    شوری خاک از مهمترین تنش های غیرزیستی است که شدیدا تولید محصول را کاهش می دهد. مطالعات اخیر نشان داده است که کاربرد بیرونی سالیسیلیک اسید به عنوان یک ملکول مهم در مسیر ترارسانی در انتقال پیام های تنش های غیرزیستی نقش دارد. با وجود این نقش آن در ترارسانی تنش شوری روشن نیست. در این تحقیق اثر غلظت های مختلف سالیسیلیک اسید، در بیان ژن های گیرنده ی شبه کینازی دو رقم جو (ریحان و نصرت) متفاوت از لحاظ مقاومت به شوری مورد بررسی قرار گرفت. نتایج نشان داد که شوری و سالیسیلیک اسید تاثیر معنی داری بر صفات گیاهچه ای جو داشته اند و موجب افزایش بیان نسبی ژن های Hv3ARK و IdiRLK2 در رقم نصرت شدند. ولی در رقم ریحان بیان نسبی ژن ها متفاوت بود. به نظر می رسد سالیسیلیک اسید با فعال کردن گیرنده های شبه کینازی در مسیر انتقال پیام شوری قرار دارد. این نتایج نشان می دهد که برخی از گیرنده های شبه کینازی، ممکن است در پاسخ های بواسطه سالیسیلیک اسید در گیاهان تحت تنش های غیرزیستی نقش داشته باشند.

    دسچورازها (EC: 1.14.19.3) کاتالیزگرهای واکنش دهیدروژناسیون بوده و باعث ایجاد پیوند دوگانه در زنجیره اسیدچرب می شوند. این آنزیم ها به دو گروه محلول و غشائی تقسیم گردیده، هر یک دارای موتیف های متفاوتی می باشند. با اینکه دلتا 6 دسچوراز آنزیم کلیدی در بیوسنتز اسیدهای چرب غیراشباع می باشد، ولی بخاطر غشایی بودن، ساختار کریستالی و سه بعدی آن تعیین نگردیده است. قارچ Mortierella alpina منبع غنی تولید اسیدچرب غیراشباع اسید آراشیدونیک بوده و دارای آنزیم دلتا 6 دسچوراز فعالی می باشد. هدف از این تحقیق، دستیابی به توالی کدکننده ژن دلتا 6 دسچوراز و تعیین جایگاه دامین های کارکردی با استفاده از آنالیزهای مقایسه ای و مدل های مولکولی و پیشنهاد یک مدل توپولوژی غشائی برای این آنزیم می باشد. بدین منظور پس از استخراج RNA و سنتز cDNA توسط آغازگرهای اختصاصی، همسانه سازی در ناقلpBlueScriptSK+ انجام و سپس توالی یابی شد. بر اساس بررسی های بیوانفورماتیکی توسط سرورهای Swissmodel، Predict و Phobius وجود دامین سیتوکروم b5 حاوی موتیف (HPGG)، 3 موتیف جعبه هیستیدینی و نواحی ترانس ممبران تعیین و سپس یک مدل توپولوژی غشائی برای توالی موردنظر پیشنهاد گردید.

    بررسی تنوع ژنتیکی بهترین راه برای استفاده از ظرفیت ژنتیکی موجود در گیاه جو جهت برنامه های به نژادی می باشد. در این بررسی تنوع ژنتیکی 63 ژنوتیپ ایرانی و غیر ایرانی جو تشریح شده است. از 30 جفت نشانگر ریزماهواره استفاده شده، 29 جفت چندشکلی مشاهده شد. در مجموع 225 آلل برای مکان های ژنی مختلف با میانگین 2/7 برای هر آغازگر شناسایی گردید. بیشترین تعداد آلل برای جایگاه های Bmag0323، Hvm54 و EBmac679 (با 13 آلل) و کم ترین تعداد برای جایگاه HVM0003 (با 2 آلل) بود. میزان PIC از 89/0 (برای جایگاه EBmac679) تا 21/0 (برای جایگاه GBM1176) و مقدار شاخص شانون از 46/2I= (برای جایگاه Bmag0323) تا 48/0I= (برای جایگاه GBM1176) متغیر بود. گروه بندی ژنوتیپ ها با روش Neighbor-net، و تجزیه خوشه ایبا استفاده از روش Bayesian انجام گردید. در این روش بهترین تعداد زیر جمعیت 4 عدد شناسایی شد که در اکثر زیرجمعیت ها ژنوتیپ های با منشاء ایران دیده شدند. نتایج گروه بندی حاصل از روش Neighbor-net با روش مبتنی بر مدل مطابقت زیادی نشان داد. نتایج این تحقیق نشان داد که نشانگرهای ریزماهواره تنوع ژنوتیپ های مختلف جو را به خوبی نمایش می دهند. همچنین با توجه به نتایج به دست آمده، ژنوتیپ های بومی ایران نیز تنوع بالایی را از خود نشان دادند.

    یکی از مکانسیم های مقاومت گیاهان به تنش های غیرزیستی تولید محلول سازگار گلایسین بتائین است. کولین پیش ساز این متابولیت مهم و همچنین یک ترکیب اصلی در حفظ یکپارچگی و سیگنالینگ غشای سلولی است. در گیاهان مهمترین مرحله تولید کولین را آنزیم سیتوپلاسمی فسفو اتانول آمین N-متیل ترانسفراز (PEAMT; EC 2.1.1.103) کاتالیز می کند. در این مطالعه ژن PEAMT از یک رقم محلی گیاه اسفناج (Spinacia oleracea Iranian landrace) با استفاده از پرایمرهای اختصاصی تکثیر و در داخل وکتور کلونینگ (pJET) همسانه سازی شد. به منظور فرابیان ژن PEAMT ، سازه ژنیPBI121GUS-9:PEAMT ساخته و در نهایت به باکتری Agrobacterium سویه GV3101(PMP90) منتقل شد. تراریزش گیاهان با استفاده از روش غوطه ورسازی گل آذین انجام و آنالیز اولیه گیاهان تراریخت احتمالی با جوانه زنی بذور در محیط انتخابی حاوی آنتی بیوتیک کانامایسین بررسی شد. گیاهچه های مقاوم به کانامایسین در سطح مولکولی با استفاده از روش PCR غربالگری و تراریختی گیاهان در سطح رونوشت برداری با RT-PCR تایید شد. متعاقبا بررسی های فنوتیپی گیاهان تراریخت حاوی ژن PEAMT در شرایط تنش شوری نشان داد که طول ریشه اصلی گیاهان تراریخت از مقدار آن در گیاهان شاهد به طور معنی داری طویل تر، ریشه های فرعی گسترده تر و رشد رویشی بهتری داشته اند. اندازه گیری متابولیت گلایسین بتائین و آنزیم پراکسیداز نیز نشان داد که گیاهان تراریخته دارای محتوای گلایسین بتائین بیشتر و فعالیت آنزیم پراکسیدازی بالاتری نسبت به گیاهان شاهد داشته اند.

    خشکی یکی از عمده ترین تنش های محیطی محسوب می شود که رشد و توسعه گیاهان را تا حد زیادی تحت تاثیر قرار می دهد. واکنش های گیاهان در برابر این تنش با بروز تغییرات زیاد در شبکه های پیچیده حاوی تعداد زیادی ژن همراه است. در پژوهش حاضر تغییرات الگوی بیان ژن ها در دو ژنوتیپ حساس و متحمل گیاه برنج (به عنوان گیاه C3) و گیاه ذرت (به عنوان گیاه C4) با استفاده از آرایه های ژنوم ذرت شامل 734/ 17پروب ست و ژنوم برنج شامل 381/57 پروب ست مورد بررسی قرار گرفت. داده های ریزآرایه ، جهت شناسایی ژن های درگیر در پاسخ به تنش در دو شرایط کنترل و تنش از بانک اطلاعاتی GEO/NCBI، گرفته شد. نتایج نشان داد که به ترتیب تعداد 1861 (49/10 درصد) و 1753 (8/9 درصد) ژن در ژنوتیپ حساس و متحمل ذرت و تعداد 9252 (16 درصد) و 7971 (8/13 درصد) ژن در ژنوتیپ حساس و متحمل برنج پس از تنش خشکی در سطح یک درصد تغییر بیان معنی داری داشتند. از این تعداد به ترتیب 1012 و 175 ژن در برگ ژنوتیپ متحمل و حساس برنج و ذرت افزایش بیان معنی داری نشان دادند. دیاگرام ون نشان داد که به ترتیب تعداد 663 و 158 ژن به ترتیب و به صورت مشترک در ارقام متحمل و حساس برنج و همچنین ذرت کاهش بیان معنی داری دارند. گیاه برنج (به عنوان گیاه C3) پنج برابر در مقایسه با گیاه ذرت (به عنوان گیاه C4) واکنش گسترده تری به تنش خشکی از خود نشان داد. گروه بندی کارکردی ژن های دارای افزایش بیان در دو گونه گیاهی مشخص کرد که در ذرت گروه کارکردی پروتئین های ریبوزومی و فسفاتازها دارای بیشترین تعداد ژن هستند در حالیکه در برنج گروهای کارکردی اتصال به فلزات، پاسخ به تنش، پاسخ به محرک های زیستی و انتقال پیام بیشترین ژن ها را شامل شدند.

    زنگ های غلات از مهم ترین بیماری های گندم در ایران و اکثر مناطق جهان می باشند. یکی از موثرترین روش های کنترل این بیماری ها استفاده از ارقام مقاوم است. این تحقیق با هدف شناسایی ژن های مقاومت به بیماری زنگ سیاه و هرمی کردن آنها در ارقام گندم بهار و پیشتاز انجام شد. برای این منظور، ابتدا بیماری زایی 18 جدایه عامل زنگ سیاه جمع آوری شده از مناطق مختلف کشور بر روی ارقام Eagle، AC Cadillac، Tr129، بهار و پیشتاز مورد بررسی قرار گرفت. نتایج نشان داد که ارقام Eagle، AC Cadillac و لاین Tr129 با دارا بودن ژن های Sr42، Sr26 و SrTr6A نسبت به اکثر نژادهای مورد بررسی مقاوم بودند. برای انتقال ژن های مقاومت به ارقام بهار و پیشتاز، پس از انجام تلاقی های اولیه و تکمیلی، گیاهچه-های F1 حاصل نسبت به بیماری زنگ سیاه ارزیابی شدند و با استفاده از نشانگرهای مولکولی حضور و یا عدم حضور ژن های مقاومت به بیماری در نتاج تلاقی های تکمیلی مورد شناسایی قرار گرفت. برای تشخیص ژن Sr26 از آغازگرهای Sr26#43 و BE518379، برای شناسایی ژن Sr42 از آغازگر Srcad از نوع STS (FSD_RSA) و برای شناسایی ژن SrTr6A از آغازگرهای Gpw2295 و Gpw4032 که در بین والدین چندشکلی نشان داده بودند استفاده شد. با انجام آزمایش های گلخانه ای و نشانگرهای مولکولی مشخص شد که در تعدادی از نتاج رقم بهار انتقال ژن های مقاومت Sr26، SrTr6A وSr42 با موفقیت کامل صورت گرفته است. همچنین با توجه به توان تشخیص نشانگرهای مورد استفاده انتقال حداقل دو ژن از سه ژن مورد بررسی در نتاج رقم پیشتاز به اثبات رسید.

    Soil salinity is one of the major abiotic stresses that dramatically reducing crop productivity in the world. Recent studies indicate that the application of exogenous salicylic acid (SA) as a key molecule in the signal transduction pathway participates in the signaling of abiotic stresses. The role of salicylic acid in salinity signalling is, however, unclear. In the present study, two barley (Hordeum vulgare L.) varieties (Reyhan and Nosrat) contrasting in their salinity tolerance were used to assess the role of various SA concentrations on the expression of Receptor-like kinases (RLKs) genes. The results demonstrated that that salinity and salicylic have a significant effect on seedling traits of barley cultivars. Salinity stress and salicylic acid increased the expression of Hv3ARK, IdiRLK2 genes in the cultivar Nosrat but in the other cultivar the relative expression of the genes were different. These results suggest the involvement of a number of RLKs in SA-mediated abiotic stress responses.

    Desaturase enzymes (EC: 1.14.19.3) catalyze dehydrogenation reactions and creates a double bond in the fatty acid chains. These enzymes classified into two groups, soluble and membrane each with different consensus motifs. Despite the Delta-6 desaturase is a key enzyme in the biosynthesis of unsaturated fatty acids but due to lack of structural information of membrane-bound desaturase, it's three-dimensional and crystal structure has not been determined. Mortierella alpina is a rich source for production of unsaturated fatty acid arachidonic and has an active Delta-6 desaturases enzyme. The aim of this study was obtain encoding sequences of Delta-6 desaturase gene, and determination of functional domains using comparative analysis and molecular model and propose a membrane topology model for this enzyme. Thus, after extraction of total RNA and cDNA synthesis using gene specific primers, PCR product was cloned into pBlueScriptSK vector and then sequenced. Based on bioinformatics analysis by Swissmodel, Predict and Phobius servers presence of functional domains cytochrome b5 carrying motif (HPGG), three His-box motifs and transmembrane regions were identified. Then we proposed a membrane topology model for the Delta-6 desaturase enzyme.

    Genetic diversity is the best way to use available genetic potential for breeding programs in barley. In this study, genetic diversity of 63 Iranian and non-Iranian genotypes have been described. 29 out of 30 pairs microsatellite markers were polymorphic. A total of 225 alleles for different gene loci with an average of 2.7 per locus were identified. The highest number of alleles for Bmag0323, Hvm54 and EBmac679 (with 13 alleles) and the lowest number for the HVM0003 (with two alleles), respectively. The PIC amount and Shannon index value were variable ranging from 0.89 (for the EBmac679) to 0.21 (for the GBM1176) and from I=2.46 (for the Bmag0323) to I=0.48 (for the GBM1176), respectively. Genotype clustering was done using Neighbor-net and Cluster analysis was performed using Bayesian methods. The best number of sub-populations was identified 4 that were seen in most sub-populations genotypes originating from Iran. The results of clustering of Neighbor-net method showed good agreement with the model-based approach. The results showed that microsatellite markers well represented different genotypes of barley diversity. Also according to the results, native genotypes showed more diversity.

    One of the plant resistant mechanisms to abiotic stresses is production of a compatible solute named glycine betaine. Choline is the precursor of this important metabolite and it is also essential compound for the structural integrity and signaling of cell membrane. In plants, the most important step of choline production is catalyzed by cytoplasmic phosphoethanolamine N-methyltransferase (PEAMT ; EC 2.1.1.103) enzyme. In this study, PEAMT gene from spinach (Spinacia oleracea Iranian landrace) was amplified using specific primers and cloned into an intermediate cloning vector (pJET). In order to overexpression of PEAMT gene, the construct of PBI121GUS-9:PEAMT was made and finally was transferred to the Agrobacterium tumefaciens GV3101 (PMP90). Floral dip method was used for transformation and initial analysis of putative transgenic plants was tested in selective medium containing kanamycin. Then resistant seedlings at the molecular level were evaluated using PCR and RT-PCR methods. Results confirmed plant transformation in the level of transcription. Subsequently, The phenotypic analysis under salt stress showed that the main root length of transgenic plants was significantly longer than control nontransgenics. In adition, glycinebetaine contents and peroxidase activity were significantly higher in transgenic compare to non-transgenics control plants.

    Drought is one of the major environmental stresses that greatly affects growth and development of plants. The plants reaction against this stress is associated with showing massive changes in complex gene networks. In the present study, the changes of gene expression patterns in two sensitive and tolerant genotypes of rice (as C3 plant) and maize (as C4 plant) were investigated using maize genome arrays containing 17,734 probe sets and the rice genome containing 57/381 probe set. The microarray data were taken from the GEO/NCBI database on both stress and control conditions to identify the genes involved in responses to the stress. The results indicated that 1861 (10.49%) and 1753 (8.9%) genes in the sensitive and tolerant maize, respectively, and 9252 (16%) and 7971 (13.8%) genes in the sensitive and tolerant rice, respectively, changed significantly after drought stress at the level of one percent. From of these genes, 1012 and 175 genes in the sensitive and tolerant genotype leaf of rice and maize, were significantly up-regulated, respectively. The Venn diagram showed that 663 genes of rice and 158 genes of maize, have significantly down-regulated. Rice plant, as a C3 plant, showed five times wider reaction to drought stress in compared with maize plant, as a C4 plant. The functional grouping of the up-regulated genes in two species revealed that functional group of ribosomal proteins and phosphatases in maize plant have the most abundant categories, whereas the functional groups of metal-binding, stress response, response to biological stimuli and signals in rice plant contained the highest percentage of the genes.

    Cereal rusts are the most important wheat diseases in Iran and worldwide. One of the most effective control measures of these diseases is deployment of resistant cultivars. The objective of this study was identification of stem rust resistance genes and pyramiding these genes in wheat cultivars Bahar and Pishtaz. For this purpose, initially the virulence of eighteen isolates of stem rust that had been collected from several parts of Iran was studied in cultivars/lines Bahar, Pishtaz, Eagle, AC Cadillac and Tr129. Results showed that cultivars AC Cadillac, Eagle, and Tr129 with Sr42, Sr26 and SrTr6A genes had good levels of resistance to all evaluated isolates. In order to transfer resistance genes to Bahar and Pishtaz cultivars, after initial and complementary crosses the F1 seedling resulted were evaluated for their resistance to stem rust and consequently the progeny of complementary crosses were screened for presence or absence of resistance genes using molecular markers. For Sr26 gene, primers Sr26#43 and BE518379; for Sr42 gene, primer STS (FSD-RSA); and for SrTr6A gene, primers GPW2295 and GPW4032 that showed polymorphism between parents were used to detect presence or absence of resistance genes. Using the seedling test in green house and also detection of molecular markers, it was found that for progeny of complementary crosses in cultivar Bahar transfer of resistance genes Sr26, Sr42 and SrTr6A have been successfully completed, and in Pishtaz based on distinctive ability of markers used, at least transfer of two genes of three genes in progenies was proved.
