مقدمه و هدف
بیماری قلبی عروقی، علت اصلی مرگ ومیر در جوامع امروزی است که از دلایل اصلی آن سبک زندگی بی تحرک و استفاده از رژیم های غذایی پرچرب و نامتعادل بیان شده است. هدف این مطالعه تعیین تاثیر 4 هفته تمرین هوازی با مکمل اکتاپامین بر بیان ژن NLRP-3 و سنتز پروتئین TRK-β در عضله قلب موش های صحرایی تغذیه شده با روغن های حرارت دیده مکرر بود.
روش کارتعداد 30 سر موش صحرایی نژاد ویستار 8 هفته ای با وزن 200 تا 250 گرم به صورت تصادفی به 5 گروه (6 تایی) شامل کنترل سالم (CO)، مصرف روغن های حرارت دیده مکرر (I)، مصرف روغن های حرارت دیده مکرر+ تمرین هوازی (H)، مصرف روغن های حرارت دیده مکرر+ مکمل اکتاپامین (B)، مصرف روغن های حرارت دیده مکرر+ تمرین هوازی+ مکمل اکتاپامین (C) تقسیم شدند. در طول دوره پژوهش روغن های حرارت دیده عمیق به صورت خوراکی (گاواژ، ml/kg10) به مدت 4 هفته، به موش های مورد آزمایش خورانده شد. 81 مول بر کیلوگرم اکتاپامین به صورت تزریق درون صفاقی به گروه های مکمل تزریق شد. موش های گروه تمرینی نیز به تمرین تردمیل با شدت متوسط در هفته ی اول 50% vo2max و در هفته ی آخر 65% vo2max پرداختند. جهت تحلیل داده ها از تحلیل واریانس یک طرفه و برای تعیین محل تفاوت از آزمون تعقیبی بن فرونی با استفاده از نرم افزار Graph pad prism نسخه 8 استفاده شد.
یافته هابیان ژن NLRP-3 در گروه تمرین نسبت به گروه کنترل که مکمل و تمرین دریافت نکرده بودند، کاهش معناداری داشت (0/05<p). دریافت مکمل اکتاپامین نیز موجب کاهش NLRP-3 شد (0/05<p). ترکیب تمرین و مکمل باهم باعث کاهش معنی دار بیان ژن NLRP-3 در مقایسه با گروه کنترل شد (0/05<p). تغییرات مقدار پروتئین TRK-β در گروه تمرین در مقایسه با گروه کنترل که مکمل و تمرین دریافت نکرده بودند، تفاوت معناداری نشان نداد (0/05>p). در گروهی که دریافت مکمل اکتاپامین نیز داشتند در مقایسه با گروه کنترل، مقادیر TRK-β تغییر نکرد و تفاوت معناداری نشان نداد (0/05>p). ترکیب تمرین و مکمل با هم موجب افزایش معنادار بیان ژن TRK-β در مقایسه با گروه تمرین شد (0/05<p).
نتیجه گیریاحتمالا تمرین هوازی همراه با مکمل اکتاپامین آثار مخرب روغن های حرارت دیده عمیق را بر بیان ژن وابسته به تخریب عضله قلب کاهش داده و موجب بهبود دگرگونی ساختار میوسیت می شود.

The aim of this study was to determine the effect of 4 weeks of aerobic exercise with octapamine supplementation on NLRP-3 gene expression and TRK-β protein synthesis in the heart muscle of rats fed with deep-fried oil.
Materials and Methods30 male Wistar rats weighing 200-250 were randomly divided into 5 groups of 6: health control (CO), deep-fried oil (I), deep-fried oil + aerobic exercise (H), deep-fried oil + octapamine, supplementation (B), and deep-fried oil + octapamine supplementation + aerobic exercise (C). During the research period, 10 ml/kg deep-fried oil was given to the rats via gavage for 4 weeks. 81 mol/kg octapamine was intraperitoneally injected to the supplement groups. The rats in the exercise group also went through moderate intensity training in the first week (50% vo2max) and last week (65% vo2max) with treadmill. One way analysis of variance was used to analyze the data and Bonferroni test was used to determine the difference, using Graph pad prism version 8.
ResultsNLRP-3 gene expression was significantly reduced in the exercise group compared to the control group who did not receive any supplements and exercise (p<0.05). Octapamine supplementation also decreased NLRP-3 (p<0.05). The combination of exercise and supplementation significantly reduced NLRP-3 gene expression compared with the control group (p<0.05). Changes in the amount of TRK-β protein in the exercise group did not show a significant difference compared to the control group that did not receive any supplements and exercise (p>0.05). In the group that also received octapamine supplementation, compared to the control group, TRK-β values did not change and did not show a significant difference (p>0.05). Combining exercise and supplementation significantly increased TRK-β gene expression compared to the exercise group (p<0.05).
ConclusionAerobic exercise training with octapamine supplementation may reduce the destructive effects of deep-heated oils on the expression of the gene associated with myocardial degradation and improve remodeling in myocytes.

Background3,4- methylenedioxymethamphetamine (MDMA) is a chemical derivative of amphetamine that can induce learning and memory impairment. Due to the effect of neurotrophins on memory and learning, the impact of MDMA was evaluated on the brain - derived neurotrophic factor (BDNF), neurotrophin- 4 (NT-4), and tropomyosin- related kinase B (Trk- β) expression in the hippocampus.
MethodsIn this study, 20 adult male Wistar rats (200-250 g) received saline (1 mL) or 10 mg/kg of MDMA intraperitoneally as single or multiple injection for 2 consecutive days per week for 2 months. Expression of BDNF, Trk-β, and NT4 were assessed using Western blotting and RT PCR methods.
ResultsOur results revealed that the expression of BDNF, Trk- β, and NT4 proteins and genes significantly decreased in MDMA groups compared to the sham group (pConclusionThe results of the present study suggest that ecstasy administration may downregulate the expression of BDNF, Trk- β, and NT-4 in hippocampus, which is more extensive in case of acute treatment.
