توکسوپلاسما گوندی انگلی است که میزبان نهایی آن، گربه و گربه سانان، نقش مهمی در گسترش بیماری دارند و ایمن سازی آن ها یکی از راه های کنترل آلودگی به شمار می رود. این مطالعه با هدف ارزیابی ایمنی زایی سوش تخفیف حدت یافته توکسوپلاسما گوندی تولید شده در موسسه رازی شیراز در گربه انجام پذیرفت.
مواد و روش هادوازده بچه گربه نر نژادDSH (Do mestic short hair) با سن 6-3 ماه به صورت تصادفی به سه گروه چهارتایی تقسیم بندی شدند. گروه اول سوش تخفیف حدت یافته تاکی زوایتتوکسوپلاسما گوندی با دوز 106 ×10 را به صورت تزریقی، گروه دوم دوز 106×25 را به صورت خوارکی و گروه آخر به عنوان کنترل تنها محیط کشت دریافت نمودند. ایمنی هومورال و سلولی به ترتیب با استفاده از آزمون آگلوتیناسیون و آزمون پوستی انجام گرفت. یک ماه بعد از ایمن سازی تمامی گربه ها توسط سووشPRU توکسوپلاسما آزموده شدند و پس از آن به مدت پنج روز مدفوع گربه ها جمع آوری و آزمون مولکولی انجام گردید.
یافته هانتایج آزمون آگلوتیناسیون و آزمایش پوستی در هر دو گروه تزریقی و خوراکی افزایش معناداری را با گروه کنترل نشان دادند. تست مولکولی نشان داد که ایمن کردن گربه با واکسن تخفیف حدت یافته توسوپلاسما گوندی در هر دو صورت خوراکی و تزریقی قادر به جلوگیری از دفع اووسیست توسط گربه می باشد
نتیجه گیریبا توجه به نتایج موفقیت آمیز پاسخ ایمنی و همچنین جلوگیری از دفع اووسیست، پیشنهاد می شود در مطالعات آتی با بررسی دقیق تر سوش، امکان استفاده از آن به عنوان واکسن در گربه مورد بررسی قرار گیرد.

MethodsTwelve male DSH (Domestic short hair) kittens aged 3-6 months were randomly divided into three groups; each contains four cats. The First group received tachyzoites of Toxoplasma gondii attenuated isolate at a dose of 10×106 by injection, the second group received a dose of 25×106 orally and the last group received culture medium as a control. The humoral immune responses and Cellular immunity evaluated by modified agglutination and skin test. One month after immunization, all cats were challenged by Toxoplasma's PRU strain, after which the cats' feces were collected and molecular tests were performed for five days.
ResultsThe results of modified agglutination and skin test showed that administration of oral and injection vaccine had a significant increase in comparison with control. Also, Molecular testing after challenge revealed that immunization of cats by oral and injection way of Toxoplasma gondii attenuated isolate could successfully prohibit oocysts shedding in cats.
ConclusionIn order to the significant immune responses and prevention of oocysts shedding, it is suggested that, using this isolation of Toxoplasma gondii as a possible candidate vaccine for cats in future studies.

Background
Clostridium perfringens is an important anaerobic bacterium found in the intestine of some livestock. It is concerned with the etiology of some diseases including enterotoxaemia. Various diseases are caused by different types of C. perfringens. Nonetheless, there is no published research on molecular typing and distribution of this pathogenic microorganism in Fars province.
ObjectivesAccordingly, our study focused on the isolation and toxin typing of C. perfringens from sheep, cattle, and goats in different parts of Fars province by the culture and the polymerase chain reaction (PCR) method.
Materials and MethodsApproximately 459 fecal samples were collected and cultured on defined media for the isolation of C. perfringens. The confirmed isolates were genotyped by the PCR method using specific primers.
ResultsC. perfringens was isolated from 30.93% of the total samples. The results of toxin typing showed a total of 76 (54%), 13 (9%), 30 (21%), and 23 (16%) isolates as types A, B, C, and D, respectively.
ConclusionOur results indicated that C. perfringens type A was the most common type in sheep, cattle, and goats while the lowest number of isolates belonged to type B. Finally, the isolation of C. perfringens and toxin typing increase our knowledge of the epidemiology of these diseases and can help in the vaccine industry and better controlling related diseases.

BackgroundThe protein listeriolysin O (LLO) encoded by hly gene, is one of the most important virulence factors of Listeria monocytogenes. This highly potent immunogenic cholesterol binding toxin has hemolytic activity, responsible for phagosomal membrane disruption and bacterial escape to the cytoplasm and facilitating the stimulation of CD8 T cells and Th1 response. Recently pathobiotechnological vaccination using probiotic bacteria have been proposed. One of these strategies is expression of LLO in non-pathogenic bacteria such as lactic acid bacteria as delivery strains.ObjectivesOur aim in this study was cloning of hly gene in a Lactobacillus species via pNZ8110, an inducible expression vector which is specific for Lactococcus species.Materials And Methodshly gene was amplified by PCR and cloned into pNZ8110 by restriction enzymes cutting and ligation method. After transformation and propagation in E. coli MC1061 intermediate host, it was successfully electrotransformed into Lactobacillus plantarum.ResultsGel electrophoresis of colony PCR, extracted plasmids and restriction analysis along with sequencing confirmed the transformation. After induction using supernatant of nisin producer Lactococcus lactis NZ9700 strain, Expression of LLO was confirmed by SDS PAGE and western blot.ConclusionHere, we have employed a nonpathogenic probiotic strain; Lactobacillus plantarum for the first time to express hly gene of Listeria monocytogenes in order to propose a new vaccine candidate.Keywords: Listeriolysin O, Cloning, Lactobacillus plantarum}

سابقه و هدفباکتری اشریشیا کلی سویه K99 مهم ترین عامل ایجاد کننده اسهال در گوساله ها می باشد. کلی سین یکی از انواع باکتریوسین ها می باشد که به وسیله سویه های مختلف اشریشیا تولید شده و خاصیت کشندگی بر روی باکتری های همان نوع یا باکتری های مشابه دارد. این مطالعه با هدف بررسی باکتری های کلی سینوژنیک به عنوان روشی در درمان عفونت اشریشیا کلی سویه K99 انجام گرفت مواد و روش ها: این مطالعه به صورت تجربی بر روی 192 بچه موش BALB/c، در 2 گروه تست وکنترل انجام شد. موش های گروه 1 کنترل تنها سوسپانسیون باکتری کلی سینوژنیک و به موش های گروه 2 کنترل تنها سوسپانسیون باکتری اشریشیا کلی K99 خورانده شد. به موش های گروه تست نیز پس از خوراندن سوسپانسیون باکتری کلی سینوژنیک، در فواصل زمانی مختلف سوسپانسیون باکتری اشریشیا کلی K99 داده شد. در نهایت میزان مرگ و میر بچه موش ها در گروه های کنترل و تست مورد بررسی قرار گرفت. یافته ها: نتایج این بررسی نشان داد که تمامی بچه موش ها در گروه 1 کنترل زنده ماندند. اما در 83% (40 مورد) از بچه موش های گروه 2 کنترل، به دلیل حضور سم ناشی از باکتری اشریشیا کلی K99، مرگ و میر مشاهده گردید. از طرفی میزان مرگ و میر در بچه موش های موجود در گروه های تست نیز به طور چشمگیری کاهش یافته بود. همچنین مشخص گردید که خاصیت مهار کنندگی باکتری های کلی سینوژنیک به غلظت باکتری و زمان تلقیح بستگی دارد.نتیجه گیریبا توجه به نقش مهم باکتری های کلی سینوژنیک در مهار باکتری اشریشیا کلی K99، کلی سین ها می توانند به عنوان گزینه ای مناسب در درمان بیماری های عفونی ناشی از این سویه باکتریایی مورد توجه قرار گیرند. از طرفی کلی سین ها می توانند در آینده نزدیک به عنوان یک پروبیوتیک، جایگزینی مناسب برای آنتی بیوتیک های متداول باشند.کلید واژگان: اشریشیا کلی K99, کلی سین, باکتری کلی سینوژنیک}Background and ObjectiveE. coli k99 is one of the major causes of diarrhea in neonatal calves. Colicin is an antimicrobial peptide produced by one strain of E. coli to suppress the growth of other strains of E. coli. The aim of this study was to control E. coli k99 infection in mice by oral administration of colicinogenic E. coli (CEC).Material and MethodsThis experimental study was conducted on two groups of control and treatment infant mice, strain BALB/c. The control group was subdivided into two groups: one group was fed with colicinogenic E. coli (CEC) and the second control group was E. coli k99. The treatment group was administrated with colicinogenic E. coli (CEC) and also with E. coli k99 in intervals. Finally, the mortality rate of the mice was assayed in both treatment and control groups.ResultResults of this study delineated that all mice in the control group who were fed with colicinogenic E. coli was survived while 83% of the second control group (40 cases) died due to production of heat-stable enterotoxin. Our study showed that the mortality rate in the treatment group was significantly reduced, and the reduction rate was increased in higher doses of colicinogenic E. coli (CEC).ConclusionBased on the antibacterial activity of CEC against E. coli k99, colicin is an appropriate choice for control of the infection. باکتری پاستورلا مالتوسیدا به صورت هم زیست در دستگاه تنفسی فوقانی و سیستم گوارشی بسیاری از حیوانات اهلی و وحشی وجود دارد. سروتیپ هایA و D این باکتری از مهم ترین عوامل ایجاد کننده ذات الریه در گوسفند و بز به شمار می روند. فیمبریه و ادهسین، کپسول، توکسین، فاکتور جذب آهن، پروتئین غشای خارجی از مهم ترین فاکتورهای بیماریزای این باکتری محسوب می شوند. هدف از این پژوهش طراحی روشMultiplex PCR برای تشخیص هم زمان و سریع مهم ترین ژن های بیماریزا، تیپ بندی کپسولی و نیز شناسایی پاستورلامالتوسیدا جدا شده از گوسفند و بز دارای علایم تنفسی در استان فارس بود.
مواد و روش هادر مجموع 500 سوآب از لوزه و بینی گوسفند و بز دارای علایم تنفسی و مشکوک به پاستورولوز تنفسی از استان فارس جمع آوری گردید. در ابتدا با استفاده از تست های های بیوشیمیایی گونه باکتری شناسایی و تایید گردید. سپس به کمک پرایمرهای اختصاصی مهم ترین فاکتورهای حدت باکتری به همراه تیپ کپسولی مورد بررسی قرار گرفتند.
یافته هادر این مطالعه 8/83 درصد از سویه های پاستورلا مالتوسیدا سروتیپ کپسولی A و 4/6 درصد سروتیپ D بودند. همچنین میزان فراوانی ژن های omph, ptfA, hgbA, toxA به ترتیب 4/77%، 19/74%، 74/67% و 74/67% گزارش گردید.
نتیجه گیریاکثر سویه های پاستورلا مالتوسیدا جدا شده از گوسفندان و بزها در استان فارس توکسیژن بودند و سروتیپ کپسولیA در بین جدایه ها شیوع بیشتری داشت. از میان ژن های بیماریزای مورد بررسی، دو ژن toxA وompH کد کننده درمونکروتوکسین و پروتئین غشای خارجی، بیشترین میزان شیوع را در سویه های جداسازی شده از گوسفند و بزها داشتند. بنابراین پایش دو ژن یاد شده می تواند نقش موثری در اپیدمیولوژی و عفونت تنفسی گوسفند و بز داشته باشد.

Serotypes A and D are responsible for pneumonia in goats and sheep, and Fimbriae, adhesions, capsule, toxin, iron uptake factor and outer membrane protein are its important virulence factors. The aim of this study was to design a multiplex PCR for fast and simultaneous detection of more important virulence factors of P. multocida isolated from sheep and goats as well as their capsular typing and identification.
Materials and MethodsTotally 500 nasal swabs were collected from the goats and sheeps suspected to respiratory signs of pasteurellasis. After isolation abd detection of P. multosida by biochemical examination, the bacteria were assayed by specific primers for identification of major important virulence factors and their capsular typing.
ResultCapsular typing of the isolates by PCR showed two capsular types A, D with 83.8% and 6.4% prevalence, respectively. The results also showed that 23 (74.19%), 21 (67.74%), 21(67.74%) and 24(77.4%) of the isolates were positive for presence of toxA, hgbA, ptfA and ompH genes, respectively.
ConclusionThe most isolated P. multocida from goat and sheep were toxigenic, and capsular type A was the most common isolates in the Fars province. The remarkable high prevalence of toxA and ompH among the afflicted sheep and goats may imply to important role of these genes in epidemiology and virulence of P. multocida. Furthermore the high prevalence of P. multocida typeA harbor toxA gene can be attributed to its important role in the respiratory infection.

It is not clear what factors determine divergent outcomes of infections caused by H. pylori. In the present study, the protein profiles of different strains of H. pylori, isolated from three groups of patients with ulcerative disease, non-ulcerative gastritis and cancer disease, were analyzed using 1D-SDS-PAGE. The patterns of different H. pylori strains were highly divergent. About 30.76% (7 bands) of the 26 observed protein bands were common in all strains isolated from 3 groups of the patients. While the similarity for the strains inside each group were 75% (15 from 20), 76.47% (13 from 17) and 78.57% (11 from 14) for cancerous, ulcerative and nonulcerative group, respectively. Some of the observed bands were significantly specific for each group. Therefore, we speculated that some H. pylori strains might be more associated with a specific disease than others, giving the clustering of some, but not all, strains within each disease group. In conclusion, this study showed that protein profile can be a characteristic in discrimination of dominant strains in different gastric clinical status. Specific and dominant proteins of different strains isolated from three groups of patients under study were candidates for further exploration for laboratory tests, which analyze disease-specific H. pylori strains, and for diagnosis of the different diseases and outcomes associated with this widespread bacterium.
