    سن شکارگر همه چیزخوارRambur Macrolophus pygmaeus در برنامه های مدیریت تلفیقی آفات، به ویژه آفات گلخانه ای جایگاه ویژه ای پیدا کرده است. در این تحقیق، به منظور بررسی امکان پرورش آن، برخی از فراسنجه های زیستی این شکارگر با تغذیه از کیست آرتمیا (Gunther Artemia urmiana) و تخم شب پره ی آرد (kuehniella Zeller Ephestia) به عنوان غذای استاندارد و غلاف لوبیاسبز L. Phaseoulus vulgaris به عنوان بستر تخم گذاری و تامین رطوبت، در دمای 1±25 درجه ی سلسیوس، رطوبت نسبی 5±65 درصد و دوره ی نوری 8 :16 ساعت تاریکی به روشنایی، با 60 تکرار بررسی شدند. نتایج نشان داد که مدت زمان نشو و نمای مرحله ی پورگی با تغذیه از تخم شب پره ی آرد و کیست آرتمیا برای حشرات نر به ترتیب 11/0±04/17 و 18/0±06/18 روز و حشرات ماده به ترتیب 09/0±31/17 و 15/0±17/18 روز، با تفاوت معنی دار بود. بررسی فراسنجه های تولیدمثلی، نرخ خالص باروری، نرخ ذاتی تولیدمثل و نرخ متناهی افزایش جمعیت در تغذیه از هر دو غذا در سطح احتمال پنج درصد تفاوت معنی دار نشان دادند. این مطالعه نشان داد که نرخ ذاتی افزایش جمعیت ( ) این شکارگر به ترتیب 005/0±080/0 و 006/0±064/0 روز1–، نرخ خالص تولیدمثل ( ) به ترتیب 97/2±12/18 و 43/2±70/10 تخم به ازای هر فرد و مدت زمان تکمیل یک نسل ( ) به ترتیب 33/0±93/35 و 53/0±66/36 روز بوده است. براساس نتایج به دست آمده، استفاده از تخم شب پره ی آرد جهت پرورش سن شکارگر مناسب تر می باشد ولی در صورت عدم دسترسی به تخم شب پره ی آرد تداوم و نگهداری جمعیت این سن شکارگر با کیست آرتمیا نیز امکان پذیر می باشد.

    شانکر باکتریایی درختان میوه ی هسته دار یکی از مهم ترین بیماری های هسته داران در جهان می باشد. در سال‏های اخیر گسترش علایم شانکر همراه با ترشح صمغ و بلاست شکوفه در مناطقی از استان آذربایجان شرقی مشاهده گردیده است. از بافت های آلوده ی جوانه، شکوفه، شاخه و تنه ی درختان باغات زردآلو بر اساس آزمون های ریخت‏شناسی، فیزیولوژیکی و بیوشیمیایی گروه LOPAT و GATTa، جدایه های فلورسنت Pseudomonas yringae pv. syringae (Pss) جداسازی و شناسایی شدند. جدایه ها ترجیح میزبانی نداشته و با درجات مختلف پرآزاری روی سرشاخه های سبز بریده‏ی زردآلو و غلاف های لوبیاسبز بیماری زایی نشان دادند. در تمامی جدایه ها قطعه 725 جفت بازی ژن syrB دخیل در سنتز زهرابه سیرینگومایسین ردیابی گردید و جدایه ها با اختلاف معنی داری از رشد رویشی پرگنه‏ی قارچ Geotrichum candidum با تولید زهرابه ممانعت کردند. با استفاده از آغازگرهای اختصاصی طراحی شده INAF/INAR بخشی از ناحیه ی ژنی inaZ تکثیر شد و فنوتیپ فعالیت هسته‏ی یخ با درجات مختلف در کلیه‏ی جدایه ها مشاهده گردید. با تعیین توالی بخشی از ناحیه 16S rRNA و ژن های rpoD و inaZ تنوع ژنتیکی در میان جدایه ها با رسم تبارنما بر اساس روش بایزین مشاهده شد. نتایج این پژوهش نشان داد که در میان جدایه های Pss عامل شانکر زردآلو هم از لحاظ فنوتیپ بیماری زایی، سنتز زهرابه و تشکیل هسته‏ی یخ و هم از لحاظ ژنتیکی تنوع وجود دارد.

    ویروسNucleopolyhedrosis virus به خاطر مزیت های فراوان مانند اثر اختصاصی روی لارو حشرات آفت، عدم تاثیر نامطلوب روی محیط زیست و جانداران غیرهدف و همچنین سهولت ترکیب و تلفیق با سایر روش های کنترلی، جایگاه ویژه ای در مدیریت کنترل آفات دارد. اولین قدم به عنوان پایه و اساس تحقیق روی این ویروس، بررسی قدرت بیمارگری آن درکنترل آفات مورد هدف است. در این تحقیق قدرت بیمارگری ویروس روی لارو سن دوم کرم قوزه ی پنبه،Helicoverpa armigera Hübner و شب پره ی پشت الماسی،Plutella xylostella L. در دمای 2± 27 درجه ی سلسیوس و رطوبت نسبی 5±65% و دوره ی نوری 16:8 (تاریکی:روشنایی) بررسی شد. غلظت کشنده ی 50 درصد (LC50) Nucleopolyhedrosis virus روی لارو سن دوم کرم قوزه ی پنبه Helicoverpa armigera OB ml-1103×2/9 و شب پره ی پشت الماسی Plutella xylastella OB ml-1104×8/3 محاسبه شد، که بیمارگری ویروس با کمترین غلظت کشنده ی 50 درصد روی کرم قوزه ی پنبه بهتربود. زمان لازم برای مرگ و میر 50 درصد (LT50) در غلظت OB ml-1106 روی لارو سن دوم،H. armigera پنج روز و در جمعیت شب پره ی پشت الماسی، P. xylostella8/4 روز تعیین گردید که تقریبا با هم مطابقت دارند. نتایج این تحقیق نشان داد که Nucleopolyhedro virus دارای قدرت بیمارگری بیشتری روی کرم قوزه ی پنبه نسبت به شب پره ی پشت الماسی است. به نظر می رسد اگرچه این ویروس روی میزبان اصلی قدرت بیمارگری بیشتری دارد ولی نتایج این مطالعه نشان داد که می تواند بعنوان عامل کنترل شب پره ی پشت الماسی نیز بکار گرفته شود.

    پیچیدگی برگ یک بیماری مهم در هلو و شلیل است.کاشت ارقام مقاوم، یکی از بهترین روش ها جهت پیشگیری و کنترل این بیماری می باشد. در بررسی حاضرحساسیت 17 رقم هلو وشلیل به قارچ عامل بیماری پیچیدگی برگ هلو بررسی گردید. نتایج ارزیابی ها در سال اول (1394) نشان داد که بیشترین میزان شدت بیماری مربوط به ارقام کال دسی2000 و جولای آلبرتا بود که بر اساس تیپ واکنش ارقام در گروه حساس (S) قرار گرفت. سپس ارقام بی بی گلد، رد اسکین، اسپرینگ تایم و آنتونی بیشترین میزان شدت بیماری را داشتند که همگی بر اساس تیپ واکنش ارقام، نیمه مقاوم (MR) طبقه بندی شدند. در مقابل، ارقام آمسدن، ردهاون، اسپرینگ کرست و فایت به ترتیب کمترین میزان شدت بیماری را داشته و همگی بسیار مقاوم (HR) طبقه بندی شدند. در سال دوم ارزیابی (1395)، ارقام اسپرینگ تایم و کال دسی 2000 بیشترین میزان شدت بیماری را داشته و بر اساس تیپ واکنش ارقام هر دو بسیار حساس به بیماری (HS) طبقه بندی شدند. ارقام جولای آلبرتا و بی بی گلد در رتبه ی بعدی قرار داشتند که هر دو حساس به بیماری (S) شناخته شدند. در مقابل، ارقام آمسدن، ردهاون و روبین کمترین میزان شدت بیماری را داشته و بر اساس تیپ واکنش ارقام بسیار مقاوم(HR) بودند. بنابراین با توجه به نتایج دو سال ارزیابی می توان گفت که ارقام کال دسی2000، اسپرینگ تایم و سپس جولای آلبرتا و بی بی گلد نسبت به سایر ارقام مورد بررسی بیشترین حساسیت و ارقام آمسدن و ردهاون بیشترین مقاومت را نسبت به بیماری پیچیدگی برگ هلو دارند.

    شب پره ی مدیترانه ای آرد، Anagasta kuehniella (Zeller) (Lep.; Pyralidae) با تغذیه از آرد سبب کاهش کیفیت نان تولیدی می شود. ایجاد اختلال در دستگاه گوارش حشرات با استفاده از مهارکننده های آنزیم های گوارشی که از گیاهان تراژن استخراج می شوند، یکی از روش های کنترل محسوب می شود. در این مطالعه، اثر عصاره ها ی پروتئینی گندم ارقام سیوند، سرداری و تریتیکاله روی فعالیت آلفا- آمیلازهای لاروهای سنین چهارم و پنجم شب پره ی مدیترانه ای آرد بررسی شد. برای استخراج پروتئین از بذرها، آمونیوم سولفات 70 درصد مورد استفاده قرار گرفت. اسیدیته و دمای بهینه ی فعالیت آلفا- آمیلاز به ترتیب 10 و 40 درجه ی سانتی گراد به دست آمد. طبق نتایج حاصل، بالاترین غلظت عصاره های پروتئینی ارقام گندم سیوند و سرداری (13میکروگرم پروتئین بر میلی لیتر) و تریتیکاله (16 میکروگرم پروتئین بر میلی لیتر) فعالیت آنزیم را به ترتیب 08/73، 07/71 و 88/65 درصد در لاروهای سن پنجم و 16/58، 14/42 و 25/50 درصد در لاروهای سن چهارم کاهش دادند. در پایین ترین غلظت عصاره های پروتئینی گندم ارقام سیوند و سرداری (812/0 میکروگرم پروتئین بر میلی لیتر) و تریتیکاله (یک میکروگرم پروتئین بر میلی لیتر) نیز فعالیت آنزیم را به ترتیب93/19، 8/17 و 26/25 درصد در لارو سن پنجم و 78/11، 81/12 و 09/10 درصد در لارو سن چهارم کاهش دادند. در ژل زایموگرام بدون استفاده از مهارکننده تنها یک باند مشاهده شد. در بالاترین غلظت عصاره ها، در لاروهای سن پنجم، باند آمیلازی ناپدید شد و در لاروهای سن چهارم، وضوح باند کم تر شد. با کاهش دز مهارکننده ها، وضوح باندها افزایش یافت. طبق نتایج به دست آمده، عصاره های پروتئینی ارقام سیوند، سرداری و تریتیکاله پتانسیل خوبی برای کاهش فعالیت آمیلازی در لارو آفت مورد نظر داشته و می تواند در برنامه های مدیریت این آفت در آینده مورد استفاده قرار گیرد.

    در این تحقیق تاثیر قارچ های Trichoderma harzianum و T. atrovirideعلیه P. lingamدر شرایط آزمایشگاه و گلخانه بررسی شد. سنجش آزمایشگاهی به صورت آزمون های کشت متقابل و تولید ترکیبات فرار انجام گردید. برای سنجش گلخانه‎ای نیز از دو روش تیمار بذر با آنتاگونیست و مایه‎زنی اندام هوایی گیاهچه‎های کلزا با سوسپانسون اسپوری عوامل آنتاگونیست استفاده شد. آزمایش در قالب دو طرح‎ کاملا تصادفی جداگانه با چهار تکرار انجام شد. درصد مرگ‎و‎میر گیاهچه‎ها و شدت آلودگی حاصل تعیین گردید. نتایج نشان داد که هر دو گونه قارچ، توان آنتاگونیستی بالایی در برابر بیمارگر دارا بودند. در آزمون کشت متقابل، جدایه‎ها اثر آنتاگونیستی بالایی بر رشد میسلیوم P. lingam نشان دادند. دو جدایه‎ی T. harzianum و T. atrovirideرشد قارچ را به ترتیب 6/58 و 4/71 درصد مهار کردند. درصد بازدارندگی از رشد دو جدایه، در آزمون ترکیبات فرار 120 و 240 ساعت پس از کشت، به ترتیب 21-15 و 39-31 درصد تعیین شد. در آزمون گلخانه‎ای هر دو گونه به طور معنی‎داری (01/0P<) شدت بیماری را نسبت به شاهد کاهش دادند. نتایج آزمایش‎های گلخانه‎ای نشان داد که در روش تیمار بذر، جدایه‏های آنتاگونیست به ترتیب 5/42 و 47 درصد بیماری را کاهش دادند. در تیمار اندام‏های هوایی با جدایه‎های T. harzianum و T. atroviride، شدت بیماری به ترتیب 13/63 و 79/78 درصد کاهش یافت. بنابراین هر دو گونه پتانسیل قابل توجهی برای مهار زیستی بیماری ساق سیاه کلزا دارند ولی در مجموع گونه T. atrovirideموثرتر از گونه دیگر بر علیه این بیمارگر عمل کرد.

    نماتدهای مولد گره ریشه (Meloidogyne spp.) از مهمترین نماتدهای انگل گیاهی است که سبب ایجاد خسارت روی اکثر محصولات کشاورزی می شوند. در این تحقیق، به بررسی اثر قارچ مایکوریز آربوسکولار Funneliformis mosseae در کنترل نماتد ریشه ی گرهی خیار، M. incognita پرداخته شد. بدین منظور آزمونی در قالب طرح کاملا تصادفی با چهار تیمار و چهار تکرار انجام شد. نتایج نشان داد که اضافه کردن قارچ مایکوریز به خاک یک ماه قبل از تلقیح نماتد، موجب کاهش معنی دار فاکتورهای بیماریزایی نماتد (تعداد گال، تعداد کیسه ی تخم، تعداد تخم در هر کیسه ی تخم و تعداد لارو سن دوم) در هر دو رقم متحمل و حساس گردید. علی رغم اینکه پارامترهای بیماریزایی نماتد در رقم متحمل در مقایسه با رقم حساس، کمتر بود اما تلقیح ریشه های حساس با قارچ مایکوریز در رقم حساس، موجب کاهش قابل توجه عوامل بیماریزایی نماتد شد. عامل تولیدمثل در گیاهان مایه کوبی شده با قارچ مایکوریز در دو رقم متحمل و حساس به ترتیب 50 و 6/66 درصد کاهش نشان داد. حضور قارچ مایکوریز قبل از تلقیح نماتد در رقم متحمل و حساس به ترتیب سبب کاهش 64 و 3/63 درصدی جمعیت لارو سن دوم در خاک گردید. همچنین مقایسه میانگین تعداد و اندازه ی سلول های غول آسا، تعداد و قطر هسته و مساحت سلول غول آسا در دو رقم، نشان دهنده ی اختلاف معنی دار آماری بین گیاهان تلقیح شده با قارچ مایکوریز و گیاهان غیرمایکوریزی بود. با توجه به اینکه قارچ مایکوریز باعث کاهش بیماریزایی نماتد ریشه ی گرهی و افزایش مقاومت رقم حساس نسبت به نماتد می گردد، می تواند به عنوان گزینه ای در جهت مهار زیستی نماتد ریشه ی گرهی مورد توجه قرار گیرد.

    Macrolophus pygmaeus Rambur is an most omnivorous predator which has considered in integrated pest management programs, especially for greenhouse pest management. In this study, biological properties and life table statistics of M. pygmaeus by feeding on Artemia urmiana Gunther cysts and Ephestia kuehniella Zeller eggs were studied under laboratory conditions (25±1°C, 65±5 % relative humidity and 16:8h L: D photoperiod) with 60 replications. The pods of green bean Phaseoulus vulgaris L. served as oviposition substrate and the moisture source. Developmental time of nymphal stages which fed by of E. kuehniella eggs and Artemia cysts were 17.04±0.11, 18.06±0.18 d for males and 17.31±0.09, 18.17±0.15 d for females respectively and were significantly different as well as intrinsic rate of increase (rm) was 0.080±0.005 and 0.064±0.006 d–1 respectively. Also, net reproductive rate (R0) was 18.12±2.97 and 10.70±2.43 eggs/individual on those food regims respectively. Furthermore, mean generation time (T) was estimated as 35.93±0.33 and 36.66±0.53 d respectively. Meanwhile, the reproductive parameters on flour moth larvae have been significantly higher than Artemia cyst. The results showed that feeding on E. kuehniella reduces nymphal period, while increases productivity and longevity of adults. According to the present findings, E. kuehniella is more suitable prey for rearing of this predator, but in absence of access to E. kuehniella, rearing on Artemia cysts is also possible.

    Bacterial canker disease of stone fruit trees is one of the most important diseases in the world. In recent years, increasing of stone fruits canker disease symptoms as well as oozing and blast has been observed in some area in East Azerbaijan province of Iran. Fluorescent Pseudomonas syringae pv. syringae (Pss) strains were isolated from apricot trees which showed infected bud, bloom, branch and stem. The isolated strains were characterized based on their morphological, physiological and biochemical properties especially LOPAT and GATTa tests. Host preference was not observed among the tested strains and they showed pathogenicity on apricot green-cut twigs and green been pods with different disease severity. A 725-bp fragment of the syrB gene which is required for synthesis of syringomycin was amplified using specific primers in all strains. The tested strains inhibited the Geotrichum candidum mycelial growth with significant differences by toxin production. Using specific designed INAF/INAR primers, the inaZ gene was partially amplified and different range of ice nucleation activity was detected in all Pss strains. Constructing dendrograms using Bayesian inference showed genetic diversity among strains based on partial sequencing of 16S rRNA, rpoD and inaZ genes. These results showed that there are diversity among the causal agent strains of apricot canker disease based on both pathogenicity, toxin production and ice nucleation activity phenotypes and genetics.

    Nucleopolyhedro virus (NPV) has an important role in pest management programs. Because of many advantages such as selective effect on larvae of insect pest, no undesirable effects on environment and non-target organisms and also compatibility with other pest management tactics. In this research, the virulence of a native isolate was evaluated on the larvae of cotton bollworm, Helicoverpa armigera Hübner and diamond back moth, Plutella xylostella at 27±2 oC, 65±5% RH and 16L: 8D photoperiod. Based on the results, 50% lethal concentration (LC50) of virus on the 2nd larval instars of H. armigera and P. xylostella were determined as 9.2×103 OB/ ml -1and 3.8×104 OB/ml -1, respectively. Also results (LC50 – values), showed that NPV was more virulent on H. armigera than on the P. xylostella. Values obtained for 50% lethal time (LT50) of NPV were almost similar 5 and 4.8 days post treatment for H. armigera and P. xylostella, respectively.

    Leaf curl is a serious disease on peaches and nectarines. Planting of resistant varieties is one of the best ways to prevent and control this disease. In this research, 17 peach and nectarine cultivars were evaluated for susceptibility to the causative agent of peach leaf curl. The results of the evaluations in the first year indicated that, the highest disease severity was related to the Caldesi 2000 and July Alberta cultivars, based on the reaction type of cultivars, they were placed in the sensitive group (S). Then, Red Skin, Baby Gold, Springtime and Antonia cultivars had the highest disease severity and all of them were classified as semi-resistant (MR) cultivars. In contrast, Amesdan, Redhaven, Springcrest and Fayette respectively had the lowest severity of the disease, and all were highly resistant (HR). In the second year of evaluation, Springtime and Caldesi2000 cultivars had the highest rate of disease severity and according to the reaction type of cultivars, both of them classified, highly susceptible (HS). Then July Alberta and Baby Gold cultivars were susceptible to disease (S). In contrast, Amesdan, Redhaven and Robine cultivars had the lowest severity of disease and according to the type of cultivars reaction were classified highly resistant (HR). Therefore, according to the results of two years evaluation, it can be said that the cultivars Caldesi2000, Springcrest,then July Alberta and Baby Gold are the most susceptible and Amesdan and Redhaven are the highest resistance to the peach leaf curl disease.

    Anagasta kuehniella Zeller (Lepidoptera: Pyralidae) causes the quality loss of the bread by feeding on flour. Disruption in insect’s digestive enzymes through transgenic plants, expressing enzyme inhibitors, is a methods of pest control. So, the aim of the current study was to investigate the inhibitory effects of proteinaceous extracts of two varieties of wheat (Sivand, Sardari) and Triticale against activity of digestive α-amylase of fourth (L4) and fifth (L5) instars larvae of A.kuehniella. Ammonium sulfate (70%) was used to extract proteins from seeds. The optimal temperature and pH of α-amylase activity was found 40°C and 10, respectively. The results showed that the highest dose of proteinaceous extracts of Sivand and Sardari (13 µg ml-1 protein) and Triticale (16 µg ml-1 protein) reduced the enzyme activity, 73.08%, 71.07% and 65.88% of L5 and 58.16%, 42.14% and 50.25% of L4, respectively. The lowest dose of proteinaceous extracts of Sivand and Sardari (0.812 µg ml-1 protein) and Triticale (1 µg ml-1 protein) inhibited the enzyme activity, 19.93%, 17.8% and 25.26% of L5 and 11.78%, 12.81% and 10.09% of L4, respectively. In the gel electrophoresis of enzyme without usage of inhibitors, one band was observed. At the highest dose of extracts, in the L5, amylase band disappeared and in the L4, waned band clearly. Band resolution was increased by reducing inhibitor dose. It was concluded that Sivand, Sardari and Triticale proteinaceous extracts can be used for management of A.kuehniella.

    In this research, the effect of Trichoderma harzianum and T. atroviride against P. lingam was investigated in the laboratory and greenhouse conditions. Laboratory tests were followed as dual cultures and testing of the fungal volatile compounds. In the greenhouse, the antagonistic effects of the isolates were tested through treating rapeseed seeds and rapeseed above-ground parts with the spore suspension of the isolates in 106 spore per ml. The experiments were conducted in complete randomized design with four replications. Percent of plant mortality and disease severity were assessed. Both isolates showed antagonistic activities against the fungal phytopathogen. The two isolates T. harzianum and T. atroviride could inhibit the growth of the pathogen by 58.6 and 71.4 respectively in dual culture. In volatile compound phase, percent of growth inhibition were 15-21% and 31-39% in 120 and 240 hours after culture. In greenhouse experiments, antagonistic species could significantly (P<0.01) reduce disease severity compared to control. The greenhouse results revealed that in the seed treatment experiment, Trichoderma isolates decreased the rapeseed blackleg to 42.5 and 47.5℅, respectively after two weeks. Treating the above-ground parts with isolates T. harzianum and T. atroviride were significantly decreased the disease rate to 63.13 and 78.79℅, respectively. So both Trichoderma species can biologically inhibit canola black leg, but T. atroviride was for the first time more effective than another one.

    Root knot nematodes (Meloidogyne spp.) are one of the most important plant parasitic nematodes that cause damage to most agricultural crops. In this study, the effect of the arbuscular mycorrhizal fungus (AMF) Funneliformis mosseae on root knot nematode M. incognita on two cucumber cultivars; tolerant and susceptible was investigated. This experiment was conducted in a completely randomized design with four treatments and four replications. The results showed that adding mycorrhizal fungus one month before inoculation of nematode caused a significant decrease in nematode pathogenicity factors (number of galls, number of eggs, eggs per egg mass and number of second stage juveniles) in both tolerant and susceptible cultivars (cv). Although the pathogenicity of the nematode in tolerant cultivars was lower than that of susceptible cvs, inoculation of susceptible roots with mycorrhizal fungus significantly reduced the pathogenicity of the nematode. The reproductive factor decreased in mycorrhizal plants in tolerant and susceptible cultivars in 50 and 66.6%, respectively. The presence of mycorrhizal in tolerant and sensitive cultivar reduced 64 and 63.3% of the second larval population in the soil, respectively. Also, the comparison of the mean number, size and area of giant cells and the number and diameter of the nucleus in 2 cvs showed a significant difference between the plants with or without mycorrhizal fungus. Since mycorrhizal fungus reduces the pathogenicity of the root knot nematode and increases the resistance of the cv susceptible to nematode, it can be considered as an option for biologic control of the root knot nematode.
