هدف
اصلاح ساختار ریشه با جذب کارآمد آب و استفاده موثر از مواد مغذی مرتبط است و استقرار بهتر گیاه در خاک را فراهم می کند. مهندسی ژنتیک برای بهبود ساختار ریشه می تواند منجر به حفظ عملکرد بالاتر در شرایط خشکی شود. این پژوهش با هدف اصلاح ساختار ریشه برنج به منظور تحمل به خشکی و بهبود کارایی جذب عناصر غذایی انجام گرفت. برای این منظور انتقال همزمان ژن های Deeper Rooting1 یا DRO1 که در تنظیم زاویه رشد ریشه ها برای انطباق با شرایط خشکی نقش دارد و Phosphorus-Starvation Tolerance1 یا PSTOL1 که در افزایش جذب فسفر و بهبود ساختار ریشه موثر است، به گیاه برنج طراحی و اجرا شد.
مواد و روش هاژن های DRO1 و PSTOL1 برگرفته از ارقام وحشی برنج طی مراحلی به ترتیب تحت پیشبرنده مختص ریشه و پیشبرنده دایمی همسانه سازی و در ناحیه T-DNA حامل دوگانه اگروباکتریومی قرار داده شدند. سازه حاصل موسوم به pUhrDroPstol به اگروباکتریوم سویه EHA105 منتقل و برای انتقال ژن به برنج رقم هاشمی مورد استفاده قرار گرفت. پس از انجام مراحل انتقال ژن، گیاهان باززاشده حاصل در محیط انتخابی حاوی 50 میلی گرم بر لیتر هیگرومایسین در مراحل مختلف کالوس زایی، باززایی و ریشه زایی زنده مانده و رشد کرده و به محلول یوشیدا و سپس به گلدان منتقل شدند. گیاهان تراریخته احتمالی توسط واکنش زنجیره ای پلیمراز (PCR) مختص سازه و مختص ژن مورد تایید قرار گرفتند و رخدادهای مستقل مشخص شدند.
نتایجدر این پژوهش 12 رخداد احتمالی برنج تراریخته حاصل شد که 10 رخداد در آنالیز PCR حضور هر دو ژن OsDRO1 و OsPSTOL1 را نشان دادند. مقایسه فنوتیپ ریشه با گیاه شاهد تفاوت ظاهری در ساختار ریشه نشان داد. گیاهان تراریخته حاصل در گلخانه تراریخته پژوهشگاه بیوتکنولوژی کشاورزی بذرگیری شدند و خلوص آنها با آنالیز مولکولی در نسل های T1 و T2 مشخص شد.
نتیجه گیریدر این تحقیق دو ژن کاندید موثر در تغییر ساختار ریشه و تحمل به خشکی با استفاده از مهندسی ژنتیک به برنج رقم هاشمی منتقل شد. تاکنون انتقال همزمان این دو ژن کاندید گزارش نشده است. گیاهان تراریخته حاصل سیستم ریشه ای بهتری را نسبت به گیاه شاهد نشان می دهند. سازه چند ژنی حاصل را می توان با هدف تغییر ساختار ریشه و تحمل به خشکی و بهبود کارایی جذب فسفر برای انتقال ژن به سایر گیاهان نیز مورد استفاده قرار داد. امید است تولید برنج تراریخته با بهبود کارایی استفاده از عناصر غذایی و با تغییر ساختار ریشه بتواند تحمل به خشکی و عملکرد را در این محصول مهم زراعی افزایش و سبب کاهش مصرف آب در کشت برنج شود.

It also provides structural support for the anchoring in soil. Genetic engineering for the improvement of plant root structure may help to maintain higher yields under drought conditions. The aim of this study was to modify the root structure of rice in order to improve drought tolerance and the efficiency of nutrient uptake. For this purpose, simultaneous transformation of Deeper Rooting1 or OsDRO1 gene, which is involved in the regulation of growth angle of the root in order to adapt to drought conditions, and Phosphorus-Starvation Tolerance1 or OsPSTOL1 gene, which is effective in increasing phosphorus uptake and improving root structure, were considered for rice root structure modification.
Materials and methodsThe OsDRO1 and OsPSTOL1 genes derived from the wild rice cultivars were cloned together in a single construct under the control of the root specific and the ubiquitin promoters, respectively. The resulting construct, pUhrDroPstol is transformed into the Agrobacterium tumefactions strain EHA105 and used for the gene transformation into Hashemi cultivar. Putative transgenic plants, survived on 50 mg/L Hygromycin during tissue culture steps, are transplanted into the Yoshida solution and then into the pots until they set seeds. Construct specific and gene specific PCR analysis are used to confirm the transgenic plants.
ResultsIn this study, 12 putative transgenic rice events were obtained, of which 10 showed the presence of both OsDRO1 and OsPSTOL1 genes in the PCR analysis. Transgenic plants show stronger root structure compared to the non-transgenic ones. Molecular analysis in the T1 and T2 generations determined the homozygous events.
ConclusionsIn this study, two candidate genes affecting root structure, nutrient uptake and drought tolerance were transferred to the Hashemi rice using genetic engineering. So far, simultaneous transfer of these two candidate genes have not been reported. Transgenic plants present better root system compared to the control plants. The mentioned construct can be used for the transformation of other crops to improve their root structure, nutrient uptake and their drought tolerance. It is hoped that the production of the transgenic rice with modified root structure and efficient phosphorus uptake increases its drought tolerance and reduce water consumption in rice cultivation.
