    Objective (s)

    Patient-derived xenograft (PDX) model becomes a more and more important tool for tumor research. This study aimed to establish a colorectal cancer PDX model and verify its applicability.

    Materials and Methods

    Fresh human colorectal cancer tissue was surgically removed and subcutaneously inoculated into immunodeficient mice to establish the PDX model. Hematoxylin and eosin (HE) staining and immunohistochemical staining were used to evaluate the model. The successful PDX model was selected to study the efficacy of capecitabine in treating colorectal cancer. 

    Results

    HE staining showed that the PDX mice model of colorectal cancer could preserve the histological characteristics of the primary tumor. Immunohistochemistry staining showed α-fetoprotein (AFP), carcinoembryonic antigen (CEA), and E-cadherin were strongly positively expressed in primary human and PDX tumor tissues, with a high degree of similarity. Capecitabine significantly inhibited PDX tumor growth and reduced the expression of AFP and CEA proteins in the tumor tissues (all Ps<0.05). 

    Conclusion

    We successfully established a colorectal cancer PDX model, and the PDX model could retain the histological and biological characteristics of the primary tumor. Using this PDX model, we revealed that capecitabine at a dose of 300–400 mg/kg can effectively treat colorectal cancer, and no significant difference in toxicity was found among different dose groups. The current work provides a feasible framework for establishing and validating the PDX tumor model to better facilitate the evaluation of drug efficacy and safety.

    Background

    The use of oncolytic viruses as therapeutic agents is a promising treatment for various human cancers. Several viruses have been extensively examined to achieve tumor cell death.

    Objectives

    This study aimed at evaluating the natural oncolytic activity of mumps Hoshino vaccine strain against two human cancer cell lines, that is, HT1080 fibrosarcoma and HeLa cervical adenocarcinoma cell lines.

    Methods

    The cytolytic activity of the virus was evaluated using an MTT assay. Apoptosis was detected by Annexin-V/propidium iodide (PI) staining and analyzed via flow cytometry. To indicate viral replication in vivo, nude mice with HeLa heterografts were treated with the Hoshino strain of mumps virus.

    Results

    It was found that human fibrosarcoma and cervical cells were more sensitive to the mumps Hoshino strain, even at a very low multiplicity of infection (MOI) compared to normal human diploid cells. The results also showed that the Hoshino strain induced apoptosis in both cancer cells. A preliminary in vivo study revealed the significant suppression of tumor growth in the group treated with the mumps Hoshino strain compared to the control group.

    Conclusions

    The Hoshino vaccine strain of mumps virus showed promising oncolytic activities against human fibrosarcoma and cervical adenocarcinoma cells.

    Background

    Meniscus plays a pivotal role in normal function of knee joint and is exposed to heavy load of pressure and trauma. The aim in this study is to investigate ultrastructure of medial and lateral meniscus of Ovis aries, in addition to comparing the findings with human meniscal structure.

    Methods

    14 samples of freshly-excised meniscus of ovis aries were provided. After conventional preparation, the samples were studied via electron microscopy (EM) and its elements were microanalyzed.

    Results

    In the macroscopic evaluation, the meniscus surface was completely smooth, but in the microscopic observation, longitudinal ridges and grooves were observed. In addition, several types of cells that were different morphologically and bundles of collagen fibers were observed. The major direction of collagen fibers was circumferentially, but there were radial fibers as well. In the microanalysis of the ovis aris meniscus, the following elements were present: sodium, carbon, and oxygen, with sodium having the highest percentage among the elements. In medial meniscus of the samples, a small amount of calcium was detected.

    Conclusion

    By comparing the present findings with those of other studies, many similarities were observed between ultrastructure of ovis aries and human knee meniscus. The compatibility in the ultrastructures will imply the possibility of application of this specimen for xenograft meniscal transplant procedure.

    Objectives

    Following tooth extraction, soft and hard tissue alterations occur; Different factors can affect this process. The objective of this study was to determine the effect of gap filling on buccal alveolar crestal bone level after immediate implant placement after 4- to 6-month observation period.

    Materials and Methods

    This   randomized clinical trial was performed on 20 patients (mean age of 38.8 years) requiring tooth extraction in a total of 27 areas in the anterior maxilla. The treatment strategy was as follows: atraumatic flapless tooth extraction, implant placement, insertion of a graft (test group) or no material (control group) between the implant and the socket wall, connection healing abutment placement and suturing the area. Clinical and cone beam computed tomographic examinations were performed before implant placement (baseline), 24 hours after surgery and 4-6 months (T2) after implant placement, to assess the buccal plate height (BH) and implant complications.

    Results

    After 4 months of healing, a reduction in different bone measurements was noticed in the two groups. No statistically significant differences were assessed in bone height measurements between the test and control groups at different time points. The study demonstrated that immediate implantation resulted in 1.30 and 1.66 mm reduction in buccal bone plate in the test and control groups, respectively.

    Conclusions

    The study demonstrated that immediate implantation in the extraction socket together with xenograft failed to prevent bone resorption.
