    Background
    Resveratrol (RSV) provides several important biological functions in wide variety of cells. In this study, we investigated the molecular mechanisms underlying anti-inflammatory effect of RSV on HepG2 cells by assessing the gene expression of RelA and c-Jun- subunits of NF-κB and AP-1 transcription factors.

    Methods
    HepG2 cells were settled in a serum- free medium with high concentrations of glucose (30 mM) and insulin (1 µM) overnight and were then incubated with RSV (5, 10, and 20 µM) for 24 and 48 hours. Real time quantitative polymerase chain reaction (qRT-PCR) was used to determine RelA and c-Jun expression.

    The findings of the present study demonstrated that RSV may be considered as a preventative and therapeutic agent for antagonizing inflammation in Hepatocellular carcinoma (HCC).

    Background
    Colorectal cancer is one of the mostly diagnosed malignancies worldwide. The main risk factors for colorectal cancer include the mutation of tumor suppressor genes or proto-oncogenes and unhealthy lifestyle. Vegetable and fruit consumption with multiple anticancer agents can reduce the risk of colon cancer. Resveratrol, is a natural polyphenolic product that inhibits proliferation and induces apoptosis through several pathways. In this study, the effects of resveratrol on β-catenin (CTNNB1) and GSK-3β expression affecting the Wnt-signaling pathway, were examined and morphology changes were analyzed in colon cancer cells with high levels of β-catenin such as HCT-116.
    Methods
    HCT-116 cells were seeded into 6-well plates and the cells were treated with various concentrations of resveratrol (25, 50 and 100 μM) for 24, 48 and 72 hours. β-catenin and GSK-3β expression were examined by Quantitative Real-time PCR and morphology changes were analyzed.
    Results
    Our result showed that resveratrol in 25 and 50 μM concentrations of resveratrol reduces β-catenin and GSK-3β expression in 24h (p-value; 0.001). Gene expressions were found to increase in 48h and 72h treatment with resveratrol in the concentration of 50 and 100 μM (p-value; 0.001).
    Conclusions
    considering our data, we suggest that low doses of resveratrol could reduce β-catenin expression, which can affect the Wnt-signaling pathway. While, high doses can increase the GSK-3β expression, playing a role in the destruction of β-catenin, inhibition of its accumulation in the cytoplasm and nuclear, apoptosis induction and cellular proliferation inhibition. On the other hand, low doses of resveratrol can decrease GSK-3β expression and suppress proliferation.

    Background
    Hyperglycemia is one of the important featurs of diabetes. In cell culture studies different methods are used to mimic the hyperglycemia condition. In this study we investigate response of human liver cancer cell line (HepG2) to high insulin, high glucose, and high insulin/ high glucose medium exposure.
    Methods
    HepG2 cells were settled in DMEM.1% FBS or DMEM free-serum medium with high concentrations of d-glucose (30 mm) and/or insulin (1μM) for 24h after an overnight starving in serum- free medium. The level of hyperglycemia was estimated in the supernatants via GOD-POD method.
    Results
    Serum-free madium with high insulin/ high glucose consentration made the higher level of hypreglycemia in HepG2 cells.
    Conclusions
    Our study interduced high insulin/ high glucose treatment as the best way to induction hyperglycemia.
