    ویروس سوختگی سیاه برگ چغندرقند (Beet black scorch virus, BBSV) یک ویروس جدید خاک زاد چغندرقند می باشد. به منظور ساخت سازه عفونت زای این ویروس و بررسی واکنش برخی ارقام چغندرقند به آن، نمونه های علائم ریشه ریشی در ریشه، از مزرعه ای در مشهد جمع آوری شد. در این نمونه با استفاده از آزمون زنجیره ای پلیمراز با ترانویسی معکوس دو ویروس BBSV و Beet necrotic yellow vein virus (BNYVV) ردیابی گردید. پس از خالص سازی و تکثیر ژنوم کامل BBSV، همسانه عفونت زای آن در ناقل pDrive ساخته شد. آران ای سنتز شده از همسانه عفونت زا توسط فاژ T7، برای آلوده سازی مکانیکی گیاه محک سلمه تره استفاده شد. به منظور آلوده سازی مکانیکی برگ گیاهچه های چغندرقند، از برگ های دارای علائم کلروتیک گیاه محک، به عنوان مایه تلقیح اولیه استفاده شد. در این تحقیق از ارقام چغندرقند داخلی شامل: شریف، آریا و شکوفا و ارقام خارجی شامل: Izabella، Pauletta و Dorothea، با تنوع ژنتیکی متفاوت کشت شده در شرایط گلخانه، استفاده شد. نتایج این بررسی نشان داد که BBSV می تواند سه هفته پس از مایه زنی، علائمی به صورت نقاط نکروتیک ریز روی برگ گیاهچه همه ارقام ایجاد نماید؛ بنابراین سازه عفونت زای ساخته شده می تواند به عنوان یک ابزار مطمئن برای غربال مقدماتی ژرم پلاسم های چغندرقند (قبل از شروع برنامه های بهنژادی جهت تهیه ارقام مقاوم به این ویروس) تحت شرایط گلخانه، مورد استفاده قرار گیرد. از آنجا که تاکنون هیچ ژن یا ژن های مقاومی در چغندرقند نسبت به BBSV گزارش نشده است، می توان با استفاده از این روش سعی در یافتن منابع مقاومت به این ویروس در چغندرهای وحشی و زراعی نمود.

    آلودگی قارچی بذر برنج به گونه های فوزاریوم مشکل بزرگی برای کشاورزان در دنیا و ایران می باشد. شناخت قدرت تهاجمی گونه های فوزاریوم مزارع برنج برای توسعه استراتژی مدیریت بیماری های ناشی از آنها مفید و موثر است. در این پژوهش بیماری زایی 47 جدایه از گونه های Fusarium fujikuroi، F. incarnatum، F. proliferatum، F. thapsinum، F. andyazi، F. oxysporum، F. graminearumو F. merismoidesکه از بذور برنج استان های مازندران، گیلان، گلستان، خوزستان و فارس جداسازی شدند، مورد بررسی قرار گرفت. بیماری زایی به روش آغشتگی بذر برنج به سوسپانسیون کنیدیوم ها در آزمایشگاه، خزانه و مزرعه برنج بررسی گردید. درصد گیاهچه های سالم، بیمار و مرده، مشاهده و ثبت شدند و درصد وقوع بیماری تعیین شد. همچنین میزان انتقال بیماری پوسیدگی طوقه توسط بذر به خزانه و مزرعه برنج تعیین شد. نتایج نشان داد که همه جدایه های مورد بررسی، روی گیاهچه برنج بیماری زا بودند و درصد جوانه زنی را در مقایسه با شاهد کاهش دادند و فقط تک جدایه F. merismoidesغیر بیماری زا بود. جدایه های F. fujikuroi بالاترین شاخص وقوع بیماری را داشتند. همچنین بین گونه های Fusarium، جدایه های درون هر گونه و جدایه های به دست آمده از پنج استان از نظر شاخص وقوع بیماری اختلاف معنی داری در سطح یک درصد وجود داشت. بر اساس میزان وقوع بیماری، 1/2 درصد جدایه ها در گروه غیر بیماری زا، 6/25 درصد جدایه ها در گروه بیماری زای ضعیف،1/34 درصد بیماری زا و 2/38 درصد در گروه بیماری زای شدید قرار گرفتند. در این پژوهش، میزان انتقال بیماری توسط بذر به خزانه، 38 درصد بود. اما پس از ضد عفونی بذر با قارچکش، میزان انتقال بیماری به خزانه از 38 به 6/3 درصد کاهش یافت. این نتایج اهمیت ضد عفونی بذر برنج با قارچکش مناسب برای مبارزه با بیماری پوسیدگی طوقه را نشان می دهد.

    بیماری شانکر باکتریایی ایجاد شده بوسیلهClavibacter michiganensis subsp michiganensis  یکی از بیماری های اقتصادی مهم گوجه فرنگی است. در این مطالعه، پس از جدا سازی و تشخیص بیمارگر، اثرات پیش‏تیمار بتا-آمینوبوتریک اسید (BABA) علیه بیماری شانکر باکتریایی در شرایط آزمایشگاهی مورد بررسی قرار گرفت.  BABAبه عنوان یکی از مواد شیمیایی توانمند القا کننده مقاومت در گیاهان بر علیه عوامل بیماریزا شناخته می شود. در این مطالعه گیاهان گوجه فرنگی با غلظت 2/0 میلی‏مولار BABA و آب مقطر سترون به عنوان شاهد تیمار شدند، سپس با جمعیت 108×1 سلول باکتری در هر میلی لیتر مایه زنی شدند. نتایج آزمایشات در فواصل زمانی مختلف نشان داد که پیش‏تیمار BABA، جمعیت باکتری و شدت بروز علائم در گیاهان مایه‏زنی شده را بطور معنی داری در مقایسه با شاهد کاهش داد، افزون بر این ظهور دیرتر علائم بیماری بیانگر دوره کمون طولانی تر در گیاهان تیمار شده توسط BABA است. همچنین افزایش قابل توجه بیان ژن‏های PR1 و کاتالاز در گیاهان تیمار شده مشاهده شد. بر اساس نتایج ما و با توجه به عدم وجود راه‏حل مدیریتی مناسب علیه این بیماری، توصیه‏ی به‏کارگیری BABA به عنوان یک عامل سازگار با محیط زیست، سهم ارزشمندی در مدیریت این بیماری خواهد داشت.

    ویروس های زردی غلات از مخرب ترین ویروس های غلات در سراسر دنیا هستند. در این تحقیق پروتئین های P0، سرکوبگرهای خاموشی، در دو ویروس کوتولگی زرد غلات، Cereal yellow dwarf virus-RPV (CYDV-RPV) وCereal yellow dwarf virus- RPS (CYDV-RPS) از نظر محل تجمع و قرارگیری در سلول های گیاه توتون مورد بررسی قرار گرفتند. بدین منظور ترادف ژن های P0 پس از تکثیر در آزمون زنجیره ای پلیمراز (PCR)، با روش Gateway همسانه سازی شدند. سپس سازه های این ژن ها در اتصال با ژن کد کننده green fluorescent protein (GFP) وارد باکتری Agrobacterium tumefaciens شدند و با تزریق باکتری حامل این سازه ها به گیاه به صورت موقتی بیان شدند. پس از 24 ساعت از زمان تزریق، محل قرارگیری این پروتئین ها در سلول های گیاه توسط میکروسکوپ کانفوکال (confocal) مورد بررسی قرار گرفت. نتایج نشان داد که محل تجمع پروتئین P0 در هر دو ویروس در هسته و سیتوپلاسم است اما تمایل آنها برای تجمع متفاوت است. به عبارت دیگر پروتئین P0 در ویروس RPV CYDV- در مقایسه با CYDV-RPS تمایل کمتری برای تجمع در هسته دارد در حالی که پروتئین P0 در ویروس CYDV-RPS بیشتر در هسته متمرکز است. در آزمون لکه برداری وسترن با استفاده از آنتی بادی پلی کلونال GFP، پروتئین های GFP:P0 ردیابی شدند ولی پروتئین GFP به تنهایی دیده نشد که نشان می دهد محل های تجمع دیده شده مربوط به پروتئین های GFP:P0 است.

    در این مطالعه قابلیت بازیابی، برخی خصوصیات ریخت شناسی و نیز شدت بیماریزائی دوازده جدایه ی قارچ عامل بیماری آنتراکنوز گردو سیزده سال پس از نگهداری بر روی کاغذ صافی در محیط کشت آرد یولاف-آگار  (OMA)و در دمای 20- درجه سلسیوس، مورد ارزیابی قرار گرفت. رشد موفق جدایه ها از صفر تا 89 درصد متغیر بوده و 75 درصد نمونه های ذخیره شده، قدرت زنده مانی و بقای خود را در طی سال های نگهداری حفظ کرده بودند. در بررسی های ماکروسکوپی و میکروسکوپی کشت های بازیابی شده، خصوصیات ظاهری پرگنه در همه جدایه ها تقریبا یکسان بود. کلیه جدایه ها توانائی تشکیل اندام های تولیدمثل غیرجنسی (آسروول حاوی کنیدیوم) را همچنان دارا بودند. در کشت های خالص قارچ، تشکیل آسکوکارپ بالغ سه ماه پس از نگهداری جدایه ها در دمای 10-8 درجه سلسیوس و تاریکی مشاهده نشد. براساس نتایج آزمون بیماریزائی، همه جدایه ها قادر به ایجاد بیماری روی دانهال های گردو بوده و اولین علائم بیماری را 16-12 روز پس از مایه زنی در گلخانه نشان دادند، اما بین جدایه های مورد مطالعه تنوع زیادی از نظر درصد آلودگی وجود نداشت. پایداری صفات ریخت شناسی، اسپورزائی و حفظ قدرت بیماریزائی جدایه ها پس از مدت طولانی نگهداری در محیط OMA نشان داد که انتقال اندام های غیرجنسی قارچ روی کاغذ صافی در محیط مذکور، می تواند روش مناسب، آسان و اقتصادی برای نگهداری طولانی مدت بیمارگر در یک کلکسیون کشت آزمایشگاهی باشد.

    گونه یPhytophthora erythroseptica ، عامل پوسیدگی صورتی سیب زمینی، یکی از اامیست های بیماری زای گیاهی است که خسارت اقتصادی قابل توجهی را در مزرعه و انبار وارد می کند. به منظور شناسایی و ردیابی دقیق و حساس P. erythroseptica ، شش توالی هسته ای و میتوکندریایی برای طراحی آغازگرهای اختصاصی مورد بررسی قرار گرفت. به دلیل شباهت زیاد توالی های P. erythroseptica به خویشاوندان نزدیکش، تنها یک توالی هسته ای (TigA) برای طراحی آغازگرهای اختصاصی مناسب تشخیص داده شد. با استفاده از آغازگرهای اختصاصی طراحی شده، شیوه ای از واکنش زنجیره ای ساده و تودرتو برای شناسایی و ردیابی P. erythroseptica ابداع شد. اختصاصیت آغازگرهای طراحی شده با استفاده از مجموعه ای از گونه های فیتوفتورا متعلق به تبارهای فیلوژنتیکی مختلف و نیز خویشاوندان نزدیک P. erythroseptica ارزیابی شد. ردیابی P. erythroseptica در دی ان ای خالص بیمارگر و دی ان ای استخراج شده از بافت های گیاهی آلوده شامل سیب زمینی، گوجه فرنگی و اسفناج با استفاده از آغازگرها با موفقیت انجام شد. آغازگرهای اختصاصی10 پیکوگرم از دی ان ای خالص P. erythroseptica را در واکنش زنجیره ای پلیمراز ساده ردیابی کردند، با این حال واکنش زنجیره ای پلیمراز تودرتو، حساسیت آغازگرها را حداقل صد برابر افزایش داد. همچنین، آغازگرهای اختصاصی قادر به ردیابی P. erythroseptica به عنوان والد پدری یا مادری در جدایه های دورگ بودند؛ این ویژگی به شناسایی یکی از والدین در دورگ های P. erythroseptica کمک شایانی می کند

    Beet black scorch virus (BBSV) is a soil-borne virus infecting sugar beet. In order to construct an infectious full-length cDNA clone of Mashhad isolate (IR-Mash1), Iran (GenBank Accession no. MK092329), and studying reaction of some sugar beet cultivars, sugar beet roots with beard lateral root growth were collected from Mashhad field and subjected to total RNA extraction and then cDNA synthesis. PCR reaction was performed to amplify the whole genome of the virus were cloned to pDrive Cloning Vector. DNA was extracted from the plasmid vector. It was used as templates for run-off transcription in vitro by use of a T7 RNA polymerase. Chenopodium quinoa was inoculated under greenhouse conditions. Necrotic local lesions formed on C. quinoa (5 dpi) leaves were used as inoculation material for inoculation of the leaves of different sugar beet cultivars grown in the greenhouse. BBSV was inoculated by rubbing the sugar beet seedling leaves. Iranian cultivar including Sharif (susceptible check), Aria, Shokoufa and foreign cultivar including Pauletta KWS (tolerant to Rhizomania and sugarbeet cyst nematode), Isabella KWS (Double Resistant to Rhizomania) and Dorothea (tolerant to Rhizoctonia) cultivars were inoculated under greenhouse conditions. Pinpoint Symptoms appeared on the leaves of all cultivars after twenty days. The result showed that the virus produces similar symptoms on leaves of susceptible and resistant cultivars to Rhizomania. So, using infectious clone, possible to screening sugar beet germplasms as part of pre-breeding programs under greenhouse conditions and to find sources of resistance to the virus in sugar beet and wild beets.

    The fungal infection of rice seeds with Fusarium species, is a major problem for farmers in Iran and the world. Determining the pathogenicity of Fusarium species in rice fields might be helpful in development of strategies for management of the relevant diseases. In this research, the pathogenicity of forty seven isolates of Fusarium belonged to Fusarium fujikuroi, F. incarnatum, F.prolifratum, F. thapsinum, F. andyazi, F. oxysporum, F. graminearum and F. merismides obtained from rice seeds in Mazandaran, Guilan, Golestan, Khuzestan and Fars provinces were investigated. The pathogenicity test was performed by rice seed dressing method using spore suspension as inoculum in invitro, nursery and the paddy field. Percentage of healthy, unhealthy and dead seedlings recorded and percentage of disease incidence was determined. Also, the transmission rate of bakanae disease by seeds from the laboratory to the nursery and the paddy was determined. The results showed that all isolates were pathogenic on rice seedling and reduced germination percentage compared to control, and only one isolate of F. merismoides was non-pathogenic. F. fujikuroi isolates had the highest disease incidence rate. In addition, there was a significant difference among Fusarium species, isolates of each species and the five provinces in terms of disease incidence. Based on the disease incidence, 2.1% of the isolates placed in the non-pathogenic group, 25.6% of the isolates were in the moderate pathogenic, 34.1% of the isolates were pathogenic and 38.2% considered as severe pathogenic group. In this research, the rate of disease transmission from contaminated seeds to the nursery was 38%., but after seed disinfection, with fungicide, the transmission rate of the disease dropped from 38% to 3.6%. These results show the importance of rice seed treatment with fungicides for the control of bakanae disease.

    Bacterial canker caused by Clavibacter michiganensis subsp. michiganensis is an economically important disease of tomato. In this study after isolation and characterization of the pathogen, effects of tomato pre-treatment with DL-β-aminobutyric acid (BABA) on bacterial canker was investigated under laboratory conditions. BABA is known as a powerful chemical resistance inducer in plants against diverse pathogens. In this study, tomato plants were treated with a 0.2 mM concentration of BABA and distilled water served as control. The plants were subsequently challenged with a 1×108 CFU ml−1 bacterial suspension. The results of experiments in different time intervals showed that BABA pre-treatment resulted in a significant reduction of bacterial population and symptoms severity of inoculated plants as compared to their controls. Furthermore, late appearance of disease symptoms indicated longer incubation periods for BABA treated plants. Also, a significant increase was observed in the expression of PR1 and catalase genes in treated plants. Based on our results and regarding the lack of proper management options against the disease, application of BABA as an environmentally safe agent is recommended as a valuable contribution to disease management.

    Cereal yellow dwarf viruses are among the most destructive cereal viruses worldwide. In this study, subcellular localizations of silencing suppressor proteins ( P0) from two cereal yellow dwarf viruses, Cereal yellow dwarf virus-RPV (CYDV-RPV) and Cereal yellow dwarf virus-RPS (CYDV-RPS), were examined via confocal microscopy. To do so, P0 genes were synthesized by polymerase chain reaction (PCR) and were cloned through Gateway cloning system. Agrobacterium. tumefaciens clones harboring GFP:P0 constructs were infiltrated into Nicotiana benthamiana plants. Transient expression of P0 proteins with N-terminal GPF fusion enabled us to examine their subcellular localizations in plant cells, 24 hours post infiltration. Results showed that both proteins are nuclear-cytoplasmic but different tendencies were observed in their localizations. In other words, P0 of CYDV-RPS was more nuclear oriented than P0 of CYDV-RPV. In the Western blot analysis using GFP polyclonal antibody only GFP:P0 proteins but no free GFP, were detected indicating that localizations were related to GFP:P0 proteins.

    In this study survivability, morphological characteristics and pathogenicity of 12 isolates of Ophiognomonia leptostyla, which is known as causal agent of walnut anthracnose, have been evaluated after 13-years of storage on filter paper in oat meal agar (OMA) and -20°C. The recovery rate varied from 0 to 89 percent and seventy-five percent of stored isolates were shown to be viable. In macroscopic and microscopic studies of revived cultures, the morphology of colonies was uniform for all isolates. Production of asexual bodies (acervular conidiomata) was preserved by all isolates. Fertile perithecia with asci and ascospores were not detected in pure fungal cultures after three months incubation at 8-10°C in darkness. In pathogenicity experiments, there was not a significant variability in infection severity for different isolates, and the first symptoms were observed on leaves 12-16 days after inoculation. Stability of morphological characteristics, ability to sporulation and retaining the pathogenicity of isolates confirmed that transferring of acervuli on filter paper in OMA medium could be a suitable, safe, simple and cost-effective tool to preserve O. leptostyla isolates in a viable state.

    Phytophthora erythroseptica, the causal agent of potato pink rot, is one of the oomycete plant pathogens that causes significant losses in field and storage. In order to develop a sensitive and rapid method for detection and identification of P. erythroseptica, six nuclear and mitochondrial gene regions were investigated to design species-specific primers. Due to the high similarity of P. erythroseptica sequences to its closely related species, only one nuclear region, TigA, was appropriate to design specific primers. Using specific primers, a simple as well as a nested-PCR based method was developed for the identification and detection of P. erythroseptica. The specificity of designed primers was examined using a collection of Phytophthora species from different phylogenetic clades as well as close relatives of P. erythroseptica. In addition to pure DNA, designed primers detected P. erythroseptica in infected plant tissues including potato, tomato and spinach. Specific primers detected 10 pg of P. erythroseptica pure DNA, however, nested PCR increased primers sensitivity at least 100 times. Moreover, specific primers designed in this study were able to detect P. erythroseptica as the maternal or paternal parent species in hybrid isolates that would make a significant help to recognize one of the parental species in hybrids of P. erythroseptica.
