The potency of a modified alum precipitated haemorrhagic septicaemia vaccine in mice, rabbit and caule was evaluated. Active mouse protection test showed that the log of protection was 7.8. Passive mouse protection test on sera obtained from vaccinated caule showed 67-100% protection up to four months and 25-67% protection 2 months post vaccination. Modified alum precipitated vaccine was potent to produce immunity against active challenge with virulent Pasteurella mu/tocida in rabbits. The antisera collected from vaccinated rabbits conferred 83-100% protection in serummized micc.

Various cell wall rractions of Pasleurella mu/toeida vaccine strain serotype A: 1 including sonicated antigen, heat stable (HS), Iipopolysaccharide-protein complex (LPS-P). olltermembrane protein (OMP), capsular (CAP) and potassium thiocyanate (KSCN~xtracted proteins were extracted. Their immllnogenecity was studied and compared with inactivated whole cell vaccine in vitro and in vivo. The protein estimation and the polypeptide pattern of these antigens were also compared by Lowery and SOS-PAGE methods. In general the immunological responses to these fractions were varied. The chickens immunized with KSCN-extract antigen and OMP showed the highest protection when they were challenged with homologous strain, while ifS and LPS antigens showed the lowest protection, These findings are in correspondence with the end-point titer of antibodies using ELISA. Cross-reaction was also observed by agar gel difussion test between the fractions and raised antisera in chickens especially with KSCN-extract. OMP and CAP antigens, The results suggcst that both OMP and KSCNextract protein as more effective immllnogens cOlild be good candidates for devclopmcnt or a subunit vaccine substitut\: for current who le œil formalized vaccine against fowl cholera infection.

The anaphylactic reactions in caule and buffaloes following haemorrhagic septicaemia vaccination in different parts of Iran were reported. The free endotoxin as an allergic substance is a component of liquid phase of the vaccine. A dense culture of bovine Pasteurella multocida type 8:2 was prepared in tri ptose phosphate broth supplemented by yeast extract. The bacterial cells were separated by centrifugation and alum gel precipitation methods. The safety trials of the vaccines conducted in mice, rabbits, guinea pigs, callie and buftàlos. The shock reaction did not occur either in laboratory or farm animais, which were tested in both controlled and field conditions.
