Hepatitis C virus is the main cause of hepatitis C infection that usually has no symptoms at early stages. Studies have shown changes in the expression level of microRNAs in various diseases such as hepatitis. MiR-199a and miR-196b have an inhibitory effect on hepatitis C virus. In a recent study, we examined the expression levels of miR-199a and miR-196b in serum samples of asymptomatic hepatitis C infected blood donors in comparison to serum samples of healthy blood donors; based on the results, the importance of microRNAs in different stages of hepatitis C infection was evaluated.
Materials and MethodsIn this case control study, first total RNA extraction from 106 serum samples of healthy blood donors (control group) and 106 serum samples of asymptomatic hepatitis C infected blood donors (case group) was carried out. After cDNA synthesis, the expression level of miR-199a and miR-196b were evaluated by SYBR Green real-time PCR, and the results were analyzed by REST 2009 software.
ResultsExpression level of miR-196b in the case group compared to the control group was lower (p 0.001). No statistically significant difference was observed in the expression level of miR-199a between the two groups.
Conclusions
With regard to lower expression level of miR-196b, and given no significant difference of miR-199a expression level in the case group in comparison to the control group, future studies on the role of miR-196b in this disease are required.

Change in the age structure of the population affects many activities and processes, and most countries are expected to enter the old population phase by 2050. Today, for various reasons, the consumption of blood and blood products has increased. The aim of this study was to investigate the age change in donor population during the years 1390 to 1397 and compare it with the societychr('39')s age change algorithm to determine if the latter affects the former.
Materials and MethodsIn this cross-sectional descriptive study, demographic information of 329631 blood donors from 1390 to 1397 from Blood Transfusion Software (Negareh software version 5/9) was extracted and the latest demographic information of the province was achieved using the website of the Statistics Center of Iran.
ResultsOf the population of Yazd and Ardakan cities, 64.7% and 65.38% are within the appropriate age range for blood donation. The donor population of 18-30 years decreased by 30.7% in 2016 and 32.3% in 2018 compared with the year 2011 while other age groups almost increased. Mean age of donors in 2014 and 2017 were 44.2 and 40.05 years, respectively.
ConclusionsDue to the aging population, and the reduction of the population under the age of 30, it is necessary to take effective programs to maintain and attract 18-30 year old population for blood donation.

Evaluation of cell survival is important in cellular studies and the result is decisive for the application of the test substance. Different methods are used to evaluate cell survival and the correlation between methods is important. Here, Vero cell line was used to evaluate the effects of HSV-1 virus on cell survival. Cell survival after HSV-1 infection using sensitive colorimetric methods of 3-, 4-, 5-dimethylthiazole-2-yl, -2, 5-diphenyltetrazolium bromide (MTT), and Trypan Blue on Vero cell line was examined.
Materials and MethodsA qualitative study was performed on Vero cell line infected with HSV-1 virus with logarithmic dilutions10-1 to 10-5 dilutions and time intervals of 0, 4, 8, 18 and 24 hours and the s in DMEM containing 2% Fetal bovine serum (FBS). Then cell viability by MTT and Trypan tests was examined . The survival percentage was used to plot the survival curve of the assays.
ResultsDifferent viral dilutions were used to determine the infectious dose of 50% (TCID50) of HSV-1virus with invert microscope. Acyclovir at a concentration of 50 mg/mL was compared as drug control for MTT and Trypan Blue. The results of regression analysis showed cell survival. Linear, positive and significant correlation was observed between the methods that showed a correlation between MTT and Trypan Blue methods (p< 0.001, r = 0.9(
Conclusions
The results showed that both methods can be used to determine cell survival and the results of MTT can be confirmed by Trypan Blue method.

Platelet activation occurs for a variety of reasons from the moment of sampling to that of injection. Activation of the platelets leads to activation of mitochondria. They move toward the membrane, sprouting and shedding freely from it, leading to the release of pro inflammatory mediators and mtDNA. Therefore, mtDNA level is considered as one of the parameters of platelet activity. The aim of this study was to compare mtDNA levels during storage.
Materials and MethodsThis experimental study was performed on 22 PRP products. We obtained consent of all donors referring to Iran Innovation Center of Iranian Blood Transfusion Organization. Samples were taken from the PRP bags on day 1, 3 and 5. Then, the fold change of mtDNA was assessed by Real time PCR. The results were analyzed by paired t-test and the P value of less than 0.05 was considered significant.
ResultsThe rate of mtDNA decreased 0.8-fold on day 3 to 1, 0.6-fold on day 5 to 3, and 0 .49 on day 5 to 1. These changes were significant on days 5 to 1 (p= 0.003) and they were not significant on days 3 to 1 (p= 0.4).
Conclusions
The amount of mtDNA increased on the first day, and then decreased during the storage of PRP products.

Only 40-50% of cross-matched blood bags are used for surgeries, which leads to wastage of blood bags. We investigated the effectiveness of antibody screening in improving blood consumption during surgeries.
Materials and MethodsThis cross-sectional study was conducted in Chamran and Kosar hospitals in1397-1398 in Shiraz. The number of requested, consumed and cross-matched blood bags and the ratio of cross-match to transfusion in two consecutive years (before and after antibody screening) in Chamran and Kowsar hospitals were compared for one month and five months. The comparison of ratio test with Medcalc-7 statistical software was used for analysis.
Results
Prior to the antibody screening program, 585 blood bags were requested, 228 crossmatched and only 84 were transfused in Chamran Hospital. After antibody screening 554 requested blood bags, 98 and 72 bags were crossmatched and transfused, respectively. The cross match -transfusion ratio was decresed from 2.71 to 1.36 (p< 0.001, 95% CI: 31-25-41 / 98: 95%). Prior to the antibody screening program, out of 3595 requested blood bags, 3377 and 1468 bags were cross-matched and transfused in Kowsar Hospital, respectively. After antibody screening, out of 2183 requested blood bags, 1658 and 1157 bags were crossmatched and transfused, respectively. The cross - match to transfusion ratio was decreased (2.30 to 1.43) (p< 0.001 < 24/28/29/32: 95% CI) after antibody screening.
Conclusions
Initiating antibody screening rationalized blood consumption. Therefore, it seems that it should be established in all hospitals, and coordination with insurance organizations should be done to allocate the budget and provide the required kits.

The effects of skipping rope on cardiovascular fitness as well as physical fitness were proved. Moreover, this exercise with high intensity may induce changes in hematological parameters and hemolysis, due to its concurrent foot strike. The aim of this study was to investigate the effect of physical training status on hemoglobin and ferritin and red blood cells counts following one session intense skipping rope exercise.
Materials and MethodsIn this quasi-experimental study, 10 male endurance athletes (Age 20.80 ± 2.34 yrs; Weight 68.30 ± 3.02 kg; Body Fat %10.10 ± 2.02) and 10 non-athlete men (Age 21.50 ± 1.71 yrs; Weight 74.40 ± 6.58 kg; Body Fat %:19.80 ± 2.85) were randomly selected from volunteers. Exercise protocol was one session skipping rope including 10-min warm up with 55% of target heart rate and then progressed to 85% for 30 min. Hemoglobin, hematocrit percent, serum ferritin and RBC count were measured pre and post-exercise.
ResultsResult showed that the increase of hemoglobin level to 14.78 ± 0.614 (p < 0.001), hematocrit percentage to 44.25 ± 1.870 (p = 0.003), and RBC to 5.26 ± 0.249 (p = 0.005) were significant only in non-athlete men. The ferritin levels did not change in both groups.
ConclusionsIt seems that the difference in responses of the two groups to acute rope skipping is related to the adaptations made in the previous exposure of endurance athletes to exercise. The use of intense skipping rope may be a good choice for those having an active lifestyle.

Platelet microparticles are microvesicles derived from platelets. Today, with the use of nanoparticles in cancer treatments, many limitations of traditional drug delivery methods are reduced. Doxorubicin, a chemotherapeutic drug available for the treatment of many cancers, has fluorescent properties and can be detected by fluorescent imaging in tissues. Aim of this study is to compare different drug loading methods into platelet microparticles.
Materials and MethodsIn this experimental study, the platelet concentrates were taken on their fifth day from Iranian Blood Transfusion Organization. Then platelet microparticles were extracted from those concentrated platelets in several centrifugation stages. Fluorescent microbeads with one-micrometer size and anti-CD41/61 antibody were used to determine the size and identity of microparticles, respectively. Doxorubicin was loaded at 10 µg/ml on platelet microparticles using three methods of incubation, cell-penetrating peptide, and Sonication. Using the auto-fluorescence property of Doxorubicin, the rate of drug loading on platelet microparticles was measured by flow cytometry method.
Results
In terms of size, 95% of the total population of microparticles was less than one micrometer. The expression levels of CD41 and CD61 were 92.39% and 80.03%, respectively. The average fluorescence light intensities calculated in each of the incubation, sonication, and CPP methods were determined to be 79.09% ± 11.37, 47.48% ± 25.12, and 56.69% ± 23.24, respectively.
Conclusions
As the incubation method has higher loading percentage, it could be an effective method for loading drug in platelet microparticles. Furthermore, the use of this method can be considered for clinical studies.
