    دوکفه ای Anodonta cygnea به دلیل مقاومت پایین در برابر تغییرات محیطی به عنوان گونه ای مناسب در ارزیابی آلودگی اکوسیستم های آبی مطرح است. امروزه به دلیل کاربرد گسترده نانومواد در صنایع مختلف، نگرانی های زیادی در خصوص اثرات منفی این مواد بر حیات سیستم های آبی وجود دارد. در مطالعه حاضر به ارزیابی اثر نانوذرات اکسید مس (II) (CuO-NPs) بر بیان ژن HSP70 در A. cygnea پرداخته شد. بدین منظور، دوکفه ای ها طی 12 روز در معرض CuO-NPs با غلظت های 0 (شاهد)، 25/0، 5/2 و ppm 25 قرار گرفتند. برای ارزیابی تغییرات بیان ژن HSP70، ابتدا توالی این ژن در گونه مورد نظر تعیین شد. در این راستا، یک قطعه 540 نوکلئوتیدی کد کننده HSP70 کلون شد. توالی نوکلئوتیدی HSP70 در A. cygnea به ترتیب دارای 8/19، 7/30، 7/26 و 9/22 درصد از نوکلئوتیدهای A، C، G و T بود. نتایج حاکی از آن بود که مواجهه با CuO-NPs منجر به القای بیان ژن HSP70 در هر دو بافت پا و آبشش شد. اما اختلاف مشاهده شده بین سطح بیان ژن HSP70 در این دو بافت معنی دار نبود (05/0P>). همچنین سطح بیان ژن در بافت های مورد بررسی، با افزایش زمان مواجهه تا 8 روز افزایش یافت اما با گذشت زمان تا روز دوازدهم روند کاهشی نشان داد (05/0≥P). در مجموع، ژن HSP70 در دوکفه ای A. cygnea دارای میزان بالاتری از نوکلئوتیدهای GC در مقایسه با سایر گونه های مورد بررسی است و سطح بیان این ژن تحت مواجهه با CuO-NPs افزایش می یابد، به نحوی که میزان این افزایش وابسته به غلظت و زمان مواجهه است.

    با توجه به پتانسیل بالای جلبک های قهوه ای در توسعه تولیدات مواد غذایی و کاربردهایی در صنعت برای انسان، دام و آبزیان، این پژوهش به تعیین خواص فیزیکوشیمیایی، پروفیل اسید چرب و ارزیابی حسی ماکروجلبک های Iyengaria stellata پرداخته است. برای ارزیابی خواص فیزیکوشیمیایی پودر جلبک، ظرفیت نگهداری آب، ظرفیت تورم و ظرفیت جذب روغن مورد بررسی قرار گرفت. پروفیل اسید چرب به روش کروماتوگرافی گازی و ارزیابی حسی با استفاده از پنل آموزش ندیده انجام شد. با افزایش دما ظرفیت تورم به صورت معنی داری کاهش یافت (05/0P<) اما ظرفیت نگهداری آب با افزایش دما کاهش معنی داری را نشان نداد (05/0P>). ظرفیت تورم و ظرفیت نگهداری آب در جلبک I. stellataپایین بود، در حالی که این جلبک از ظرفیت جذب روغن بالایی برخوردار بود. ماکروجلبک I. stellata به ترتیب حاوی %73/38 اسید چرب اشباع (SFAs)، %86/42 اسید چرب غیراشباع با یک پیوند دوگانه (MUFAs) و %03/5 اسید چرب غیراشباع با چند پیوند دوگانه (PUFAs) بود. نتایج ارزیابی حسی نشان داد که ماکروجلبک مورد مطالعه از طعم و بوی قابل قبولی برخوردار بود. طبق نتایج این مطالعه، ماکروجلبک‏ I. stellata را می توان به عنوان منبع بالقوه از اسیدهای چرب ضروری و همچنین به عنوان افزودنی در صنایع غذایی مورد استفاده قرار داد.

    در مطالعه حاضر، ابتدا با استفاده از آنزیم آلکالاز از ماهی کیلکای معمولی (Clupeonella cultriventris) پروتئین هیدرولیز شده (FPH) به دست آمد. سپس فعالیت آنتی اکسیدانی پروتئین هیدرولیز شده ماهی به وسیله آزمون های قدرت مهارکنندگی رادیکال های آزاد DPPH و ABTS، قدرت کاهندگی آهن، فعالیت شلاته کنندگی یون آهن و فعالیت آنتی اکسیدانی کل در 5 غلظت متفاوت (1، 2، 3، 4 و 5 میلی گرم در میلی لیتر)، مورد ارزیابی قرارگرفت. نتایج بیان کننده آن بود که با افزایش غلظت پروتئین، خاصیت آنتی اکسیدانی FPH به طور معنی داری افزایش یافت. بالاترین فعالیت مهارکنندگی رادیکال DPPH، قدرت کاهندگی آهن، شلاته کنندگی یون آهن و فعالیت آنتی اکسیدانی کل مربوط به غلظت 5 میلی گرم بر میلی لیتر بود. در مجموع می توان گفت هیدرولیز آنزیمی ماهی کیلکا منجر به تولید پروتئین هایی با خواص آنتی اکسیدانی می شود که می توانند بعد از تایید مطالعات کلینیکی به عنوان افزودنی مواد غذایی مورد استفاده قرار گیرند.

    هدف از مطالعه حاضر، تعیین سمیت حاد یا غلطت کشندگی (50LC 96 ساعته) عصاره هیدروالکلی اکالیپتوس در بچه ماهیان آمور Ctenopharyngodon idella و بررسی تاثیر این عصاره بر روی باکتری Aeromonas hydrophila بوده است. 180 قطعه بچه ماهی آمور با میانگین وزنی 5 گرم به صورت تصادفی در 5 گروه تیمار با غلظت های 2000، 2300، 2600، 3000 و ppm3500 عصاره هیدروالکلی اکالیپتوس و یک گروه شاهد تقسیم شدند (هر گروه با سه تکرار). محدوده کشندگی مقادیر عصاره هیدروالکلی اکالیپتوس در مدت زمان 96 ساعت به روش لگاریتمی برابر ppm56/2478 تعیین شد. مطالعات آسیب شناسی بافت های آبشش و کبد بچه ماهیان آمور که در معرض غلظت تحت کشنده عصاره هیدروالکلی اکالیپتوس قرار داشتند نشان دهنده بیشترین ضایعات شامل پرخونی و خونریزی بود. برای تعیین حداقل غلظت مهار کنندگی رشد (MIC) از روش رقت لوله ای و برای تعیین حداقل غلظت کشندگی ماده ضد میکروبی ( MBC) از محیط کشت استفاده شد. نتایج حاصل مشخص کرد که عصاره هیدروالکلی اکالیپتوس دارای حداقل غلظت مهارکنندگی از رشد (MIC) mg/mL 25/0 و حداقل غلظت کشندگی ( MBC) mg/mL 5/0 بود. در مجموع، عصاره هیدرو الکلی اکالیپتوس از رشد باکتری A. hydrophila در محیط کشت جلوگیری کرد و با توجه به تاثیرات سوء آنتی بیوتیک ها می تواند به عنوان جایگزین آن ها در کنترل و نابودی باکتری A. hydrophila مورد استفاده قرار گیرد.

    در پژوهش حاضر، ناپلی آرتمیا با غلظت های مختلف (2/0 و 4/0 گرم در لیتر) عصاره الکلی برگ گیاه حرا (Avicennia marina) غنی سازی و همراه با غذای پایه کنسانتره در تغذیه پست لارو میگوی سفید غربی (میانگین طول کل 05/0±2/0 سانتی متر و میانگین وزن 20/0±10/10 میلی گرم) به مدت 30 روز استفاده شد. عملکرد رشد شامل میزان طول کل، وزن و ضریب رشد ویژه در روزهای 1، 15 و 30 و درصد بازماندگی در انتهای دوره پرورش تعیین شد. آزمایش استرس شوری و استرس فرمالین نیز در روزهای 15 و 30 دوره پرورش انجام شد. نتایج نشان داد استفاده از عصاره برگ حرا در غلظت های 2/0 و 4/0 گرم در لیتر بر عملکرد رشد میگوها نسبت به تیمار شاهد تاثیر مثبت داشت. میگوهای تغذیه شده با آرتمیای غنی شده بطور معناداری از بازماندگی بیشتری برخوردار بودند. تحمل استرس شوری در میگوهای تغذیه شده با آرتمیای غنی شده بیشتر از تیمار شاهد بود. در تست فرمالین نیز بازماندگی میگوهای تغذیه شده با عصاره برگ حرا نسبت به تیمار شاهد بیشتر بود، هر چند اختلاف معنی دار نبود. در مجموع، استفاده از آرتمیای غنی شده با عصاره برگ حرا در مرحله لاروی میگوی سفید غربی می تواند موجب بهبود کیفیت پست لاروها شود.

    این مطالعه با هدف بررسی افزودن سطوح متفاوت نانوذرات آهن صفر ظرفیتی به جیره غذایی و تاثیر آن بر رشد و شاخص های خونی بچه ماهی انگشت قد کپور معمولی انجام شد. 150 قطعه ماهی با وزن متوسط 10/0±14/6 گرم به مدت 8 هفته با جیره های حاوی صفر، 50، 100، 150 و mg/Kg200 نانوذره آهن صفر ظرفیتی تغذیه شدند. نتایج نشان می دهد که مقادیر شاخص های وزن نهایی، وزن به دست آمده، طول نهایی، نرخ رشد ویژه، درصد افزایش وزن بدن، درصد رشد روزانه، کارایی غذا و کارایی پروتئین و چربی در تیمار mg/Kg150 نانوذره آهن اختلاف معنی داری را با سایر تیمارها نشان دادند (05/0>P). شاخص کبدی در تیمار صفر دارای بیشترین مقدار بود (05/0>P). بیشترین مقادیر گلبول های قرمز، غلظت هموگلوبین و درصد هماتوکریت در تیمار mg/Kg150 و کمترین آن ها در تیمار صفر مشاهده شد (05/0>P). تعداد گلبول های سفید و شاخص MCV در تیمار mg/Kg150 دارای کمترین مقدار بود (05/0>P)؛ اما مقادیر MCH، MCHC، مونوسیت و ائوزوفیل در بین تیمارها تفاوت معنی داری را در بر نداشت (05/0 کلیدواژگان: آهن صفر ظرفیتی، شاخص های رشد، گلبول های قرمز، جیره غذایی

    Anodonta cygnea is considered as a suitable species in evaluating the pollution of aquatic ecosystems because of the low resistance to changes in environmental conditions. Nowadays, due to the wide application of nanomaterials in different industries, the negative effect of these materials on aquatic life is of great concern. In the present study, the effect of copper oxide nanoparticles (II) (CuO-NPs) on HSP70 gene expression was evaluated in A. cygnea. For this purpose, the mussels were exposed to the concentrations of 0 (control), 0.25, 2.5 and 25ppm of CuO-NPs during 12 days. To analyze the changes in HSP70 gene expression, the sequence of this gene was determined in this species. In this regard, a fragment of 540 nucleotide coding HSP70 was cloned. The nucleotide sequence of HSP70 in A. cygnea contained 19.8, 30.7, 26.7 and 22.9 percent of A, C, G and T, respectively. The results indicated that exposing to CuO-NPs induced HSP70 gene expression in both tissues of foot and gill. Hereof, there was no significant difference (P>0.05) in gene expression level of HSP70 among foot and gill tissues. Also, the gene expression level in the studied tissues increased with increasing exposure time up to 8 days while it showed decrease over the time up to 12th day (P≤0.05). In conclusion, the HSP70 gene in A. cygnea showed higher level of GC nucleotides in comparison to the other studied species and the expression level of this gene increases under exposing to CuO-NPs so that this increase is dependent on the concentration and time.

    Considering the high potential of brown algae in the development of food production and applications in the industry to humans, livestock and aquatics, this study designated to the physicochemical properties, fatty acid profile and sensory evaluation of macroalgae Iyengaria stellata. Water holding capacity, swelling, and oil holding capacity were investigated for evaluation of physicochemical properties of algae powder. The fatty acid profile and sensory profiling were analyzed by gas chromatography and using a non-trained panel, respectively. The swelling capacity was decreased significantly with increasing temperature (P0.05). The capacities of swelling and water holding of the algae I. stellata were down while oil holding capacity was high in this seaweed. I. stellata consisted of 38.73% saturated fatty acids (SFAs), 42.86% monounsaturated fatty acids (MUFAs) and 5.03% polyunsaturated fatty acids (PUFAs). The results of the sensory evaluation showed that taste and smell of the studied macroalgae were acceptable. In conclusion, macroalgae I. stellata can be a source of essential fatty acids as well as was used as additives in the food industry.

    In the present study, fish protein hydrolysates (FPH) prepared by hydrolysis of common Kilka (Clupeonella cultriventris­) using the Alcalase enzyme. The antioxidant activity of FPH was evaluated by DPPH and ABTS radical scavenging abilities, reducing power assay, ferrous ion-chelating and total antioxidant activity tests in five different concentrations (1, 2, 3, 4 and 5 mg/mL). The results indicated that with increasing concentration (5 mg/mL), the antioxidant properties of FPH were significantly increased. The highest DPPH radical scavenging activity, ferrous ion-chelating, ferric reducing antioxidant power and total antioxidant activity was related to the concentration of 5 mg/mL. In conclusion, Kilka fish enzymatic hydrolysis leads to the hydrolysate production with antioxidant properties and can be used as food additives after confirming by clinical studies.

    The purpose of this study was to determine the acute toxicity or 50% lethal concentration within 96 hours (LC50 96 h) of eucalyptus extract on the juvenile grass carp (Ctenopharyngodon idella) and the impact of the extraction on Aeromonas hydrophila. 180 grass carp with an average weight of 5g were distributed randomly in the five treatment groups with eucalyptus concentrations of 2000, 2300, 2600, 3000, 3500 ppm and one control group (with three replications). Lethal concentration of eucalyptus extract using logarithmic was found 2478.56ppm. Histopathological studies of gill and liver that were exposed to lethal concentrations of eucalyptus extracts, showed the lesions consisted of hyperemia and bleeding. The minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of the antimicrobial agent were determined using tube dissolution method. The MIC and MBC of extract of eucalyptus against A. hydrophila were recorded as 0.25mg/mL and 0.5mg/mL, respectively. In conclusion, the extract of eucalyptus can inhibit the growth of A. hydrophila in a culture medium. Due to the adverse effects of antibiotics, they can be used as an alternative to controlling and destroying A. hydrophila.

    In the present study, nauplius of Artemiafranciscana was enriched with different levels (0.2 and 0.4 g/L) of ethanolic leaf extract of grey mangrove Avicennia marina and then was fed by Pacific white shrimp Penaeus vannamei postlarvae (total length average 0.2±0.05 cm and weight average 10.10±0.20 mg) by adding to basal diet for 30 days. Growth performance including total length, weight, and specific growth rate was determined on days 1, 15 and 30 and survival percentage at the end of the period. The tests of salinity and formalin stress resistance were achieved on days 15 and 30 of the culture period. Results showed that the use of extract at concentrations of 0.2 and 0.4g/L had positive effects on shrimp growth in relation to the control. Shrimps fed with enriched Artemia had significantly higher survival than control. Salinity stress resistance in shrimps fed with enriched Artemia was higher than control. In formalin stress test, the survival in shrimps fed with enriched Artemia was higher compared to control, although this was not significantly different. In conclusion, use of Artemia enriched by mangrove leaf extract in the larval stage of Pacific white shrimp could improve the quality of postlarvae.

    The aim of this study was to assess the effects of different doses of zero-valent iron nanoparticles in the diet of common carp fingerling on growth performance and blood parameters. 150 fish with an average initial weight of 6.14±0.10g were fed one of the experimental diets for 8 weeks. The diets contained 0, 50, 100, 150 and 200 mg/Kg zero-valent iron nanoparticles. The results showed that fish were fed by the diet supplemented with 150 mg/Kg zero-valent iron nanoparticles displayed significant difference in final weight, weight gain, final length, specific growth rate, body weight increase, average daily growth, feed efficiency, protein efficiency ratio and lipid efficiency ratio (P0.05). Based on the results, due to the increased surface to volume ratio, 150 mg/Kg zero-valent iron nanoparticles in the diet has more influence than larger scales, it is recommended in the diet of common carp fingerlings.
