    مقدمه

    رسوبات آبی رودخانه ها و دریای خزر به علت ایجاد شرایط مناسب، زیستگاه های میکروسکوپی متعددی را برای جامع بزرگی از اعضای میکروبی فراهم می کند که فعالانه به تولید آنزیم های سوختی بپردازند.

    مواد و روش‏‏ها:

     نمونه برداری رسوبات سطحی برخی رودخانه های مسیر شهرستان های رشت و انزلیرودخانه سفیدرود، سیاه رود، رود هراز و دریای خزر انجام شد. نمونه ها در محیط YGC آگار به روش پور پلیت کشت داده شدند. قارچ های حاصل از رشد روی پلیت های یادشده ازنظر حضور آنزیم های پروتیاز، آمیلاز، لیپاز، Dnase، کیتیناز، لسیتیناز و پکتیناز بررسی شدند؛ در ادامه، افتراق بین جدایه های مخمری با محیط های کروم آگار انجام و شناسایی مولکولی جدایه های جداسازی شده به روش PCR و در ادامه، توالی یابی محصولات PCR انجام شد.

    نتایج

    چهار جدایه بررسی شده طی سه مرحله نمونه برداری از چهارده ایستگاه از رسوبات رودخانه ها و دریای خزر پس از تهیه رقت های پی درپی و کشت روی YGC آگار جدا شدند. همه جدایه ها آنزیم لیپاز، DNase و آمیلاز و دو جدایه آنزیم پروتیاز و لسیتیناز را تولید می کردند. هیچ یک از جدایه ها توانایی تولید آنزیم کیتیناز و پکتیناز را نداشتند. دو جدایه SH002 و SH007 بیشترین تنوع آنزیمی را داشتند. شناسایی بیوشیمیایی و ریخت شناختی چهار جدایه و سپس شناسایی ژنتیکی دو جدایه SH002 و SH014 انجام شد که به ترتیب به گونه های Candida tropicalisو Candida fameta شبیه بودند.

    بحث و نتیجه‏ گیری: 

    بر اساس بررسی های انجام شده در منابع در دسترس، گزارش هایی از فعالیت پکتینازی و کیتینازی در مخمرهای جداشده از منابع مختلف مشاهده می شوند که در توضیح این مشاهده می توان گفت علاوه بر تاثیر عوامل، منطقه ای که مخمر از آن جداسازی شده است، ممکن است بر تولیدشدن یا تولیدنشدن این آنزیم ها تاثیرگذار باشد.

    مقدمه

    باکتری Pseudomonas syringae pv syringaeعامل چندین بیماری باکتریایی روی گیاهان زراعی و باغی علفی و چوبی است. افزایش مقاومت های آنتی بیوتیکی و عوارض جانبی نامطلوب ناشی از مصرف آنها تمایل به یافتن روش های جایگزین نظیر به کارگیری ترکیبات چون کیتوزان را که رویکردی بی خطر در مهار بیمارگرهای باکتریایی گیاهی است، افزایش داده است.

    مواد و روش‏‏ها: 

    در پژوهش حاضر، اثر رقت های مختلف کیتوزان با وزن مولکولی پایین روی سیستم حدنصاب احساس باکتری گزارشگر Pss B728a ارزیابی شد و درنهایت اثر کمترین رقت انتخابی موثر بر حرکت تجمعی و بیوفیلم باکتریPss 3289 بررسی شد.

    نتایج

    غلظت 01/0 گرم بر لیتر کیتوزان با وزن مولکولی پایین نه تنها توانست سیستم حدنصاب احساس در باکتری گزارشگر را مهار کند، بلکه میزان حرکت تجمعی و تشکیل بیوفیلم در باکتری Pss 3289 را که نقش مهمی در کلنیزاسیون گیاه دارد و تعیین کننده قدرت بیماری زایی این باکتری است نیز در حد مطلوبی مهار کرد.

    بحث و نتیجه‏ گیری: 

    باتوجه به توانایی کیتوزان در مهار سیستم تنظیمی حدنصاب احساس باکتری Pss، میزانحرکت تجمعی و تشکیل بیوفیلم در آن که دو خصوصیت مهم در بیماری زایی این باکتری است و با درنظرگرفتن روند افزایش مقاومت آنتی بیوتیکی در بیمارگرهای باکتریایی، به کارگیری ترکیباتی چون کیتوزان می تواند جایگزین مناسبی برای مهار این بیمارگرها باشد.

    مقدمه

    امروزه، آلودگی فلزات سنگین یکی از مهم ترین مشکلات محیط زیست است و باتوجه به هزینه بربودن روش های متداول پالایش، استفاده از زیست توده میکروبی برای پالایش فلزات سنگین پیشنهاد می شود. مطالعه حاضر با هدف ارزیابی میان کنش مخمر رودوترولا تورلوییدس سویه IR-1395 با جیوه انجام شد.

    مواد و روش‏‏ها:

     در مطالعه حاضر، میزان رشد و انواع برهم کنش سویه IR-1395 با جیوه در محیط ساکارز براث بررسی و مقدار جیوه تجمع یافته در محیط و در مخمر به روش اسپکتروفتومتری اندازه گیری شد؛ سپس به منظور تعیین بهترین شرایط جذب زیستی، تاثیر شاخص های اسیدیته، غلظت جیوه، غلظت زیست توده، زمان مجاورسازی و دما سنجیده شد؛ همچنین میزان جذب توسط مخمر تیمارشده با 2 و 4 دی نیتروفنل و اتوکلاو بررسی و درنهایت، جذب زیستی جیوه توسط مخمر با میکروسکوپ الکترونی نگاره مجهز به طیف سنجی پراش انرژی پرتو ایکس بررسی شد.

    نتایج

    بررسی ها نشان دادند بیشترین رشد مخمر طی 36 ساعت رخ می دهد و پس از آن، وارد فاز سکون می شود؛ همچنین این سویه قادر به جذب (جذب زیستی و تجمع زیستی) 86/61 درصد جیوه و فرارسازی 17/10 درصد جیوه از محیط دارای 10 میلی گرم برلیتر کلریدجیوه طی هفت روز است. در بررسی عوامل تاثیرگذار بر جذب زیستی جیوه مشخص شد اسیدیته بهینه برای این مخمر برابر 4 است و بیشترین میزان جذب در غلظت 300 میلی گرم برلیتر جیوه و در دمای 15 درجه سانتی گراد به مدت10 دقیقه مشاهده می شود. در بررسی اثر تیمار 2 و 4 دی نیتروفنل و اتوکلاو مشخص شد میزان جذب توسط مخمر تیمارشده کاهش می یابد. نتایج طیف سنجی پراش انرژی پرتو ایکس جذب زیستی جیوه توسط مخمر را تایید کردند.

    بحث و نتیجه‏ گیری: 

    بررسی انواع برهم کنش های سویه IR-1395 با جیوه و پتاسیل آن در جذب زیستی نشان داد این سویه، ریزموجود باارزشی در فرایندهای زیست پالایی جیوه از محیط های آبی آلوده است.

    مقدمه

    عوامل ژنتیکی و محیطی مختلفی با بیماری مالتیپل اسکلروزیس (MS)و پیشرفت آن ارتباط دارند؛ ازجمله عوامل محیطی موثر می توان به عوامل عفونی و به ویژه قارچ ها اشاره کرد. مبنای ایمونولوژیک بیماری MS و آثار عفونت های قارچی بر سیستم ایمنی سبب شد فراوانی نسبی ابتلا به عفونت های قارچی در بیماران مبتلا به MS با افراد شاهد مقایسه شود.

    مواد و روش‏‏ها:

     تعداد 100 نمونه سرم بیمار مبتلا به بیماری MS و افراد شاهد جمع آوری شدند. پس از استخراج DNA، اختصاصیت و حساسیت آزمون PCR در زمینه آغازگرهای عمومی قارچی بررسی و سپس آزمون PCR بهینه روی نمونه های شاهد و بیمار انجام شد.

    نتایج

    قطعه 575 جفت بازی به کمک آغازگرهای عمومی تکثیر و روی ژل آگارز مشاهده شد. آزمون PCR به منظور بررسی اختصاصیت با DNAهای قارچ، باکتری و ویروس انجام شد و تکثیر تنها با DNA قارچی مشاهده شد و تکثیری با سایر DNAها مشاهده نشد. تعداد 11 نمونه از 100 نمونه بیمار و تعداد 4 نمونه از 100 نمونه سالم مثبت شد؛ همچنین حد تشخیص در این واکنش برابر 40 کپی DNA به دست آمد. بررسی نتایج با نرم افزار SPSS و آزمون دقیق فیشر نشان دادند تفاوت معناداری (p value=0.052) بین گروه بیمار و شاهد وجود دارد.

    بحث و نتیجه‏ گیری: 

    بیماران MS به علت سیستم ایمنی ضعیف تر و استفاده از داروهای سرکوب کننده سیستم ایمنی، فراوانی نسبی بیشتری در میزان آلودگی به عفونت های قارچی دارند. وجود عامل عفونی فعال در این بیماران می تواند مدت زمان تخریب سیستم عصبی را افزایش دهد؛ ازاین رو، تشخیص عامل عفونی فعال و تجویز رژیم دارویی متناسب می تواند عامل کمک کننده ای در جلوگیری از پیشرفت بیماری تلقی شود و بنابراین، لازم است مطالعه های بیشتری در زمینه انواع گونه های قارچی موثر و سازوکار بیماری زایی آنها انجام شود.

    مقدمه

    آلودگی ایجادشده از استخراج و فراوری نفت، موجب تحریک رشد باکتری های تجزیه کننده هیدروکربن های نفتی شده است. زیست پالایی فناوری امیدبخشی برای تصفیه آب و خاک های آلوده با بهره مندی از میکروارگانیسم های تجزیه کننده ترکیبات نفتی است. هدف از این پژوهش، جداسازی باکتری های تجزیه کننده ترکیبات نفتی و بررسی قابلیت استفاده از آن در زیست پالایی خاک های آلوده به ترکیبات نفتی است.

    مواد و روش‏‏ها:

     نمونه برداری از خاک آلوده یک منطقه پالایشگاهی صورت گرفت. از محیط کشت معدنی حاوی n-هگزان برای جداسازی استفاده شد. از روش های بیوشیمیایی و تعیین توالی 16S rRNAباکتری های جداسازی شده شناسایی شدند. آزمون سمیت خاک با اندازه گیری میزان درصد جوانه زنی بذر گیاه شاهی صورت گرفت. زیست پالایی خاک آلوده با استفاده از سویه های منتخب در آزمایشگاه و در محیط، طی یک دوره سه ماهه انجام شد.

    نتایج

    تعداد 19 سویه باکتریایی با قابلیت رشد برروی محیط کشت معدنی حاوی n-هگزان جداسازی شدند. از میان سویه های جداسازی شده، چهار سویه از توانایی بیشتری برای تجزیه زیستی ترکیبات نفتی آنتراسن، فنانترن و نفتالین برخوردار بودند. با روش شناسایی مولکولی، سویه های منتخب با عنوان Stenotrophomonas maltophilia (SB10)،Bacillus subtilis subsp. spizizenii (SB7)، Streptomyces ambofaciens (SB14)و Cupriavidus respiraculi(SB16)شناسایی شدند. تیمار خاک آلوده نفتی با استفاده از سویه S. maltophilia (SB10)، موجب افزایش جمعیت باکتری های تجزیه کننده هیدروکربن های نفتی از 103×2/5 به 104×4/1 در پایان ماه سوم شد. میزان تجزیه زیستی کل هیدروکربن های نفتی در پایان ماه سوم، به میزان 80درصد و قدرت جوانه زنی گیاه شاهی در خاک تیمارشده به 68 درصد رسید.

    بحث و نتیجه‏ گیری:

     افزودن سویه های باکتریایی منتخب به خاک آلوده به ترکیبات نفتی، موجب افزایش جمعیت باکتری های تجزیه کننده شد.استفاده از باکتری های تجزیه کننده همراه با خاک اره موجب بهبود پاک سازی خاک های آلوده نفتی شد.

    مقدمه

    یکی از روش های نوین برای کاهش آلودگی های ناشی از کاربرد علف کش ها و سایر سموم شیمیایی کشاورزی، استفاده از جدایه های باکتریایی متحمل و تجزیه کننده این علف کش هاست. پژوهش حاضر به منظور جداسازی و شناسایی جدایه های باکتریایی مقاوم یا متحمل در برابر علف کش های اتللو، آتلانتیس و پوماسوپر از خاک مزارع گندم انجام شد.

    مواد و روش‏‏ها: 

    نمونه برداری از خاک مزارع گندم تیمار شده با سه علف کش اتللو، آتلانتیس و پوماسوپر انجام شد. جدایه های باکتریایی با استفاده از محیط کشت سوکسینات استاندارد حاوی 10 درصد از علف کش های آزمایش شده جداسازی شدند. جدایه های نماینده بر اساس آزمون های بیوشیمیایی و تکثیر ژن 16S rRNA شناسایی شدند. کمترین غلظت مهارکنندگی و کمترین غلظت کشندگی برای هر سه علف کش آزمایش شده محاسبه شد.

    نتایج

    درمجموع، تعداد 17 جدایه توانایی رشد روی محیط های رشد حاوی 10 درصد از علف کش های آزمایش شده را داشتند. بر اساس کلیدهای باکتری شناسی معتبر و نتایج آزمون های مولکولی روی جدایه های نماینده، استنوتروفوموناس مالتوفیلیا، انسیفر ادهایرنس، زانتوموناس، سودوزانتوموناس و اسینتوباکتر شناسایی شدند. کمترین غلظت مهارکنندگی و کمترین غلظت کشندگی در علف کش های اتللو (به ترتیب 28/0 و 4/0) و آتلانتیس (به ترتیب 35/0 و 5/0) برای اسینتوباکتر و در علف کش پوماسوپر برای جنس زانتوموناس (به ترتیب 45/0 و 67/0) محاسبه شد.

    بحث و نتیجه‏ گیری: 

    باتوجه به نتایج پژوهش حاضر، شناسایی جدایه های متحمل و تعیین میزان تحمل آنها به علف کش های یادشده، گامی مهم در پژوهش های زیست فناوری آینده به منظور پالایش آلاینده ها از محیط های آلوده است و می تواند مشکلات زیست محیطی ناشی از این علف کش ها در اکوسیستم را کاهش دهد؛ ازاین رو، پژوهش های بیشتر برای تعیین توانایی جدایه های باکتریایی مقاوم در تجزیه کنندگی علف کش های یادشده پیشنهاد می شود.

    Introduction

    The aquatic sediments of the rivers and the Caspian Sea provide numerous microscopic habitats for a large community of microbial organisms that are actively producing fuel enzymes due to favorable conditions.

    Materials and methods

    Surface sediment sampling of the Sefidrood, Siahrood, and Haraz rivers as well as the Caspian Sea was carried out. The samples were cultured in YGC agar medium by the pour plate method. Fungi grown on these plates were examined for the presence of Protease, Amylase, Lipase, Dnase, Chitinase, Lectinase and Pectinase. Then, the differentiation between yeast isolates was performed by chromagar media; the molecular identification of the isolated yeasts was performed by PCR and sequencing of PCR products.

    Results

    A set of four isolates was isolated from fourteen stations in three stages by sampling of rivers and Caspian sediments after continuous sequencing and cultivation on YGC Agar. All isolates produced Lipase, DNase and Amylase. Two isolates produced Protease and Lisetinase. None of the isolates had the ability to produce Chitinase and Pectinase. Two isolates, SHOO2 and SHOO7, had the most diverse enzyme production. Biochemical and morphological identification of four isolates was also performed accurately, and then genetic identification of two isolates SHOO2 and SHOO14 was performed, which were similar to Candida tropicalis species and Candida fameta, respectively.

    Discussion and conclusion

    Based on the available sources, there have been reports of Pectinase and Kinase activity in yeasts isolated from different sources. To justify this observation, it can be said that the region was affected by the isolation and the enzymatic activity of the isolated yeasts.Aquatic sediments can be a good source of yeasts with the ability to produce a wide range of useful enzymes needed in the industry.

    Introduction

    Pseudomonas syringae pv. syringae is the causal agent of several bacterial diseases on a wide range of horticultural and agricultural herbaceous or woody plants. Increasing resistance to antibiotics and undesirable side effects arising from their usage, enhance the trend of finding alternative methods in controlling plant bacterial pathogens using compounds such as chitosan as a safe way.

    Material and methods

    In this research the effect of different dilutions of Low Molecular Weight (50-190 kDa) chitosan were evaluated on the quorum sensing system of P. syringae pv. syringae B728a biosensor. Finally the effect of the lowest selected dilution was assessed on P. syringae pv. syringae 3289 swarming motility and biofilm formation.

    Results

    0.01gr/l LMW chitosan not only could control quorum sensing system in biosensor bacterium, but also decreased P. syringae pv. syringae 3289 swarming motility and biofilm formation (that plays important role in plant colonization and determine virulence severity of this bacterium).

    Discussion and conclusion

    According to chitosan role in controlling P. syringae pv. syringae quorum sensing system and its swarming motility and biofilm formation (that are two important factors in this bacterium virulence) and with increasing resistance to antibiotices in bacterial pathogeneses, using compounds such as chitosan can be a good replacement for controlling these pathogens.

    Introduction

    Nowadays, heavy metals pollution is one of the most important environmental problems. Due to the high cost of common refining methods, the use of microbial biomass is recommended to clean up heavy metals. The aim of this study was the evaluation of interactions of Rhodoterula toruloides strain IR-1395 with mercury.

    Materials and methods

    In this study, the growth rate and interactions of strain IR-1395 with mercury in Sucrose Broth medium were investigated and the amount of mercury accumulated was measured using the Spectrophotometry method. Then, in order to determine the best biosorption conditions, the effect of parameters such as pH, mercury concentration, biomass concentration, contact time and temperature were measured. Also, the biosorption was investigated by treated cells with 2, 4 dinitrophenol and autoclave. Finally, the biosorbent was characterized by Scanning Electron Microscope with Energy Dispersive X-Ray Analysis.

    Results

    Studies showed that the maximum growth rate of strain IR-1395 was after 36h and then it enters the stationary phase. Also, this strain was able to the biosorption and bioaccumulation (61.86%) and 10.17% biovolatilization of mercury from the medium at the concentration of 10 mg/l of chloride mercury during seven days. In study of factors affecting on the mercury biosorption, it is indicated that the optimal pH was 4. The highest biosorption was observed at 300 mg /l of mercury in 10 minutes at 15 ˚C. Mercury adsorption using treated biomass by autoclaved and 2,4 dinitrophenoltreatment has reduced than control samples. Finally, the results of the EDX confirmed the biosorption of mercury by this yeast.

    Discussion and conclusion

    By study of the various interactions of strain IR-1395 with mercury and its potential in biosorption, it was found that this strain is a valuable microorganism in mercury bioremediation processes from contaminated aquatic environments.

    Introduction

    Various genetic and environmental factors are associated with multiple sclerosis (MS) and its progression. Environmental factors include infectious agents especially fungi. The immunological basis of MS and the effects of fungal infections on the immune system led us to compare the relative frequency of fungal infections in the serum of MS patients with controls.

    Materials and methods

    One-hundred serum samples of patients with MS and control individuals were collected. After the DNA extraction, the specificity and sensitivity of the PCR test for universal fungal primers were investigated. Then, an optimal PCR test was performed on control and patient samples.

    Results

    In this study, 575 base pairs (bp) of genes were considered as the target product using universal fungal primers. The PCR test was performed to check for the specificity with fungal, bacterial, and viral DNAs, along with fungal DNA replication and other reproductive DNAs. The positive cases were 11 and 4 among 100 patients and 100 healthy samples, respectively. Also, the detection limit in this reaction was 40 copies of DNA. The results using SPSS software and Fisher's exact test showed a significant difference between the two groups (P value= 0.052).

    Discussion and conclusion

    MS patients have a higher relative frequency of fungal infections due to weaker immune systems and immunosuppressive drugs. The presence of an active infectious agent in such patients can increase the duration of the destruction of the nervous system. Therefore, diagnosing the active infectious agent and prescribing an appropriate medication regimen can be considered as a contributing factor in preventing the progression of the disease. Therefore, more studies are needed on the types of effective fungal species and their pathogenesis mechanism.

    Introduction

    Pollution caused by oil extraction processing stimulates the growth of petroleum hydrocarbon-degrading bacteria. Bioremediation, a process that utilizes the capability of microorganisms to degrade petroleum hydrocarbon compounds, is emerging as a promising technology for the treatment of soil and groundwater contamination. This paper aimed to separate petroleum-degrading bacteria and to investigate their applicability in the bioremediation of petroleum-contaminated soils.

    Materials and methods

    Sampling was done from the soil of a refinery area. A mineral culture-medium environment containing n-hexane was used for isolation. Isolated bacteria were identified by biochemical methods and 16S rRNA sequencing. Phytotoxicity test was performed by measuring the percentage of Lepidium sativumm seed germination. Contaminated soil bioremediation was performed using the selected strains in the laboratory and the environment over three months.

    Results

    A total of 19 bacteria isolates capable of growing on mineral medium containing n-hexane were isolated. Among the isolated strains, four strains had the highest ability to biodegrade the petroleum compounds of anthracene, naphthalene, and phenanthrene. By the molecular identification method, the selected strains were identified as Stenotrophomonas maltophilia (SB10),  Bacillus subtilis subsp.spizizenii (SB7),  Streptomyces ambofaciens (SB14), andCupriavidus respiraculi (SB16).  The treatment of petroleum-contaminated soils using S. maltophilia (SB10) increased the population of petroleum-degrading bacteria from 5.2×103 to 1.4×104 at the end of the third month. The rate of biodegradation of the total petroleum hydrocarbons reached 80% and the germination power of Lepidium sativumm reached 68% at the end of the third month.

    Discussion and conclusion

    The addition of the selected bacterial strains to soil contaminated with petroleum compounds increased the population of degrading bacteria. The use of degrading bacteria along with sawdust improved the purification of petroleum-contaminated soils.

    Introduction

    One of the new methods in reducing the pollution caused by the use of herbicides and other agricultural chemical pesticides is the use of resistant and degrading bacterial isolates of these herbicides. This study was conducted to isolate and identify resistant bacterial strains from the soil of wheat fields using the three herbicides of Othello, Atlantis, and Puma Super.

    Materials and methods

    To isolate and characterize resistant bacteria, samples were collected from wheat fields treated with Othello, Atlantis, and Puma Super herbicides. Isolates were selected based on the growth on the Standard Succinate Medium containing 10 percent of the mentioned herbicides. The representative isolates were identified based on biochemical tests and amplification of the 16S rRNA gene. Also, the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of the representatives were measured.

    Results

    A total of 17 isolates could grow on a growth medium containing 10% of the tested herbicides. Based on the valid biochemical and molecular test results, the representative strains were identified as Stenotrophomonas maltophilia, Ensifer adhaerens, Xanthomonas sp., Pseudoxanthomonas sp., and Acinetobacter sp. The Minimum inhibitory concentration (MIC) and Minimum Bactericidal Concentration (MBC) were attributed to Othello (0.28 and 0.4) and Atlantis herbicides (0.35 and 0.5) belonging to Acinetobacter and Xanthomonas (0.45 and 0.67) for Puma Super herbicides.

    Discussion and conclusion

    According to the results of this study, herbicide-resistant bacteria identified in this study can be useful in plant biotechnology applications in polluted areas and can reduce environmental pollution regarding using these herbicides. However, further studies are needed in order to identify the degrading potential of the resistant strains.
