    Background

    Previous evidence suggests that exposure to organochlorine pesticides (OCPs) may contribute to the development of type 2 diabetes.

    Objectives

    The relationship between OCPs and type 1 diabetes (T1D) in children and adolescents is not well understood. We aimed to explore the association between organochlorine pesticide levels and T1D in this population.

    Methods

    In this case-control study, we included 147 newly diagnosed T1D cases and 147 healthy controls. Spot urine samples were collected from children under 15 years old in both groups to measure organochlorine pesticide levels. We analyzed six OCPs: β-HCH, heptachlor, aldrin, heptachloro epoxy, α-endosulfan, and p,p'-DDD, categorizing them into tertiles. We examined the relationship between urinary OCP levels and T1D, adjusting for age, sex, duration of breastfeeding, Body mass index (BMI), family history of diabetes, and five dietary patterns.

    Results

    The average urinary concentration of p,p'-DDD was significantly higher in cases than in controls (0.15 (0.07 - 0.3) vs. 0.09 (0.04 - 0.2) μg/g creatinine, P < 0.001). After adjustments for age, sex, breastfeeding duration, BMI, family history of diabetes, and dietary patterns, a significantly positive association was observed between the highest levels of p,p'-DDD, and T1D (odds ratio (OR) = 4.9; 95% confidence interval (CI): 2 - 12.3). Additionally, participants in the middle tertile of urinary β-BHC had a higher OR for diabetes compared to those in the lowest tertile (OR = 2.9; 95% CI: 1.2 - 6.8). No association was found between other OCPs and T1D.

    Conclusions

    These findings highlight a potential role for p,p'-DDD, and β-BHC in the development of T1D, urging further investigation into the mechanisms of this association and potential preventive strategies in this area.

    Background

    Methylation alterations of tumor suppressor gene promoters are essential aspects of epigenetic changes in gastric cancer (GC). Cadherin1 (CDH1) encodes a protein with essential roles in cell-cell adhesions. In this study association of organochlorine pesticides (OCPs) serum levels with the methylation profile of this gene was investigated in gastric cancer (GC), intestinal metaplasia (IM), and functional dyspepsia (FD) patients.

    Methods

    GC (n=34), IM (n=8), and FD (n=48) patient serums were analyzed for the determination of OCP levels by gas chromatography equipment. The methylation status of the CDH1 gene promoter was examined by methylation-specific PCR (MSP) method. In order to confirm reduced protein expression of this gene in methylated samples, immunohistochemistry (IHC) was used.

    Results

    Our findings revealed significant hypermethylation of CDH1 gene promoter along with reduced expression in GC patients compared with IM and FD patients. Furthermore, there was significant association between CDH1 promoter hypermethylation and 2,4-DDT (odds ratio [OR]:1.183;95%CI:1.001-1.398, p=0.048) serum levels in GC patients.

    Conclusion

    Our results could suggest the association of 2,4-DDT OCP levels in the GC patient serums with CDH1 gene promoter hypermethylation. Additionally, this gene promoter methylation may play a role in the progression of pre-cancerous IM towards GC.

    In this study, sixteen (16) organochlorine (OC) pesticides in sediment, soil and fish (Clarias anguillaris) samples of River Owan, Edo State, Southern Nigeria were investigated in accordance with acceptable analytical methods. Our findings confirmed the presence of the 16 OCPs in different concentration values in soil, sediment, and fish (Clarias anguillaris). The ∑OCP in the sediments samples were between 0.0033-0.0319 (ng g-1) with total value of 0.1506 (ng g-1) for the sediment sample 1 (SE1); 0.0021-0.0587(ng g-1) with total value of 0.0291(ng g-1) for the sediment sample 2 (SE2). The level of the OCPs in the soil ranged between 0.0069-0.0626 (ng g-1), total concentrations of 0.3986(ng g-1) for soil sample 1 (SO1); 0.0026-0.0630 (ng g-1), total concentration of 0.3678 (ng g-1) for soil sample 2 (SO2). The concentration level of the OC pesticides were between 0.0046-0.1707 (ng g-1) in the fish sample with total concentration of 0.1490 (ng g-1). These pesticides were found in higher concentrations in soil than in sediment samples. The level of OC pesticide residues detected in the fish was greater than the sediment samples. The hazard quotient is moderate (1.49) when compared with the guidelines.

    Six rice brands BG-213, Notore, Das, Kwandala, Jeep and Jamila brands were collected from various locations in Kura local government area of Kano, Nigeria. Rice brands were analyzed for proximate compositions, macro elements, heavy metals and pesticides using standard analytical techniques. Results of proximate compositions analyses indicate that the average mean value of moisture content, crude protein, crude fat, ash content and crude fiber in rice brands were; 10.27, 9.17 3.82, 1.93 and 0.34 % respectively. The average mean concentrations of macro elements (Calcium (Ca) and Magnesium (Mg)) are; 53.5 mg/kg and 46 mg/kg, while mean levels of Heavy Metals: Zinc (Zn), Copper (Cu), Nickel (Ni), Chromium (Cr), Cadmium (Cd) and lead (Pb) were; 11.87, 4.04, 0.06, 0.00, 0.044 and 0.00 mg/kg respectively. Pearson Correlation Coefficient analysis performed on rice brands using MATLAB student version VI software shows results of macro elements and heavy metals were at a significant level of p≥0.05. The GC/MS analysis of organochlorine pesticides in rice brands revealed that the level of pesticides was below the detection limit (BDL) this indicates that brands of rice obtained from the Kura local government area were safe and free from heavy metals and pesticides contamination could be attributed to restriction or regulation for compliance on the application and economy because most of these pesticides are very costly.

    Persistent organic pollutants, (POPs), are vast distributed compounds in environment which are recognized as one of the global pollution problems. These groups of materials being dangerous due to their high stability are accumulated in animal tissues and occurring in the food chain. One of the major paths through which persistent organic combinations access to human body is consuming polluted foods, particularly, fishes. Among aquatic animals, trout as one of the mostly consumed fishes in Tehran’s food basket was studied. In this study two categories of persistent organic pollutants: Organochlorine pesticides (OCPs) including HCB, Dieldrin, Methoxychlor, α-, ϒ-Chlordane, α-, β-Endosulfan and o,p’-DDE, p,p’-DDE, o,p’-DDT, p,p’-DDT and the second group Polychlonitated biphenyls (PCBs) including seven PCB congeners which are called indicator PCBs (IUPAC nos.: 28,52,101,118,138,153 and 180) were determined in trout by GC-MS/MS in MRM monitoring mode and LLE extraction. The average recoveries of OCPs and PCBs at five concentration levels were in the range of 73-112%. The relative standard deviations of POPs in fish were in the range of 1.4-17.9% for all of the concentration levels. Limit of detections (LODs) and limit of quantitations (LOQs) were between 0.6-8.3 and 2-25 µg/kg, respectively. The results indicated the presence of organochlorine pesticides in trout and the levels of p,p’-DDE and p,p’-DDT were within the range of < LOQ -12.83 and < LOQ -10.2 ng/g ww (wet weight), respectively. According to the results, OCPs residues were lower than maximum residue levels set by European Council Directives.

    The present study aimed to assess the levels of organochloride pesticides (OCPs) in the human breast milk obtained from the nursing women admitted for delivery to a hospital in Marivan county, located in Kurdistan, Iran. Milk samples were collected from 30 women. Gas chromatography analysis was performed using a Dani 1000 (Italy) gas chromatograph equipped with the Ni electron capture detector (ECD). The findings showed the high concentrations of OCPs in the samples. On the other hand, DDTs were ranked as the most abundant OCPs, followed by HCHs and HCBs, with the concentrations estimated at 2,345, 2,617, and 570 ng/g lipid/wt, respectively. Therefore, it could be concluded that the tested women might have been exposed to OCPs. However, further investigation is required on larger sample sizes to take proper measures.
