Chronic liver disease (CLD) affects millions of people and its impact on bone loss has become a subject of interest. Nitric oxide and endogenous opioids are suggested to increase during cholestasis/cirrhosis and may impact bone resorption by different mechanisms. The receptor activator of nuclear factor-κB (RANK)/RANK-ligand (RANKL)/osteoprotegerin (OPG) signaling pathway regulates bone resorption, but its role in metabolic bone disease subsequent to CLD is unknown. We aimed to investigate the involvement of nitrergic and opioidergic systems in bone loss relative to the RANK/RANKL/OPG pathway, in bile duct-ligated (BDL) rats. Eighty BDL/sham-operated (SO) rats received injections of 3 mg/kg/day Nω-Nitro-L-arginine methyl ester ± naltrexone (10 mg/kg/day) or saline for 28 days. Plasma bone turnover markers, OPG, RANK, and RANKL along with mRNA expression levels of the latter three were assessed. Plasma bone turnover markers and OPG level increased, but RANKL decreased in the BDL group compared with their SO controls (both: P ≤ 0.05). Administration of naltrexone reduced bone turnover markers and OPG level while increased RANKL content in comparison to BDL rats (P ≤ 0.05). As compared to untreated BDL rats, nitric oxide inhibition showed no effect on bone turnover marker i.e. OPG, RANK, and RANKL levels. BDL significantly increased RANK mRNA, but had no significant effect on RANKL and OPG mRNA expression. The lack of association between plasma levels and quantitative gene expression of RANKL and OPG suggests an indirect function of these markers in BDL rats. Considering that opioid receptor blockage by naltrexone in BDL animals caused a significant decrease in OPG and an increase in RANKL plasma contents, it could be postulated that the opioidergic system may have a regulatory effect on these bone markers.Keywords: Cirrhosis, Bone loss, Nitrergic system, Opioidergic system, BDL rats, RANK, RANKL, OPG axis}

سابقه و هدفهورمون های جنسی می توانند درک درد را در جنس نر و ماده تغییر دهند. در رابطه با نحوه عمل هورمون های جنسی نر و ماده بر درک درد اطلاعات کافی وجود ندارد. هدف این مطالعه بررسی میزان تاثیر اوپیوئیدهای درون زا بر درک درد در موش های سوری نر و مراحل مختلف سیکل جنسی ماده (پرواستروس، استروس، مت استروس و دی استروس) می باشد.مواد و روش هادر مطالعه پژوهشی حاضر از موش سوری نر و ماده بالغ (در مراحل محتلف سیکل جنسی) با وزن 3±30 گرم در گروه های کنترل، شاهد تزریق و دریافت کننده نالوکسان (3 میلی گرم/کیلو گرم، داخل صفاقی) استفاده شد. در همه گروه ها زمان پاسخ دهی به محرک دردناک با تست صفحه داغ ارزیابی شد.نتایجزمان بی دردی در فاز دی استروس گروه های کنترل و شاهد تزریق موش های ماده بیشترین مقدار بوده و با فاز استروس (5 0/0>P) و نرها (001/0> P) اختلاف داشت. نالوکسان باعث کاهش معنی دار زمان بی دردی هم در نرها (001/0> P) و هم در فازهای پرواستروس، استروس، مت استروس و دی استروس ماده ها به ترتیب (001/0 >P<0/001) (P<0/001) (P<0/05) (P) گردید. هم چنین، نالوکسان موجب تفاوت در زمان بی دردی موش های نر نسبت به ماده ها در فاز پرو استروس و دی استروس (5 0/0 >P) شد.نتیجه گیریبه نظر می رسد بخشی از اثرات هورمون های جنسی موش های نر و ماده در مکانیسم درک درد توسط اوپیوئیدهای درون زا واسطه گری شود.

There are few information about the role of sex hormones on nociception. The aim of this study was to evaluate the effect of endogenous opioids on nociception in male mice compared with female mice during different phases of the estrous cycle (proestrus، estrus، metestrus and diestrus).Materials And MethodsIn this study، adult male and female mice (in different phases of estrous cycle (weighing 30±3gr were assigned to the control group، vehicle and naloxone (3mg/kg، i. p.) groups. In all groups analgesia time was evaluated by hotplate test.ResultsAnalgesia time in diestrus phase of control and vehicle groups of female rats was more than the other phases and different from the estrus phase (P<0. 05) and male (P<0. 001) groups. In the naloxone group، there was a significant reduction in analgesia time among males (P<0. 001) and also in the proestrus، estrus، metestrus، diestrus phases (P<0. 001، P<0. 05، P<0. 001، P<0. 001، respectively) among females. Moreover، in the naloxone group، a significant difference was seen in analgesia time in male mice compared with females in proestrus and diestrus phases (P<0. 05).ConclusionIt seems that the effects of sex hormones in male and female mice on pain perception mechanisms can be mediated by endogenous opioids.Keywords: Hot plate test, Pain, Opioidergic system, Estrus cycle, Mouse} -
مقدمهسیستم اوپیوئیدرژیک درون زاد نقش مهمی در بیان حساسیت به درد، ادراک درد و پاسخ به برخی شرایط استرس زا ایفا می نماید. مطالعه حاضر نقش احتمالی سیستم اوپیوئیدرژیک درون زاد را در محرومیت غذایی بر رفتارهای دردی القا شده با فرمالین در موش های بزرگ صحرای جنس نر و ماده مورد بررسی قرار داده است.روشجهت ایجاد محرومیت غذایی کوتاه مدت، حیوانات از مصرف غذا به مدت 48 ساعت پیش از انجام آزمون فرمالین محروم شدند، در حالی که آب در دسترس آن ها بود. در گروه شاهد حیوانات به غذا و آب آزادانه دسترسی داشتند. تزریق فرمالین در کف پای راست انجام شد. نالوکسان جهت بررسی دخالت سیستم اوپیوئیدرژیک درون زاد بر اثر محرومیت غذایی روی رفتارهای دردی تزریق شد.یافته هاتزریق فرمالین به کف پای راست حیوان سبب ایجاد رفتارهای دردی در دو فار شد. فاز اول و دوم به وسیله اینترفاز که رفتارهای درد کاهش می یابد از هم تفکیک می شوند. 48 ساعت محرومیت غذایی سبب افزایش رفتارهای دردی در موش های نر و ماده در پاسخ به فرمالین شد، در حالی که رفتارهای دردی در فاز 2 با تاخیر به پایان رسید. تزریق نالوکسان از افزایش رفتارهای دردی در مرحله اینترفاز در موش های نر و مرحله اول در موش های ماده بعد 48 ساعت محرومیت غذایی جلوگیری کرد.نتیجه گیریمطالعه حاضر نشان داد که محرومیت غذایی از طریق ایجاد تحمل در سیستم اوپیوئیدرژیک، سبب بروز رفتارهای دردی القا شده توسط فرمالین در مرحله اینترفاز می شود که توسط نالوکسان قابل برگشت است.

The present study examined the probable role of endogenous opioidergic system in effects of food deprivation on formalin-induced nociceptive behaviors in male and female rats.MethodsTo make short-term food deprivation, the rats did not receive food for 48 hours prior to the formalin test. However, they could freely access water. Afterward, 50 µl formalin 2% was injected into the bottom of the rats’ right paw. Naloxone was injected to evaluate the involvement of the endogenous opioidergic system in effects of food deprivation on formalin-induced nociceptive behaviors.ResultsFormalin injection caused nociceptive behaviors in two phases. The first and second phases were separated by a brief interphase where nociceptive behaviors decreased. As a result of 48-hour food deprivation, nociceptive behaviors in male and female rats were increased and the second phase was finished with delay. Naloxone administration blocked the pronociceptive effect of the 48-hour food deprivation in the interphase of male rats and in the first phase of female rats.ConclusionThe present study indicated that food deprivation increased formalin-induced nociceptive behaviors through affecting the opioidergic system.Keywords: Food deprivation, Opioidergic system, Formalin test, Naloxone}
