Abstract:
Introduction
Tissue engineering is a new method for replacing tissue fragments degraded by biodegradable polymers, which is provided as a three-dimensional scaffold for the growth and propagation of stem cells. In this study, chitosan was used to evaluate the migration and proliferation of fibroblasts in the presence of hyaluronic acid.Material and
Method
In this study, scaffold chitosan was prepared for the growth of fibroblastic cells. Then, for further studies, scaffolds were divided into the following groups: Group 1: Chitosan scaffold with hyaluronic acidGroup 2: Chitosan without hyaluronic acid scaffold
Group 3 (control 1): Hyaluronic acid fibroblast cell
Control group 2: Hyaluronic acid-free fibroblastic cell
The human foreskin was prepared and the fibroblast cells in the dermal layer were centrifuged after separation, and the cells were transferred to the culture flasks with DMEM medium and stored in a CO2-encoded incubator. After several passage cells, 10,000 cells were transferred to 96 wells containing DMEM medium and MTT and DAPI staining were investigated and the fibroblasts were infiltrated on the chitosan scaffold. The obtained results were analyzed by SPSS software, ANOVA and post hoc Tukey test.
Results
The results showed that the mean survival rate of chitosan without hyaluronic acid scaffold in 24 hours was significantly higher than hyaluronic acid and chitosan groups with hyaluronic acid (P Conclusion
The results of this study showed that chitosan scaffolds with regard to hydrophilicity, better biocompatibility with fibroblast cells, suggest that chitosan may be synthesized in structures that are synthesized to repair affected areas of skin. Have a proper and complementary role. Keywords:
