed that the average number of polymorphic bands per primer was 3.95 and the highest amount of polymorphic information content and marker index was belonged to ET18-6 primer. According to the Jaccard's similarity coefficient, the range of similarity among studied genotypes was varied from 0.42 to 0.89. Cluster analysis based on UPGMA method divided genotypes to 5 groups at cut of line in 0.71 similarity coefficient. The principle coordinates analysis showed that the first three components explained 74.65% of total variance indicating the restricted distribution of these ISJ markers at the grass pea genome. Present study is the first report on application of ISJ markers in grass pea, as important forage plant, which is hoped to be effective in the development of molecular breeding programs of this plant.

    Leptin is produced by adipocytes and plays a critical role in the regulation of body-weight through reducing food intake and stimulating energy expenditure. Leptin gene can participate in many physiological functions and regulation of food intake. In order to identify the polymorphism in a region of the second intron of the leptin gene, blood samples were taken randomly from 150 Raeini cashmere goats. Genomic DNA was extracted and a fragment with a length of 442 bp was amplified from the second intron of leptin gene using PCR-RFLP method. The statistical analysis identified three band patterns of AA, AB and BB with frequencies of 57.0, 3.0 and 13.0, respectively. The A and B alleles showed frequency of 72.0 and 28.0, respectively. Shannon index and effective number of alleles for the leptin gene were 5898/0 and 6673/1 respectively. Analysis of the body size related traits with leptin genotypes showed that while birth weight was not associated with this gene weaning weight, body length, body height and chest circumference were significantly associated with identified polymorphisms in this gene (P

    Fig mosaic disease is considered as the most common fig viral disease, which occurs worldwide. Fig badnavirus-1 (FBV-1) is one of the most important viruses involved in this viral disease. In the present research, phylogenetic relationship between the Iranian and other detected FBV-1 isolates was studied. A genomic fragment with an expected size about 1090 bp was amplified from the movement protein coding region of twelve virus isolates using specific primers. Amplified fragments were cloned in plasmid pTZ57R/T, and sequenced. Obtained sequences were phylogenetically compared with the corresponding isolates available in the GenBank after multiple alignments. The phylogenetic tree was drawn for the FBV-1 isolates based on the movement protein gene sequence. The studied isolates were categorized into two distinct phylogroups in which Iranian isolates were separated into two different subgroups in group I with 99% identity at the nucleotide level. Phylogeny on the basis of the amino acid sequences categorized isolates with 98-99٪ identity into three different groups. According to the results of this study, it could be concluded that genetic variation between Iranian isolates is less than that of American isolates, and additionally the genetic variation between the isolates is affected by the geographical condition.

    Sugar beet is one of the most important plants playing a key role in human food production in the world. Sugar beet has nutritional value as rice, corn, wheat, potato and beans. The estimation of genetic diversity of plant material is the first step for identification, conservation of genetic resources and designing of breeding programs. In order to study the genetic diversity of 20 sugar beet genotypes, 20 ISSR primers were used. After DNA extraction and PCR amplification, using ISSR primers the scores (0 and 1 for absent and present band, respectively) of resulting bands subjected to statistical analyses. Genetic diversity indices of ISSR markers in the studied sugar beet genotypes showed that UBC853 and UBC847 had the highest (0.38) and lowest (0.13) values of polymorphism information content respectively. Also UBC830 locus had the highest rate (4.32) of Shannon diversity index, which represents the genetic diversity among populations, whereas UBC847 had the lowest (0.58) Shannon index. Cluster analysis based on molecular data using Jaccard's similarity coefficient and UPGMA method, classified the sugar beet genotypes into four major groups. Based on the results of principal components analysis, the first three principal components explained 72.67, 5.99 and 4.05 percent of total (82.72%) total variation indicating ISSR markers have good distribution in the whole genome. The classification of the sugar beet genotypes based on the first principal components, had similarities and differences with cluster analysis. Results of this study can be used in breeding programs of sugar beet.
