Background
Psidium guajava (guava tree) is widely used in Nigeria to treat diseases. However, a paucity of information exists on the safety of the plant.
ObjectivesThis study determined the safety of P. guajava leaves collected in Birnin Kebbi, Nigeria.
MethodsThe methanolic extract of the plant’s leaves was subjected to phytochemical and heavy metal screening using standard protocols, and thereafter, subjected to a cytogenetoxicity test using the Allium cepa toxicity assay. Twenty-one A. cepa bulbs divided equally into seven groups were grown over beakers containing distilled water (negative control), formaldehyde (positive control), as well as 0.25, 0.5, 1, 2, and 4 g of the extract, respectively, for five days. The root-tip cells of the A. cepa bulbs were treated and then examined for chromosomal aberrations.
ResultsThe phytochemical screening revealed high levels of saponins, and moderate levels of phenols, tannins, and flavonoids, while quinones and terpenoids were sparingly available. The heavy metal analysis showed non-permissible levels of cadmium and zinc, while two other tested heavy metals (lead and copper) were undetected. Except for the A. cepa treated with 0.25 and 0.5 g, the extract induced dose-dependent root growth and mitotic index inhibition (P<0.05). The extract also induced cytogenetic effects, mainly sticky, vagrant, and fragmented chromosomes as well as anaphase bridges.
ConclusionsIt can be inferred from the results that low to medium doses of the extract are safe but may elicit harmful effects at high doses. Advice from a phytomedicine or phytotherapy expert should be sought before using it.

مقدمه
ترکیبات فنلی و فلاونوییدها خواص بیولوژیکی متعددی مانند خاصیت آنتی اکسیدانی، به دام انداختن رادیکال های آزاد و خاصیت ضدالتهاب دارند. حلال های مختلف قابلیت متفاوتی در استخراج مواد فنلی و خواص آنتی اکسیدانی دارند؛ بنابراین، در این تحقیق سعی شد ضمن بررسی و مقایسه گیاهان گوناگون حوزه جنوب کشور از لحاظ محتوای فنلی و خواص آنتی اکسیدانی، تاثیر نوع حلال بر موارد یادشده نیز بررسی گردد.
مواد و روش هاتعداد 15 ژنوتیپ (انجیر معابد، لوز [گارم زنگی]، انجیر، سپستان، شاه توت، فالسا، گواوا، انبه 1، انبه 2، اکالیپتوس، جمبو، کنار 1، هندوانه ابوجهل، کنار 2، انار) از گیاهان دارویی جنوب کشور، بر اساس دو نوع عصاره متانلی و استونی از لحاظ میزان خواص آنتی اکسیدانی و مواد فنلی و فلاونوییدی در طرحی به صورت فاکتوریل، در قالب کاملا تصادفی و در سه تکرار ارزیابی شدند. تجزیه وتحلیل داده ها با استفاده از نرم افزار Statistic vol.10 و مقایسه میانگین ها با استفاده از حداقل تفاوت معنی دار (LSD) در سطح احتمال 1 درصد انجام گردید.
یافته هانتایج تجزیه واریانس نشان داد که اثر متقابل گیاه و خواص آنتی اکسیدانی (بر اساس DPPH) برای عصاره استونی و متانولی، در سطح احتمال 1 درصد معنی دار بود. در استفاده از حلال استونی، بیشترین میزان فنل در اکالیپتوس (mg/gFW43/7) و سپس جمبو (mg/gFW52/6)، فلاونویید در انبه 1(mg/gFW21/23) و سپس لوز (گارم زنگی) (mg/gFW75/18) و اکالیپتوس (mg/gFW36/15) و بیشترین خواص آنتی اکسیدانی در گواوا (24/85 درصد) و سپس کنار 1(68/82 درصد) و لوز (گارم زنگی) (31/82 درصد) به دست آمد. در استفاده از حلال متانلی، بیشترین میزان فنل از جمبو (mg/gFW82/7) و سپس اکالیپتوس (mg/gFW34/7)، فلاونویید از لوز (گارم زنگی) و انبه 1(به ترتیب mg/gFW46/24 و mg/gFW06/25) و سپس فالسا (mg/gFW07/16) و کنار 1 (mg/gFW51/13) و بیشترین خواص آنتی اکسیدانی از لوز (گارم زنگی) (07/82 درصد) و سپس انار (97/78 درصد) و کنار 1 (16/78 درصد) حاصل شد.
بحث و نتیجه گیریحلال استون موثرترین حلال در استخراج موادی فنلی و فلاونوییدی با خاصیت اکسیدانی بالاست و کنار و لوز (گارم زنگی) موثرترین گیاهان از لحاظ میزان مواد فنلی و خاصیت آنتی اکسیدانی هستند.

Different solvents have different capabilities in extracting phenolic materials and antioxidant properties. This study aimed to investigate and compare the different plants in the south of Iran regarding phenolic content and antioxidant properties; moreover, it was attempted to evaluate the effect of the solvent type on the mentioned issues.
Material & MethodsA total of 15 genotypes of medicinal plants (Ficus religiosa L, Terminalia catappa, Ficus carica, Cordia myxa, Black mulberry, Grewia asiatica, Psidium guajava, Mangifera 1, Mangifera 2, Eucalypteae, Syzygium cumini, Ziziphus 1, Citrullus colocynthis, Ziziphus 2, Punica granatum) were obtained from Roodān region (Hormozgan Province in Iran) and were also evaluated based on two types of methanolic and acetone extracts in terms of antioxidant properties, as well as phenolic and flavonoid substances in a factorial design in a completely randomized format with three replications. Data were analyzed using Statistic software (version 10), and the means were compared using the least significant difference at a 1% probability level.
FindingsThe results of the analysis of variance showed that the mutual effect of the plant and antioxidant properties (based on the DPPH) for acetone and methanolic extracts was significant at a 1% probability level. In the use of acetone solvent, the highest amount of phenol was in eucalyptus (7.43 mg/gFW), followed by Syzygium cumini (Java Plum) (6.52 mg/gFW). Furthermore, the highest amount of flavonoids was in mango 1 (23.21 mg/gFW), followed by Terminalia catappa (Indian-almond) (18.75 mg/gFW) and eucalyptus (15.36 mg/gFW). The most antioxidant properties were in Psidium guajava (Guava) (85.24%), followed by Ziziphus mauritiana 1 (Jujube 1) (82.68%) and Terminalia catappa (Indian-almond) (82.31%). In the use of methanol solvent, the highest amount of phenol was in Syzygium cumini (Java Plum) (7.82 mg/gFW), followed by eucalyptus (7.34 mg/gFW). In addition, the highest amount of flavonoids was in Terminalia catappa (Indian-almond) (mangroves) and mango 1 (24.46 mg/gFW and 25.06 mg/gFW, respectively) and then Grewia asiatica (Phalsa) (16.07 mg/gFW) and Ziziphus mauritiana 1(Jujube 1) (13.51 mg/gFW). The highest antioxidant properties were obtained from Terminalia catappa (Indian-almond) (82.07%), pomegranate (78.97%), and Ziziphus mauritiana 1(Jujube 1) (78.16%).
Discussion & ConclusionThe most critical solvent for the extraction of phenol and Flavonoid substances with high oxidative properties is acetone. The most useful plants in terms of the presence of materials and antioxidant properties were Terminalia catappa (Indian-almond) and Ziziphus mauritiana (Jujube).

استیوآرتریت یکی از ملاحظات بهداشت عمومی است که به علت درد مزمن، سفتی، اختلال خواب و افسردگی منجر به ناتوانی می شود. در میان محصولات گیاهی، میوه زیتون محلی (Psidium Guajava L) به دلیل ویژگی های آنتی اکسیدانی آن می تواند به عنوان یک داروی طبیعی بالقوه در نظر گرفته شود. هدف از این مطالعه بررسی اثر درمانی میوه زیتون محلی (Psidium Guajava L ) بر استیوآرتریت زانو در موش بود.
روش کارچهل سر ی با استیوآرتریت ناشی از تزریق داخل مفصلی 500 واحد کلاژناز نوع 2 در زانوی چپ در چهار گروه درمانی (10 تایی) قرار گرفتند (A) کنترل،(B) پیاسکلدین (10 میلی گرم در کیلوگرم) به عنوان کنترل مثبت، (C) تجویز خوراکی عصاره هیدروالکلی میوه زیتون محلی (Psidium Guajava L) (500 میلی گرم در کیلوگرم)، و (D) تجویز خوراکی عصاره هیدروالکلی میوه زیتون محلی (Psidium Guajava L) (1000 میلی گرم در کیلوگرم). پس از 8 هفته استفاده روزانه، نتیجه با ارزیابی های بافت شناسی و رادیوگرافی مورد بررسی قرار گرفت. داده ها با استفاده از نرم افزار Kruskal-Wallis در نسخه SPSS 23.00 مورد تجزیه و تحلیل قرار گرفت و P <0.05 از نظر آماری معنی دار تلقی شد.
نتایجعصاره هیدروالکلی میوه زیتون محلی (Psidium Guajava L) فعالیتهای خوب رادیکال با IC50 از 0.13 ± 0.46 میلی گرم / میلی لیتر (کویرستین به عنوان یک کنترل مثبت) نشان داد. ارزیابی های هیستوپاتولوژیکی غضروف، سطح مفصلی صاف و یکنواخت با توزیع سلول هایی در یک ستون را در گروه آزمایش دریافت 1000 میلی گرم بر کیلوگرم عصاره هیدروالکلی میوه زیتون محلی (Psidium Guajava L) نشان داد که در مقایسه با سایر گروه ها بخصوص گروه کنترل بهبودی بهتری داشت (P-value = 0.02). ارزیابی رادیولوژیکی مفصل زانو یافته های مشابه ارزیابی بافت شناسی نشان داد.
بحثعصاره هیدروالکلی میوه زیتون محلی (Psidium Guajava L) می تواند به عنوان یک مکمل موثر و درمان جایگزین برای آرتروز زانو در موش در نظر گرفته شود.

Among the herbal products Psidium Guajava L. fruit could be regarded as a potential natural remedy due to its anti-oxidant characteristics. The aim of this study was to evaluate the healing effect of Psidium Guajava L. fruit on osteoarthritis of the knee in rats.
MethodsForty male Sprague-Dawley rats with induced osteoarthritis by intra-articular injection of 500U of type 2 collagenase in the left knee were recruited in four treatment groups (n=10): (A) control, (B) Piascledine (10mg/kg) as positive control, (C) oral administration of hydro-alcoholic extract of Psidium Guajava L. fruit (500mg/kg), and (D) oral administration hydro-alcoholic extract of Psidium Guajava L. fruit (1000mg/kg). After 8 weeks of daily administration, the outcome was evaluated by histopathological and radiographic assessments. The data were analyzed using Kruskal-Wallis nonparametric in SPSS version 23.00, and P<0.05 was considered statistically significant.
ResultsPsidium Guajava extract exhibited good radical scavenging activities with IC50 of 0.46 ± 0.13 mg/mL (quercetin as a positive control). Histopathological assessments of the cartilage showed smooth and continuous articular surface with columnar cell distribution in the experimental group that received 1000mg/kg Psidium Guajava L. fruit hydro-alcoholic extract which healed better than others, especially in the control group (p-value =0.02). The radiological assessments of the knee joints showed similar findings in histopathological assessment.
ConclusionPsidium Guajava L. fruit hydro-alcoholic extract might be regarded as an effective complementary and alternative treatment for osteoarthritis of the knee in rats.

BackgroundSeveral plants have been shown to possess antioxidant and estrogenic properties that can be useful in postmenopausal bone-loss prevention. The present study aimed to investigate the anti-osteoporotic effects of the hydroalcoholic extract of the Psidium guajava (PG) fruit in ovariectomized (OVX) rats.MethodsSixty female Sprague–Dawley rats were randomly divided into 6 groups: a control positive group, a sham-operated group, an OVX group given normal saline (OVX-only group), and 3 treatment groups comprising 2 OVX groups treated orally with 500 and 1000 mg/kg/d of the hydroalcoholic extract of the PG fruit respectively and an OVX group treated with an injection of 0.15 mg/kg of estradiol. The study was conducted over a 12-week period. Samples from the animals’ blood, femoral bones, and uteri were collected for stereological and biochemical analyses. The data were analyzed using SPSS, version 19. A P value equal to or less than 0.05 was considered statistically significant.ResultsThe results revealed a significant decrease in the levels of calcium, total antioxidant capacity, and phosphorus as well as uterus weight, femoral ash density, femoral volume and weight, and numbers of osteocytes and osteoblasts. Moreover, there was an increase in the levels of alkaline phosphatase and urine deoxypyridinoline together with a rise in the number of osteoclasts in the OVX-only group compared to the control and treatment groups (P≤0.05). The hydroalcoholic extract of the PG fruit increased femoral weight and volume, femoral ash density, numbers of osteocytes and osteoblasts, and trabecular volume of the bones in comparison with the OVX-only group in a dose-dependent manner. No significant difference was observed between the groups in the levels of malondialdehyde and interleukin-6.ConclusionThe hydroalcoholic extract of the PG fruit prevented OVX-induced bone loss in the rats, with no proliferative effect on atrophic uteri; it should, therefore, be considered for treatment purposes.Keywords: AMP1 protein, Psidium guajava, Ovariectomy, Osteoporosis, Rat}

پیشزمینهاستئوآرتریت به عنوان بیماری دژنراتیو مفصلی شناخنه می شود. داروهای سنتتیک در دسترس برای درمان استئوآرتریت، عوارض جانبی خطرناک و اثربخشی کم دارند. بنابراین، استفاده از داروهای گیاهی، جزء مهم طب مکمل و جایگزین است، سلول های بنیادی مزانشیمی از جایگاه ویژه ای در درمان بیماری ها برخوردار می باشند. برگ گواوا به عنوان آنتی اکسیدان، ضد التهاب و ضد درد استفاده می شود. هدف از این مطالعه، بررسی تاثیر تزریق داخل مفصلی عصاره هیدرو الکلی برگ گواوا همراه با سلول بنیادی در زانوی رت می باشد.مواد و روش هاابتدا 90 سر رت نر انتخاب و برای ایجاد استئوآرتریت در مفصل زانو، کلاژناز نوع 2 تزریق شد. سپس به 9 گروه تقسیم شدند. که شامل: شم، کنترل، درمان با هایالگان، درمان با سلول های بنیادی، درمان با پلاسمای غنی از پلاکت، درمان با عصاره برگ گواوا، درمان با عصاره برگ گواوا و پلاسمای غنی از پلاکت، درمان با عصاره برگ گواوا و سلول بنیادی و درمان با عصاره برگ گواوا، سلول بنیادی و پلاسمای غنی از پلاکت می باشد. پس از 5 ماه از درمان، نتایج توسط رادیولوژیست و پاتولوژیست مورد ارزیابی قرار گرفت.یافته هاارزیابی رادیوگرافی و هیستوپاتولوژیکی، ترمیم غضروف مفصلی در گروه عصاره برگ گواوا همراه با سلول بنیادی و پلاسمای غنی از پلاکت را بهتر از گروه کنترل نشان داد (P<0.05).نتیجه گیریبه نظر می رسد عصاره هیدروالکلی برگ گواوا را به دلیل دارا بودن ترکیبات پلی فنولی زیاد، می توان به عنوان طب مکمل و جایگزین مناسب در استئوآرتریت پیشنهاد کردکلید واژگان: استئوآرتریت, پسیدیوم گواوا, پلاسمای غنی از پلاکت, سلول های بنیادی سینوویوم, رت}BackgroundOsteoarthritis (OA) recognized, as degenerative arthritis. The goals of scientific research of this study were to the evaluate the regeneration articular cartilage effect of injection of the hydroalcoholic extract of leave of Psidium Guajava with Stem Cell into the knee joints of rats.MethodsFirstly, ninety male rats were choosen, and collagenase type II was prepared for an intra-articular knee injection, for them to induce OA in the animal models. Then, they were divided into nine treatment groups. (N = 10): (a) Sham, (b) Control, (c) injection of hyaluronic Acid intra-articular, (d) Injections of Stem Cell intra-articular, (e) Injections of PRP, (f): Injections of the extract Psidium guajava leaves (g) Injections of the extract Psidium guajava leave with PRP, (h) Injections of the extract Psidium guajava leaves with Stem Cell (i) Injections of the extract Psidium guajava leaves with Stem Cell and PRP. After 5 months, the results were assessments by a Radiologist and Histopathologist.ResultsRadiological and histopathological findings were in which the improvement of the articular cartilage was better in the leaves with Stem Cell and PRP compared with the control group(PConclusionsIt seems that Psidium guajava leaves can be suggested as an effective complementary and alternative treatment for OA of the knee. BackgroundThe aim of this in vitro study was to assess antimicrobial efficacy of Acacia nilotica, Murraya koenigii (L.) Sprengel, Eucalyptus hybrid, Psidium guajava extracts, and their combination on Streptococcus mutans and Lactobacillus acidophilus.Materials And MethodsThe branches of four plants were collected, identified, and authenticated by a taxonomist. The plants were rinsed in water, healthy leaves were separated and shade dried over a period of 34 weeks. Soxhlet apparatus using ethanol was employed for extraction procedure. The combinations of plant extracts were prepared by mixing equal quantities of 10% solutions of each of these extracts. 0.2% chlorhexidine and dimethyl sulfoxide were used as positive and negative controls, respectively. The antimicrobial efficacy testing was done using agar well‑diffusion method under anaerobic conditions. The mean diameter of inhibition zone was computed and compared between different categories using one‑way analysis of variance and Tukeys post‑hoc test. A qualitative assay was carried out to identify the various phytochemical constituents in the plants. The data was assessed by SPSS version 20. The statistical significance was fixed at 0.05.ResultsAll the plants extracts and their combinations inhibited S. mutans and L. acidophilus. However, the quadruple combination of A. nilotica M. koenigii (L.) Sprengel Eucalyptus hybrid P. guajava produced the maximum inhibition zone (23.5 ± 2.2 mm) against S. mutans. Although, 0.2% chlorhexidine produced the highest inhibition zone against L. acidophilus (18.8 ± 1.2 mm), A. nilotica extract produced maximum inhibition among the various plant extracts and their combinations (14.1 ± 1.8 mm).ConclusionAll the individual plant extracts and their combinations were effective against S. mutans and L. acidophilus. These could be tried as herbal alternates to chlorhexidine. However, these in vitro results have to be further evaluated for any toxicity of the polyherbal combinations in animal models and effectiveness has to be assessed using in vivo studies on humans.Keywords: Acacia, nilotica, Eucalyptus, hybrid, Lactobacillus acidophilus, Murraya, koenigii (L.)
