    بومادران جزء قدیم ترین گیاهان دارویی شناخته شده می باشد که به دلیل وجود ترکیبات شیمیایی خاص هم در طب سنتی هم در طب مدرن استفاده های گوناگونی برای آن ذکر شده است. یکی از مهم ترین خواص دارویی بومادران، تاثیرات ضد میکروبی آن بر طیف گسترده ای از عوامل بیماری زا در انسان و حیوان است. در این مطالعه هدف استخراج و شناسایی اجزای اصلی اسانس برگ بومادران و ارزیابی فعالیت ضد میکروبی و آنتی اکسیدانی اسانس می باشد. نتایج حاصل از بررسی اجزاء اسانس به روش GC/MS تعداد 58 ترکیب را در اسانس برگ اثبات کرد که بورنئول (%32/12)، 1، 8 سینئول (%51/10)، β- پینن (%33/9) و α- پینن (%82/8) ترکیبات عمده را تشکیل می دادند. اسانس به شدت رادیکال دی فنیل پیکریل هیدرازیل (IC50 =0.94 μg/ml) را کاهش داد. در رابطه با فعالیت ضد میکروبی اسانس، حساس ترین میکروارگانیسم ها قارچ ها و سپس باکتری گرم مثبت استافیلوکوکوس اورئوس و مقاوم ترین آن ها باکتری های گرم منفی تشخیص داده شدند (MIC=0.45-7.20 mg/ml). این مطالعه تایید کرد که اسانس برگ بومادران دارای ویژگی های آنتی اکسیدانی و ضد میکروبی در In Vitro بود. بنابراین می توان استفاده از آن را به عنوان یک ترکیب آنتی اکسیدان و نگهدارنده در فرآورده های غذایی پیشنهاد نمود.

    به منظور بررسی و مقایسه تاثیر سرد کردن با روش غوطه وری در آب خنک، محلول پراکسید هیدروژن و گاز اوزون بر کاهش بار میکروبی لاشه طیور مجموع 45 لاشه مرغ گوشتی کشتار شده شده از یک واحد تولیدی در کشتارگاه طیور استان چهارمحال و بختیاری جمع آوری و به سه گروه تقسیم شدند. تعداد 15 قطعه لاشه به مدت 60 دقیقه در محلول خنک کننده آب سرد (کنترل) و 15 قطعه لاشه به مدت 60 دقیقه در خنک کننده حاوی 02/0 پراکسید هیدروژن و 15 قطعه لاشه دیگر داخل محفظه ای تحت تاثیر گاز اوزون با غلظت 7 میلی گرم در دقیقه قرار داده شدند. برای ارزیابی بار میکروبی از هر لاشه متعلق به گروه های آزمایشی یک سوآپ از ناحیه پوست سینه جمع آوری گردید و نمونه ها تحت آزمایش های استاندارد بیوشمی قرار گرفتند. نتایج نشان داد در گروه های مصرف کننده پراکسید هیدروژن و اوزون کاهش معنی داری در تعداد کلی فرم ها و استافیلوکوکوس اورئوس نسبت به گروه کنترل مشاهده گردید. همچنین بررسی ها نشان دهنده کاهش قابل توجهی در تعداد کلی کپک ها و مخمر ها در اثر مصرف پراکسید اکسیژن و گاز اوزون بود. استفاده از هیدروژن پروکساید در مقایسه با گاز اوزون در خصوص کاهش تعداد کلی فرم ها، کپک ها و مخمر ها و استافیلوکوکوس اورئوس موثر تر واقع شد. به صورت کلی می توان بیان کرد استفاده از پراکسید هیدروژن وگاز اوزون اثرات قابل توجهی بر کاهش بار میکروبی لاشه طیور و حفظ کیفیت آن در خط تولید کشتارگاه دارد.

    مطالعه حاضر با هدف تایید مولکولی، تعیین تیپ کپسولی و بررسی 12 عامل مهم حدت شامل ompH، oma87، sodA، sodC، hgbA، hgbB، exBD-tonB، nanB، nanH، ptfA، pfhAوtoxA 18 جدایه طیوری پاستورلا مولتوسیدا از مناطق مختلف ایران به همراه یک سویه از ارگانیسم که از موارد همه گیری پاستورلوز در مناطق شمالی کشور جداسازی شده بود، و نیز بررسی کشندگی جدایه های حاد در جنین و نیمچه ماکیان طراحی گردید. برای تایید مولکولی جدایه های مورد آزمایش از پرایمرهای اختصاصی ژن kmt1 و برای تعیین تیپ کپسولی و شناسایی عوامل حدت آن ها از روش PCR چندگانه و پرایمرهای اختصاصی ژن های مسئول بیوسنتز کپسول و ژن های کدکننده عوامل حدت ارگانیسم بهره برده شد. تمام جدایه ها از نظر مولکولی تایید شدند ولی از نظر تیپ کپسولی، 16 جدایه (8/88 درصد) به همراه سویه جداشده از موارد همه گیری پاستورلوز متعلق به تیپ A بوده و دو جدایه (1/11درصد) غیرقابل تیپ بندی بودند. آزمایش شناسایی عوامل حدت نشان داد که 100 درصد جدایه ها دارای شش ژن حدت sodC، hgbA، hgbB، nanB، nanHو ptfAمی باشند. ژن های oma87، exBD-tonB، ompH، sodAو pfhAنیز به ترتیب در 4/94، 4/94، 3/83، 6/66 و 7/27 درصد از جدایه ها حضور داشتند درحالی که هیچ کدام از آن ها حامل ژن toxAنبودند. جهت تعیین میزان کشندگی 50 درصد جدایه ها برای جنین ماکیان (Chicken embryo lethal dose 50) (CELD50) و نیز محاسبه دوز کشندگی 50 درصد آن ها در ماکیان (Lethal dose 50) (LD50)، رقت های مختلف پنج جدایه حاد واجد اکثر ژن های حدت و نیز سویه جداشده از موارد همه گیری پاستورلوز به ترتیب به تخم مرغ های جنین دار و نیمچه های تخم گذار تلقیح گردیدند. بر اساس نتایج محاسبه CELD50، با اینکه هر شش جدایه قادر به تلف نمودن جنین ماکیان بودند ولی سویه جداشده از موارد همه گیری پاستورلوز به عنوان حادترین جدایه مشخص گردید. نتایج آزمایش تعیین LD50 نشان داد که هیچ کدام از پنج جدایه حاد قادر به تلف نمودن پرندگان تحت آزمایش نبودند در حالی که میزان LD50 سویه جداشده از موارد همه گیری پاستورلوز 101×5 واحد تشکیل دهنده پرگنه به ازای هر پرنده (cfu/case) محاسبه گردید و این سویه به عنوان حادترین جدایه تحت بررسی شناسایی شد. یافته های پژوهش جاری نشان داد با اینکه جدایه های پاستورلا مولتوسیدای طیور ایران حاوی اغلب ژن های حدت بوده و حادترین آن ها قادر به تلف نمودن جنین ماکیان نیز بودند ولی هیچ کدام از این جدایه ها توانایی عفونی سازی نیمچه ماکیان را نداشته و حدت سویه های جداشده از پرندگان مبتلا به شکل فوق حاد بیماری در طی همه گیری ها، به مراتب بالاتر می باشد. داده های مطالعه حاضر می تواند در طراحی واکسن های جدید و کارآمدی که قادر به ایجاد محافظت قابل قبول در جمعیت های دامی باشند، مفید واقع گردد.

    هدف از این مطالعه بررسی اثرات ضد میکروبی عصاره آبی، اتانولی، متانولی، استونی، کلروفورمی و اتیل استات حاصل از برگ گیاه صبر زرد (Aloe vera) بر روی دو باکتری بیماری زای ماهی، ویبریو کلرا (Vibrio cholerae) و لیستریا مونوسایتوژنز (Listeria monocytogenes) بود. عصاره گیاه صبر زرد با استفاده از دستگاه روتاری بدست آمد و باکتری های ویبریو کلرا و لیستریا مونوسایتوژنز از کلیکسیون قارچ و باکتری ایران تهیه شدند. حداقل غلظت کشندگی (MBC) و حداقل غلظت مهار کنندگی (MIC) عصاره ها با روش رقت سازی در چاهک تعیین گردید. همچنین قطر هاله مهاری با استفاده از روش دیسک گذاری تعیین شد. نتایج مطالعه حاضر نشان داد که عصاره آبی گیاه صبر زرد، علی رغم خواص مهاری و کشندگی علیه لیستریا مونوسایتوژنز، هیچگونه تاثیری بر ویبریو کلرا نداشت. بیشترین خواص مهار کنندگی و کشندگی علیه هر دو باکتری مورد مطالعه، مربوط به عصاره کلروفورمی صبر زرد بود، هرچند تاثیر ضد باکتریایی آن بر لیستریا مونوسایتوژنز، بیشتر بود. عصاره کلروفومی حتی در رقیق ترین حالت نیز دارای خاصیت مهارکنندگی بود. به همین ترتیب، بیشترین قطر هاله مهاری عصاره صبر زرد مربوط به حلال کلروفورم و علیه لیستریا مونوسایتوژنز بود. با توجه به اینکه صبر زرد به راحتی به صورت خوراکی مصرف می شود و با توجه به قابل دسترس بودن و قیمت مناسب آن، امکان استفاده از آن به عنوان جایگزین آنتی بیوتیک های مرسوم و مواد شیمیایی ضد عفونی کننده استخر های پرورشی وجود دارد.

    لاکتوکوکوس گارویه عامل بیماری لاکتوکوکوزیس یکی از مهمترین باکتری های بیماری زا در ماهیان به حساب می آید. بررسی حاضر با هدف مطالعه اثر بازدارندگی غلظت های مختلف اسانس گیاه مرزه بختیاری و مرزه خوزستانی بر بیان ژن کپسول (epsD) باکتری لاکتوکوکوس گارویه عامل بیماری لاکتوکوکوزیس ماهیان انجام پذیرفت. به منظور انجام این مطالعه، باکتری دارای ژن epsD در مجاورت حداقل غلظت مهارکنندگی (MIC) اسانس ها برابر با μl/ml 12/0 قرار گرفت و میزان بیان ژن مربوطه با استفاده از Real Time PCR بررسی گردید. نتایج نشان داد که بیان ژن در باکتری های مجاورت داده شده با غلظت MIC اسانس ها کاهش یافت. نتایج همچنین حاکی از کاهش بیان ژن مذکور متعاقب افزایش غلظت اسانس بود. اگر چه میزان تاثیر مرزه بختیاری بر بیان ژن کپسول بیش از مرزه خوزستانی بود. از این رو استفاده از اسانس مرزه بختیاری و مرزه خوزستانی به منظور کنترل و پیشگیری بیماری لاکتوکوکوزیس به واسطه ممانعت از بیان ژن کپسول توصیه می شود.

    فلور باکتریایی آب و سطح بدن ماهی نقش مهمی در بروز بیماری های باکتریایی داشته و بعنوان عوامل بیماریزای ثانویه باکتریایی نامیده می شوند. این باکتری ها در صورت ایجاد شرایط مساعد ناشی از استرس های مختلف فعال شده و می توانند تلفات بالایی را ایجاد نمایند. در این تحقیق از سطح بدن (پوست و باله) و آبشش 70 قطعه ماهی قزل آلای رنگین کمان 300 تا 500 گرمی از 7 مزرعه پرورش ماهی قزل آلای رنگین کمان واقع در جاده هراز حد فاصل لاریجان-پلور توسط سواپ های استریل نمونه برداری انجام شد. آب کلیه مزارع از رودخانه هراز تامین می شد. سوآپ های سطح بدن و آبشش هر ماهی بطور جداگانه در ظروف شیشه ای درپوش دار حاوی PBS (Phosphate Buffer solution) استریل به آزمایشگاه حمل شدند و روی محیط های کشت TSA (Tryptic Soy Agar)، Plate Count Agar و MacConkey Agar کشت داده شدند. هر کدام از نمونه ها در دو پلیت TSA جهت نگهداری در شرایط هوازی و بیهوازی در دمای 25 درجه سانتیگراد و یک پلیت مک کانکی فقط در شرایط هوازی جهت اطمینان از عدم حضور باکتری های گرم منفی کشت داده شد. برای جداسازی باکتری های مزوفیل و ساکروفیل از محیط Plate count agar در شرایط دمایی 13 و 37 درجه سانتیگراد استفاده شد. 9 گونه باکتری از سطح بدن و آبشش ماهیان جدا شد (شامل: Staph. areus, Staph. saprophyticus, Staph. epidermidis, E.coli, Bacillus cereus, Aeromonas hydrophila, Proteus mirabilis, Pasteurella multocida, Psuedomonas aeruginosa). باکتری E. coli در تمامی نمونه های کارگاه ها از پوست و آبشش جداشد. درکارگاه های مختلف اختلاف معنی دار از نظر تعداد گونه باکتری جدا شده بین سطح بدن و آبشش مشاهده نشد (P>0.05). بین کارگاه های مختلف نیز، اختلاف معنی داری بین نوع باکتری های جداشده وجود ندارد (P>0.05). بجز باکتری E. coli که در همه کارگاه ها از پوست و آبشش تمامی نمونه ها جدا گردید و نیز باکتری P. aeruginosa که از فراوانی منظمی برخوردار نبود، در بقیه موارد درصد آلودگی به گونه های جداشده از کارگاه های بالادست بسمت پایین از روند افزایشی برخوردار بود. جداسازی این باکتری ها می تواند نشان از آلودگی آب رودخانه به فاضلاب های شهری و خانگی باشد.

    توکسوپلاسما گوندی تک یاخته ای اجباری داخل سلولی است که می تواند طیف وسیعی از حیوانات خونگرم را به عنوان میزبان های واسط آلوده کند. گربه ها میزبان نهایی هستند. شیوع سرمی آنتی بادی این انگل در پرندگان می تواند معیاری مناسب برای آلودگی محیط باشد. در این مطالعه تیتر سرمی ایمونوگلوبولین G (IgG) به روش الایزا در 168 نمونه سرم جوجه های گوشتی ارجاعی به کشتارگاه صنعتی طیور سمنان و 30 نمونه سرم مرغ های با پرورش آزاد که از محل های نگهداری مختلف در سمنان جمع آوری شده بودند، سنجیده شد. %9/39 جوجه های گوشتی ارجاعی به کشتارگاه سمنان و %7/96 مرغ های با پرورش آزاد دارای تیتر سرمی IgG توکسوپلاسما بودند. در مقایسه دو گروه مورد مطالعه در مربع کای، اختلاف معنی داری بین دو گروه مشاهده گردید (P<0.0001). نتایج این مطالعه نشان می دهد که شیوع سرمی آنتی بادی توکسوپلاسما در مرغ های سمنان نسبتا بالا می باشد که در مرغ های با پرورش آزاد به آلودگی خاک به اووسیست انگل و در جوجه های گوشتی به مشکلات مدیریتی در مرغداری ها و دسترسی گربه به عنوان میزبان نهایی به منابع آب و غذای پرندگان پرورشی مربوط می شود. کلمات کلیدی: توکسوپلاسما گوندی،Toxoplasma gondii، طیور، الایزا، ایران

    دراین پژوهش 42راس گاو واکل (گروه آزمایشی) و18راس گاوسالم (گروه شاهد) آماده تلقیح هلشتاین فحل ازیکی ازدام پروری های خراسان رضوی فراهم شد. خون از ورید زیردمی اخذ وسرم آن ها پس ازجداسازی منجمد وهمراه با لیزابه فحلی به آزمایشگاه ارسال شد. در آزمایشگاه، غلظت استرادیول سرم به کمک روش RIA و دیگر مشخصات لیزابه با مشاهده مستقیم (کش آمدن و شفاف بودن) ویا به کمک میکروسکوپ (سرخسی شدن و نظم کریستال ها) و همچنین نتیجه کشت باکتری شناسی تعیین شد. نتایج نشان داد که میانگین کش آمدن (cm) (9/0=P ,3/0±3/3 ,4/0±1/3 ,3/0± 25/3 ,2/0± 15/3)، میانه سرخس شدن ساده (3/0=P ,3 ,2 ,3 ,5/2)، میانه سرخسی شدن بررسی شده بافتوشاپ(1/0=P ,4 ,4 ,3 ,3)، میانه منظم بودن کریستال ها (85/0=P ,2 ,2 ,2 ,2)، میانه شفاف بودن لیزابه (06/0=P ,2 ,2 ,5/1 ,2) و میانگین غلظت استرادیول(pg/ml) (7/0=P، 3 ± 3/5، 5/1 ± 4، 7/1 ± 7/4، 4/2 ± 8) به ترتیب ازچپ به راست درگروه واکل غیرآبستن(34راس)، واکل آبستن(8 راس)، سالم غیرآبستن (10 راس) وسالم آبستن (8 راس) بود. توزیع دام ها دردرجات مختلف شاخص های بررسی شده در لیزابه فحلی معنی دار نبود(05/0<P). آزمون اسپرمن تنها میان میزان سرخسی شدن بررسی شده با فتوشاپ و گروه های آزمایشی و شاهد ارتباط معنی داریافت (017/0=P , 3/0-=r) به گونه ای که درجه 3 سرخسی شدن (معادل سرخسی شدن40 تا60% لیزابه) برای باروری به ترین و درجات کم تر یا بیش تر از آن نامناسب تلقی شد. نتایج باکتری شناسی نشان داد که میان باکتری های جدا شده و درجات سرخصی شدن، کش آمدن، شفاف بودن و منظم بودن کریستال ها ارتباطی وجود نداشت (P>0.05) درحالیکه اشرشیا کلی، کلبسیلا، تروپرلا پیوژنز، استافیلوکوک های بتا همولتیک به طور معنی داری از گروه واکل ها بیشتر جدا شد (P=0.00) .از این مطالعه نتیجه گیری شد که از میان شاخص های بررسی شده تنها درجه متوسط سرخسی شدن بررسی شده با فتوشاپ با میزان آبستن شدن و واکل نبودن دام ها ارتباط داشت.

    یکی از عوامل مهم ایجاد تلفات تخم های ماهیان در تفریخگاه ها آلودگی با قارچ ساپرولگنیا پارازیتیکا است. مالاشیت سبز پرکاربرد ترین دارو در مبارزه با این قارچ بوده که امروزه سرطانزا بودن آن تایید شده است. هدف از این پژوهش بررسی اثرات ضد قارچی عصاره های بابونه و بومادران در مقایسه با مالاشیت سبز در کنترل آلودگی تخم های قزل آلای رنگین کمان با قارچ ساپرولگنیا است. در این مطالعه با استفاده از روش های Micro Broth Dilution Test و کشت قارچ در محیط کشت جامد، حداقل غلظت مهار کننده رشد (MIC) و حداقل غلظت کشنده (MFC) در برابر قارچ ساپرولگنیا پارازیتیکا برای عصاره بابونه به ترتیب 100 میلی گرم در میلی لیتر و 150 میلی گرم در میلی لیتر و برای عصاره بومادران بترتیب 150 میلی گرم در میلی لیتر و 200 میلی گرم در میلی لیتر تعیین شد. سپس تخم های قزل آلای رنگین کمان در چهار تیمار (شاهد، دو تیمار عصاره گیاهی و یک تیمار مالاشیت سبز) تقسیم شدند و در دوره انکوباسیون روزانه یک ساعت تحت تاثیر غلظت های MFC هر عصاره و 1 میلی گرم در لیتر مالاشیت سبز قرار گرفتند. تلفات تخم ها در مراحل لقاح تا چشم زدگی، چشم زدگی تا تفریخ و کل دوره انکوباسیون مقایسه شدند. نتایج نشان داد تلفات تخم ها در هر مرحله در تیمار مالاشیت سبز بطور معنی داری کمتر از شاهد و بومادران بود (05/0p˃) اما با تیمار بابونه تفاوت معنی داری نداشت (05/0p˃). معنی دار نبودن تلفات و درصد تفریخ تخم ها در تیمار بابونه (%26/0±90/75) نسبت به مالاشیت سبز (%32/2±97/77)، نشان داد که عصاره بابونه با اثرات ضد قارچی، داروی طبیعی مناسبی برای جایگزینی مالاشیت سبز در تفریخگاه های تخم ماهیان است.

    اشرشیاکلی یکی از مهمترین میکروب های بیماری زا در انسان و پرندگان محسوب می شود. جدایه های اشرشیاکلی بیماریزای پرندگان (APEC) و اشرشیاکلی بیماریزای دستگاه ادراری (UPEC) قادر به بیماریزایی خارج از محیط روده می باشند. این جدایه ها صفات مشترکی دارند. این احتمال وجود دارد که انتقال برخی از خصوصیات مشترک بین میزبان های مختلف، در اپیدمیولوژی بیماری در انسان و طیور حائز اهمیت باشد. در مطالعه حاضر، شیوع 4 ژن وابسته به حدت (sitA، iutA، traT و tsh) در 26 جدایه APEC و 25 ایزوله UPEC جدا شده از موارد بالینی مشکوک به عفونت اشرشیاکلی در آذربایجان غربی به روش واکنش زنجیره ای پلیمراز (PCR) بررسی گردید. آنالیز آماری با استفاده از نرم افزار minitab 15 انجام گرفت. فراوانی ژن های حدت در جدایه های APEC و UPEC به ترتیب در مورد ژن traT %2/96 و %64، ژن sitA %5/88 و %76، ژن iutA %6/84 و %68، و tsh %5/61 و %16 می-باشد. ژن های حدت traT و sitA به ترتیب در جدایه های APEC و UPEC بالاترین فراوانی را داشتند. تنوع زیادی از حضور ژن-های حدت در این جدایه ها مشاهده شد. ترکیب iutA-sitA-tsh-traT بیشترین فراوانی ژن های حدت را در بین جدایه های بیماریزای پرندگان داشت. در مورد جدایه های بیماریزا در عفونت ادراری انسان، بالاترین فراوانی را ترکیب iutA-sitA-traT داشت. این احتمال وجود دارد که ژن های حدت بین طیور و انسان انتقال یابند. ضروری است تا مطالعات بیشتری انجام گیرد و اهمیت چنین احتمالاتی را در اپیدمیولوژی بیماری در انسان و طیور ارزیابی نماید.

    چکیده ویروس برونشیت عفونی (IBV) عامل بیماری ویروسی حاد تنفسی در طیور همراه با رال های تنفسی و سرفه و عطسه می باشد. کرونا ویروس ها طیف گسترده ای از بیماری های تنفسی، روده ای، کبدی، عصبی با حدت های مختلف در گربه ها، سگ ها، خوک ها، جوندگان، گاو، پرندگان و انسان ایجاد می کنند. ویروس برونشیت عفونی عامل بسیار واگیردار بیماری برونشیت عفونی در طیور می باشد. کرونا ویروس ها توانایی نوترکیبی و جهش زیادی دارند. توالی یابی ژنS1  برای مطالعات اپیدمیولوژی مولکولی و مشخصات ژنوتیپی ویروس برونشیت عفونی کاربرد دارد. به منظور فراهم شدن درک بهتری از اپیدمیولوژی مولکولی ویروس برونشیت عفونی در ایران جدایه های ژن S1 ویروس برونشیت عفونی طیور که در مزارع گوشتی در استان اردبیل جمع آوری شده بود، در این مطالعه توالی یابی شد. در سال 1395 در محدوده استان اردبیل 30 عدد سواب نای از گله های گوشتی به صورت تصادفی برای ردیابی و تعیین ژنوتیپ ویروس جمع آوری گردید. سپس RNA آنها با استفاده از کیت تخلیص RNA استخراج گردید. ساخت cDNA با روش RT-PCR صورت گرفت و محصول RT-PCR با روش Nested- PCR با پرایمرهای کد کننده5'UTR، تکثیر شد. این کار جهت شناسایی ویروس برونشیت عفونی صورت گرفت.برای تعیین ژنوتیپ نمونه های مثبت (توالی یابی ژن گلیکوپروتئین S) با ارسال به شرکت کره ای انجام شد. 70 درصد نمونه های نایی مثبت بودند که در ادامه شناسایی سروتیپ ها بر اساس پروتکل های موجود انجام شد. در این مطالعه مراحل استخراج و RT-PCR بر روی نمونه های مشکوک مثبت چندین بار تکرار شد. می توان نتیجه گرفت که کرونا ویروس در بین گله های گوشتی کشور در حال چرخش است. در طول مراحل آزمایش از دو کنترل منفی شامل کنترل منفی استخراج و کنترل منفی واکنش RT-PCR استفاده گردید. آنالیز ژنتیکی بر اساس توالی نوکلئوتیدی بخش بسیار تغییر پذیر ژن S1 نشان داد که میزان کلی عفونت 65-70% بود و این ژنوتیپ ها با این درصد مشخص شد: (واریانت2 (IS/1494), 52.3%، 793/B  33.5% , MASS 9.5%، QX 4.7%)

    چکیده
    آدنوویروس های پرندگان به طور گسترده ای در گله های تجاری جهان پراکنده هستند و گستره ی وسیعی از حدت و علائم بالینی را نشان می دهند. برخی جدایه های آدنوویروس مرغی می توانند عامل بیماری هپاتیت به همراه گنجیدگی (IBH) باشند. اهمیت حضور ویروس عامل IBH علاوه بر خسارات اقتصادی، همراهی آن با سایر بیماری های ویروسی هست. تضعیف سیستم ایمنی به دنبال درگیری با ویروس بیماری بورس عفونی (IBDV) بیماری زایی ویروس عامل IBH را تسهیل می کند و علاوه بر این، درگیری هم زمان ویروس کم خونی عفونی ماکیان (CIAV) با ویروس عامل IBH میزان جراحات قلبی و تلفات را افزایش می دهد. میزان مرگ و میر در بیماری ویروس عامل IBH ممکن است به 10 ٪ برسد، اما در بعضی موارد تا 30 ٪ هم گزارش شده است. مطالعه حاضر برای اولین بار به شناسایی ویروس IBH به روش واکنش زنجیره ای پلیمراز (PCR) و حضور ژن هگزون در ناحیه متغیر LOOP1 در ماکیان گوشتی استان اصفهان پرداخته شد. در این راستا 69 نمونه کلینیکی از 35 مرغداری مختلف در هشت شهرستان استان، با علائم کبدی موردبررسی قرار گرفت، درمجموع پنج نمونه مثبت (25/7 ٪) از سه شهرستان کاشان، اردستان و گلپایگان در آزمایش PCR تایید شدند و وجود آلودگی برای اولین بار در استان اصفهان تایید گردید.

    Achillea millefolium is one of the oldest known medicinal plants, due to the presence of certain chemical compounds are used traditional and modern medicine. One of the most important medicinal properties of Yarrow, its antimicrobial effects on a wide range of pathogens in humans and animals. The in vitro antimicrobial and antioxidant activities of the essential oil from leaves of Achillea millefolium subsp. millefolium were examined. GC-MS analysis of the essential oil resulted in the identification of fifty-eight compounds constituting 98.71% of the total essential oil of leave. The essential oil of leaves were characterized by a high number of monoterpenes borneol (12.32%), 1, 8-cineole (10.51%), β -pinene (9.33%) and α -pinene (8.82%). Regarding anti-oxidation, the investigated essential oil from leaves strongly reduced the diphenylpicrylhydrazyl radical (IC50=0.94 mg/ml). This essential oil showed antimicrobial activity as well against Staphylococcus aureus, Salmonella enteritidise, Escherichia coli, Penicillium glaucum and Saccharomyces cervesiae (MIC values 0.45-7.20 mg/ml). This study confirms that the essential oil from leaves of Achillea millefolium possessed antioxidant and antimicrobial properties in vitro.

    The study was carried out to determine the reducing microbial load status of broiler carcasses throughout of cold water chilling, hydrogen peroxide solution and ozone therapy. A total number of 45 broilers carcasses from automatic poultry slaughter plant in Chaharmahal and Bakhtiari Province taken from one batch (The same flock) and it was expected to have a high degree of similarity after complete preparation (Slaughtering-scalding-defeathring evisceration) and just before placing in the chillers. Fifteen carcasses were placed in the ordinary chiller containing cold water for 60 min, other 15 were placed in another similar chiller containing aqueous solution of 0.2% hydrogen peroxide and others were treated by ozone (7 mg per min) for the same time period. Total colony count, mould and yeast count, coliform count and Staphylococcus aureus counts were determined in all groups. The reduction percent in the microbial counts were detected. The result showed that highly significant reduction in the numbers of total colony count, coliform count and Staphylococcus aureus count and non significant decrease in the total mould and yeast count in comparison with the untreated ones. Data from this study showed that hydrogen peroxide and ozone therapy induced microbial. Additionally illustrate a very high reduction percent in the total counted colonies, coliform count and Staphylococcus aureus count and little reduction in the total mould and yeast count as a result of addition of hydrogen peroxide into the water used for chilling broiler carcasses. It was concluded that hydrogen peroxide and ozone therapy indicate superior activity as a powerful and versatile sanitizer for poultry carcass.

    Present study was designed in order to molecular confirmation, capsular typing and detection of 12 important virulence factors include ompH, oma87, sodA, sodC, hgbA, hgbB, exBD-tonB, nanB, nanH, ptfA, pfhA and toxA in 18 Pasteurella multocida avian isolates from different areas of Iran with a strain of the organism was isolated from the epidemic pasteurellosis in the Northern region, and also survey of acute isolates lethality in poultry embryos and pullet. Molecular confirmation of tested isolates was performed using specific primers of kmt1. As well, capsular typing and detection of virulence factors of isolates were performed by application of multiplex PCR method and using specific primers of capsule biosynthesis genes and genes encoding virulence factors of organism. All isolates were confirmed molecularly but based on the results of capsular typing, 16 (88.8%) isolates with strain isolated from epidemic pasteurellosis were identified as type A and 2 (11.1%) as untypeable isolates. Detection of virulence genes showed that all studied isolates has six virulence genes of sodC, hgbA, hgbB, nanB, nanH and ptfA. The oma87, exBD-tonB, ompH, sodA and pfhA genes were respectively present in 94.4, 94.4, 83.3, 66.6 and 27.7% of the isolates, but there was not any toxA positive isolates. To determine the chicken embryo lethal dose 50 (CELD50) and lethal dose 50 (LD50) of the isolates, different dilutions of five acute isolates containing the majority of virulence genes as well as strain isolated from epidemic pasteurellosis were respectively inoculated to embryonated chicken eggs and lying pullets. Based on the results of CELD50, though all six isolates were able to waste the poultry embryos, but strain isolated from epidemic pasteurellosis was recognized as the most acute isolate. Results of LD50 test showed that none of the five acute isolates were able to waste the studied pullets while the LD50 of strain isolated from epidemic pasteurellosis was determined 5 × 101 colony forming unit per bird (cfu/case), and this strain was identified as the most acute understudy isolate. The findings of this study showed that, despite the presence of the majority of virulence genes in P. multocida avian isolates in Iran, and that the most acute isolates were able to waste the poultry embryos but they were not infectious to lying pullets, and virulence of strains isolated from birds with hyperacute form of the disease is far higher. Our data could be useful in the design of new and efficient vaccines that would be acceptable protection in animal populations.

    The aim of the present study was to evaluate the antimicrobial effects of aqueous, ethanol, methanol, acetone, chloroform and ethyl acetate extracts of Aloe vera leaves on two fish infectious bacteria, Vibrio cholerae and Listeria monocytogenes. The Aloe vera extracts obtained through rotary apparatus and V. cholerae and L. monocytogenes provided from Iranian fungi and bacterial collection. Minimum bactericidal concentration (MBC) and minimum inhibitory concentration (MIC) of the extracts were determined using dilution in well method. Moreover, the diameters of inhibition zones was determined through disk diffusion method. Results of this study showed while aqueous extract of Aloe vera had inhibitory and lethal effects on L. monocytogenes, but have no influence on V. cholerae. The most inhibitory and lethal characteristics of studied extracts on both bacteria was due to chloroform solvent, while had more influence on L. monocytogenes. Chloroform extract tend its inhibitory property even in the most diluted form. Like inhibitory properties, the biggest diameters of inhibition zones was from chloroform extract against L. monocytogenes. Acording to edibility of Aloe vera, its availability and reasonable value, it is possible to use Aloe vera as an alternative for conventional antibiotics and chemical disinfectants in aquaculture rearing pools.

    Lactococcus garvieae, the causative agent of lactococcosis, is one of the most important fish pathogens all around the world. The aim of this study was to investigate the inhibitory effects of Satureja bachtiarica and Satureja khuzestanica essential oils against the expression of capsule gene (epsD) of L. garvieae. Bacterial isolates containing epsD gene were exposed to the minimum inhibitory concentrations of the essential oils (0.12 μl/ml) and the expression of epsD gene was investigated by real-time PCR. The results showed that the expression of epsD gene was suppressed in the studied bacterial isolates under MIC concentration of the essential oils. The results also revealed that the gene expression was decreased by increasing the essential oil concentration. However, S. khuzestanica was more effective on the expression of epsD gene than S. khuzestanica. Therefore, the use of S. bachtiarica and S. khuzestanica essential oils can be recommended to prevent lactococcosis in aquaculture via suppression of capsule gene.

    Bacterial microflora in water and fish body surface has an important role for fish bacterial diseases outbreak and are known as secondary bacterial infections. These bacteria can be pathogenic and develop high mortality in stress conditions. In this study, samples were collected from body surface and gills of 70 rainbow trout (300-500 g) in seven fish farms on Haraz road between Larijan and Poloor regions by sterile swabs. Water resource was Haraz River. Skin and gill swabs were transferred to the lab in capped tubes containing sterile PBS (Phosphate Buffer solution), individually and were cultured in TSA (Tryptic Soy Agar), Plate Count Agar and MacConkey Agar media. Each sample were cultured in two TSA medium plates for conserving in aerobic and anaerobic conditions in 25°C, and one MacConkey Agar plate to reassure of gram negative bacteria absence. Plate count agar culturing was applied for mesophilic and psychrophilic bacteria isolation in 13 and 37° C. Nine bacterial species were isolated from gills and fish body surface (including: E.coli, Staph. areus, Staph. saprophyticus, Staph. epidermidis, Bacillus cereus, Aeromonas hydrophila, Proteus mirabilis, Pasteurella multocida, Psuedomonas aeruginosa). E. coli were identified in all samples. There was no significant difference between gills and skin in all facilities (P>0.05). Among the facilities, there was no significant difference between isolated bacterial species (P>0.05). Except E.coli that was identified in all farms, and irregularity frequency for P. aeruginosa in farms, in other cases, bacterial frequency increased up to downstream fish farms. Isolation of such bacteria may be an indicator for urban and home waste water contamination.

    Toxoplasma gondii is an obligatory intracellular protozoan which can infect broad spectrum of warm blooded animals as intermediate hosts. Cats are definitive hosts. Seroprevalence of this parasite among birds is an effective criterion for environmental contamination to Toxoplasma oocysts. In this research, T. gondii Immunoglobulin G (IgG) were detected by ELISA in 168 broilers sera which were referred to Semnan industrial abattoir and 30 freerange chicken sera which were collected from different places in Semnan. 39.9% of broilers and 96.7% of free-range chickens were positive in Toxoplasma IgG antibody. There was significant difference between two different groups in Chi-square test (P<0.0001). According to the results, seroprevalence of Toxoplasma is relatively high among chickens in Semnan. In free-range chickens, soil contamination to the parasite oocysts and in broilers some management problems and cat access as a definitive host to the water and food supply of chickens can effective on high prevalence of the infection.

    In this article; 42 repeated breeders (test group), and 18 healthy (control group) and on heat Holstein cows were prepared in a farm of Khorasan-Razavi province. Their blood samples were obtained from the coccygeal vein and the isolated serum samples were frozen and sent to a laboratory, together with the heat discharge samples. In the laboratory; estradiol concentrations of serum samples were determined with RIA technique, and microbiology and the other discharge characteristics were determined grossly (transparency and stretching levels), or with a light microscope (fernization and crystals regularity). Results showed that; the mean of stretching levels (cm) (3.15±0.2, 3.25±0.3, 3.1±0.4, 3.3±0.3, P=0.9), the median of simple fernization (2.5, 3, 2, 3, P=0.3), the median of fernization levels evaluated with Photoshop software (3, 3, 4, 4, P=0.1), the crystals regularity (2, 2, 2, 2, P=.85), the median of levels of transparency (2, 1.5, 2, 2, P=0.06), the mean of concentrations of estradiol-17-β (pg/ml) (8±2.4, 4.7±1.7, 4±1.5, 5.3±0.3, P=0.7) were determined in non-pregnant (N=34) and pregnant (N=8) repeat breeder, and non-pregnant (N=10) and pregnant (N=8) normal cows, respectively. Spearman correlation test showed only a significant correlation between fernization levels evaluated with Photoshop software and different (test and control) animal groups (r= - 0.3, p=0.017). So that the third degree of fernization (equivalent to 40-60% of fernization) was considered the best degree, and the lower or the higher than it considered as unsuitable degrees. There was no significant correlation between the result of microbiologic findings and fernization, transparency, stretching levels and crystals regularity (P>0.05). However, E. coli, Klepsiella, Trueperella pyogenes and beta-hemolytic Staphylococci were significantly more isolated from repeat breeder cows (P=0.00).     One of the important factors causing the loss of fish eggs in the hatcheries is contamination with Saprolegnia parasitica. Today, Green Malachite is the most widely used drug against this fungi, which has been confirmed to be carcinogenic. The aim of this study is to investigate the antifungal effects of Matricaria chamomilla and Achillea officinolis extracts to control of infected rainbow trout eggs with Saprolegnia parasitica in comparison with Green Malachite. In this study minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) were mesured by Micro Broth Dilution test and the growth of fungi in a solid medium, and determined as 100 and 150 mg/mL for M. chamomilla and 150 and 200 mg/mL for A. officinolis, respectively. The eggs of rainbow trout were divided into four treatments (control, two plant extract and Green Malachite treatments) and were exposed with MFC of each extract and 1 mg/L Green Malachite for 1 hour/day during incubation period. The mortality of eggs were compared in stages of fertilization to sighting, sighting to hatching and total incubation period. The results showed that the mortality of eggs in each stage in Green Malachite treatment was significantly lower than control and A. officinolis (p˂0.05), but did not show difference with M. chamomilla treatment (p>0.05). The lack of significance difference of mortality and hatching percentage in M. chamomilla (75.90±0.26%) compared to Green Malachite (77.97±2.32%) showed that M. chamomilla extract with antifungal effects is a suitable natural remedy for replacing Green Malachite in fish eggs hatcheries.

    Escherichia coli is one of the most important bacterial pathogens of human and poultry. Avian pathogenic Escherichia coli (APEC) and uropathogenic Escherichia coli (UPEC) isolates cause diseases outside intestine environment. These strains share some common traits. It has been suggested that transfer of these commonalities between different hosts is of importance in the epidemiology of disease in poultry and human. In the present study, presence of 4 virulence associated genes (sitA, iutA, traT, and tsh) were investigated among 26 APEC and 25 UPEC isolates by polymerase chain reaction (PCR) assay. Statistical analysis was also carried out using minitab 15. The frequencies of the presence of these selected genes in APEC and UPEC isolates were 96.2% and 64% for traT, 88.5% and 76% for sitA, 84.6% and 68% for iutA, and 61.5% and 16% for tsh, respectively. traT and sitA were the most prevalent genes among APEC and UPEC isolates, respectively. Diverse combinations of virulence associated genes were also detected among these strains. Combinations of virulence genes of iutA-sitA-tsh-traT and iutA-sitA-traT were the most frequent in APEC and UPEC isolates, respectively. It supports possibility of virulence genes transfer between poultry and human. Further studies are needed to ascertain the importance of such possibilities in the epidemiology of the infection in human and poultry.

    Infectious bronchitis virus (IBV) is the cause of acute respiratory viral disease in poultry with respiratory rats and coughing and sneezing. Corona viruses are created a wide range of respiratory, intestinal, liver and nervous disease with different virulence in cats, dogs, pigs, rodents, cows, birds and human. Corona viruses are able lots of recombination's and mutations. This ability gives them the opportunity to be active in different hosts and different climatic conditions, such as avian infectious bronchitis disease in poultry that can cause losses in broilers. S1 gene sequencing is used for molecular epidemiology and genotype characteristics of infectious bronchitis virus. For better understand the molecular epidemiology of infectious bronchitis virus in Iran, the IBVs gene isolates of broiler poultries from Ardebil province were sequenced in this study. A total of 30 tracheal swabs were collected from broilers in random in Ardebil province in 2016. For detection and genotyping of virus, RNA extraction was performed using RNA extraction kit. RT-PCR was used in one process for the production of cDNA, and then, RT-PCR product were amplified to identify infectious bronchitis virus using Nested-PCR with primers encoding 5’UTR, then, for genotyping of positive samples, gene sequencing of spike glycoprotein was performed 70% of tracheal swabs were positive and strain identification was done according to existing protocols. It can be concluded that the corona virus is spinning among broiler flocks in Iran. In this study, extraction and RT-PCR on suspected positive samples was repeated several times. During the test two negative controls was used including extraction negative control and RT-PCR negative control. Genetic analysis based on the highly variable nucleotide sequence of S1 gene showed that, the overall rate of infection was 70% and detected three genotypes in this area: Variant 2 (IS/1494), 793/B and MASS and QX overall percentages in respectively 52/3%, 33/5%, 9/5% and 4/7%.

    Avian Adenoviruses are widely distributed in commercial flocks in the world and show wide variety of virulence and clinical signs. Some of isolates of fowl adenoviruses can cause Inclusion Body Hepatitis (IBH). The importance of IBH agents are economic losses and its association with other viral diseases. Immunosuppression after Infectious Bursal Disease (IBD) can facilitate disease of IBHV.Also simultaneous infection with chicken infectious anemia virus(CIAV) and IBH increase the heart injuries and mortality. Mortality rate in IBH can reach to 10% but sometimes it can be over 30%. In this study identification of IBHV by PCR method and presence of Hexon gene in variable area of Loop1 was studied in Isfahan province for the first time. 69 liver samples from suspected 35 flocks and cities gathered and sent to laboratory. Five samples from Kashan, Ardestan and Golpayegan were positive in PCR. So presence of IBHV confirmed in this province.
