    سیاری از مطالعات انواع مختلفی از افزودنی های خوراک را مورد بررسی قرار داده اند و در بین این افزودنی ها مواد هیومیک (HS) نیز در جیره دام و طیور مورد استفاده قرار گرفته اند. از آن جایی که مواد هیومیک از تجزیه مواد آلی حاصل می شوند، به طور معمول در طبیعت حضور دارند و در خاک و آب طبیعی یافت می شوند. اجزای فعال HS شامل هیومیک اسید (HA)، هیومس، اولمیک اسید، فولویک اسید، هیومین و برخی میکرو المنت های خاص است. هیومیک اسید به طور گسترده به عنوان یک محرک رشد جایگزین آنتی بیوتیک ها در بهبود سلامت و کارآیی طیور استفاده شده است. علاوه بر این، اضافه کردن مواد تجاری به عنوان یک منبع HS به آب آشامیدنی و یا جیره، سبب بهبود مصرف خوراک، بازده خوراک و افزایش وزن جوجه های گوشتی و همچنین بهبود وزن تخم مرغ، توده تخم مرغ و تولید تخم مرغ در مرغ های تخم گذار شده است. این مقاله مروری به توصیف کاربردهای مفید و جنبه های اطلاعاتی اخیر مرتبط با HA شامل نحوه عملکرد و کاربردهای سودمند مختلف آن در بهبود تولید و حفظ سلامت دام و طیور می پردازد.

    پیشینه:

    استفاده از پپتید ناتریورتیک تیپ C (CNP) به همراه سیستئامین در شرایط پیش از بلوغ آزمایشگاهی (IVM) می تواند جهت فراهم کردن یک سیستم pre-IVM موثر، کمک کند.

    هدف

    این مطالعه جهت بررسی اثرات CNP در ترکیب با سیستئامین بر بلوغ میوتیک و قابلیت تکامل تخمک های گاو در شرایط آزمایشگاهی طراحی شد.

    روش کار

    تخمک ها به مدت 6 ساعت در محیط کشت pre-IVM غنی شده با nM  100 از CNP با یا بدون µM 100 سیستئامین، کشت و به دنبال آن به مدت 24 ساعت با روش استاندارد IVM کشت داده شدند. تخمک هایی که به مدت 24 ساعت به وسیله روش IVM استاندارد (بدون pre-IVM) بالغ شده بودند، به عنوان گروه شاهد انتخاب شدند. درصد تخمک های متوقف شده در مرحله وزیکول ژرمینال (GV) در هر یک از گروه های تیمار، در ساعت صفر پس از انجام IVM، ارزیابی و سپس میزان بلوغ هسته ای با بررسی درصد تخمک های رسیده به مرحله متافاز ІІ)ІІ (M در ساعت 24 پس از انجام IVM، اندازه گیری شد. پس از 24 ساعت از انجام IVM، تخمک ها تحت لقاح آزمایشگاهی (IVF) و کشت آزمایشگاهی جنین (IVC) قرار گرفتند. نرخ کلیواژ، 24 ساعت پس از تلقیح ارزیابی شد. نرخ تشکیل بلاستوسیت ها نیز 8 روز پس از IVF اندازه گیری شد. علاوه بر این گلوتاتیون (GSH) داخل تخمکی و همچنین مقدار گونه های فعال اکسیژن (ROS) برای هر گروه تیمار در ساعت 0 و 24 پس از انجام IVM اندازه گیری شد.

    نتایج

    هیچ تفاوتی بین گروه های تیمار pre-IVM همراه با CNP و شاهد از نظر نرخ بلوغ تخمک های گاوی وارد شده به مرحله M II، کلیواژ و نرخ تشکیل بلاستوسیت ها پس از لقاح آزمایشگاهی و همچنین سطوح داخل تخمکی GSH مشاهده نشد. جالب توجه اینکه، حضور سیستئامین در طی کشت pre-IVM همراه با CNP به طور معنی داری سبب بهبود نرخ تکامل جنین ها به مرحله بلاستوسیست، پس از لقاح و بلوغ آزمایشگاهی شد‎‏، به علاوه سبب افزایش سطح GSH و کاهش ROS در تخمک های گاو گردید. نتیجه گیری: بهبود قابلیت تکامل تخمک های گاوی در روش IVM به وسیله pre-IVM همراه با ترکیب CNP و سیستئامین ممکن است ناشی از افزایش قابلیت دفاع آنتی اکسیدانی باشد. بنابراین، این رویکرد ممکن است گزینه مناسبی برای فراهم سازی یک سیستم pre-IVM باشد.

    پیشینه: ارزیابی شاخص های سوخت و ساز انرژی در گاوهای هولشتاین دوره میانی شیردهی جهت نظارت بر وضعیت سلامتی ضروری است.

    هدف: هدف از این مطالعه بررسی اثرات افت کم (75/0≤) در مقابل زیاد (75/0>) در نمره وضعیت بدنی (BCS) تا روز 60 پس از زایمان بر شاخص های سوخت و ساز انرژی در دوره میانی شیردهی گاوهای هولشتاین بود. روش کار: بیست و هشت راس گاو هولشتاین از روز زایمان تا روز 120 شیردهی وارد مطالعه شدند. نمونه های خون کامل در روزهای 60، 90 و 120 شیردهی (DIM) اخذ شد. سطح انسولین، گلوکز، اسیدهای چرب غیر استریفیه (NEFA) و بتا-هیدروکسی بوتیرات (BHB) سرم اندازه گیری شد. نتایج: در گاوهای با افت کم BCS (گروه LoD)، انسولین در روزهای 60 تا 120 شیردهی تغییر معنی داری نکرد، اما در گاوهای با افت زیاد BCS (گروه HiD)، سطح انسولین افزایش یافت (P<0.001). غلظت گلوکز در گاوهای LoD به طور خطی کاهش یافت (P=0.039) و در گروه HiD در روز 90 افزایش درجه دوم نشان داد (P=0.028). غلظت NEFA در گروه HiD تغییرات خطی (P=0.04) و درجه دوم (P=0.002) داشت. گاوهای HiD در روز 120 انسولین بالاتری نسبت به گاوهای LoD داشتند (P=0.017). میزان گلوکز در گاوهای گروه HiD در روز 60 DIM کمتر بود (P<0.01). غلظت NEFA در گاوهای HiD در روز 90 بالاتر بود (P<0.01). شاخص های جایگزین مقاومت به انسولین (محاسبه شده بر اساس غلظت متابولیت های اندازه گیری شده) بین دو گروه، در روز 90 شیردهی متفاوت بود که نشان دهنده کاهش حساسیت به انسولین در گاوهای HiD می باشد.

    نتیجه گیری: کاهش زیاد ذخایر بدن در اوایل شیردهی ممکن است منجر به اختلالاتی در سوخت و ساز انرژی در مرحله میانی شیردهی شود. کنترل افت BCS در اوایل شیردهی ممکن است به کاهش چنین تغییراتی، احتمالا از طریق تغییر حساسیت بافت ها به انسولین کمک کند.

    پیشینه:

    کبک چوکار (الکتوریس چوکار) به علت ارزش تغذیه ای بالا و گوشت نسبتا گرانش پرورش داده می شود.

    هدف

    این مطالعه برخی اندام های لنفاوی کبک چوکار در سنین قبل تا پیرامون بلوغ را با رهیافت هیستومورفومتریک ارزیابی می نماید.

    روش کار

    در این مطالعه 30 قطعه کبک چوکار یک روزه از دو جنس نر و ماده مورد استفاده قرار گرفتند. در سنین 80، 100 و120 (پیرامون بلوغ) از بورس کلواکی، تیموس، طحال، لوزه سکومی (CT)، لوزه پیلور و همچنین لوزه مری نمونه گیری انجام شد. ده برش عرضی از هر نمونه اخذ شده از هر پرنده تهیه و با رنگ آمیزی هماتوکسیلین و ائوزین (H&E) جهت اندازه گیری ارتفاع پلیکا، قطر فولیکول همچنین ضخامت قشر و مرکز فولیکول در بورس کلواکی، طول کرک، قطر واحد فولیکولی (ND) و ارتفاع آن در CT، قطر لوبول، ضخامت قشر و مرکز آن در تیموس، ضخامت پالپ سفید در طحال، قطر فولیکول در لوزه پیلور و لوزه مری گراتیکول خطی مورد استفاده قرار گرفت و تعداد فلیکول های هر پلیکا در بورس کلواکی و تعداد فولیکول هر ND در CT، رنگ آمیزی و زیر میکروسکوپ نوری ارزیابی شدند.

    نتایج

    تغییرات شدیدی در پارامترهای هیستومورفومتریک بافت های لنفاوی در پیرامون بلوغ به ویژه در بورس کلواکی و تیموس دیده شد که به صورت کاهش معنی دار در تمام پارامترهای مورد مطالعه نشان داده شد. تحلیل بافتی همچنین در لوزه مری و لوزه پیلور دیده شد. نتیجه گیری: تحلیل بافتی در بورس و تیموس در پیرامون بلوغ در مقایسه با مراحل قبل از بلوغ در کبک رخ می دهد. این تغییرات همچنین در CT و لوزه پیلور و لوزه مری نیز قابل ملاحظه است در حالی که در طحال تغییر چهره بافتی قبل و پیرامون بلوغ دیده نمی شود.

    پیشینه:

    پنی باسیلوس لاروا عامل ایجاد کننده بیماری لوک آمریکایی (AFB)، کشنده ترین بیماری در لارو زنبور عسل (آپیس ملیفرا)، می باشد.

    هدف

    هدف از انجام تحقیق حاضر مطالعه بررسی اثر ضد میکروبی مایع رویی عاری از سلول (CFS) پروبیوتیک ها بر روی تشکیل بیوفیلم یک جدایه ایرانی بود.

    روش کار

    یک سویه بومی پنی باسیلوس لاروا بر اساس تعیین توالی 16S rRNA شناسایی شد. اثر ضد میکروبی تعدادی از پروبیوتیک ها با روش قطره پلیت، حداقل غلظت مهار کننده (MIC) و حداقل غلظت کشنده باکتری (MBC) و آزمون زمان کشندگی ارزیابی شد. توانایی تشکیل بیوفیلم پنی باسیلوس لاروا و اثر مهاری ایجاد بیوفیلم توسط CFS با روش میکروتیتر پلیت و میکروسکوپ الکترونی نگاره (SEM) مورد مطالعه قرار گرفت. ماهیت متابولیت های ثانویه در CFS با آزمون MODA بررسی شد.

    نتایج

    نتایج توالی یابی 16S rRNA پنی باسیلوس لاروا KB10 (شماره دسترسی بانک ژن MH000685.1) با پایگاه داده، نشان دهنده شباهت بیش از 97% با     پنی باسیلوس لاروا بود. بیشترین اثر ضد باکتری در CFC لاکتوباسیلوس روتری ATCC23272با قطر هاله عدم رشد (ZOI) 2/3 ± 75/12 میلی متر در رقت µL/ml 1000 برای MIC و MBC مشاهده شد. آزمون زمان کشندگی نشان داد که CFS قادر به از بین بردن     CFU/ml 108 × 5/1 پنی باسیلوس لاروا KB10 در مدت زمان دو ساعت مواجهه بود. نتایج میکروتیتر پلیت و SEM نشان داد که CFS (در غلظت زیر MIC) قادر به مهار کردن تشکیل بیوفیلم توسط پنی باسیلوس لاروا بود. نتایج حاصل از آزمون MODA نشان داد که فعالیت ضد میکروبی مربوط به تولید اسیدهای آلی موجود در آن می باشد. نتیجه گیری: مایع رویی عاری از سلول لاکتوباسیلوس روتری ATCC23272، به دلیل ماهیت اسیدی، دارای اثرات مهار کننده بر روی رشد و تشکیل بیوفیلم باسیلوس لاروا KB10 بود. نتایج به دست آمده می تواند جهت جایگزین کردن آنتی بیوتیک در کنترل و درمان بیماری لوک آمریکایی مورد استفاده قرار گیرد.

    پیشینه:

    استرپتوکوکوس آگالاکتیه پاتوژن اصلی ایجاد کننده ورم پستان گاوی (BM) است و ضررهای اقتصادی قابل توجهی را به صنعت لبنیات در سراسر دنیا وارد می کند. واکسن های علیه استرپتوکوکوس آگالاکتیه نقش بسیار مهمی در پیشگیری از بیماری ایفا می کنند.

    هدف

    هدف از انجام این مطالعه ارزیابی ایمنی محافظتی پروتئین های فیوژنی پیلوس آیلند استرپتوکوکوس آگالاکتیه شامل پروتئین 1 فرعی-پروتئین 2 فرعی (AP1-AP2)، پروتئین 1 فرعی-پروتئین استخوان (AP1-BP)، پروتئین استخوان-پروتئین 2 فرعی (BP-AP2) و پروتئین 1 فرعی-پروتئین استخوان-پروتئین 2 فرعی (AP1-BP-AP2) در موش های بالب سی بود.

    روش کار

    چهار نوع از آنتی ژن های فیوژنی با حجم مساوی از ادجوانت کامل فروند به منظور ساخت نمونه های ایمنی زای پروتئین فیوژنی به شدت با هم مخلوط شد. موش ها در 4 زمان (روزهای 0، 7، 14 و 28) با این نمونه ها با دوز ایمنی زای 50 میکروگرم به ازای هر موش، ایمن شدند. پس از چهارمین مرحله ایمن سازی، آزمون های سرولوژی جهت ارزیابی تولید پادتن مورد استفاده قرار گرفت. عیار پادتن تولید شده توسط آنتی ژن فیوژنی AP1-BP-AP2 دارای بیشترین میزان و حداکثر تا 1:25600 بود. نیم میلی لیتر از استرپتوکوکوس آگالاکتیه های جدا شده از موارد بالینی با غلظت CFU/ml 104 × 2 در روز 50 تلقیح شد و موش ها به صورت روزانه تا 7 روز مورد مشاهده قرار گرفتند.

    نتایج

    تحلیل های آماری نشان داد که این 4 نوع از آنتی ژن های فیوژنی ایمنی محافظتی خوبی ایجاد کردند. در بین آن ها آنتی ژن فیوژنی AP1-BP-AP2 دارای بهترین ایمنی محافظتی در موش های بالب سی با شاخص حفاظت (PI) ایمنی 80% بود. نتیجه گیری: این مطالعه مبانی نظری قابل اعتمادی را جهت غربالگری آنتی ژن های نامزد تولید واکسن های زیر واحد و فرآورده های  آنتی ژن های شناساگر مربوط به استرپتوکوکوس آگالاکتیه فراهم می کند.

    پیشینه: پرندگان را می توان به عنوان مخزن قارچ های بیماری زا و غیر بیماری زا در نظر گرفت. فضولات کبوتر قابلیت انتشار این قارچ ها در طبیعت را دارا می باشند. گونه های کریپتوکوکوس از قارچ های مهم موجود در فضولات کبوتر می باشد. در این میان گونه های مختلفی از مخمر در فضولات کبوتر وجود دارد که برای سلامتی انسان و حیوانات مهم است.

    هدف

    هدف از این مطالعه شناسایی مخمرهای غیر کریپتوکوکوس جدا شده از فضولات کبوتر در شیراز، جنوب ایران می باشد.

    روش کار

    تعداد 100 مخمر ناشناخته که قبلا با روش های معمول آزمایشگاهی به عنوان مخمر غیر کریپتوکوکوس شناخته شده و از فضله کبوتران جدا شده بودند مورد مطالعه قرار گرفت. شناسایی جدایه ها با روش های متداول آزمایشگاهی و آنالیز سکانس ناحیه فاصله گزار در نسخه برداری (ITS) ژن DNA ریبوزومی انجام شد. نتایج سکانس با نتایج موجود در بانک اطلاعاتی NCBI مقایسه و با استفاده از نرم افزار ابزار اصلی جستجوی آنالیز توالی (BLAST) بررسی گردید.

    نتایج

    تعداد 16 گونه از هفت جنس قارچی شامل جنس های کاندیدا 51% (8 گونه)، رودوتورولا 24%، ترایکوسپورون 21% (3 گونه)، رودوسپوریدیوم 2%، ساکارومایسس 1%، رایزوکتونیا 1% و میروزیما 1% شناسایی شدند. رودوتورولا روبرا (24%)، کاندیدا فوماتا (20%) و ترایکوسپورون آساهی (13%) از شایع ترین گونه های جدا شده بودند. سایر گونه ها شامل رودوسپوریدیوم کراتوچویلوا 2 (2%)، ساکارومایسس سرویسیه 1 (1%)، رایزوکتونیا سولانی 1 (1%) و     میروزوما کاریبیکا 1 (1%) بودند. نتیجه گیری: فضولات کبوترهای مورد بررسی در این مطالعه حاوی گونه های مختلفی از مخمرهای فرصت طلب بود که می توانند برای انسان و حیوانات مستعد، بیماری زا باشند.

    پیشینه: 

    بروز رسانی سالیانه اطلاعات درباره شیوع استافیلوکوکوس اورئوس و ژن های انتروتوکسین استافیلوکوکی (SEs) در هر منطقه جغرافیایی لازم است.

    هدف

    این مطالعه جهت بررسی شیوع این باکتری و انواع ژن های انتروتوکسین های مرتبط با آن در مواد غذایی مختلف، با استفاده از آزمون واکنش زنجیره ای پلیمراز (PCR) چندگانه، انجام شد.

    روش کار

    برای رسیدن به این هدف، 310 نمونه، که به سه گروه (محصولات لبنی، گوشت، و گروه شیرینی سنتی) تقسیم شده بودند، جمع آوری شدند. پس از تعیین شیوع استافیلوکوکوس اورئوس، میزان حضور ژن های 16s rRNA، sea، seb، sec، sed، و see با استفاده از آزمون PCR چندگانه ارزیابی شدند.

    نتایج

    استافیلوکوکوس اورئوس از 103 نمونه (33%) بررسی شده جدا شد. همچنین، نمونه های در دسته گوشت بالاترین آلودگی را با استافیلوکوکوس اورئوس نشان دادند. در همین راستا، نمونه های کباب (5/61%) و به دنبال آن همبرگر (3/47%) و بستنی (8/33%) بیشترین محصولات آلوده بودند. در میان این 103 جدایه استافیلوکوکوس اورئوس، 72 جدایه (9/69%) حداقل حامل یک ژن SEs کلاسیک بودند. ژن انتروتوکسین نوع A به میزان بالاتری نسبت به سایر ژن های  SEsشناسایی شد. نتیجه گیری: یافته ها نشان دادند که دستکاری نامناسب نمونه ها در مراحل آماده سازی و فرآوری، به خصوص در محصولات گوشتی، می تواند منجر به انتشار بیشتر باکتری ها در محصولات شود. شیوع نسبتا بالای بعضی از ژن های انتروتوکسین کلاسیک در جدایه ها، قدرت بالقوه این باکتری در تولید انتروتوکسین، که می تواند منجر به مسمومیت غذایی شود را، نشان داد.

    پیشینه:

    کتواسیدوز دیابتی (DKA) در واقع یک اختلال در سوخت و ساز کربوهیدرات ها است که سبب تکرر ادرار، لاغری مفرط، خستگی شدید و کم آب شدن بدن می شود. هیچ گونه اطلاعات یا اطلاعات محدودی در رابطه با DKA در بز نر (باک) در دسترس است. مطالعه حاضر به منظور گزارش یک مورد نادر از DKA در یک بز نر انجام شد. توصیف بیمار: یک راس بز نر 5/1 ساله با بی اشتهایی و سرفه ارجاع داده شد. در معاینه بالینی تب، گیجی، وضعیت بدنی نامناسب و همچنین رنگ پریدگی غشای مخاطی ملتحمه مشاهده شد.

    یافته ها/درمان و نتیجه درمان:

    اسمیر خون محیطی عفونت مخلوط با گونه های تیلریا و آناپلاسما را نشان داد. تابلوی خونی نشان دهنده آنمی (کم خونی) و لوکوسیتوز (افزایش تعداد گلبول های سفید خون) بود. حیوان با استفاده از بوپارواکون با دوز mg/kg 5/2 و اکسی تتراسیکلین طولانی اثر با دوز mg/kg 20 درمان شد. ارزیابی پس از درمان 7 روز بعد از درمان اولیه انجام شد. مصرف غذای حیوان اندکی بهبود یافته بود‏، دمای بدن طبیعی شده بود ولی تاکی کاردی (تند تپشی) همراه با ضربان ضعیف مشاهده شد. علاوه بر این تکرر ادرار و لاغری مفرط شدید نیز دیده شد. آزمایش ادرار گلیکوزاوری (گلوکز ادراری ادرار)، کتو اوری (کتون ادراری) و ادرار اسیدی (pH = 6.0) را نشان داد. آزمایش بیوشیمیایی سرم، هایپرگلایسمی (افزایش قند خون)، هایپوانسولینمی (کاهش انسولین خون)، افزایش سطح فروکتوزآمین و تری گلیسریدها را نشان و رخداد DKA خود به خودی را تایید کرد. حیوان با انسولین ایزوفان دو فازی با دوز IU/kg 1، دو بار در روز، به طور منظم درمان شد. سطح گلوکز خون پس از درمان با انسولین به حالت طبیعی برگشت. میزان مصرف خوراک به مقادیر مناسب رسید و بهبود در میزان هموگلوبولین (Hb)، حجم متراکم گلبول های قرمز (PCV)، شمارش تام گلبول های قرمز (TEC) و وزن گیری مشاهده شد.

    نتیجه گیری:

    این مطالعه گزارش موردی، به لحاظ بهبود موفقیت آمیز حیوان پس از درمان با انسولین، با اهمیت به نظر می رسد، اما برای مدیریت دیابت شیرین وابسته به انسولین (IDDM) در این بز، نیاز به درمان با انسولین در تمام طول عمر می باشد.

    پیشینه:

    جسم خارجی در مری پرندگان به ندرت در مطالعات گزارش شده است. رایج ترین محل گیر کردن جسم خارجی در ماکیان سانان پیش معده و سنگدان است. هدف از این مطالعه تشریح تشخیص وجود یک جسم خارجی بزرگ تیز در یک سوم انتهایی گردن با پرتونگاری ساده و خارج سازی آن از طریق ازوفاگوتومی و زنده ماندن پرنده متعاقب عمل جراحی است. توصیف بیمار: یک قطعه ماکیان اهلی نژاد آسیل  (گالوس دومستیکوس) با تاریخچه تورم در ناحیه گردن و بی اشتهایی متعاقب آن از 24 ساعت، اما با مصرف طبیعی آب به بخش جراحی ارجاع داده شد. بررسی پرتونگاری حاکی از وجود یک استخوان بزرگ ماهی در مری بود. لبه های تیز جسم خارجی منجر به تصمیم برای انجام ازوفاگوتومی به جای روش های مدیریتی معمول نظیر خارج کردن جسم از طریق دهان و یا هدایت جسم به چینه دان و انجام اینگلوویوتومی، شد.

    یافته ها/درمان و نتیجه درمان:

    ازوفاگوتومی تحت بی حسی موضعی با استفاده از محلول رقیق شده لیگنوکائین انجام شد. برش ازوفاگوتومی با استفاده از نخ پلی گلاکتین 910 شماره 0-3 در دو لایه دوخته شد. پس از عمل جراحی به صاحب پرنده توصیه شد که انروفلوکساسین را با دوز mg 10 به ازای هر کیلوگرم وزن بدن (BW) به مدت هفت روز و ترامادول را (10 mg/kg BW, BID) به مدت سه روز در آب آشامیدنی به پرنده تجویز کند. این مورد به مدت 8 ماه مورد بررسی و پیگیری قرار گرفت. صاحب پرنده به مدت یک ماه مواد غذایی را فقط به شکل مایع به پرنده داد. پس از یک ماه، تغذیه به حالت طبیعی برگردانده شد. صاحب پرنده متوجه هیچ گونه عارضه ای نشد.

    نتیجه گیری:

    می توان نتیجه گرفت که مداخله سریع و مدیریت جراحی جهت خارج کردن جسم خارجی تیز در بخش انتهایی مری در ناحیه گردن ماکیان اهلی ممکن است لازم باشد.

    پیشینه:

    پمفیگوس پارانئوپلاستیک یک بیماری پوستی تاول زای خود ایمن همراه با نئوپلازی هم زمان است که در دامپزشکی به ندرت دیده می شود.

    توصیف بیمار:

    این گزارش موردی، یک قلاده سگ نژاد لابرادور رتریور ماده 10 ساله مبتلا به مو ریختگی و سبوره عمومی، جراحات اولسراتیو و پوسته پوسته در نواحی اطرف چشم و دهان، لب ها و کشاله ران را توصیف می نماید.

    یافته ها/درمان و نتیجه درمان:

    آزمایش های هماتولوژی افزایش قابل توجه تعداد لنفوسیت ها را نشان داد در حالی که سایر پارامترها طبیعی بودند. نمونه برداری از پوست انجام و یافته های هیستولوژیک وجود پمفیگوس همراه با لنفوم اپی تلیوتروپیک را تایید کرد. سپس آزمایش های ایمونوهیستوشیمی انجام شد. رسوب ایمونوگلوبین G (IgG) در غشای پایه و فضای داخل سلولی مشاهده شد که با ویژگی های الگوی رنگ آمیزی ایمونولوژیک مربوط به پمفیگوس اریتماتوز (PE) مطابقت داشت. نمونه های لنفوم از نظر وجود CD3 مثبت شدند و به عنوان یک لنفوم اپی تلیوتروپیک ناشی از سلول T دسته بندی شد.

    نتیجه گیری:

    بر اساس اطلاعات موجود، این اولین مورد از وقوع هم زمان PE و لنفوم پوستی در یک سگ است که موارد ارتباط بین بیماری های وابسته به ایمنی و سرطان را در سگ ها گسترش می دهد و همچنین از ارتباط احتمالی بین بیماری های خود ایمن و نئوپلازی در این گونه حمایت می کند.

    پیشینه:

    تراوش پریکاردی (PE) ناشی از متاستاز ثانویه به ندرت در سگ ها گزارش شده است. توصیف بیمار: این گزارش موردی، نشانه های بالینی و تشخیص بیشتر در مورد کارسینوم متاستاتیک با منشا اولیه نامشخص (CUP) را در PE مقاوم به درمان در یک قلاده سگ توصیف می کند.

    یافته ها/درمان و نتیجه درمان:

    یک قلاده سگ 9 ساله نر اخته شده از نژاد شیتزو برای معاینه و بررسی سرفه و تنگی نفس ارجاع داده شد. در معاینه بالینی، تاکی پنه، سرفه های متناوب و صدا های قلبی گنگ ثبت شد. رادیوگرافی قفسه سینه، الکتروکاردیوگرافی و اکوکاردیوگرافی، ابتلا به PE را تایید کرد. هیچ گونه ضایعه توده ای در قاعده قلب، آئورت یا دهلیز راست (RA) مشاهده نشد. بررسی مایع پریکارد، سیتولوژی هموراژیک را نشان داد و به طور موقت PE هموراژیک با علت نامعلوم تشخیص داده شد. سگ به درمان های محتاطانه با استروئید و داروهای مدر پاسخ داد ولی نشانه های بالینی مجددا باز گشت. بررسی بیشتر با مقطع نگاری رایانه ای با آشکارسازهای چند ردیفی (MDCT)، ارزش تشخیصی نداشت. سگ 457 روز پس از معاینه اولیه تلف شد. کالبدگشایی و هیستوپاتولوژی حاکی از کارسینوم متاستیک با منشا اولیه نامعلوم بود.

    نتیجه گیری:

    این مورد یک علت نادر PE بازگشت کننده در سگ ها را نشان داد.

    Many studies tested different feed additives, among these additives, humic substances (HS) have been used in livestock and poultry diets. Humic substances commonly present in nature as they are created from the organic matter decomposition, and are normally found in the soil and natural water. Active components of HS consist of humic acid (HA), humus, ulmic acid, fulvic acid, humin and certain microelements. Humic acid is widely used as an alternative growth promoter for antibiotics in improving poultry performance and health. Moreover, supplementation of a commercial substance as a source of HS through the drinking water or diet improved the feed consumption, feed efficiency and weight gain of broiler chickens, and also improved egg weight, egg mass, and egg production of laying hens. This review describes the useful applications and recent facets of HA including its modes of action and various valuable uses in improving the production and health safeguarding of livestock and poultry.

    Background

    The use of C-type natriuretic peptide (CNP) combined with cysteamine during pre-in vitro maturation (IVM) can help establish an effective pre-IVM system.

    Aims

    This study was designed to investigate the effects of CNP combined with cysteamine on meiotic maturation and developmental competence of bovine oocytes in vitro.

    Methods

    Oocytes were cultured in pre-IVM medium supplemented with 100 nM CNP for 6 h in the absence or presence of 100 μM cysteamine, followed by standard IVM for 24 h. Oocytes matured by standard IVM for 24 h (no pre-IVM) were used as the control. The percentage of oocytes arrested at germinal vesicle (GV) stage in each treatment group was examined at 0 h of IVM, and the nuclear maturation rate was assessed by evaluating the percentages of oocytes that reached metaphase II (M II) stage at 24 h of IVM. After 24 h of IVM, oocytes were subjected to in vitro fertilization (IVF) and in vitro embryo culture (IVC). Cleavage rates were assessed 48 h post-insemination. Blastocyst production rates were recorded on day 8 after IVF. In addition, intra-oocyte glutathione (GSH) and reactive oxygen species (ROS) content for each treatment were tested at 0 h and 24 h of IVM.

    Results

    No differences were observed between the CNP pre-IVM treatment and control groups in the rate of bovine oocytes maturing to the M II stage, cleavage and blastocysts production rates after IVF, and intra-oocyte GSH levels. Notably, the presence of cysteamine during pre-IVM culture with CNP significantly improved the rate of embryos developed to the blastocyst stage after in vitro maturation and fertilization, moreover, it increased the levels of GSH and reduced the levels of ROS in bovine oocytes.

    Conclusion

    The improvement to IVM bovine oocyte developmental competence through pre-IVM with CNP combined with cysteamine may be associated with an increased antioxidant defense. Therefore, such an approach may be a good option for establishing a pre-IVM system.

    Background

    Evaluation of energy metabolism indices in mid-lactation Holstein cows is critical to monitor health status.

    Aims

    The objective of this study was to assess the effects of low (≤0.75) vs. high (>0.75) drops in body condition score (BCS) until day 60 post-partum on energy metabolism indices during mid-lactation in Holstein cows.

    Methods

    Twenty-eight Holstein cows were included in the study from the day of calving to day 120 of lactation. Whole blood samples were taken on 60, 90, and 120 days in milk (DIM). Serum was analyzed for insulin, glucose, non-esterified fatty acids (NEFA) and beta-hydroxybutyrate (BHB) levels.

    Results

    In cows with low BCS drop (LoD group), insulin did not change significantly through days 60 to 120 of lactation, but increased in high drop cows (HiD group) (P<0.001). Glucose concentrations decreased linearly in the LoD cows (P=0.039) and showed a quadratic increase in the HiD group on day 90 (P=0.028). Concentrations of non-esterified fatty acids showed both linear (P=0.04) and quadratic (P=0.002) changes in the HiD group. The HiD cows had significantly higher concentrations of insulin on day 120 (P=0.017) compared to the LoD group. Glucose concentration was lower (P<0.01) in HiD cows on 60 DIM. The concentration of non-esterified fatty acids was higher in HiD cows on day 90 (P<0.01). Surrogate indices of insulin resistance (calculated based on the concentrations of the measured metabolites) were different between the groups on day 90, indicating decreased insulin sensitivity in the HiD cows.

    Conclusion

    Greater depletion of body reserves during early lactation may result in some inconsistencies in energy metabolism during mid-lactation periods. Controlling BCS loss during early lactation may help alleviate such alterations possibly through modifying insulin sensitivity of the tissues.

    Background

    Chukar partridge (Alectoris chukar) is raised for its highly nutritional and relatively expensive meat.

    Aims

    This study evaluates selected lymphoid organs of juvenile and peripubertal Chukar partridges using a histomorphometric approach.

    Methods

    Thirty one-day-old male and female Chukar partridges were included in the study. Samples were taken from cloacal bursa, thymus, spleen and the caecal tonsil (CT) as well as oesophageal and pyloric tonsils at 80, 100, and 120 days (peripuberty) of age. Ten transverse sections were then taken from each sample of each bird and stained with haematoxylin and eosin (H&E) in order to measure plicae height and follicular width as well as thickness of follicular cortex and medulla in cloacal bursa, villus height and nodular unit (ND) width and height in CT, lobular width as well as lobular cortex and medullar width in thymus, white pulp width in spleen and follicular width in oesophageal and pyloric tonsils, a linear graticule was used, the number of follicles in each plicae in cloacal bursa and the number of follicles in each ND in CT were determined under a light microscope.

    Results

    Drastic changes in histomorphometrical parameters of lymphoid tissues were observed in peripuberty, prominently in cloacal bursa and thymus as demonstrated by the significant decrease in all assayed parameters. Involutionary changes were also observed in oesophageal and pyloric tonsils.

    Conclusion

    Involution occurs in bursa and thymus of partridges around puberty rather than their juvenile stages. These changes are also appreciable in CT, oesophageal and pyloric tonsils, whereas the spleen keeps the same histological features before and around puberty.

    Background

    Paenibacillus larvae is the etiological agent of American foulbrood (AFB) disease, the most lethal disease in honeybee (Apis mellifera) larvae.

    Aims

    The aim of the present work was to study the antimicrobial effect of cell free supernatant (CFS) of probiotics on an Iranian isolate’s biofilm formation.

    Methods

    A local strain was identified by 16S rRNA sequencing. The antibacterial effect of some probiotics was evaluated through drop plate method, minimum inhibitory concentrations (MIC), minimum bactericidal concentrations (MBC) and time-kill assay. The biofilm formation ability of P. larvae and the inhibition of biofilm formation by CFS were studied by microplate and scanning electron microscopy (SEM). The nature of the secondary metabolites in CFS was examined by microscale optical density assay (MODA).

    Results

    Alignment of the results of P. larvae KB10 (GenBank accession number MH000685.1) 16S rRNA with the database revealed more than 97% identity with P. larvae. The most antibacterial effect was observed in the CFS of Lactobacillus reuteri ATCC23272 with 12.75 ± 3.2 mm for zone of inhibition (ZOI) at 1000 µL/ml for MIC and MBC. Time-kill assay revealed that CFS eliminated 1.5 × 108 CFU/ml P. larvae KB10 at 2 h of exposure. Microtitre plate and SEM results revealed that CFS (at sub-MIC concentration) was able to inhibit biofilm formation by P. larvae. The results of MODA assay showed that antimicrobial activity were related to the production of organic acids.

    Conclusion

    Cell free supernatant from L. reuteri ATCC 23272 had inhibitory effects on P. larvae KB10 growth and biofilm production due to its acidic nature. The obtained results can be used for antibiotic substitution in AFB control and treatment.

    Streptococcus agalactiae is the main causal pathogen of bovine mastitis (BM), causing considerable economic loss to the dairy industry worldwide. Vaccines against S. agalactiae play an important role in preventing disease.

    Aims

    The aim of this study was to evaluate the immunoprotection of S. agalactiae pilus island fusion proteins, ancillary protein 1-ancillary protein 2 (AP1-AP2), ancillary protein 1-bone protein (AP1-BP), bone protein-ancillary protein 2 (BP-AP2), and ancillary protein 1-bone protein-ancillary protein 2 (AP1-BP-AP2) in Balb/c mice.

    Methods

    Four kinds of fusion antigens and the same volume of Freund’s complete adjuvant were mixed vigorously to prepare fusion antigen immuno-samples. The mice were immunized 4 times (on the 0th, 7th, 14th, and 28th days) with these samples with an immunizing dose of 50 mg/mouse. After the 4th immunization, serology tests were used to evaluate the antibody. The antibody titre produced by AP1-BP-AP2 fusion antigen was the highest, at up to 1:25600. The mice were then injected with 0.5 ml of 2 × 104 CFU/ml clinically isolated S. agalactiae at day 50 and observed daily for the following 7 days.

    Results

    Statistical analyses showed that these 4 kinds of fusion antigens had good protective immunity. Among them, AP1-BP-AP2 fusion antigen had the best protective immunity in Balb/c mice, with an immune protection index (PI) of 80%.

    Conclusion

    This research provides a reliable theoretical basis for screening candidate antigens of the subunit vaccine and detecting antigen preparations of S. agalactiae.

    Background

    Birds are considered as a reservoir for pathogenic and non-pathogenic fungi. Pigeon droppings have the potential for spreading these fungi to the environment. Cryptococcus species are important fungi associated with pigeon droppings. In this regard, there are many types of yeast associated with guano that is important for human and animal health.

    Aims

    The main objective of this study is the identification of non-Cryptococcus yeasts isolated from pigeon dropping in Shiraz, Southern Iran.

    Methods

    A total of 100 unknown yeasts, which were previously screened and identified as non-Cryptococcus from pigeon guano through the conventional methods, were used in this study. Identification of the isolates was performed based on conventional methods and DNA sequence analysis of internal transcribed spacer (ITS) rDNA gene region. The sequence results were deposited in NCBI database using the Basic Local Alignment Search Tool (BLAST).

    Results

    A total of 16 species belonging to 7 genera were identified as Candida spp. 51% (8 species), Rhodotorula sp. 24%, Trichosporon spp. 21% (3 species), Rhodosporidium 2%, Saccharomyces 1%, Rhizoctonia 1%, and Meyerozyma 1%. The predominant isolates were Rhodotorula rubra (24%), Candida famata (20%), and Trichosporon asahii (13%). The other species were Rhodosporidium kratochvilovae 2 (2%), Saccharomyces cerevisiae 1 (1%), Rhizoctonia solani 1 (1%), and Meyerozyma caribbica 1 (1%).

    Conclusion

    Pigeon excreta examined in this study were associated with several kinds of opportunistic yeasts which could cause diseases in prone human and animals.

    Background

    An annual update of information about the prevalence of Staphylococcus aureus and staphylococcal enterotoxins (SEs) genes is required in every geographic area.

    Aims

    This study was conducted to investigate the prevalence of the bacterium and type of associated enterotoxin genes in different food samples, using multiplex polymerase chain reaction (PCR) assay.

    Methods

    In order to achieve these goals, 310 samples, divided into three groups (dairy products, meat, and traditional sweets groups), were collected. After determination of the prevalence of S. aureus, the existence of 16s rRNA, sea, seb, sec, sed, and see genes were evaluated using multiplex PCR assay.

    Results

    Staphylococcus aureus was isolated from 103 (33%) samples. Furthermore, the meat category had the most contamination rate of S. aureus. Additionally, the kebab samples (61.5%) were the most contaminated products, followed by hamburger (47.3%), and ice cream (33.8%). Of these 103 S. aureus isolates, 72 isolates (69.9%) harbored at least one type of the classical SEs genes. The prevalence of the type A enterotoxin gene was detected higher than the other SEs genes.

    Conclusion

    The results indicated that inappropriate handling of the samples in the preparation and processing steps, especially for the meat products, can lead to the spread of more bacteria. The relatively high prevalence of some classical enterotoxin genes in the isolates revealed the potential power of this bacterium to produce enterotoxins, which can lead to food-poisoning.

    Background

    Diabetic ketoacidosis (DKA) is a disorder of carbohydrate metabolism that causes frequent urination, emaciation, extreme tiredness and dehydration. There is little or no information available on DKA in male goat (buck). The present study was carried out to report a rare case of DKA in a buck. Case description: A 1.5 year old buck was presented with anorexia and cough. On physical examination of buck showed fever, dullness, poor body condition and pale conjunctival mucous membrane.

    Findings/treatment and outcome:

    The peripheral blood smear revealed the mixed infection of Theileria sp. and Anaplasma sp. The blood picture showed anaemia and leukocytosis. The animal was treated with buparvaquone (2.5 mg/kg) and long acting oxytetracycline (20 mg/kg). Post treatment evaluation was done 7 days after initial treatment. Animal showed mild improvement in feed intake, the body temperature becomes normal, but showed tachycardia with weak pulse. Subsequently, animal showed severe emaciation with frequent urination. Urinalysis revealed glycosuria, ketonuria and acidic urine (pH = 6.0). Serum biochemistry revealed hyperglycemia, hypoinsulinemia, increased level of fructosamine and triglycerides and confirmed spontaneous DKA. It was treated with biphasic isophane insulin (1.0 IU/kg) twice a day, regularly. The blood glucose level becomes normal after insulin therapy. Animal resumed adequate feed intake, improvement in haemoglobulin (Hb), packed cell volume (PCV), total erythrocyte count (TEC), and weight gain was observed.

    Conclusion

    This case study gains significance, due to its successful recovery after insulin treatment, but it requires lifelong insulin therapy to manage insulin dependent diabetes mellitus (IDDM) in this goat.

    Background

    Foreign body in esophagus in birds is rarely reported in literature. Most common site of foreign body in birds is proventriculus and ventriculus. The purpose of this study is to discuss the diagnosis of a large sharp foreign body in the distal third of the neck by plain radiography and its retrieval through esophagotomy and subsequent survival of the fowl. Case description: An Aseel breed of domestic fowl (Gallus domesticus) was referred to the surgery department with a history of swelling in the neck and subsequent anorexia since 24 h, but normal water intake. Radiological examination revealed a large fish bone in esophagus. Because of the sharp edges of the foreign body esophagotomy was performed rather than per os (po) retrieval or milking of the foreign body into the crop and performing an ingluviotomy.

    Findings/treatment and outcome:

    Esophagotomy was performed under local infiltration using a diluted solution of lignocaine. Fish bone pieces were retrieved from the esophagus of the fowl. The esophagotomy incision was closed in two layers using polyglactin 910 no: 3-0. Postoperatively, the owner was advised to administer Enrofloxacin po at 10 mg/kg body weight (BW)/day for seven days and Tramadol (10 mg/kg BW, BID) for three days in water. This case was followed up for 8 months. There were no complications noticed by the owner.

    Conclusion

    It could be concluded that prompt intervention and surgical management may be necessary for retrieval of sharp esophageal foreign bodies lodged in the distal cervical esophagus of domestic fowl.

    Background

    Paraneoplastic pemphigus is an autoimmune blistering skin disease associated with concurrent neoplasia that is rarely observed in veterinary medicine.

    Case description:

    This case report presents a ten-year-old female Labrador Retriever dog with generalized seborrhea and alopecic, crusting and ulcerative lesions in the periocular and perioral regions, the lips and the groins.

    Findings/treatment and outcome:

    Hematology analysis showed a marked increase in the number of lymphocytes, while the rest of the values were normal. Skin biopsies were collected and the histological findings were consistent with pemphigus in association with an epitheliotropic lymphoma. Immunohistochemical analysis was performed. There was deposition of immunoglobulin G (IgG) in the basement membrane and in the intercellular space that corresponded with an immunostaining pattern characteristic of pemphigus erythematosus (PE). The lymphoma was positive to CD3 cells and was classified as a T-cell epitheliotropic lymphoma.

    Conclusion

    To our knowledge, this is the first case of PE and cutaneous lymphoma coexisting in a dog, expanding the list of associations between immune-mediated diseases and cancer in dogs, and providing support to the eventual connection between autoimmunity and neoplasia in this species.

    This case describes clinical signs and further diagnostics regarding metastatic carcinoma of unknown primary origin (CUP) in refractory PE of a dog. Findings/treatment and outcome: A nine-year-old, castrated male Shih Tzu dog was referred for evaluation of cough and dyspnea. On presentation, tachypnea, intermittent cough, and muffled heart sounds were noted. Thoracic radiography, electrocardiography, and echocardiography confirmed a PE. No mass lesion was detected at the heart base, aorta, or right atrium (RA). Analysis of the PE showed hemorrhagic cytology, and an idiopathic hemorrhagic PE was tentatively diagnosed. The dog responded to conservative treatment with steroid and diuretics, but the clinical sign recurred. Further evaluation with multi-detector computed tomography (MDCT) was non-diagnostic. The dog died 457 days after initial presentation. Necropsy and histopathology revealed metastatic CUP origin.

    Conclusion

    This case illustrated a rare cause of recurrent PE in dogs.
