Objective
Choroid plexus epithelial cells (CPECs) have the epithelial characteristic, produce cerebrospinal fluid, contribute to the detoxification process in the central nervous system (CNS), and are responsible for the synthesis and release of many nerve growth factors. On the other hand, studies suggest that normobaric hyperoxia (HO) by induction of ischemic tolerance (IT) can protect against brain damage and neurological diseases. We examined the effect of combination therapy of encapsulated CPECs and HO to protect against ischemic brain injury.
Materials and MethodsIn this experimental study, six groups of adult male Wistar rats were randomly organized: sham, room air (RA)+middle cerebral artery occlusion (MCAO), HO+MCAO, RA+MCAO+encapsulated CPECs, HO+MCAO+encapsulated CPECs, RA+MCAO+empty capsules. RA/HO were pretreatment. The CPECs were isolated from the brain of neonatal Wistar rats, cultured, and encapsulated. Then microencapsulated CPECs were transplanted in the neck of the animal immediately after the onset of reperfusion in adult rats that had been exposed to 60 minutes MCAO. After 23 hours of reperfusion, the neurologic deficit score (NDS) was assessed. Next, rats were killed, and brains were isolated for measuring brain infarction volume, blood-brain barrier (BBB) permeability, edema, the activity of superoxide dismutase (SOD), and catalase (CAT) and also, the level of malondialdehyde (MDA).
ResultsOur results showed that NDS decreased equally in HO+MCAO, RA+MCAO+encapsulated CPECs, and HO+MCAO+encapsulated CPECs groups. Brain infarction volume decreased up 79%, BBB stability increased, edema decreased, SOD and CAT activities increased, and MDA decreased in the combination group of HO and transplantation of encapsulated CPECs in the ischemic brain as compared with when HO or transplantation of encapsulated CPECs was applied alone.
ConclusionThe combination of HO and transplantation of encapsulated CPECs for stroke in rats was more effective than the other treatments, and it can be taken into account as a promising treatment for ischemic stroke.

مقدمهسکته مغزی مهمترین بیماری نورولوژیک و سومین عامل مرگ و میر بعد از بیماری های قلبی و سرطان در افراد مسن می باشد. با وجود تلاش های زیاد در جهت درمان یا فهم پاتوفیزیولوژی سکته مغزی هنوز درمان موثری برای آن یافت نشده است. گیاه زیره سبز با نام علمی Cuminum cyminum در درمان بیماری ها به کار می رود. این مطالعه با هدف بررسی اثر عصاره زیره سبز بر کاهش آسیب سد خونی- مغزی حاصل از سکته مغزی انجام گرفت.روش کاردر این مطالعه تجربی از 5 گروه 6 تایی موش های صحرایی نر استفاده شد. گروه ها شامل شم، کنترل و 3 گروه آزمایشی بودند که دوز های 25 ،50 و100میلی گرم بر کیلو گرم وزن بدن عصاره زیره سبز را به صورت خوراکی به مدت 30 روز دریافت نمودند. 2 ساعت بعد از آخرین گاواژ گروه های کنترل و آزمایشی تحت جراحی انسداد شریان میانی مغز به منظور القای ایسکمی و اندازه گیری استحکام سد خونی- مغزی قرار گرفتند.نتایجغلظت اوانس آبی در نیمکره آسیب دیده (نیمکره راست) مغز در گروه کنترل در مقایسه با دوزهای 25 و 50 عصاره تفاوت معنی داری نداشت. اما دوز 100 نسبت به گروه کنترل تفاوت معنی داری را نشان داد به طوری که میزان اوانس آبی در نیمکره راست کاهش یافت (05/0>p).نتیجه گیریپیش تیمار با عصاره خوراکی زیره سبز با دوز 100 (میلی گرم بر کیلو گرم وزن بدن) باعث کاهش آسیب سد خونی- مغزی حاصل از سکته مغزی می گردد.کلید واژگان: سکته مغزی, زیره سبز, سد خونی, مغزی, موش صحرایی}Introductionstroke is most important neurological disease and the third cause of mortality after heart diseases and cancer in the elderly. Despite many efforts for the treatment or understanding the pathophysiology of stroke, an effective treatment has not been found yet. The Cumin plant with the scientific name "Cuminum cyminum" is used for the treatment of diseases. This study was conducted the effect of cumin on reduce the damage of blood-brain barrier resulting from stroke.
Methods & Materials: In this experimental study, the male rats were divided in 5 groups of 6. Groups including sham, control and 3 experimental groups that were received 25, 50, 100 mg/kg of cumin extract orally for 30 days. 2h after the last gavage, control and experimental groups were underwent middle cerebral artery occlusion surgery to induce stroke and measure the stability of blood-brain barrier.ResultsConcentration of Blue Evans in the damaged hemisphere (right hemisphere) in the control group had no significant difference compared with the doses of 25 and 50 of cumin extract. But dose of 100 showed a significant difference compared to the control group as Blue Evans in the damaged hemisphere decreased (PConclusionPre-treatment with 100 mg/kg of cumin extract reduces damage of the blood-brain barrier resulting from stroke.Keywords: Stroke, Cumin, Blood, brain Barrier, Rat}
