    سابقه و هدف
    در سال های اخیر دریافت کمک زیست یارها و زیست یارها به علت اثرات مفید بر سلامتی به طور گسترده ای تایید شده است. کمک زیست یارها کربوهیدرات های غیرقابل هضمی هستند که رشد/ و یا فعالیت باکتری های مفید روده بزرگ را افزایش داده و موجب بهبود سلامتی می شوند. علاوه بر این، افزودن زیست یارها به مواد غذایی، به علت اثر ایمن سازی و تولید اسیدهای آلی و سایر ترکیباتی که جذب مواد مغذی و سلامت کلی دستگاه گوارش را افزایش می دهند، رو به افزایش است. یافته ها و نتیجه گیری: راهبرد زیست فناورانه برای تولید کمک زیست یارها و خالص سازی آن ها به منظور تامین نیاز به مواد اولیه برای فرموله کردن غذاهای فراسودمند پیشنهاد شده است. انواع رشد مایه ها و روش های تخمیر، خالص سازی و تعیین ویژگی های آنها بررسی شده اند. گذشته از این، ما مزایای زیست یارها، روش های جداسازی و تعیین و ارزیابی ویژگی های آنها که موجب استفاده از آنها در صنعت غذا شده است را نشان خواهیم داد. با پیشرفت های فناورانه انتقال کمک زیست یارها و زیست یارها به مواد غذایی به منظور تامین نیاز مصرف کننده، با هزینه پایین و داشتن ویژگی های حسی مطلوب و اثرات مفید سلامتی، امکان پذیر شده است.

    سابقه و هدف
    صنعت فرآوری حیوانات در 28 کشور اتحادیه اروپا، شامل کشتارگاه ها، شرکت های تبدیل مواد مازاد (ذوب چربی) ، و سایرین، حجم زیادی از پساب حاوی حدود 500،000 تن چربی به همراه مقادیر قابل توجهی ضایعات لاشه و پودر استخوان و گوشت تولید می کنند. این مواد باید با روش هایی مانند زیست دگرگونی زیست بسپارهای پلی هیدروکسی آلکانوآت (PHA) با ترکیب های مولکولی متنوع و ویژگی های شبه پلاستیک ارزش بیشتری پیدا کنند. نوآوری مقاله حاضر این است که برای اولین بار پژوهش های پیشین و فعلی در خصوص تولید PHA از پساب های حیوانی خلاصه و انتخاب گونه های میکروبی تولیدکننده مناسب، فرآیندهای بالا دستی مواد خام برای تولید منابع کربنی در دسترس، سینتیک زیست فرآیند، تعیین ویژگی های زیست بسپارها با ساختار مولکولی دگرگون شده، ارزیابی فرآیند محیطی، و امکان سنجی اقتصادی بیان شده است. یافته ها و نتیجه گیری: مطالعات مقایسه موردی به وضوح نشان می دهند استفاده از ضایعات فرآیند حیوانی به عنوان نسل دوم مواد اولیه برای تولید زیست بسپارها به دلیل استفاده از گونه های میکروبی بهینه، خوراک دهی مناسب، کوتاه بودن مسیر انتقال، و طرح های کسب و کار شفاف برای تجاری سازی محصول نهایی می تواند یقینا می تواند فرآیندی مناسب و با ارزش اقتصادی باشد. بیشتر از همه، استفاده از پسماند حیوانی برای تولید زیست بسپارها و سوخت های زیستی نسل دوم، با حفظ ارزش تغذیه ای مواد اولیه، در ایمنی مواد غذایی می تواند نقش داشته باشد.

    سابقه و اهداف
    آنزیم بتا-گالاکتوزیداز موجب آبکافت لاکتوز به گلوکز و گالاکتوز و تولید فرآورده های شیری بدون لاکتوز می شود. میزان چربی و یون های گوناگون در شیر می توانند به عنوان مهار کننده یا فعال کننده آنزیم های بتا-گالاکتوزیداز فعالیت کنند. در مطالعه قبلی ژن آنزیم بتا-گالاکتوزیداز فعال در سرما (BGalP) حاصل از پلانوکوکوس sp-L4، در Pichiapastoris بیان داده شد تا لاکتوز شیر ضمن نگهداری یخچالی آبکافت شود. در این مطالعه، برای تایید ظرفیت این آنزیم در آبکافت لاکتوز شیر، اثر ترکیبات عمده موجود در شیر بر سینتیک آنزیمی بررسی شده است.
    مواد و روش ها
    فعالیت آنزیم BGalP در غلظت های گوناگون از یون های سدیم کلرید (NaCl) ، پتاسیم کلرید (KCl) ، منیزیم کلرید (MgCl2) و کلسیم کلرید (CaCl2) و در شیر حاوی مقادیر کم، متوسط و زیاد چربی تعیین شد. در این آزمون ها، اورتو-نیترفنیل بتا- گالاکتوزید به عنوان رشد مایه مورد استفاده قرارر گرفت. همچنین، بعد از 24 ساعت گرمخانه گذاری در دمای اتاق، میزان گلوکز تولید شده، به عنوان فرآورده به دست آمده، اندازه گیری شد.

    سابقه و هدف
    اخیرا، علاقه ای رو به رشد در زمینه مواد غذایی تولید شده بدون افزودن نگهدارنده های شیمیایی وجود دارد. باکتری های لاکتیک اسید توانایی زیادی در کنترل رشد ریزاندامگان های ناخواسته دارند. در این کار، فعالیت ضدقارچی هشت سویه لاکتوباسیل جدا شده از منایع گیاهی و مواد غذایی (سس تارتار، آرد گندم، آرد جو، خمیر ترش) در برابر فوزاریوم کالموروم DMF301 و پنی سیلیوم اکسپانسوم بررسی شد.
    مواد و روش ها
    فعالیت ضدقارچی سلول های زنده لاکتوباسیل و مایع رویی حرارت دیده آنها با روش انتشار در آگار در درجه حرارت 30 درجه سلسیوس تعیین و هاله رشد اندازه گیری شد. تولید اسید آلی سویه های مورد آزمون با HPLC بررسی شد. یافته ها و نتیجه گیری: سلول های زنده سه سویه لاکتوباسیلوس پلانتاتاروم بیشترین فعالیت ضدقارچی را از خود نشان دادند. فوزاریوم کلاموروم DMF301 به فعالیت لاکتوباسیل ها بیشتر حساس بود. مایع رویی باکتری های حرارت دیده (100 درجه سانتی گراد به مدت 10 دقیقه) نیز مورد آزمون قرار گرفتند، و به میزان 10 یا 20 درصد حجمی حجمی به محیط کشت اضافه شدند؛ رشد فوزاریوم کلاموروم DMF301 به میزان 80 تا 90 درصد در غلظت 10 درصد مایع رویی و به طور کامل در غلظت 20 درصد آن مهار شد. هرچند پس از خنثی سازی، فقط مایع رویی حرارت دیده لاکتوباسیلوس پلانتاروم CCDM 583 فعالیت ضدقارچی نسبتا موثری داشت. اثر مهاری مشترک سویه های لاکتوباسیل فعالیت ضدقارچی آنها را کاهش داد. در فعالیت سویه های لاکتوباسیلوس پلانتاروم CCDM 583 و MP2 به تنهایی یا در ترکیب با هم در برابر فوزاریوم کلاموروم DMF301 تفاوت های آماری معنی داری یافت شد. برای استفاده در ترکیب با سایر کشت ها، لازم است سازگاری سویه های گوناگون لاکتوباسیل مورد تایید قرار گیرد.

    سابقه و هدف
    اسپیرولینا پلاتنسیس یکی از منابع عمده ترکیبات فراسودمند با خواص غذا دارویی است. این محصول بسیار فسادپذیر است و لازم است بلافاصله پس از برداشت فرآوری شود. بنابراین، هدف اصلی مطالعه حاضر بررسی اثر شرایط متفاوت فرآیند بر مهمترین ویژگی های کیفی اسپیرولینا پلاتنسیس بود.
    مواد و روش ها
    اسپیرولینا پلاتنسیس تازه با روش های گوناگون (خشک کردن در سایه، خشک کردن در آفتاب، خشک کردن در گرمخانه، خشک کردن در مایکروویو، خشک کردن درگرمخانه تحت خلا، خشک کردن انجمادی، خشک کردن پاششی و انجماد با و بدون آنزیم بری) تحت فرآوری قرار گرفت و تغییرات ویژگی های کیفی آن (مواد معدنی، ترکیب اسیدهای چرب، میزان کل ترکیبات فنولیک و فعالیت ضداکسایشی) نمونه ها بررسی شد. یافته ها و نتیجه گیری: شرایط فرآوری اثر معنی داری بر ویژگی های کیفی نمونه ها داشت. نمونه خشک شده در گرمخانه تحت خلا بیشترین فعالیت ضداکسایشی و بالاترین محتوی ترکیبات فنولیک را داشت، چرا که انجام فرایندهای شیمیایی تخریبی وابسته به اکسیژن و واکنش های قهوه ای شدن آنزیمی در این روش کمتر است. میزان مواد معدنی در نمونه های خشک شده تفاوت معنی دار آماری باهم نداشتند، در حالی که میزان سدیم، پتاسیم، منگنز، منیزیم، کلسیم و فسفر نمونه های منجمد به طور معنی داری کاهش یافته بود. اسیدهای چرب ضروری غیراشباع گوناگون مانند آلفا لینولنیک اسید، گاما لینولنیک اسید، آراشیدونیک اسید، ایکوزاپنتانوئیک اسید و دوکوزاهگزانوئیک اسید در اسپیرولینا شناسایی شدند. براین اساس، خشک کردن پاششی و خشک کردن انجمادی، بهترین روش های فرآوری برای حفظ اسیدهای چرب غیر اشباع بودند، و از نظر حفظ ترکیبات فنولیک کل و ویژگی های ضد اکشایشی اسپیرولینا پلاتنسیس، روش خشک کردن در گرمخانه تحت خلا ارجح بود.

    سابقه و هدف
    گاما-آمینوبوتیریک اسید آمینواسیدی غیرپروتئینی تولید شده توسط باکتری های لاکتیک اسید در مواد غذایی تخمیر شده می باشد و عملکرد منحصر به فردی در سامانه زیستی انسان دارد. هدف این مطالعه بهینه سازی محیط کشت برای تولید گاما-آمینوبوتیریک اسید در زیست یارهای جدا شده از فرآورده های شیری بومی غرب ایران با استفاده از دو محیط کشت MRS broth و پروتئین آب پنیر بوده است.
    مواد و روش ها
    تولید بالقوه گاما-آمینوبوتیریک اسید توسط لاکتوباسیل پاراکازئی، لاکتوباسیلوس پلانتاروم و پدیوکوکوس اسیدی لاکتیس که به ترتیب از دوغ، ماست و پنیر جدا شده بودند با استفاده از محیط های کشت MRS broth و پروتئین آب پنیر و کروماتوگرافی مایع با کارایی بالا ارزیابی شد. به منظور افزایش تولید گاما-آمینوبوتیریک اسید، محیط های کشت در pH (4-6) ، درجه حرارت (30-50°C) ، مدت زمان (72-12 ساعت) و غلظت گلوتامیک اسید (250-25 میلی مول) بهینه شدند. یافته ها و نتیجه گیری: نتایج نشان داد که در شرایط درجه حرارت کشت37°C ، مدت زمان گرمخانه گذاری 60 ساعت، pH برابر با 5 در MRS brothحاوی50 میلی مول گلوتامیک اسید، لاکتوباسیلوس پلانتاروم جدا شده از دوغ بیشترین تولید گاما- آمینوبوتیریک اسید (24/115 میلی گرم در کیلوگرم) را داشت. پس از بهینه سازی محیط کشت لاکتوباسیلوس پلانتاروم تولید گاما-آمینوبوتیریک اسید تا170/492 افزایش یافت. شرایط بهینه شامل غلظت گلوتامیک اسید 250 میلی مول، درجه حرارت °C27/37، 19/5pH و مدت زمان گرمخانه گذاری 72 ساعت بود. بر اساس نتایج به دست آمده، کاربرد جدایه های فرآورده های شیری بومی غرب ایران و بهینه سازی شرایط رشد توانایی تولید گاما-آمینوبوتیریک اسید را افزایش داد.

    Background and objective
    Prebiotics and probiotics intake have been widely recognized in past recent years due to possessing multiple health benefits. Prebiotics are non-digestible carbohydrates that promote the growth and/or activity of beneficial bacteria in the colon which improves the health. Moreover, the incorporation of probiotics in food has also been a growing practice due to its immunomodulatory effect, the production of organic acids and other compounds that promotes the absorption of nutrients and the general health of the digestive system. Results and conclusion: Biotechnological strategies have been proposed for prebiotic production and purification in order to meet the demand to be included as ingredients in functional food formulation. Different aspects related to the substrates and different fermentation systems for their production as well as the purification and characterization processes are addressed. Also, we will present the benefits promoted by probiotics, the methods of isolation and characterization, as well as the evaluation of these attributes, so that they can be used in the food industry. With the technological developments in prebiotics and probiotics, it will be possible to deliver foods that respond to consumer demand with low cost and with pleasant sensory characteristics as well as providing beneficial health effects.     Background and objective
    Animal processing industry in the EU-28 states, encompassing slaughterhouses, rendering companies, and others, generates high quantities of waste streams containing about 500,000 t of lipids plus considerable amounts of offal material and meat and bone meal. These materials need to be utilized in a value-creating way, such as via bioconversion towards polyhydroxyalkanoate biopolyesters of diverse molecular composition and various plastic-like features. As a novelty, the present article summarizes for the first time previous and current efforts to utilize these animal-based waste streams for polyhydroxyalkanoate production in terms of selection of suitable microbial production strains, upstream processing of the raw material to generate accessible carbon sources, kinetics of the bioprocess, characterization of the produced biopolyesters of diverse molecular architecture, environmental process assessment, and economic feasibility. Results and conclusion: The compared case studies clearly demonstrate that utilization of animal processing waste as a second generation feedstock for biopolyester production can definitely become an economically viable and sustainable process provided the utilization of optimized microbial strains, tailored feeding regime, short transportation distances, and clear business plans for commercialization of the final products. Most of all, using animal based waste for generation of second generation biopolyesters and second generation biofuel contributes to food security by preserving raw materials of nutritional value.     Background and objective
    β-galactosidase enzymes hydrolyze lactose into glucose and galactose for production of lactose free dairy products. However, different ions and fat content in milk may act as the inhibitor or activator for β-galactosidase enzymes. A cold-active β-galactosidase enzyme (BGalP), originally from Planococcus sp. L4, was previously expressed in Pichia pastoris to perform lactose hydrolysis in the refrigerated milk. In this study, the effects of milks major ingredients were evaluated on the enzymatic kinetics to confirm its capacity for hydrolyzing milk lactose. Material and methods: The activity was determined in different concentrations of NaCl, KCl, MgCl2, and CaCl2 as well as in the milk with low, medium or high-fat content. In these experiments ortho-Nitrophenyl β-galactoside was used as the substrate. Additionally, glucose was measured as the product after incubation of milk with BGalP enzyme for 24 h at room temperature. Results and conclusion: This study demonstrated that ions and fat content did not adversely affect the enzyme activity in the concentration corresponding to the milk contents. Ca (27.5-32.5 mM), Cl (25.3-30.9 mM), Na (15.2-39.1 mM) and Mg (3.75-5.83 mM) had no inhibitory effects, but KCl decreased the enzyme activity. Since Cl existed in MgCl2, and CaCl2 exerted no inhibitory effects, it can be concluded that inhibitory effects of KCl resulted from potassium rather than chloride. The results indicate that BGalP enzyme was not inhibited by milks major ingredients and has the potential to be used for the production of lactose-free dairy products.

    Background and objective
    Presently, there is a growing interest in food produced without the addition of chemical preservatives. Lactic acid bacteria have a high potential to control the growth of undesirable microorganisms. In this work, the antifungal activity of eight strains of lactobacilli isolated from plant sources and foodstuffs (tartar sauce, wheat flour, barley flour, sourdough) were tested against Fusarium culmorum DMF301 and Penicillium expansum DMF04.
    Materials and methods
    Antifungal activity of live cells of lactobacilli and their heat-treated supernatants was determined by agar diffusion method at 30°C, the radial growth of fungi was measured. Organic acid production of tested strains was examined by HPLC. Results and conclusion: Live cells of three Lactobacillus plantarum strains showed the highest antifungal activities. Fusarium culmorum DMF301 was more sensitive to the activity of lactobacilli. Heat-treated bacterial supernatants (100°C, 10 min) were also tested, being added at 10 or 20% v v-1 to the culture medium; growth of Fusarium culmorum DMF301was inhibited by range of 80-90% compared to controls at 10% supernatant concentration and fully at 20%. However, after neutralization, only the heat treated supernatant from Lactobacillus plantarum CCDM 583 had partially effective antifungal activity. The mutual inhibition of lactobacilli strains reduced their antifungal activity. Statistically significant differences in activity against Fusarium culmorum DMF 301 were found using individual strains of Lactobacillus plantarum CCDM 583 and MP2 or their combination. For use in combination with other cultures, it is therefore necessary to verify the compatibility of various strains of lactobacilli.

    Background and objective
    A Spirulina platensis is one of the major sources of functional food ingredients with nutraceutical properties. It is a very perishable and should be processed immediately after harvesting. Therefore, the main purpose of the present study was investigation the effect of different processing condition on the most important qualitative features of Spirulina platensis. Material and methods: Fresh Spirulina platensis was processed (shade, sun, oven, microwave, vacuum oven, freeze and spray-drying and freezing with and without blanching) and changes in its qualitative characteristics (minerals and fatty acids composition, total phenolic compounds and antioxidant activity) of samples were analyzed. Results and conclusion: Processing conditions significantly (p≤0.05) affected the qualitative properties of the sample. The vacuum-oven dried sample had the highest level of total phenolic compounds and antioxidant activity because of the lower possibility of oxygen dependent degradation and enzymatic browning reactions. The mineral was not significantly different (p>0.05) in dried samples, while Na, K, Mg, Mn, Ca and P content of the frozen samples were reduced significantly. Various unsaturated essential fatty acids like α-linolenic acid, γ-linolenic acid, arachidonic acid, eicosapentaenoic acid, and docosahexaenoic acid were detected in Spirulina. In this regards, spray and freeze-drying were the best processing methods in protecting UFA, and vacuum oven-drying was preferred in protecting total phenolic compounds and antioxidant activity of the Spirulina platensis.

    Background and objective
    Gamma-aminobutyric acid is a non-protein amino acid produced by lactic acid bacteria in fermented foods and includes unique functions in the human biological system. The aim of this study was optimization of culture media for gamma-aminobutyric acid production in probiotics extracted from local dairy products in west of Iran using two culture media of MRS broth and whey protein. Material and methods: The potential of gamma-aminobutyric acid production was assessed in Lactobacillus paracasei, Lactobacillus plantarum and Pediococos acidilactici, respectively extracted from doogh, yogurt and cheese using MRS broth and whey protein media and high performance liquid chromatography. To increase gamma-aminobutyric acid production, these media were optimized as pH (4-6), temperature (30-50°C), time (12-72 h) and glutamic acid concentration (25-250 mM). Results and conclusion: Results have shown that Lactobacillus plantarum extracted from doogh includes the highest potential of gamma-aminobutyric acid production (115.24 mg kg-1) under the following conditions of a culture temperature of 37°C, incubation time 60 h at pH 5 in MRS broth containing 50 mM of glutamic acid. After optimization of Lactobacillus plantarum media, gamma-aminobutyric acid production increased to 170.492 mg kg-1. The optimum conditions included a glutamic acid concentration of 250 mM, culture temperature at 37.27°C, pH=5.19 and an incubation time of 72 h. Based on the results, use of local isolated dairy products in west region of Iran and optimization of growth conditions increased the ability of gamma-aminobutyric acid production.
