مقدمهبیماری پارکینسون با از دست رفتن انتخابی گروهی از سلول های دوپامینرژیک در جسم سیاه ایجاد می شود. یکی از روش های درمانی این بیماری، جایگزینی سلول های از دست رفته توسط سلول های بنیادی است. این مطالعه با هدف تاثیر القاکننده های عصاره مغز نوزاد رت و دپرنیل در تمایز سلول های کارسینومای جنینی P19به سلول های عصبی انجام گرفت.روش بررسیسلول های بنیادی P19 در محیط کشت α-MEM همراه با سرم 10% FBS و پس از آن سلول ها بر روی ظروف کشت با چسبندگی پایین جهت تولید اجسام شبه جنینی کشت داده شدند. جهت تمایز، سلول های شبه-جنینی در محیط کشت حاوی سرم 3% FBS به همراه عصاره مغز نوزاد رت، دپرنیل و ترکیبی از هر دو به مدت 28 روز کشت داده شدند. برای ردیابی پروتئین های ویژه سلول های عصبی مانند سیناپتوفیزین، بتاتوبولین3 و نستین در سلول های تمایزیافته از روش ایمنوفلورسنس و به منظور سنجش بیان ژن های اختصاصی عصبی از Real-time PCR استفاده شد.یافته هادر روش ایمنوفلورسنس سلول های P19 تمایز یافته با هرسه گروه القاگر نسبت به آنتی باد های نستین، سیناپتوفیزین و بتاتوبولین3 پاسخ مثبت نشان دادند. بررسی بیان نسبی فاکتورهای رونویسی نستین، سیناپتوفیزین در سلول های تمایزیافته با هرسه القاگر با استفاده از روش Real-time PCR حاکی از بیان بالای هر دوی این ژن ها تحت تاثیر هر سه گروه القاگر در هفته دوم بود و در هفته های سوم و چهارم کاهش معنی داری را نشان داد.نتیجه گیریسلول های بنیادی کارسینومای جنینی P19 تحت شرایط آزمایشگاهی و تحت تاثیر عوامل القاگر قابلیت تمایز به سلول های عصبی به ویژه فنوتیپ دوپامینرژیک را دارند.کلید واژگان: سلول های کارسینومای جنینیP19, دپرنیل, عصاره مغز نوزاد رت, تمایز عصبی}IntroductionParkinson's disease cause by selective loss of a group of dopaminergic cells in the Substantial nigra. One of the treatments for this disease is the displacement of lost cells by stem cells. The purpose of this study was to investigate the effect of inducers of extract of neonatal rat brain and deprenyl on neural differentiation of P19 embryonic carcinoma cells.Materials and MethodsP19 stem cells were cultured in alpha-MEM medium supplemented with 10% FBS serum. Then, the cells were cultured on low-adhesion culture plates to create pseudo-embryonic bodies. In order to differentiate, germ cells were cultured in medium containing 3% FBS serum and extract of neonatal rat brain, deprenyl and mixture of both extract for 28 days. Immunofluorescence assay and Real-time PCR was applied to detect and evaluate the expression specific proteins of neurons including synaptophysin, beta-tubulin 3 and nestin, respectively.ResultsRegarding to immunofluorescence results, differentiated P19 cells, positively affected by three inducers such as nestin, synaptophysin and beta-3. Relative expression of transcription factors of nestin, synaptophysin in cells differentiated by all three inducer using Real-time PCR demonstrated that in P19 embryonic carcinoma cells, Nestin and Synaptophysin expression increased in the second week, while, thier expression significantly reduced in the third and fourth weeks.ConclusionThe results of this study indicated P19 embryonic carcinoma stem cells are potential to differentiate into neurons, especially dopaminergic phenotype, following laboratory conditions and inducing factorsKeywords: P19 embryonic carcinoma cells, deprenyl, extracts of neonatal rat brain, neural differentiation}

ObjectiveThere is longstanding experimental and clinical evidence that supports the idea that replacement of dopaminergic (DAergic) neurons can ameliorate functional disabilities of Parkinson’s disease (PD). The purpose of the present study is to examine the efficacy of transplantation of rat bone marrow stromal cell (BMSCs)-derived tyrosine hydroxylase-positive (TH+) cells induced by deprenyl into 6-hydroxydopamine (6-OHDA)-lesioned rat models, using behavioral tests and immunohistochemical evaluations.Materials And MethodsIn this experimental study, undifferentiated BrdU-labeled BMSCs were incubated in serum-free medium that contained 10-8 M deprenyl for 24 hours. Afterwards, BMSCs were cultured for 48 hours in α-minimal essential medium (α-MEM) supplemented with 10% FBS, then differentiated into TH+ neurons. We randomly divided 24 hemiparkinsonian rats as follows: group 1 (control) received only medium, while groups 2 and 3 were injected with 2×105 BMSCs and deprenyl-treated cells in 4 μl medium. Injections were made into the injured strata of the rats. Rotational behavior in response to apomorphine was tested before transplantation and at 2, 4, and 6 weeks post-graft. Animals were then sacrificed, and the brains were extracted for immunohistochemical and electron microscopic studies.ResultsApomorphine-induced rotation analysis indicated that animals with grafted cells in groups 2 and 3 exhibited significantly less rotational behavior than those in the control group at 2, 4, and 6 weeks after transplantation. Immunohistochemical analysis demonstrated that BrdU-labeled cells expressed specific neuronal markers, such as NF 200 and TH, at the implantation site. The presence of TH+ cells in conjunction with the reduction in rotation might show the capacity of grafted cells to release dopamine. Ultrastructural analysis revealed the presence of immature neurons and astrocyte-like cells at the graft site.ConclusionTH+ neurons induced by deprenyl can be considered as a cell source for PD autograft therapy.Keywords: Bone Marrow Stromal Cells, Deprenyl, Cell Therapy, Parkinson's Disease, 6, Hydroxydopamine}

زمینه و هدفاستفاده از القاگرهای شیمیایی برای تمایز سلول های بنیادی مزانشیمی مغز استخوان بالغ (BMSCs) به سلول های عصبی مورد توجه محققین است و در ابتدا لازم است تا اثر سمیت القاگر شیمیایی بر سلول های تحت القاء بررسی گردد. بدیهی است افزایش درصد سلول های زنده پس از القاء می تواند بهترین معیار برای تعین مناسب ترین القاء کننده باشد. این مطالعه به منظور تعیین اثرات القایی دپرنیل و دی متیل سولفوکساید (DMSO) بر تکثیر و بقاء سلول های بنیادی مزانشیمی مغزاستخوان انجام شد.روش بررسیاین مطالعه تجربی روی 20 سر موش صحرایی بالغ نژاد ویستار در دانشکده زیست شناسی دانشگاه دامغان انجام شد. BMSCs از مغز استخوان موش صحرایی بالغ استخراج و در محیط αMEM حاوی FBS ده درصد کشت داده شدند. در پاساژ سوم تعیین هویت سلولی برای آنتی ژن های سطحی CD71 و CD90 به روش ایمونوسیتوشیمی و قابلیت چندتوانی BMSCs با تمایز به سلول های چربی و استخوان انجام شد. سلول ها به مدت 24 ساعت در معرض عوامل القاگر (محیط کشت تکمیل شده با 2درصد دی متیل سولفوکساید و محیط کشت تکمیل شده با دپرنیل 10 به توان منفی 8 مولار) قرار گرفتند و بعد از آن به محیط αMEM حاوی FBS ده درصد منتقل شدند. تکثیر و بقاء سلولی پس از 24، 48، 72 و 96 ساعت با روش آزمون MTT مورد ارزیابی قرار گرفت. داده ها با استفاده از نرم افزار آماری SPSS-18 و آزمون های One-Way ANOVA و Tukey تجزیه و تحلیل شدند.یافته هاسلول های چسبنده به فلاسک کشت که از مغز استخوان جداشدند؛ علاوه بر بیان آنتی ژن های سطحی CD71 و CD90 توانایی تمایز به سلول های چربی و استخوان را داشته و نتایج حاصل از آزمون MTT نشان داد که بقاء و تکثیر سلول های القاء شده با دپرنیل و دی متیل سولفوکساید در زمان های 48، 72 و 96 ساعت پس از القاء نسبت به گروه کنترل منفی به طور معنی داری افزایش داشته است (p<0.05).نتیجه گیریدپرنیل موجب افزایش بقاء و قابلیت تکثیر سلولی در مقایسه با دی متیل سولفوکساید شد و می توان این ترکیب را به عنوان القاگر سلولی مورد استفاده قرار داد.

Plasticity and easy accessibility of bone marrow mesenchymal stem cells is a unique charactristic for treatment of neural disorderies. This study was desgined to determine the inductive effect of Deprenyl and Dimethyl sulfoxide on proliferation and survival of the mesenchymal stem cells.Materials And MethodsIn this experimental study، BMSCs isolated from the adult rat bone marrow and cultured in αMEM containing 10% FBS. Cell identity for surface antigens was performed in third passage by immunocytochemistry and multipotancy capacity of BMSCs was done by BMSC differentiation into adipocytes and osteocytes. The cells were exposed to chemical agents (a: the αMEM medium supplemented with 2% DMSO، b: the αMEM medium supplemented with 10-8M Deprenyl) for 24 houres and then transferred to αMEM containing 10% FBS cell survival and proliferation was evaluated after the 24، 48، 72 and 96 houres by MTT [3- (4-5-Dimethylthiazolyl-2-y1) -2،5-diphenyltetrazolium bromid] test. Data were analyzed using SPSS-16، One-Way ANOVA and Tukey tests.ResultsIn addition to expression the surface antigens and adipogenic and osteogenic differentiation by BMSCs، MTT test results showed that proliferation and survival of induced-deprenyl and DMSO cells within 48، 72 and 96 hours after the induction was increased significantly than negative control group.ConclusionDeprenyl increases survival and cell proliferation compared to Dimethyl Sulfoxide.
