رپلیزوم یوکاریوت ها یک کمپلکس چند جزئی بوده که همانندسازی DNA را با سرعت حدود Kb 2-3 در دقیقه انجام می دهد. این مسئله نشان می دهد که ساختار کروماتین به میزان 15-10 نوکلئوزوم در دقیقه و در جلوی هر رپلیزوم فعال متلاشی می شود. برای تشکیل یک الگوی کروماتینی مشابه بر روی DNA های تازه شکل گرفته، هیستون ها و یک سری از فاکتورهای متصل شونده به کروماتین، از روی رشته مادری به روی رشته های دختری انتقال داده می شوند. به علاوه، هیستون های جدید نیز برای ایجاد و حفظ تراکم نوکلئوزومی موردنظر تجمع یافته و اصلاحات هیستون های جدید نیز بر مبنای الگو گرفتن از هیستون های مادری و قدیمی انجام می گیرد. به طور کلی بررسی اینترکشن های میان اجزای رپلیزوم با پروتئین های کروماتین، به درک صحیح نحوه پیشرفت چنگال همانندسازی و ارتباط آن با کروماتین کمک می نماید. در این مطالعه به بررسی مکانیسم های تنظیمی در همانندسازی کروماتین و فرآیند حفظ اپینوم پرداخته می شود.

کلبسیلاپنومونیه از عوامل شایع عفونت های ناشی از آنزیم های بتالاکتامازی در بیمارستان ها است. هدف از این مطالعه تعیین میزان فراوانی ژن های مولد مقاومت آنتی بیوتیکی TEM و IMP-1در سویه های کلبسیلاپنومونیه است.
مواد و روش ها94 نمونه کلبسیلاپنومونیه جهت تایید فنوتیپی ESBLs مورد استفاده قرار گرفتند. میزان مقاومت آنتی بیوتیکی جدایه ها مشخص و میزان شیوع ژن های TEM و IMP-1با روش واکنش PCR تعیین شدند.
یافته ها6/77% جدایه ها دارای فنوتیپ ESBL و 3/22% نمونه ها فاقد فنوتیپ ESBL بودند. نتایج نشان داد که 1/4% نمونه ها دارای ژن IMP-1 و 8/43% نمونه ها دارای ژن TEM هستند. 8/43% نمونه ها نیز به طور مشترک واجد هر دو ژن TEM و IMP-1 بودند.
نتیجه گیریبا توجه به این که ژن های TEMو IMP-1 به طور مشترک در تعداد زیادی از نمونه های مقاوم وجود داشتند، استفاده از راه کارهای درمانی مناسب، تجویز درست و منطقی آنتی بیوتیک ها توسط پزشکان در این زمینه پیشنهاد می گردد.

قارچ لنتینولا ادودز (شی تاکه)، به دلیل وجود متابولیت های ثانویه معجزه آسای موثر در درمان سرطان اهمیت دارویی به سزایی دارد. در این مقاله تلاش می شود تا با انتخاب محیط مناسب جهت رشد بهینه و شناسایی و بررسی عوامل تاثیر گذار بر تولید متابولیت های ثانویه، تولید ترکیب های آنتی اکسیدان بهینه سازی گردد.
مواد و روش هاابتدا بانک سلول های قارچی در محیط جامد تهیه شد و سپس نمونه ها به محیط مایع انتقال داده شد. عصاره الکلی از نمونه ها تهیه شد. جهت بررسی فعالیت آنتی اکسیدانی از دو روش 2و2 دی فنیل 1- پیکریل هیدرازیل (DPPH) به منظور بررسی قابلیت حذف رادیکال های آزاد و قدرت احیاءکنندگی فریک (FRAP) به منظور بررسی قابلیت احیای یون فریک استفاده شد. جهت اندازه گیری میزان پلی ساکارید و فنل نیز به ترتیب از روش های فنول-سولفوریک اسید و Folin-Ciocalteu استفاده شد. میزان سمیت سلولی عصاره نیز با تست MTT سنجیده شد.
یافته هابهینه سازی خاصیت آنتی اکسیدانی قارچ شی تاکه با ارائه محیط غنی شده حاوی عصاره چوب بلوط انجام شد و میزان بازدارندگی رادیکال DPPH آن 24/5 برابر و میزان قدرت احیای یون فریک آن 9/1 برابر افزایش یافت. هم چنین سمیت سلولی بسیار مناسب (IC50 برابر 8 میکروگرم بر میلی لیتر) را بر روی رده سلولی MCF-7 نشان داد.
نتیجه گیریاین مطالعه نشان داد که عصاره چوب بلوط دارای خاصیت تحریک کنندگی بر تولید مواد آنتی اکسیدان در قارچ شی تاکه است. پس می تواند به عنوان محرک مناسبی برای افزایش میزان آنتی اکسیدان ها درسلول های قارچ شی تاکه در محیط های کشت به کار رود.

آلوپسیا آره آتا (AA) یک بیماری چند عاملی است که با ریزش موی سر به خصوص در ناحیه سر شناسایی می شود که 5 میلیون نفر را تحت تاثیر خود قرار داده است. نواحی خاصی ازHLA ها شناسایی شده که با پاتوژنز بیماری آلوپسیا آره آتا در ارتباط است. در این تحقیق اثر فراوانی آلل HLA-DQB2 در بروز بیماری آلوپسیا آره آتا در جامعه بیمار و کنترل بررسی شده است.
مواد و روش هااین مطالعه شامل 30 بیمار AA (13 زن و17 مرد، میانگین سنی 3/26) و 15 نفر کنترل سالم (5 زن و 10 مرد با میانگین سنی 30) است DNA از نمونه های خون با استفاده از روش DNG plus استخراج شد و با روش PCR-SSP تشخیص HLA(HLA-DQB2) انجام شد. علاوه بر این ارتباط بین این آلل HLA با فاکتورهای دموگرافیک بررسی شد.
یافته هافرکانس آلل HLA-DQB2 در مقایسه با افراد کنترل (7/26 %) در بیماران به طور چشم گیری بالاتر بوده است. از بین فاکتورهای دموگرافیک، فقط در معرض آفتاب بودن با وجود آللHLA-DQB2 ارتباط معنی داری نشان داده است. هم چنین هیچ ارتباط معنی داری بین ترس، استرس، سابقه خانوادگی در بیمارانی با بیماری AA با وجود این آلل یافت نشده است.
بحثداده های ما هیچ ارتباطی بین آلل HLA-DQB و وقوع بیماری آلوپسیا آره آتا نشان نمی دهند، گرچه میزان وجود آن در بیمارانAA به طرز چشم گیری نسبت به افراد سالم بیش تر است.

استافیلوکوکوس اورئوس عامل عفونت های مختلفی در انسان است. عفونت باکتریایی دهانه رحم می تواند در تغییر پارامترهای اسپرماتوزوا نقش داشته باشد و ممکن است منجر به ناباروری در زنان شود. هدف این تحقیق بررسی حضور سوپر آنتی ژن های استافیلوکوکوس اورئوس در زنان نابارور با علت ناشناخته بود.
مواد و روش هاصد نمونه واژینال از زنان نابارور بدون علت مشخص جمع آوری شد. نمونه های مثبت استافیلوکوکوس اورئوس با حداقل سه مقاومت آنتی بیوتیکی، به وسیله PCR بررسی شدند. جهت بررسی تاثیر عفونت بر ناباروری، پس از انکوباسیون نمونه های تازه اسپرم با سویه های دارای یکی از ژن های seg , sei و یا Tsst-1 ، پارامترهای اسپرم اندازه گیری شد.
یافته هاباکتری استافیلوکوکوس اورئوس از 52 نمونه جدا شد. پس از انجام تست آنتی بیوگرام، 19 نمونه استافیلوکوکوس اورئوس مقاوم به سه یا بیش تر آنتی بیوتیک، شناسایی شدند. فراوانی ژن های Tsst-1، sei و seg به ترتیب 3/26%، 1/21% و 5/10% بود. در 11 سویه (9/57%) ، ژن های seg , sei و یا Tsst-1 حضور داشتند. هم چنین در 5/10٪ نمونه ها، هر دو ژن sei و seg وجود داشت. یک سویه (3/5%) دارای هر سه ژن seg , sei و Tsst-1، توانایی اگلوتیناسیون 50٪ اسپرم های تازه را بدون تغییر قابل ملاحظه ای در حرکت و قابلیت زنده ماندن داشت.
نتیجه گیرینتایج نشان داد استافیلوکوکوس اورئوس عامل عفونت واژ ینال می تواند سبب آسیب به مورفولوژی و تحرک اسپرم شود. در نتیجه، درمان واژینوز باکتریال جهت پیشگیری از ناباروری ها با علل ناشناخته در زوجین نابارور می تواند نقش موثری ایفا نماید.

مشخصه التهاب در کبد تهاجم سلول های التهابی است. این سلول ها با ترشح سایتوکین ها التهابی مثل اینترلوکین-6 و فاکتور توموری نکروز آلفا باعث پیش روی بیماری و آسیب بیش تر کبد می شوند. در این مطالعه اثر ضدالتهابی عصاره زنجبیل و آب لیموترش بر فاکتور التهابی IL-6 در شرایط سمیت کبدی القا شده توسط تیواستامید در موش های صحرایی مورد برسی قرار گرفت.
مواد و روش هابه منظور انجام آزمایش از 55 موش آزمایشگاهی هم سن با وزن تقریبی 250-200 گرم تهیه شده از دانشگاه اصفهان استفاده شد. نمونه ها به طور تصادفی به 9 گروه تیمار، یک گروه کنترل و یک گروه شاهد تیواستامید (کنترل مثبت) (هر گروه با 5 موش) تقسیم شدند. پس از تیمار هر گروه با دوزهای مختلف عصاره زنجبیل و آب لیمو خون گیری از قلب تحت شرایط بی هوشی انجام شد و به منظور اندازه گیری فاکتور التهابی IL-6 از تست الایزا استفاده شد. در نهایت مقایسه آماری میانگین مقادیر سرمی IL-6 در گروه های مختلف با استفاده از نرم افزار SPSS V.20و آزمون های آنالیز واریانس یک طرفه (ANOVA) انجام شد.
یافته هانتایج مطالعه حاضر نشان داد، مصرف عصاره زنجبیل و آب لیمو ترش موجب کاهش معنی دار سطح سرمی IL-6 شده است. در خصوص IL-6 سطح سرمی قبل از تیمار و بعد از تیمار با تیواستامید بیش ترین میزان کاهش در گروه های تیمار ترکیبی، زنجبیل 500 میلی گرم بر کیلوگرم و آب لیمو 100 درصد بود. کاهش سرمی IL-6در گروه های مورد مطالعه وابسته به دوز بود (P<0.001).
بحثتیواستامید باعث مسمومیت و در نهایت التهاب کبد می شود. عصاره زنجبیل وآب لیمو بخاطر داشتن جینجرول و فلاونوئیدها که خاصیت ضدالتهابی دارند به صورت ترکیبی می تواند پارامترهای کبدی را در التهاب کبدی بهبود بخشد.
نتیجه گیریعصاره زنجبیل و آب لیمو ترش خاصیت ضدالتهابی دارند و به صورت ترکیبی می تواند پارامترهای کبدی را در التهاب کبدی بهبود بخشد و با کاهش سطح سرمیIL-6 باعث کاهش التهابات می شوند.

هدف از این تحقیق بررسی تئوری تاثیر پلی مورفیسم تک نوکلئوتیدیrs141831067 ژن دی هیدروفولات ردوکتاز بر پاسخ درمانی افراد مبتلا به سرطان پروستات به داروی متوترکسات و مطالعه فراوانی این پلی مورفیسم در جمعیت ایرانی است.
مواد و روش هادر این پژوهش خون محیطی از 50 فرد سالم و 50 فرد مبتلا به سرطان پروستات گرفته شد. سپس DNA ژنومی استخراج و جداسازی شد. برای بررسی وجود پلی مورفیسم موردنظر در افراد مورد مطالعه، از تکنیک ARMS-PCR استفاده شد. در نهایت بررسی آماری نتایج به دست آمده به واسطه بررسی فراوانی آللی، درجه هتروزیگوتی، آنالیز فاکتور PIC و بررسی تعادل هاردی واینبرگ صورت پذیرفت.
یافته هامقایسه فراوانی آلی برای پلی مورفیسم rs141831067 در دو گروه سالم و بیمار نشان داد که فراوانی آللی این مارکر در گروه بیمار به صورت معناداری بیش تر از گروه سالم است (002/0P-Value=). به علاوه آنالیزهای بیوانفورماتیکی برهم کنش آنزیم دی هیدروفولات ردوکتاز و داروی متوترکسات نشان داد که پلی مورفیسم تک نوکلئوتیدیrs141831067 ژن دی هیدروفولات ردوکتاز در برهم کنش با دارو تغییر ایجاد می کند و موجب خروج نسبی متوترکسات از جایگاه فعال دی هیدروفولات ردوکتاز می شود.
نتیجه گیریچند شکلی تک نوکلئوتیدی rs141831067 موجود در ژن دی هیدروفولات ردوکتاز در احتمال بروز سرطان پروستات نقش مهمی دارد و این چند شکلی تک نوکلئوتیدی می تواند باعث بروز مقاومت دارویی به متوترکسات در سرطان پروستات گردد.

صفحات 93-101سابقه و هدفآرتریت باکتریایی یکی از انواع آرتریت های شناخته شده است که به سرعت باعث تخریب مفاصل می شود. از بین باکتری های مولد آرتریت عفونی، مایکوباکتریوم تربرکلوزیس (MTB) یکی از باکتری هایی است که می تواند به طور نادر ایجاد کننده این عارضه باشد. هدف از این مطالعه بررسی وجود مایکوباکتریوم تربرکلوزیس در مایع مفصلی بیماران مبتلا به آرتریت از طریق روش PCR است.مواد و روش هااین مطالعه بر روی مایع مفصلی 70 بیمار مراجعه کننده به بیمارستان شریعتی تهران که دچار آرتریت بوده اند صورت گرفت. DNA از مایع مفصلی این 70 بیمار با روش فنل کلروفرم استخراج شد. پرایمر، برای ژن هدف IS6110 انتخاب و بعد از تایید حساسیت و اختصاصیت، تست PCR بهینه بر روی نمونه ها انجام و مورد ارزیابی قرار گرفتند.یافته هاتست PCR بهینه و آمپلیکون bp 317 به وسیله آگاروزژل الکتروفورز %5/1 مشاهده گردید. حد تشخیص تست 100 و با ویژگی صد درصد ارزیابی گردید. DNA مایکوباکتریوم توبر کلوزیس در 4 نمونه مایع مفصلی بیماران مبتلا به آرتریت (%7/5) دیده شد.نتیجه گیریاولین قدم در درمان بیماری، تشخیص درست و سریع آن است. مایکوباکتریوم توبرکلوزیس مشخصه هایی از قبیل رشد کند دارد که شناسایی این باکتری را از طریق کشت و تست های بیوشیمیایی مشکل و طاقت فرسا می کند. نتایج این مطالعه موید آرتریت های مایکوباکتریوم توبرکلوزیس موجود است که تشخیص سریع و به موقع آن ها از طریق PCR امکان درمان صحیح آن ها را مهیا می نماید.کلیدواژگان: آرتریت، PCR، مایکو باکتریوم تربرکلوزیس، مایع مفصلی، تشخیص

The eukaryotic replisome is a multi-component complex that drives DNA replication with a speed of approximately 2 to 3 kb per min. This implies that chromatin is disrupted at a rate of around 10 to 15 nucleosomes every minute a head of each active replisome. To reproduce a similar chromatin environment on new DNA, histones and other chromatin-bound factors are transferred from the parental strand to the daughter strands. In addition, new histones are incorporated to maintain nucleosome density, and their modification signature should be assimilated to nearby old histones in the local chromatin environment. In general interaction between the components of the replisome and chromatin proteins can help to understand the proper way of improving the replication fork and its relationship to chromatin. In this study, the regulatory mechanism of chromatin replication and epigenome maintenance are evaluated.

Klebsiella pneumoniae is a common cause of β-lactamase-associated infections in hospitals. The present study aimed to determine the frequency of antibiotic resistance genes in Klebsiella pneumoniae strains producing IMP-1 and TEM β-lactamase.
Materials and methodsThe present research identified 94 samples of K. pneumoniae, using antibiogram for the phenotypic confirmation of ESBLs. The antibiotic resistance of the isolates and the prevalence of TEM and IMP-1 genes were determined using PCR method.
FindingsOf 94 samples, 77.6% were ESBL-positive and 22.3% ESBL-negative. A total of 4.1% of the samples carried the IMP-1 gene and 43.8% the TEM gene, while 43.8% of the samples carried both genes.
ConclusionGiven that TEM and IMP-1 genes were commonly present in a large number of the resistant samples, physicians are recommended to use therapeutic measures properly, and to prescribe antibiotics rationally.

Lentinula edodes (Shiitake) mushroom have a significant pharmacological importance due to the presence of secondary metabolites with a miraculous effect on cancer treatment. The aim of this study was optimizing the production of antioxidant compounds by selecting the appropriate growth medium and investigating the affecting factors on the production of secondary metabolites.
Material and methodsThe bank of fungal cells was prepared in a solid medium and then the samples were transferred to the liquid medium. Alcoholic extract of the samples was prepared. For antioxidant activity, two methods were used: 1- DPPH assay to investigate the capability of removing free radicals and 2- FRAP assay to investigate the ability of ferric ion reduction. Polysaccharide and phenol levels were measured using phenol-sulfuric acid and Folin-Ciocalteu methods, respectively. The cytotoxicity of the extract was measured by MTT assay.
ResultsOptimization of antioxidant properties of Shiitake was done by introducing oak extract-enriched medium and the scavenging activity of DPPH radical and reduction potential for ferric ion increased by 5.24 and 1.9 times, respectively. Also, it has showed an appropriatecytotoxicity (IC50 value of 8 μg/ml) on the MCF-7 cell line.
ConclusionThe results represented that oak wood crude extract could be used in Shiitake growth medium as an appropriate elicitation of increasing antioxidant activity.

Alopecia Areata (AA) is a common immune-mediated disease which considered as the second most common cause of hair loss in humans. This hair loss targets hair follicles, and has a genetically complex inheritance. HLA genes such as HLA class II can influence the development of AA. The aim of the study was to investigate whether HLA-DQB2 allele is associated with AA.
Materials and MethodsAlopecia Areata patients and control subjects were enrolled in this study. A total of 30 AA patients (13 female and 17 male with mean age 26.3±12.5) and 15 healthy controls (5 Female and 10 Male with mean age 30 ±5.88) were included and analyzed in a case–control study. Genomic DNA was prepared using DNG plus method. Polymerase chain reaction with sequence specific primers technique (PCR-SSP) used to detect HLA-DQB2. Association of HLA-DQB2 allele with family history, age of onset, and stress were assessed by logistic regression analysis.
ResultsHLA-DQB2 allele did not show a significant association with susceptibility to AA and had a frequency of 56.7% in AA patient vs 26.7% in healthy controls (OR = 3.596, 95% CI = 0.929-13.916, p value = 0.064). Furthermore, this study did not show any association of HLA-DQB2 allele with family history (OR =0.3, 95% CI = 0.06 – 2.32, p value = 0.3), phobia (OR = 1.57, 95% CI = 0.38-6.4 p value = 0.5) and stress (OR = 1.36, 95% CI = 0.28-6.48 p value = 0.6), family history (OR=0.4, 95%CI=0.06, p value=0.3). the significant association found with sunlight exposure with HLA-DQB2 allele in AA (OR=4.46,95%CI=0.95-20.83, p value=0.05).
ConclusionOur data do not show a correlation between the HLA-DQB2 allele and occurrence of AA and clinical data in Iranian population. To our knowledge this is the first study to frequency of HLA-DQB2 allele in Iranian AA patients.

Staphylococcus aureus can be caused different infections in human. Bacterial infection of the cervix can be effective in spermatozoid parameters change and may be come infertility in women. The aim of this study was identification of Staphylococcus aureus super-antigenes from infertile women with unexplained infertility.
Materials and MethodsHundred vaginal samples were isolated from unexplained infertile women. Positive samples for Staphylococcus aureus with at least three antibiotic resistances were analyzed by PCR. For identification of infection effect on infertility, after incubation of fresh sperm samples ubated with the Staphylococcus aureus consist one of seg, sei or Tsst-1 genes, were measured sperm parameters.
ResultsStaphylococcus aureus was selected from 52 samples. After antibiogram test, 19 Staphylococcus aureus samples were identified with resistance to three or more antibiotics. Frequency of Tsst-1, sei and seg genes were, (26.3%) ,(21.1%) and (10.5%), respectively. There were in 11 (57.9%) isolates, sei, seg or Tsst-1 genes. As also, 10.5% of isolates had both of seg and sei genes. One strain (5.3%) with seg, sei and Tsst-1 genes, could be agglutinated 50% of fresh sperms without any significant changes on motility and viability.
ConclusionThe results were indicated Sphylococcus aureus that is caused vaginal infection could be damage the morphology and motility of male spermatozoa. In summary, treatment of bacterial vaginosis could be effective for prevention of idiopathic infertilities in infertile couples rates.

The characteristic of inflammation in the liver is the invasion of inflammatory cells. These cells, with the secretion of inflammatory cytokines such as IL-6 and the alpha-induced tumor necrosis factor, contribute to the disease and damage to the liver. In this study, the anti-inflammatory effect of ginger and lemon juice on an inflammatory factor of IL-6 in liver toxicity induced by thioacetamide in rats was investigated.
Material and methodsIn order to perform the experiment, 55 mouse with an approximately weight of 250-200 grams collected from Isfahan University were used. The samples were randomly divided into 9 treatment groups, one control group and one thioacetamide control group (positive control) (each group with 5 mice). After treatment, each group received different doses of ginger extract and lemon juice, blood samples were taken from the heart under anesthetized conditions and ELISA technique was used to measure the inflammatory factor of IL-6. Finally, the statistical comparison of mean serum IL-6 levels in different groups was performed using SPSS V.20 software and one-way ANOVA test.
ResultsThe results showed that consumption of ginger extract and lemon juice significantly decreased serum IL-6 levels. The highest reduction of IL-6 serum levels after thioacetamide treatment was shown in 500 mg/kg ginger and 100% lemon juice combination treatment. This reduction was dose-dependent (P <0.001).
ConclusionThioacetamide causes poisoning and ultimately inflammation of the liver. Extract of ginger and lemon juice in combination with ginger and flavonoids that have anti-inflammatory properties can improve liver parameters in liver inflammation. Ginger extract and sour lemon juice have anti-inflammatory properties and can improve liver parameters in liver inflammation and decrease inflammation by decreasing serum levels of IL-6.

The aim of this study was to investigate the effect of rs141831067 Single nucleotide Polymorphism existence on therapeutic response of patients diagnosed with Prostate Cancer to Methotrexate and investigation of the polymorphism Frequency in Iranian population.
Material and methodsIn this study, peripheral blood samples were taken from 50 healthy individuals and 50 patients suffering from prostate cancer. Then genomic DNA of each sample was extracted and isolated. ARMS-PCR and direct sequencing techniques were used to determine the presence of the polymorphism in populations. Finally, the statistical analysis of the results was performed by analyzing allelic frequency, heterozygosity, PIC analysis and Hardy Weinberg equilibrium analysis.
ResultsA comparative study of rs141831067 polymorphism existence showed that the allele frequency of this marker was significantly higher in the patients relative to healthy group (P-value = 0.002). In addition the bioinformatics analyzes of the interactions of the dihydrofolate reductase enzyme and methotrexate showed that existence of the polymorphism may alter the interaction patterns and result in the relative displacement of methotrexate position from the active site of dihydrofolate reductase.
ConclusionRs141831067 Single nucleotide Polymorphism in Dihydrofolate reductase gene plays an important role in the prostate cancer occurrence and this Single nucleotide Polymorphism can cause drug resistance to methotrexate in prostate cancer.

Pages 93-101Aim andBackgroundBacterial arthritis is one of the arthritis diseases known that can rapidly cause joint damage. Among the bacteria causing septic arthritis, Mycobacterium tuberculosis (MTB) is one of those that rarely produce arthritis. The aim of this study is to examine the presence of MTB in synovial fluid of patients with arthritis using Polymerase Chain Reaction (PCR)Materials and MethodsThis study was carried out on 70 synovial fluid samples gathered from Shariati Hospital. DNA was extracted using Phenol-Chloroform standard extraction technique. PCR test optimized on the basis of IS6110 target gene. Samples were analyzed by PCR test after evaluation of specificity and sensitivity of PCR.RESULTPCR test was optimized and the 317-bp amplicon detected by 1.5% agarose gel electrophoresis. Limit of detection (LOD) was estimated as 100 copy/Reaction. MTB DNA was detected in 4 (5.7%) synovial fluid samples of patients with arthritis.
CONLUSION: First step in treatment is rapid and accurate diagnosis. MTB have some characteristics including slow generation making the identification difficult and exhausting through culturing and biochemical tests that sometimes leads to ambiguous results. Results of this study confirm that MTB could play a role in bacterial arthritis and rapid diagnosis using PCR provides us with accurate treatment.Keywords: Mycobacterium tuberculosis, Diagnosis, Synovial, PCR
