Evaluation of the effect of Myoinositol-Sodium citrate and serum on developmental competency of zona-free bovine cloned embryos

Abstract:
The aim of this study was to improve culture conditions of bovine embryos produced by the Hand-made cloning (HMC) method with the use of serum and myoinositol. For this purpose, after oocyte in vitro maturation followed by elimination of the zona pellucida, bovine oocytes were enucleated by separation of the metaphase spindle protrusion using a very thin pulled Pasteur pipette. After attachment of adult fibroblasts as donor cell into the enucleated oocyte and subsequent fusion and activation, reconstructed embryos were cultured by Well of the Well (WOW) method in six groups, including IVCSOF, IVCSOF-CSS, IVCSOF-ci, IVCSOF-ciCSS, IVCSOFci and IVCSOFci-CSS. In these groups, the embryos were cultured in IVCSOF with or without myoinositol-sodium citrate media until the third and then CSS and/or myoinositol-sodium citrate was added.
