مقدمه
فلاونوییدها، گروهی از ترکیبات طبیعی، در گل ها، میوه ها، ریشه ها و ساقه ها یافت می شوند. این دسته از ترکیبات طبیعی به دلیل اثرات دارویی مفید شناخته شده اند، از این جهت، جداسازی و شناسایی فلاونوییدها از گیاهان ارزشمند است. فلاونوییدها در حال حاضر کاربردهای گسترده ای در صنایع غذایی، دارویی و آرایشی دارند. آپیژنین، کویرستین و آپیژنین-7-O-گلیکوزید دو گروه از فلاونوییدها هستند که دارای خواص درمانی نیز هستند.
هدفهدف از این مقاله خالص سازی سه ترکیب آپیژنین، کویرستین و آپیژنین-7-O-گلیکوزید از گیاهان و شناسایی آنها با کمک NMR, HPLC UV-visible است.
روش بررسیدر این مطالعه عصاره تام سه گیاه کرفس، جعفری و پیاز با کمک حلال هایی با قطبیت مختلف به بخش های مختلف حلالی تقسیم شدند. برای خالص سازی این فراکسیون ها از کروماتوگرافی ستونی استفاده شد. در نهایت از تکنیک های 1H-NMR، 13C-NMR و طیف سنجی جرمی برای شناسایی استفاده شد. همچنین برای شناسایی این ترکیبات از تکنیک HPLC و استانداردسازی استفاده شد.
نتایجدر کار حاضر، آپیژنین از فراکسیون اتیل استات دانه کرفس استخراج شد. آپیژنین-7-O-گلیکوزید با خلوص بالا از عصاره بوتانولی قسمت های هوایی جعفری خالص سازی شد. همچنین، کویرستین از فراکسیون اتیل استاتی پیاز جدا شد. آنالیز داده های به دست آمده از 1H-NMR، 13C-NMR و مقایسه این داده ها با منابع خلوص ترکیبات اثبات شد.
نتیجه گیری:
روش های به کار رفته در این پژوهش توانسته اند این سه ترکیب را با درصد خلوص بالا، جهت استفاده در صنعت به عنوان استاندارد تولید کنند.

These natural products are well known for their health-beneficial effects. Hence, isolation and purifying of flavonoids from the ingredients is worthwhile. Flavonoids are now considered indispensable in various nutraceutical, pharmaceutical, medicinal and cosmetic applications. Apigenin, quercetin, and apigenin-7-O-glycoside have therapeutic properties that these two are in the flavonoid group.
ObjectivesIn this study, apigenin, quercetin, and apigenin 7-O-glycoside were purified by column chromatography and identified by NMR and HPLC UV-visible techniques.
MethodsTotal extracts of three plants (Apium graveolens, Petroselinum crispum, Allium cepa) were divided into different solvent fractions. Column chromatography is used to purify these fractions. Finally, 1H-NMR, 13C-NMR, and mass spectrometry techniques were used for identification. Also, the HPLC technique was used for identifying these compounds and standardized.
ResultsIn the present work, apigenin was extracted from ethyl acetate fraction of Apium graveolens seeds. High purity of apigenin 7-O-glycoside isolated from the butanol fraction of Petroselinum crispum aerial parts. Also, ethyl acetate fraction isolated quercetin from the Allium cepa. Identification of the data obtained from 1H-NMR and 13C-NMR, which were proofed by comparison with the sources of purified compounds.
ConclusionThe methods used in this research were able to produce these three compounds with a high percentage of purity to be used in industry as standards.

IntroductionPetroselinum crispum (P. crispum) is an associate of Umbelliferae family that has several therapeutic attributes. Ischemia/reperfusion (I/R) is one of the main challenges in acute kidney damage. This study was designed to assess the anti-inflammatory and protective effects of P. crispum extract against I/R-induced renal disorders.MethodsForty male rats were randomly divided into five groups (n=8) namely normal control (saline) and I/R control group, and three groups of I/R intraperitoneally pretreated with various doses of P. crispum (100, 150 and 200mg/kg). The I/R-induced renal inflammation was evaluated by determining leukocyte infiltration and mRNA expression level of intercellular adhesion molecule-1 and tumor necrotic factor-alpha. Antioxidant capacity of kidneys and thiobarbituric acid reactive species were measured in kidneys for the evaluation of oxidative stress. In addition, the diameters of renal glomeruli, kidney function indicators and serum nitrite oxide levels were respectively determined by morphometric analysis, autoanalyzer device and Griess technique.ResultsThe I/R increased all measured parameters except for the tissue ferric reducing/antioxidant power (FRAP) level, which was decreased compared to the normal control group. However, pretreatment with P. crispum extract in all doses significantly reduced blood urea nitrogen, kidney malondialdehyde, creatinine, glomerular diameter, leukocyte infiltration, levels of tumor necrotic factor-alpha, adhesion molecule-1 expression, and nitrite oxide as well as increased tissue FRAP compared to the I/R control group.ConclusionIt seems that P. crispum administration improves I/R-induced acute kidney injury.Keywords: Petroselinum crispum, Injury, Kidney

Apigenin is best known as an irreversible inhibitor of monoamine oxidase (MAO), an intracellular enzyme associated with the outer membrane of mitochondria. The purpose of this study is to establish a reliable and quick method for the assignment of Apigenin in Cosmos bipinnatus, Apium graveolens and Petroselinum crispum by high performance liquid chromatography with ultraviolet detection (HPLC-UV). A rapid and sensitive high performance liquid chromatographic (HPLC) method has been developed for determination of Apigenin in human apigenin. Mobile phase was composed water-acetonitrile (55:45 v/v) with a flow rate of 1 ml/min. The eluted peaks were detected by a UV detector was set at wavelength of 340 nm. The method was validated in the range of Apigenin concentrations from 0.01 to 500 µg/ml. The limits of detection (LOD) and quantitation (LOQ) of the method were 0.005 and 0.01 µg/ml, respectively. The average recovery was 97.28 percent throughout the linear concentration range. The average within-run and between-run accuracy values of 99.32 and 96.79 respectively. مقدمهبا توجه به پیشرفت روزافزون دیابت و استفاده از داروهای جایگزین و کم خطر گیاهی، در این مطالعه تجربی اثر عصاره هیدروالکلی برگ گیاه جعفری Petroselinum crispum بر فعالیت آنزیم های کبدی و آسیب شناسی بافتی بررسی گردید.مواد و روش هادر این مطالعه 60 سر موش صحرایی نر از نژاد ویستار به طور تصادفی به 5 گروه تیماری تقسیم شدند. پس از انجام تیمارهای لازم، از حیوانات تحت آزمایش نمونه های خونی و مقاطع بافت کبد اخذ گردید. فعالیت آنزیم های کبدیAST ، ALT ،ALP مورد سنجش قرار گرفت. مقاطع میکروسکوپیک از کبد تهیه و از نظر آسیب شناسی بافتی بررسی گردید.
یافته های پژوهش: فعالیت آنزیم های کبدی(ALT، AST، ALP) در گروه دیابتی در مقایسه با کنترل غیر دیابتی افزایش معنی داری نشان داد(P<0.01). مداخله عصاره گیاهی موجب کاهش معنی داری در فعالیت آنزیم ها و ترمیم بافت آسیب دیده کبد در موش های دیابتی گردید(P<0.01).بحث و نتیجه گیرییافته های این تحقیق نشان داد که عصاره هیدروالکلی برگ جعفری در موش های دیابتی قادر است فعالیت آنزیم های کبدی را تعدیل نماید. اثر ترمیمی این عصاره در حالت ابتلاء به دیابت بر بافت شناسی کبد قابل توجه بود. از این خواص فارماکولوژیکی و اثر ضد التهابی عصاره جعفری شاید بتوان از اثر حفاظتی آن در جلوگیری از التهاب کبدی ناشی از دیابت استفاده نمود.کلید واژگان: گیاه جعفری, آنزیم های کبدی, دیابت, موش صحراییIntroductionAccording to the progress of diabetes and the use of alternative herbal medicines, in this study, the effect of Petroselinum crispum parsley leaf hydroalcoholic extract on histopathological and liver enzymatic activity were examined.Materials and MethodsIn this study, 60 male wistar rats with the weight range of 250-300 g were divided randomly into 5 groups of treatment. After the necessary treatments tested on animals , blood and liver samples were collected . Activity of liver enzymes AST, ALT, ALP was measured . The microscopic sections were prepared from the liver and the pathological tissue was investigated .
Findings: Activity of liver enzymes (ALT, AST, ALP) in the diabetic group compared to non-diabetic controls showed a significant increase (pDiscussion &ConclusionsThe findings showed that the extract of parsley leaves in diabetic rats can modulate the activity of the hepatic enzymes. Healing effect of the extract in the case of diabetes on liver histopatology was remarkable. Background
Sperm dysfunction is one of the main causes of male infertility. Petroselinum crispum (P.crispum) is a member of umbelliferae family that contains different vitamins and minerals and has numerous therapeutic properties. The aim of this study was to evaluate P. crispum effect on sperm parameters, testis tissue and serum nitric oxid levels in mice.
Materials and MethodsHydroalcoholic extract of P. crispum was prepared and administered intraperitoneally (0,100, 150 and 200 mg/kg) to 40 mice, which were divided into four groups (n = 10), one control group and three experimental groups, for 14 consequent days. The sperm parameter such as motility, sperm count, morphology, and seminiferous tubules diameter, and weight of prostate and testis, and serum nitric oxide levels were analyzed.
ResultsP. crispum administration (100, 150 and 200 mg/kg) significantly increased mean percentage of sperm motility, testis and prostate weight and serum nitric oxide compared to the control group (P < 0.05). However, no significant effect was reported for different doses of P. crispum extract on sperm parameters.
ConclusionAdministrating hydroalcoholic extract of P. crispum has positive effects on some reproductive parameters.

ObjectivePresent study investigated the effects of parsley extract on pregnant rat kidneys which have undergone clinical abortion using prostaglandins. The renal protective effect of parsley extract was evaluated in pregnant rats which had an abortion. Parsley was used due to its antioxidant properties.Materials And MethodsFifty-four female rats were divided in 9 groups of 6: control pregnant, two pregnant groups which received parsley extract and prostadin, two non-pregnant groups treated with parsley extract and prostadin, a group administered with both treatments, and three groups which received parsley extract in pre-implantation, implantation, and post-implantation periods of embryos. Ethanolic extract (5 mg/kg) was given daily to animals for 18 days of pregnancy period. Parameters such as malondialdehyde (MDA), total antioxidant statues (TAS), creatinine, and urea were measured using biochemical assays. Histopathologic studies were also done with Hematoxylin-Eosin staining method.ResultsAfter 18 days of treatment, significant differences were observed in serum creatinine, urea, and MDA and TAS levels. Kidney cross-sections showed edema in prostadin-treated rats while improvements in parsley + prostadin -treated rats were observed.ConclusionThese results suggested that ethanolic extract of Petroselinum crispum reduced the dysfunction in rats kidney caused by prostadin-induced abortion and could have beneficial effect in reducing the progression of prostaglandin-induced edema.Keywords: abortion, Dysfunction, Kidney, Petroselinum crispum

Phytochemical Screening, Antibacterial and Cytotoxic Activities of Petroselinum crispum Leaves Grown in Oman.Petroselinum crispum belongs to the Apiaceae family, is a well-known spice and vegetable. The essential oil obtained from the fruit has also strong action on the central nervous system. Present study is designed to evaluate the phytochemical screening, antibacterial and cytotoxic activity of the leaves of Petroselinum crispum collected from Oman. Results of phytochemical screening indicate the presence of primary and secondary metabolites. Antimicrobial activity was measured using disc diffusion method against S.aureus, E.coli, P.aeruginosa and K. pneumonia. Brine shrimp test was used to estimate cytotoxic activity. In antibacterial assay, Petroselinum crispum leaves extracts showed very strong results, inhibition zones ranged from 7 – 20 mm. So this plant can be used as a good source of potential antimicrobial agent. Hydro alcoholic and Petroleum ether extracts showed the highest antibacterial activity. Furthermore, Ethyl acetate and Hydro alcoholic extracts have almost killed 90% of the shrimp larvae at higher concentration of 1000 µg/ml. LC50 values for the two extracts was 51.95 and 88.15 µg/ml, respectively.
