    ابداع دستگاه های میکروفلوییدیکی (ریزتراشه سیالی) منجر به تحول شگرفی در حوزه مهندسی، پزشکی و زیست پزشکی شده است. دستگاه های میکروفلوییدیکی شرایط کشت سلول را در ابعاد واقعی بدن فراهم می کنند. در مطالعه حاضر، ریزتراشه سیالی با قابلیت زنده نگه داشتن سلول در شرایط جریان دینامیک ساخته شد. این ریزتراشه از یک میکروکانال اصلی تشکیل شده است که سلول ها در کف آن کشت داده می شوند. با عبور محیط کشت از روی سلول مقادیر مختلف تنش برشی به سلول ها اعمال می شود. نتایج شبیه سازی میدان جریان نشان می دهد که در محدوده دبی 1 تا 100میکرولیتر بر دقیقه توزیع تنش برشی در ریزتراشه، یکنواخت است. در این محدوده، تنش برشی از 0/005434 تا 0/5432دین بر سانتی مترمربع تغییر می کند که در محدوده مجاز برای سلول ها قرار دارد. مقادیر بزرگ تر تنش برشی مانند دبی 1000میکرولیتر بر دقیقه منجر به پارگی جداره سلول و نهایتا متلاشی شدن آن می شود. نتایج آزمایشات تایید می کند که روند رشد و تکثیر سلول ها به ازای مقادیر مختلف فاکتور رشد به عنوان یک فاکتور شیمیایی متفاوت است. سلول ها با فاکتور رشد 15% در روز پنجم کشت، کف میکروکانال را پر کردند، در حالی که در نمونه بدون فاکتور رشد، میکروکانال در روز هفتم پر شد. نتایج به دست آمده نشان می دهد که این ریزتراشه قابلیت رشد و نگهداری سلول ها به مدت بیش از یک هفته را دارد و با تنظیم دبی سیال می توان مقادیر مختلف تنش برشی را به سلول ها اعمال کرد. بنابراین این ریزتراشه قابلیت اجرای آزمایشات مختلف سلولی و بررسی اثر تنش برشی بر واکنش آنها را دارد.

    اهداف

    واکسن رایج هپاتیت ب بر پایه ادجوانت آلوم است که به دلیل نقص در تحریک ایمنی سلولی کارآمد، نیازمند بهینه سازی است. آلفا- توکوفرول از اجزای اصلی ویتامین E است که به عنوان یک تعدیل کننده پاسخ ایمنی توانایی تنظیم پاسخ های سیستم ایمنی در جهت کاهش پاسخ های التهابی را دارد. از این رو در مطالعه حاضر به بررسی اثر آلفا- توکوفرول بر بهبود پاسخ های ایمنی واکسن هپاتیت ب پرداخته شد.

    مواد و روش ها:

     واکسن تجاری هپاتیت ب با دوزهای 1، 5 و 10میکروگرم از آلفا- توکوفرول فرموله شد. واکسن در سه نوبت به فاصله دو هفته به موش های BALB/C تزریق شد. 10 روز پس از آخرین تزریق خونگیری از گروه های موشی انجام شد. میزان سایتوکاین های IFN-γ، TNF-α، IL-4، IL-2 و همچنین توتال IgG و ایزوتایپ های IgG1 و IgG2a به روش الایزا مورد بررسی قرار گرفت.

    یافته ها:

     واکسن فرموله شده در دوز 10میلی گرم از آلفا- توکوفرول توانایی بیشتری در افزایش معنی دار سایتوکاین های IFN-γ، TNF-α، IL-2 و در دوز پایین منجر به تقویت ایمنی هومورال شد.

    نتیجه گیری:

     پاسخ های ایمنی ایجادشده توسط واکسن به دوز آلفا- توکوفرول وابسته هستند، به طوری که در دوز 10میلی گرم آلفا- توکوفرول افزایش پروفایل سایتوکاینی Th1 و در دوز یک میلی گرم منجر به افزایش تیتر آنتی بادی می شود.

    اهداف

    سالمونلوز یکی از بیماری های عفونی مشترک بین انسان و دام است که از مصرف گوشت طیور و فرآورده های آن ایجاد می شود. ژن invA نقش بسیار مهمی در بیماری زایی سالمونلا اینفتیس دارد. موجودات دریایی از جمله خیار دریایی به دلیل داشتن متابولیت های ثانویه موثر دارای ترکیباتی با خواص آنتی اکسیدانی، ضدسرطانی، ضدباکتری و ضدالتهاب شناسایی و مطالعه شده اند. هدف مطالعه حاضر، بررسی اثر عصاره خیار دریایی بر باکتری سالمونلا اینفنتیس و بیان ژن حدت invA در این جدایه ها است.

    مواد و روش ها:

     نمونه برداری از گوشت طیور انجام شد و سویه های سالمونلا اینفنتیس واجد ژن invA جداسازی شدند. عصاره هگزانی از بافت روده خیار دریایی استخراج و اثر آن بر سالمونلا اینفنتیس در چندین غلظت با روش MIC بررسی شد. سپس تغییرات بیان ژن invA در این سویه ها با تکنیک Real-time PCR مورد بررسی قرار گرفت.

    یافته ها:

     از بین 450 نمونه، 12 ایزوله باکتری سالمونلا اینفنتیس جداسازی شد. با تکنیک PCR هر 12 ایزوله مورد نظر واجد ژن حدت invA بودند. میزان MIC در این ایزوله ها 256میکروگرم بر میلی لیتر به دست آمد. میزان تغییر بیان ژن invA 1/21 بود.

    نتیجه گیری:

     عصاره هگزانی استخراج شده از روده خیار دریایی با کاهش بیان ژن حدت invA، خاصیت آنتی ویرولانسی بر روی باکتری سالمونلا اینفنتیس دارد و می تواند به عنوان مکمل درمانی علیه این باکتری مورد استفاده قرار گیرد.

    اهداف

    کف پای صاف یکی از عارضه های شایع در افراد است. بررسی نیروهای عکس العمل زمین در فعالیت های مختلف از اهمیت کلینیکی برخوردار است. بنابراین هدف مطالعه حاضر بررسی مولفه های نیروهای عکس العمل زمین هنگام دویدن در افرادی با عارضه کف پای صاف در مقایسه با افراد سالم است.

    مواد و روش ها: 

    12 مرد سالم و 12 مرد مبتلا به کف پای صاف در این مطالعه شرکت کردند. با استفاده از دو صفحه نیرو (1000هرتز)، مولفه های نیروهای عکس العمل زمین در فاز استقرار دویدن مورد ارزیابی قرار گرفتند. برای بررسی تفاوت های بین گروهی داده ها از آزمون تی مستقل با سطح معنی داری 0/05p≤ استفاده شد.

    یافته ها: 

    نتایج نشان داد بین گروه ها در مولفه های Fx1 (0/001 =p)، Fx3 (0/03 =p)، Fy1 (0/001 =p)، Fz1 (0/03 =p) و Fz3 (0/01 =p) اختلاف معنی داری وجود دارد و اختلاف میانگین ها حاکی از کمتربودن این مولفه ها در گروه کف پای صاف نسبت به گروه سالم بوده است.

    نتیجه گیری:

     کاهش برخی از مولفه های نیروی عکس العمل سطح در افراد کف پای صاف ممکن است متاثر از دامنه حرکتی و گشتاور عضلات ضدجاذبه متفاوت و یا اتخاذ استراتژی خشکی زانو در این افراد باشد.

    اهداف

    سلول های زنده دارای بار الکتریکی هستند که به واسطه حضور یون ها و رادیکال های آزاد ایجاد می شوند. میدان های مغناطیسی با برهمکنش با یون ها و به ویژه مواد فرومگنتیک نظیر آهن بر سلول های زنده تاثیر می گذارند. ویژگی مشترک حدود 20 بیماری مختلف، تجمع پروتئین در قالب رسوباتی با ساختار آمیلویید است. در مطالعه حاضر تاثیر میدان مغناطیسی ایستا (SMF) بر میزان تشکیل و سمیت ساختارهای آمیلوییدی مورد بررسی قرار گرفت.

    مواد و روش ‏ها: 

    سلول های CHO در مواجهه با میدانی با قدرت 6میلی تسلا در سه روز متوالی قرار گرفتند و تاثیر SMF در تشکیل ساختارهای آمیلوییدی در پروتئین های ذاتی این سلول ها نسبت به نمونه کنترل به کمک آزمون اتصال تیوفلاوین T (ThT) مورد بررسی قرار گرفت. میزان تشکیل ساختارهای آمیلوییدی در سلول های CHO بیان کننده ProIAPP انسانی نیز در مواجهه با میدان به کمک مشاهده تجمعات پروتئینی حاصل از proIAPP متصل به پروتئین GFP مورد بررسی قرار گرفت. تاثیر SMF بر سمیت حاصل از الیگومرهای لیزوزیم بر سلول های CHO و Hela نیز نسبت به نمونه کنترل بررسی شد.

    یافته ها:

     قرارگرفتن سلول های CHO در معرض میدان مغناطیسی تاثیر معنی داری بر تشکیل ساختارهای آمیلوییدی در پروتئین های ذاتی سلول های CHO و میزان این ساختارها در سلول های CHO بیان کننده پروتئین proIAPP ندارد، اما می تواند سمیت الیگومرهای لیزوزیم را بر روی سلول های CHO و Hela افزایش دهد.

    نتیجه‏ گیری: 

    میدان مغناطیسی با قدرت 6میلی تسلا بر تشکیل ساختارهای آمیلوییدی تاثیر معنی داری ندارد، اما بر سمیت این ساختارها می افزاید.

    پروتئین های فیوژن ویروسی به دلیل نواحی حفاظت شده شامل پپتیدهای فیوژنی برای فعالیت ورود به واسطه ادغام غشای ویروس های مختلف و غشای سلول میزبان ضروری هستند. پدیده فیوژن در سطح سلول میزبان یا در ترکیبات سیتوزولی و در اجزای سیتوپلاسمی اتفاق می افتد. پروتئین های فیوژن ویروسی بر حسب تغییراتی که طی فیوژن متحمل می شوند در سه دسته قرار می گیرند. دسته اول شامل ویروس هایی مانند اورتومیکسو، پارامیکسو، فیلو، کرونا و رترو است. در این دسته از ویروس ها پروتئین های فیوژن پس از برش و حذف بعضی نواحی به شکل بالغ و عملکردی خود می رسند. پروتئین های فیوژن در ویروس هایی مانند آلفا و فلیوی، بدون ایجاد برش به شکل عملکردی خود تبدیل می شوند. دسته سوم شامل ویروس هایی مانند وزیکولو استوماتیت، هرپس سیمپلکس و باکولوویروس ها، دارای ویژگی های مشترکی با دسته های اول و دوم هستند.بررسی تغییرات پروتئین های فیوژن در سطوح قبل و بعد از فیوژن، شرح فیوژن پروتئین ها در ویروس هایی مانند آنفولانزا، فیلو و ریو به عنوان پروتوتایپ ویروس های دارای این پروتئین و کاربردهای درمانی پروتئین های فیوژن و مکانیزم اثر داروهایی مانند لاکتوفرین و اینفیوورتاید در جلوگیری از وقوع این پدیده- مسایل بسیار مهم در همانندسازی و ورود ویروس را رقم می زند که بررسی آن بسیار مهم است.

    استیوآرتریت یا آرتروز شایع ترین بیماری مفصلی است که کیفیت زندگی مبتلایان را به شکل قابل توجهی تحت تاثیر قرار می دهد. از آنجا که غضروف بافتی بدون رگ و اعصاب است، درمان آن با مشکل روبه رو است. روش های رایج درمان از قبیل استفاده از داروهای ضدالتهابی غیراستروییدی و جراحی تنها موجب کنترل بیماری می شوند و با گذشت زمان مفصل کارآیی خود را مجددا از دست می دهد. روش های نوین مانند سلول درمانی و مهندسی بافت همگام با استفاده از زیست مواد مختلف در تلاش برای یافتن راه حلی هستند که بافت تخریب شده غضروف ترمیم شود. رویکرد دیگر، استفاده از RNAهای مداخله گر است که با هدف قراردادن توالی های تخریب گر، عوامل موثر در تخریب بافت غضروف را سرکوب می کند و در پی آن، بافت آسیب دیده به مرور زمان تعادل بین فرآیندهای آنابولیک و کاتابولیک را دوباره به دست می آورد. شناسایی ژن های موثر در تخریب ماتریس غضروف و سرکوب آنها نتایج امیدبخشی را ارایه کرده است. در مطالعه حاضر، بعد از معرفی بافت غضروف، فرآیند تخریب غضروف و ایجاد استیوآرتریت، به مرور پژوهش هایی پرداخته شده است که تاثیر RNAهای مداخله گر بر ترمیم بافت غضروف آسیب دیده را بررسی کرده اند.

    The invention of microfluidic devices has led to a dramatic change in engineering, medicine, and biomedicine. Microfluidic devices provide the conditions for cell culture in real body dimensions. In the present study, a microfluidic chip was fabricated that is capable of keeping cells alive under dynamic flow conditions. This microchip consists of a microchannel in which cells are cultured. Different amounts of shear stress are exerted to the cells by passing culture media. The results of the flow field simulation show that in the flow rate of 1 to 100 microliters per minute, the shear stress distribution is uniform. In this range of flow rate, shear stress varies from 0.005434 to 0.5432dyn/cm2, which is within the allowable shear stress for cells. Large shear stresses, such as a flow rate of 1000 microliters per minute, cause the cell wall to rupture, and eventually disintegration. The experimental results confirm that the growth and proliferation of cells vary for different amounts of growth factor as a chemical factor. The cells filled the microchannel for a growth factor of 15% on the fifth day of culture, while without growth factor, the microchannel was filled on the seventh day. The results indicate that this microchip can maintain cells alive for more than a week. Also, by adjusting the flow, different amounts of shear stress can be applied to the cells. Therefore, this microchip can perform various cellular tests to investigate the effect of shear stress on the cells.

    Aims

    The common hepatitis B vaccination was designed based on alum adjuvant. This adjuvant is failed to stimulate cellular immunity so that, optimization is necessary for better stimulation of cellular immune responses. α--tocopherol is a key component of vitamin E that is considered as immunomodulatory agent to regulate immunity in order to decrease inflammation immune responses. Therefore, in the present study, the effect of α-tocopherol on the improvement of immune responses in hepatitis B vaccination was evaluated.

    Materials & Methods

    commercial hepatitis B vaccine was formulated with α-tocopherol at the doses of 1, 5, and 10mg. The vaccine was injected into BALB/C mice three times two weeks apart. Ten days after the last injection, blood-drawing was performed from mice groups. The levels of IFN-γ, TNF-α, IL-4, and IL-2 cytokines and IgG total antibody, as well as IgG1 and IgG2a antibody isotypes, were measured by ELISA method.

    Findings

    Formulated vaccine at the dose of 10mg of α-tocopherol is more potent in the significant increase of IFN-γ, TNF-α, and IL-2 cytokines and low dose result in improvement of humoral immune response.

    Conclusion

    Immune response of vaccine formulated with α-tocopherol was dose-dependent and at the dose of 10mg demonstrated a significant increase of Th1 response and at the dose of 1mg result in increase of antibody responses.

    Introduction

    The invA gene plays a critical role in the pathogenicity of Salmonella infantis. Marine organisms, including sea cucumbers due to their effective secondary metabolites, have been identified and studied with compounds with antioxidant, anti-cancer, antibacterial and anti-inflammatory properties. The aim of this study is to determine effect of sea cucumber extraction (Holothuria leucospilota)  on S. infantis invA gene expression .

    Material & Methods

    Poultry meat was sampled. The S. infantis strains containing invA gene were isolated. The Hexane extract was extracted from sea cucumber colon tissue. Its effects on S. infantis and its effect on gene expression were investigated by MIC and Real-time PCR, respectively.

    Results

    Morphological and biochemical characteristics of these bacteria were confirmed. From 450 samples, 12 S.infantis isolates were isolated. The PCR technique was used to identify the invA encoding gene. All 12 isolates have invA virulence genes. MIC was determined 256 µg/ml. The effect of sea cucumber extract on invA gene expression in S.infantis was evaluated, and the rate of change for the invA gene is estimated -1.21.

    Discussion & Conclusion

    According to our results, hexane extract extracted from the sea cucumber (H. leucospilota) caused reduction ofinvA gene expression in salmonella infantis. So, it can be used as a therapeutic supplement against S. infantis

    Aims

    Flatfoot is one of the common complications in individuals. The study of ground reaction forces (GRF) in different activities has clinical importance. Therefore, the aim of the present study was to analyze the GRF during running in individuals with flatfoot compared with healthy subjects.

    Materials & Methods

    12 healthy men and 12 male patients with flatfoot participated in this study. By two force plates (1000Hz), GFR components during the stance phase of running were measured. Independent t-test was used to investigate differences between groups. The significance level was p≤0.05.

    Findings

    The results showed that there is a significant difference between the two groups in ground reaction force of Fx1 (p= 0.001), Fx3 (p= 0.03), Fy1 (p= 0.001), Fz1 (p= 0.03), Fz3 (p= 0.01), and the mean difference between these components was lower in the group of flatfeet compared to the healthy group.

    Conclusion

    Reducing some of the surface reaction force components in flatfoot individuals may be affected by the range of motion and torque of different anti-gravity muscles or adopting a strategy of stiffening the knee in these individuals.

    Aims

    Living cells have an electrical charge created by the presence of ions and free radicals. Magnetic fields interact with ions, especially ferromagnetic materials such as iron that affect living cells. A common feature of about 20 different diseases is the aggregation of proteins in the form amyloid structure. In the current study, the effect of static magnetic field (SMF) on the formation and the toxicity of amyloid structures was investigated.

    Materials & Methods

    CHO cells were exposed to 6mT SMF in three consecutive days, and the effect of SMF on the formation of amyloid structures in the intrinsic proteins of these cells related to the control was investigated using thioflavin T (ThT) binding assay. The formation of amyloid structures in CHO cells expressing human ProIAPP cells was analyzed by observation of proIAPP protein aggregates linked to GFP protein. The effect of SMF on the toxicity of lysozyme oligomers on CHO and Hela cells was also compared with the controls.

    Findings

    Exposure of CHO cells to magnetic fields does not have a significant effect on the formation of amyloid structures in the intrinsic proteins of CHO cells and the amount of these structures in CHO cells expressing proIAPP protein but can increase the toxicity of lysozyme oligomers on CHO and Hela cells.

    Conclusion

    The magnetic field does not have a significant effect on the formation of amyloid structures in the 6mT SMF strength, but it adds to the toxicity of these structures.

    Viral fusion protein through protected areas called fusion peptide are essential for entry of the virus into the host cell by membrane integration. Fusion phenomenon occurs at the host cell surface or in the cytoplasmic compounds and in cytoplasmic components. The fusion proteins are divided into three categories according to variations that are likely during the fusion. In the first category, which includes viruses such as orthomixo, paramyxo, filo, corona, and retroviruses, definite domains of fusion proteins are cleaved and removed and they become mature and functional. The second group of fusion proteins is alpha and flaviviruses become their functional form without cleavage. The third category also includes viruses such as vesicular stomatitis, herpes simplex, and baculovirus, they have common features of the first and second categories.The changes in fusion protein in the levels before and after fusion, description of fusion proteins in viruses such as influenza, filo, and reoviruses as a prototype of fusion protein viruses and their therapeutic applications of fusion protein as potential drugs such as Lactoferrin and Enfuvertide in preventing the occurrence of fusion phenomenon is an important issue for consideration about multiplication and virus entry.

    Osteoarthritis is the most common articular disease that has significantly affected the patients’ quality of life. As cartilage doesn’t have any blood vessels and neurons, its treatment is a difficult task to do. Traditional therapeutic approaches, including the use of non-steroidal anti-inflammatory drugs (NSAIDs) and surgical interventions, can only control the disease, and the joint will lose its functionality after a short period. Consequently, modern methods such as cell therapy and tissue engineering along with using various biomaterials are being attempted to repair degenerated cartilage tissue. Using interfering RNAs is another approach that targets specific destructive or malfunctioned RNA sequences and suppresses the responsible factors for cartilage tissue destruction. Hence, the degenerated tissue can gradually retain the balance between anabolic and catabolic activities. Identification of the affecting genes in degeneration or malfunctioning and their suppression has provided promising results for the treatment of diseases. In the current study, after introducing the tissue, the process of cartilage degeneration and osteoarthritis development, the researches that have investigated the effect of interfering RNAs on rehabilitating cartilage tissue via inhibition of cartilage matrix destruction are reviewed.
