    هدف از انجام این پژوهش، نقشه یابی جایگاه های ژنی موثر بر وزن زنده و اندازه بدن و شناسایی نشانگرهای زیستی مهم مرتبط با سرعت رشد در گوسفند نژاد لری بختیاری بود. آنالیز نواحی ژنومی موثر بر صفات کمی مورد بررسی (QTL) در یک جمعیت گوسفند نژاد لری بختیاری با استفاده از تحلیل پیوستگی به روش مکان یابی درون فاصله ای مبتنی بر رگرسیون انجام گردید. جمعیت مورد بررسی شامل 162 حیوان مربوط به 5 خانواده ناتنی پدری بود. داده های فنوتیپی شامل اندازه گیری های وزن تولد (BW)، وزن یک ماهگی (W1)، وزن شیرگیری (WW)، وزن شش ماهگی (W6)، دور سینه در شش ماهگی (HG6)، طول بدن در شش ماهگی (BL6)، ارتفاع جدوگاه در شش ماهگی (HT6)، وزن نه ماهگی (W9) و وزن دوازده ماهگی (W12) بودند. 5 والد نر و نتاج آنها برای شش نشانگر ریزماهواره تعیین ژنوتیپ شدند. داده ها در دو مرحله، آنالیز هر خانواده به صورت انفرادی و آنالیز توام تمام خانواده ها به کمک یک مدل تک QTLتجزیه و تحلیل شدند. براساس آنالیزهای انفرادی خانواده ها، QTL مرتبط با صفت وزن یک ماهگی (روی کروموزوم شماره 1 در موقعیت 6/210 سانتی مورگان) در نزدیکی پرایمر INRA011در خانواده دوم شناسایی گردید. در خانواده ی سوم نیز QTL های مرتبط با صفات وزن یک ماهگی و وزن شیرگیری به ترتیب در موقعیت های 6/252 و 6/223 سانتی مورگان در نزدیکی نشانگرهای LSCV105 و MCM137 شناسایی شدند. همچنین در خانواده چهارم نیز یک QTL موثر بر صفت وزن یک ماهگی در موقعیت 6/254 سانتی مورگان در نزدیکی نشانگر LSCV105شناسایی شد. با انجام آنالیز به صورت همزمان بر روی تمام خانواده ها یک QTL مرتبط با صفت وزن یک ماهگی در موقعیت 6/254 سانتی مورگان در نزدیکی نشانگر LSCV105شناسایی شد. با توجه به آن که ژن های ترانسفرین و PIT1 بر روی کروموزوم 1 قرار دارند این دو ژن کاندیداهای قوی برای اثرات مشاهده شده ی QTL برای صفات رشد در این نژاد هستند.

    شناسایی ویژگی های ژنتیکی تمساح پوزه کوتاه ایرانی (گاندو)، در برنامه ریزی مدیریتی به منظور حفاظت و نگهداری آن نقش بسیار مهمی دارد، لذا این پژوهش با هدف، بررسی ژنتیکی کروکودیل پوزه کوتاه ایران بر اساس مطالعه جمعیتی نواحی ثابت و متغیر ژنوم میتوکندری انجام گردید. بدین منظور، 12 نمونه بافت پوست و خون از سه منطقه مختلف شهرستان چابهار جمع آوری و پس از استخراج DNA، نواحی ثابت و متغیر ژنوم میتوکندری Cyt b و D-loop با استفاده از روش PCR تکثیر گردید. محصولات PCR توالی یابی و سپس آنالیز نتایج با استفاده از نرم افزارهای بیوانفورماتیک و مبتنی بر داده های نوکلئوتیدی و کدون های پروتئینی انجام گرفت. نتایج این تحقیق نشان داد که تمامی نمونه های مورد مطالعه، دارای توالی نوکلوتیدی یکسان هستند و هیچ تنوع ژنتیکی در بین این نمونه ها یافت نشد. نزدیکترین گونه به گاندو، گونه Crocodylus palustris و سپس گونه C. porosus بدست آمد. مقایسه توالی ها نشان دهنده وجود 12 جایگاه پلی مورفیک در بین 1156 جایگاه مورد مطالعه بود و 5 هاپلوگروپ معرفی گردید. لذا، با توجه به عدم مشاهده تنوع ژنتیکی در بین نمونه های مورد مطالعه، استفاده از برنامه های حفاظتی جهت افزایش تنوع ژنتیکی در گاندو امری بسیار ضروری به نظر می رسد.

    کادمیوم یکی از فلزات سنگین است که در گیاهان تنش اکسیداتیو ایجاد می کند و گیاهان برای حفاظت در برابر این تنش مجهز به یک سیستم جاروب کننده رادیکا ل های آزاد می باشند. این سیستم شامل آنزیم های آنتی اکسیدان و نیز سیستم آنتی اکسیدانی غیر آنزیمی است. در این مطالعه تغییر در الگوی بیان ژن و فعالیت آنزیم گلوتاتیون ردوکتاز در پاسخ به غلظت های (0، 600،300 و 900 میکرو مولار) کلرید کادمیوم در قالب یک طرح کاملا تصادفی به ترتیب با 2 و 5 تکرار در گیاه ماریتیغال (Silybum marianum) بررسی شد. طبق نتایج بدست آمده از آنالیز نیمه کمی RT-PCR، میزان بیان این ژن در برگ گیاهچه های تحت تنش نسبت به گیاهچه های شاهد، تغییرات معنی دار نشان داد به طوری که کمترین و بیشترین میزان بیان به ترتیب در سطح شاهد و غلظت 900 میکرو مولار کلرید کادمیوم مشاهده شد. بنابراین می توان گفت آنزیم گلوتاتیون ردوکتاز در سیستم دفاع آنتی اکسیدانی آنزیمی در پاسخ به سمیت کادمیوم در گیاه ماریتیغال نقش ایفا می کند.

    شوری به عنوان یکی از مهمترین تنش های غیر زیستی تولید محصولات کشاورزی در مناطق خشک و نیمه خشک را به شدت کاهش می دهد. کلزا به دلیل کیفیت بالای روغن دانه، مقادیر زیاد اسیدهای چرب اشباع نشده و عملکرد بیشتر روغن در واحد سطح نسبت به دیگر دانه های روغنی، برتری دارد. گیاهان در مواجهه با شرایط تنش، با سنتز برخی متابولیت های ضروری، پروتئین های خاص ساختاری یا آنزیم های مسیرهای متابولیکی با تنش وارد شده مقابله می کنند. جهت شناسایی مکانیسم های مولکولی تحمل تنش شوری، تغییرات بیان پروتئین های رقم حساس Option500 تحت تنش شوری مورد بررسی قرار گرفت. شوری از نوع کلرید سدیم با سطوح صفر (شاهد)، 150 و 300 میلی مولار باعث کاهش معنی دار وزن خشک اندام هوایی، ارتفاع بوته، پتاسیم برگ و نسبت K+/Na+ برگ و از طرفی باعث افزایش میزان سدیم و پرولین برگ گردید. با انجام الکتروفورز دو بعدی 110 لکه ی پروتئینی تکرار پذیر شناسایی شد، که از بین آنها 44 لکه براساس شاخص فاکتور القا (Induction Factor)، دارای تغییرات بیان معنی‫دار بودند، که هفت لکه پروتئینی همچنین در سطح احتمال 5% معنی‫دار شدند. به طور کلی یک لکه افزایش بیان و شش لکه کاهش بیان نشان دادند. با روش طیف سنجی LC-MS/MS این پروتئین ها، که در چرخه تولید انرژی و فتوسنتز نقش کلیدی ایفا می کنند، شناسایی شدند. در این پژوهش نقش احتمالی پروتئین های شناسایی شده در واکنش به تنش شوری مورد بحث قرار گرفت.‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬‬

    گروه ژن های RNase در فرایندهای حیاتی سلولی همچون همانند سازی DNA، رونویسی، پیرایش و کنترل ترجمه، شرکت می کنند.S-Like ریبونوکلئازها (S-Like RNase)، همولوگS ریبونوکلئاز (S-RNase) ها هستند، ولی آنها در ناسازگاری گیاه مشارکتی ندارند. در گیاهان دو لپه ای S-Like RNase ها نقش مهمی در انتقال مجدد فسفات طی فرایند پیری دارند، همچنین آنها در فرایندهای کمبود فسفات معدنی، دفاع سلولی و زخم های فیزیکی القا می شوند. اثرات RNase ها در پاسخ به خشکی می تواند به دلیل اثرات متقابل بین چندین فعالیت مختلف آنها باشد. در این تحقیق پروموتر ژن S-Like RNase در بالادست ژن بتا گلوکرونیداز (GUS) قرار داده شد سپس سازه مورد نظر توسط الکتروپوریشن به آگروباکتریوم تومفاسینس، سویه LBA 4404، انتقال داده شد و سپس به کمک هم کشتی، گیاه تراریخت ایجاد شد. با استفاده از روش رنگ آمیزی هیستوشیمیائی فعالیت پروموتر ژن S-Like RNase در بافت ریشه، برگ و بساک برنج بررسی گردید. نتایج نشان داد که پروموتر این ژن دارای فعالیت بالایی در بساک برنج می باشد درحالیکه میزان فعالیت آن در برگ کم و در ریشه نیز ناچیز بوده است. بررسی بروز پروموتر این ژن در بافت های مختلف، نتایج بیان بالای ژن S-Like RNase در بساک در مقایسه با بافت-های دیگر که با استفاده از آنالیز q-PCR در مطالعات قبلی مشخص شده بود را تکمیل و تائید می کند.

    گیاهان به دلایل متعددی به عنوان بیوراکتورهای ارزشمند برای تولید انواع پروتئین های نوترکیب (به ویژه آنتی بادی های مونوکلونال و...) می توانند مورد بهره برداری قرار گیرند. استفاده از ناقل های ویروسی گیاهی برای بیان موقت، یک استراتژی مفید برای تولید پروتئین های مهم دارویی از قبیل آنتی بادی ها در مقیاس بالا و در یک دوره زمانی خیلی کوتاه پیشنهاد می شود. امروزه، سرطان دومین عامل مرگ و میر در جامعه بشری است. شواهد قانع کننده نشان می دهد که فاکتور رشد اندوتلیال عروقی (VEGF) به دلیل نقش ضروریش در رگزایی یک هدف مهم برای درمان سرطان می باشد. در این مطالعه قطعات مختلف نانوبادیAnti-VEGF (Nb42) به طور موقت تحت کنترل راه انداز قوی ویروسی در گیاه کدو با استفاده از ناقل ویروسی مبتنی بر ZYMV بیان گردید. این قطعات شامل توالی معمولی نانوبادی (Nb42I) و توالی بهینه شده نانوبادی برای بیان در گیاه (Nb42II) بودند. پس از مایه کوبی گیاهان، حضور رونوشت های قطعات مختلف ژنی Nb42 در گیاهان مایه کوبی شده کدو با استفاده از آزمون RT-PCR شناسایی گردید. همچنین بیان پروتئین نوترکیب با استفاده از آزمون های لکه گذاری نقطه ای، SDS-PAGE، لکه گذاری وسترن و الایزا تائید گردید. بر اساس نتایج الایزا، میزان بیان نانوبادی Nb42Iو Nb42II به ترتیب 2 و 8/6 میکروگرم در هر گرم بافت برگی بود. در مجموع، نانوبادی نوترکیب Nb42 می تواند به طور موثر در گیاه کدو بیان گردد و سیستم بیانی مبتنی بر ZYMV یک سیستم تولیدی مناسب برای بیان پروتئین های نوترکیب در گیاهان خانواده کدوئیان خواهد بود.

    زیره سبز (Cuminum cyminum L.) گیاهی گلدار از خانواده چتریان (Apiaceae) است که بومی مناطق شرق مدیترانه تا هند می باشد. به رغم اهمیت دارویی فراوان زیره سبز، اطلاعات بسیار اندکی از ژنوم و مکانیسم های بیوشیمیایی ترکیبات موجود در این گیاه وجود دارد. در مطالعه حاضر جهت ارزیابی توالی رونوشت زیره سبز، از اندام گل (به دلیل تجمع بالای ترکیبات آلدئیدی و محل اصلی بیوسنتز آن)، جهت استخراج RNA از چهار نمونه و انجام آنالیز های RNA-Seq استفاده شد. در نهایت، بیش از 153 میلیون خوانش به طول 50 باز از توالی یابی این نمونه ها حاصل شد. سرهم بندی خوانش ها به منظور ایجاد رونوشت های بیان شده توسط نرم افزار Trinity (نسخه 3 . 1 . 1) و با مقادیر kmer 25 و 32 انجام شد. بهترین سرهم بندی بر اساس کامل بودن توالی رونوشت ها با استفاده از نرم افزار BUSCO (نسخه 3) شناسایی شد. بعد از سرهم بندی خوانش ها 50973 توالی ژن (کانتیگ) با میانگین طول 725 باز و مقدار N50 برابر با 1136 باز به دست آمد. همچنین از این تعداد ژن، 53103 رونوشت شناسایی شد. از این تعداد، 35860 رونوشت دارای حداقل یک همولوگ در بانک اطلاعاتی Nr بودند. بیش از 7/66 درصد رونوشت ها حداقل دارای یک همولوگ در بانک اطلاعاتی GO (فرآیند های بیولوژیک، عملکرد های مولکولی و اجزاء سلولی) بودند. بیشتر ژن های شناسایی شده مرتبط با تنظیم رونویسی و فعالیت های غشایی بودند. در مطالعه حاضر نخستین پروفایل توالی رونوشت در گیاه زیره گزارش شد که می تواند در مطالعات بعدی به منظور شناسایی ژن های دخیل در مسیر بیوسنتزی ترکیبات ثانویه مختلف و دیگر مطالعات ژنتیکی در این گیاه مورد استفاده قرار گیرد.

    ریحان (Ocimum basilicum L.) از مهمترین گیاهان دارویی متعلق به خانواده نعناعیان است که اسانس آن سرشار از ترکیبات فنیل پروپانوئیدی می باشد. تنش های محیطی مانند خشکی می توانند درصد و میزان ترکیبات اسانس را تغییر داده و بر بیان ژن های دخیل در سنتز آن نیز تاثیر بگذارند. به منظور مطالعه اثر تنش خشکی بر بیان برخی ژن های کلیدی دخیل در بیوسنتز فنیل پروپانوئیدها از جمله فنیل آلانین آمونیالیاز (PAL)، اوژنول سینتاز1 (EGS1) و کافئیک o- متیل ترانسفراز ((COMT در رقم کشکنی لولوی ریحان، آزمایشی بر پایه طرح کاملا تصادفی با سه تکرار در گلخانه اجرا شد. تنش خشکی در سه سطح 100 (کنترل)، 75 و50 درصد ظرفیت زراعی در مرحله 6 تا 8 برگی اعمال شد. بیان ژن های مورد نظر با استفاده از تکنیک Real time PCR بررسی شد. تجزیه داده های مربوط به بیان ژن های موردنظر و مقایسه میزان بیان ژن EGS1 در تیمار 75 و 50 درصد ظرفیت زراعی نسبت به شاهد نشان داد که میزان بیان این ژن در تیمار 75 درصد تغییر چندانی نسبت به کنترل نداشته ولی در تیمار 50 درصد، 74/9 برابر کنترل افزایش یافت. میزان بیان ژن COMT در هر دو تیمار تنش خشکی نسبت به شاهد، تغییر چندانی نکرد ولی میزان بیان ژن PAL در هر دو تیمار تنش خشکی نسبت به کنترل، 5/1 تا 2 برابر افزایش یافت. البته میزان بیان این ژن بین دو تیمار تنش خشکی معنی دار نبود. بنابراین احتمال می رود بتوان از طریق اعمال تنش خشکی و افزایش بیان ژن های EGS1 و PAL محتوای ترکیبات فنیل پروپانوئیدی را در ریحان افزایش داد.

    توتون به عنوان گیاهی مدل، امروزه به طور چشمگیری در تحقیقات مولکولی با هدف درک مبانی تعاملات بیمارگر و گیاه استفاده می شود. بیماری باکتریایی آتشک توتون گسترش جهانی دارد و خسارت قابل توجهی به توتون وارد می کند. عامل این بیماری Pseudomonas syringae pv. tabaci (Pst) می باشد. القای ژن های مرتبط با بیماری زایی که در مقاومت به بیماری ها ایفای نقش می کنند می تواند در کاهش شدت بیماری موثر باشد. شواهد نشان می دهد که هورمون های گیاهی در بیان این ژن ها در گیاهان موثر بوده و موجب افزایش تولید متابولیت های ثانویه می گردند. علاوه بر این سیستم دفاعی گیاه را از طریق فعال سازی رونویسی ژن های مرتبط با مکانیسم دفاعی گیاه را تحریک نموده و باعث تنظیم مقاومت القایی می گردند. در این تحقیق، بیان ژن های PR1، PR5، PAL، Catlase و WRKY12 در گیاه توتون تیمار شده با هورمون اسید سالیسیلیک با غلظت 3 میلی مولار و اسید جاسمونیک با غلظت 5/0 میلی مولار و گیاه شاهد، پس از آلودگی با باکتری Pseudomonas syrigae pv. tabaci در چند بازه زمانی با استفاده از تکنیک Quantitative Real time PCR (QRT-PCR) مورد بررسی قرار گرفت. نتایج نشان داد که تیمار اسید جاسمونیک و به خصوص اسید سالیسیلیک باعث تغییرات قابل توجه در بیان این ژن ها پس از تزریق باکتری شده است.

    وزن بدن از مهمترین معیارهای تعیین کننده سود اقتصادی در صنعت پرورش گوسفند است. در مطالعات پیوستگی و انتخاب ژنومی تعیین میزان و گستره عدم تعادل پیوستگی (LD) در تشخیص تعداد نشانگر مورد نیاز و اندازه نمونه، اهمیت بسزایی دارد. علاوه براین، انتخاب برای افزایش فراوانی موتاسیون های جدیدی که فقط در برخی از زیرجمعیت ها سودمند هستند باعث باقی گذاشتن نشانه هایی در سطح ژنوم می شود. اغلب این مناطق با ژن ها و QTLهای مرتبط با صفات مهم اقتصادی در ارتباط هستند. بنابراین، هدف این تحقیق مطالعه گستره LD و شناسایی مناطقی از ژنوم گوسفندان نژاد زندی با وزن بدن متفاوت بود که در طی سال های مختلف به صورت مصنوعی یا طبیعی انتخاب شده اند. بدین منظور، از 73 راس گوسفند زندی نمونه خون تهیه شد و با استفاده از آرایه های SNPChip 50 K شرکت ایلومینا ژنوتیپ 54241 نشانگر بر مبنای آخرین نسخه Oar_4.0 ژنوم گوسفند تعیین شد. پس از مراحل کنترل کیفی، در نهایت 40879 نشانگر SNP مربوط به 71 حیوان آنالیز شدند. مقدار LD با محاسبه آماره r2 بین تمام جفت جایگاه ها از طریق نرم افزار PLINK محاسبه شد. برای شناسایی نشانه های انتخاب بر پایه روش های عدم تعادل پیوستگی از آزمون هموزیگوسیتی هاپلوتایپی بسط داده شده جمعیت تلاقی (XP-EHH) استفاده شد. در این مطالعه گستره مفید عدم تعادل پیوستگی در 40K برابر با 2/0 r2= برآورد شد. نتایج این تحقیق 10 منطقه ژنومی روی کروموزوم های 1، 2، 3، 5، 6، 9، 12، 13، 21 و 22 را شناسایی کرد. تجزیه و تحلیل بیوانفورماتیکی نشان داد که برخی از این مناطق ژنومی با ژن های موثر بر صفات رشد، متابولیسم چربی، توسعه سیستم اسکلتی، متابولیسم انرژی و کیفیت گوشت مانند ژن های FBP1، FAM184B، PKD2، FGF19، SPP1، MEPE، CAPN2 همپوشانی دارند.

    The aim of this study was to map loci affecting live weight and body size in Lori-Bakhtiari sheep and identification of biomarkers linked to growth rate in sheep. Quantitative trait loci (QTL) linkage analysis in a Lori-Bakhtiari sheep population was conducted using the regression-based interval mapping method. The mapping population consisted of 162 animals related to 5 paternal half families. Phenotypic data were measurements of birth weight (BW), weight at one month of age (W1), weaning weight (WW), weight at six months of age (W6), chest circumference (HG6), body length (BL6), wither height at six months of age (HT6), weight at nine months of age (W9) and yearling weight (W12). Five sires and their progeny were genotyped for microsatellite markers in a candidate region on sheep chromosome 1. Data were analyzed in two phases, individual family analysis and combined family’s analysis using a single-QTL model. Analysis based on individual families revealed QTL related to W1 (210.6 cM), near INRA011 marker segregating in the second family. In the third family, QTL affecting W1 and WW were identified at 252.6 and 223.6 cM, nearby MCM137 and LSCV105 markers, respectively. In addition, in the fourth family, a QTL underlying W1 was detected at 256.6 cM near LSCV105 marker. Combined analysis of all the families resulted to the identification of a QTL associated with W1 at position 254.6 cM relative to the centromere near the LSCV105 marker. There were two strong candidate genes close to the location of the detected QTL, the transferrin and PIT1 genes, thus these genes may account for the observed QTL effects for growth traits in Lori-Bakhtiari sheep.

    Identification of the genetic features of Persian mugger crocodile, the Gando, is crucial in management planning for conservation and preservation of this endangered species. Therefore, this study is aimed to investigate the genetic characteristics of Gando in Iran, based on population studies of the conserved and variable regions of the mitochondrial DNA. For this purpose, 12 samples of skin tissue and blood were collected from three regions of Chabahar city, south east of Iran. After DNA extraction, conserved and variable regions of the mitochondrial genome Cyt b and D-loop were amplified using PCR. The PCR products were sequenced and results were analyzed using bioinformatics software based on nucleotide data as well as protein codons. Results revealed that all samples had similar nucleotide sequences and no genetic variation was found among the samples. The closest species to Gando were Crocodylus palustris and then C. porosus. Comparison of sequences with other Crocodile species revealed the presence of 12 polymorphic loci among 1156 loci studied and also 5 haplogroups were introduced. As we didn’t find any nucleotide diversity among studied samples, therefore, to increase the genetic diversity of the Gando, it is essential to include specific conservational plans for this endangered species.

    Cadmium (Cd) is a one of the heavy metals that causes oxidative stress in plants and Plants equipped with a scavenging ROS system to protect against this stress. In this study, changes of expression profile and enzyme activity of glutathione reductase (GR) were analyzed in milk thistle (Silybum marianum) seedlings in response to cadmium chloride concentrations (0, 300, 600 and 900 μM) using completely randomized design with two and five replications, respectively. Results of semi quantitative RT- PCR showed that gene expression of GR in stress plants leaves was significantly different from control plants. The lowest and highest gene expression was in control and 900 μM concentration of cadmium chloride, respectively. It could be concluded that glutathione reductase enzyme plays important role in enzymatic antioxidant defense system in response to cadmium toxicity in Silybum marianum.

    Salinity is one of the most important problems in arid and semi-arid areas that causes serious decrement in agricultural productions. Rapeseed is a superior oilseed due to the high quality of oil, large amounts of polyunsaturated fatty acids and oil yield. To deal with abiotic stress conditions, plants synthesize some essential metabolites, special structural proteins or metabolic enzymes pathways to overcome the stress. To identify the molecular mechanisms of salt responsiveness in rapeseed, proteome changes of Option500 a salt-sensitive genotype under salt stress was studied. An increase in proline and the Na of leaf and a reduction in shoot dry weight, plant height, K and Kﳖ were observed under salt stress (0, 150, 300 mM). Over 110 protein spots were identified by two-dimensional electrophoresis (2-DE) gels. 44 spots showed significant expression changes based on induction factor, which 7 spots were recognized significantly at 5% probability level. 1 and 6 spots were up and down-regulated, respectively. By using LC-MS/MS mass spectrometry analysis proteins that involved in energy production and photosynthesis were identified. Role of these proteins to salt stress response will be discussed.

    RNases participate in vital cellular functions such as DNA replication, transcription, splicing, and control of translation. S-like ribonucleases (S-like RNases) are homologous to S-ribonucleases (S-RNases), but are not involved in self-incompatibility in plants. In dicotyledonous plants, S-like RNases play an important role in phosphate recycling during senescence and are induced by inorganic phosphate-starvation and in response to defense and mechanical wounding. Potensial effects of RNases in drought response might refer to intraction of several different functions. In this study, the promoter of S-Like RNase gene was inserted upstream of β-glucuronidase gene (GUS), Agrobacterium tumefaciens, transformed by electroporation with these constructs were used for co-culture for plant transformation. Histochemical staining method on root, Leaf and anther tissue of rice were done for measering promoter activity. Based on these results, we report tissue speciefic activity of S-Like RNase promoter in the anther of rice. Results indicate the high activity of S-Like RNase promoter in the anther tissue of rice however Expression level in the leaf was low and in the root was negligible. These results is confirmed by previeos studies using q-PCR that indicated high expression of S-Like RNase gene at the anther in comparison with other tissues.

    Plants can be exploited for the production of various recombinant proteins as valuable bioreactors because of many reasons. The use of plant viral vectors for the transient expression offers as a useful strategy for the large-scale production of proteins with pharmaceutical importance, such as antibodies within a very short time period. Today, cancer is the second cause of death in human society. Compelling evidence suggests that vascular endothelial growth factor (VEGF), due to its essential role in angiogenesis, is a critical target for cancer treatment. In This study, Anti-VEGF (Nb42) nanobody different fragments were transiently expressed under the control of a viral strong promoter in cucurbit plant using ZYMV-based viral vector. These fragments included natural nanobody sequence (Nb42I) and optimized nanobody sequence (Nb42II) for expression in plant. After inoculation of plants, the presence of transcripts of the Nb42 gene fragments in cucurbit inoculated plants was detected by using RT-PCR. Also, the recombinant protein expression was confirmed by Dot blotting, SDS-PAGE, Western blotting and ELISA. Based on the ELISA results, the expression level of Nb42I and Nb42II nanobody fragments was 2 and 6.8 μg/g of leaf tissue respectively. Taken together, recombinant Nb42 nanobody could be efficiently expressed in cucurbit plant and ZYMV-based expression system would be an appropriate platform for production of recombinant proteins in cucurbit plants.

    Cumin (Cuminum cyminum L.) is a flowering plant from Apiaceae family and native to the East Mediterranean to India. The main component of essential oil in cumin seeds is cumin aldehyde (63% of total oil). Despite the importance of the cumin derivative drugs little information is available on the genome and the molecular mechanisms involved in metabolic pathway of this plant. Transcriptomic studies have greatly contributed to better understand in metabolic pathways of medicinal plants. At the moment¡ the use of next-generation sequencing techniques¡ especially RNA-seq technique were considered as the suitable promising and most accurate methods of transcriptomic evaluation. In the present study¡ we report cumin transcriptome for the first time. Flower tissue was used to extract RNA from four samples for RNA-seq analysis. According to the results¡ more than 153000000 reads with length of 50 NT were achieved. Trinity software¡ using 25 and 32 K-mers¡ was used to assemble the reads. Selection of the best assembly was followed using BUSCO software based on the integer transcript sequences. After assembly of reads¡ 50973 genes with an average length of 725 NT and N50 value of 1136 NT were obtained. Moreover¡ 53103 transcripts were identified from all genes. From this number¡ 35860 transcripts had at least one homologous in Nr database. More than 66.7% of all transcripts had at least one homologous in GO database (biological process¡ molecular function¡ cellular compound). Most of the genes were related to transcriptional regulation and membrane activities. In the present study¡ the first transcriptome profile is reported in cumin which data can be used in subsequent studies to assess expression of genes and other genetic studies in this plant.

    Basil (Ocimum basilicum L.) is one of the important medicinal plants belonging to the mint family which is a rich source of phenylpropanoid compounds such as methylchavicol and methyleugenol. Environmental stresses such as drought could change the percentage and content of the essential oils and expression level of the genes involved in their biosynthesis. In the current study, an experiment based on completely randomized design (CRD) with three replications was conducted in greenhouse to study the effect of drought stress on the expression of key genes involved in phenylpropanoids biosynthesis in O. basilicum c.v. Keshkeni luvelou. The genes studied, were phenylalanine ammonialyase (PAL), eugenol synthase 1 (EGS1) and caffeic acid O-methyltransferase (COMT). Plants were exposed to three levels of 100 (control), 75 and 50 % field capacity (FC) at the 6-8 leaf stage. The expression level of the studied genes were determined by real time PCR technique in plant leaves at the flowering stage. Analysis of gene expression data indicated that 50% FC increases the expression level of EGS1 by about 9.74, whereas those of COMT relatively remained unchanged. The expression level of PAL was increased 1.5 to 2 times under drought stress, although no significant difference for its expression was observed between 50% and 75% FC. Thus, it is possible to enhance the content of the phenylpropanoid compounds in basil through the application of controlled drought stress and consequently increasing the expression levels of EGS1 and PAL genes.

    Tobacco as a model plant (owing to certain features) is extensively used in molecular research with the aims to understand the fundamental plant and pathogen interactions. Fire blight bacterial disease of tobacco (Pseudomonas syringae pv. tabaci (Pst)) causes considerable damage to tobacco worldwide. Induction of genes associated with virulence that play a role in resistance to diseases can be effective in reducing the severity of disease. The evidence suggests that plant hormones are effective in gene expression in plants and increase the production of secondary metabolites. In addition, they stimulate the immune system of plant through the activation of transcription of genes linked with plant defense mechanisms that regulate the induced resistance. In this study, the expression of PR1, PR5, Pal, Catalase and WRKY12 in tobacco plants treated with salicylic acid at a concentration of 3 mM and tobacco plant treated with jasmonic acid at a concentration of 0.5 mM and also control plant in times 0, 12, 24, 48 and 72 after injection of Pseudomonas syrigae pv. tabaci was investigated by Quantitative Real-time PCR (QRT-PCR) technique. The results showed that jasmonic acid and particularly salicylic acid treatments caused significant changes in the expression of these genes after bacterial injection.

    Body weight is the most economically important trait in sheep industry. In genome-wide association study and genomic selection, determining of extent and level of linkage disequilibrium (LD) is critical in sample size and marker density. Moreover, selection for increasing of frequency in new mutations that are advantageous only in a subset of populations leaves some signatures in the genome. Locations of selection signatures are often correlated with genes and QTLs affecting economically important traits. Therefore, the objectives of this research were to study LD pattern and identify the genomic regions that have been under artificial and natural selection in Zandi sheep breeds with different body weight. For this purpose, the blood samples were collected form 73 Zandi sheep and 54241 markers were genotyped by using Illumina Ovine SNP50K BeadChip array based on latest assembly (Oar_4.0) sheep genome. After quality control, 40879 SNPs belonging to 71 animals were used in the final analysis. LD was calculated between all pairs were calculated with r2 by PLINK software. To detect the selection sweep, due to linkage disequilibrium, associated with these signatures we carried out the cross-population Extended Haplotype Homozygosity (XP-EHH) test. In this study, the extent of LD in this study was 40 kb with r2=0.2. The results revealed ten genomic regions on 1, 2, 3, 5, 6, 9, 12, 13, 21 and 22 chromosomes. Bioinformatics analysis demonstrated that some of these genomic regions overlapped with reported genes included in the growth traits, fat metabolism, development of the skeletal system, energy metabolism and meat quality traits such as FBP1, FAM184B, PKD2, FGF19, SPP1, MEPE CAPN2 genes.
