Background
Due to frequent exposure to surface water and contact with animals, children represent a group susceptible to zoonotic diseases.
ObjectivesThe present study aims to determine the presence and prevalence of the main zoonotic agents in R. norvegicus populations in Tehran, Iran.
MethodsIn the present study, 100 R. norvegicus were captured within a time span of one year from five districts of Tehran, Iran. Fecal and blood samples were collected from rodents and serum was recovered after centrifugation. The presence of specific IgG antibodies against Leptospira spp. and Rabies virus was detected using a commercial qualitative rat ELISA kit. A conventional PCR assay was employed to detect the presence of Vibrio vulnificus in the commensal R. norvegicus population.
ResultsIn general, 80% (n = 80/100) and 20% (n = 20/100) of rats were males and females, respectively. The results of the ELSA assay showed that of the 100 R. norvegicus captured in Tehran, 7% (n = 7/100) and 1% (n = 1/100) were positive for Leptospira spp. and Rabies virus, respectively. Leptospira spp. revealed the highest frequency (20%; 4/20) among R. norvegicus collected from the eastern part of Tehran. Rabies virus was detected only from the southern (5%; 1/20) part of Tehran. Results of the PCR method showed that the percentage of the rats tested positive for V. vulnificus was 5%. Overall, the surveyed zoonotic microorganisms had the highest (n = 5/20; 25%) and lowest (n = 1/20; 5%) frequency rates in the eastern and northern parts of Tehran, respectively.
ConclusionsThe results accentuate the necessity of implementing rodent control programs and regular disinfection as well as avoiding contact with rodent populations in urban environments.

مجله طب جنوب، سال هجدهم شماره 6 (بهمن و اسفند 1394)، صص 1140 -1148زمینهلپتوسپیروزیس نوعی بیماری مشترک بین انسان و دام است، که توسط باکتری لپتوسپیزا اینتروگانس ایجاد می شود. ژن بیان کننده LipL21 یکی از ژن های شناسایی شده در باکتری لپتوسپیرا است که فقط در سویه های بیماری زا وجود دارد. هدف از این تحقیق کلونینگ و آنالیز تعیین توالی ژن کد کننده لیپوپروتئین سطحی LipL21 در 5 سرووار واکسینال باکتری لپتوسپیرا اینتروگانس در ایران می باشد.مواد و روش هاسرووارهای بیماری زای لپتوسپیرا اینتروگانس در محیط کشت اختصاصیEMJH و 10 درصد سرم خرگوش کشت داده شدند. بعد از استخراج DNA ژنومیک با استفاده از پرایمرهای اختصاصی PCR گذاشته شد و محصول PCR در درون وکتور E.Coli DH5&alpha انتقال داده شد. پلاسمیدهای نوترکیب جهت تعیین توالی ارسال گردید.یافته هاآنالیز توالی ژن lipL21 در مورد سرووارهای واکسینال داخلی و مقایسه آن ها با دیگر سرووارهای موجود در بانک ژنی نشان داد که 3 سرووار واکسینال سرجوهاردجو، کانی کولا و پومونا دارای 100 درصد تشابه با همدیگر می باشند و سرووار گریپوتیفوزا دارای بیشترین اختلاف با سرووارهای واکسینال می باشد. در کل نتایج نشان داد که این ژن یک ژن بسیار حفاظت شده در سرووارهای واکسینال بومی و سرووارهای موجود در بانک ژنی با درصد تشابه بالای 7/ 95 درصد می باشد.نتیجه گیرینتایج نشان داد که ژن lipL21 در میان سرووارهای واکسینال بسیار حفاظت شده (4/ 96 درصد< شباهت) می باشد. بنابراین ژن کد کننده پروتئین سطحی LipL21 می تواند به عنوان یک تست سرولوژیک مناسب با ویژگی و حساسیت بالا برای تشخیص درست و به موقع لپتوسپیروزیس در نمونه های کلینیکی و هم در آینده به عنوان کاندیدایی برای واکسن های زیرواحد موثر مورد استفاده قرار گیرد.

Gene expressing LipL21 is one of the genes identified in the bacterium, existing only in the pathogenic strains. The aim of this study was to cloning and analyzing the sequence of the gene encoding surface lipoprotein, LipL21, in five vaccinal leptospira serovars in Iran.Material And MethodsPathogenic Leptospira interrogans serovars were cultured in EMJH medium with 10% rabbit serum. After genomic DNA extraction, PCR with specific primers was employed and the resulting product inserted in a vector then transferred into E. Coli DH5&alpha. The recombinant plasmids were finally sent for sequencing.ResultsThe analysis of gene lipL21 in domestic vaccinal serovars and comparison of them with other serovars in the GenBank database revealed that three vaccinal serovars serjo hardjo, canicola and pomona had 100% similarity with each other and grippotyphosa serovar had the highest difference with the vaccinal serovars. In general, the results showed that this gene is a highly conserved gene in the domestic vaccinal serovars and serovars in the GenBank database with more than 95.7 percent similarity.ConclusionThese results showed that the gene, lipL21, is highly conserved in the vaccinal serovars (similarities > 96.4 %). Therefore, the gene encoding surface protein LipL21 can serve as a useful serologic test with high specificity and sensitivity for diagnosis of leptospirosis in clinical samples and in future as an effective subunit vaccine candidate to be used.Keywords: Leptospirosis, Leptospira Spp., lipL21 gene, cloning}
