Comparing two aerobic training intensity (low intensity and high intensity) on PLIN2 protein expression in skeletal muscle, serum glucose levels and insulin in Streptozotocin-induced diabetic rats

Background And Aims
Lipid metabolism disorder plays a important role in creating insulin resistance in skeletal muscle and It was shown PLIN2 is effective in regulating the accumulation and metabolism Intramuscular lipid. On the other hand, one of the proposed pathways for the effects of exercise training on metabolic diseases is an Intramuscular lipolysis pathway. The purpose of this study was compared the effect of aerobic exercise intensity in PLIN2 protein expression skeletal muscle diabetic rat.
Method
24 male Wistar rats were randomly divided into three groups. Low-intensity endurance training (n=8) and high-intensity endurance training (n=8). The control group (n=8) did not receive any intervention. Diabetes mellitus induced by intraperitoneal injection (55 mg per kg body weight) Streptozotocin, and endurance training was applied three times a week for eight weeks. Low-intensity exercise group were trained to the treadmill by running at a speed of 18 meters per minute (equivalent to 55-65 percent of vo2 max) and high intensity training with a speed of 34 meters per minute(equivalent to 85% Vo2max). The expression of the plin2 protein was analyzed by Western blot technique. To determine the significance of differences between the groups, the results were analyzed using one-way ANOVA and Tukey post-hoc test (α= 0.05).
Results
Direct comparison between the groups by ANOVA show significant differences in PLIN2 (p=0/037). Tukey's post hoc test showed that there was a statistical difference between the mean values of the diabetic control group and high intensity endurance group (P = 0.028). PLIN2 not significantly increased in low-intensity exercise compared to the control group (P=0/28), No significant between-group differences were found for changes in PLIN2 occurring during the 8-week low-intensity and high-intensity aerobic exercise (P=0/38). Also, comparison between groups showed, there was no significant difference between the three groups. The serum levels of glucose and insulin (respectively p=0/09 and p=/028).
Conclusion
The results of the present study showed that the Effects of with high-intensity endurance training increase the expression PLIN2 In diabetes rats.
