    در پژوهش حاضر، ابتدا تاثیر افزودن عسل و عرق نعناع در سطوح 0، 1، 2 و 3 درصد به نوشیدنی تخمیری فراسودمند بر پایه سورگوم جوانهزده ی تهیه شده توسط باکتری های پروبیوتیک لاکتوباسیلوس اسیدوفیلوس و لاکتوباسیلوس پلانتاروم با نسبت 50:50، بر ویژگی های حسی نظیر طعم، بو، پس طعم، رنگ و پذیرش کلی بررسی شد. سپس بهترین نمونه انتخاب شده، از نظر ویژگی های کیفی نظیر pH،اسیدیته، بریکس، میزان فنول کل، میزان فعالیت ضداکسایشی، خصوصیات حسی و زندهمانی باکتری های پروبیوتیک در طول پانزده روز نگهداری در فاصله زمانی 5 روز، در دمای یخچال با نمونه شاهد (فاقد افزودنی) مورد مقایسه قرار گرفت. مطابق نتایج حاصل از ارزیابی حسی، نمونه حاوی 3 درصد عسل و عرق نعناع به عنوان بهترین نمونه انتخاب شد. نتایج بررسی ویژگی های کیفی، حسی و زنده مانی این نمونه، در طول پانزده روز نگهداری در دمای یخچال، نتایج نشان داد که افزودن عسل و عرق نعناع سبب افزایش معنی داری (05/0p) در ویژگی های کیفی، حسی و زنده مانی باکتری های پروبیوتیک در نوشیدنی تخمیریانتخاب شده می گردد.

    فرایند ریزپوشانی پروبیوتیک ها در نمونه های غذایی به عنوان یک فن نوین برای افزایش زنده مانیو قابلیت تحمل پروبیوتیک های پوشش دهی شده در شرایط دستگاه گوارش ثابت شده است. با این حال تحقیقات در زمینه ترکیبات بهینه ریزپوشانی به منظور افزایش بقای باکتری های پروبیوتیک در مواد غذایی و شرایط شبیه سازی معده و روده ادامه دارد. در این پژوهش با ایجاد ریزپوشینه متشکل از ماتریکس کنسانتره پروتئین آب پنیر، کربوکسی متیل سلولز، پکتین، اینولین و صمغ فارسی، به منظور افزایش زنده مانی باکتری لاکتوباسیلوس اسیدفیلوس[1]و باسیلوس کوآگولانس[2] در شرایط معده (پپسین، 5/1pH)و روده (پانکراتین و نمک های صفراوی، 8pH)، به ترتیب راندمان ریزپوشانی 50/76 و 55/84 درصد حاصل گردید. ریزپوشینه ها در شرایط شبیه سازی شده در بازه های زمانی صفر،10، 30، 60 و90 minدر دمای°C 37 قرار گرفتند. نتایج آنالیز واریانس و مقایسه میانگین نشان داد که اختلاف معنی داریدر شرایط شبیه سازی معده (05/0P) در کاهش سیکل لگاریتمی جمعیت لاکتوباسیلوس اسیدفیلوس ریزپوشانی شده نسبت بهآزاد (نسبت 3 به 8 Log Cfu/ml) و در باسیلوس کوآگولانس (نسبت 3 به 7 Log Cfu/ml) حاصل گردید در حالی که در شرایط بازی روده نسبت کاهش لگاریتمی جمعیت باکتری های ریزپوشانی شده بهسلول هایآزاد آن در سویه های مذکور به ترتیب نسبت 3 به 9 و 3 به 8 Log Cfu/mlبود.[1]Lactobacillus acidophilus[2]Lactobacillus coagulans

    هدف از این مطالعه، بررسی امکان استفاده از پسماندهایمختلف کشاورزی و صنایع وابسته به منظور تولید بهینه آنزیم آلفاآمیلاز توسطسویه بومی باسیلوس سوبتلیس[1] T41aو انتخاب اجزاء مناسب و اقتصادی به منظور دست یابی به بیشینه بازدهی تولید این آنزیم بوده است. استفاده از پسماندهایکشاورزی و صنایع وابسته در تولید فرآورده های میکروبی به عنوان منابع تامین کننده کربن و نیتروژن موردنیاز در محیط کشت، ضمن ایجاد ارزش افزوده بالا، موجب کاهش هزینه های تولید و نیز کاهش معضل زیست محیطی ناشی از انباشت پسماندها در محیطمی شود. در مطالعه حاضر، در گام نخست و به منظور انتخاب هدفمند پسماندهایکشاورزی، تاثیر منابع متخلف کربن و نیتروژن بر تولید آلفا آمیلاز ارزیابی شد. بر اساس نتایج ثبت شده لاکتوزو پپتون سویا به ترتیب به عنوان مناسب ترین منابع کربن و نیتروژن بیشینه تولید آنزیم را باعث شدند. در گام بعدی تاثیر انواع پسماندهای کشاورزی شامل پسماند مالت، سبوس برنج و سبوس گندم، پودر آب پنیر، ملاس، کنجاله سویا، کنجاله کلزا، کنجاله ذرت، شربت خیسانده ذرت، ویناس الکل و ضایعات گلوتن به میزان 1% در محیط کشت پایه بر تولید آلفا آمیلاز در سامانه تخمیر غوطه ور با سوبسترای معلقبررسی و مناسب ترین ترکیب پسماندها برای طراحی محیط کشت اقتصادی و کارا انتخاب شد، به طوری که با جایگزینی منابع کربنی و نیتروژنی محیط کشت پایه با 3 درصد از پسماندهای کشاورزی منتخب سه گانه متشکل از 1 درصد از ضایعاتحاصل از فرایند مالت گیری، 1 درصد کنجاله کلزا و 1 درصد از ویناس الکل، میزان تولید آنزیم به 7/1 برابر تولید در محیط کشت پایه حاوی ترکیبات گران قیمت رسید. تولید آلفا آمیلاز در محیط کشت منتخب نهایی به U/L4113 معادل72 درصد افزایش تولید نسبت به محیط شاهد رسید، ضمن این که با حذف منابع کربن و نیتروژن سنتزی، هزینه تولید کاهش یافت.     در این تحقیق جدایه 3Aاز مخمر رودوترولا موسیلاجینوزا[1] برای کنترل کپک آبی سیب پنیسیلیوم اکسپنسیوم[2] به کار گرفته شد. کپک آبی سیب از بافت آلوده سیب جداسازی شد. آزمون تست تقابل، متابولیت های خارج سلولی و مواد فرار در آزمایشگاه برای ارزیابی قدرت بیوکنترلی مخمرها به کار گرفته شد. مخمر آنتاگونیست توانست رشد قارچ را در آزمایشگاه کاهش دهد. میزان کاهش رشد عامل کپک آبی سیب، در آزمون کشت تقابل 38/53 درصد، در مورد گاز فرار41/84 درصد، و در تست متابولیت های خارج سلولی 16/86 درصد بود. نمک کلرید کلسیم با دو درصد حجمی به طور معنی داری از رشد عامل بیمارگر جلوگیری کرد در حالی که اثر سوئی بر روی تعداد کلنی های مخمری در محیط PDBنداشت. غلظت Cell/ml107 از مخمررودوترولا موسیلاجینوزا 3Aبر روی جوانه زنی و همچنین طول لوله جوانه زنی اسپور عامل بیمارگر پنیسیلیوم اکسپنسیومدر آزمایشگاه اثر کنترلی گذاشت، و همچنین از ظهور بیماری و توسعه بیماری نیز جلوگیری کرد. ارتباط معنی داری بین غلظت مخمر به کار رفته و کنترل قارچ بیمارگر وجود داشت. با اضافه کردن کلرید کلسیم به مخمر نرخ جوانه زنی اسپورها و همچنین طول لوله جوانه زنی اسپورها در آزمایشگاه، و متعاقبا ظهور بیماری و قطر لکه ها در مقایسه با تیمار مخمر تنها کاهش پیدا کرد. زمانی که سوسپانسیون سلول های مخمری به غلظت Cell/ml108 رسید، رشد بیمارگر به طور کلی در آزمایشگاه متوقف شد، و هیچ آلودگی ای در سیب های تیمار شده با یا بدون نمک کلرید کلسیم یافت نشد، علاوه بر این نمک کلرید کلسیم قادر به افزایش فعالیت آنزیم های آنتی اکسیدان مانند پراکسیداز و پلی فنل اکسیداز شد، که این افزایش در مورد کاربرد مخمر آنتاگونیست بر روی سیب های آلوده نیز اتفاق افتاد، ولی مقدار القای مقاومتی که به وسیله نمک کلرید کلسیم اتفاق افتاد بسیار بیشتر از مخمر به تنهایی بود.[1]Rhodotorula mucilaginosa[2]Penicillium expansum

    جداسازی و شناسایی باکتری های پروبیوتیک ازسویه های بومی، نقش بسیار مهمی در صنعت غذایی و همچنین افزایش سلامت افراد جامعه دارد. با توجه به اهمیت و سودمندی لاکتوباسیلوسهابه عنوان باکتری های پروبیوتیک و آغازگر در صنایع غذایی مخصوصا لبنی، بر آن شدیم تا با جداسازی وشناسایی سویه های لاکتوباسیلوس بومی از محصولات لبنی منطقه غرب ایران گامی درجهت شناسایی پروبیوتیکهای مطلوب ورشد آن ها برداریمهدف ازاین مطالعه، شناسایی و بررسی خواص پروبیوتیکی و فنوتیبی باکتری های اسیدلاکتیک بومی جدا شده ازمحصولات لبنی منطقه غرب ایران بود.جهت شناسایی اولیه آزمون های تشخیصی شامل رنگ آمیزیگرم، کاتالاز و آزمون اکسیدازروی50 نمونه ازباکتری های اسیدلاکتیک جدا شده از محصولات لبنی بومی (ماست، دوغ، پنیروکره) منطقه غرب ایران انجام شد و سپس روی نمونه های منتخب آزمون های مربوط به بررسی خواص پروبیوتیکی شاملمقاومت به اسید، صفرا، شیره معده، عدم فعالیتهمولیتیک و هیدرولیزال آرژنین و آزمونشناسایی بهوسیله ی پرایمر هایعمومیSrDNA16همراه با تعیین توالی ورسم درخت فیلوژنی صورت پذیرفت.نتایج آزمون های تشخیصی نشان داد در میان 50 گونه از باکتری های اسیدلاکتیک مورد بررسی 3 سویه گرم مثبت، کاتالاز منفیو اکسیدازمنفی بودند. مطابق با نتایج خواص پروبیوتیکی هر3 سویه منتخب دارای خواص پروبیوتیکی مطلوبی بودند. نتایجواکنش زنجیرهای پلیمراز SrDNA16 نشان داد 3 باکتری مورد بررسیمتعلق بهگونه هایجدید لاکتوباسیل کازئیلاکتوباسیلوس پلانتاروم و پدیوکوکوساسیدلاکتیک بودند.

    آمین های بیوژنیک ترکیبات مهم برای عملکردهای فیزیولوژیکی و متابولیکی در تمام ارگانیسم های زنده هستند که در اثر متابولیسم برخی از اسیدهای آمینه و واکنش دکربوکسیلاسیون در طول تخمیر و یا فساد مواد غذایی تولید می شوند.کنترل تجمع آمین ها در فراورده های تخمیری که بخش اعظم آمین های بیوژن در مواد غذایی را هیستامین تشکیل می دهد یکی از چالش های حاضر در صنعت غذا می باشد. محدود نمودن رشد میکروبی و فعالیت دکربوکسیلازی میکروارگانیسم ها با استفاده از سویه هایی که فاقد آنزیم آمینواسید دکربوکسیلاز بوده و یا سویه های اکسیدکننده آمین ها در کشت های آغازگر از روش های کنترل تشکیل آن ها می باشند. به منظور افزایش کنترل مواد غذایی و احتمالا برای کمک به روشن شدن نقش هیستامین در رژیم غذایی برای سلامت انسان، امروزه از روش های سریع و تحلیلی آسان و مقرون به صرفه مانند روش الایزا برای تعیین کمی آمین های بیوژنیک استفاده می شود. لذا در این پژوهش اثر دوسویه باسیلوس سرئوسzk2و باسیلوس تورنجینسیسILBB224جداشده از لجن لبنی به تنهایی و همراه با کشت آغازگر پنیر در کاهش هیستامین به دو صورت کیفی و کمی موردبررسی قرار گرفت. بررسی کیفی از طریق آزمایش دکربوکسیلاسیون سویه های باکتریایی در محیط لیزین دکربوکسیلاز براثانجام پذیرفت. نتایج حاکی از آن بود که سویه ها فاقد آنزیم دکربوکسیلاز بوده و قادر به شکستن گروه کربوکسیل از اسیدآمینه نمی باشند. با مصرف قند و تولید اسید، رنگ محیط کشت را به رنگ زرد تغییر دادند. همچنین، تاثیر سویه های باسیلوس سرئوسzk2باسیلوستورنجینسیسILBB224و کشت آغازگر پنیر بر کاهش غلظت های ppm150، ppm125،ppm100،ppm75ppm50هیستامین محیط با استفاده از آزمون الایزا انجام شد. نتایج نشان داد که با افزایش غلظت هیستامین، درصد کاهش هیستامین توسط سویه های مختلف کاهش یافت. همچنین، مشخص شد بیشترین میزان کاهش هیستامین در هنگام استفاده از مخلوط سویه های باسیلوسبه همراه آغازگر پنیر در غلظت ppm50 هیستامین بود. بنابراین، بر اساس یافته های این پژوهش می توان از این سویه ها به عنوان کشتی با پتانسیل مناسب جهت کاهش آمین های بیوژنیک در فراورده های تخمیری مختلف استفاده نمود.

    In the current study the effect of honey and mint distillate (0, 1, 2 and 3%) addition to fermented beverage based on germinated sorghum by probiotic bacteria (Lactobacillus plantarum and Lactobacillus acidophilus in ratio of 50:50) on sensory attributes was first evaluated. Then, the selected sample was evaluated for quality characteristics including pH, acidity, total phenol content, antioxidant activity and probiotic bacteria viability at 5 days interval during 15 days storage at 4ºC. The sensory evaluation results indicated that the beverage contains 3% of honey and mint distillate shows higher sensory scores among other samples. Furthermore, the results obtained from the selected sample and control (without additives) evaluation during 15 days storage show that addition of honey and mint distillate increased significantly (p≤0.05) quality and sensory characteristics and bacteria viability of the selected fermented beverage.

    Encapsulation of probiotics in food samples has been proven as a novel technique for enhancing the viability and stability of encapsulated probiotics in simulated gastrointestinal environment. However, investigations on the optimized compositions of encapsulation to enhance the viability of the probiotic bacteria in food and simulated gastrointestinal environment are intending to increase. In the present study, by creating a matrix composed of whey protein concentrate, Carboxymethylcellulose, Pectin, Inulin and Persian gum, in order to increase the viability of Lactobacillus acidophilus and Bacillus coagulants in stomach environmental condition (pepsin, pH= 1.5) and intestine (pancreatin and bile salts, pH =8), the encapsulation efficiency of 76.58 and 84.55%, was obtained, respectively. The encapsulated probiotics were subjected to the simulated environment at 0, 10, 30, 60 and 90 min intervals at 37 °C.     In this study the use of suitable agro-industrial residues for higher and economical production of alpha amylase from Bacillus subtilis T41a was investigated. Using agricultural waste as a substrate rich in protein and carbohydrate for microbial fermentation resulted in value added production and reduction in production cost as well. At first step, the effect of different carbon and nitrogen sources on enzyme production was studied. The results showed that lactose and soy peptone were the best carbon and nitrogen sources for maximum enzyme production. Next, alpha amylase production has been investigated by solid suspended fermentation using agroindustrial residues including rice bran, wheat bran, brewer's spent grain, molasses, soy meal, canola meal, corn meal, corn liquor steep, vinasse, gluten, and whey powder at 1%concentration in production culture medium. Based on the experimental results, the optimum composition of agricultural wastes was selected and substituted the carbon and nitrogen sources of basal medium, accordingly in 3% concentration of selected agricultural waste that was composed of 1 percent brewer's spent grain, 1 percent canola meal and 1 percent vinasse, the enzyme production increased 1.7 times in comparison with control medium. The synthetic medium was successfully substituted by agricultural waste and enzyme production increased to 4113 U/L. Optimization of culture medium caused 72 percent enhancement in enzyme production in comparison to control sample and decreased the production cost.

    The antagonistic yeast, Rhodotorula mucilaginosa A3 was evaluated in order to control the blue mold of apple caused by Penicillium expansum. Dual culture, cell free metabolite and volatile test were used in vitro assay. Yeast strains of two genus inhibited growth of P.expansum, inhibition was 53.38 %, in dual culture, 84.41% in volatile metabolite and 86.16% in cell free metabolite test. Calcium chloride (2% w/v) significantly inhibited the growth of the pathogen Penicillium expansum, but did not affect the colony-forming units (CFU) of yeast Rhodotorula mucilaginosa A3 in potato dextrose broth. The concentration 107 cell/mL of yeast suspension influenced spore germination and germ tube growth of Penicillium expansum in vitro, as well as disease incidence and lesion development in fruits. There were significant negative relationships between the suspension concentrations of the yeasts and the growth as well as infectivity of the pathogen. The addition of calcium resulted in lower spore germination rates and slower growth of germ tubes in vitro, as well as in lower disease incidences and smaller lesion diameters compared with treatments with yeast antagonists alone. When yeast cell suspensions reached a concentration of 108 CFU mL-1, growth of the pathogen was completely limited in vitro, and no infection was found in apple fruits treated with or without calcium. In addition, the calcium chloride salt could increase the activity of antioxidant enzymes such as peroxidase and polyphenol oxidase, Calcium chloride salt was induced more than of the application of yeast alone.

    Isolation and identification of probiotic bacteria from indigenous strains plays important role in the food industry and increases the health of the community. Considering the importance and usefulness of lactobacill as probiotic in the food industry, especially dairy products thus we decided to isolate and identify the native Lactobacillus strains from dairy products in the western part of Iran. The aim of this study was to identify probiotic in phenotypic properties of native lactic acid bacteria isolated from dairy products in western Iran. For initial identification of diagnostic tests including positive gram, catalase and oxidation test. From 50 samples of lactic acid bacteria isolated from native dairy products (yogurt, dough, butter and cheese) in west of Iran were tested probiotic properties including acid resistance, bile, gastric juice, hemolytic inactivation, hydrogel l-arginine inactivation, and identification by primer of 16SrDNA with sequencing of phylogeny tree. The results of the diagnostic tests showed that among the 50 species of lactic acid bacteria, 3 strains of gram positive, negative catalase and oxidizing agents were studied. According to the results of properties, all 3 selected strains had favorable probiotic properties. The results of 16SrDNA polymerase chain showed that 3 bacteria were Lactobacillus casei, Lactobacillus plantarum, and Pediococcus acidilactici.

    Biogenic amines are important compounds for physiological and metabolic functions in all living organisms that are produced by the metabolism of some amino acids and decarboxylation reactions during fermentation or food spoilage. Control of amine accumulation in fermentation products is one of the challenges in the food industry. Limiting microbial growth and decarboxylase activity of microorganisms by strains that lack of the enzyme amino acid decarboxylase or the oxidative strains of amines in starter cultures are their control methods. Therefore, in this study, the effect of Bacillus cereus strain zk2 and Bacillus thuringiensis strain ILBB224 isolated from dairy sludge alone and with cheese starter culture was investigated in both quantitative and qualitative reduction of histamine. A qualitative study was carried out through decarboxylation testing of bacterial strains in Lysine decarboxylase broth. The results indicated that strains lacked decarboxylation properties; thus, they were able to degrade histamine and reduce it, and by changing sugar and acid production, the color of the culture medium changed to yellow. Also, the effects of strains of Bacillus cereus strain zk2, Bacillus thuringiensis strain ILBB224 with starter culture of cheese on reduction of 50, 75, 100, 125 and 150 ppm of histamine were performed by ELISA. The results showed that with increasing histamine concentration, the reduction in histamine content was reduced by different strains. It was also found that the highest reduction in histamine was observed when using Bacillus strains with cheese starter at a concentration of 50 ppm histamine. Therefore, based on the findings of this study, these strains can be used as culture with the potential to reduce biogenic amines in different fermentation products.
