    Sarcocystis is a mandatory intracellular parasite that is common between humans and animals, which causes huge losses in the livestock and meat industries. This study aimed to determine the prevalence of Sarcocystis infection in goats slaughtered in Tabriz slaughterhouse. For this purpose, the carcasses of 240 slaughtered goats were examined for the presence of macrocysts by eye observation and then by microscopic by dob-smear and digestive methods. Based on the results, 2.5% of the studied carcasses were diagnosed with macrocytic observation. In the microscopic examination by dob-smear and digestion methods, 35.83% and 100% respectively of the cases were positively diagnosed. There is a significant relationship between contamination and the studied method (p<0.05), which has higher validity digestion than the dob-smear and macroscopic method. There was also a statistically significant difference between the contamination rate among different muscles (p<0.05). As well, the highest microcyst contamination was observed in diaphragm muscles and the lowest microcyst contamination was observed in the heart muscle. This study showed the necessity of microscopic observation of the Sarcocystis contamination in the goat carcasses.

    Background

    We aimed to evaluate Sarcocystis contamination in conventional and industrial raw beef burger samples from butcheries and retail stores in Hamadan, western Iran.

    Methods

    Overall, 80 samples including 30 conventional and 50 industrial hamburgers were randomly obtained from different butcheries and supermarkets. All specimens were studied by digestion method following microscopic examination. Samples` genomic ribosomal DNA were amplified and nucleotide sequences were analyzed by BLAST for comparison with the sequences in the gene bank of the NCBI.

    Results

    Sarcocystis bradyzoites were detected in 46 of 80 (57.6%) samples. Positive specimens were included as 46 (57.6%) and 30 (37.5%) by digestion and molecular method, respectively. Differences between two studied (digestion and molecular) methods was statistically significant (P=0.00). Twenty-six (86.5 %) of 30 conventional beef burgers and 20 (40%) of 50 industrial burgers were positive for Sarcocystis sp. by digestion method. There was a significant difference between Sarcocystis infested conventional and industrial beef burgers (P=0.01).

    Conclusion

    The parasitic contamination of beef burgers implied a high level of infection in cattle. Felids as the definitive hosts for S. bovifelis urged on the improvement of the hygienic conditions of keeping and feeding livestock in order to reduce the infection. Molecular techniques confirm species in meat products with high sensitivity and distinguish it from human species.

    The genus Sarcocystis consists of intracellular coccidian protozoan parasites with the ability to invade muscle tissue and mature into sarcocysts, causing the zoonotic disease sarcocystosis. These parasites have an obligatory two-host life cycle, which correlates with prey-predator relationship. The distribution and prevalence of Sarcocystis in reptiles remains unclear, despite several previous reports. In this study, 54 faecal samples of various snake species and four faecal samples of several lizard species in Malaysia were examined for Sarcocystis through PCR amplification of the 18S rDNA sequence. Fourteen snake faecal samples were positive via PCR; however, only eight samples (14%) were found positive for Sarcocystis species, whereas four were positive for other genera and the identity of another three samples were unable to be determined. Further phylogenetic analysis of the 18S rDNA sequences revealed that the snakes were infected with either S. singaporensis, S. lacertae, or undefined Sarcocystis species which are closely related to either S. singaporensis or S. zuoi. Sarcocystis nesbitti infection was not identified in any of the infected snakes. This is the first report of identification of S. lacertae in the black-headed cat snake.
