    سابقه و هدف

    استافیلوکوکوس اوریوس از مهمترین عوامل مسمومیت غذایی در جهان است. هدف از این پژوهش ارزیابی فراوانی ژن های انتروتوکسینو شناسایی جدایه های دارای مقاومت چند دارویی در محصولات لبنی سنتی استان فارس می باشد. 

    مواد و روش ها:

     این پژوهش به صورت مقطعی_توصیفی بر روی 300 نمونه محصولات لبنی سنتی(پنیر، شیر، کشک، دوغ و سرشیر) انجام گرفت. شناسایی و تایید جدایه های استافیلوکوکوس اوریوس با استفاده از آزمون های استاندارد بیوشیمیایی و تکثیرژن هایnucو 23S rRNA با روش PCR انجام شد. سپس به منظور ارزیابی ژن های انتروتوکسین از روشPCRچندگانه استفاده شد. همچنین حساسیت جدایه ها برای 16 آنتی بیوتیک با روش دیسک دیفیوژن مطابق معیار CLSI ارزیابی گردید. 

    یافته ها:

     از مجموع نمونه های مورد بررسی، 120 جدایه (40%) استافیلوکوکوس اوریوس تشخیص داده شدند. بیشترین فراوان یآلودگی مربوط به پنیر 38 نمونه (66/31%) و کمترین آن مربوط به دوغ 10 نمونه (33/8%) بود. 91 نمونه (83/75%) از جدایه های آلوده حاوی ژن انتروتوکسین بودند. بیشترین فراوانی ژن های انتروتوکسین، مربوط به sea (17/54%) و به دنبال آن به ترتیب seb ، seg ،  see،  sedو sec  با فراوانی50%، 33/28%، 67/16%، 17/4% و 50/3% بود. همچنین از 64 سویه استافیلوکوکوس اوریوس (33/56%) دارای مقاومت چند دارویی، 54 جدایه (38/84%) واجد ژن های انتروتوکسین بودند. 

    نتیجه گیری: 

    نتایج پژوهش نشان دهنده فراوانی قابل توجه جدایه های استافیلوکوکوس اوریوس دارای مقاومت چند دارویی مولد انتروتوکسین در منطقه مورد بررسی بود. از این رو پایش مداوم  مولکولی جدایه های استافیلوکوکوس اوریوس و همچنین مقایسه پروفایل ژنتیکی جدایه ها با نمونه های کلینیکی پیشنهاد می گردد.

    سابقه و هدف

    شناسایی باکتری های عامل عفونت خونی و تعامل آن با سیستم ایمنی،اهمیت بسیار زیادی دارد.RAGE  نوعی گیرنده سلولی است که مولکول های با منشا درونی و خارجی را شناسایی می کند و CXC11  نیز درالقای پاسخ های ایمنی مناسب علیه میکروب ها شرکت می کند. هدف از این تحقیق،تعیین میزان سطح بیان این مولکول ها در بیماران با عفونت خونی و افراد سالم می باشد.  

    مواد و روش ها

    دراین مطالعه مورد-شاهدی، سطح بیان ژن RAGE و CXCL11 در 40 بیمار مبتلا به عفونت خونی و 40 فرد سالم به عنوان کنترل با استفاده از تکنیک  Real time-PCRمورد بررسی قرار گرفت. تشخیص عفونت خونی با انجام کشت خون و بکارگیری آزمون های بیوشیمیایی باکتریایی انجام شد.

    یافته ها

    سطح mRNA ژن RAGE در بیماران آلوده به سودوموناس آیروژینوسا در مقایسه با  اشریشیا کلی ، استافیلوکوکوس اوریوس و اسینتوباکتر بامانی، به طور قابل توجهی افزایش داشت.اما سطح بیان CXCL11 در بین بیماران و گروه کنترل سالم افزایش معنی داری نداشت. همچنین در مقایسه باکتری های عامل عفونت ارتباط معنی داری مشاهده نگردید.

    نتیجه گیری

    نتایج نشان داد که درعفونت خونی RAGE گیرنده مهمی در برابر سودوموناس آیروژینوسا می باشد. از این رو روش های کاهش میزان بیان مولکول هایRAGE ، می تواند نقش کاربردی در پاک سازی خون از این باکتری داشته باشد.

    سابقه و هدف

    سالمونلوز یکی از بیماری های عفونی و مشترک بین انسان و حیوانات است که به وسیله سویه های مختلف سالمونلا ایجاد می گردد. پیشرفت در روش های تشخیص مولکولی، شناسایی پاتوژن های غذایی را دقیق تر و راحت کرده است. هدف از این پژوهش استفاده از روش HRM به منظور شناسایی سریع و دقیق باکتری سالمونلا با استفاده از ژن inv A می باشد.  

    مواد و روش ها

    در این تحقیق شناسایی باکتری سالمونلا با واکنش زنجیره ای پلی مراز با روش منحنی ذوب با شفافیت بالا (PCR-HRM) به وسیله توالی ژن invA، به عنوان یک نشانگرانجام شد.در مجموع تعداد 9 سویه رفرانس سالمونلا با استفاده از یک جفت آغازگراختصاصی ژن invA برای تشخیص باکتری سالمونلا استفاده شدند.  

    یافته ها

    قطعات موردانتظار 284 جفت بازی پس از تکثیر ژن invA   توسط PCR به دست آمد. تمام نمونه های مورد آزمون قادر به تولید پیک منحنی ذوب اختصاصی سالمونلا با شفافیت بالا در بازه دمایی   87/8  تا 87/9 درجه سلسیوس بودند.

    نتیجه گیری

    نتایج نشان داد که روش شفافیت منحنی ذوب با استفاده از آغازگر های اختصاصی ژن invA می تواند به عنوان یک روش دقیق و مطمین به منظور تعیین جنس های سالمونلا مورد استفاده قرار گیرد.

    سابقه و هدف

    رنگ های طبیعی فیکواریترین سیانوباکتری ها، دارای ویژگی های درمانی بالقوه ای هستند. امروزه جایگزینی آنتی اکسیدان های مصنوعی با آنتی اکسیدان های طبیعی بی ضرر مورد توجه می باشد. هدف ازاین تحقیق، جداسازی و خالص سازی رنگدانه ارغوانی خوراکی فیکواریترین و ارزیابی فعالیت آنتی اکسیدانی و ضد میکروبی سیانوباکتری خاکزی  FA1Nostoc sp. می باشد.  

    مواد و روش ها

    استخراج و خالص سازی رنگدانه فیکواریترین در چهار مرحله آماده سازی عصاره خام، رسوب گیری با آمونیوم سولفات 65 درصد، دیالیز و کروماتوگرافی تبادل آنیونی انجام گردید. سپس فعالیت آنتی اکسیدانی با روش نیتریک اکساید و فعالیت ضد میکروبی با روش انتشار در دیسک مورد بررسی قرار گرفت.   

    یافته ها

    غلظت و خلوص رنگدانه فیکواریترین پس از هر مرحله خالص سازی تا 4 برابر (از 5/1 تا 22/6) افزایش یافت. بیشترین فعالیت ممانعت کنندگی رنگدانه فیکواریترین در مقابل باسیلوس سوبتلیس (5/10± 28/0) و کاندیدا آلبیکنس (98/10±006/0) یافت گردید. علاوه بر آن، نتایج حاصل از بررسی فعالیت آنتی اکسیدانی، افزایش معناداری را با افزایش میزان غلظت رنگدانه نشان داد.

    نتیجه گیری

    نتایج این تحقیق می تواند زمینه ساز معرفی رنگدانه های طبیعی خوراکی سیانوباکتری های خاکزی دارای ویژگی های آنتی اکسیدانی و آنتی میکروبی با قابلیت استفاده در صنایع غذایی و دارویی باشد.

    سابقه و هدف
    پسماند های جامد و نیمه جامد به عنوان لجن نفتی در مراحل مختلف پالایش نفت خام تولید می شود. تجمع پسماند نفتی در پالایشگاه منجر به کاهش کارایی پالایش نفت و  رهاسازی آن موجب آلودگی های زیست محیطی می شود. هدف از انجام این تحقیق روان سازی  نفت خام در خطوط لوله و بازیافت نفت با استفاده از بیوسورفاکتانت تولید شده توسط یک سویه ی بومی می باشد.
    مواد و روش ها
    جدایه های مولد بیوسورفاکتانت از مخازن نفت پالایشگاه اصفهان جداسازی شدند و غربال گری آن توسط آزمون گسترش نفت انجام شد. همچنین کشش سطحی با استفاده دستگاه تنسیومتر اندازه گیری شد. ساختار شیمیایی بیوسورفاکتانت با استفاده از آنالیزهای شیمیایی شناسایی شد. بازیافت نفت از لجن نفتی در شرایط ثابت اندازه گیری شد. تاثیرروان سازی  بیوسور فاکتانت بر روی نفت خام در خطوط لوله در شرایط آزمایشگاهی ارزیابی گردید. همچنین پایداری بیوسورفاکتانت در شرایط محیطی مختلف اندازه گیری شد.
    یافته ها
    بهترین باکتری تولید کننده ی بیوسورفاکتانت به عنوان اسینتوباکتر جانسونی ABR6شناسایی شد و توالی ژنومی 16S rDNA آن در بانک ژن با شماره ی دسترسی 100470  MK ثبت شد. آنالیز های شیمیایی  اثبات کرد که بیوسورفاکتانت تولید شده لیپوپپتیدی است. به وسیله ی بیوسورفاکتانت حاصل از این باکتری  50 درصد نفت خام از لجن نفتی بازیافت شد و همچنین سرعت انتقال نفت خام در خطوط لوله از64 ثانیه به 35 ثانیه کاهش یافت. همچنین این بیوسورفاکتانت پایداری بالایی درw/v 5% نمک ،  گسترهpH  8 تا 10 و دمای 60 درجه سلسیوس و شرایط اتوکلاو را داشت.
    نتیجه گیری
     نتایج این پژوهش نشان داد که بیوسورفاکتانت تولید شده توسط سویه ی بومیاسینتوباکتر جانسونی ABR6علاوه بر فعالیت در شرایط افراطی،  قابلیت بازیافت نفت و افزایش سرعت انتقال نفت خام در خطوط لوله را نیز دارد. بنابراین استفاده از بیوسورفاکتانت حاصل در صنایع نفت توصیه می شود.

    آب میوه ها به دلیل استفاده رایج و داشتن ترکیبات فعال زیستی بستر مناسبی برای تولید غذاهای فراسودمند سین بیوتیک می باشند. بنابراین امکان زنده مانی باکتری پروبیوتیک و تاثیر تکنولوژیک باکتری و اینولین بر کیفیت آب انار بررسی شد. اینولین در درصد های مختلف (5/1، 3 و 5) و باکتری لاکتوباسیلوس پلانتاروم معادل 5/0 مک فارلند به آب انار اضافه شدند و در مقایسه با نمونه شاهد، نمونه پروبیوتیک و نمونه پری بیوتیک حاوی 3 درصد اینولین، تعداد باکتری و توانایی اینولین و باکتری در جلوگیری از رشد قارچ ها و حفظ ویژگی های آنتی اکسیدانی آب انار با توجه به ممنوعیت استفاده از نگه دارنده در محصول فراسودمند بررسی شد. نتایج نشان داد که آب انار سین بیوتیک حاوی 3 درصد اینولین ضمن کسب بالاترین امتیاز حسی، توانست ماندگاری باکتری لاکتوباسیلوس را تا  cfu/ml107 ×5/1 در هفته چهارم حفظ نماید. کاهش ویژگی آنتی اکسیدانی و شمارش قارچ در هفته چهارم به ترتیب در نمونه شاهد 4/25 درصد و cfu/ml 150، در نمونه پری بیوتیک حاوی 3 درصد اینولین 38/0 درصد و cfu/ml 90 و در سین بیوتیک حاوی 3 درصد اینولین 26/0 درصد و cfu/ml 65 بود. از این رو می توان نتیجه گرفت که تولید آب انار سین بیوتیک با 3 درصد اینولین می تواند ضمن ایجاد محصولی فراسودمند با حفظ تعداد مناسب باکتری پروبیوتیک، موجب حفظ کیفیت آنتی اکسیدانی و افزایش ماندگاری محصول نیز به مدت 4 هفته گردد.

    سابقه و هدف

    با استفاده از اطلاعات به دست آمده از نوع و پراکندگی میکروارگانیسم ها در نمونه های خاک های سطحی می توان به میزان توزیع مخازن نفت و گاز پی برد. هدف از این پژوهش، جداسازی و شناسایی باکتری های اکسید کننده متان از خاک های آلوده به نفت در خوزستان و بهینه سازی دما و اسیدیته در حضور متان بود.

    مواد و روش ها

    باکتری ها در محیط نمک های معدنی و نیترات (NMS) در حضور 50 درصد هوا و 50 درصد متان جداسازی شدند. محیط های کشت به مدت 10 روز بر روی شیکر در دمای °C 30 در تاریکی گرمخانه گذاری شدند و هر 2 روز یک بار مخلوط گازها تجدیدکشت شد. جدایه ها با استفاده از ویژگی های بیوشیمیایی و مولکولی شناسایی شدند. همچنین بهینه سازی رشد باکتری ها در دما و اسیدیته های گوناگون در محیط کشت NMS در حضور متان به عنوان منبع کربن انجام شد.

    یافته ها

     در شرایط مورد بررسی، سه باسیل گرم منفی از نمونه های خاک جداسازی گردید. جدایه ها متعلق به گونه هایی از اکروموباکتر و اسفنگوموناس بودند. تمامی سویه ها قادر به رشد در محیطNMS  دارای غلظت بالای متانول (3%) بودند. بهترین شرایط pH و دمای بهینه رشد باکتری های جداسازی شده به ترتیب 4/7 و °C 30  به دست آمد.

    نتیجه گیری

    سویه هایمتانوتروف جداسازی شده در بررسی حاضر قادر به رشد و اکسیداسیون متان در محدوده وسیع اسیدیته و دما بودند و می توانند برای حذف ترکیبات تک کربنه مانند متان و به عنوان شناساگر بیولوژیک مخازن نفت و گاز پیشنهاد شوند.

    سابقه و هدف

    ویروس موزاییک خیار((CMV یک کوکوموویروس با انتشار جهانی و یکی از ویروس های مهم کدوییان و سایر محصولات است که سبب کاهش قابل توجه محصول می گردد. این پژوهش به منظور به دست آوردن اطلاعاتی در مورد انتشار، اهمیت و ویژگی مولکولی CMV در مزارع جالیزی منطقه جهرم انجام شد.  

    مواد و روش ها

    147 نمونه گیاهی مشکوک به علایم CMV  از برگ های بوته های ارقام مختلف خیار، کدو، هندوانه و خربزه از مزارع مختلف کدوییان و گلخانه های خیار شهرستان جهرم جمع آوری و با استفاده از آزمون سرولوژیکی الایزا(DAS-ELISA) و آزمون مولکولی RT-PCR مورد بررسی قرار گرفتند. محصولات PCR مربوط به دو جدایه از روستاهای قطب آباد و هورموج تعیین توالی  و با 18 توالی NCBI مقایسه شدند. سپس درخت فیلوژنتیکی با کمک نرم افزار های ClustalW و MEGA6 با روش  Neighbor-Joining   ترسیم گردید.  

    یافته ها

    با استفاده از آزمون الیزا 59 نمونه آلوده به CMV تشخیص داده شد. واکنش RT-PCR نیز آلودگی ها را تایید نمود. آنالیز توالی ها نشان داد که دو جدایه  CMV-JS1و  CMV-JS2شهرستان جهرم همراه با جدایه های M21464.1، AJ242585، AJ866272.1،L40953.1 به ترتیب از استرالیا، چین، هند و آفریقای جنوبی در یک زیر گروه قرار می گیرند.  

    نتیجه گیری

    نتایج نشان داد که آلودگی قابل توجهی به ویروس موزاییک خیار در مزارع جالیزی منطقه جهرم وجود دارد که می تواند کاهش محسوس محصول را به دنبال داشته باشد. به دلیل دامنه میزبانی وسیع CMV و امکان انتقال به وسیله شته ها، از این رو توصیه می گردد که علاوه بر کنترل علف های هرز در مزارع مبارزه با شته ها نیز انجام شود.

    Background & Objectives

    Staphylococcus aureus is one of the most importantfactors of food poisoning in the world. The objective of this research is to investigate the frequency amount of enterotoxin genes in traditional dairy products with multidrug resistance isolates in Fars province.  

    Materials and Methods

    This cross-sectional descriptive study was performed on 300 samples of traditional dairy products.  In order to identify Staphylococcus aureus isolates in addition to standard biochemical tests, according to PCR method, nuc and 23S rRNA genes proliferation were confirmed. Enterotoxin genes were identified by using of the multiple PCR method. The susceptibility of the isolates for 16 antibiotics was investigated by using of the disk diffusion method according to CLSI criteria.  

    Results

    120 isolates (40%) were identified as Staphylococcus aureus. The highest frequency of contamination is related to Cheese with 38 samples (31.66%) and its lowest relates to Doogh with 10 samples (8.33%). 91 samples (75.83%) of contaminated isolates were included of the enterotoxin gene, the highest frequency of enterotoxin gene was related to sea (54.17%) and its following, seb, seg, see, sed and sec respectively were included the frequency 50%, 33.33%. 28%, 16.67%, 4.17% and 3.50%. Also out of 64 strains of Staphylococcus aureus (56.33%) with multidrug resistance, 54 isolates (84.38%) had enterotoxin genes.  

    Conclusion

    The finding indicated that a significant frequency of Staphylococcus aureus isolates with enterotoxin producing multidrug resistance. Therefore, continuous molecular monitoring of Staphylococcus aureus isolates and also evaluation of the association of genetic profiles with clinical specimens is recommended.

    Background & Objectives

    Identifying the bacteria causing the infection and interacting with the immune system is too essential. RAGE is a receptor, which recognizes several endogenous and exogenous molecules and CXCL11 participates in the induction of appropriate immune responses against microbes. The aim of the study was expression of the molecules in the patients suffering from septicemia versus healthy controls.

    Materials and Methods

    In this case-control study, the expression levels of RAGE and CXCL11 in 40 patients with septicemia and 40 healthy subjects, were evaluated using Real time-PCR technique. Diagnosis of septicemia was performed by blood culture and bacterial biochemical tests.

    Results

    RAGE mRNA levels were significantly increased in patients infected with Pseudomonas aeruginosa compared with Escherichia coli, Staphylococcus aureus and Acinetobacter bamanii. However, no significant increase in CXCL11 expression level was observed in patients and healthy controls and also in comparison with bacteria causing infection.

    Conclusion

    Results showed that RAGE is a critical receptor against Pseudomonas aeruginosa during septicemia, Therefore, methods to reduce the expression of RAGE molecules can play a practical role in cleansing the blood from this bacterium.

    Background & Objectives

    Salmonellosis is an infectious and common disease between humans and animals that is caused by different strains of Salmonella. Progress in molecular diagnostic methods, has led to accurate and easy detection and characterization of food microbial agent. The purpose of this research was to use HRM technique to access more accurate and rapid diagnosis of Salmonella bacteria by means of invA gene. 

    Material & Methods:

    In this study, diagnosis of Salmonella was done by polymerase chain reaction and high resolution melting curve ( PCR-HRM) using the sequence on invasion A gene (invA) as a marker. In total 9 Salmonella reference strains using a specific primer pair of genes invA were used to detect Salmonella.  

    Results

    The expected size of PCR amplified fragments of invA gene was determined as 284bp. All tested strains were able to show a Salmonella specific melting curve with high resolution at thermal interval of 87.8-87.9°C.

    Conclusion

    The results showed that HRM using specific primers of invA gene can be used as an accurate and reliable technique for diagnosis Genus of Salmonella.

    Background & Objectives

    Phycoerythrin (PE( found in cyanobacteria are natural dyes which have shown potential therapeutic properties. Nowadays, substitution of artificial oxidants with harmless natural antioxidants is crucial. Hence, the present study was focused on purification process of PE from a soil strain of Nostoc sp.FA1 and evaluating its antioxidant and antimicrobial activity.    

    Materials and Methods

    Extraction and purification of PE was performed in four stages of preparation of crude extract, 65% ammonium sulfate precipitation, dialysis and anion exchange chromatography. Subsequently, antioxidant and antimicrobial activity was determined by nitric oxide scavenging and paper disk diffusion method respectively.    

    Results

    It was found that concentration and purity of PE were increased after each purification step by almost four times (from 1.5 to 6.22). PE had the strongest inhibitory activity against Bacillus subtilis(10.5 ± 0.28) and Candida albilancs (10.98 ± 0.006). In addition, the results of antioxidant surveys showed that PE is a potent free radical scavenger and this activity increased significantly with the increasing pigment concentration.  

    Conclusion

     The results of this study can be the basis for the introduction of natural edible pigments of soil cyanobacteria with antioxidant and antimicrobial properties that can be used in food and pharmaceutical industries.

    Background and Objectives
    Solid and semi-solid wastes are produced as oil sludge at different stages of crude oil refining. Accumulation of oil waste in the refinery reduces the efficiency of oil refining and its release causes environmental pollution.  The purpose of this study is to lubricate crude oil in pipelines and recycle oil using a biosurfactant produced by an indigenous strain.  
    Materials and Methods
    The biosurfactant-producing isolates were obtained from petroleum reservoir in Isfahan Oil Refinery, Isfahan, Iran. Screening was performed by oil displacement method. Also the surface tension was measured by tensiometer. Biosurfactant chemical structure was identified by using chemical analysis. Oil recovery from the sludge was measured under controlled condition. The effect of biosurfactant lubrication was investigated on crude oil in the pipelines in vitro. The stability of biosurfactant in different environmental conditions was also determined.
    Results
    The best biosurfactant-producing bacterium was identified as Acinetobacter johnsonii ABR6, and its 16S-rDNA genomic sequence was deposited in GenBank under the accession number of MK100470. Chemical analysis of TLC and FTIR confirmed that the produced biosurfactant was lipopeptide. The biosurfactant obtained from this bacterium recovered 50% of crude oil from petroleum sludge and also reduced transportation speed from 64 to 35 seconds. This biosurfactant had high stability in 5% w/v NaCl, pH range of 8 to 10, temperature of 60°C and autoclave conditions.  
    Conclusion
    The results of this study showed that the biosurfactant produced by the native strain of Acinetobacter Johnson ABR6, in addition to operating in extreme conditions, also has the ability to recover oil and increase the transfer rate of crude oil in pipelines. Therefore, the use of this biosurfactant can be considered as an asset in the petroleum industry

    Natural juices are an excellent substrate for the production of synbiotic functional food for the high nutritional value and common use. Therefore, the survival of the probiotic bacteria and the technological effects of bacteria and inulin were studied on the pomegranate juice quality. Inulin in different percentages (1.5, 3, 5) and Lactobacillus plantarum bacteria equivalent to 0.5 McFarland were added to pomegranate juice. The number of bacteria and the ability of inulin and bacteria in inhibition of fungi and preservation of antioxidant quality of pomegranate juice was considered in compare to the control sample, probiotic sample and prebiotic sample with 3% inulin due to the prohibition of preserver usage in functional food. Based on the results, the pomegranate juice sample containing 3% inulin was able to protect the number of lactobacilli bacteria to 1.5 × 107 cfu/ml in the 4th week in addition to the highest sensory score. Decrease of the antioxidant activity and counting of the fungi were 25.4%, 150 cfu/ml in the control sample, 0.38%, 90 cfu/ml in the prebiotic sample with 3% inulin and 0.26%, 65 cfu/ml in the symbiotic sample with 3% inulin, respectively, in the fourth week. Finally, production of the pomegranate juice with 3% inulin can effect on the maintenance of the antioxidant quality and increase of the product shelf life up to 4 weeks meanwhile making a functional product with preserving of the suitable number of probiotic bacteria.

    Background & objectives

    Based on information about the types and numbers of microorganisms in the surface soil samples, the distribution range of underlying oil and gas reservoirs can be judged. In the present study, methane oxidizing bacteria were isolated and characterized from oil contaminated soils in Khuzestan, Iran and their growth was optimized in the presence of methane as sole carbon and energy sources. 

    Materials & Methods:

    Bacteria were isolated in nitrate mineral salt (NMS) medium in the presence of 50% air and 50% methane. The cultures were incubated on a shaker for 10 days at 30°C in dark place, and every two days, the gas mixture was replaced. The isolated bacteria were characterized based on biochemical and molecular identification tests. Then the optimum growth conditions was detected in different pH values and incubation temperatures in NMS medium containing methane.

    Results

    Three Gram-negative rods were isolated from soil samples that were able to grow in isolation condition. The isolates were characterized as Achromobacter and Sphingomonas spp. The strains could also grow in the NMS medium with a high methanol concentration (3%). The optimal pH and temperature for the isolates were 7.4 and 30°C respectively.

    Conclusion

    Methanotrophic strains that were isolated in the present study were able to grow and oxidize methane in high ranges of temperature and pH and can be proposed for the removal of mono-carbon compounds such as methane and as biological detectors for prospecting for oil and gas reservoirs

    Background and Objectives

    The Cucumber Mosaic Virus (CMV) is a worldwide distributed Cucumovirus, and one of the important viruses of cucurbits and other agricultural crops, which causes a significant reduction of yield. This study was done to determine some information about the distribution, importance, and molecular characteristics of CMV in cucurbitaceous fields of the Jahrom area.

    Material and Methods

    147 plant samples suspected of CMV symptoms were collected from leaves of different varieties of cucumber, pumpkin, watermelon, and melons from various farms of cucurbits and cucumber greenhouses of the Jahrom area and were investigated using DAS-ELISA and RT-PCR methods. The PCR products of two-isolates were sequenced from the Ghotbabad and Hormoj villages. These isolates sequences were compared with 19 selected sequences of NCBI. Using Clustal W and Mega6 software, the phylogenetic tree was drawn via the Neighbor-Joining method.

    Results

    59 samples were detected as CMV infected by DAS-ELISA test. The RT-PCR reaction confirmed the infection. Sequences analysis showed that the two isolates of CMV-JS1 and CMV-JS2 from the Jahrom area with the isolates of M21464.1, AJ242585, AJ866272.1, and L40953.1, respectively from Australia, China, India, and South Africa are located in an individual subgroup.

    Conclusion

    The results of this study showed that there where a significant infection in cucurbit fields of the Jahrom area and may cause significant yield losses. Since the CMV has a wide host range and is transmitted by aphids, hence it is recommended to control weeds as well as vector aphids.
