    پروتئین CD80 به عنوان عضوی از ابرخانواده ایمنوگلوبولین ها، یک پروتئین تراغشایی است که در سطح سلول های ارایه کننده آنتی ژن (APC) بیان می شود. این پروتئین دارای دو گیرنده CTLA-4 و CD28 در سطح سلول های T است. بر اثر اتصال این پروتئین به این گیرنده ها به ترتیب مسیر مهاری و تحریکی در سلول های T آغاز می شود. در حالت طبیعی، پروتئین های CD80 دارای تمایل اتصال بیشتری به CTLA-4 نسبت به CD28 هستند و این از عوامل خاموش کننده سلول های T در سیستم ایمنی، به منظور جلوگیری از خودایمنی است. هدف از مطالعه حاضر، ایجاد واریانتی از پروتئین CD80 است که دارای تمایل اتصال افزایش یافته به CD28 است تا با قدرت بیشتری به این گیرنده متصل شود و مسیرهای تحریکی را بیشتر از نوع وحشی این پروتئین (پروتئین CD80 اولیه) در سلول های T القا کند. برای شناسایی این واریانت ابتدا توالی وحشی با کمک محیط برنامه نویسی R، در جایگاه های 31 و 92 با اسیدهای آمینه ای که نقش مهمی در شکل گیری پیوندهای هیدروژنی دارند، جهش داده شد. 100 توالی خروجی نرم افزار R، با سرور SWISS-MODEL مدل سازی شدند. سپس مدل های خروجی، به صورت تک تک با سرور HADDOCK داک شدند و در نهایت انرژی های اتصالی از جمله انرژی های الکترواستاتیک و واندروالسی بین گیرنده ها و لیگاندها محاسبه شدند. بین تمامی مدل های ساخته شده، توالی جهش یافته K31Y, R92F دارای بهترین انرژی الکترواستاتیک و واندروالسی است و در مقایسه با نوع وحشی پروتئین CD80، با قدرت بالاتری به پروتئین CD28 متصل می شود.

    اهداف

    سرطان سینه شایع ترین نوع بدخیمی زنان در سراسر جهان است. از میان روش های مختلف در پیشگیری و درمان سرطان، ترکیبات طبیعی گیاهی دارای مزایای بیشتری نسبت به داروهای شیمیایی هستند و عوارض جانبی کمتری دارند. اخیرا مطالعات بسیاری در ارتباط با خواص آنتی اکسیدانی، ضدسرطانی، ضدتکثیری و ضدالتهابی لیگنان های گیاهی انجام شده است که نشان دهنده اهمیت این ترکیبات در پیشگیری و درمان انواع بیماری ها است. در مطالعه حاضر اثرات سمیت سلولی و القای آپوپتوز توسط ترکیبات لیگنانی پینورسینول و لاریسی رسینول بر رده سلولی سرطان سینه SKBr3 بررسی شد.

    مواد و روش ها:

     سلول های SKBr3 با غلظت های مختلفی از پینورسینول و لاریسی رسینول به مدت 72 ساعت تیمار شدند. سپس قابلیت حیات سلولی و تغییرات مورفولوژیکی سلول ها به ترتیب با ارزیابی MTT و میکروسکوپ نوری معکوس تعیین شدند. همچنین القای آپوپتوز به وسیله آنالیز فلوسایتومتری و با استفاده از کیت تشخیص آپوپتوز آنکسین V-FITC بررسی شد.

    یافته ها:

     تیمارهای پینورسینول و لاریسی رسینول هر دو در حالت وابسته به غلظت موجب القای تغییرات مورفولوژیکی، کاهش قابلیت رشد، حیات، تکثیر سلولی و افزایش معنی دار میزان القای آپوپتوز در رده سلولی SKBr3 نسبت به سلول ها شدند.

    نتیجه گیری: 

    القای آپوپتوز و جلوگیری از رشد و تکثیر سلول های سرطانی از مکانیزم های مهم در درمان سرطان است. پینورسینول و لاریسی رسینول می توانند از طریق کاهش تکثیر سلولی و افزایش القای آپوپتوز در پیشگیری و درمان سرطان سینه مفید باشند.

    سورفکتانت های زیستی از جمله فرآورده های میکروبی ارزشمند هستند که کاربردهای قابل توجهی در صنایع مختلف دارند. از مزیت های این ترکیبات نسبت به انواع شیمیایی خود می توان به زیست تخریب پذیری، سمیت پایین، فعالیت در شرایط محیطی حاد و قابلیت تولید از پسماندها اشاره کرد. در مطالعه حاضر، توانایی سویه بومی Halomonas sp. MM93 در تولید بیوسورفکتانت با استفاده از روش گسترش روغن و همولیز در محیط کشت نوترینت براث و دمای °C30 پس از 72 ساعت مورد بررسی قرار گرفت. فعالیت امولسیون کنندگی سورفکتانت زیستی نیز در یک محیط تولید تعریف شده طی 96 ساعت، توسط آزمون سنجش فعالیت امولسیون کنندگی بررسی شد. اثر روغن زیتون، ان- هگزان و نفت سفید به عنوان منابع کربنی آب گریز در القای تولید سورفکتانت زیستی توسط این سویه مورد بررسی قرار گرفت. به واسطه اهمیت پایداری سورفکتانت های زیستی در استفاده های صنعتی، پایداری مایع رویی محیط کشت باکتری در شرایط افراطی دما، pH و غلظت نمکی سنجیده شد. این سویه در آزمون گسترش روغن، هاله ای به قطر 5/2سانتی متر ایجاد کرد و شاخص E24 آن برابر 45% محاسبه شد. Halomonas sp. MM93  به واسطه تولید این سورفکتانت زیستی قادر به کاهش کشش سطحی محیط کشت از 70 به 40میلی نیوتون بر متر بود. همچنین سورفکتانت زیستی تولیدشده پایداری قابل ملاحظه ای در دمای بالا (°C100)، شرایط اسیدی و بازی حاد (12-2=pH) و غلظت نمک بالا (20گرم بر لیتر) را نشان داد. طبق نتایج حاصل، باکتری نمک دوست بومی Halomonas sp. MM93 می تواند به عنوان یک سویه توانمند در تولید سورفکتانت زیستی پایدار در صنایع مختلف به ویژه فرآیندهای افزایش بازیافت میکروبی نفت که نیازمند ترکیبات دارای فعالیت سطحی با پایداری زیاد هستند، مورد توجه قرار گیرد.

    پساب حاصل از استفاده آب در مصارف خانگی، صنایع و کارخانه ها سبب عوارضی زیست محیطی می شود که با ایجاد بوی نامطبوع، محیط مناسبی برای رشد باکتری های بیماری زا فراهم می کند. در این پژوهش پس از جداسازی میکروارگانیسم های بومی پساب صنایع عرق گیری گیاهی، توانایی آن ها برای کاهش میزان اکسیژن خواهی زیستی BOD و  میزان اکسیژن خواهی شیمیایی COD پساب بررسی شد.از قسمت های مختلف پساب صنایع عرق گیری کاشان نمونه برداری شد. پس از انتقال نمونه ها به آزمایشگاه، از محیط کشت  لیزوژنی براث، برای جداسازی باکتری ها استفاده شد. جدایه ها بر اساس ویژگی ظاهری و زیست شیمیایی تفکیک شدند. از میان 69 جدایه ها، چهار جدایه برای سنجش توانایی کاهش BOD و COD انتخاب شده و به پساب کارخانه عرق گیری افزوده شدند. کاهش BOD پس از پنج روز با استفاده از دستگاه  BODمتر و کاهشCOD به روش تیتراسیون اندازه گیری شد. جدایه ها بر اساس آزمون های زیست شیمیایی شناسایی شدند.پس از افزودن سویه های  منتخب به پساب، میزان BOD و COD ، به ترتیب به 82/71- 43/47 درصد و 79/44-50/56 درصد کاهش یافت. همچنین با افزودن کنسرسیوم سویه ها به پساب، میزان BOD و COD  به ترتیب 21/83-6/76 و 29/57-32/38 درصد کاهش یافت.طبق نتایج بدست آمده، پساب حاوی سویه های باکتریایی است که توانایی بالایی در کاهش میزان BOD وCOD  و همچنین تجزیه مواد آلی موجود در پساب دارند، به گونه ای که با استفاده مجدد از پساب تصفیه شده، امکان سرمایه گذاری در بخش کشاورزی و صنعت وجود دارد. بنابراین می توان از این سویه ها برای تصفیه پساب صنایع عرق گیری گیاهی استفاده نمود.

    کیندلینگ الکتریکی یکی از محبوب ترین تکنیک های مدل صرعی است که باعث ایجاد تشنج هایی مانند صرع لوب گیجگاهی می شود. تاکنون برای درمان صرع، از تکنیک های درمانی مختلفی استفاده شده است. از بین این تکنیک ها، تحریک الکتریکی با فرکانس پایین (LFS) ، به طور گسترده ای برای بهبود صرع مقاوم به دارو مورد توجه قرار گرفته است، اما مکانیزم اثر آن به خوبی روشن نشده است. از آنجا که وارد شدن کلسیم به سیتوپلاسم و افزایش غلظت آن یکی از مهمترین دلایل بروز تشنج است، گیرنده متابوتروپیک گلوتامات (mGluR1)، گیرنده دوپامین (D1) و ADPR سیکلاز (CD38)، سه گیرنده ای که از مسیرهای مختلف باعث افزایش کلسیم در سیتوپلاسم می شوند، انتخاب شدند. با این هدف که با بررسی تغییر بیان این گیرنده ها کمک به سزایی به مشخص شدن مسیر بهبود بخشی LFS شود. در این مطالعه از هیپوکمپ موش های صحرایی استفاده شد و تغییر بیان ژن ها با تکنیک real-time PCR مورد بررسی قرار گرفت. نتایج نشان داد که بیان هر سه ژن انتخابی پس از کیندلینگ به طور معنی داری افزایش یافته و بعد از اعمال تحریکات LFS میزان بیان هر سه به مقدار کنترل بازگشته است. از این رو یکی از مسیرهایی کهLFS  برای بهبود بخشی تشنج های کیندلینگ مداخله می نماید، ممکن است مربوط به مسیر ورود کلسیم به سیتوپلاسم باشد

    اهداف

    پراکسیدازها در طیف وسیعی از فرآیند های زیست فناوری مورد استفاده قرار می گیرند که بیشتر آنها در دما و pH های بالا انجام می شوند. از آنجا که بیشتر پراکسیدازهای رایج در شرایط قلیایی و دمای بالا ناپایدار و غیرفعال بوده یافتن پراکسیدازهای ترموآلکالوفیلیک برای اهداف کاربردی امری ضروری به نظر می رسد.

    مواد و روش ها

    در این مطالعه تولید برون سلولی آنزیم پراکسیداز در سویه بومی Bacillus tequilensis مورد مطالعه قرار گرفت. برای این منظور، فعالیت آنزیم با استفاده از دو سوبسترای 2,4-DCP و پیروگالول در کشت مایع باکتری سنجش و  تاثیر مدت زمان کشت بر تولید آنزیم و همچنین تاثیر پارامترهای pH و دما بر میزان فعالیت آنزیم بررسی  شد. خالص سازی نسبی آنزیم با استفاده از روش کروماتوگرافی تعویض یونی با سفادکس DEAE A50 انجام و پارامترهای کینتیکی فعالیت آنزیم  بررسی شد. در این مطالعه پارامترهای کینتیکی از قبیل Km و Vmax محاسبه شدند.

    نتایج

    اندازه گیری فعالیت آنزیم در  زمان های مختلف کشت نشان داد که بیشترین میزان تولید آنزیم پراکسیداز 72 ساعت بعد از کشت باکتری بدست می آید.

    به منظور ارزیابی تنوع ژنتیکی، 32 خانواده ناتنی از گونه فسکیوی پابلند (Festuca arundinacea) در سال زراعی 94-1393 در شهرستان تبریز مورد بررسی قرار گرفت. براساس نتایج تجزیه واریانس تنوع معنی داری بین ژنوتیپ های مورد بررسی برای صفات ارتفاع بوته، قطر تاج پوش، روز تا خوشه دهی، روز تا گرده افشانی، قطر طوقه، عملکرد تر علوفه، عملکرد خشک علوفه، تعداد ساقه و عملکرد بذر در چین اول و در صفات قطر تاج پوش، قطر طوقه، عملکرد تر علوفه و عملکرد خشک علوفه در چین دوم و در سطح احتمال یک درصد مشاهده گردید. براساس نتایج به دست آمده از مقایسات میانگین، بیشترین عملکرد خشک علوفه در چین اول با 5/758 گرم در ژنوتیپ 32 به دست آمد. تجزیه به مولفه های اصلی با در نظر گرفتن مقادیر ویژه بزرگتر از یک موجب معرفی سه مولفه شد که 5/80 درصد از تغییرات بین نمونه ها را توجیه نمودند. در تجزیه خوشه ای بیشترین تمایز بین گروه ها با سه خوشه حاصل گردید و با برش دندروگرام ژنوتیپ ها در سه گروه قرار گرفتند. با توجه به نتایج، خوشه سوم از نظر اکثر صفات برتر از دو خوشه دیگر بود. ژنوتیپ های خوشه سوم با توجه به ارزش بیشتر این خوشه از نظر صفات مهم نظیر عملکرد علوفه و عملکرد بذر در برنامه های اصلاحی از اهمیت ویژه ای برخوردار خواهند بود. در به نژادی گیاهان علوفه ای، موفقیت در گزینش، به تنوع ایجاد شده از نوترکیبی ژنتیکی و هتروزیس بستگی دارد. با توجه به فاصله بین گروه های یک و سه، احتمالا بیشترین موفقیت در تلاقی بین ژنوتیپ های این دو گروه به دست خواهد آمد.

    اهداف

    از آنجا که یکی مشکلات مهم در ترمیم استخوان احتمال بروز عفونت های باکتریایی است، در سالهای اخیر استفاده از داربست های نانوکامپوزیتی حاوی آنتی بیوتیک در کاربردهای مهندسی بافت استخوان مورد توجه پژوهشگران زیادی واقع شده است.

    مواد و روش ها

    در این پژوهش داربست های کامپوزیتی پلی کاپرولاکتونی حاوی 10 درصد حجمی از نانوذرات دی اکسید تیتانیم  nm) 21~) و بیوگلاس µm) 6 (~ به صورت فاقد دارو و نیز حاوی غلظتهای 57/0 و 15/1 میلی گرم بر میلی لیتر داروی تتراسایکلین هیدروکلراید به روش ریخته گری محلول جهت کاربرد در مهندسی بافت استخوان ساخته شدند. مشخصه یابی ساختاری با مشاهدات میکروسکوپ الکترونی روبشی و نیز طیف سنجی تبدیل فوریه مادون قرمز به منظور تایید اتصال ذرات و دارو بر روی داربست انجام شد. همچنین بررسی سمیت سنجی سلولی با استفاده از آزمون MTT و مطالعه خواص آنتی باکتریال با استفاده از روش نفوذ در چاهک آگار انجام شد.

    یافته ها

    در این مطالعه داربست های پلیمری و کامپوزیتی حاوی ذرات داروی توزیع شده بر سطح تولید شده اند. همچنین مشاهده شد که داربست های کامپوزیتی بایوگلاس/ پلی کاپرولاکتون حاوی57/0 میلی گرم بر میلی لیتر تتراسایکلین علاوه بر خواص آنتی باکتریال مطلوب، میزان قابل قبولی از زنده ماندن سلولها را دارند.

    نتیجه گیری

    در نتیجه این داربست ها بالقوه می توانند به عنوان داربستهای مهندسی بافت با خاصیت آنتی باکتریال مورد توجه قرار گیرند.

    اهداف

    گرافن کوانتوم دات ها) (GQDبا داشتن ویژگی های منحصر به فرد مانند قابلیت حل شدن در آب، ویژگی فتولومینسانس، زی ست سازگاری خوب، ویژگی های فیزیکی، شیمیایی و الکترونیکی توجه زیادی را به خود جلب کرده ا ست. باتوجه به ویژگی های ذکر شده، GQDها از قابلیت استفاده در انواع کاربردهای زیستی، سنسور، فتوکاتالیست ها و جاذب برخوردار می باشند. هدف از انجام این پژوهش سنتز و انجام اصلاحات شیمیایی GQD به منظور افزایش ویژگی سطحی باشد.

    مواد و روش ها

    در این مطالعه از سیتریک اسید به عنوان پیش ماده کربن و همچنین اوره استفاده گردید و سیتریک اسید با روش هیدروترمال در دمای 160 °Cبه مدت 4ساعت طی فرآیند خودآرایی به ساختار گرافن تبدیل شد. سپس GQDسنتز شده کربونیزه شده و به صورت شیمیایی با روش KOHفعال سازی شد. ناحیه سطحی و ساختار حفره ها با ایزوترم های جذب/ واجذب موردبررسی قرار گرفت.

    یافته ها

    نتایج این تحقیق ن شان داد که ناحیه سطح ویژه گرافن کوانتوم دات کربونیزه و فعال شده) (CA-GQDاز 0/06 m2/gبه 12043 m2/gافزایش یافت و ساااختار حفره ها به طور قابل مظح ه ای تقویت شااد. الگوی XRDمربوط به GQDساااختار پایه مربوط به لایه گرافیت را تایید کرد. عکس های TEMمورفولوژی یکنواخت GQDرا نشاان داد و اندازه GQDکمتر از 5بدست آمد.

    نتیجه گیری

    روش به کار گرفته شااده در این تحقیق، روش موثری برای ایجاد GQDهای کارآمد با سااطح BETوساایو و توزیو باریکی از ساختار حفره -ها می باشد که می تواند برای مقاصد زیست پزشکی مورد استفاده قرار گیرد.

    جیوه از خطرناکترین آلاینده های زیست-محیطی است که در صورت ورود به بدن انسان متابولیزه نشده و با تشکیل کمپلکس های پایدار با مولکول های زیستی اختلالات عصبی، تنفسی و متابولیکی متعددی را در پی خواهد داشت بنابراین توسعه روش های دقیق، سریع و ارزان برای تشخیص جیوه درمحیط زیست اهمیت زیادی دارد. نقطه کوانتومی کربنی یک ماده فلورسنت جدید با خواص فیزیکی و شیمیایی منحصر به فردی است که برای کاربردهای تشخیصی به خصوص برای تشخیص فلزات سنگین در غلظت های کم مورد توجه قرار گرفته است. در این بررسی نقاط کوانتومی کربنی با نشر فلورسنت آبی با استفاده از روش پیرولیز سنتز و مشخصه یابی شد. نشر فلورسنت نقاط کوانتومی کربنی سنتز شده در میانکنش با جیوه به میزان قابل توجهی کاهش یافت. در ادامه برای سهولت استفاده از این ماده برای تشخیص جیوه در هیدروژل آلژینات سدیم انکپسوله شد. امکان خاموش سازی توسط جیوه در ساختار هیدروژل ارزیابی شد و کمترین غلظت قابل شناسایی جیوه در ساختار جدید به میزان  یک میکرومولار به دست آمد. استفاده از ساختار هیدروژلی علاوه بر اینکه سبب گسترش کاربردهای حسگر جیوه می شود، می توان از آن برای حذف جیوه از محیط نیز بهره برد.

    قارچ ها مهم ترین عامل فرسودگی زیستی در موزه ها، کتابخانه ها، آرشیو ها و مخازن اسناد مکتوب می باشند. هدف از مطالعه حاضر بررسی تنوع قارچی نسخه های خطی آسیب دیده در یکی از مخازن مرکز اسناد و کتابخانه آستان قدس رضوی و تعیین آلودگی هوای مخزن و ارتباط آن با آلودگی نسخ می باشد. نمونه گیری از نسخه های خطی با استفاده از سوآب و کاغذ نیتروسلولز استریل و هوای مخزن به روش پلیت باز، انجام شد. القاء فرسودگی اسپورهای قارچی روی کاغذ، در آزمایشگاه مدل سازی شد. چهارده جدایه قارچی از سه نمونه ی بستر مختلف خطی و شش جدایه قارچی از نمونه ی هوای مخزن به دست آمد. در بین جدایه های قارچی نسخ خطی Aspergillus sp. با بیشترین فراوانی (%36) و Penicillium sp. (5/21%) و Alternaria sp. (14%) به ترتیب در مقام دوم و سوم قرار داشتند. نمونه های قارچی P. chrysogenum، Cladosporium cladosporioides، Talaromyces diversus و Aspergillus sp. از نمونه کاغذهای پوستی جدا شده اند که می توانند در ارتباط با قارچ های با فعالیت آبی کم و نمک دوست باشند. بیشترین قارچ های جداشده مخزن از محیط MEA (53%)  به دست آمد. این قارچ ها شامل Purpureocillium lilacinum، Talaromyces diversus، Cladosporium sp. و Aspergillus sp. می باشند.تعداد کم قارچ های جدا شده از فضای مخزن، در مقایسه با اتاق بافری می تواند به عوامل محیطی مناسب کنترل شده درون مخزن نسبت داده شود. گونه های شناسایی شده مشترک بستر و هوا می تواند ناشی از انتقال و استقرار اسپورهای قارچی هوا به سطح نسخ باشد. حضور لکه های رنگی روی مدل های کاغذی آغشته به اسپورهای قارچی فرسایش قارچی را در بستر کاغذی اثبات می کند.

    آنژیوژنز[1] غیرطبیعی با بیماری های مختلف نظیر سرطان و متاستاز آن، رتینوپاتی و آرتریت روماتوئید در ارتباط است. VEGF-A[2] مهمترین واسطه ی آنژیوژنز در میان تمام فاکتورهای رشد است. فعالیت زیستی VEGF توسط دو گیرنده تیروزین کینازی VEGFR-1 و VEGFR-2 سلول های اندوتلیال وساطت می شود. سیگنالینگ VEGF از طریق VEGFR-2 مسیر اصلی آنژیوژنز است که منجر به تحریک رشد سریع سلول های اندوتلیال به داخل تومور می شود. این رسپتور از یک بخش خارج سلولی، یک بخش سیتوپلاسمی و یک دمین ترانس ممبران تشکیل شده است. بخش خارج سلولی متشکل از هفت دمین شبه ایمونوگلوبولین (D1-D7) است که دمین های اول تا سوم به عنوان جایگاه اتصال لیگاند عمل می کنند. در این مطالعه دمین های خارج سلولی 3-1 گیرنده (KDR1-3) بصورت نوترکیب در Pichia pastoris بیان شد. یک قطعه DNA 975 نوکلوتیدی حاوی دمین های 3-1 براساس توالی نوکلئوتیدی در GenBank و توالی پروتئین در Swiss-Prot طراحی شد. وکتور بیانی ترشحی نوترکیب (pPinkαHC/KDR1-3) ساخته شد و به روش الکتروپوریشن به داخل مخمر منتقل شد. سلول های نوترکیب با بیان بالا از طریق تکمیل اگزوتروفی آدنین شناسایی و کشت شدند. KDR1-3 تحت القا با متانول 1٪ بیان و با SDS-PAGE و تکنیک وسترن بلات تایید شد. پس از تخلیص با کروماتوگرافی تمایلی با رزین Ni-NTA، اتصال محصول بیان شده به hVEGF165 با الایزای مستقیم و الایزای مبتنی بر گیرنده اثبات شد. نتایج نشان داد که دمین خارج سلولی 3-1 گیرنده VEGFR-2 انسانی با ساختار پروتئین یوکاریوتی، که هیچ گزارشی در مورد آن وجود ندارد، با موفقیت بیان شد.

    برهمکنش میان نانوذراتی همچون نانولوله های کربنی با پروتئین ها مورد توجه زیادی قرار گرفته است.  کاربردهای بیومولکولی پیشرفته نانولوله های کربنی نیاز به درک متقابل برهمکنش آنها با مولکول های زیستی دارند. برهمکنش غیر کووالانسی پپتیدهای خونی مانند هپسیدین با نانولوله های کربن، اثرات  مهمی در طیف گسترده ای از کاربردهای بیولوژیکی دارد، که از طریق آنالیز پارامترهای ترمودینامیکی برهمکنش بین نانولوله های کربنی و پپتید ها شناسایی می شوند. علاوه بر این، اثرات پارامترهای مختلف در نحوه برخورد نانولوله ها با پپتید و تغییرات ساختاری و پایداری پپتید مورد بررسی قرار می گیرند. در این مطالعه که بر اساس شبیه سازی دینامیک مولکولی صورت گرفت، تغییرات ساختاری هپسیدین 20 در برهم کنش با نانولوله ی دوجداره کربوکسیله مورد بررسی قرار گرفت. نتایج حاصل از شبیه سازی نشان داد که نانولوله های کربنی موجب  بازشدن ساختار هپسیدین 20 و ایجاد تغییرات ساختاری در این پپتید میشوند. از طرفی باز شدن ساختار هپسیدین احتمالا  منجر به تغییر  میزان فعالیت آن می شود. نتایج حاکی از این بود که تغییرات قابل توجهی در ساختار هپسیدین 20 در حضور نانولوله ی کربنی ایجاد می گردد.که این اختلاف مقادیر به خاطر تحرکات و جابجایی های لوپ وN - ترمینال  پپتید هپسیدین 20 می باشد.

    coli Escherichia یک باکتری گرم منفی، دومین باکتری از نظر فراوانی در روده و شایع ترین عامل عفونت ادراری و از عوامل اصلی مسمومیت غذایی و اسهال است. همچنین به عنوان شاخص اصلی آلودگی آب شهری اهمیت ویژهای دارد. مقاومت دارویی این باکتری به آنتیبیوتیک ها به عنوان یک چالش جهانی مطرح میشود. به فرآیند انتقال ماده ژنتیکی در مسیری غیر از انتقال ژنها به صورت رابطه والد- فرزندی انتقال افقی ژن (HGT (گفته میشود. این فرآیند میتواند روی عملکرد ژنها از جمله ژنهای ایجادکننده مقاومت به آنتیبیوتیک ها تغییر ایجاد کند. ایجاد مقاومت در باکتری coli. E میتواند از طریق ژنهای متحرک به گونه های دیگر باکتریها هم انتقال پیدا کند. امروزه از ابزارهای بیوانفورماتیکی برای فهم و ردیابی ژنهای انتقال یافته با مکانیزم انتقال افقی استفاده میشود. در این مطالعه با استفاده از ابزارهای بیوانفورماتیکی مانند PredictBias، Alien‐ و PAI‐IDA ،Mobil Genetic Elements (MGEs) ،ACLAME Hunter به بررسی برخی ژنهای دخیل در مقاومت به آنتیبیوتیک در باکتری coli. E پرداخته شد. نتایج بررسیهای بیوانفورماتیکی و روش MIC انتقال افقی ژنهای مقاومت را نشان داد. 26 ژن از 32 ژن در کلیه نرمافزارها انتقال را نشان دادند. بیشتر ژنهای شناساییشده دارای درصد بازهای GC بالای 50 %بودند. ژنهایی مانند P put با 178 توالی، blaCMY با حدود62 توالی، BlaTEM با 43 توالی و 6‐aac با 66 توالی در سایتهای PredictBias و ACLAME به دست آمدند. mobC ،mobA و mobB و خانواده ژنی rep بیشترین احتمال انتقال افقی را بین سویه های بیماری زا نشان دادند. بیشترین مقاومت در تست های آزمایشگاهی را ژنهای blaSHV و blaCHV روی پلاسمید blaCMY نشان دادند.

    سرطان سینه یک عامل پرخطر برای سلامت زنان محسوب می شود که حدود 23 درصد میزان سرطان ها و دومین عامل مرگ در بین تمامی سرطان ها به شمار می رود. به خاطر پرهزینه و زمان بربودن روش های سنجش فعلی سرطان سینه ، نیاز به توسعه روش های حساس تر و اختصاصی ترمی باشد. بیوسنسورها به خاطر پاسخ سریع ناشی از محاسبه مستقیم در مایعات فیزیولوژیکی (بزاق، خون، سرم، شیر و ادرار) حائز اهمیت هستند. بیوسنسورهای برپایه آپتامر نسبت به روش های سنتی برای شناسایی سلول های سرطانی مزیت هایی از جمله سرعت، سادگی و ارزان بودن را دارا می باشند. در این مطالعه، با اتصال دزوکسی ریبوزیم و آپتامر یک روش رنگ سنجی برای شناسایی سلول های سرطان سینه ارائه شد. آپتامرهای MUC1 و PTK7 نیز بعنوان آپتامرهای اختصاصی برای اتصال به سلول های سرطانی سینه استفاده شدند. این روش ممکن است نیازی به تغییر DNA و استفاده از برچسب را نداشته باشد که می تواند تشخیص سلول های سرطانی را ساده تر و ارزان تر انجام دهد. نتایج به دست آمده خاطر نشان ساخت که این آپتامرها فعالیت قابل توجهی برای تشخیص سلول های سرطانی را نشان دادند درحالی که در نمونه های کنترل فعالیتی مشاهده نشد. علاوه براین، نتایج نشان دادند که یک رابطه خطی بین مقدار سلول های سرطانی و سیگنال رنگ سنجی وجود دارد. در نهایت، نتایج به دست آمده یک روش ارزان و سهل انجام را برای تشخیص سلول های سرطانی در آینده را خاطر نشان می سازد.

    The CD80 protein, a member of the super-family of immunoglobulin, is a transmembrane protein expressed on the surface of the antigen-presenting cells (APC). This protein has two receptors on the surface of T cells (CTLA-4 and CD28), due to the binding of this protein to these receptors, the inhibitory and stimulatory pathway in the T cells begin, respectively. Naturally, CD80 proteins tend to have more binding affinity to CTLA-4 than CD28, and this is a factor in the extinction of T cells in the immune system in order to prevent autoimmunity. The aim of the present study is to create a variant of the CD80 protein that has an increased binding affinity to CD28 to bind to this receptor more strongly and induce more simulate pathways than the wild type of this protein (primary CD80 protein) in T cells. To identify this variant, first, the ancestral sequence was mutated by R software at positions 31 and 92 with amino acids that play an important role in the formation of hydrogen bonds. The R software output sequences were modeled with the SWISS-MODEL server. Then, each output model was docked with the HADDOCK server, and finally, the electrostatic and van der Waals energies between the receptors and the ligands were calculated. Among all the built-in models, the mutated K31Y, R92F has the best electrostatic and van der Waals energies and has the ability to have a much better connection to its CD28 receptor compared to the ancestral type of CD80.

    Aims

    Breast cancer is the most common malignancy in women throughout the world. Among the various methods for preventing and treating cancer, plant natural compounds have more benefits than chemical drugs and have less side effects. Recently, many studies have been carried out on the antioxidant, anticancer, antiproliferation, and antiinflammatory properties of plant lignans, indicating the importance of these compounds in the prevention and treatment of various diseases. In the present study, the cytotoxic effects and inducing apoptosis of pinoresinol and lariciresinol lignans were investigated on SKBr3 breast cancer cell line.

    Materials & Methods

    SKBr3 cells were treated with different concentrations of both pinoresinol and lariciresinol, separately for 72 hours. Then, cell viability and cells morphological changes were determined using MTT assay and inverted light microscope, respectively. Also, apoptosis induction was analyzed by flow cytometry using an Annexin V-FITC apoptosis detection kit.

    Findings

    Both pinoresinol and lariciresinol treatments induced morphological changes, decreased cell growth, survival, proliferation and a significant increase in apoptosis induction in SKBr3 cell line compared to control cells in a concentration-dependent manner.

    Conclusion

    Inducing apoptosis and preventing the growth and proliferation of cancer cells are important mechanisms in the treatment of cancer. Pinoresinol and lariciresinol can be used to reduce cell proliferation and increase apoptosis induction in prevention and treatment of breast cancer.

    Biosurfactants are valuable microbial metabolites that have considerable applications in different industries. They offer so many advantages over their synthetic counterparts such as biodegradability, low toxicity, activity at extreme conditions, ability to be produced from renewable wastes and by-products. In the present study, biosurfactant production of Halomonas sp. MM93 in nutrient broth medium at 30°C after 72h was investigated using oil spreading and hemolysis tests. The emulsification capacity of the biosurfactant was also evaluated in a defined production medium during 96h. Effect of olive oil, n-Hexan, and kerosene as hydrophobic carbon sources to induction of biosurfactant production by the strain MM93 was also investigated. Due to the importance of stability in the case of industrial use, the effect of extreme temperature, pH and salinity on the stability of bacterial culture supernatant was evaluated. This strain created a clear zone of 2.5cm diameter in an oil-spreading test and its E24 index was 45%. Halomonas sp. MM93 could reduce the surface tension of the culture medium from 70 to 40 mN/m. Also, the produced biosurfactant showed remarkable stability at high temperature (100°C), extreme acidic and alkaline conditions (pH=2-12), and high salinity (20g/L). According to obtained data, native isolated moderately halophilic bacterium, Halomonas sp. MM93 could be considered as a potent strain in terms of producing stable biosurfactants for various industries especially the processes of increasing microbial recovery of oil that need Compounds with High surface activity and high stability.

    The waste produced by households, industry and factories to use as wash water and cooling systems and equipment, is caused environmental complications. Waste created a suitable environment for the growth of odor and pathogenic bacteria. This study aimed to isolate microorganisms in the wastewater and use them for the reduction of BOD (biochemical oxygen demand) and COD (chemical oxygen demand).The samples were collected aseptically from different parts of wastewater of herbal distillation industry. The bacterial strains were isolated from the samples in LB media. The isolates were distinguished based on the morphology and biochemical characteristics. Among the total 69 isolates, four isolates were selected to measure the ability to reduce BOD and COD and added to the waste water. Reduction of BOD was measured using BOD meter. Also, COD was determined by titration method. The isolates were identified by biochemical tests. The amount of BOD and COD reduction after adding selected strains to wastewater was 47.43 - 71.82% and 44.79 - 56.5% respectively. Also, the consortium of bacterial strains showed better ability to reduce BOD and COD (38.32-57.29%and 76.6-83.21% respectively). The results determined that wastewater contains bacterial strains which have shown significant reduction in BOD and COD and organic matter decomposition in wastewater and reuse it in agricultural and industrial sectors. Therefore, we can use these bacteria for wastewater treatment.

    Electrical Kindling is one of the most popular epileptic model techniques that cause seizures such as temporal lobe epilepsy. So far, various therapies have been used to treatment of epilepsy. Among these treatments, low-frequency stimulation (LFS) has been widely considered for improving effect on drug-resistant epilepsy, but its mechanism is not well understood. Since calcium entering to the cytoplasm and increasing its concentration is one of the reasons for seizure, metabotropic glutamate receptor (mGluR1), dopamine receptor (D1) and ADPR cyclase (CD38), which increased calcium in the cytoplasm from different pathways, were selected. With this aim that by examining the change in the expression of these genes, we help to clarify the LFS improvement effect.  In this study, the hippocampus of rats was used and the changes in genes expression were investigated using real-time PCR technique. The results showed that the expression of all selected genes  increased significantly after kindling and then after the LFS the expression of all was returned to sham value. Hence, one of the ways in which LFS interferes may be related to the pathway for calcium entering to the cytoplasm.

    Aims

    Peroxidases are used in a wide range of biotechnological processes, most of which are carried out at high temperatures and high pH levels. Since most of the commonly used peroxidases are unstable and inactive in alkaline conditions and high temprature, it is necessary to find thermoalkalophilic peroxidases for practical purposes.

    Materials and Methods

    In this study, extracellular production of peroxidase in the native strain Bacillus tequilensis was studied. for this purpose, Enzyme activity was evaluated using two substrates 2,4-DCP and pyrogallol in bacterial liquid culture and the effect of culture time on enzyme production, as well as the effect of parameters such as pH and temperature on enzyme activity investigated. The relative purification of the enzyme was performed using ion exchange chromatography with sephadex DEAE A50 and the kinetic parameters of enzyme activity were evaluated. In this study, kinetic parameters such as Km and Vmax were calculated.

    Results

    Measurement of enzyme activity at different times of culture indicated that the highest amount of peroxidase production was obtained 72 h after bacterial culture.

    This study was conducted to assess genetic diversity among the 32 tall fescue half-sib families using a randomized complete block design (RCBD) experiment in the four replicates. Based on analysis of variance, significant differences were observed among studied genotypes at the probability of 1% for plant height, canopy diameter, days to heading, days to pollination, crown diameter, fresh forage yield, dry forage yield, number of stem and seed yield in first harvest and in canopy diameter, crown diameter, fresh forage yield and dry forage yield in second harvest. Based on the results of mean comparisons, highest dry forage yield in the first harvest was obtained in genotype 32 by 758.5 grams. Principal component analysis by considering eigenvalues greater than one, caused to introduction of three components which determined 80.5% of the variation among the samples. In cluster analysis, the greatest of distinction between the groups was achieved with three clusters, and by cutting the dendrogram genotypes in three groups. According to the results, the third cluster was superior to other two clusters in terms of most traits. The genotypes of third cluster, according to the value of this cluster in terms of forage yield and seed yield will be of particular importance in breeding programs. In the breeding of cross-pollinated forage crops, success in selection depends on creating diversity by genetic recombination and achievement of heterosis. Due to the distance between groups 1 and 3, probably the most successful crosses will be achieved among genotypes of these two groups.

    Since one of the main problems in bone tissue repair is the bacterial infections, recently the development of drug-eluting nanocomposite scaffolds for bone regenerative medicine applications has attracted significant attention. In this study Polycaprolactone (PCL)-based composite scaffolds containing 10vol% of titanium dioxide nanoparticles (~21nm), and bioactive glass particles (~6µm), were prepared without drug and also loaded by Tetracycline hydrochloride (TCH) antibiotic (0.57, 1.15 mg/mL) through solvent casting method for bone tissue engineering applications. Structural characterizations based on Scanning Electron Microscopy (SEM), and FTIR analysis were utilized to study the chemical bonds of glass/ceramic particles, and antibiotic crystals on the surface. In addition, in vitro cytotoxicity, and antibacterial analysis were performed by MTT, and Agar well-diffusion assays, respectively. In this study polymeric and composite scaffolds were fabricated with TCH clusters decorated on the surface. It was shown that the bioactive glass/PCL scaffolds loaded by 0.57 mg/mL of TCH revealed significant antibacterial effect, despite the acceptable cell viability. These scaffolds seem to be of interest as a potential candidate in drug-eluting scaffolds for bone tissue engineering applications.

    Graphene quantum dots (GQDs) have attracted increasing attention due to their unique properties such as high water solubility, photoluminescence activity, good biocompatibility, physical, chemical and electrical properties which makes them appropriate candidates for use in a variety of bio-applications, sensors and photocatalysts. The objective of this study is synthesis of GQDs and improving their surface properties via chemical modification.Here, urea and citric acid as carbon precursor were used.  Citric acid was self-assembled into graphene framework via hydrothermal method at 160 °C for 4 h.  Then, the synthesized GQDs were carbonized and chemically activated by KOH treatment. The surface area and pore structures of GQDs were analyzed by nitrogen adsorption/desorption isotherms. The results showed that the specific surface area of carbonized-activated graphene quantum dots (CA-GQDs) have been increased from 0.06 to 1204.0 m2/g and pore structures have been enhanced significantly. The XRD pattern of GQDs confirmed the basic structure of graphite layer. The TEM images indicated the unique morphology of GQDs and the sizes of GQDs  were less than 5 nm. Thus, our applied method is an effective approach in the formation of GQDs with large BET surface area and narrow pore structures which reveals their potential applicability in biomedical field.

    Mercury is one of the most harmful pollutants in the environment, which in the event of ingestion into the human body, it is not metabolized and severe nervous, respiratory and metabolic disorders will occur due to the formation of stable complexes with biological molecules. Therefore, the development of precise, fast and inexpensive methods for mercury detection in the environment is of great importance. Carbon quantum dot is a new fluorescent substance with unique physical and chemical properties which is taken into consideration for diagnostic applications, especially for heavy metals detection at low concentrations. In this study, carbon dots with blue fluorescent emission were synthesized using the pyrolysis method and characterized. Fluorescent emission intensity of synthesized carbon dots was decreased significantly in interacting with mercury. Then for providing an easy-to-use mercury detection method, carbon dot was encapsulated in sodium alginate hydrogel. Quenching fluorescent intensity of encapsulated carbon dots in the hydrogel structure was evaluated in the presence of mercury and 1micromolar detection limit was obtained. Encapsulating carbon dots in hydrogel structure not only extend the use of carbon dot as a mercury sensor but also can be used as an adsorbent for mercury pollutant from the environment.

    Fungi are the most important agents of biodeterioration in museums, libraries, and repositories. The objectives of the paper were to evaluate the microbial diversity in biodeteriorated manuscripts located in a repository of the central library of Astan Quds Razavi (AQR) and to estimate the fungal occurrence of the repository air. The sterile cotton swabs and nitrocellulose membranes were used for sampling the manuscripts, while the sedimentation method was used for the microbial sampling of air. To evaluate the biodeteriorative impacts of fungi, fungal spore’s suspension inoculated on paper strips. Fourteen and six fungal isolates were collected from the three different deteriorated substrates and the repository air samples, respectively. Among the fungi isolates, Aspergillus sp. was isolated in high frequency (36%), followed by Penicillium sp. (21/5%) and Altelnania sp. (14%). Fungi species including P. chrysogenum, Cladosporium cladosporioides, Talaromyces diversus, and Aspergillus sp. were isolated from B1 sample as a parchment. The most fungal isolates (53%) in the air repository including Purpureocillium lilacinum, Talaromyces diversus, Cladosporium sp., and Aspergillus sp were achieved from MEA medium. The low number of isolated fungi from repository air can be attributed to the efficiently controlled environment factors of AQR repository. The combination of finding provides some support for the conceptual premise that it could be a direct relationship between the isolated microorganisms from air and those isolated from the manuscripts. The presence of color spots on paper strips can approve the biodeterioration of paper via fungal activities.

    Abnormal angiogenesis is associated with various diseases such as solid tumors and metastasis, retinopathies, and rheumatoid arthritis. VEGF-A is the most important mediator of angiogenesis among all growth factors. The bioactivity of VEGF is mediated by two tyrosine kinase receptors VEGFR-1 and VEGFR-2 present on endothelial cells. VEGF signaling through VEGFR-2 is the major angiogenic pathway that leads to stimulation of endothelial cell ingrowths into the tumor. It comprised of an extracellular portion, a cytoplasmic portion, and a short transmembrane domain. The extracellular portion consists of seven Ig-like domains (D1–D7), of which the 1st to 3rd domains function as ligand-binding sites. In the present work, a soluble recombinant extracellular domain 1-3 of VEGFR-2 was expressed in Pichia pastoris to inhibit the VEGF-induced angiogenesis. The 975 bp DNA fragment containing extracellular domain 1-3 kdr, was designed according to the nucleotide sequence at GenBank and protein sequence at Swiss-Prot. The recombinant secretory expression vector (pPinkαHC/KDR1-3) was constructed and transferred into yeast by electroporation. The high expression transformants were identified through complementation of adenine auxotrophy and cultured. KDR1-3 was expressed under the induction of %1 methanol and confirmed by using SDS-PAGE and western blot techniques. After being purified by affinity chromatography using Ni-NTA resins, binding of expressed product to hVEGF165 was proved by two direct ELISA and ELISA receptor binding assays. The data showed that human VEGFR-2 extracellular domain 1-3 with eukaryotic protein structure, that there is no reported paper about, was successfully expressed.

    The interactions between carbon nanotubes (CNTs) and proteins were considered much attention. Advanced CNT applied biomolecules require mutual understanding of their interactions with biological molecules. Enhanced biomedical applications of CNTs have necessitated the need for the understanding their interaction with biomolecules. Non-covalent interactions of blood peptides, such as hepcidin, with carbon nanotubes, have important effects in a wide range of biological applications that are detected by analyzing the thermodynamic parameters of the interaction between CNTs and peptides. In addition, the effects of different parameters in order to evaluate how the interaction of CNTs with peptide affects and structural changes and stability of peptides were studied. In this study, based on molecular dynamics (MD) simulation, the structural changes of hepcidin 20 in interaction with multi walled carbon nanotubes (MWCNTs-COOH ) were investigated. The simulation results revealed that carbon nanotubes cause to loose the hepcidin structure and make structural changes in this peptide. On the other hand, the loose of the hepcidin structure may lead to a change in its activity. The results indicated that significant changes were made in the structure of hepcidin 20 in the presence of carbon nanotubes. The difference of parameter amounts calculated in heptidine 20 is related to their N-terminal, and loop regions.

    Escherichia coli is a Gram-negative bacterium, the second most common bacteria in the intestine and the main indicator of urban water pollution, is the most common cause of urinary tract infection and also is one of the main factors in food poisoning and diarrhea. Drug resistance of this bacterium to antibiotics is a global challenge. Horizontal gene transfer (HGT) is the movement of genetic material between unicellular and other gene transfer pathways which is an important factor in the evolution of many organisms, antibiotic resistance in bacteria, gene function. Antibiotic resistance in E. coli can be transferred to another species of bacteria through HGT mechanisms. Today, Bioinformatics methods have been used to understand of gene transfer from HGT mechanism. In this study, we used bioinformatics tools such as PredictBias, ACLAME, Mobil Genetic Elements (MGEs) PAI-ID, and Alien_Hunter in order to genes analysis that related from antibiotic resistance in E. coli. Bioinformatics and MIC assays result show that from 26 to 30 genes have been identified in all safthwers. Most of genes that identified show over 50 percent of GC content. put P gene with 178, blaCMY with 62, BlaTEM with 43, and aac-6 with 66 homology in the PredictBias website identified. Also in the ACLAME website, mob (A-C) and rep (A-C) gene family are highest number of horizontal gene transfer from infection bacteria strain. Those cluster genes are the highest resistant of laboratory tests which carries resistant genes such as  blaSHV and blaCHV on the blaCMY plasmid.

    Breast cancer is a serious health concern for women. It contributes to about 23% of the cancer cases, and accounts for the second largest number of deaths among all cancers. Expensive and time consuming recognition methods currently available for recognition of breast cancer potentiates the need for improvement of novel, specific and ultrasensitive strategies. Biosensors are sensitive, specific and cost effective procedures. These also display the benefit of quick response due to direct calculation in physiological fluids (saliva, blood, serum, milk, urine etc.) in a non-invasive way. Aptamer-based biosensors for cancer cell recognition have shown advantages of rapidness, simplicity and cost-efficiency over traditional approaches. In this study, by linking DNAzyme and aptamer together, we established colorimetric biosensor for the detection of MCF7 breast cancer cells.  MUC1 and PTK7 aptamers used as specific aptamers to binding to the breast cancer cells. This manner may also evade the modification of DNA and the use of labels, which can intensely rise the cost-efficiency and simplicity of cancer cell discovery. Our results indicated that these aptamers showed good activity for breast cancer cells detection in which in the control cells no activity was observed. Results also indicated that there is a good linear relationship between the cancer cell values and colorimetric signal. Finally, the obtained results indicated a cost effective and conveniently operated approach for cancer diagnosis in future.
