صفحات 986-999زمینه و هدفتشخیص سرطان پستان و درمان های مرتبط با آن تجربه عاطفی آسیب زایی برای زنان محسوب شود. پژوهش حاضر باهدف شناسایی پیامدهای روان شناختی سرطان پستان در مطالعات گذشته مبتنی بر مرور نظام مند انجام شد.مواد و روش هاپژوهش حاضر یک مرور توصیفی و با توجه به روش اجرا جزء مطالعات مرور نظام مند است. جستجوی سابقه پژوهشی با استفاده از پایگاه های داده های الکترونیکی زیر شامل پایگاه اطلاعات علمی (sid) ،magiran، medlib، sciencedirect، pubmed، google scholar، web of science، scopus، cinahl و medline از سال های 1370 تا 1396 (1991 تا 2017) در رابطه با پیامدهای روان شناختی مرتبط با سرطان پستان در ایران با استفاده از کلیدواژه های سرطان، اختلال خلقی، اختلال اضطرابی، اختلال روانی، کیفیت زندگی، بدعملکردی جنسی، اختلال خواب، سلامت روان، بی خوابی، نگرانی، تصویر بدنی، اختلال روان پزشکی، نشانگان رفتاری، درماندگی روان شناختی، رفتار مقابله ای، اختلال استرس پس از سانحه و سرطان پستان انجام شد. محتوای مقالات، بعد از استخراج، با چک لیست پریزما (prisma) مورد ارزیابی کیفی قرارگرفته و جهت بررسی داده ها از روش تحلیل محتوا استفاده شد.نتایجپس از جستجو و ارزیابی مطالعات، تحلیل نهایی بر روی 73 مقاله انجام گرفت. یافته های این پژوهش نشان داد که پیامدهای روان شناختی سرطان پستان در 8 طبقه کیفیت زندگی، اضطراب، استرس و تنیدگی، افسردگی، عملکرد جنسی، تصویر بدنی، راهبردهای مقابله ای، خستگی و کیفیت خواب جای می گیرند.نتیجه گیریبا توجه به نتایج به دست آمده، با ترسیم حیطه های مشخص شده در حوزه پیامدهای روان شناختی سرطان پستان می توان از آن به عنوان یک نقشه مفهومی برای انجام مطالعات منسجم تر و طراحی برنامه های مداخله ای گسترده استفاده نمود.کلیدواژگان: پیامدهای روان شناختی، سرطان پستان، مرور نظام مند

صفحات 1000-1011زمینه و هدفمتاستاز علت اصلی مرگ ومیر ناشی از سرطان است و حدود 90% مرگ ومیر ناشی از سرطان را تشکیل می دهد. میزان درمان سرطان به طور قابل توجهی بهبود یافته است، این پیشرفت عمدتا به دلیل تشخیص زودهنگام و مهار تکثیر سلول سرطان است. مهم ترین ویژگی یک سلول توموری بدخیم، توانایی فعالیت تهاجم و مهاجرت سلولی است. مطالعه مهاجرت سلولی و فاکتورهای موثر در آن در تشخیص سرطان، پیش آگهی، توسعه دارو و درمان سرطان ارزشمند است. تکنیک ها و روش های نوینی جهت بررسی قدرت تهاجم و مهاجرت سلولی در محیط آزمایشگاه و نیز جهت بررسی فعالیت آنزیم های درگیر در فرآیند تهاجم وجود دارند. اثر و کارآمد بودن داروها و ترکیبات مختلف ضد سرطانی را با استفاده از این روش ها در محیط آزمایشگاه و مدل حیوانی می توان موردبررسی قرار داد.نتیجه گیریدر این مقاله خلاصه ای از آزمون های مهاجرت در in-vitro شامل trans well migration assay، scratch wound assay، microfluidic chamber assay، exclusion zone assay، fence assay،micro carrier bead assay و spheroid migration assay و یک روش در in-vivo، Chick chorioallantoic membrane (CAM) assay ارائه می کنیم.در این مقاله همچنین روش های In situ Zymography،ELISA ،FRET based measurement of MMP activity ،Substrate Zymography به منظور اندازه گیری میزان آنزیم ماتریکس متالوپروتئیناز به عنوان آنزیم اصلی در تخریب ماتریکس خارج سلولی توضیح داده شده است.کلیدواژگان: ماتریکس متالوپروتئیناز، بررسی مهاجرت سلولی، تومور بدخیم

صفحات 1012-1024زمینه و هدفماکروفاژها تحت تاثیر ریز محیط، عملکردهای متنوع و به تبع آن فنوتیپ های گوناگونی را نشان می دهند. از طرفی ملاتونین به لطف عملکردهای چندگانه می تواند سبب توسعه سرطان و یا محدودیت رشد سرطان در هماهنگی با دیگر فاکتورها شود. بااین حال، هیچ اطلاعاتی در مورد نقش ملاتونین در شکل دادن به ماکروفاژها در محیط تومور وجود ندارد. هدف این مطالعه تعیین اثر سلول های ماکروفاژ تیمار شده با ملاتونین بر روی سلول های سرطانی رده K562. است.مواد و روش هابرای این منظور سلول های RAW264.7 در پلیت 24 خانه با تراکم سلولی 104× 6 برای 24 ساعت کشت داده شدند. سلول ها با غلظت های صفر، 50، 75، 100، 150 و 200 میکرومولار ملاتونین به مدت 24 ساعت تیمار شدند. سپس سلول های K562 با نسبت 1: 10 به سلول های RAW264.7 تحت تیمار اضافه شدند. قدرت زنده مانی سلول های K562 با روش MTT و NR تعیین شد و میزان آپوپتوز و نکروز در این سلول ها به وسیله ی رنگ آمیزی با آکریدین اورنج و اتیدیوم برماید ارزیابی گردید. همچنین میزان MPO و NO در سلول های RAW264.7 تیمار شده با ملاتونین موردسنجش قرار گرفت. داده ها با استفاده از آزمون کراسکال والیس و من ویتنی مورد ارزیابی قرار گرفتند (05/0P<).نتایجمیزان رشد و زنده مانی سلول های K562 در مجاورت سلول های RAW264.7 تحت تیمار با ملاتونین در غلظت های 50 و 75 میکرومولار افزایش یافت. هرچند در غلظت های بالاتر این روند تغییر یافت. میزان MPO و NO نیز با افزایش غلظت ملاتونین کاهش یافت.نتیجه گیرینتایج حاکی از افزایش قابل توجه میزان زنده مانی و سایر قابلیت های حیاتی سلول های K562 در مجاورت سلول های RAW264.7 تحت تیمار با غلظت های پایین ملاتونین است.کلیدواژگان: سیتوتوکسیتی، ماکروفاژ رده RAW264، 7، ملاتونین، K562

صفحات 1025-1035زمینه و هدفباکتری اشرشیا کلی انتروتوکسیژنیک از عوامل شایع اسهال است. فاکتورهای کلونیزاسیون و انتروتوکسین ها از عوامل اصلی حدت زا در این ارگانیسم هستند. باکتری مذکور از طریق فاکتورهای کلونیزاسیون به سطح سلول های اپیتلیال روده متصل می شود و انتروتوکسین ها را تولید می کند که این سموم باعث ترشح بیش ازحد مایعات و الکترولیت ها در لومن روده می شوند که درنهایت منجر به اسهال می گردند. به دلیل درمان مشکل و شیوع بالای این بیماری طراحی واکسن علیه این ارگانیسم یکی از اهداف سازمان بهداشت جهانی است. پروتئین CooD – CotD به عنوان زیر واحدهای راسی CS1 و CS2 نقش مهمی را در اتصال باکتری به سلول های اپیتلیال روده ایفا می کنند. در این مطالعه بیان و تخلیص ژن کد کننده پروتئین کایمریک CooD – CotD به عنوان کاندیدای واکسن های زیرواحدی انجام شد.مواد و روش هادر این مطالعه ، بهینه سازی کدونی ژن کایمریک cooD – cotD با نرم افزار Gene Designer انجام شد. ژن موردنظر با روش PCR تکثیر و در وکتور کلونینگpJET1.2/blunt همسانه سازی گردید و به منظور بیان در وکتور بیانیpET28a زیرهمسانه سازی شد. پروتئین با روش کروماتوگرافی نیکل تخلیص و با وسترن بلات ارزیابی شد.نتایجحضور باند 82 کیلودالتونی در ژل SDS-PAGE 10 درصد بیان پروتئین کایمریک CooD – CotD را نشان داد که با روش وسترن بلات تایید گردید. میزان پروتئین تخلیص شده در این تحقیق 121 میکروگرم در میلی لیتر بود.نتیجه گیریمطالعات بیان و تخلیص پروتئین نشان داد که این پروتئین دارای بیان در میزبان همولوگ است.کلیدواژگان: انتروتوکسیژنیک اشرشیا کلی، CS1، CS2، بیان نوترکیب

صفحات 1036-1045زمینه و هدفتاکنون کم تر مطالعه ای به بررسی ارتباط بار اسیدی رژیم غذایی با الگوهای غذایی سالم پرداخته است. هدف از این مطالعه بررسی ارتباط بار اسیدی رژیم غذایی با امتیاز رژیم مدیترانه ای و رژیم DASH (dietary approaches to stop hypertension) است.مواد و روش هادر این مطالعه مقطعی، 270 خانم 50-20 سال مراجعه کننده به مراکز بهداشتی شرکت داشتند. دریافت مواد غذایی با استفاده از پرسشنامه بسامد خوراک 168 آیتمی ارزیابی شد. نمایه های بار اسیدی رژیم غذایی به همراه امتیاز رژیم مدیترانه جایگزین و رژیم DASH برای افراد محاسبه شد. فعالیت بدنی با استفاده از ثبت 3 روزه فعالیت بدنی برای افراد بر اساس Met.h/day ارزیابی گردید.نتایجمیانگین سن ± انحراف معیار کلی افراد شرکت کننده 44/8 ±64/32 سال بود. میانگین امتیاز رژیم مدیترانه ای در سهک های PRAL، NEAP و DAL به طور معنی داری متفاوت بود ](001/0>P)، (01/0=P) و (003/0=P) به ترتیب برای PRAL، NEAP و [DAL. همچنین افرادی که در سهک میانی بار اسیدی رژیم غذایی قرار داشتند امتیاز DASH پایین تری نسبت به سهک اول دارا بودند ](007/0P=)، (03/0=P) و (005/0=P) به ترتیب برای PRAL، NEAP و [DAL. این نتایج برای هر دو امتیاز رژیمی حتی بعد از تعدیل کردن مخدوش گرهای مختلف همچنان معنادار باقی ماند (05/0˂p).نتیجه گیریمطالعه حاضر رابطه ای معنادار و مستقل بین نمایه های بار اسیدی رژیم غذایی و امتیاز رژیم مدیترانه جایگزین و رژیم DASH نشان می دهد. برای تایید این یافته ها اجرای مطالعات آینده نگر ضروری است.کلیدواژگان: بار اسیدی رژیم غذایی، رژیم مدیترانه جایگزین، رژیم غذایی DASH

صفحات 1046-1055زمینه و هدفآموزش بالینی، فعالیت های حمایت کننده یادگیری در محیط های بالینی است که هدف آن نائل شدن دانشجو به اهداف آموزشی جهت انجام مراقبت های بالینی و بسترسازی مناسب جهت همسوسازی دانش نظری به مهارت های عملی است، به گونه ای که شناخت وضعیت فعلی می تواند بستری جهت ارتقا کیفیت عملکرد دانشجویان اتاق عمل در آینده را فراهم سازد. ازاین رو پژوهش حاضر باهدف ارزیابی کیفیت آموزش بالینی از دیدگاه دانشجویان اتاق عمل انجام شد.مواد و روش هااین مطالعه توصیفی- تحلیلی در نیمسال دوم 1396 – 1395 به صورت سرشماری بر روی 81 نفر از دانشجویان اتاق عمل در دانشگاه علوم پزشکی فسا انجام گرفت. اطلاعات توسط پرسشنامه معتبر شامل متغیرهای دموگرافیک و دیدگاه دانشجویان درباره آموزش بالینی جمع آوری گردید. داده ها توسط نرم افزار SPSS نسخه 22 و با استفاده از روش های آماری توصیفی تجزیه وتحلیل شد.نتایجدر این مطالعه 22 نفر (2/27 درصد) مرد و 59 نفر (8/72 درصد) زن بودند. 37% دانشجویان، اعتماد مربیان بالینی به کارآموزان، 6/55% آماده سازی دانشجویان قبل از ورود به محیط بالینی، 1/48% اعتماد اساتید به دانشجویان و علاقه و رغبت به یادگیری، 6/50% اعتمادبه نفس و نگرش مثبت به رشته تحصیلی، 6/44% آشنایی کارکنان بااخلاق حرفه ای و 42% امکانات محیطی را نامطلوب گزارش نمودند.نتیجه گیریبر اساس نتایج مطالعه حاضر، آموزش بالینی در رشته اتاق عمل دارای مشکلات و کمبودهایی است که برطرف کردن مشکلات و ضعف های موجود نیازمند همت عمومی از طرف مسئولان، اساتید و کارشناسان آموزش بالینی است.کلیدواژگان: آموزش بالینی، دانشجو، اتاق عمل، ارزیابی، دیدگاه

صفحات 1056-1067زمینه و هدفآیریزین مایوکاینی است که در اثر فعالیت ورزشی حاد از عضله ترشح شده و مصرف انرژی را افزایش می دهد. هدف از پژوهش حاضر تعیین پاسخ غلظت سرمی آیریزین و مقاومت به انسولین به یک جلسه تمرین حاد پیلاتس در زنان چاق غیرفعال است.مواد و روش هاپژوهش حاضر از نوع نیمه تجربی با طرح پیش آزمون-پس آزمون بود. جامعه آماری این تحقیق را زنان چاق غیرفعال شهر اهواز تشکیل دادند که 21 آزمودنی با (میانگین سنی 0/10±1/39 سال و میانگین BMI 4/3±9/35 کیلوگرم بر مترمربع) به طور تصادفی به دو گروه تجربی (12 نفر) و کنترل (9 نفر) تقسیم شدند. گروه تمرینی به 60 دقیقه تمرین پیلاتس با شاخص درک فشار بورگ 16-14 پرداختند، درحالی که گروه کنترل هیچ گونه فعالیت ورزشی نداشتند. نمونه های خونی قبل (پس از حداقل 8 ساعت ناشتایی) و بلافاصله پس از فعالیت پیلاتس برای اندازه گیری سطوح آیریزین، گلوکز، انسولین و مقاومت به انسولین جمع آوری شد. برای تجزیه و تحلیل داده های آماری از آزمون تی وابسته و تحلیل کواریانس استفاده شد و ارتباط بین متغیرهای تحقیق با آزمون هم بستگی پیرسون محاسبه گردید.نتایجنتایج نشان داد، پس از یک جلسه تمرین پیلاتس سطوح سرمی گلوکز (23/0(P=، انسولین (44/0(P=، مقاومت به انسولین (50/0(P= و آیریزین (14/0(P= در گروه تمرین در مقایسه با گروه کنترل تغییر معناداری پیدا نکرد (05/0<P). همچنین، هیچ رابطه ای بین آیریزین با گلوکز، انسولین و شاخص مقاومت به انسولین مشاهده نشد (05/0<P).نتیجه گیریاگرچه مطالعه در این زمینه در آغاز راه است، اما به نظر می رسد تمرین حاد پیلاتس تاثیری بر سطوح گلوکز، انسولین، مقاومت به انسولین و آیریزین ندارد.کلیدواژگان: آیریزین، تمرین حاد پیلاتس، چاقی، مقاومت به انسولین

صفحات 1068-1078زمینه و هدفهورمون آنژیوپوئیتین شبه پروتئین-8 که از بافت چربی و تحت تاثیر مسیرهای پایین دست PGC-1α ترشح می شود، در بازتولید سلول های بتا نقش دارد. هدف از تحقیق حاضر ارزیابی تاثیر یک دوره تمرین استقامتی بر بیان ژن PGC-1α بافت چربی، غلظت سرمی ANGPTL8 و عملکرد سلول های بتا در رت های دیابتی بود.مواد و روش هاتحقیق حاضر به روش تجربی انجام شد. تعداد 24 رت نر نژاد ویستار به طور تصادفی در سه گروه کنترل سالم، کنترل دیابتی و ورزش استقامتی تقسیم شدند. پس از القای دیابت با استرپتوزوتوسین، گروه تمرین استقامتی چهار هفته بر روی تردمیل جوندگان به تمرین پرداخت. مقادیر نسبی بیان ژن PGC-1α بافت چربی احشایی به روش Real-time PCR، غلظت سرمی ANGPTL8 به روش الایزا و تعداد سلول های بتا با ارزیابی بافت پانکراس به روش رنگ آمیزی ائوزین- هماتوکسیلین شمارش شد. یافته ها به روش تحلیل واریانس یک طرفه مورد تجزیه وتحلیل قرار گرفت.نتایجمیزان بیان ژن PGC-1α بافت چربی در گروه تمرین استقامتی به طور معناداری بالاتر بود (00/0=P). علیرغم بالاتر بودن سطوح ANGPTL8 به دنبال چهار هفته تمرین استقامتی، این تغییر معنادار نبود (47/0=P) عملکرد سلول های بتا تغییر معناداری در گروه تمرین استقامتی نشان نداد (08/0=P) بااین حال تعداد سلول های بتا در گروه تمرین استقامتی به طور معناداری بالاتر بود (001/0=P). ارتباط مثبت و معناداری بین مقادیر نسبی ژن PGC-1α و سطوح سرمی ANGPTL8 مشاهده شد (008/0=P، 65/0r=).نتیجه گیریعلیرغم ارتباط مثبت و معنادار بین مقادیر نسبی ژن PGC-1α و هورمون ANGPTL8، افزایش سطوح ANGPTL8 معنادار نبود، بااین حال همین مقدار افزایش در گروه تمرین استقامتی همراه با افزایش تعداد سلول های بتا بود. بررسی های بیشتر با برنامه های تمرینی متفاوت توصیه می شود.کلیدواژگان: تمرین استقامتی، تکثیر سلول های بتا، ANGPTL8، PGC-1?

صفحات 1079-1087زمینه و هدفرزیستین آدیپوکینی مترشحه از بافت چربی و سلول های ایمنی است. ارتباط میان رزیستین و سندرم متابولیک و اجزای آن مورد اختلاف است، هدف از این مطالعه تعیین سطح سرمی رزیستین، مقاومت به انسولین و برخی متغیرها در افراد مبتلا و غیر مبتلا به سندرم متابولیک است.مواد و روش هادر این مطالعه مقطعی که در سال 1394 انجام شد، تعداد 56 فرد در دسترس، شامل 31 فرد مبتلا به سندرم متابولیک و 25 فرد به عنوان گروه کنترل انتخاب شدند. غلظت سرمی رزیستین با روش الایزا اندازه گیری و ارتباط آن با شاخص های سندرم متابولیک بررسی شد. داده ها با استفاده از آزمون های آماری t مستقل، کای دو و آزمون پیرسون مورد تجزیه وتحلیل قرار گرفتند.نتایجسطح سرمی رزیستین در گروه بیمار به طور معنی داری نسبت به گروه کنترل بالاتر بود (002/0=p-Value). بررسی ارتباط میان رزیستین و سایر عوامل در افراد مبتلا به سندرم متابولیک نشان داد که ارتباط معنی داری بین سطح سرمی رزیستین با شاخص توده بدنی (015/0p=؛ 338/0r=)، دور کمر (005/0p=؛ 387/0r=)، فشارخون سیستولی (001/0>p؛ 575/0=r)، فشارخون دیاستولی (001/0=p؛ 449/0=r)، کلسترول (018/0=p؛ 421/0=r) و LDL (044/0=p؛ 283/0=r) وجود دارد.نتیجه گیریسطح سرمی رزیستین در افراد مبتلا به سندرم متابولیک بالاتر بوده و در این افراد با برخی اجزای سندرم متابولیک ارتباط دارد، ممکن است رزیستین با شدت این سندرم و عوارض ناشی از آن مرتبط باشد.کلیدواژگان: رزیستین، سندرم متابولیک، مقاومت به انسولین

صفحات 1088-1096زمینه و هدفتشنج در دوران بارداری سبب آسیب نورونی در سیستم لیمبیک مغز جنین می گردد که نقص در یادگیری و اختلال شناختی را در نوزادان به همراه دارد. این مطالعه به بررسی اثر تشنجات تونیک-کلونیک پریناتال و تیمار با لیتیوم کلراید بر یادگیری و حافظه نوزادان موش های صحرایی می پردازد.مواد و روش هاموش های صحرایی باردار با تزریق درون صفاقی پنتیلن تترازول (PTZ) با دوز 40 میلی گرم/کیلوگرم وزن بدن حیوان) از روز 13 بارداری به مدت 7 روز متوالی کیندل شدند. موش های صحرایی تیمار شده با لیتیوم کلراید در گروه های PTZ+Li0.04 و PTZ+Li0.08 به ترتیب میزان 04/0 و 08/0 درصد لیتیوم را از طریق آب آشامیدنی از روز 10 بارداری تا روز 24 پس از تولد نوزادان دریافت نمودند. یادگیری فضایی به کمک ماز آبی موریس در 30 روزگی ارزیابی شد. پس از مطالعات رفتاری، سنجش سطح هیپوکامپی فاکتور نوروتروفیک مشتق از مغز (BDNF) به روش الایزا انجام شد.نتایجدر مقایسه با گروه کنترل، در گروه PTZ مدت زمان و مسافت پیمایش تا رسیدن به سکوی مخفی در آزمون ماز آبی موریس افزایش و سطح هیپوکامپی BDNF کاهش معنی داری را نشان داد. از طرفی، حیوانات دریافت کننده لیتیوم کلراید کاهش مدت زمان و مسافت پیمایش مسیر را در ماز آبی و افزایش معنی دار BDNF را در مقایسه با گروه PTZ+Saline نشان دادند.نتیجه گیریلیتیوم با افزایش سطح BDNF در هیپوکامپ سبب بهبود حافظه فضایی در مدل حیوانی تشنجات پریناتال می گردد.کلیدواژگان: تشنج، لیتیوم، دوران بارداری، موش صحرایی

صفحات 1097-1105زمینه و هدفسرطان پروستات شایع ترین سرطان مردانه و هشتمین علت مرگ در ایران است. آنژیوژنز عامل اصلی متاستاز و مقاومت به داروئی در این سرطان است. هدف این تحقیق بررسی تاثیر ده هفته تمرین تناوبی با شدت بالا بر سطوح استراحتی برخی عوامل آنژیوژنز و عملکرد ریوی مردان مبتلابه سرطان پروستات بود.مواد و روش هااین مطالعه نیمه تجربی بر روی مردان مبتلابه سرطان پروستات انجام شد. 20 مرد مبتلابه سرطان پروستات با دامنه سنی 55 تا 62 سال انتخاب و به صورت تصادفی به دو گروه کنترل (10 نفر) و تجربی (10 نفر) تقسیم شدند. گروه تجربی به مدت 10 هفته تمرین تناوبی با شدت بالا، هر هفته سه جلسه و هر جلسه 40 دقیقه با شدت 70 تا 75 ضربان قلب بیشینه را انجام دادند. 48 ساعت قبل و بعد از اجرای پروتکل تمرین تناوبی، عامل رشد اندوتلیالی عروق (VEGF)، عامل رشد فیبروبلاست (FGF)، نیتریک اکساید (NO) و عملکرد ریوی (FVC) اندازه گیری شد. تغییرات درون گروهی و بین گروهی به ترتیب با استفاده از آزمون تی وابسته و تی مستقل مورد تجزیه وتحلیل قرار گرفت.نتایجنتایج نشان داد انجام 10 هفته تمرین تناوبی با شدت بالا باعث افزایش معنی داری در سطوح VEGF(001/0P=)، FGF (001/0P=) و NO (001/0P=)، در گروه تجربی نسبت به گروه کنترل شد. همچنین، 10 هفته تمرین تناوبی باعث افزایش معنی داری در FVC(002/0P=) شد.نتیجه گیریبه نظر می رسد 10 هفته تمرین تناوبی با شدت بالا باعث افزایش عوامل آنژیوژنز و عملکرد ریوی در بیماران مبتلابه سرطان پروستات می گردد.کلیدواژگان: تمرین تناوبی با شدت بالا، سرطان پروستات، فاکتورهای آنژیوژنز، عملکرد ریوی

صفحات 1106-1115زمینه و هدفهسته های آزول، بخش بسیار مهمی از داروهای ضد میکروبی، ضد درد و ضدالتهاب را تشکیل می دهند. آزول ها گروهی از ترکیبات ضد باکتری و ضد قارچ هستند که به دلیل پایین بودن سمیت و اثربخشی بالا، دارای طیف گسترده ای از فعالیت می باشند. امروزه تلاش های فراوان برای توسعه داروهای ضد باکتری و ضد قارچ با سمیت کمتر، اثربخشی بیشتر و مکانیسم عمل جدید صورت گرفته است. در این مطالعه تلاش شده است تا اثرات ضد باکتریایی و ضد قارچی مشتقات جدید آزولی را بررسی کنیم.مواد و روش هااین مطالعه به صورت توصیفی - مقطعی انجام شد. فعالیت ضد باکتریایی و ضد قارچی ترکیبات طراحی شده به ترتیب بر روی سوش های استاندارد گرم منفی و گرم مثبت و قارچ های آسپرژیلوس نیجر، آسپرژیلوس فلاووس، کاندیدا آلبیکانز به کمک روش حداقل غلظت مهارکنندگی انجام شد.نتایجاز میان ترکیبات موردمطالعه، ترکیب حاوی حلقه هتروسیکل 2- متیل، 4- ایمیدازول اثر ضد باکتریایی قوی را در برابر سودوموناس آئروژینوزا، اشرشیاکلی و استافیلوکوک اورئوس، باسیلوس سابتلیس به اثبات رساند، درحالی که ترکیبات حاوی حلقه هتروسیکل ایمیدازول و بنزایمیدازول اثرات ضد قارچی در برابر آسپرژیلوس نیجر، آسپرژیلوس فلاووس، کاندیدا آلبیکانز نشان دادند. حداقل غلظت مهارکنندگی از رشد این باکتری ها و قارچ ها از 5/12 تا 200 میلی گرم بر میلی لیتر متغیر است.نتیجه گیرینتایج حاکی از آن است که ترکیبات طراحی شده حاوی حلقه های هتروسیکل 2- متیل، 4- ایمیدازول، ایمیدازول و بنزایمیدازول در مطالعه ی حاضر اثرات ضد باکتریایی و ضد قارچی امیدبخشی از خود نشان دادند. از طرفی ترکیبات مورد تست دارای طیف اثر مناسبی بودند، بدین جهت که بر روی هر دو سویه گرم مثبت و گرم منفی اثر ضد باکتریایی نشان دادند.کلیدواژگان: عوامل ضد باکتری، عوامل ضد قارچ، آزول ها، سمیت، حداقل غلظت مهارکنندگی رشد

صفحات 1116-1126زمینه و هدفسطح بیان فاکتور NF-κB (فاکتور رونویسی که باعث افزایش بیان ژن های التهابی می شود) اغلب در سرطان های مختلف انسانی افزایش می یابد، لذا مهارکننده های این فاکتور مانند سلاسترول می توانند جهت جلوگیری از پیشرفت سرطان کاربرد داشته باشند. این مطالعه باهدف بررسی اثر ضد سرطانی سلاسترول بر روی سلول های K562 انجام شد.مواد و روش هاابتدا سلول های K562 کشت داده شدند و اثرات سیتوتوکسیسیته ترکیب سلاسترول با آزمون MTT تعیین شد. از رنگ آمیزی هوخست و الکتروفورز DNA برای بررسی وقوع آپوپتوز استفاده شد. تجزیه وتحلیل داده ها توسط نرم افزار SPSS ویرایش 16 و آزمون آماری ANOVA انجام گردید (05/0(P< .نتایجبررسی آماری داده های به دست آمده از آزمون MTT نشان داد که رشد سلول های تیمار شده با غلظت های مختلف سلاسترول به صورت معنی داری کاهش پیدا کرده است (05/0(P< که این تاثیر مهاری سلاسترول وابسته به غلظت و مدت زمان تیمار است، لذا بیشترین تاثیر در غلظت μM 8 و 72 ساعت مشاهده شد. غلظت IC50 سلاسترول برابر با 4 میکرومولار به دست آمد. نتایج و تصاویر به دست آمده از الکتروفورز DNA و رنگ آمیزی هوخست سلول های تیمار شده، قطعه قطعه شدن DNA و هسته سلول را نشان داد.نتیجه گیریبر اساس نتایج به دست آمده، سلاسترول باعث کاهش بقای سلولی (05/0(P< و القا آپوپتوز در سلول های K562 می شود و تاثیر آن وابسته به غلظت ترکیب و مدت زمان تیمار است. درنتیجه احتمالا از این ترکیب می توان به عنوان یک داروی ضد سرطان در درمان لوسمی میلوئید مزمن استفاده کرد.کلیدواژگان: سلاسترول، رده سلولی K562، اثرات ضد سرطانی، لوسمی

صفحات 1127-1135زمینه و هدفگیرنده ی کبدی ایکس آلفا (LXRα) از خانواده ی بزرگ گیرنده های هسته ای هورمونی است که در هموستاز کلسترول نقش دارد. هدف از پژوهش حاضر، تاثیر ده هفته ورزش شنای استقامتی متوسط و بلند مدت، بر بیان ژن گیرنده ی کبدی ایکس آلفا و بررسی سطوح سرمی کلسترول و تری گلیسرید در موش های صحرایی نر بود.مواد و روش هاتعداد 18 سر موش نر صحرایی در محدوده ی وزنی 25±275 به صورت تصادفی به سه گروه شش تایی کنترل، شنای متوسط و طولانی تقسیم شدند. گروه های تمرینی به مدت ده هفته و 5روز در هفته در آب 32 درجه شنا کردند. گروه متوسط هر جلسه یک ساعت و گروه طولانی هر جلسه سه ساعت شنا کردند. پس از اتمام تمرین، نمونه گیری خونی از سیاهرگ زیرین و بافت برداری از کبد موش ها انجام گرفت. برای سنجش بیان ژن LXRα از روش real-time PCR توسط مستر میکس Ampliqon و کیت سنتز DNA استفاده شد. تفاوت ها با روش ANOVA One-Way، و مقایسه ی گروه ها توسط تست تعقیبی Tukey تعیین گردید. از همبستگی پیرسون جهت تعیین همبستگی سطوح بیان ژنLXRα با کلسترول و تری گلیسیرید استفاده شد. نرم افزار SPSS نسخه20 در سطح 05/0p≤ به کاربرده شد.نتایجگروه تمرین متوسط در مقایسه با هر دو گروه کنترل و طولانی، افزایش معنادار ژن LXRα، و کاهش معنی دار کلسترول و تری گلیسرید را نشان داد (001/0>p). همچنین بیان ژن LXRα در این گروه، همبستگی منفی و معناداری با کلسترول (rs= -0.912) و تری گلیسرید (rs= -0.862) داشت.نتیجه گیریبه نظر می رسد مکانیسم مثبت ناشی از شنای متوسط از طریق افزایش بیان ژن LXRα، منجر به خروج شاخص های لیپیدی سلولی می گردد.کلیدواژگان: تمرین شنای استقامتی، ژن LXR?، کلسترول، تری گلیسرید

صفحات 1136-1145زمینه و هدفکولیت اولسراتیو از علل شایع بیماری های دستگاه گوارش در کشورهای توسعه یافته است. این بیماری باعث ازکارافتادگی به خصوص در بالغین جوان می شود و بار اقتصادی و اجتماعی زیادی را به جامعه تحمیل می کند. کنترل التهاب و کاستن از اثرات سوء داروها از اهداف مهم درمان کولیت اولسراتیو است. هدف از مطالعه حاضر بررسی اثرات فاسیولا هپاتیکا در مدل کولیت اولسراتیو ناشی از اسید استیک در رت است.مواد و روش هابعد از القای کولیت، رت ها در سه گروه ده تایی (گروه بدون تیمار، گروه تیمار شده با پردنیزولون (mg/Kg5/0-زیر جلدی) و درمان شده با انگل فاسیولا هپاتیکا (mg/Kg 100-زیر جلدی) تقسیم شدند. پس از 10 روز رت ها آسان کشی شده و ازنظر ضایعات ماکروسکوپی و تولید واسطه های التهابی در بافت روده و میزان تولید سایتوکاین های TNF-α و IL-6 موردبررسی قرار گرفتند.نتایجنتایج به دست آمده نشان داد که کاهش میزان تولید میلوپراکسیداز بافتی در گروه درمانی فاسیولاهپاتیکا در مقایسه با پریدنوزولون تفاوت آماری معناداری ندارد. میزان تولید سایتوکاین های TNF-α و IL-6 در گروه های درمانی فاسیولا هپاتیکا و پردنیزولون نسبت به گروه بیمار کاهش معناداری یافته بود. البته میزان کاهش در گروه درمان شده با پریدنیزولون ازنظر آماری بیشتر بود. بالعکس کاهش میزان پراکسیداسیون لیپیدی در گروه درمان شده نسبت به گروه پردنیزولون بیشتر بود. بااین حال شدت کاهش علائم گراس و همچنین شاخص بقای رت ها در هر دو گروه درمان شده با پریدنیزولون و درمان شده با فاسیولا هپاتیکا یکسان بود.نتیجه گیریازآنجایی که دست یابی به فاسیولا هپاتیکا ساده و سریع است، ممکن است از آن به عنوان یک روش درمانی جهت تخفیف علائم کولیت اولسراتیو استفاده شود.کلیدواژگان: کولیت اولسراتیو، اسید استیک، رت، فاسیولا هپاتیکا، التهاب

صفحات 1146-1156زمینه و هدفطی مطالعات پیشین اثرات محدودیت جیره های غذایی بر تغییر پاسخ های ایمنی و همچنین محتوای دوپامین مغز مشخص شده است. از طرفی نشان داده شده است که سلول های ایمنی علاوه بر تولید دوپامین، خود نیز دارای گیرنده های دوپامینی هستند. هدف از این تحقیق ارزیابی مهار گیرنده های دوپامینی بر عملکردهای سلول های تک هسته ای خون محیطی موش صحرائی تحت محدودیت غذایی بوده است.مواد و روش هادر این مطالعه تجربی، تعداد 36 سر موش صحرائی نر بالغ نژاد ویستار، با میانگین وزنی 250-200 گرم در شش گروه شش تایی شامل گروه های کنترل، محدودیت غذایی 25%، محدودیت غذایی 50%، محدودیت غذایی 75%، دریافت کننده سولپیراید و محدودیت غذایی 75% و دریافت کننده سولپیراید، قرار گرفتند. سولپیراید با غلظت μg/rat50 به صورت درون بطن مغزی یک بار در روز 21 پس از آغاز تیمار تزریق گردید. در انتها، از موش ها خون گیری شد و سلول های تک هسته ای خون محیطی به روش گرادیان فایکول جداسازی شدند.نتایجمحدودیت غذایی موجب کاهش معنی دار شاخص فعالیت سلول های مونوسیت موجود در گرادیان سلول های تک هسته ای خون محیطی از قبیل تست برداشت نوترال رد و قابلیت انفجار تنفسی (تست احیای NBT) هم زمان با کاهش قابلیت تکثیری سلول های لنفوسیت موجود در گرادیان سلول های تک هسته ای خون محیطی نسبت به میتوژن فیتوهماگلوتین شد. تجویز سولپیراید همراه با رژیم غذایی دارای محدودیت 75% منجر به برگشت این عملکردهای سلول های مونوسیت و همچنین قابلیت تکثیر لنفوسیتی شد.نتیجه گیریتجویز داخل بطن مغزی آنتاگونیست گیرنده های D2 دوپامینی (سولپیراید)، به نحو موثری از اثرات مخرب محدودیت شدید رژیم غذایی بر مهار عملکردهای ایمنی ممانعت به عمل می آورد.کلیدواژگان: دوپامین، سلول های تک هسته ای خون محیطی، محدودیت غذایی، سولپیراید، موش صحرائی

صفحات 1157-1166زمینه و هدفلوسمی میلوئیدی حاد (Acute myeloid leukemia, AML)، سومین سرطان شایع در ایران و پنجمین رتبه را در جهان دارد و حدود 8% از کل سرطان ها را به خود اختصاص داده است. مسیر Janus kinas و فعال کننده رونویسی (JAK-STAT)، در تنظیم تکثیر سلولی، تمایز، آپاپتوز و همچنین در ایجاد بعضی از بیماری های خونی نقش دارد. مسیر JAK/STAT یک محور سیگنالینگ مهم است که فعال شدن غیرطبیعی این مسیر باعث پیشرفت در مسیر ابتلا به سرطان خون می گردد. یکی از ژن های مهم این مسیر، ژن JAK2 است و هدف از این مطالعه، ارزیابی بیان JAK2 در بیماران مبتلابه AML است.مواد و روش هادر این مطالعه شاهد- موردی، نمونه خونی از 30 بیمار مبتلابه AML مراجعه کننده به بیمارستان میرزا کوچک خان جنگلی، تهران در سال 1395 و 20 نمونه سالم به عنوان کنترل جمع آوری گردید. پس از استخراج RNA و سنتز cDNA، بیان ژن JAK2 با روش Real time PCR به روش محاسباتی CT∆∆ بررسی گردید. تجزیه وتحلیل آماری با نرم افزار Grafpad prism 6 و با بکار بردن t-test صورت گرفت. مقادیر P<0.05 معنادار در نظر گرفته شد.نتایجبررسی نتایج نشان می دهد که سطح بیان ژن JAK2 در بیماران در مقایسه با گروه کنترل افزایش چشمگیری داشته است که ازنظر آماری نیز معنی دار بوده است (P=0.0001). درحالی که ارتباط معناداری بین سن (P=0.753) و جنسیت بیماران(P=0.494) با افزایش بیان ژن JAK2 در جمعیت موردمطالعه وجود ندارد.نتیجه گیریبررسی میزان بیان ژن JAK2 می تواند در تعیین پیش آگهی بیماران مبتلابه AML نقش مهمی داشته باشد.کلیدواژگان: لوسمی میلوئیدی حاد، ژن JAK2، Real-time PCR

صفحات 1167-1176زمینه و هدفبیان ژن عامل رشد عصبی مشتق شده از مغز و گیرنده آن در اثر تمرینات مقاومتی در بیماران آلزایمری به خوبی روشن نشده است. هدف از تحقیق حاضر بررسی تاثیر یک دوره تمرین مقاومتی بر بیان ژن BDNF و گیرنده های TrkB در موش های صحرایی نر نژاد ویستار مدل آلزایمری بود.مواد و روش هادر این مطالعه تجربی 32 سر موش صحرایی نر بالغ با میانگین وزن 230 تا 280 گرم انتخاب و به دو گروه اصلی آلزایمری و شم تقسیم شدند. حیوانات در گروه شم نرمال سالین دریافت نمودند و در گروه آلزایمری به وسیله تزریق داخل بطنی STZ آلزایمری شدند. سپس به چهار زیر گروه: 1. شم استراحت 2. شم ورزش 3. آلزایمر استراحت 4. آلزایمر ورزش تقسیم شدند. در گروه های تمرین، رت ها به مدت 8 هفته و هر هفته سه روز تمرین کردند. تمرین شامل بالا بردن وزنه از نردبان بود. موش ها پس از 24ساعت از آخرین جلسه تمرین قربانی شدند و هیپوکامپ آن ها جهت انجام آزمایش های سلولی مولکولی برداشته شد. به منظور بررسی بیان ژن، از روش RT-PCR و برای تحلیل آماری داده ها از آزمون t مستقل استفاده شد.نتایجبین میزان BDNF و TrkB در گروه موش های استراحت کننده و ورزش کننده تفاوت معنی داری وجود داشت (05/0>p) و در گروه موش های آلزایمری ورزش بسیار بیشتر بود.نتیجه گیریبه نظر می رسد تمرین مقاومتی باعث افزایش بیان ژن BDNF و گیرنده TrkB در موش های نر نژاد ویستار مدل آلزایمری می گردد.کلیدواژگان: آلزایمر، بیان ژن، تمرین مقاومتی، BDNF، گیرنده TrkB، موش صحرایی

The present study aimed to identify the psychological consequences of breast cancer in previous studies based on systematic review.Materials & MethodsThe present study is a descriptive survey and, according to the method of implementation, is a systematic review study. A literature search was conducted by using following electronic databases including scientific information databases (SID), Magiran, Medlib, Sciencedirect, PubMed, Google Scholar, Web Of Science, Scopus, cinahl and medline from 1991 through 2017 regarding the psychological consequences associated with breast cancer in iran Using keywords cancer, mood disorder, anxiety disorder, mental disorder, life quality, sexual dysfunction, sleep disorder, mental hygiene, insomnia, anxiety, body image, mental disorder, behavioral symptom, psychological distress, coping behaviors, traumatic stress disorder and breast cancer. The content of all articles, after extraction from the bases, was evaluated by the prisma checklist and the Content analysis was used to examine dataResultsAfter the search and evaluation of the studies, the final analysis was carried out on 73 articles. The findings of this study showed that the psychological consequences of breast cancer in 8 categories are quality of life, anxiety, stress and stress, depression, sexual function, body image, coping strategies, fatigue and quality of sleep.ConclusionsBased on the results, drawing on the areas identified in the field of psychological outcomes of breast cancer can be used as a conceptual map for more coherent studies and the design of extensive interventions.Keywords: Psychological Consequences, Breast Neoplasms, Systematic Review

New methods are available to investigate the invasion, migration and the activity of enzymes involved in the invasion process in the laboratory. Due to the multiple functions, Melatonin can promote the cancer progression or cancer regression in relation to other factors. However, there is no information about the role of melatonin on the formation of macrophages in the tumor environment. The purpose of the present study is to evaluate the effects of melatonin treated RAW264.7 macrophages on the growth rate of erythroleukemia cell line K562.Materials & MethodsRAW264.7 cells were cultured in the 24-well plates at a density of 6 × 104 cells per well. After culturing for 24 h, the cells were treated for 24 h with melatonin at concentrations 0, 50, 75, 100, 150, 200 μmol/L. After removal of the supernatant, macrophages and K562 were co-cultured in 1:10 ratio. K562 vitality was then determined by MTT and NR assay. The rate of apoptosis in the cell population was evaluated by staining with acridine orange and ethidium bromide colors. Also, NO and MPO were measured in the supernatant of macrophages RAW264.7. Data were statistically analyzed using Kruskal-Wallis and Mann–Whitney U test (P<0.05).ResultsThe growth rate and vitality of K562 cells co-cultured with melatonin-treated macrophages was increased at concentrations of 50 and 75 μmol/L. Nevertheless, at higher concentrations of melatonin, this process was reversed. The levels of MPO and NO were decreased by increasing concentrations of melatonin.ConclusionThese results indicated that treatment of macrophage with melatonin, especially at lower doses (50 and 75μmol/L) could remarkably increase the K562 viability and vitality.Keywords: Cytotoxicity, Melatonin, RAW264.7 Macrophage, K562

Enterotoxins and the colonization factors (CFs) are major virulence factors in ETEC infections. The bacterium binds to the intestinal epithelial cell surface through colonization factors and produces enterotoxins that cause excessive fluid and electrolyte secretion in the lumen of the intestine, which ultimately leads to diarrhea. Due to the difficult of treatment and the high prevalence of this disease, the design of the vaccine against this organism is one of the goals of the World Health Organization. The CooD-CotD protein, as adhesion tip subunits of CS1 and CS2, plays an important role in bacterial attachment to the intestinal epithelial cells. In this study, the expression and purification of chimeric protein CooD-CotD was carried out with the aim of investigating as candidate vaccine.Materials & MethodsIn this study, codon optimization of cooD-cotD chimeric gene was performed by Gene Designer software. The gene was amplified by PCR and cloned into pJET1.2 / blunt then it was subcloned into pET28a to express chimeric protein. Recombinant protein was purified following expression, using Ni-NTA affinity chromatography and confirmed by western blotting analysis.ResultsThe presence of 82kDa in the SDS-PAGE gel showed that expression of CooD-CotD chimeric protein and confirmed by western blotting analysis. Pages 1036-1045Background & ObjectiveThere are few studies that have been examined the association between dietary acid load and healthy diet patterns. The purpose of this study was to evaluate the association between dietary acid load with alternative Mediterranean diet score (aMED) and dietary approaches to stop hypertension (DASH).Matrrials & MethodesThis cross-sectional study was conducted among 270 women with age range 20-50 years in Tehran, Iran. Usual dietary intakes were assessed by a semi-quantitative food frequency questionnaire, which contains 168 food items. Potential renal acid load (PRAL) and Net Endogenous Acid Production (NEAP) and Dietary acid load (DAL) was calculated for each person. The scores of aMED and DASH were estimated for participants.ResultsThe mean ± SD of age was 32.64 ±8.44 years. AMED scores were significant difference among the PRAL, NEAP and DAL tertiles (P<0.001, P=0.01 and P=0.003 for PRAL, NEAP and DAL respectively). Also, the women who were in the middle tertiles of dietary acid load indices, have lower DASH scores compared with the lower tertiles (P=0.007, P=0.03 and P=0.005 for PRAL, NEAP and DAL respectively). Thus the aim of this study was to evaluation the quality of clinical education from Perspectives of Operating Room StudentsMaterials & MethodsThis descriptive and cross-sectional study was conducted on 81 operating room students of Fasa University of Medical Sciences Using consensus sampling method in 2015 -2016. Data were collected using a valid questionnaire including demographic variables and assessing the viewpoints of students about clinical education. Data were analyzed using descriptive statistics in SPSS v. 22.ResultsIn this study, 22 (27.2%) men and 59 (72.8%) were female. According to study results from the perspective of students (37%) confidence clinical instructors to students, 6/55% to prepare students to enter the clinical setting, 48.1% confidence and willingness of teachers to students and to students, 55.6% Prepare students to enter the learning, 50.6% confidence and a positive attitude to education, 44.6% Personnel familiar with the professional ethics and42% Adverse environmental features.ConclusionBased on the results of this study, clinical education in the operating room has problems and shortcomings that needs to be addressed by authorities, professors and clinical education experts in order to resolve existing problems and weaknesses.Keywords: Clinical education, Operating Room, Students, Evaluate, Perspectives

The purpose of this study was to determine the response of irisin serum and insulin resistance to acute pilates training sessions pilates training in obese sedentary women.Materials & MethodsThis research is a semi experimental study with pre-and post-test design. The statistical population of this study was made up of disabled obese women in Ahwaz, with 21 subjects (mean age 39.1±10.0 years and BMI 35.9±3.4 kg/m2) were randomly divided into Pilates group (n=12) and control group (n=9). The training group performed 60-minute Pilates training with a14-16 Borg perceived perception index, while the control group did not exercise. After at least 8 hours-fasting, blood samples were collected before and immediately after each activity, to measure the levels of Irisin, glucose, insulin and insulin resistance. Data were analyzed with paired-sample t-test and ANCOVA and the relationship between the variables was calculated by Pearson correlation test.ResultsThe results indicated no significant changes in blood glucose, insulin, insulin resistance index and serum Irisin levels in Pilate’s group compared with the control group after a Pilates exercise session (p≥0.05). Furthermore, correlation was not between Irisin to, glucose, insulin, and insulin resistance index (p≥0.05).ConclusionAlthough research on this regard is a beginning pathway. Pages 1068-1078Background & ObjectiveAngiopoietin-like Proteins 8 (ANGPTL8) which secreted form adipose tissue due to downstream PGC-1α pathways, is the main factor for regeneration of beta cell. The aim of this study was to investigate the effect of 4 endurance training program on PGC-1α expression in adipose tissue, ANGPTL8 serum concentration and beta cell function (HOMA.B) in diabetic rats.Materials & methodsMale Wistar rats (N=24) divided in 3 groups include healthy control (HC), diabetic control (DC) and endurance training (ET). After induction of diabetes with STZ, training groups performed 4 weeks endurance training and real-time polymerase chain reaction (Real-Time PCR) method used for the relative expression of PGC-1α in visceral adipose tissue, the enzyme-linked immunosorbent assay (ELISA) method for measuring serum ANGPTL8 levels. Also beta cell number was counted with pancreases hematoxylin-eosin evaluation. Results analyzed with ANOVA.ResultsThe relative expression of mRNA PGC-1α was significantly increased in ET (P=0.001). Although ANGPTL8 levels increased in ET group, this change wasn’t significant (P=0.47). HOMA.B didn’t showed any significant change in ET (P=.0.08) but the number of beta cells in this group significantly increased (P=0.001). There was a positive correlation between relative mRNA PGC-1α and ANGPTL8 levels.ConclusionDespite the positive and significant correlation between relative expression of mRNA PGC-1α and ANGPTL8, this increase wasn’t significant, but this could increases the number of beta cell in endurance training group. Pages 1079-1087Background &ObjectiveResistin is an adipokin secreted from adipose tissue and immunity cells. Relationship between resistin and metabolic syndrome and its components is controversial. The aim of this study is measurement of serum resistin level, insulin resistance and other factors in subjects with and without metabolic syndrome.Materials & Methodsthis cross-sectional study was conducted in 2015, 56 availableperson including; 31subjects were enrolled as the study group and 25 subjects were recruited as the control group of the study. Serum resistin concentrations were measured by ELISA and its association with metabolic syndrome markers was measured. Data were Analyzed using independent T test, Pearson correlation Test and, Chi-squared TestResultsSerum resistin levels were significantly higher in the metabolic syndrome subject compared with control group (p-value=0.002). Correlation analysis revealed body mass index (p=0.018; r=0.480), waist circumference (p=0.015; r=0.431), systolic blood pressure (p=0.008; r=0.467), diastolic blood pressure (p=0.001; r=0.562), cholesterol (p=0.018; r=0.421) and LDL (p=0.003; r=0.515) were positively correlated with plasma resistin.ConclusionSerum resistin levels are elevated in subjects with metabolic syndrome and are associated with some of its components in metabolic syndrome patients, resistin may be associated with the severity of this syndrome and its complications.Keywords: Resistin, metabolic syndrome, Insulin Resistance

Pages 1088-1096Background & ObjectivePrenatal seizures cause neuronal damage in the limbic area of fetal brain leading to learning deficits and cognitive impairment in newborns. This study examined the effect of prenatal tonic-clonic seizure and lithium chloride (LiCl) therapy on the learning and memory of rat’s pups.Materials & MethodsPregnant Wistar rats were kindled by (i.p) injections of 40 mg/kg/BW of Penthylentetrazole (PTZ) on embryonic day (ED) 13 for 7 consecutive days. LiCl treated rats in PTZ+Li0.04, and PTZ+Li0.08 groups received 0.04% and 0.08% of LiCl in tap water from ED10 to postnatal day (PND) 24, respectively. The spatial performance learning was evaluated at PND 30 using Morris water maze (MWM). After behavioral study, the hippocampal level of BDNF was measured using ELISA.ResultsIn comparison to controls, PTZ group showed a significant increase in latency time and distance to hidden platform in MWM test and decrease level of hippocampal BDNF. LiCl-treated rats showed a significant lower latency time and distance to hidden platform in MWM and higher level of BDNF than PTZ group.ConclusionThe LiCl with elevated of BDNF in hippocampus can ameliorate spatial memory in prenatal seizure model.Keywords: Seizure, Lithium, Pregnancy, Rat

Pages 1097-1105Background & ObjectiveProstate cancer is the most common malignancy and the eighth cause of death in Iran. Angiogenesis is the major cause of metastatic and drug resistance in this cancer. The aim of this study was to investigate the effects of 10 weeks of high intensity periodic exercises on resting levels of some angiogenesis markers and also the pulmonary function of men with prostate cancer.Materials & Methoed20 male patients with prostate cancer aged 55 to 62 years old were randomly divided into two groups (n = 10) and experimental (n = 10). The experimental group subjected to 10 weeks of periodic exercise with intensity progressed weekly, three sessions for 40 minutes and intensity of 70-75 was set for each session. 48 hours before and after exercise, the serum levels of VEGF, FGF, nitric oxide (NO) and also pulmonary function were measured. Intra and intergroup changes were analyzed using dependent t-test and independent t-test, respectively.ResultsThe results showed that 10 weeks of severe periodic training significantly increased the levels of VEGF (P = 0.001), FGF (P = 0.001) and NO (P = 0.001) in the experimental group compared with the control group. Moreover, considering the results, FVC in patients with periodic exercise has increased significantly (P = 0.002).ConclusionAs for results, it can be concluded that 10 weeks of periodic exercise increases the angiogenesis and pulmonary function in patients suffered from prostate cancer.Keywords: High-intensity interval training, prostate cancer, angiogenesis markers, pulmonary function

The azole class of compounds is the most popular among the antibacterial and antifungal classes because of its lower toxicity, higher efficacy and a broad spectrum of activity. Today, Efforts have focused on the development of new, less toxic and more efficacious antifungal and antibacterial drugs, with the novel mechanism of action. In this study, we tried to investigate the antibacterial and antifungal effects of the new azole derivatives.Materials & methodsThis research is a descriptive – cross sectional study. The antibacterial and antifungal activity of compounds was evaluated using the minimum inhibitory concentration (MIC) method on a series of gram positive and gram negative bacterial strains and Aspergillus niger, Aspergillus flavus, Candida albicans. The Minimum Inhibitory Concentration from the growth of this bacteria and fung ranges from 12.5 mg/ml to 200 mg/ml.ResultsAmong the tested compounds, 4- Nitro, 2- methyl imidazole hetrocycle-pathic compound exerted potent in vitro antibacterial activity against Pseudomonas aeruginosa, Escherichia coli, Bacillus subtilis and Staphylococcus aureus while compounds (imidazole, benzimidazole hetrocycle-pathic compound) exhibited potent in vitro antifungal activity against Candida albicans, Aspergillus flavus and Aspergillus niger.ConclusionOur results indicated that the designed derivatives (4- Nitro, 2- methyl imidazole, imidazole and benzimidazole hetrocycle-pathic compounds) showed effective antibacterial and antifungal activity mainly against a series of gram positive and gram negative bacterial strains. Pages 1116-1126Background and ObjectiveThe level of NF-κB factor expression (a transcriptional factor which increases the expression of inflammatory genes) is often increased in various human cancers. Therefore, NF-κB inhibitors such as Celastrol may prevent cancer development. The purpose of this study was to evaluate the anticancer effects of Celastrol on K562 cells proliferation.Materials and MethodsFirst, the K562 cells were cultured and cytotoxicity effects of celastrol were determined by MTT assay. Hoechst staining and DNA electrophoresis are used to check apoptosis. Data analysis was performed using SPSS, version 16 and ANOVA test (P<0.05)ResultsStatistical analysis of MTT assay data showed that the growth of treated cells with different concentrations of the Celastrol significantly decreased (P<0.05) and inhibitory effect of Celastrol was time and concentration–dependent; so in higher concentrations (8 µM) and 72 hours, the maximum effect has occurred. The IC50 value of Celastrol was obtained 4 µM. Also, the results of Hoechst staining and DNA electrophoresis showed that Celastrol caused fragmentation of cell nucleus and DNA.ConclusionBased on the results, Celastrol decreases cells viability (P<0.05) and induces apoptosis in K562 cells, its effect is time and dose-dependent. The purpose of the present study was to investigate the effect of ten weeks of moderate and long term endurance exercise on the expression of the LXRα gene and the evaluation of serum levels of cholesterol and triglyceride in male rats.Materials & Methods18 male Wistar rats were randomly divided in to three groups of six, control, moderate and long swimming, in the range of 275±25g. The training groups swam in 32 degrees water for ten weeks and 5 days a week. The moderate group swam for one hour and a long group each session for three hours. After the completion of the exercise, blood sampling from the underlying vena-cava and tissue from the rat liver was performed. Real-time PCR was used to measure the of LXRα gene expression by Ampliqon mixer and DNA synthesis kits. Differences were determined by ANOVA One-Way method and the groups were compared by Tukey follow-up test. Pearson correlation was used to determine the correlation between levels of expression of LXRα gene with cholesterol and triglyceride. SPSS software version20 was used at the significance level of p≤0.05.ResultsThe moderate exercise group showed a significant increase in LXRα gene and a significant reduction in cholesterol and triglyceride compared to both control and prolonged groups (p=0.001). Also, LXRα gene expression in this group had a negative and significant correlation with cholesterol (rs=-0.912) and triglyceride (rs=-0.862).ConclusionIt seems that the positive mechanism induced by moderate swelling by increasing the expression of the LXRα gene results in the release of cell lipid makers.Keywords: Endurance swimming training, LXR? gene, Cholesterol, Triglyceride

This disease causes disability, especially in young adults, and imposes many social and economic burdens on the community. The aim of this study was to investigate the effects Effect of Fasciola hepatica extract on acetic acid induced ulcerative colitis in rat model.Materials & MethodsThirty adult male Wistar rats were divided into 3 equal groups. Colitis was induced by administration of 4% acetic acid. Control group just was administrated phosphate-bufferedsaline (S.C.) and the other groups were received Prednisolone (0.5 mg/kg) and Fasciola hepatica extract (100 mg/kg) for 10 consecutive days.Since then, the animals were euthanized and intestinal tissue and blood samples were taken. Macroscopic and microscopic features, inflammatory mediators as well as cytokines, TNF-α and IL-6 were assessed.ResultsThe amount of the myeloperoxidase production was the same between the two treatment groups. As compared to the control, both of the treatment groups produced lower amount of TNF-α and IL-6, however this reduction was more pronounced in Prednisolone treated animals than those of extract treated. On the other hand, it has been shown that immune cells, in addition to dopamine production, also have dopamine receptors. The purpose of this study was to evaluate the effect of inhibition of D2 dopamine receptors on several functions of monocytes of peripheral blood in rat under food restrictionMaterials & methodsIn this experimental study, 36 male Wistar rats (weighing 200-250 gr) were allocated into six groups (n=6), including control groups, food restriction (25%), food restriction (50%), food restriction (75%), food restriction 75% and Sulpiride and rats treated with Sulpiride. Sulpiride was injected Intracerebroventricular at a concentration of 50 μg / rat on day 21 after the study initiation. At the end, the Rats were bled and peripheral blood mononuclear cells were isolated by ficoll gradient method.ResultsFood restriction caused a significant decrease in the activity of monocyte cells of gradient of peripheral blood mononuclear cells like neutral red uptake test and respiratory burst (NBT reduction test) simultaneously with decreasing lymphocytes proliferation after stimulation with phytohemagglutinin. Administration of Sulpiride with a 75% Food restriction resulted in the improvement of these functions of monocyte cells of gradient of peripheral blood mononuclear cells as well as lymphocyte proliferation.ConclusionIntracerebroventricular administration of dopamine D2 receptor antagonists (sulpiride) effectively inhibited the effects of a severe dietary restriction on the suppression of immunity system.Keywords: Dopamine, Peripheral blood mononuclear cells, Food restriction, Sulpiride, Rat

Pages 1157-1166Background & ObjectiveAcute myeloid leukemia (AML) is the third most prevalent type of cancer in Iran and fifth in the world, which includes 8% of the whole type of cancers. The Janus kinase and transcription activator (JAK-STAT) pathway plays pivotal roles in the regulation of cell proliferation, differentiation, migration and apoptosis, which is closely related with the development of some hematological diseases. The JAK-STAT is an important signaling pathway which abnormal inactivation of that is involved in leukemia development. One the important gene in this signaling pathway is JAK2 and the purpose of this study was to evaluate the JAK2 gene expression in AML patients and compare it with control group.Materials & methodsIn this case-control study, blood sample from 30 AML patients admitted to Mirzakochak Khan Jangli Hospiyal, Tehran in 1395 and 20 healthy individual as control were collected. RNA was extracted and after cDNA synthesis, the expression of JAK2 gene was evaluated using Real time PCR (ΔΔCT computational). Statistical analysis was performed software Graphpad prism 6, using t-test and P<0.05 was considered significant.ResultsThe results showed there was a statistically significant association between increasing in JAK2 gene expression in patients compared to the control group (P=0.0001). Pages 1167-1176Background & ObjectiveThis study is intended to evaluate the effect of One Session of resistance exercises on expression of BDNF gene and TrkB receptor in Alzheimer model male Wistar rats.Materials & Methods32 mature male Wistar rats with mean weight of 230 to 280 grams were chosen and divided into Alzheimer and Sham groups. Rats in Sham group received normal saline while rats in Alzheimer group received STZ via intraventricular injection. These rats were then divided into the following four subgroups: 1. Resting Sham, 2. Exercising Sham, 3. Resting Alzheimer, and 4. Exercising Alzheimer. The two exercising rat subgroups, exercised 3 times a week for a period of 8 weeks. Exercise included lifting weight from the ladder. At the end of 8th week and 24 hours after last exercise session, the rats scarified by head separation. Hippocampus tissue was precisely extracted and samples were sent to laboratory for molecular and cellular tests. To determine the gene expression, RT-PCR method was used for analyzing the data and ANOVA was used.ResultsThe amount of BDNF, and TrkB between exercising rats and resting rats.
