    Background and objectives
    Medicinal plants have effectively treated Alzheimer’s disease (AD). In this study, the oleo-gum-resin of Ferula persica known as sagapenum was selected to investigate its inhibitory activity toward enzymes involved in the creation and progression of AD. Also, the phytochemical analysis, which was not previously reported in the literature, was conducted.
    Methods
    The in vitro inhibitory activity of dichloromethane, methanol, and aqueous extracts was investigated toward acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) according to the modified Ellman’s method. Moreover, the phytochemical analysis of the most potent extract was conducted using different column chromatography techniques.
    Results
    The dichloromethane extract showed selective BuChE inhibitory activity (IC50 = 23.41 µg/mL), compared with donepezil as the reference drug (IC50 = 1.97 µg/mL). Phytochemical analysis of the related extract led to the isolation and identification of aurapten, farnesiferol A, umbelliprenin, farnesiferol C, farnesiferone A, karatavicinol, ferocaulidin, and ligupersin A, which were assayed toward cholinesterases (ChEs). Farnesiferol A was the most potent and selective inhibitor of BuChE (IC50 = 31.46 µg/mL). Moreover, it showed good β-secretase 1 (BACE1) inhibitory activity (IC50 = 5.14 µM), compared with the positive control, OM99-2 (IC50 = 0.014 µM) to be considered a multi-target directed ligand against AD.
    Conclusions
    Selective anti-BuChE activity of the dichloromethane extract of sagapenum, as well as farnesiferol A and its good anti-BACE1 activity, may play a significant role in the development of anti-AD supplements.

    مقدمه

    آنقوزه یک گیاه دارویی انحصاری ایران است. اولیو گام رزین آن یا صمغ آنقوزه، فرآورده گیاهی مهمی است که استانداردسازی آن برای اطمینان از موثر بودن و ایمنی آن اهمیت دارد.

    هدف

    به دلیل فقدان داده های دقیق و کامل در مورد صمغ آنقوزه ایرانی، هدف از این مطالعه این است که با گزارش بعضی خواص فیزیکوشیمیایی و فیتوشیمیایی یک نمونه صمغ آنقوزه ایرانی، دانش موجود توسعه داده شود.

    روش بررسی

    خوص ارگانولپتیک با ارزیابی حسی تعیین شدند. همچنین خاکستر تام، خاکستر نامحلول در اسید، باقیمانده خشک و نیز محتوای تام فنولی و فلاونوییدی با روش های پذیرفته شده اندازه گیری شد تا بعضی از خواص فیزیکوشیمیایی و فیتوشیمیایی این اولیو گام رزین تعیین گردد. آنالیز اسانس آن بوسیله GC/MS و نیز ارزیابی بار میکربی انجام گردید.

    نتایج

    صمغ آنقوزه توده ای قهوه ای مایل به زرد با بوی تند گوگرد- مانند شبیه به بوی سیر، با خاکستر تام و خاکستر نامحلول در اسید به ترتیب برابر با 2/83 درصد و 2/06 درصد و باقیمانده خشک 3/43 درصد بود. محتوای تام فنلی برابر با 0/26 ± 4/15 میلی گرم گالیک اسید در هر گرم صمغ خشک بود. ترکیبات عمده در آنالیز اسانس آن شامل 1-(E) -پروپنیل سکو بوتیل دی سولفید، 1-(Z)  -پروپنیل سکو بوتیل دی سولفید و 4,2 -تیازلیدین دیون بودند. بار میکربی در محدوده قابل قبول فارماکوپه بود.

    نتیجه گیری

    یافته های حاضر اطالعاتی را در مورد پاره ای خواص فیزیکوشیمیایی و فیتوشیمیایی صمغ آنقوزه ایرانی فراهم می سازد که برای کامل کردن دانش موجود کاربردی است. اگرچه اطلاعات بیشتری برای استانداردسازی مورد نیاز است که از مطالعات وسیع تر حاصل خواهد شد.

    Background

    Ferula assa-foetida is an endemic medicinal plant of Iran. Its oleo-gum resin or assafoetida gum (AG) is an important herbal product whose standardization is important to ensure effectiveness and safety.

    Objective

    Due to the lack of accurate and complete data about Iranian AG, the objective of this study is to extend the available knowledge by reporting some physicochemical and phytochemical properties of an Iranian AG sample.

    Methods

    Organoleptic properties were determined by sensory evaluation. Total ash, acid-insoluble ash, dry residue as well as total phenolic and flavonoid content were measured by accepted methods to determine some physicochemical and phytochemical properties of AG. Analysis of its essential oil by GC/MS as well as microbial load assessment were performed.

    Results

    The AG was a yellowish brown mass with a sulfur-like pungent odor similar to garlic, with total ash and acid-insoluble ash of 2.83 % and 2.06 %, respectively, and a dry residue of 3.43 %. Total phenol content was 4.15 ± 0.26 mg gallic acid equivalent/g in dry AG. (E)-1-Propenyl sec-butyl disulphide, (Z)-1-propenyl sec-butyl disulphide, and 2,4-thiazelidinedione were the major essential oil component of this oleo-gum resin. Microbial load was in acceptable range according to Pharmacopeia.

    Conclusion

    The current findings provide data on some of the physicochemical and phytochemical properties of Iranian AG which is applicable for completing current knowledge. Although more information is needed for establishing Iranian AG standardization criteria, which will be obtained from more extensive researches.

    Background and objectives

    The oleo-gum resin from Ferula gummosa</em> Bioss. is commercially important. Variability of geographic location has shown a deep effect on essential oil of different genus of plants. Thus, evaluation of oleo-gum resin of Ferula gummosa</em> from different locations is important for further studies. In the present study, phytochemicals profile of essential oil of oleo-gum resins from F. gummosa</em> in different geographic location and various bioclimates of Iran, were studied.

    Methods

    The oleo-gum resin of F. gummosa</em> was collected from different regions of Iran. The essential oils of the samples were extracted by Clevenger apparatus and were analyzed by GC-MS.

    Results

    The highest amount of essential oil compared was observed in the samples from Damghan area (24.0%), Firuzkuh area (22%) and Kalat-e-Naderi area (22%). The variability in the oil composition of different samples exhibited a notable difference in monoterpenes content from 39.15 % to 87.48%. Based on GC-MS analysis, the major components were β</em>-pinene (5.11-63.80 %), sabinene (3.37-30.69%), α</em>-pinene (4.43-12.72%) and δ</em>-carene (2.38-10.23 %). The highest concentrations of β</em>-pinene (63.80%) and sabinene (30.69%) were observed in the Shiraz and Torbat-e-Jam essential oils, respectively.  The highest amounts of α</em>-pinene (12.72%) and δ-</em>carene (10.23%) were recorded in Firuzkuh and Shiraz areas, respectively.

    Conclusion

    Significant variation in the component of the essential oils from different oleo-gum resins can be used to improve the knowledge about the endemic medicinal plants.

    In this experiment, the chemical composition of the essential oils obtained from Ferula assa-foetida oleo-gum resin collected from two different sites of Yazd province (Tabas and Yazd) in the center of Iran, were identified. The gas chromatography mass-spectroscopy (GC/MS) data showed that the qualitative composition of the components appeared to be constant in two different regions. Moreover, no remarkable variations were found in the amounts of the essential oil major constituents. A total thirty-nine components, comprising 91.52% and 95.61% of the total oil, were characterized in Tabas and Yazd samples, respectively. The hydrodistilled oils contained E-1-propenyl sec-butyl disulfide (40.15 and 44.36% in Tabas and Yazd samples, respectively), Z-1-propenyl sec-butyl disulfide (23.93 and 27.98%), Guaiol (5.50 and 3.14%) and Carotol (5.14 and 1.63%) as major constituents.
